Abstract

Cancer genomes are frequently characterized by numerical and structural chromosomal abnormalities. Here we integrated a centromere-specific inactivation approach with selection for a conditionally essential gene, a strategy termed CEN-SELECT, to systematically interrogate the structural landscape of mis-segregated chromosomes. We show that single-chromosome mis-segregation into a micronucleus can directly trigger a broad spectrum of genomic rearrangement types. Cytogenetic profiling revealed that mis-segregated chromosomes exhibit 120-fold-higher susceptibility to developing seven major categories of structural aberrations, including translocations, insertions, deletions, and complex reassembly through chromothripsis coupled to classical non-homologous end joining. Whole-genome sequencing of clonally propagated rearrangements identified random patterns of clustered breakpoints with copy-number alterations resulting in interspersed gene deletions and extrachromosomal DNA amplification events. We conclude that individual chromosome segregation errors during mitotic cell division are sufficient to drive extensive structural variations that recapitulate genomic features commonly associated with human disease.

Abstract

The mitotic checkpoint ensures accurate chromosome segregation through assembly of the mitotic checkpoint complex (MCC), a soluble inhibitor of the anaphase-promoting complex/cyclosome (APC/C) produced by unattached kinetochores. MCC is also assembled during interphase by Mad1/Mad2 bound at nuclear pores, thereby preventing premature mitotic exit prior to kinetochore maturation and checkpoint activation. Using degron tagging to rapidly deplete the AAA+ ATPase TRIP13, we show that its catalytic activity is required to maintain a pool of open-state Mad2 for MCC assembly, thereby supporting mitotic checkpoint activation, but is also required for timely mitotic exit through catalytic disassembly of MCC. Strikingly, combining TRIP13 depletion with elimination of APC15-dependent Cdc20 ubiquitination/degradation results in a complete inability to exit mitosis, even when MCC assembly at unattached kinetochores is prevented. Thus, mitotic exit requires MCC produced either in interphase or mitosis to be disassembled by TRIP13-catalyzed removal of Mad2 or APC15-driven ubiquitination/degradation of its Cdc20 subunit.

Abstract

Background This study aimed to investigate the risk factors for community-acquired urinary tract infection (UTI) caused by extended-spectrum beta-lactamase (ESBL)-positive bacteria in infants. Methods We retrospectively reviewed the medical records of infants aged < 1 year with first UTI from 2018 to 2019 at two tertiary centers in Korea. Data analyzed included clinical findings, birth history, delivery mode, milk type, use of postpartum care center, and previous use of antibiotics both in the patient and mother. Results Of 265 patients, 62 (23.4%) were diagnosed with first UTI caused by ESBL-positive bacteria at the median age of 3.6 (interquartile range (IQR) 2.3-5.4) months. Maternal use of antibiotics during pregnancy (29.0 vs. 10.3%, p < 0.001) and Klebsiella species (19.4% vs. 4.9%, p < 0.001) were significantly associated with ESBL-positive UTIs and remained valid in the multivariate analysis (odds ratio [OR], 3.40; 95% confidence interval [CI] 1.61-7.19, p = 0.001, and OR 5.26; 95% CI 2.03-13.13, p = 0.001, respectively). Previous antibiotic exposure of patients, previous hospitalization, prematurity, delivery mode, milk type, and use of postpartum care center were not significantly different between ESBL-positive and ESBL-negative groups. With respect to the clinical course of UTI, the ESBL-positive group presented a higher number of blood leukocytes (p = 0.041) and longer hospital stay (p < 0.001) than the ESBL-negative group. Conclusions About one-fourth of infantile UTI cases were ESBL-positive. Prenatal antibiotic exposure of mothers and Klebsiella species were associated with community-acquired UTI caused by ESBL-positive bacteria. 247. Towards an analysis of passive verbs in Q’antel K’iche’

Abstract

We give a description of the morphology and syntax of passive verbs in the Q’antel dialect of K’iche’, a Mayan language spoken in the department of Quetzaltenango (Aissen et al. 2017). Our description is based on two years of fieldwork carried out both ex situ and in situ, and is the first published work on the Q’antel dialect to our knowledge. We show that, like other dialects the Q’antel dialect has three formally- distinct patterns by which the passive is formed, each of which behave differently with regards to syntax and argument structure (Mondloch 1981). We further present new data about passives of intransitive verbs, ditransitive verbs, and verbs with anaphoric arguments. Last, we use these data to build towards an analysis that supports the traditional analysis of the passive, rather than the smuggling analysis presented in Collins 2005.

249. Light and emotion : exploring human affect in lighting

Abstract

This thesis starts by questioning the efficiency of the current basis of lighting design practice on modern workspaces. The study argues the importance of shifting the basis of guidance from the types of spaces, and activities into human psychological states in the Introduction. Both visual perceptions of the space and human emotion states are theoretically examined as a potential basis for lighting design practice. As a result, the human emotion state, ‘affect’, from the concept of the circumplex model of affect has been chosen as a better exploratory tool. Human ‘affect’ states of ‘lively’ and ‘relaxing’ were hypothesized mental states that people could benefit from in the workspace. A detailed field study, which consisted of two separate phases, was carried out to test the hypothesis. In the first phase of the field study, a group of lighting designers was invited to devise a set of lighting design concepts that explore activation and pleasantness in the circumplex model of affect with the use of smart control of lighting. Then, the thesis investigated the design elements of the lighting settings as well as their photometric and colorimetric characteristics. The result indicated that although the design elements showed particular groupings, such characteristics were not effectively expressed using the current photometric and colorimetric variables. In the second phase of the field study, 15 lighting settings in a controlled interior environment were set up based on the results of the first phase. A group of participants were invited to assess the settings by rating of their visual perceptions of the space and their emotion state. The results indicated that the settings inspired by the designers effectively affect human emotion states while providing high quality visual perceptions of the space. However, the two dimensions of human emotion, activation and pleasantness, did not provide, in detail, a statement of human emotion. Therefore, the thesis has proposed a new indicator that involved four different zones, defined by the two dimensions as an effective tool of measuring human emotional experiences from lighting. In conclusion, the thesis successfully explores the possibilities of an emotion-based lighting design approach and explains their impacts by the field experiment as well as developing and investigating the effectiveness of a new indicator of human affect. The work described in this thesis would work as a fundamental but crucial step to promote human well-being in workspaces by fulfilling various human needs and the potentials of smart lighting technologies.
