This study was undertaken to determine the effects of induced molt on basal functional activities of heterophils from aging hens. For this purpose, heterophils from both molted and unmolted hens were examined by in vitro bioassays for functional responsiveness and efficiency. We evaluated the ability of the heterophils to migrate to chemotactic stimuli, phagocytize opsonized and nonopsonized Salmonella-enteritidis (SE), and generate an oxidative burst in response to inflammatory agonists. A significant (P < 0.001) heterophilia was found in the molted hens within 2 days after feed withdrawal and remained throughout the length of the experimental feed withdrawal period. No significant differences were found in the random migration of heterophils from either group. The chemotactic movement of heterophils from molted hens was not affected until 8 days after feed withdrawal when compared with heterophil chemotaxis from unmolted hens. A significant decrease in chemotaxis by the heterophils from molted hens was observed days 8-12 after feed withdrawal (P < 0.05). Significantly (P < 0.05) fewer heterophils from molted hens were able to phagocytize opsonized (59% vs. 38%) and nonopsonized (26% vs. 15%) SE within 2 days after feed withdrawal. Likewise, significantly (P < 0.05) fewer bacteria were phagocytized per heterophil from the molted hens when compared with the number of bacteria per heterophil from the unmolted hens. The oxidative burst of heterophils stimulated by either opsonized zymosan A or phorbol myristate acetate of heterophils from molted hens was significantly (P < 0.05) reduced when compared with that generated by heterophils from the unmolted hens. These results indicate that feed withdrawal to induce molt alters the number and function of peripheral blood heterophils. This decreased efficiency of heterophil functional activity appears to play a role in the increased susceptibility of molting hens to SE infections.

Salmonella arizonae (SA) and S. gallinarum (SG) are of economic importance to international poultry production because of their pathogenesis in young poultry during the first week after hatching. Previous studies from our laboratory have shown immune lymphokines (ILK) produced by S. enteritidis (SE)-immunized chickens provide protection against SE organ invasion in day-old chickens and turkey poults. Previous studies have also demonstrated that SG organ invasion was significantly decreased by administration of ILK to broiler chicks. The objective of the present study was to determine the effect of ILK on the incidence of horizontal transmission of SA in turkey poults and of SG in broiler chicks. The effect of ILK administration on horizontal transmission of SA in poults and SG in chicks was assessed in a seeder/contact model. Seeders were challenged with the appropriate bacterium (SA turkeys, SG chicks), contacts were either untreated or administered ILK. Seeders and contacts cohabited within an experimental group throughout the experiment. Mortality and organ invasion as a result of horizontal transmission were determined. There were no significant differences in mortality between non-treated and ILK-treated contact poults. In contrast, SG was extremely pathogenic to young broiler chicks. Non-treated contact chicks had a mortality rate of approximately 68% whereas significant (P < 0.05) reduction in mortality was demonstrated in the contact chicks treated with ILK (15%). Horizontal transmission, as determined by organ invasion, of SA to contact turkey poults and SG to contact broiler chicks was also significantly (P <0.05) decreased by immunoprophylactic administration of ILK. Bacterial recovery of SA from the liver/spleen and the cecal tonsil in contact poults and SG from contact chicks treated with ILK was dramatically reduced when compared to non-treated contact poults and chicks. Our results strongly suggest the immunoprophylactic administration of SE-immune lymphokines to young turkey poults and broiler chicks significantly reduces the horizontal transmission of Salmonella in poultry. These results suggest the possibilities of using a non-vaccine immunologically-based preventative strategy against Salmonella in poultry.

Experiments involving 132 weaned piglets were conducted to evaluate the efficacy of a Salmonella enteritidis-immune lymphokine (PILK) derived from the T cells of Salmonella enteritidis (SE)-immunized pigs to protect weaned piglets from Salmonella choleraesuis (SC) infection. Fourteen-to-seventeen day-old piglets were weaned and randomly placed into 1 of 5 groups: (1) noninfected controls, (2) PILK 3X noninfected, (3) SC infected controls, (4) PILK 1X SC infected, and (5) PILK 3X SC infected. PILK was given orally either one time (PILK 1X) or three times (PILK 3X) over 14 days. One hour after the first PILK administration on day 0, piglets were orally challenged with 10(7) cfu of SC. Weights were recorded on day 0, day 7, and day 14. On day 14, pigs in groups 3, 4, and 5 were sacrificed and organs and lymph tissue were cultured for the presence of SC. Three replicates of this experiment were pooled and anlayzed. A significant reduction in the number of pigs positive for SC in the liver, lung, and spleen was found in group 5 (PILK 3X) when compared to group 3 (inf. cont. p < 0.001[). The number of SC positive cecal contents was dramatically reduced in group 5 group when compared to group 3, with the PILK 3X group showing 13% positive pigs versus 55.2% in the infected controls (p < 0.05). Weight gain over the 14 day study in the infected PILK 3X group (group 5) was found to be comparable to the gain observed in the group 1 (noninfected controls). The pigs receiving PILK 3X (group 2) with no SC challenge gained significantly more weight than all other groups, including the noninfected controls (group 1 p < 0.05[). The results of these experiments indicate that PILK protects against SC infection in weaned pigs while enhancing performance in the presence of an SC infection.

Objective: To determine functional responses of neonatal chicken and turkey heterophils to various inflammatory agonists., Animals: 100 one-day-old chickens and turkeys., Procedure: Blood heterophils were isolated and stimulated for 30 minutes at 39 C with ionomycin, phorbol myristate acetate (PMA), opsonized zymosan (OZ), or formyl-methionyl-leucyl-phenylalanine (FMLP). Functional responses (shape change, adherence, phagocytosis, influx of intracellular calcium, and oxidative burst) of stimulated heterophils were measured and compared with responses of unstimulated (control) heterophils., Results: Turkey and chicken heterophils did not respond to FMLP stimulation. Stimulation of chicken and turkey heterophils with ionomycin resulted in significant increases in adherence, percentage of cells with a shape change, phagocytosis, intracellular calcium concentration, and oxidative burst. Turkey heterophils did not respond to PMA stimulation, whereas stimulation of chicken heterophils with PMA resulted in significant increases in adherence, percentage of cells with a shape change, phagocytosis, and oxidative burst but not intracellular calcium concentration. Stimulation of chicken and turkey heterophils with OZ resulted in significant increases in oxidative burst., Conclusions: Mechanisms regulating initiation of heterophil activation in neonatal chicken and turkey heterophils are consistent with those described for heterophils isolated from mature birds. The biochemical and cytoskeletal systems of neonatal avian heterophils undergo functional alterations following stimulation with inflammatory agonists., Clinical Relevance: Understanding heterophil activation and regulation should eventually lead to methods for controlling bacterial diseases in poultry.

A commercial competitive exclusion (CE) culture (PREEMPT) was evaluated for its ability to decrease mortality during the first 10-12 days posthatch resulting from the causative agent of fowl typhoid, Salmonella gallinarum. In the first experiment, chicks provided the CE culture on day of hatch and challenged with 10(5) S. gallinarum on day 3 had a significant decrease in mortality compared with non-CE-treated S. gallinarum-challenged chicks in all four replicates. Mortality for control chicks when averaged across all four replicates was 74% compared with 7.5% for the CE-treated chicks. A second experiment was performed that was designed to measure the efficacy of the CE culture in decreasing the horizontal transmission of this pathogen from infected to uninfected chicks when commingled. Day-of-hatch chicks that were directly infected (seeders) with 10(5) S. gallinarum and provided no CE culture averaged 86% S. gallinarum organ positive and 80% mortality during the first 12 days posthatch across four replicates. Untreated contact (not directly infected) chicks that were commingled with the seeder chicks averaged 84% S. gallinarum organ positive and 54% mortality, and the commingled CE-treated contact chicks (provided CE culture on day of hatch) average 35% S. gallinarum organ positive and 9% mortality during the same time period. These results are of importance to the poultry industry in geographic areas where poultry production is adversely affected by fowl typhoid and indicate that treating newly hatched chicks with this commercial CE culture may be a novel way of decreasing economic losses associated with this highly pathogenic organism.

Heterophils are important mediators of innate resistance in poultry, especially in young birds that have not yet developed an acquired immune response. Invasion of the intestinal mucosa by Salmonella spp. initiates the recruitment of large numbers of heterophils to the lamina propria. Thus, the heterophilic response can control, but not eliminate, bacterial numbers in the bird until development of acquired immunity. Unfortunately, chicks and turkey poults are highly susceptible to Salmonella infections during the first 4 d posthatch due to the functional immaturity of both the innate and acquired immune systems. We have previously shown that the administration of Salmonella enteritidis (SE)-immune lymphokines (ILK) into either 18-d-old developing embryos or day-of-hatch chicks and poults conferred increased resistance to SE organ invasion. In this review, we present evidence that the protection induced by ILK is mediated by vigorous recruitment and activation of heterophils. These activated heterophils migrate rapidly to the site of bacterial invasion where they phagocytize and kill the SE. Specifically, in vitro studies demonstrate an enhancement of functional activities of the heterophils including chemotaxis, adherence, phagocytosis, and bacterial killing. In addition, during the activation process, membrane expression of adhesion molecules rapidly changes from L-selectins to beta2 integrins (CB11b/CD18) on the cells that become activated. These results further demonstrate the validity of preventive activation in poultry to induce the migration of large numbers of activated phagocytic cells to the site of infection by a pathogenic organism. Importantly, this immunopotentiation of the inflammatory response by ILK, as described here, induces the functional maturation of heterophils during the first 4 d posthatch.

We have previously reported that the prophylactic administration of factor(s) from T-cell supernatants derived from Salmonella enteritidis-immune chickens (ILK) have a favorable effect in controlling or eliminating salmonellosis in neonatal poultry. Experimentally, we have used the intraperitoneal injection as the standard method of administering ILK to neonatal poultry. However, this method is neither easy, practical, nor economical for the poultry industry. In the present study, we evaluated the effectiveness of oral (p.o.), intranasal (i.n.), and subcutaneous (s.c.) administration of ILK for ease of delivery, induction of protective resistance against Salmonella enteritidis (Se) organ invasion, and the ability to activate peripheral blood heterophils in day-old chickens. In the first experiments, delivery of ILK p.o., i.n., and s.c. significantly (P < 0.01) increased the resistance of day-old chickens to Se organ invasion. The level of protection was equivalent to that induced by the i.p. route. Administration of a comparable protein control (bovine serum albumin, BSA) by the 3 routes induced no protective effect against Se organ invasion. Likewise, a significant increase was found in the number of circulating heterophils within 4 h of administration of the ILK by all routes. In the 2nd experiment, the function of the heterophils from ILK-treated birds was compared with that of the control cells in adherence, chemotaxis, and phagocytosis assays. The heterophils from birds given ILK i.p., s.c., p.o., or i.n. had significantly (P < 0.01) increased functional activities when compared to the activities of the heterophils from the control birds. These studies indicate that the delivery of ILK either orally or parenterally, routes which can be used by the poultry industry, can confer protection to chickens against a localized enteric Se organ invasion by potentiating the systemic heterophilic innate response.

Previous experiments have shown that the administration by intraperitoncal (i.p.) injection of Salmonella enteritidis-immune Iymphokines (LK) derived from chicken splenic T cells protect both chicks and turkey poults from Salmonella enteritidis (SE) organ invasion. This protection was mediated by the avian heterophil. The present experiments evaluated the ability of SE-immune Iymphokines derived from a virally transformed chicken T cell line (VILK) in protecting day-old turkeys against SE liver invasion, inducing peripheral blood heterophilia, and functionally activating heterophils when delivered by subcutaneous (s.c), oral (p.o.) or intranasal (i.n.) routes when compared to i.p. injection. All routes of administration of VILK showed dramatic reductions in SE liver invasion (P < 0.01) and significant elevations in the number of heterophils in the peripheral blood (P < 0.01). The rise in peripheral blood heterophils was accompanied by a significant increase in the functional activity of the heterophils. Chemotactic movement of heterophils from all VILK groups was two to three times that observed in the control heterophils using chicken serum and recombinant human interleukin-8 (rhuIL-8) as the chemoattractants (P<0.01). The number of heterophils phagocytizing SE and the number of bacteria per heterophil were significantly higher in all VILK administered groups (P < 0.01). Adherence to bovine serum albumin (BSA) revealed significant increases in adherence of heterophils from all VILK administered groups when compared to control heterophils from poults (P<0.01). The results of these experiments clearly show that VILK delivered s.c, p.o. and i.n. is as effective in protecting day-old poults from SE invasion as an i.p. injection and that this protection is also mediated by the activation of an increased number of heterophils in the circulation.

Heterophils, the functional equivalent to the mammalian neutrophil, are important mediators of natural resistance against invasive pathogens in poultry. Young poultry are susceptible to pathogens, such as Salmonella enteritidis, during the first week post-hatch. No studies have evaluated the ontogeny of heterophil function in turkeys during the first few weeks post-hatch. Previous studies from our laboratory have shown day-old poults were protected against S. enteritidis organ invasion following immunoprophylactic administration of chicken S. enteritidis immune lymphokines. Therefore, the objective in the present study was to characterize the development of phagocytosis and bacterial killing by turkey heterophils during the first 3 weeks of life and to compare the effect of immune lymphokines on the development of heterophil phagocytosis and killing during the first 3 weeks post-hatch. Both functional phagocytosis and killing activities were age-dependent events. During the first 1-7 days post-hatch, little functional activity was demonstrated which apparently is associated with susceptibility. Optimal heterophil phagocytosis and killing activities were reached 14-21 days post-hatch. Administration immune lymphokines significantly potentiated phagocytosis (P < 0.01) and killing (P < 0.001) by turkey heterophils. In fact, immune lymphokine administration to 1-7-day-old poults augmented phagocytosis and killing activities of heterophils equivalent to levels found in functionally mature 14-21-day-old poults. These results demonstrate the ontogeny of the functional activity of the turkey heterophil is an age-related phenomenon, with inefficient phagocytosis and killing during the first week post-hatch. Prophylactic administration of immune lymphokines significantly potentiated the functional activity of the heterophil poults during the first 3 weeks of life. Most importantly the administration of immune lymphokines enhanced the functional activity of heterophils from 1-7-day-old poults to levels comparable to that of an immunologically mature bird.

Hematopoietic colony stimulating factors (CSF) regulate the growth and development of phagocytic cell progenitors and also augment functional activation of phagocytes. Granulocyte-CSF (G-CSF) is the CSF that acts specifically upon granulocyte progenitor cells and mature granulocytes. We have shown that lymphokines (ILK) from T cells of birds immunized against Salmonella enteritidis (SE) induce a granulocytic (PMN) inflammatory response in chicks challenged with SE. This inflammatory response was characterized by: (a) a dramatic emigration of granulocytic cells from the bone marrow into the peripheral blood, (b) an enhancement of the biological functions of the circulating PMNs, and (c) a directed influx of these activated PMNs to the site of bacterial invasion. In the current study, we determined the presence of G-CSF in ILK by Western blot analysis using a goat polyclonal antihuman G-CSF antibody (Ab). Using this Ab, we then evaluated the role of G-CSF in the ILK-induced protective inflammatory response in chickens against SE. Pretreatment of ILK with the Ab totally abolished the colony-stimulating activity of the ILK. Furthermore, Ab treatment of ILK resulted in: (a) an elimination of the ILK-induced peripheral blood heterophilia with a dramatic inhibition of ILK-mediated protection against SE organ invasion and (b) an elimination of accumulation of inflammatory PMNs in the peritoneum with subsequent decrease in the survival rate of chicks challenged i.p. with SE. Taken together these studies demonstrate for the first time the contribution of G-CSF to avian PMN activation and the immunoprophylaxis of SE infection by ILK in neonatal chickens.

Newly hatched chicks are susceptible to organ invasion by various serovars of Salmonella. We have previously shown that intraperitoneal administration of Salmonella enteritidis-immune lymphokine (SEILK) increased resistance to organ invasion by S. enteritidis (SE) in day-of-hatch chicks. In the present study, we determined that Salmonella-immune lymphokines can provide protection against organ invasion by serovars that are unrelated to the original serovar used as an immunogen. Immune lymphokines were produced from the splenic T cells of mature hens immunized with either viable SE or Salmonella anatum (SA). Day-of-hatch chicks were treated with either SA-immune lymphokine (SAILK) or SEILK and challenged per os with either Salmonella typhimurium (ST) or SE 1 hr later. Liver samples were aseptically collected 20 hr later and were cultured to detect salmonella. We found that SAILK conferred resistance against invasion by either SE, a serogroup D organism, or ST, a serogroup B organism. Furthermore, SEILK reduced organ invasion by ST. These observations together reveal that the protection against organ invasion conferred by Salmonella-immune lymphokines is unrelated to the serovar-specific antigenic characteristics of the immunizing bacteria.

Research on the control of intestinal and tissue colonization of breeder and table-egg producing flocks by invasive Salmonella enteritidis (SE) has focused on the advancement of anti-salmonella feed additives, microbiological strategies, and the development of vaccines. Recent investigations in our laboratories have concentrated on the development of immunoprophylactic measures to control Salmonella infections. We have found an increased resistance to Salmonella enteritidis (SE) organ infectivity in chickens conferred by the prophylactic administration of SE-immune lymphokines (SE-ILK). Fowl typhoid, caused by Salmonella gallinarum (SG), is a septicemic disease of domestic birds resulting in morbidity with moderate to very high mortality within the first 2 weeks of age. The objective of the present studies was to evaluate the effect of a prophylactic treatment of neonatal broiler chicks with lymphokines derived from S. enteritidis (SE)-immunized chickens (SE-ILK) on the birds' resistance to an experimental infection with S. gallinarum (SG). On the day-of- hatch, chicks were intraperitoneally administered either SE-ILK, control nonimmune lymphokines (NILK), or nothing. Thirty min later, all chicks were gavaged with either 10(4) cfu or 10(6) cfu SG. For 10 days after challenge, the chicks were observed twice daily for morbidity and mortality. Chicks that died during the experiment had their livers cultured for SG. Chicks that survived throughout the 10 day experimental period were killed and their livers, spleens, and cecal tonsils cultured for SG. The prophylactic treatment of chickens with SE-ILK induced significant protection against an extraintestinal SG infections when compared to NILK as evidenced by: 1) a significant reduction (P < 0.005) in the mortality of chicks challenged with either 10(4) and 10(6) cfu sg; 2) increased average weight gains of chicks challenged with either 10(4) and 10(6) efu SG; and 3) a significant (P < 0.001) reduction in the total number of SG-organ-culture positive chicks. The results suggest that the prophylactic administration of SE-ILK can non-specifically confer protection to chicks against a pathogenic salmonellae as seen by reduced morbidity, mortality, and organ infectivity of SG in broiler chicks while enhancing weight gain during the first ten days of life.

The prophylactic treatment of neonatal broiler chicks with lymphokines derived from S. enteritidis-immurazed chickens (SE-ILK) was evaluated for its effect on the birds' resistance to an experimental infection S. enterica ser. gallinarum (SG). On the day of hatch, chicks were injected intraperitoneally with either SE-ILK, control non-immune lymphokines (NILK), or were left untreated. Thirty minutes later, all chicks were orally gavaged with either 10(4) colony forming units (CFU) or 10(6) CFU SG. The chicks were observed twice daily for 10 days for morbidity and mortality. Chicks that died during the experiment had their livers cultured for SG. The survivors were killed and their livers, spleens and caecal tonsils cultured for SG. The prophylactic treatment of chickens with SE-ILK induced significant protection against extraintestinal SG infection when compared to the NILK-treated or non-treated controls as evidenced by: (1) a significant reduction (P< 0.005) in the mortality of chicks challenged with either 10(4) and 10(6) CFU SG; (2) an increased average weight gains of chicks challenged with either 10(4) and 10(6) CFU SG; and (3) a significant (P< 0.001) reduction in the number of chicks with organs culture-positive for SG. The results suggest that the prophylactic administration of SE-ILK can confer non-specific protection to chicks against a pathogenic species of Salmonella resulting in reduced morbidity, mortality, and organ infectivity caused by SG infections of broiler chicks, while enhancing performance during the first 10 days of Ufe.

This investigation was designed to characterise the specific cytokine activity from the conditioned medium of concanavalin A-stimulated avian T cells derived from Salmonella enteritidis-immune chickens, S enteritidis-immune lymphokine (ILK). Studies were designed to determine first, whether colony-stimulating activity was present in ILK, second, the type(s) of colonies from the bone marrow that were supported in vitro by the potential colony-stimulating factors in ILK and, third, whether colony-stimulating activity was present in serum from chicks treated with ILK and challenged with S enteritidis, and to use physicochemical treatment as a means of identifying the potential colony-stimulating factor(s) in ILK. Both ILK alone and serum from chicks treated with ILK and challenged with S enteritidis caused significant increases in the number of colony-forming units (CFU) from the bone marrow in vitro. After 10 days of incubation, ILK alone supported the in vitro growth of granulocytic bone marrow colonies. The colony-stimulating activity from serum derived from chicks treated with ILK and challenged with S enteritidis peaked two hours after the challenge. When ILK was either heated at 100 degrees C or treated with trypsin or acid and then injected into chicks, all the chicks responded with significant increases in circulating polymorphonuclear leucocytes (PMNs). However, when assayed for in vitro colony-stimulating activity, only trypsinisation destroyed the activity in ILK. The results indicate that a colony-stimulating factor which preferentially supported the growth of granulocytic bone marrow colonies was present in ILK and that the factor was stable to heat and acid but sensitive to trypsin.

We have shown previously that increased resistance to Salmonella enteritidis (SE) organ infectivity in 1-day-old chicks was conferred by the immunoprophylactic administration of SE-immune lymphokines (SEILK). These lymphokines have been found to be present in the cell culture media of concanavalin A-stimulated splenic lymphocytes obtained from SE-immunized chickens. In the present study we evaluated whether turkeys also produced SEILK and whether these lymphokines could protect 1-day-old chicks and turkey poults against SE liver invasion. In addition, we tested the ability of our original chicken SEILK to reduce SE liver invasion in turkey poults. Day-of-hatch chicks and turkey poults were injected intraperitoneally with immune lymphokines of either chicken or turkey origin. One hour later the birds were challenged per os with SE, and 20 hours later their livers were examined by bacteriological methods for the presence of SE. We found that SEILK induced from the splenic lymphocytes of SE-immunized turkeys reduced SE liver invasion in both chicks and turkey poults. Conversely, we also determined that SEILK produced by chicken splenic lymphocytes conferred protection against invasion by SE in turkey poults. This research is the first report of the production of SEILK in turkeys and also the first report on the cross-species activity of these effector molecules in chickens and turkeys.

We used an anti-inflammatory dose of dexamethasone (DEX) and Salmonella enteritidis (SE)-immune lymphokines (ILK) followed by oral SE challenge to chicks to determine the effects of these treatments on SE organ invasion and in vitro function of PMNs derived from peripheral blood. Endpoints included percent protection against SE organ invasion, numbers of peripheral blood PMNs, and in vitro PMN adherence, chemotaxis, and SE killing. SE organ invasion was significantly reduced in chicks treated with either ILK alone or DEX + ILK compared to controls. Chicks treated with either DEX alone or DEX + ILK responded with a significant increase in numbers of peripheral blood PMNs as compared to controls, while numbers of PMNs in the peripheral blood from chicks treated with ILK alone were not significantly increased. PMN adherence and percent SE killing by PMNs derived from chicks treated with either ILK alone or DEX + ILK were significantly increased compared to controls. Chemotaxis of PMNs derived from chicks treated with either ILK alone or DEX alone significantly increased 2-fold over control levels. Interestingly, chemotaxis of PMNs derived from chicks that received DEX + ILK was similar to controls. Generally, ILK abated the anti-inflammatory effects of DEX on PMNs in these assays, except for chemotaxis. We interpret these data to suggest that ILK may confer protection to chicks against the early phase of SE organ invasion by inducing an inflammatory response predominated by activated PMNs.

We previously reported that the prophylactic, intraperitoneal administration of supernatants from concanavalin A-stimulated T cells derived from Salmonella enteritidis (SE)-immune White Leghorn hens (i.e., SE-immune lymphokines or ILK), conferred protection to neonatal White Leghorn chicks against SE organ invasion. In the present study, we evaluated the effects of in ovo administration of ILK on hatchability, hatch weight, in vitro bactericidal activity of heterophils, and protection against SE organ invasion in neonatal White Leghorn chicks. On Day 18 of embryogenesis, injections were made into the amnion with either ILK or nonimmune ILK (NILK) or were not injected (untreated). On the day of hatch, whole blood was collected from 20 of the chicks per treatment group for heterophil isolation. All remaining chicks were orally challenged with 5 x 10(4) cfu SE. Twenty-four hours after SE challenge, organs (liver and spleen) from the chicks were cultured for SE. Hatchability of ILK- and NILK-treated chicks was not different from that of untreated chicks. Hatch weights of ILK-treated chicks were approximately 1 g less (P < .05) than that of NILK-treated or untreated chicks. In vitro bactericidal activity of peripheral blood heterophils derived from ILK-treated chicks was increased (P < .05) above activity from heterophils derived from NILK-treated or untreated chicks. Organ invasion with SE was markedly and significantly decreased in the ILK-treated chicks as compared with chicks treated with NILK or untreated chicks. These results suggest that in ovo administration of ILK confers protection to neonatal chicks against SE organ infectivity at hatch.

Investigations in our laboratories have indicated that when Salmonella enteritidis (SE)-immune lymphokines--supernatants from concanavalin-A-stimulated T cells derived from SE-immune adult chickens--were administered intraperitoneally to 1-day-old chicks before SE challenge, they conferred protection against SE organ invasion within 24 hr. This resistance mediated by SE-immune lymphokines was associated with a concomitant increase in peripheral blood polymorphonuclear leukocytes that peaked 4 hr after SE challenge. In the present study, we evaluated efficacy of SE-immune lymphokines in protecting chicks against SE organ invasion and alterations in peripheral blood polymorphonuclear leukocyte counts. Administration of SE-immune lymphokines to chicks either 30 min or 6 days before SE challenge caused a significant reduction in SE organ invasion. However, when SE-immune lymphokines were administered 2 days after SE challenge, there was no reduction in SE organ invasion. Both prophylactic (before SE challenge) and therapeutic (after SE challenge) administration of SE-immune lymphokines caused a significant increase in numbers of peripheral blood polymorphonuclear leukocytes. Results from these studies suggest that SE-immune lymphokines have potential value as an effective prophylactic but not as a therapeutic modulator of early resistance to SE organ invasion in neonatal leghorn chicks.

We characterized the inflammatory cell influx in day-old chicks induced by the i.p. administration of live Salmonella enteritidis (SE) and lymphokines from concanavalin A-stimulated SE-immune T lymphocytes (ILK). An i.p. injection of ILK along with 5 x 10(3) cfu SE increased the survival rate of chicks 48 h later from 70% (ILK-treated controls) compared with 25% (saline-treated). The injection of both the ILK and live SE (but not formalin-killed SE) resulted in an increased influx of inflammatory heterophils into the peritoneum that peaked at 4 h after the injections with no increase in peritoneal macrophages. The heterophil accumulation was not influenced by polymyxin B, but was sensitive to heat treatment (100 C for 1 h) of the ILK, suggesting that lipopolysaccharide (LPS) did not contribute to the induced accumulation of heterophils. Treatment of the chicks with nordihydroguaiaretic acid or indomethacin did not abrogate the induced heterophil accumulation, suggesting that arachidonic acid metabolites were not involved in the SE/ILK-induced accumulation of peritoneal heterophils. The results from the current studies indicate that 1) ILK-mediated resistance to SE-induced mortality is mediated by a rapid influx of inflammatory heterophils to the site of infection; 2) live SE, during invasion, are vital for the site-directed migration of the heterophils; and 3) the mechanisms of induced heterophil accumulation are unknown but involve neither LPS nor arachidonic acid metabolites.

We have previously shown that increased resistance to Salmonella enteritidis organ infectivity in day-old chicks was conferred by the immunoprophylactic administration of S. enteritidis-immune lymphokines (ILK). This resistance was associated with a significant increase in the number of circulating heterophils 4 h after ILK injection. The objective of the present study was to evaluate heterophil function following the administration of ILK in day-old chicks. Significant increases (P < 0.001) in adherence, chemotaxis, and phagocytosis of S. enteritidis were found with heterophils isolated from ILK-injected chickens compared to the heterophils isolated from birds injected with either pyrogen-free saline or lymphokines from non-immune T cells. After phagocytosis, the heterophils from the ILK-injected chickens were also able to kill significantly greater numbers of S. enteritidis more rapidly than did the heterophils from the saline-injected control birds (within 30 min, control cells killed 21.89% of the bacteria whereas ILK-treated cells killed 88.22%). We also found that the heterophils from the ILK-injected birds were more efficient killers of S. typhimurium, S. gallinarum, and E. coli. These results strongly suggest that the protection against S. enteritidis organ invasion induced by the prophylactic treatment of day-old chicks with ILK involves activated heterophils which migrate rapidly to the inflammatory stimulus where they phagocytize and kill the bacteria.

The effect of either low doses of the chicken coccidium, Eimeria tenella (ET) or high doses of the turkey coccidium, Eimeria adenoeides (EA) on Salmonella enteritidis (SE) organ invasion, as well as histological and morphometric changes in the ceca of Leghorn chicks, was investigated. In these studies, chicks were inoculated orally with either saline solution (control) or one of three doses of sporulated oocysts of either ET or EA at 1 d of age. Five days later, 10(4) cfu of SE were administered per os to all chickens. Chicks were killed and organs cultured for SE 6 d following bacterial challenge. A clear relationship of decreased SE organ invasion was observed by increasing the dose of sporulated oocysts of either ET or EA. Furthermore, the increased resistance to organ infectivity in both experiments was matched with an increase in lamina propria thickness based on morphometric analysis (P < .05), probably due to a marked infiltration of inflammatory cells in the cecal mucosa. In both trials, a significant correlation was found (r = -.98 for ET; r = -.99 for EA) between the rate of bacterial organ infectivity and lamina propria thickness of the cecum. These data indicate that the increased resistance to SE organ invasion following subclinical coccidial infections were associated with morphological changes in the ceca of Leghorn chicks.

Chickens injected with 5-fluorouracil (5-FU) were found to have a three- to nine-fold reduction in circulating heterophils without a reduction in the number of circulating mononuclear cells. A 50% organ invasive dose (ID50) for orally and intravenously administered Salmonella enteritidis (SE) was established in the heteropenic chickens. When challenged orally, about 150-fold fewer SE cells were required for organ invasion in the 5-FU-treated chickens; whereas an intravenous challenge of the heteropenic chickens required about 4000-fold fewer bacteria for organ invasion than found in the control birds. Significant (P < 0.0001) SE-dose-dependent reductions in body weight with increased mortality and lesions in the eye, heart, and thymus were found in the 5-FU-treated birds versus the control birds. Alternatively, the control birds had significantly more intestinal lesions than the 5-FU-treated birds. No consistent significant hepatic lesions were observed in either treatment group. Under the conditions used in these experiments, 5-FU treatment of chickens caused otherwise subclinical SE infections to rapidly become clinical infections with more severe extraintestinal organ infections, whereas the control chickens had infections that were primarily restricted to the intestine. These findings suggest that the heterophil is extremely important in controlling both initial SE organ invasion and subsequent disease pathogenesis in chickens.

Cytokines, derived from either concanavalin A-stimulated Salmonella enteritidis-immune chicken T lymphocytes [SE-immune Lymphocyte Stimulated Soluble Factor (LSSF)] or lipopolysaccharide-stimulated SE-immune chicken macrophages [SE-immune Macrophage Stimulated Soluble Factor (MSSF)], were evaluated for their ability to increase resistance to SE organ invasion in day-old Leghorn chicks. In Trial 1, day of hatch chicks were injected i.p. with either SE-immune LSSF or SE-nonimmune LSSF (control). In Trial 2, chicks were similarly injected with either SE-immune MSSF, SE-nonimmune MSSF, or SE-immune LSSF (positive control). Thirty minutes postinjection, all chicks were gavaged with an invasive dose of SE. Twenty-four hours later, livers and spleens from all chicks were cultured for SE. In Trial 1, SE-immune LSSF caused a rapid and marked protection (P < .01) against SE infection as determined by the number of chicks that were culture positive regardless of challenge dose. In Trial 2, SE-immune MSSF was not associated with protection against SE organ infection. These experiments demonstrate that SE-immune LSSF, but not MSSF, are able to confer protection against SE organ invasion in day-old Leghorn chicks. Thus, it appears that the stimulated immune T cell, and not the macrophage, is responsible for producing the soluble products that protected the chicks.

Chickens were treated prophylactically with the soluble products from Con A-stimulated T-lymphocytes from Salmonella enteritidis-infected chickens in order to investigate the effect of such prophylactic treatment on organ invasion by S. enteritidis. At 18 days of age, chicks were injected intraperitoneally with one of the following: A) the Amicon YM 10 ultrafiltrate from immunized chickens, B) the Amicon YM 10 ultraretentate material from immunized chickens, or C) the Amicon YM 10 ultraretentate material from control nonimmune chickens. Thirty minutes after lymphokine injection, all birds were challenged per os with 10(8) colony-forming units of S. enteritidis. At both 1 and 6 days post-challenge, prophylactic treatment of chickens with the immune retentate resulted in a 51-60% reduction in S. enteritidis organ invasion. The reduction in S. enteritidis organ invasion was associated with a significant increase in lamina propria thickness based on morphometric analysis (P < 0.05). These results demonstrate that the prophylactic administration of S. enteritidis-immune lymphokines induces protection against S. enteritidis organ invasion, which is associated with a measurable microanatomical change in the cecal mucosa.

Two-week-old Leghorn chicks injected intravenously with 5-fluorouracil (5-FU) were found to have a three- to fivefold reduction (P < .05) in circulating polymorphonuclear leukocytes (PMN) 5 to 10 days later. The reduction in PMN was dose-dependent with a dosage of 200 mg/kg body weight of 5-FU, causing suppression of PMN to almost undetectable levels. Increasing the dosage of 5-FU to 400 mg/kg had no further effect on reducing the number of PMN in the circulation nor were overt clinical signs of toxicity observed. Single per os administration of 10(6) viable Salmonella enteritidis (SE) to individual groups of chicks treated or not treated with 5-FU at 2, 4, 6, 8, 10, or 12 days postinjection resulted in a two- to sixfold increase in the incidence of SE organ invasion (liver and spleen) beginning on Day 4 postinjection and continuing through Day 12 postinjection. The greatest increase in organ invasion occurred on Days 8 through 10 postinjection and was inversely proportional to the greatest reduction in the number of circulating PMN. Using 5-FU, a granulocytopenic chicken model was developed that can be used to study the defensive role of avian heterophils against infectious agents. The results from these preliminary experiments indicate that PMN play a significant role in reducing organ invasion by SE in Leghorn chicks.

Two-week-old broiler chickens were inoculated orally with various doses of avian Eimeria that infect different sections of the intestine. The coccidia used were E. acervulina (duodenum), E. maxima (jejunum), E. brunetti (ileum) and E. tenella (caecum). The measurements evaluated were weight gains, intestinal lesion scores and changes in serum alkaline phosphatase (SAP) activity 6 days after infection. SAP activity was inversely proportional to both oocyst dose and gross intestinal lesion score, and directly proportional to weight gains with infections of E. acervulina, E. maxima and E. tenella. However, no significant changes in the SAP activity were found in those birds infected with E. brunetti. Taken together, the data suggest that SAP activity may be a sensitive marker of the pathogenesis of certain coccidial infections, especially in the duodenum, jejunum and caecum, that may be used as an additional factor in evaluating coccidial control measures.

We assayed two classes of immunoregulatory cytokines, colony-stimulating factors (CSF) and interferon (IFN), during and immediately after a primary coccidial infection in chickens. Coccidial infection induces significant alterations in serum colony-stimulating activity (CSA) and these alterations immediately precede the characteristic biphasic leukocytosis. CSA rose sharply during the first 24 h post-inoculation (PI), but returned to control levels by 48 h PI. At this time, we detected an increase in peripheral blood leukocytes which peaked at 96 h PI. A second phase of CSA increase began 96 h PI and peaked at 120-144 h PI which again preceded the second phase of leukocytosis. We also examined the production of IFN during the first 20 days PI. Splenic T cells from Eimeria maxima-infected chickens produced significantly less IFN on day 5 PI compared to T cells from the coccidia-free controls. By days 10 and 15 PI, there was no significant difference in IFN production between the T cells of infected and non-infected chickens. However, by day 20 PI, IFN production by the T cells of the infected birds produced significantly more IFN than the control T cells. The results of our studies indicated the differential production of two different cytokines by chickens during and following a primary coccidial infection. Based on these experiments, CSF may be some of the first cytokines produced during an E. maxima-infection, while IFN may be one of the later cytokines produced.

The role of T lymphocytes on the development of turkey coccidia in chickens was investigated to further document the involvement of immune mechanisms in the reactions of non-specific hosts to infections with species of Eimeria. All chickens treated with the T cell-specific immunosuppressive drug, cyclosporin A (CsA), 1, 2 or 3 days before or for 3 consecutive days immediately before inoculation with sporulated oocysts of Eimeria from the turkey, shed oocysts in their feces 7-12 days after inoculation. Those chickens treated with CsA after inoculation produced no oocysts although in one experiment a small number of oocysts were discharged by birds injected with CsA 1 day after inoculation. High serum levels of CsA were maintained in the CsA-treated chickens for at least 24 h post-inoculation with the oocysts. Immunosuppressive activity of the drug was determined by measuring lymphocyte blastogenesis in response to concanavalin A. These results demonstrate that the suppression of T lymphocyte activity in a non-specific host early during an infection with a heterologous species of Eimeria permits the complete intracellular development of the parasite. Our findings are suggestive of a role for T lymphocytes in preventing the development of Eimeria in non-specific hosts possibly via a lymphokine-mediated mechanism.

During the past ten years, subtotal thyroidectomy for hyperthyroidism was performed upon 43 children at Childrens Hospital of Los Angeles. There were no deaths, no recurrent laryngeal nerve injuries and no permanent hypoparathyroidism. During the one to ten year follow-up period, one patient had recurrent hyperthyroidism develop and was treated with 131I. Twenty-five patients are hypothyroid and require thyroid supplement; 14 are euthyroid and receiv no medication. Postoperative thyroid function did not correlate well with gland remnant size, degree of fibrosis or the extent of lymphoid follicle formation. Lymphocytic infiltration was more severe in patients who had hypothyroidism develop postoperatively. Transient hypocalcemia developed in 22 patients. The effectiveness and safety of the surgical treatment for hyperthyroidism in children is reaffirmed, and it is advocated for consideration over 131I or prolonged medical therapy.

When moderately halophilic bacteria adapt phenotypically to altered salinity, they modify the composition of their cell envelopes. These alterations are discussed in the context of how external salt concentration could be sensed and the adaptive changes triggered. A general model for salt sensing and signal transduction is suggested.

Cultured animal cells were pretreated with crude cytokines from E. tenella-immune chicken spleen cells to determine whether the invasion of these cells by sporozoites of E. tenella could be affected. Treatment of both primary chicken kidney (CK) cells and the established Madin-Darby bovine kidney cell line (MDBK) with the crude avian cytokines caused an increased invasion of the cultures by the parasite. Apparently, the cytokine treatment of the cells increased the susceptibility or sensitivity of the host cells to invasion as evidenced by the increased number of the total cells infected and not by an increased number of sporozoites/infected cell. This increased host cell susceptibility to infection induced by the crude avian cytokines was: (a) dose-dependent; (b) non-host species specific, since it affected both avian and bovine cells; and (c) non-parasite species specific, since cytokines from E. tenella and E. maxima-immune chicken spleen cells and E. nieschulzi-immune rat spleen cells also increased the invasion of the CK and MDBK cells by E. tenella. However, treatment of MDBK cells with lymphokines induced from purified E. tenella-immune splenic T lymphocytes inhibited the invasion of the cells by E.tenella sporozoites. Therefore, the increased susceptibility of the host cells is caused by a B cell-or macrophage-mediated factor. Although the identity of the factor or factor(s) is unknown, this in vitro-lymphokine-E.tenella system should provide an excellent model to aid in the identification of cellular binding sites and/or penetration mechanisms for cellular invasion by the Eimeria.

Oocysts were found in the feces of chickens (recipients) dosed orally with whole blood, liver, lung, or heart homogenates from chickens and turkeys (donors) inoculated 3 and 4 days previously with a mixture of 3.5 X 10(6) oocysts of chicken Eimeria. No oocysts were found in the feces of recipients given spleen homogenates from these same chickens and turkeys and none were found in the feces of recipients given similar material from uninoculated donors. Intracellular sporazoites were found in the peripheral blood of a turkey inoculated with chicken Eimeria. The results indicate that a small number of sporozoites are capable of invading and surviving for at least 4 days in the peripheral blood of chickens and turkeys.

Five children with end-stage renal disease resulting from cystinosis received seven cadaver renal allografts. Four recipients have functioning grafts eight to 55 months after receiving the transplant and one recipient lost two grafts at 17 and 26 months after the transplant. There was no florid recurrence of the Fanconi syndrome although proximal renal tubular dysfunction developed in two patients, in one in association with chronic rejection and in one without apparent etiology. Free cystine content of white blood cells, cultured skin fibroblasts and allograft tissue was significantly increased. Cystine crystals were observed in the mesangium of two grafts and in the interstitial tissue of all grafts; however, no cystine crystals were found in the tubules. The location of the cystine crystals, as well as the fact that the highest free level of cystine of allograft tissue was observed in the graft undergoing chronic rejection. led to the hypothesis that recipient cells infiltrating the graft were the source of cystine deposition. The data indicate that successful cadaveric transplantation does not correct the primary metabolic defect in cystinosis, thereby explaining the persistence of the extrarenal clinical manifestations, such as photophobia and hypothyroidism, after renal transplantation in cystinosis.

We evaluated gonadal function in 18 female and eight male patients with galactosemia due to transferase deficiency; it was normal in the males, but 12 females had signs of hypergonadotropic hypogonadism. All female patients had a 46,XX karyotype, normal levels of thyroid hormone and prolactin, and no anti-ovarian antibodies. The biologic activity of urinary gonadotropins was normal. Ultrasonography of the pelvis revealed that ovarian tissue was diminished or absent. Total estrogens increased in one of two patients after administration of human menopausal gonadotropin. The frequency of hypergonadotropic hypogonadism was higher in females in whom dietary treatment for galactosemia was delayed. Clinical course and mean erythrocyte galactose-1-phosphate and urinary galactitol levels did not correlate with ovarian function. We conclude that female patients with galactosemia have a high incidence of ovarian failure due to acquired ovarian atrophy. Galactose or its metabolites may be toxic to the ovarian parenchyma, particularly during the immediate neonatal period.

Proteolysis rates in vivo were measured in Escherichia coli cultures during treatment with dihydrostreptomycin and under various other conditions. Dihydrostreptomycin treatment caused an increase in the proteolysis rate, compared to untreated controls. The proteolytic system in vivo responsible for the elevated proteolysis in the early stages of dihydrostreptomycin treatment, or that during canavanine and puromycin treatment, were not inhibited by addition of phenylmethanesulphonyl fluoride. This agent did inhibit proteolysis rates in cultures whose growth was inhibited by starvation, or had been completely stopped by dihydrostreptomycin. It seems, therefore, that the extremely high proteolysis rates in cultures at this stage of dihydrostreptomycin treatment were due to the action of two protease systems: the one concerned with the breakdown of abnormal proteins, and the other concerned with normal protein turnover and active during a non-specific decline of growth. The proteolytic rate at complete growth inhibition brought about by dihydrostreptomycin was intermediate between those induced by canavanine and puromycin at the same stage of treatment. This indicated a similar hierarchy in the extent and nature of abnormality in the proteins synthesized under these conditions. The relationship between the abnormality of proteins induced by dihydrostreptomycin and the importance of this in the antibiotic mechanism is discussed.

Four children, treated with diazoxide for idiopathic leucine-sensitive (ILS) hypoglycemia of infancy, had follow-up studies at 2 to 10 yr of age to assess: (1) persistence of leucine sensitivity in later childhood. (2) pancreatic glucagon responses, and (3) the hormonal and glycemic effects of diazoxide therapy. On the third day after diazoxide therapy was stopped, the mean +/- S.E. baseline plasma glucose level (65.3 +/- 3.4 mg/dl) was significantly (P less than 0.005) lower than that of the controls (80.1 +/- 3.1 mg/dl). Corresponding mean plasma immunoreactive insulin (IRI) and immunoreactive glucagon (IRG) values were higher than control values but the differences were not significant. After the oral administration of leucine (50 mg/kg) in the ILS children, the mean plasma IRI level rose from 15.2 +/- 4.1 to 59 +/- micro U/ml, the mean plasma glucose concentration fell to 36.0 +/- 3.3 mg/dl and the mean plasma IRG level rose from 196 +/- 16 to 261 +/- 41 pg/ml. These responses were significantly greater (P less than 0.05 to 0.005) than those of control children who received 150 mg/kg of leucine. Intravenous arginine administration caused similar changes in mean plasma glucose, IRI and IRG values in the ILS and control children. During diazoxide therapy in the ILS children, the baseline mean plasma glucose level (89.5 +/- 4.2 mg/dl) was significantly (P greater than 0.005) higher than without therapy. Corresponding mean plasma IRI and IRG values decreased with therapy but the differences were not significant. Diazoxide therapy blunted the changes induced by leucine administration in the ILS children but did not significantly change their response to arginine infusion. Our results indicate that marked sensitivity to leucine persists after infancy in the ILS children. Their IRG responses are appropriate to the stimuli, indicating that their pancreatic alpha cells do not share the abnormality of the beta cells. Diazoxide therapy increases baseline plasma glucose levels and inhibits IRI responses to leucine in ILS children, but it has little if any effect on IRG responses.

Two lines of Eimeria tenella (PS and FS65) and one line of E. necatrix (FS144) of U.S. origin were passaged in chicken embryos. The embryo-passaged line of E. tenella PS was significantly less pathogenic than the parent line when tested in chickens after 33 embryo passages. After 29 embryo passages, E. tenella FS65 was just as pathogenic to chickens as the parent line. A comparison of the immunogenicity of the embryo-adapted lines and the parent lines in chickens showed that embryo-passaged E. tenella PS and E. tenella FS65 were as immunogenic as parent lines. The embryo-passaged line of E. necatrix FS144 was significantly less pathogenic when tested in chickens after 30 embryo passages. Although there was also some loss of immunogenicity in the embryo-passaged line of E. necatrix, immunity to this parasite could be achieved by increasing the dose of oocysts.

The serial passage of 3 strains of Eimeria from the chicken in embryonating turkey embryos is reported here. All 3 strains increased their reproduction in turkey embryos; reproduction being equal to or better than the same lines maintained in chicken embryos. Pathogenicity and immunogenicity trials conducted in chickens using these 3 strains of Eimeria showed that all 3 turkey embryo-adapted lines were significantly less pathogenic than the parent lines. Only 1 of the lines (E. tenella (PS) turkey embryo-adapted) maintained its immunogenic potential.

The diagnosis of diabetic ketoacidosis must be suspected and the initiation of treatment should be prompt to provide a satisfactory outcome in the treatment of diabetic ketoacidosis. Corrections of fluid and electrolyte deficiencies should be made slowly; rapid "push"injections or large infusions of sodium bicarbonate should avoided and ample amounts of potassium should be given early. Precautions should be taken so that blood glucose concentrations do not fall rapidly, and so that blood glucose levels of 250-300 mg/100 ml are maintained by the administration of 5-10% glucose solutions. Bicarbonate therapy is indicated only in severe acidosis (pH less than or equal to 7.1). Physicians who are trained in the care of diabetes mellitus should supervise the treatment. In our hospital the same staff physicians and fellows attend all patients with diabetes. In addition the efforts of our house staff and nurses have contributed significantly to the care of these patients.

After the infusion of fructose, 0.25 g/kg body wt, the mean peak plasma uric acid level was 5.4 +/- 0.7 (SEM) mg/100 ml in six normal children and was not significantly increased compared with that of the mean basal value of 4.1 +/- 0.5 mg/100 ml. The mean blood inorganic phosphate (Pi) levels were significantly less than the mean fasting value after fructose. Blood glucose, lactic acid, and fructose levels were significantly increased after fructose, but serum magnesium levels did not change. In two patients with hereditary fructose intolerance (HFI) the peak blood uric acid levels were 12.1 and 7.6 mg/100 ml, respectively, after fructose. In both patients the blood glucose concentrations decreased 69 and 26 mg/100 ml below the fasting levels after fructose. The serum Pi level decreased 2.3 and 1.2 mg/100 ml below fasting values, decrements greater than the mean decrement in serum Pi of 0.8 +/- 0.2 mg/100 ml which occurred in six normal children. The mean uric acid excretion, expressed as milligrams per mg urinary creatinine, was 0.6 +/- 0.1 (SEM) before fructose in the normal children and increased significantly to 1.0 +/- mg/mg creatinine after fructose. In two patients with HFI the uric acid excretion increased four- to fivefold after fructose administration; the increased uric acid excretion in HFI exceeded that of normal children. In three patients with galactosemia, increases in blood uric acid levels after galactose ingestion were similar to those in normal children after fructose, but less than those in patients with HFI after fructose. The serum Pi levels decreased less in galactosemic patients after galactose administration than in patients with HFI after fructose infusion. These studies support the hypothesis that fructose-induced hyperuricemia results from degradation of adenosine monophosphate. This effect appears to be specific for fructose. The lack of hyperruricemia in galactosemia patients after galactose ingestion may be explained by the observation that galactose is phosphorylated more slowly than fructose.

Thyroid glands from 33 children with hyperthyroidism and nine with juvenile lymphocytic thyroiditis were examined histologically and for IgG, IgA, IgM, and C3 by immunofluorescent staining. There was no significant difference between glands with JLT and those with hyperthyroidism in the degree of lymphoid infiltration or lymphoid follicle formation. In thyroiditis there was no correlation between the degree of histologic abnormalities and the presence of immunofluorescent staining for IgG, IgM, or IgA. In hyperthyroidism there was a correlation between the degree of histologic abnormalities and the presence of IgG. In both groups of patients LI and LFF were distinctly more severe in glands positive for C3. Postsurgical hypothyroidism correlated with LI but not with LFF, IgG, or C3.
