Abstract Beer is made via the fermentation of an aqueous extract predominantly composed of malted barley flavoured with hops. The transforming microorganism is typically a single strain of Saccharomyces cerevisiae, and for the majority of major beer brands the yeast strain is a unique component. The present yeast used to make Guinness stout brewed in Dublin, Ireland, can be traced back to 1903, but its origins are unknown. To that end, we used Illumina and Nanopore sequencing to generate whole-genome sequencing data for a total of 22 S. cerevisiae yeast strains: 16 from the Guinness collection and 6 other historical Irish brewing. The origins of the Guinness yeast were determined with a SNP-based analysis, demonstrating that the Guinness strains occupy a distinct group separate from other historical Irish brewing yeasts. Assessment of chromosome number, copy number variation and phenotypic evaluation of key brewing attributes established Guinness yeast-specific SNPs but no specific chromosomal amplifications. Our analysis also demonstrated the effects of yeast storage on phylogeny. Altogether, our results suggest that the Guinness yeast used today is related to the first deposited Guinness yeast; the 1903 Watling Laboratory Guinness yeast.

We propose and analyze an algorithmic framework for "bias bounties": events in which external participants are invited to propose improvements to a trained model, akin to bug bounty events in software and security. Our framework allows participants to submit arbitrary subgroup improvements, which are then algorithmically incorporated into an updated model. Our algorithm has the property that there is no tension between overall and subgroup accuracies, nor between different subgroup accuracies, and it enjoys provable convergence to either the Bayes optimal model or a state in which no further improvements can be found by the participants. We provide formal analyses of our framework, experimental evaluation, and findings from a preliminary bias bounty event.

Abstract Flood exposure has been linked to shifts in population sizes and composition. Traditionally, these changes have been observed at a local level providing insight to local dynamics but not general trends, or at a coarse resolution that does not capture localized shifts. Using historic flood data between 2000-2023 across the Contiguous United States (CONUS), we identify the relationships between flood exposure and population change. We demonstrate that observed declines in population are statistically associated with higher levels of historic flood exposure, which may be subsequently coupled with future population projections. Several locations have already begun to see population responses to observed flood exposure and are forecasted to have decreased future growth rates as a result. Finally, we find that exposure to high frequency flooding (5 and 20-year return periods) results in 2-7% lower growth rates than baseline projections. This is exacerbated in areas with relatively high exposure to frequent flooding where growth is expected to decline over the next 30 years.

219. Exposure of the US population to extreme precipitation risk has increased due to climate change

Abstract The magnitude and frequency of extreme precipitation events in the early twenty-first century have already proven to be increasing at a rate more quickly than previously anticipated. Currently, the biggest consequence of the change in extreme precipitation is the lack of a climate-adjusted national standard taking into account these recent increases that could be used to prevent life and property loss from catastrophic precipitation-driven floods. Here, we address how severe the change in extreme precipitation compares against the current national standard for precipitation climatology (NOAA Atlas 14) and how much of the population is affected by the underestimation of this risk in the contiguous United States (CONUS). As a result, extreme precipitation in the early twenty-first century has outpaced our current national standard in half of CONUS, and the heavy precipitation events experienced recently are quickly becoming a “new normal”, which will increase in severity and frequency in a continually changing climate. Over three-quarters of the U.S. population will likely experience this new normal occurrence of extreme precipitation. As much as one-third of the population is expected to experience the current definition of a 1-in-100-year storm as often as three times in their lifetime. Additionally, the current precipitation standards for designing transportation infrastructure and urban stormwater drainage systems that are built upon Atlas 14 may be insufficient to protect the public's safety and personal/community property from severe flooding. Areas where flood risk is mitigated by operating hydraulic and adaptation structures urgently need to assess the impact of the increased-hourly extreme precipitation and reevaluate their applicable operation rules. Understanding and predicting patterns and the likelihood of short-duration heavy precipitation would be beneficial in preparing for severe precipitation-driven disasters, such as flash floods and landslides, which would happen more frequently in a changing climate. Following the results of this analysis, accelerating the development and dissemination of the next generation of the national standard that has been climatically adjusted to adapt to the new normal is strongly recommended.

The Module-0 Demonstrator is a single-phase 600 kg liquid argon time projection chamber operated as a prototype for the DUNE liquid argon near detector. Based on the ArgonCube design concept, Module-0 features a novel 80k-channel pixelated charge readout and advanced high-coverage photon detection system. In this paper, we present an analysis of an eight-day data set consisting of 25 million cosmic ray events collected in the spring of 2021. We use this sample to demonstrate the imaging performance of the charge and light readout systems as well as the signal correlations between the two. We also report argon purity and detector uniformity measurements and provide comparisons to detector simulations.

PurposeMultiple sclerosis (MS) is an immune-mediated, neuroinflammatory disease of the central nervous system and in industrialised countries is the most common cause of progressive neurological disability in working age persons. While treatable, there is substantial interindividual heterogeneity in disease activity and response to treatment. Currently, the ability to predict at diagnosis who will have a benign, intermediate or aggressive disease course is very limited. There is, therefore, a need for integrated predictive tools to inform individualised treatment decision making.ParticipantsEstablished with the aim of addressing this need for individualised predictive tools, FutureMS is a nationally representative, prospective observational cohort study of 440 adults with a new diagnosis of relapsing-remitting MS living in Scotland at the time of diagnosis between May 2016 and March 2019.Findings to dateThe study aims to explore the pathobiology and determinants of disease heterogeneity in MS and combines detailed clinical phenotyping with imaging, genetic and biomarker metrics of disease activity and progression. Recruitment, baseline assessment and follow-up at year 1 is complete. Here, we describe the cohort design and present a profile of the participants at baseline and 1 year of follow-up.Future plansA third follow-up wave for the cohort has recently begun at 5 years after first visit and a further wave of follow-up is funded for year 10. Longer-term follow-up is anticipated thereafter.

225. Text Types and Their Relation to Efficacy in Beginning Reading Interventions

- Abstract
Researchers disagree about the value of controlling the decodability of texts for students with reading difficulty, specifically what type of text they should read: "decodable texts" (words limited to taught patterns), "nondecodable" texts (those not limited by instruction), or "both." We analyzed the effects of reading intervention for elementary-age students with reading difficulty (k = 119) to determine whether effects varied by the type of texts students read--decodable, nondecodable, or both--compared with interventions without text reading. Inadequate information was available to code text type for 22 interventions including text reading; effect sizes were calculated for 97 studies. Effects for interventions with decodable or nondecodable reading did not differ from no-text interventions. For both types of interventions, the effect (g = 0.28) approached significance versus no-text, 95% CI [-0.09, 0.65]. Disaggregating effects by whether the measures were standardized or researcher-designed showed a significant both-types effect, g = 0.45, 95% CI [0.02, 0.89] relative to no-text. Disaggregating by whether outcomes were for word recognition or reading comprehension showed a positive both-types effect for word recognition outcomes; data were inadequate to examine comprehension. A possible confounding effect of time spent reading was tested but was uncorrelated with the intervention effect. The both-types finding suggests the possible value of varied reading experiences in intervention, but this analysis did not account for other factors that might be correlated with text type and the intervention effect. Furthermore, more comprehensive reporting about text types is important for replication and meta-analytic review.

226. Using the Features of Written Compositions to Understand Reading Comprehension

- Abstract
The current study is an exploration of the association between the characteristics of students' written compositions and their reading comprehension performance. We address the empirical question about the degree to which writing is predictive of reading comprehension by comparing the utility of several popular written composition metrics. These include the 6+1 Trait® Model of Writing, a metric from curriculum-based measurement in written expression, Coh-Metrix, and automated counts of word features. The dependent variable for all data analytic models was the reading comprehension score (RC score) from the FAIR-FS. First, we conducted two random forest regressions, one containing the reading skills, namely, scores on the vocabulary, syntax, word recognition, and reading fluency tasks on the FAIR-FS and the other containing these reading skills in addition to all of the writing scores calculated from the written composition task. Second, we use the importance indices from the random forest regression to select skills to include in an explanatory regression model. In addition to finding that the writing related skills added to the importance, we further found that the most important skills in the random forest regression were reading-related followed by the various instantiations of curriculum-based measures, number of morphemes, and incidence of gerund density. These signify a unique contribution between curriculum-based measures and reading comprehension when accounting for comprehension-related reading skills thus furthering our understanding of the interdependency between the components of writing and reading.

This article presents the rationale, evidence base, and strategies for teachers and related service personnel to implement peer-assisted Augmentative & Alternative Communication (AAC) modeling with their students with complex communication needs within inclusive settings across the school day. Specific steps for teaching the student's same-age peers to use aided AAC modeling are provided, as well as overall recommendations for implementation. Aided AAC modeling examples and strategies for collecting data on student AAC use are also included.

Through a Multitiered Systems of Support (MTSS) framework that incorporates sociocultural theory, school psychologists can collaborate with other educators and community members to support these students and foster academic success.

229. Promising Strategies for Teaching Augmentative and Alternative Communication in Inclusive Educational Settings: A Systematic Review

- Abstract
Purpose: The purpose of this article was to conduct a systematic review of the literature on teaching or increasing the use of augmentative and alternative communication (AAC) by students with significant intellectual disabilities and complex communication needs (CCNs) within inclusive school settings. Method: A systematic review of research literature from 1998 to 2022 was completed using multiple electronic databases, as well as citation chaining and cited author reference searches. Three hundred two articles were located with 17 meeting criteria for inclusion in this systematic review. Articles were initially screened by one author for potential inclusion; the remaining 83 were coded by one author and reviewed for consensus by three authors for inclusion in this review. Articles were analyzed and rated regarding both level of research design and quality of methodology. Results: All studies reported positive outcomes, with students with significant intellectual disabilities and CCN, of AAC intervention implemented in an inclusive setting. Fourteen over 17 studies were single-case designs with an average rating of 76% on the Single-Case Experimental Design measure of methodological quality. Less than half of the studies assessed generalization of subject learning. Review of interventions used in the studies, however, revealed the majority of studies utilized packages of interventions, making it difficult to determine the effectiveness of individual approaches. Conclusions: This systematic review indicates there are promising interventions within inclusive settings for improved use of AAC by students with significant intellectual disabilities and CCN. Further research is needed, however, to obtain more data on generalization of student gains as well as to determine which specific interventions might be the most successful for this population of students in inclusive settings.

- Abstract
We share insights from a mixed methods study to describe the experience of educational developers participating in a community of practice (CoP). This CoP consisted of 19 leaders at eight institutions, serving collectively as facilitators of a hybrid, across-institutional, seven-week workshop series on course design for graduate students and postdoctoral scholars. From results of a survey examining the functional and sustaining features of the community, we illustrate that members experience the community aspect of our CoP primarily as a sense of satisfaction/personal fulfillment and, to an extent, in the effectiveness/productiveness of collaborations between members. Members of our CoP experience several community-related benefits specific to their across-institution collaborations. We demonstrate the importance of members' sense of belonging, overall professional development, and well-being for sustaining our CoP.

Radioactivity induced by cosmic muon spallation is a dominant source of backgrounds for $\mathcal{O}(10)~$MeV neutrino interactions in water Cherenkov detectors. In particular, it is crucial to reduce backgrounds to measure the solar neutrino spectrum and find neutrino interactions from distant supernovae. In this paper we introduce new techniques to locate muon-induced hadronic showers and efficiently reject spallation backgrounds. Applying these techniques to the solar neutrino analysis with an exposure of $2790\times22.5$~kton.day increases the signal efficiency by $12.6\%$, approximately corresponding to an additional year of detector running. Furthermore, we present the first spallation simulation at SK, where we model hadronic interactions using FLUKA. The agreement between the isotope yields and shower pattern in this simulation and in the data gives confidence in the accuracy of this simulation, and thus opens the door to use it to optimize muon spallation removal in new data with gadolinium-enhanced neutron capture detection.

The viability of a new drug molecule is a time and resource intensive task that makes computer-aided assessments a vital approach to rapid drug discovery. Here we develop a machine learning algorithm, iMiner, that generates novel inhibitor molecules for target proteins by combining deep reinforcement learning with real-time 3D molecular docking using AutoDock Vina, thereby simultaneously creating chemical novelty while constraining molecules for shape and molecular compatibility with target active sites. Moreover, through the use of various types of reward functions, we can generate new molecules that are chemically similar to a target ligand, which can be grown from known protein bound fragments, as well as to create molecules that enforce interactions with target residues in the protein active site. The iMiner algorithm is embedded in a composite workflow that filters out Pan-assay interference compounds, Lipinski rule violations, and poor synthetic accessibility, with options for cross-validation against other docking scoring functions and automation of a molecular dynamics simulation to measure pose stability. Because our approach only relies on the structure of the target protein, iMiner can be easily adapted for future development of other inhibitors or small molecule therapeutics of any target protein.

A new search for the diffuse supernova neutrino background (DSNB) flux has been conducted at Super-Kamiokande (SK), with a $22.5\times2970$-kton$\cdot$day exposure from its fourth operational phase IV. The new analysis improves on the existing background reduction techniques and systematic uncertainties and takes advantage of an improved neutron tagging algorithm to lower the energy threshold compared to the previous phases of SK. This allows for setting the world's most stringent upper limit on the extraterrestrial $\bar{\nu}_e$ flux, for neutrino energies below 31.3 MeV. The SK-IV results are combined with the ones from the first three phases of SK to perform a joint analysis using $22.5\times5823$ kton$\cdot$days of data. This analysis has the world's best sensitivity to the DSNB $\bar{\nu}_e$ flux, comparable to the predictions from various models. For neutrino energies larger than 17.3 MeV, the new combined $90\%$ C.L. upper limits on the DSNB $\bar{\nu}_e$ flux lie around $2.7$ cm$^{-2}$$\cdot$$\text{sec}^{-1}$, strongly disfavoring the most optimistic predictions. The Deep Underground Neutrino Experiment (DUNE) will produce world-leading neutrino oscillation measurements over the lifetime of the experiment. In this work, we explore DUNE's sensitivity to observe charge-parity violation (CPV) in the neutrino sector, and to resolve the mass ordering, for exposures of up to 100 kiloton-megawatt-years (kt-MW-yr). The analysis includes detailed uncertainties on the flux prediction, the neutrino interaction model, and detector effects. We demonstrate that DUNE will be able to unambiguously resolve the neutrino mass ordering at a 3$\sigma$ (5$\sigma$) level, with a 66 (100) kt-MW-yr far detector exposure, and has the ability to make strong statements at significantly shorter exposures depending on the true value of other oscillation parameters. We also show that DUNE has the potential to make a robust measurement of CPV at a 3$\sigma$ level with a 100 kt-MW-yr exposure for the maximally CP-violating values $\delta_{\rm CP}} = \pm\pi/2$. Additionally, the dependence of DUNE's sensitivity on the exposure taken in neutrino-enhanced and antineutrino-enhanced running is discussed. An equal fraction of exposure taken in each beam mode is found to be close to optimal when considered over the entire space of interest.

In order to improve Super-Kamiokande's neutron detection efficiency and to thereby increase its sensitivity to the diffuse supernova neutrino background flux, 13 tons of $\rm Gd_2(\rm SO_4)_3\cdot \rm 8H_2O$ (gadolinium sulfate octahydrate) was dissolved into the detector's otherwise ultrapure water from July 14 to August 17, 2020, marking the start of the SK-Gd phase of operations. During the loading, water was continuously recirculated at a rate of 60 m$^3$/h, extracting water from the top of the detector and mixing it with concentrated $\rm Gd_2(\rm SO_4)_3\cdot \rm 8H_2O$ solution to create a 0.02% solution of the Gd compound before injecting it into the bottom of the detector. A clear boundary between the Gd-loaded and pure water was maintained through the loading, enabling monitoring of the loading itself and the spatial uniformity of the Gd concentration over the 35 days it took to reach the top of the detector. During the subsequent commissioning the recirculation rate was increased to 120 m$^3$/h, resulting in a constant and uniform distribution of Gd throughout the detector and water transparency equivalent to that of previous pure-water operation periods. Using an Am-Be neutron calibration source the mean neutron capture time was measured to be $115\pm1$ $\mu$s, which corresponds to a Gd concentration of $111\pm2$ ppm, as expected for this level of Gd loading.
