We built an automatic chamber system to measure greehouse gas (GHG) exchange in forested peatland ecosystems. We aimed to build a system robust enough which would work throughout the year and could measure through a changing snowpackin addition to producing annual GHG fluxes by integrating the measurements without the need of using models. The system worked rather well throughout the year, but it was not service free. Gap filling of data was still necessary. We observed problems in carbon dioxide (CO2) flux estimation during calm summer nights, when a CO2 concentration gradient from soil/moss system to atmosphere builds up. Chambers greatly overestimated the nighttime respiration. This was due to the disturbance caused by the chamber to the soil-moss CO2 gradient and consequent initial pulse of CO2 to the chamber headspace. We tested different flux calculation and measurement methods to solve this problem. The estimated flux was strongly dependent on (1) the type of the fit (linear and polynomial), (2) the starting point of the fit after closing the chamber, (3) the length of the fit, (4) the speed of the fan mixing the air inside the chamber, and (5) atmospheric turbulence (friction velocity, u*). The best fitting method (the most robust, least random variation) was linear fitting with the period of 120-240 s after chamber closure. Furthermore, the fan should be adjusted to spin at minimum speed to avoid the pulse-effect, but it should be kept on to ensure mixing. If nighttime problems cannot be solved, emissions can be estimated using daytime data from opaque chambers. [ABSTRACT FROM AUTHOR]

Intramammary infection (IMI), also known as mastitis, is the most frequently occurring and economically the most important infectious disease in dairy cattle. This study provides a validation of the analytical specificity and sensitivity of a real-time PCR-based assay that identifies 11 major pathogen species or species groups responsible for IMI, and a gene coding for staphylococcal β-lactamase production (penicillin resistance). Altogether, 643 culture isolates originating from clinical bovine mastitis, human, and companion animal samples were analyzed using the assay. The isolates represented 83 different species, groups, or families, and originated from 6 countries in Europe and North America. The analytical specificity and sensitivity of the assay was 100% in bacterial and β-lactamase identification across all isolates originating from bovine mastitis (n = 454). When considering the entire culture collection (including also the isolates originating from human and companion animal samples), 4 Streptococcus pyogenes, 1 Streptococcus salivarius, and 1 Streptococcus sanguis strain of human origin were identified as Streptococcus uberis, and 3 Shigella spp. strains were identified as Escherichia coli, decreasing specificity to 99% in Strep. uberis and to 99.5% in E. coli. These false-positive results were confirmed by sequencing of the 16S rRNA gene. Specificity and sensitivity remained at 100% for all other bacterial targets across the entire culture collection. In conclusion, the real-time PCR assay shows excellent analytical accuracy and holds much promise for use in routine bovine IMI testing programs. This study provides the basis for evaluating the assay's diagnostic performance against the conventional bacterial culture method in clinical field trials using mastitis milk samples. BACKGROUND: Depth-of-anesthesia monitoring with the electroencephalogram has become widely used in anesthesia practice. Generally, the methods presented are based on the spectral changes of the electroencephalogram. In this study, the authors evaluate the influence of remifentanil on the relation of timely occurrence of clinical endpoints and the spectral behavior of the electroencephalogram. METHODS: Twenty-seven patients scheduled to undergo a surgical procedure were randomly assigned to three groups. Patients blindly received equal volumes of saline or remifentanil (7.5 or 30 microg.kg.h) 1 min before induction of anesthesia with infusion of propofol (30 mg.kg.h). The occurrence of loss of counting, loss of obeying verbal command, and loss of reaction to tetanic stimulation was assessed. The electroencephalogram was recorded from electrode Fz referenced to the common average, and an iterative algorithm was applied to solve the underlying frequency progression pattern. The positions of the clinical endpoints on the pattern were analyzed. RESULTS: The administration of remifentanil during induction of anesthesia with propofol led to an earlier occurrence of the clinical endpoints on the frequency progression pattern. A significant difference (P < 0.05) was observed between the saline and high-dose patient groups in all three endpoints. The effect of remifentanil was proportional to the infusion rate. CONCLUSIONS: The infusion of remifentanil during propofol anesthesia significantly modifies the mutual relations of the electroencephalographic spectral characteristics and the endpoints in a predictable and quantifiable manner. This finding suggests that the electroencephalographic phenomena and the endpoints may not be identical but rather to some extent separate manifestations of hypnotic drug effect. Compilation and analysis of information from the literature regarding cross-species microsatellite amplification and polymorphism success, and relating this to source-target species genetic distance as estimated by pairwise cytochrome b ( cytb) divergence, enabled an in-depth investigation of factors affecting avian cross-species microsatellite amplification. Source-target species cytb distances provided accurate estimates of cross-species microsatellite amplification/polymorphism success rates not only in birds, but also in taxa where microsatellites cross-amplify across contrasting levels of taxonomic classification (frogs and cetaceans). As cytb is one of the most commonly sequenced DNA regions, pairwise cytb genetic distances should therefore be useful for predicting cross-species microsatellite success across a range of taxonomic groups. While the most important factor affecting cross-species microsatellite amplification/polymorphism success was a negative association with source-target species genetic distance, associations with additional features affecting cross-species amplification/polymorphism success included: decreasing PCR annealing temperature significantly increasing the chance of successful cross-species amplification, and a significant positive association between source species polymorphism and the proportion of target species in which a locus revealed polymorphism. No association between cross-species amplification and repeat motif (di-, tri-, or tetranucelotide) or repeat structure (perfect, imperfect, or compound) was observed. A set of nine loci which cross-amplified across an unusually broad range of passerine bird species were also identified, and could serve as a good starting point for cross-species amplification testing in passerine species for which insufficient loci are available. We apply the current spin-density functional formalism (CSDFT) of Vignale and Rasolt to two-dimensional quantum dots in magnetic fields. Avoiding any spatial symmetry restrictions of the solutions, we find that a broken rotational symmetry of the electronic charge density can occur in high magnetic fields. [ABSTRACT FROM AUTHOR]

The concept of shell structure has been found useful in the description of semiconductor quantum dots, which today can be made so small that they contain less than 20 electrons. We review the experimental discovery of magic numbers and spin alignment following Hund’s rules in the addition spectra of vertical quantum dots, and show that these results compare well to model calculations within spin density functional theory. We further discuss the occurrence of spin density waves in quantum dots and quantum wires. For deformable two-dimensional quantum dots (for example, jellium clusters on surfaces), we study the interplay between Hund’s rules and Jahn–Teller deformations and investigate the effect of magnetic fields on the ground-state shapes. Using the density functional Kohn-Sham method we show that the internal shapes of fermion clusters are nearly independent of the interparticle interactions. The odd-even staggering of the total energy of small nuclei and of sodium clusters are shown to be of the same origin. Tetrahedral deformations are predicted for 40, 70 and 112 atom sodium clusters. The diagnostic value of I-phenylpentadecanoic acid (IPPA) metabolic cardiac imaging was studied in a group (n=29) of patients with angiographically confirmed CAD using single photon emission computed tomography (SPECT). A symptom-limited exercise test was first done with IPPA, and 2 days later with thallium. Medications were not withheld during testing. Fourteen healthy control subjects participated in parallel IPPA and 15 in thallium tests. Data acquisition and output were comparable in the two imaging modalities. By testing various relatively simple criteria for abnormality we found that the semiquantitative interpretation was more accurate than the visual readings. The best compromise of accuracy with the scored criteria consisted of a sensitivity of 86% and a specificity of 86%, obtained with IPPA polar tomograms (mild exercise defect) and a sensitivity of 86% and a specificity of 80% obtained with thallium (regionally decreased washout). With visual interpretation alone, a sensitivity of 83% and a specificity of 71% was detected with IPPA (mild exercise defect) and 72% and 73%, respectively, with thallium (partial reversibility). The sensitivity of the exercise ECG alone was 62%. The results of this study imply that IPPA imaging could be a rational, uncomplicated clinical method for non-invasive diagnosis of CAD. Molar teeth of four groups of Osborne-Mendel rats, kept on a cariogenic dict, were treated once per day for 48 days with either distilled water (solution C), a solution containing 0.05% chlorhexidine gluconate and 0.04% NaF, pH 5.8 (solution CXF), CXF plus 1000 ppm Sr (solution CXFS), or CXFS plus 1000 ppm Zn (solution CXFSZ). Caries of the hemi-sectioned lower molars was scored according to grades of severity, and the extent of the buccal plaque was estimated. Inflammatory cell infiltration, vascularity, and thickness of the oral mucosa were estimated from stained sections of the buccal oral mucosa to detect possible adverse effects of the treatments. Fissure caries lesions with involvement of the dentin were reduced by 57% (p < 0.01) with the CXF treatment and by 68%o (p < 0.001) with the CXFS treatment. The CXFSZ treatment reduced the fissure caries least (44%, p < 0.05), but it appreciably reduced the extent of plaque. The mucosal changes were greatest in the group receiving the CXFSZ treatment, but they were generally small and at a non-pathological level. Equilibrium geometries of quasi two-dimensional jellium systems are calculated in the local density approximation, closely following the 'Ultimate Jellium Model' of [1]. The background charge is assumed to be fully deformable in a layer between two parallel planes, whereas the wave functions in the direction perpendicular to such a 'jellium slice' are confined to their ground state. Like for jellium clusters in three dimensions [1], we find that for various system sizes, a trend towards a breaking of axial and inversion symmetries is observable. [ABSTRACT FROM AUTHOR]

The local spin-density approximation is used to calculate ground- and isomeric-state geometries of jellium clusters with 2 to 22 electrons. The positive background charge of the model is completely deformable, both in shape and in density. The model has no input parameters. The resulting shapes of the clusters exhibit breaking of axial and inversion symmetries; in general the shapes are far from ellipsoidal. Those clusters which lack inversion symmetry are extremely soft against odd-multipole deformations. Some clusters can be interpreted as molecules built from magic clusters. The deformation produces a gap at the Fermi level. This results in a regular odd-even staggering of the total energy per electron and of the HOMO level. The strongly deformed 14-electron cluster is semimagic. Stable isomers are predicted. The splitting of the plasmon resonance due to deformation is estimated on a classical argument. The origin of the plasmon excitation in small metal clusters is studied within the jellium model through ab initio electronic-structure calculations based on the nuclear shell model. In the limit of infinite size, the plasmon classically represents pure harmonic motion of the center of mass of the valence electrons. Tamoxifen-induced DNA adducts were searched in leucocyte DNA from breast cancer patients. Total white blood cell DNA from tamoxifen-treated and control patients was analysed by 32P-postlabelling using HPLC-radioactivity detection. Rat liver DNA was used as a positive standard. In blinded analysis four of the six treated patients showed DNA adducts; none of the five controls were positive. The identity of fraction as a tamoxifen adduct was confirmed by using different chromatographic systems, each with spiked rat liver samples. The level of adducts in the treated patients was 5.5 adducts/10(9) nucleotides as compared to an apparent level of 1.9/10(9) in the controls. [ABSTRACT FROM PUBLISHER]

Serum concentrations of C-reactive protein were studied in 23 patients with acute myocardial infarction. In 14 patients who did not receive thrombolytic treatment there was a linear relation between infarct size (determined by serial creatine kinase-MB determinations and thallium-201 isotope emission tomography) and the C-reactive protein response. The correlation coefficient between the concentration-time integrals of creatine kinase-MB and C-reactive protein was 0.96. The correlation coefficient between the creatine kinase-MB concentration-time integral and the peak serum value of C-reactive protein was 0.93. In the nine patients who received intravenous streptokinase treatment there was also a positive correlation between the concentration-time integrals of creatine kinase-MB and C-reactive protein. The relation, however, depended on the success of the treatment. In patients with successful reperfusion the C-reactive protein response was only approximately 20% of that in patients in whom reperfusion failed or who received no thrombolytic treatment and who were matched by infarct size. When thrombolysis was successful the correlation coefficient between the concentration-time integrals of creatine kinase-MB and C-reactive protein was 0.86. Daily measurement of serum C-reactive protein is useful in evaluating infarct size in patients with acute myocardial infarction who do not receive thrombolytic treatment.
