Abstract

An overview of matter-coupled ${\cal N}=2$ supergravity theories with 8 real supercharges, in 4,5 and 6 dimensions is given. The construction of the theories by superconformal methods is explained from basic principles. Special geometry is obtained and characterized. The relation between the theories in those dimensions is discussed. This leads to the concepts of very special geometry and quaternionic-K\"ahler manifolds, whose structures are explained., Comment: 196 pages. Abstract

We propose to build and operate a detector that, for the first time, will measure the process $pp\to\nu X$ at the LHC and search for feebly interacting particles (FIPs) in an unexplored domain. The TI18 tunnel has been identified as a suitable site to perform these measurements due to very low machine-induced background. The detector will be off-axis with respect to the ATLAS interaction point (IP1) and, given the pseudo-rapidity range accessible, the corresponding neutrinos will mostly come from charm decays: the proposed experiment will thus make the first test of the heavy flavour production in a pseudo-rapidity range that is not accessible by the current LHC detectors. In order to efficiently reconstruct neutrino interactions and identify their flavour, the detector will combine in the target region nuclear emulsion technology with scintillating fibre tracking layers and it will adopt a muon identification system based on scintillating bars that will also play the role of a hadronic calorimeter. The time of flight measurement will be achieved thanks to a dedicated timing detector. The detector will be a small-scale prototype of the scattering and neutrino detector (SND) of the SHiP experiment: the operation of this detector will provide an important test of the neutrino reconstruction in a high occupancy environment., Comment: Letter of Intent

Abstract

Archived specimens, taken by standardized procedures in clinical practice, represent a valuable resource in translational medicine. Their use in retrospective molecular-based studies could provide disease and therapy predictors. Microfluidic array is a user-friendly and cost-effective method allowing profiling of hundreds of microRNAs (miRNAs) from a low amount of RNA. However, even though tissue miRNAs may include potentially robust biomarkers, non-uniformed post-analytical pipelines could hinder translation into clinics. In this study, epidermal RNA from archival skin biopsy specimens was isolated from patients with peripheral neuropathy and healthy individuals. Unbiased miRNA profiling was performed using RT-qPCR-based microfluidic array. We demonstrated that RNA obtained from archival tissue is appropriate for miRNA profiling, providing evidence that different practices in threshold selection could significantly influence the final results. We showed the utility of software-based quality control for amplification curves. We revealed that selection of the most stable reference and the calculation of geometric mean are suitable when utilizing microfluidic arrays without known references. By applying appropriate post-analytical settings, we obtained miRNA profile of human epidermis associated with biological processes and a list of suitable references. Our results, which outline technical and post-analytical considerations, support the broad use of archived specimens for miRNA analysis to unravel disease-specific molecular signatures.

Abstract

An event topology with two secondary vertices compatible with the decay of short-lived particles was found in the analysis of neutrino interactions in the Opera target. The observed topology is compatible with tau neutrino charged current (CC) interactions with charm production and neutrino neutral current (NC) interactions with $c\overline{c}$ pair production. However, other processes can mimic this topology. A dedicated analysis was implemented to identify the underlying process. A Monte Carlo simulation was developed and complementary procedures were introduced in the kinematic reconstruction. A multivariate analysis technique was used to achieve an optimal separation of signal from background. Most likely, this event is a $\nu_{\tau}$ CC interaction with charm production, the tau and charm particle decaying into 1 prong and 2 prongs, respectively. 218. Conformal boundary conditions for a 4d scalar field

Abstract

We construct unitary, stable, and interacting conformal boundary conditions for a free massless scalar in four dimensions by coupling it to edge modes living on a boundary. The boundary theories we consider are bosonic and fermionic QED$_3$ with $N_f$ flavors and a Chern-Simons term at level $k$, in the large-$N_f$ limit with fixed $k/N_f$. We find that interacting boundary conditions only exist when $k≠ 0$. To obtain this result we compute the $\beta$ functions of the classically marginal couplings at the first non-vanishing order in the large-$N_f$ expansion, and to all orders in $k/N_f$ and in the couplings. To check vacuum stability we also compute the large-$N_f$ effective potential. We compare our results with the the known conformal bootstrap bounds.

Abstract

Food residues are a promising resource for obtaining natural pigments, which may replace artificial dyes in the industry. However, their use still presents challenges due to the lack of suitable sources and the low stability of these natural compounds when exposed to environmental variations. In this scenario, the present study aims to identify different food residues (such as peels, stalks, and leaves) as potential candidates for obtaining natural colorants through eco-friendly extractions, identify the colorimetric profile of natural pigments using the RGB color model, and develop alternatives using nanotechnology (e.g., liposomes, micelles, and polymeric nanoparticles) to increase their stability. The results showed that extractive solution and residue concentration influenced the RGB color profile of the pigments. Furthermore, the external leaves of Brassica oleracea L. var. capitata f. rubra, the peels of Cucurbita maxima, Cucurbita maxima x Cucurbita moschata, and Beta vulgaris L. proved to be excellent resources for obtaining natural pigments. Finally, the use of nanotechnology proved to be a viable alternative for increasing the stability of natural colorants over storage time.

Abstract

(1) Mesenchymal stem cells (MSCs) are a valuable cell model to study the bone pathology of Osteogenesis Imperfecta (OI), a rare genetic collagen-related disorder characterized by bone fragility and skeletal dysplasia. We aimed to generate a novel OI induced mesenchymal stem cell (iMSC) model from induced pluripotent stem cells (iPSCs) derived from human dermal fibroblasts. For the first time, OI iMSCs generation was based on an intermediate neural crest cell (iNCC) stage. (2) Skin fibroblasts from healthy individuals and OI patients were reprogrammed into iPSCs and subsequently differentiated into iMSCs via iNCCs. (3) Successful generation of iPSCs from acquired fibroblasts was confirmed with changes in cell morphology, expression of iPSC markers SOX2, NANOG, and OCT4 and three germ-layer tests. Following differentiation into iNCCs, cells presented increased iNCC markers including P75NTR, TFAP2A, and HNK-1 and decreased iPSC markers, shown to reach the iNCC stage. Induction into iMSCs was confirmed by the presence of CD73, CD105, and CD90 markers, low expression of the hematopoietic, and reduced expression of the iNCC markers. iMSCs were trilineage differentiation-competent, confirmed using molecular analyses and staining for cell-type-specific osteoblast, adipocyte, and chondrocyte markers. (4) In the current study, we have developed a multipotent in vitro iMSC model of OI patients and healthy controls able to differentiate into osteoblast-like cells.

Abstract

Amyotrophic lateral sclerosis (ALS) is a progressive, lethal neurodegenerative disease mostly affecting people around 50–60 years of age. TDP-43, an RNA-binding protein involved in pre-mRNA splicing and controlling mRNA stability and translation, forms neuronal cytoplasmic inclusions in an overwhelming majority of ALS patients, a phenomenon referred to as TDP-43 proteinopathy. These cytoplasmic aggregates disrupt mRNA transport and localization. The axon, like dendrites, is a site of mRNA translation, permitting the local synthesis of selected proteins. This is especially relevant in upper and lower motor neurons, whose axon spans long distances, likely accentuating their susceptibility to ALS-related noxae. In this work we have generated and characterized two cellular models, consisting of virtually pure populations of primary mouse cortical neurons expressing a human TDP-43 fusion protein, wt or carrying an ALS mutation. Both forms facilitate cytoplasmic aggregate formation, unlike the corresponding native proteins, giving rise to bona fide primary culture models of TDP-43 proteinopathy. Neurons expressing TDP-43 fusion proteins exhibit a global impairment in axonal protein synthesis, an increase in oxidative stress, and defects in presynaptic function and electrical activity. These changes correlate with deregulation of axonal levels of polysome-engaged mRNAs playing relevant roles in the same processes. Our data support the emerging notion that deregulation of mRNA metabolism and of axonal mRNA transport may trigger the dying-back neuropathy that initiates motor neuron degeneration in ALS.

223. Non-tariff Protectionism in Mercosur: Analysis and Recommendations on the Last Three Decades

Abstract

Abstract This paper aims to discuss and demonstrate the non-tariff challenges to Mercosur’s trade integration. Its Common External Tariff (CET) and exceptions will be discussed, as well as the evolution of its non-tariff protectionism framework in the last three decades. This is an unprecedented exploratory exercise on all 5019 non-tariff measures (NTM) notified by Mercosur members to the WTO from 1995 to 2020. As main results, we see that (i) members with fewer exceptions to the CET are the ones that most use NTMs to protect their markets; (ii) the stock of NTMs targeted at members and non-members varies accordingly; (iii) of the 10 countries most targeted by NTMs imposed by Mercosur members, its own members figure in the 3rd, 4th, 7th, and 8th positions; (iv) the bloc lacks deep regulatory harmonization, especially with regard to sanitary and phytosanitary barriers in 11 sectors. Based on these results, two critical policy action plans are suggested to the bloc.

Abstract

Summary: Background: While vaccines have established utility against COVID-19, phase 3 efficacy studies have generally not comprehensively evaluated protection provided by previous infection or hybrid immunity (previous infection plus vaccination). Individual patient data from US government-supported harmonized vaccine trials provide an unprecedented sample population to address this issue. We characterized the protective efficacy of previous SARS-CoV-2 infection and hybrid immunity against COVID-19 early in the pandemic over three-to six-month follow-up and compared with vaccine-associated protection. Methods: In this post-hoc cross-protocol analysis of the Moderna, AstraZeneca, Janssen, and Novavax COVID-19 vaccine clinical trials, we allocated participants into four groups based on previous-infection status at enrolment and treatment: no previous infection/placebo; previous infection/placebo; no previous infection/vaccine; and previous infection/vaccine. The main outcome was RT-PCR-confirmed COVID-19 >7–15 days (per original protocols) after final study injection. We calculated crude and adjusted efficacy measures. Findings: Previous infection/placebo participants had a 92% decreased risk of future COVID-19 compared to no previous infection/placebo participants (overall hazard ratio [HR] ratio: 0.08; 95% CI: 0.05–0.13). Among single-dose Janssen participants, hybrid immunity conferred greater protection than vaccine alone (HR: 0.03; 95% CI: 0.01–0.10). Too few infections were observed to draw statistical inferences comparing hybrid immunity to vaccine alone for other trials. Vaccination, previous infection, and hybrid immunity all provided near-complete protection against severe disease. Interpretation: Previous infection, any hybrid immunity, and two-dose vaccination all provided substantial protection against symptomatic and severe COVID-19 through the early Delta period. Thus, as a surrogate for natural infection, vaccination remains the safest approach to protection. Funding: National Institutes of Health.

Abstract

PurposeRecent advancements in obtaining image-based biomarkers from CT images have enabled lung function characterization, which could aid in lung interventional planning. However, the regional heterogeneity in these biomarkers has not been well documented, yet it is critical to several procedures for lung cancer and COPD. The purpose of this paper is to analyze the interlobar and intralobar heterogeneity of tissue elasticity and study their relationship with COPD severity.MethodsWe retrospectively analyzed a set of 23 lung cancer patients for this study, 14 of whom had COPD. For each patient, we employed a 5DCT scanning protocol to obtain end-exhalation and end-inhalation images and semi-automatically segmented the lobes. We calculated tissue elasticity using a biomechanical property estimation model. To obtain a measure of lobar elasticity, we calculated the mean of the voxel-wise elasticity values within each lobe. To analyze interlobar heterogeneity, we defined an index that represented the properties of the least elastic lobe as compared to the rest of the lobes, termed the Elasticity Heterogeneity Index (EHI). An index of 0 indicated total homogeneity, and higher indices indicated higher heterogeneity. Additionally, we measured intralobar heterogeneity by calculating the coefficient of variation of elasticity within each lobe.ResultsThe mean EHI was 0.223 ± 0.183. The mean coefficient of variation of the elasticity distributions was 51.1% ± 16.6%. For mild COPD patients, the interlobar heterogeneity was low compared to the other categories. For moderate-to-severe COPD patients, the interlobar and intralobar heterogeneities were highest, showing significant differences from the other groups.ConclusionWe observed a high level of lung tissue heterogeneity to occur between and within the lobes in all COPD severity cases, especially in moderate-to-severe cases. Heterogeneity results demonstrate the value of a regional, function-guided approach like elasticity for procedures such as surgical decision making and treatment planning.

Abstract

The OPERA experiment has conclusively observed the appearance of tau neutrinos in the muon neutrino CNGS beam. Exploiting the OPERA detector capabilities, it was possible to isolate high purity samples of $\nu_{e}$, $\nu_{\mu}$ and $\nu_{\tau}$ charged current weak neutrino interactions, as well as neutral current weak interactions. In this Letter, the full dataset is used for the first time to test the three-flavor neutrino oscillation model and to derive constraints on the existence of a light sterile neutrino within the framework of the $3+1$ neutrino model. For the first time, tau and electron neutrino appearance channels are jointly used to test the sterile neutrino hypothesis. A significant fraction of the sterile neutrino parameter space allowed by LSND and MiniBooNE experiments is excluded at 90% C.L. Abstract

A four-dimensional Abelian gauge field can be coupled to a 3d CFT with a $U(1)$ symmetry living on a boundary. This coupling gives rise to a continuous family of boundary conformal field theories (BCFT) parametrized by the gauge coupling $\tau$ in the upper-half plane and by the choice of the CFT in the decoupling limit $\tau \to \infty$. Upon performing an $SL(2,\mathbb{Z})$ transformation in the bulk and going to the decoupling limit in the new frame, one finds a different 3d CFT on the boundary, related to the original one by Witten's $SL(2, \mathbb{Z})$ action [1]. In particular the cusps on the real $\tau$ axis correspond to the 3d gauging of the original CFT. We study general properties of this BCFT. We show how to express bulk one and two-point functions, and the hemisphere free-energy, in terms of the two-point functions of the boundary electric and magnetic currents. We then consider the case in which the 3d CFT is one Dirac fermion. Thanks to 3d dualities this BCFT is mapped to itself by a bulk $S$ transformation, and it also admits a decoupling limit which gives the $O(2)$ model on the boundary. We compute scaling dimensions of boundary operators and the hemisphere free-energy up to two loops. Using an $S$-duality improved ansatz, we extrapolate the perturbative results and find good approximations to the observables of the $O(2)$ model. We also consider examples with other theories on the boundary, such as large-$N_f$ Dirac fermions --for which the extrapolation to strong coupling can be done exactly order-by-order in $1/N_f$-- and a free complex scalar., Comment: 69 pages

Abstract Objective Spinal cord degeneration is a hallmark of amyotrophic lateral sclerosis. The assessment of gray matter and white matter cervical spinal cord atrophy across clinical stages defined using the King's staging system could advance the understanding of amyotrophic lateral sclerosis progression. Methods We assessed the in vivo spatial pattern of gray and white matter atrophy along cervical spinal cord (C2 to C6 segments) using 2D phase‐sensitive inversion recovery imaging in a cohort of 44 amyotrophic lateral sclerosis patients, evaluating its change across the King's stages and the correlation with disability scored by the amyotrophic lateral sclerosis functional rating scale revised (ALSFRS‐R) and disease duration. A mathematical model inferring the potential onset of cervical gray matter atrophy was developed. Results In amyotrophic lateral sclerosis patients at King's stage 1, significant cervical spinal cord alterations were mainly identified in gray matter, whereas they involved both gray and white matter in patients at King's stage ≥ 2. Gray and white matter areas correlated with clinical disability at all cervical segments. C3–C4 level was the segment showing early gray matter atrophy starting about 7 to 20 months before symptom onset according to our model. Interpretation Our findings suggest that cervical spinal cord atrophy spreads from gray to white matter across King's stages in amyotrophic lateral sclerosis, making spinal cord magnetic resonance imaging an in vivo assessment tool to measure the progression of the disease.

Abstract

Long non-coding RNAs (lncRNAs) regulate gene expression through different molecular mechanisms, including DNA binding via the formation of RNA:DNA:DNA triple helices (TPXs). Despite the increasing amount of experimental evidence, TPXs investigation remains challenging. Here we present 3plex, a software able to predict TPX interactions in silico. Given an RNA sequence and a set of DNA sequences, 3plex integrates 1) Hoogsteen pairing rules that describe the biochemical interactions between RNA and DNA nucleotides, 2) RNA secondary structure prediction and 3) determination of the TPX thermal stability derived from a collection of TPX experimental evidences. We systematically collected and uniformly re-analysed published experimental lncRNA binding sites on human and mouse genomes. We used these data to evaluate 3plex performance and showed that its specific features allow a reliable identification of TPX interactions. We compared 3plex with the other available software and obtained comparable or even better accuracy at a fraction of the computation time. Interestingly, by inspecting collected data with 3plex we found that TPXs tend to be shorter and more degenerated than previously expected and that the majority of analysed lncRNAs can directly bind to the genome by TPX formation. Those results suggest that an important fraction of lncRNAs can exert its biological function through this mechanism. The software is available at https://github.com/molinerisLab/3plex.

Abstract

The paper deals with the separation of power system into coherent areas; this is a relevant issue for managing the network in both normal operating conditions and during anomalous events. In particular, the attention is focused on partitioning the power system in such a way as to group together frequency signals, measured by means of phasor measurement units (PMU), exhibiting similar oscillatory behavior after the occurrence of a fault or disturbance. Unfortunately, the increasingly massive presence of renewable energy sources is undermining the clustering methods defined so far, requiring new solutions to the problem. To overcome the considered drawbacks, the authors propose hereinafter to ( ${i}$ ) improve the grouping capabilities of an iterative spectral clustering method thanks to the definition of new parameters for similarity estimation (Modified Bray Curtis index) and cluster thresholding (weighted Fiedler value) as well as (ii) enhance its robustness with respect to both measurement noise and uncertainty affecting the PMUs by means of a deep test procedure. To this aim, particular attention is paid in the design and assessment stage to the definition of both filtering algorithm and measurement parameters (e.g., the length of the analysis window). Once defined these parameters, the method is capable of 100% correctly separating transmission network sections oscillating with similar trends in a number of tests conducted on simulated and actual signals, so highlighting the promising performance of the method highlighting its reliability and efficacy in different test conditions.

Abstract

The geographic position, isolation, and the long and dynamic history of colonization created a human context in Sicily that allows for a particular anthropological study; information about “migratory flow” and “population influx” could be investigated in the cranial morphology of a localized geographical region. The research goals are the identification of temporal trends in facial morphology in order to assess the adaptations and the microevolutionary trends and to verify if the cranial morphology of humans was modified by the various genetic contributions and more or less related to the intense and significant migratory flows. This work includes a diachronic morphometrics study of 3D models of 95 Sicilian skulls coming from 19 populations (from the Paleolithic to the Contemporary Age), providing an overview of human biodiversity and variability in Sicily. To achieve this, a geometric morphometrics analysis of the facial features of adult human skulls was performed. The approach used allows for the identification of the main micro-anatomical and micro-evolutionary features. Considering sample size/composition, it has been possible to discriminate between prehistorical and historical populations. The results highlight a series of morphological changes related to different migratory flows that have followed one another with different intensities and effectiveness starting from the Prehistory up to the Contemporary Age. The human peopling of Sicily is a subject of continuous debate; however, this study points to the coexistence of microevolutionary patterns and population dynamics, with the latter being one of the main causes of the morphological variations.

Abstract

Background Lynch syndrome (LS) is an autosomal dominant condition due to the germline mutation in the mismatch repair (MMR) genes including MLH1, MSH2, MSH6, and PMS2 (post-meiotic segregation increased 2). The MMR mutation carriers have high risk for cancers. Pathogenic PMS2 variants are rarely reported in LS-associated colorectal cancer (CRC) with colorectal polyps. The aim of the study was to investigate the genetic etiology of CRC in an individual with CRC with multiple colorectal polyps and a family history of cancers. Patients and Methods The index patient was an African male affected by CRC with multiple colorectal polyps. The clinical diagnostic for LS was based on the Amsterdam II criteria and pedigree. Next-generation sequencing with inherited cancer genes panel was used to detect the pathogenic variant. Results The patient fulfilled the Amsterdam II criteria and the pedigree revealed a family history of recurrent CRC. A deleterious PMS2 germline heterozygous mutation c.2192_2196delTAACT was detected. Conclusion Our study supports the notion that LS may be associated with polyps and shows the predisposition of PMS2 heterozygous mutation in LS-associated CRC at young age.

Abstract

O objetivo deste artigo é discutir os resultados de uma maior participação nas cadeias globais de valor (CGVs) para a redução dos índices de pobreza na América Latina e Caribe (ALC), especialmente tendo em vista o aumento da miséria no contexto pós-pandêmico. A hipótese testada no artigo é a de que quanto maior for a inserção da região da ALC nas cadeias globais de valor, menor será o nível de pobreza. Para testá-la, realiza-se uma análise de regressão linear múltipla com dados sobre a inserção da região em CGVs, oriundos da United Nations Conference on Trade and Development (UNCTAD), bem como com informações de pobreza extraídas do Banco Mundial, tendo por período de análise o intervalo entre 1991 e 2018. Como resultados, observa-se um decréscimo tímido na inserção da ALC em CGVs. Apesar disso, identifica-se uma correlação entre a participação em CGVs e a redução da pobreza, confirmando-se, com ressalvas, a hipótese proposta.

Abstract

Abstract Cell microencapsulation has been utilized for years as a means of cell shielding from the external environment while facilitating the transport of gases, general metabolites, and secretory bioactive molecules at once. In this light, hydrogels may support the structural integrity and functionality of encapsulated biologics whereas ensuring cell viability and function and releasing potential therapeutic factors once in situ. In this work, we describe a straightforward strategy to fabricate silk fibroin (SF) microgels (µgels) and encapsulate cells into them. SF µgels (size ≈ 200 µm) were obtained through ultrasonication-induced gelation of SF in a water-oil emulsion phase. A thorough physicochemical (SEM analysis, and FT-IR) and mechanical (microindentation tests) characterization of SF µgels were carried out to assess their nanostructure, porosity, and stiffness. SF µgels were used to encapsulate and culture L929 and primary myoblasts. Interestingly, SF µgels showed a selective release of relatively small proteins (e.g., VEGF, molecular weight, MW = 40 kDa) by the encapsulated primary myoblasts, while bigger (macro)molecules (MW = 160 kDa) were hampered to diffusing through the µgels. This article provided the groundwork to expand the use of SF hydrogels into a versatile platform for encapsulating relevant cells able to release paracrine factors potentially regulating tissue and/or organ functions, thus promoting their regeneration. Abstract

Abstract Regeneration of the neuromuscular junction (NMJ) leverages on extensive exchange of factors released from motor axon terminals (MATs), muscle fibers and perisynaptic Schwann cells (PSCs), among which hydrogen peroxide (H2O2) is a major pro-regenerative signal. To identify critical determinants of NMJ remodeling in response to injury, we performed temporal transcriptional profiling of NMJs from 2 month-old mice during MAT degeneration/regeneration, and cross-referenced the differentially expressed genes with those elicited by H2O2 in SCs. We identified an enrichment in extracellular matrix (ECM) transcripts, including Connective Tissue Growth Factor (Ctgf), which is usually expressed during development. We discovered that Ctgf levels are increased in a Yes-associated protein (YAP)-dependent fashion in response to rapid, local H2O2 signaling generated by stressed mitochondria in the injured sciatic nerve, a finding highlighting the importance of signals triggered by mechanical force to motor nerve repair. Through sequestration of Ctgf or inactivation of H2O2, we delayed the recovery of neuromuscular function by impairing SC migration and, in turn, axon-oriented re-growth. These data indicate that H2O2 and its downstream effector Ctgf are pro-regenerative factors that enable axonal growth, and reveal a striking ECM remodeling process during nerve regeneration upon local H2O2 signaling. Our study identifies key transcriptomic changes at the regenerating NMJ, providing a rich source of pro-regenerative factors with potential for alleviating the consequences of peripheral nerve injuries.

Abstract A significant body of literature has been generated related to the detection of measurable residual disease (MRD) at the time of achieving complete remission (CR) in patients with hairy cell leukemia (HCL). However, due to the indolent nature of the disease as well as reports suggesting long-term survival in patients treated with a single course of a nucleoside analog albeit without evidence of cure, the merits of detection of MRD and attempts to eradicate it have been debated. Studies utilizing novel strategies in the relapse setting have demonstrated the utility of achieving CR with undetectable MRD (uMRD) in prolonging the duration of remission. Several assays including immunohistochemical analysis of bone marrow specimens, multi-parameter flow cytometry and molecular assays to detect the mutant BRAF V600E gene or the consensus primer for the immunoglobulin heavy chain gene (IGH) rearrangement have been utilized with few comparative studies. Here we provide a consensus report on the available data, the potential merits of MRD assessment in the front-line and relapse settings and recommendations on future role of MRD assessment in HCL.

Abstract

Abstract Background This study aimed to examine associations of early childhood physical fitness and physical activity (PA) with PA during later childhood/early adolescence while accounting for gender differences. Methods We selected data of N = 4329 children from the IDEFICS/I. Family cohort (age 2.4–11.7 years) with data on baseline fitness and accelerometer measurements. At baseline, physical fitness tests were conducted including Flamingo balance, Backsaver sit and reach, Handgrip strength, Standing Long Jump, 40-m sprint and 20-m Shuttle run (to estimate cardio-respiratory fitness levels). PA was measured with Actigraph accelerometers over 3 days at baseline (ActiTrainer or GT1M) and 7 days at follow-up (GT3X). Evenson cutpoints were used to determine moderate-to-vigorous PA (MVPA) time, and children with ≥60mins/day of average MVPA were deemed as having met WHO guidelines at baseline and follow-up. Linear and logistic regressions were performed to examine longitudinal associations between meeting WHO guidelines, MVPA, and physical fitness tests at baseline with meeting WHO guidelines and MVPA at follow-up. Models were conducted on the entire sample, the sex-stratified sample, and stratified by sex and pubertal status at follow-up. Results Results showed that meeting WHO guidelines for MVPA at baseline was positively associated with MVPA (Standardized Beta (B) = 0.13, 95%CI:(5.6;11.1)) and meeting WHO guidelines at follow-up for the entire sample (OR = 2.1, 95%CI:(1.5; 3.14), and stratified by males (OR = 2.5, 95%CI:(1.5; 4.1)) and females (OR = 1.8, 95%CI:(1.0; 3.2)). This was also found for both male pre/early pubertal and pubertal groups but only in the female pre/early pubertal group, and not the female pubertal group (MVPA: B = .00, 95%CI:(− 6.1; 5.6), WHO: OR = 0.61, 95%CI:(0.23;1.6)). Models indicated that Standing Long jump, 40-m sprint, Shuttle run and Flamingo balance at baseline were associated with MVPA and meeting the guidelines at follow-up. Conclusions Meeting WHO guidelines and certain fitness tests at baseline were strongly associated with MVPA and meeting WHO guidelines at follow-up, but this association varied with sex and pubertal status. Consequently, these findings underline the importance of ensuring sufficient physical activity in terms of quality and quantity for children at the earliest stages of life.
