Abstract Increasing the application of nitrogen fertilizer is the main approach to increase rice production, but it also brings problems of environmental pollution and increases agricultural production costs. Cultivating high‐yielding and high nitrogen use efficiency (NUE) rice varieties is an important approach to solving this problem. The rice varieties carrying dep1 (dense and erect panicle 1) have both high grain yield and high NUE. However, their plant traits have not been fully explored. In this study, two rice near‐isogenic lines carrying dep1 (NIL‐DEP1 and NIL‐dep1) were grown in paddy fields under 0, 120 and 270 kg N ha−1. We analyzed agronomic traits of panicle type, plant type, leaves and roots, and physiological traits of vascular bundles, photosynthetic rate and carbon and nitrogen transport. The results showed that the NIL‐dep1 exhibited higher grain yield and NUE than NIL‐DEP1, mainly due to the higher spikelet number per panicle, grain filling percentage and dry matter production. Compared with NIL‐DEP1, NIL‐dep1 had improved flag leaf morpho–physiological traits, including erect flag leaves, greater leaf thickness and specific leaf weight, higher root dry weight, root length, root volume and root surface area, and a better canopy structure, as reflected by a lower light interception percent and canopy extinction coefficient, leading to better photosynthetic performance and dry matter production. In addition, NIL‐dep1 exhibited better vascular bundle traits of peduncle and enhanced dry matter, stem carbon and nitrogen translocation during grain filling. In conclusion, NIL‐dep1 had high grain yield and NUE by improved agronomic and physiological traits and increasing carbon and nitrogen translocation during grain filling. These traits mentioned above could be used to select and breed high grain yield with high NUE rice varieties.

Abstract Background Bone or brain metastases may develop in 20–40% of individuals with late‐stage non‐small‐cell lung cancer (NSCLC), resulting in a median overall survival of only 4–6 months. However, the primary lung cancer tissue's distinctions between bone, brain and intrapulmonary metastases of NSCLC at the single‐cell level have not been underexplored. Methods We conducted a comprehensive analysis of 14 tissue biopsy samples obtained from treatment‐naïve advanced NSCLC patients with bone (n = 4), brain (n = 6) or intrapulmonary (n = 4) metastasis using single‐cell sequencing originating from the lungs. Following quality control and the removal of doublets, a total of 80 084 cells were successfully captured. Results The most significant inter‐group differences were observed in the fraction and function of fibroblasts. We identified three distinct cancer‐associated fibroblast (CAF) subpopulations: myofibroblastic CAF (myCAF), inflammatory CAF (iCAF) and antigen‐presenting CAF (apCAF). Notably, apCAF was prevalent in NSCLC with bone metastasis, while iCAF dominated in NSCLC with brain metastasis. Intercellular signalling network analysis revealed that apCAF may play a role in bone metastasis by activating signalling pathways associated with cancer stemness, such as SPP1‐CD44 and SPP1‐PTGER4. Conversely, iCAF was found to promote brain metastasis by activating invasion and metastasis‐related molecules, such as MET hepatocyte growth factor. Furthermore, the interaction between CAFs and tumour cells influenced T‐cell exhaustion and signalling pathways within the tumour microenvironment. Conclusions This study unveils the direct interplay between tumour cells and CAFs in NSCLC with bone or brain metastasis and identifies potential therapeutic targets for inhibiting metastasis by disrupting these critical cell–cell interactions.

BackgroundEpigenome-wide association studies (EWAS) have identified CpG sites associated with HIV infection in blood cells in bulk, which offer limited knowledge of cell-type specific methylation patterns associated with HIV infection. In this study, we aim to identify differentially methylated CpG sites for HIV infection in immune cell types: CD4+ T-cells, CD8+ T-cells, B cells, Natural Killer (NK) cells, and monocytes.MethodsApplying a computational deconvolution method, we performed a cell-type based EWAS for HIV infection in three independent cohorts (Ntotal = 1,382). DNA methylation in blood or in peripheral blood mononuclear cells (PBMCs) was profiled by an array-based method and then deconvoluted by Tensor Composition Analysis (TCA). The TCA-computed CpG methylation in each cell type was first benchmarked by bisulfite DNA methylation capture sequencing in a subset of the samples. Cell-type EWAS of HIV infection was performed in each cohort separately and a meta-EWAS was conducted followed by gene set enrichment analysis.ResultsThe meta-analysis unveiled a total of 2,021 cell-type unique significant CpG sites for five inferred cell types. Among these inferred cell-type unique CpG sites, the concordance rate in the three cohorts ranged from 96% to 100% in each cell type. Cell-type level meta-EWAS unveiled distinct patterns of HIV-associated differential CpG methylation, where 74% of CpG sites were unique to individual cell types (false discovery rate, FDR ConclusionOur findings uncovered computationally inferred cell-type specific modifications in the host epigenome for people with HIV that contribute to the growing body of evidence regarding HIV pathogenesis.

BackgroundEvidence shows people living with CHB even with a normal ALT (40U/L as threshold) suffer histological disease and there is still little research to evaluate the potential benefit of antiviral benefits in them.MethodsWe retrospectively examined 1352 patients who underwent liver biopsy from 2017 to 2021 and then obtained their 1-year follow-up data to analyze.ResultsALT levels were categorized into high and low, with thresholds set at >29 for males and >15 for females through Youden’s Index. The high normal ALT group showed significant histological disease at baseline (56.43% vs 43.82%, p< 0.001), and better HBV DNA clearance from treatment using PSM (p=0.005). Similar results were obtained using 2016 AASLD high normals (male >30, female >19). Further multivariate logistic analysis showed that high normal ALT (both criterias) was an independent predictor of treatment (OR 1.993, 95% CI 1.115-3.560, p=0.020; OR 2.000, 95% CI 1.055-3.793, p=0.034) Both of the models had higher AUC compared with current scoring system, and there was no obvious difference between the two models (AUC:0.8840 vs 0.8835)ConclusionMale >30 or female >19 and Male >29 or female>15 are suggested to be better thresholds for normal ALT. Having a high normal ALT in CHB provides a potential benefit in antiviral therapy.

214. Outcomes of lung transplantation from donors with hepatitis C viremia with outpatient initiation of antiviral therapy

Transplantation of lungs from donors with active hepatitis C viremia with early initiation of antiviral therapy has been shown to have similar short- and medium-term outcomes compared to transplantation of lungs from nonviremic donors. Consideration of hepatitis C viremic lungs is particularly helpful in patients with anticipated prolonged time on the waiting list. Whether clinical outcomes remain favorable with delay of initiation of antiviral therapy to the outpatient setting or in patients with higher severity of illness is not well understood. Our transplant center considered hepatitis C nucleic acid testing positive (NAT+) donors for all waitlisted lung transplant candidates without chronic liver disease. For those transplanted with hepatitis C NAT+ lungs, we initiated antiviral therapy in the outpatient setting and continued treatment for 12 weeks. In a retrospective single-center study of 15 lung transplant recipients receiving hepatitis C NAT+ lungs and 88 recipients receiving nonviremic lungs, we tested the hypothesis that deferral of antiviral therapy after transplantation of lungs from hepatitis C NAT+ donors to the outpatient setting would result in similar 1-year survival compared to transplantation of lungs from nonviremic donors. Patients receiving hepatitis C NAT+ lungs had similar baseline characteristics but had longer index hospital lengths of stay (24 vs 13 days, p = 0.021). Patients receiving hepatitis C NAT+ lungs had fewer episodes of acute cellular rejection in the first year. Patients receiving hepatitis C NAT+ lungs had similar 1-year survival to patients receiving lungs from nonviremic donors, after controlling for age and lung allocation score (p = 0.638). In this small single-center study, outpatient initiation of antiviral therapy for donor-derived hepatitis C is associated with acceptable clinical outcomes and can be considered in patients with high severity of illness.

Purpose: Treatment of triple-negative breast cancer (TNBC) remains challenging. Intermittent fasting (IF) has emerged as a promising approach to improve metabolic health of various metabolic disorders. Clinical studies indicate IF is essential for TNBC progression. However, the molecular mechanisms underlying metabolic remodeling in regulating IF and TNBC progression are still unclear. Methods: In this study, we utilized a robust mouse model of TNBC and exposed subjects to a high-fat diet (HFD) with IF to explore its impact on the metabolic reprogramming linked to cancer progression. To identify crucial serum metabolites and signaling events, we utilized targeted metabolomics and RNA sequencing (RNA-seq). Furthermore, we conducted immunoblotting, real-time quantitative polymerase chain reaction (RT-qPCR), cell migration assays, lentivirus-mediated Mmp9 overexpression, and Mmp9 inhibitor experiments to elucidate the role of decanoylcarnitine/Mmp9 in TNBC cell migration. Results: Our observations indicate that IF exerts notable inhibitory effects on both the proliferation and cancer metastasis. Utilizing targeted metabolomics and RNA-seq, we initially identified pivotal serum metabolites and signaling events in the progression of TNBC. Among the 349 serum metabolites identified, decanoylcarnitine was picked out to inhibit TNBC cell proliferation and migration. RNA-seq analysis of TNBC cells treated with decanoylcarnitine revealed its suppressive effects on extracellular matrix-related protein components, with a notable reduction observed in Mmp9. Further investigations confirmed that decanoylcarnitine could inhibit Mmp9 expression in TNBC cells, primary tumors, lung, and liver metastasis tissues. Mmp9 overexpression abolished the inhibitory effect of decanoylcarnitine on cell migration. Conclusion: This study pioneers the exploration of IF intervention and the role of decanoylcarnitine/Mmp9 in the progression of TNBC in obese mice, enhancing our comprehension of the potential roles of various dietary patterns in the process of cancer treatment.

This study carried out a research on the distribution characteristics of the geostress field of ultra-deep strike-slip faults, aiming to improve the exploration efficiency and development benefits of ultra-deep fault-controlled reservoirs. This study combines multiple geological information, drilling information, seismic data, and seismic attribute analysis. Taking the FI12 fault zone in the Fengman Oilfield as an example, it carried out numerical simulation of the geostress field, clarified the distribution pattern of geostress, analyzed the effectiveness of fracture mechanics, and proposed a reservoir quality evaluation method and production increase strategy based on geostress analysis. The study includes the following three aspects: 1) research on the heterogeneity of fault-controlled fractured carbonate reservoirs: Due to the existence of faults, fractures, and pores, carbonate reservoirs exhibit strong heterogeneity. These discontinuous structures lead to local stress field decreases to varying degrees. By analyzing the changes in geostress, it is possible to infer the development of faults, fractures, and pores. This provides important basis for predicting and evaluating reservoirs. 2) research on the distribution pattern of geostress: The geostress of fractured bodies exhibits a “shell-type” distribution pattern. Inside the fractured body, the geostress values are lower, indicating that this part is a favorable reservoir body. On the outside of the fractured body, the geostress exhibits high-value concentration, and this part has relatively poor permeability and can be regarded as an unfavorable drilling target. 3) research on the relationship between fault-fracture mechanical activity and reservoir quality and production capacity: Geostress and its influence on fault-fracture mechanical activity are directly related to the quality and production capacity of fault-controlled fractured carbonate reservoirs. When deploying well locations and optimizing well trajectories, geostress factors should be fully considered, and reservoir reformation efficiency should be taken into account to promote single well production increases and reservoir economic devel.

The effective operation of intangible assets in commercial sports events can bring long-term, sustainable, and large-scale economic benefits to sports events. However, currently, in the operation of commercial sports events, the lack of experience in hosting events often hinders the development of intangible assets that should have created higher profits, and cannot achieve the expected value. The intangible assets operational management performance (IAOMP) evaluation of commercial sporting events is the multiple-attribute group decision-making (MAGDM). The Logarithmic TODIM (LogTODIM) and TOPSIS technique was brought forward the MAGDM. The interval-valued intuitionistic fuzzy sets (IVIFSs) are brought forward as the useful technique for coping with uncertain and fuzzy information during the IAOMP evaluation of commercial sporting events. In this study, the interval-valued intuitionistic fuzzy number Logarithmic TODIM-TOPSIS (IVIFN-LogTODIM-TOPSIS) technique is brought forward the MAGDM under IVIFSs circumstances. Finally, the numerical example for IAOMP evaluation of commercial sporting events is brought forward to verify the IVIFN-LogTODIM-TOPSIS technique. The main contribution of this study is brought forward: (1) the LogTODIM-TOPSIS was extended to IVIFSs in light with MEREC model; (2) the MEREC model is brought forward to derive weight in light with score information values under IVIFSs circumstances. (3) the IVIFN-LogTODIM-TOPSIS is brought forward for MAGDM under IVIFSs circumstances; (4) the numerical example for IAOMP evaluation of commercial sporting events and several different comparative analysis is brought forward to verify the IVIFN-LogTODIM-TOPSIS model.

Bi2NiMnO6 (BNMO) epitaxial thin films with a layered supercell (LSC) structure have emerged as a promising single‐phase multiferroic material recently. Because of the required strain state for the formation of the LSC structures, most of the previous BNMO films are demonstrated on rigid oxide substrates such as SrTiO3 and LaAlO3. Here, the potential of BNMO films grown on muscovite mica substrates via van der Waals epitaxy, spotlighting their suitability for cutting‐edge flexible device applications is delved. Comprehensive scanning transmission electron microscopy/energy‐dispersive X‐ray analyses reveal a layered structure in the BNMO film and a pristine interface with the mica substrate, indicating high‐quality deposition and minimal interfacial defects. Capitalizing on its unique property of easily cleavable layers due to weak van der Waals forces in mica substrates, flexible BNMO/mica samples are fixed. A standout feature of the BNMO film grown on mica substrate is its consistent multiferroic properties across varied mechanical conditions. A novel technique is introduced for thinning the mica substrate and subsequent transfer of the sample, with post‐transfer analyses validating the preserved structural and magnetic attributes of the film. Overall, this study illuminates the resilient multiferroic properties of BNMO films on mica, offering promising avenues for their integration for next‐generation flexible electronics.

ObjectiveThis study aimed to assess the risk factors for symptomatic osteonecrosis (ON) in systemic lupus erythematosus (SLE) and identify clinical characteristics and laboratory markers for predicting symptomatic ON occurrence in SLE patients.MethodsSeventy (6.0%) of 1175 SLE patients diagnosed with symptomatic ON were included in this study. An equal number of SLE patients without symptomatic ON, matched in terms of age and gender, were enrolled in the control group. Clinical symptoms, routine laboratory examinations, lymphocyte subsets, and treatments of these patients were retrospectively reviewed and compared between the two groups. Logistic regression analysis was employed to identify risk factors associated with symptomatic ON in SLE.ResultsAmong the 70 cases in the symptomatic ON group, 62 (88.6%) patients experienced femoral head necrosis, with bilateral involvement observed in 58 patients. Bone pain was reported in 32 cases (51.6%), and 19 cases (30.6%) presented with multiple symptoms. Univariate analysis revealed significant differences between the two groups in various factors, including disease duration (months), cumulative steroid exposure time, history of thrombosis, neurological involvement, the number of affected organs, myalgia/myasthenia, and the use of medications such as glucocorticoids, immunosuppressants, aspirin, and statins (P

Abstract Keratins are an integral part of cell structure and function. Here, it is shown that ectopic expression of a truncated isoform of keratin 81 (tKRT81) in breast cancer is upregulated in metastatic lesions compared to primary tumors and patient‐derived circulating tumor cells, and is associated with more aggressive subtypes. tKRT81 physically interacts with keratin 18 (KRT18) and leads to changes in the cytosolic keratin intermediate filament network and desmosomal plaque formation. These structural changes are associated with a softer, more elastically deformable cancer cell with enhanced adhesion and clustering ability leading to greater in vivo lung metastatic burden. This work describes a novel biomechanical mechanism by which tKRT81 promotes metastasis, highlighting the importance of the biophysical characteristics of tumor cells.

221. Photoactive Poly-L-Lysine gel with resveratrol-magnesium metal polyphenol network: A promising strategy for preventing tracheal anastomotic complications following surgery

Postoperative complications at the anastomosis site following tracheal resection are a prevalent and substantial concern. However, most existing solutions primarily focus on managing symptoms, with limited attention given to proactively preventing the underlying pathological processes. To address this challenge, we conducted a drug screening focusing on clinically-relevant polyphenolic compounds, given the growing interest in polyphenolic compounds for their potential role in tissue repair during wound healing. This screening led to the identification of resveratrol as the most promising candidate for mitigating tracheal complications, as it exhibited the most significant efficacy in enhancing the expression of vascular endothelial growth factor (VEGF) while concurrently suppressing the pivotal fibrosis factor: transforming growth factor-beta 1 (TGF-β1), showcasing its robust potential in addressing these issues. Building upon this discovery, we further developed an innovative photosensitive poly-L-lysine gel integrated with a resveratrol-magnesium metal polyphenol network (MPN), named Res-Mg/PL-MA. This design allows for the enables sustained release of resveratrol and synergistically enhances the expression of VEGF and also promotes resistance to tensile forces, aided by magnesium ions, in an anastomotic tracheal fistula animal models. Moreover, the combination of resveratrol and poly-L-lysine hydrogel effectively inhibits bacteria, reduces local expression of key inflammatory factors, and induces polarization of macrophages toward an anti-inflammatory phenotype, as well as inhibits TGF-β1, consequently decreasing collagen production levels in an animal model of post-tracheal resection. In summary, our novel Res-Mg/PL-MA hydrogel, through antibacterial, anti-inflammatory, and pro-vascularization mechanisms, effectively prevents complications at tracheal anastomosis, offering significant promise for translational applications in patients undergoing tracheal surgeries.

Abstract The therapeutic effects of pharmaceuticals depend on their drug concentrations in the cochlea. Efficient drug delivery from the systemic circulation into the inner ear is limited by the blood‐labyrinth‐barrier (BLB). This study investigated a novel noninvasive sound conditioning (SC) strategy (90 dB SPL, 8–16 kHz, 2 h sound exposure) to temporally enhance BLB permeability in a controllable way, contributing to maximizing the penetration of pharmaceuticals from blood circulation into the cochlea. Trafficking of Fluorescein Isothiocyanate conjugated dextran and bovine serum albumin (FITC‐dextran and FITC‐BSA) demonstrated that paracellular leakage of BLB sustained for 6 h after SC, providing a controllable time window for systemic administration. Cochlear concentrations of dexamethasone (DEX) and dexamethasone phosphate (DEX‐P), respectively transported by transcellular and paracellular pathways, showed a higher content of the latter one after SC, further confirming the key role of paracellular pathway in the SC‐induced hyperpermeability. Results of high‐throughput RNA‐sequencing identified a series of tight junction (TJ)‐associated genes after SC. The expressions of TJ (ZO‐1) were reduced and irregular rearrangements of the junction were observed by transmission electron microscopy after SC. We further determined the inhibiting role of Rab13 in the recruitment of ZO‐1 and later in the regulation of cellular permeability. Meanwhile, no significant change in the quantifications of endothelial caveolae vesicles after SC indicated that cellular transcytosis accounted little for the temporary hyperpermeability after SC. Based on these results, SC enhances the BLB permeability within 6 h and allows systemically applied drugs which tend to be transported by paracellular pathway to readily enter the inner ear, contributing to guiding the clinical medications on hearing loss.

Both Bactrocera minax and Bactrocera dorsalis are phytophagous insects, and their larvae are latent feeders, which cause great damage and economic losses to agriculture production and trade. This study aimed to provide a scientific reference for researching and developing the feasible countermeasures against these two pests. Based on the distribution data of B. minax and B. dorsalis in China, obtained from the Chinese herbaria, investigation and literature. Four niche models (Garp, Bioclim, Domain, and Maxent) were used to analyze the key environmental factors affecting the distribution of both pests and to build prediction models of the potential distribution in Sichuan Basin. Combined with two statistical standards, area under the receiver operating characteristic curve (AUC) and Kappa, the validity of prediction models were analyzed and compared. The results show that: the average AUC values of the four models are all above 0.90, and the average Kappa values are all above 0.75, indicating that the four models are suitable for predicting the potential distribution area of B. minax and B. dorsalis. The annual range of temperature, the mean temperature in the driest quarter, the mean temperature in the warmest quarter, the annual precipitation, and the precipitation in driest month are the key environmental factors affecting the distribution of B. minax, while the mean diurnal temperature range, the mean temperature in the driest quarter, the seasonal temperature variations and the precipitation in driest month affect the potential distribution of B. dorsalis. The suitable areas for B. minax are mainly concentrated in the eastern of Sichuan Basin, while the suitable areas for B. dorsalis are concentrated in the southeastern. Except for the Bioclim model, the highly-suitable area for both pests predicted by the other three models are all greater than 15.94 × 104 km2 and the moderately-suitable areas are greater than 13.57 × 104 km2. In conclusion, the suitable areas for both pests in Sichuan Basin are quite wide. Therefore, the relevant authorities should be given strengthened monitoring of both pests, especially in areas with high incursion rates.

224. Failure modes and mechanical properties of double-layer rock-like composite specimens with a single fissure under triaxial compression

Introduction: Geotechnical engineering disasters often result from instability failures in layered and heterogeneous fissured rock masses. However, the key mechanisms governing mechanical properties and crack propagation in these rock masses remain unclear.Methods: This study presents triaxial compression tests on double-layer rock-like specimens composed of limestone and sandstone materials, containing a single fissure, to investigate the effects of fissure angles and positions on the strength and failure modes of these double-layer specimens under varying confining pressure.Results and Discussion: The experimental results reveal that the intact composite rock approaches the strength of sandstone but is deformation-limited by limestone. Under constant confining pressure (σ3 = 5 MPa), the fissure angle affects initial crack initiation, and fissure position dictates the failure mode and extent, while increased confining pressure induces overall shear failure in the composite rock, with the failure mode being predominantly influenced by confining pressure. Concerning mechanical deformation, augmenting the fissure angle and confining pressure substantially enhances the elasticity and ductility of the composite rock. Regarding volumetric deformation, the extent of volume shrinkage in the composite rock is influenced by both fissure angle and confining pressure, while volume expansion is influenced by fissure position. Under uniaxial compression, fissured composite rock exhibits the most unstable crack propagation, resulting in early failure. Triaxial compression shows that a higher fissure angle stabilizes crack propagation while confining pressure variation affects stability only when the fissure is in limestone. When the fissure is in sandstone, crack propagation stability remains at its highest. Furthermore, an increase in fissure angle, higher confining pressure, and changes in fissure position from sandstone through the contact interface to limestone contribute to an increasing trend in the peak strength and elastic modulus of the composite rock. Fissure-induced rock degradation is primarily influenced by the fissure angle. These findings are significant for guiding engineering construction and design, providing valuable insights to geotechnical engineers, and enhancing safety in rock engineering projects.

Objective. During in vitro fertilization-embryo transfer (IVF-ET) treatment, the reproductive endocrine regulatory mechanisms hold pivotal importance. Specifically, the serum estradiol (E2) level during ovulation emerges as a critical factor influencing pregnancy outcomes. This retrospective study aimed to comprehensively compare two common clinical regimens based on the grouping of serum E2 levels and the number of oocytes retrieved on the trigger day. Our objective was to evaluate the pregnancy outcomes in IVF-ET patients across different ovarian response groups, exploring the efficacy of the dual-trigger and single-trigger regimens to provide valuable insights for optimizing clinical strategies in the context of IVF-ET. Methods. A retrospective analysis was conducted on the clinical data of 2778 infertile patients who underwent ART (IVF/ICSI). Subsequently, a detailed statistical analysis was performed on 1032 patients following an antagonist regimen. Participants were categorized into single-trigger and dual-trigger groups based on real-world trigger protocols, considering different ovarian responses. Comprehensive statistical assessments were conducted on baseline characteristics, ovulation induction, and pregnancy outcomes. Results. Baseline characteristics and cycle parameters among the three patient groups (high ovarian response, normal response, and poor response) exhibited no significant differences between the dual-trigger and single-trigger regimen groups. Despite the dual-trigger regimen utilizing a significantly lower HCG dose, no notable discrepancies were observed in laboratory results and pregnancy outcomes (embryo transfer rate, pregnancy rate, and live birth rate) for normal and high responders. Remarkably, E2 levels were higher in the dual-trigger group compared to the single-trigger group. In high and normal responders, the dual-trigger regimen demonstrated increased oocyte counts and oocyte acquisition rates, coupled with decreased transfer cancellation rates attributed to ovarian hyperstimulation syndrome (OHSS). Intriguingly, patients with a poor ovarian response experienced no graft cancellations due to OHSS prevention in either group. Conclusion. For patients with high and normal ovarian responses, the utilization of a dual-trigger regimen on the trigger day effectively mitigates the risk of OHSS. Our large sample study supports the substitutability of the dual-trigger regimen over the single-trigger regimen without compromising pregnancy outcomes. However, this conclusion is not applicable to patients with poor ovarian responses. The results of this study highlight the necessity of adopting a customized and individualized treatment approach that should be based on the patient’s ovarian response. Additionally, recognizing the pivotal role of the endocrine environment in influencing pregnancy outcomes and the occurrence of OHSS, further exploration of the effects of different triggering regimens on endocrine parameters is warranted. Such investigations will contribute to enhancing the reproductive outcomes of IVF-ET technology.

226. Spatial differences and underlying mechanisms in electronic word of mouth in the foodservice industry: A case of Sanya, China.

Studying the electronic word-of-mouth (eWOM) in the foodservice industry can not only provide guidance for merchants, but also spatially optimize the urban foodservice industry, restaurants' location selection, and customers' purchasing decisions. In this study, taking Sanya city as the research object, using big data crawling technology to collect the directory and their attribute information of 2107 restaurants with more than 100 reviews. Kernel density analysis, grid analysis and the geographically weighted regression (GWR) model were applied to reveal the distribution characteristics and influencing factors of eWOM in the foodservice industry in Sanya, China. The main results are as follows. The foodservice industry in Sanya extends along the southern coastline and is characterized by little dispersion and agglomeration at the macro level. The overall eWOM score of the foodservice industry is low. Market popularity, restaurant rating, transportation conditions, and commercial development all have a positive impact on the eWOM of the foodservice industry. Population and price have both positive and negative effects and the public services has a nonsignificant impact on the eWOM. This study not only improves the theoretical understanding of the foodservice industry, but also provides a general reference for its development in other industries and cities.

227. JWST’s First Glimpse of a z > 2 Forming Cluster Reveals a Top-heavy Stellar Mass Function

Clusters and their progenitors (protoclusters) at z ∼ 2 − 4, the peak epoch of star formation, are ideal laboratories to study the formation process of both the clusters themselves and their member galaxies. However, a complete census of their member galaxies has been challenging due to observational difficulties. Here we present new JWST/NIRCam observations targeting the distant cluster CLJ1001 at z = 2.51 from the COSMOS-Web program, which, in combination with previous narrowband imaging targeting H α emitters and deep millimeter surveys of CO emitters, provide a complete view of massive galaxy assembly in CLJ1001. In particular, JWST reveals a population of massive, extremely red cluster members in the long-wavelength bands that were invisible in previous Hubble Space Telescope (HST)/F160W imaging (HST-dark members). Based on this highly complete spectroscopic sample of member galaxies, we show that the spatial distribution of galaxies in CLJ1001 exhibits a strong central concentration, with the central galaxy density already resembling that of low- z clusters. Moreover, we reveal a “top-heavy” stellar mass function for the star-forming galaxies (SFGs), with an overabundance of massive SFGs piled up in the cluster core. These features strongly suggest that CLJ1001 is caught in a rapid transition, with many of its massive SFGs likely soon becoming quiescent. In the context of cluster formation, these findings suggest that the earliest clusters form from the inside out and top to bottom, with the massive galaxies in the core assembling first, followed by the less massive ones in the outskirts.

Objective. Osteosarcoma (OS) represents a prevalent primary bone neoplasm predominantly affecting the pediatric and adolescent populations, presenting a considerable challenge to human health. The objective of this investigation is to develop a prognostic model centered on anoikis-related genes (ARGs), with the aim of accurately forecasting the survival outcomes of individuals diagnosed with OS and offering insights into modulating the immune microenvironment. Methods. The study’s training cohort comprised 86 OS patients sourced from The Cancer Genome Atlas database, while the validation cohort consisted of 53 OS patients extracted from the Gene Expression Omnibus database. Differential analysis utilized the GSE33382 dataset, encompassing three normal samples and 84 OS samples. Subsequently, the study executed gene ontology and Kyoto encyclopedia of genes and genomes enrichment analyses. Identification of differentially expressed ARGs associated with OS prognosis was carried out through univariate COX regression analysis, followed by LASSO regression analysis to mitigate overfitting risks and construct a robust prognostic model. Model accuracy was assessed via risk curves, survival curves, receiver operating characteristic curves, independent prognostic analysis, principal component analysis, and t-distributed stochastic neighbor embedding (t-SNE) analysis. Additionally, a nomogram model was devised, exhibiting promising potential in predicting OS patient prognosis. Further investigations incorporated gene set enrichment analysis to delineate active pathways in high- and low-risk groups. Furthermore, the impact of the risk prognostic model on the immune microenvironment of OS was evaluated through tumor microenvironment analysis, single-sample gene set enrichment analysis (ssGSEA), and immune infiltration cell correlation analysis. Drug sensitivity analysis was conducted to identify potentially effective drugs for OS treatment. Ultimately, the verification of the implicated ARGs in the model construction was conducted through the utilization of real-time quantitative polymerase chain reaction (RT-qPCR). Results. The ARGs risk prognostic model was developed, comprising seven high-risk ARGs (CBS, MYC, MMP3, CD36, SCD, COL13A1, and HSP90B1) and four low-risk ARGs (VASH1, TNFRSF1A, PIP5K1C, and CTNNBIP1). This prognostic model demonstrates a robust capability in predicting overall survival among patients. Analysis of immune correlations revealed that the high-risk group exhibited lower immune scores compared to the low-risk group within our prognostic model. Specifically, CD8+ T cells, neutrophils, and tumor-infiltrating lymphocytes were notably downregulated in the high-risk group, alongside significant downregulation of checkpoint and T cell coinhibition mechanisms. Additionally, three immune checkpoint-related genes (CD200R1, HAVCR2, and LAIR1) displayed significant differences between the high- and low-risk groups. The utilization of a nomogram model demonstrated significant efficacy in prognosticating the outcomes of OS patients. Furthermore, tumor metastasis emerged as an independent prognostic factor, suggesting a potential association between ARGs and OS metastasis. Notably, our study identified eight drugs—Bortezomib, Midostaurin, CHIR.99021, JNK.Inhibitor.VIII, Lenalidomide, Sunitinib, GDC0941, and GW.441756—as exhibiting sensitivity toward OS. The RT-qPCR findings indicate diminished expression levels of CBS, MYC, MMP3, and PIP5K1C within the context of OS. Conversely, elevated expression levels were observed for CD36, SCD, COL13A1, HSP90B1, VASH1, and CTNNBIP1 in OS. Conclusion. The outcomes of this investigation present an opportunity to predict the survival outcomes among individuals diagnosed with OS. Furthermore, these findings hold promise for progressing research endeavors focused on prognostic evaluation and therapeutic interventions pertaining to this particular ailment.

230. Retrospective Analysis of the Efficacy and Safety of Ultrasound-Guided Radiofrequency Ablation in the Treatment of Papillary Thyroid Microcarcinoma: A Follow-Up Study of Continuous Postoperative Surveillance and Large-Sample Data

Objective. To retrospectively evaluate the efficacy and safety of ultrasound-guided radiofrequency ablation (RFA) in the treatment of papillary thyroid microcarcinoma (PTMC) through a follow-up study of continuous postoperative surveillance and large-sample data. Methods. The efficacy of ultrasound-guided RFA was evaluated by measuring the tumor volume reduction rate (VRR), tumor disappearance rate, and disease progression in 358 patients with low-risk unifocal PTMC who underwent ultrasound-guided RFA at Hangzhou Weja Hospital, while the safety was evaluated by measuring their complications. Results. The VRR was −745.69 ± 1012.69 (%), −150.35 ± 395.5 (%), 46.47 ± 138.74 (%), 92.95 ± 27.88 (%), 97.78 ± 10.99 (%), and 99.27 ± 3.82 (%), respectively, at 1, 3, 6, 12, 18, and 24 months after RFA. The corresponding tumor disappearance rate was 1.68%, 9.78%, 43.85%, 82.68%, 92.59%, and 95.63%, respectively. No local recurrence, new tumors, lymph node metastasis, distant metastasis, and deaths caused by recurrent/metastatic PTMC were found at the last follow-up. Except for 3 cases (0.84%) with thickening or hoarseness of voice and 3 cases (0.84%) with coughing during drinking water, no other complications were found. Conclusions. Ultrasound-guided RFA has good efficacy and safety for the treatment of low-risk unifocal PTMC and can be promoted for use in patients who meet the required indications.

Background: Cystatin C, a low-molecular-weight protein, belongs to cysteine protease inhibitors produced primarily by nucleated cells. Its serum concentration, independent of sex, age, or muscle mass, is a good predictor of renal dysfunction in older adults. This study evaluated the association between all-cause mortality and preoperative cystatin C levels in hip fractures. Materials and methods: Data describing the demographic and clinical characteristics of the patients were gathered specifically from older individuals who had suffered hip fractures. The study used linear and non-linear multivariate Cox regression models to investigate the association between preoperative cystatin C levels and mortality. The analyses were conducted using the R and EmpowerStats software. Results: In total, two thousand three hundred and ninety-four patients were included in this study. A total of 790 patients (33 %) died of all causes. The mean follow-up was 37.62 months. The preoperative cystatin C was 0.91 ± 0.41 mg/L. Linear multivariate Cox regression analysis revealed a significant association between preoperative cystatin C level and death, with a hazard ratio (HR) of 2.19 (95 % confidence interval [CI]: 1.72–2.79, P

The most prevalent and devastating form of organ damage in systemic lupus erythematosus (SLE) is lupus nephritis (LN). LN is characterized by glomerular injury, inflammation, cell proliferation, and necrosis, leading to podocyte injury and tubular epithelial cell damage. Assays for urine biomarkers have demonstrated significant promise in the early detection of LN, evaluation of disease activity, and tracking of reaction to therapy. This article focuses on studies examining urinary biomarkers associated with podocyte injury in LN, offering fresh perspectives on the application of biomarkers in the early detection and management of LN.

Introduction: Ice plugs acts as isolate maintenance sections during pipeline repairs in nuclear power plants can effectively reduce maintenance costs, which is formed by the cooling of liquid nitrogen. The single-jacket freezing method takes a long time for the formation of ice plugs with a certain load-bearing capacity, increasing the liquid nitrogen consumption.Methods: In this study, a multiple-jacket method for the formation of ice plugs was proposed, and the numerical simulation using CFD (Computational Fluid Dynamics) was conducted. The effects of total jacket length and jacket number on freezing time and the consumption of liquid nitrogen were analyzed.Results and Discussion: Research indicated that the multiple-jacket method obtained a 11%∼15% decrease in freezing time and a 18%∼26% decrease in liquid nitrogen consumption than the single-jacket method. For the multiple-jacket method, the gap between the jackets presented a certain influence on the freezing time and liquid nitrogen consumption. As the gap between the jackets increases, there exist a small increase in the freezing time, but the liquid nitrogen consumption will be reduced by 2% to 5%.

Tumor-associated macrophages (TAMs) are critical in the tumor microenvironment (TME) of hepatocellular carcinoma (HCC). Major vault protein (MVP) mediates multidrug resistance, cell growth and development, and viral immunity. However, the relationship between MVP and TAMs polarization has not been clarified in HCC. We found that MVP significantly increased M2-TAMs infiltration levels in tumor tissues of HCC patients. MVP promoted HCC proliferation, metastasis, and invasion by regulating M2 polarization in vivo and in vitro. Mechanistically, MVP associated with signal transducer and activator of transcription 6 (STAT6) and enhanced STAT6 phosphorylation. STAT6 translocated from the cytosol to the nucleus and regulated M2 macrophage-associated gene transcription. These findings suggest that MVP modulates the macrophage M2 transcriptional program, revealing its potential role in the TAMs of TME.

The Monocyte chemoattractant protein-1 (MCP-1), also referred to as chemokine ligand 2 (CCL2), belongs to the extensive chemokine family and serves as a crucial mediator of innate immunity and tissue inflammation. It has a notable impact on inflammatory conditions affecting the kidneys. Upon binding to its receptor, MCP-1 can induce lymphocytes and NK cells’ homing, migration, activation, differentiation, and development while promoting monocytes’ and macrophages’ infiltration, thereby facilitating kidney disease-related inflammation. As a biomarker for kidney disease, MCP-1 has made notable advancements in primary kidney diseases such as crescentic glomerulonephritis, chronic glomerulonephritis, primary glomerulopathy, idiopathic proteinuria glomerulopathy, acute kidney injury; secondary kidney diseases like diabetic nephropathy and lupus nephritis; hereditary kidney diseases including autosomal dominant polycystic kidney disease and sickle cell kidney disease. MCP-1 not only predicts the occurrence, progression, prognosis of the disease but is also closely associated with the severity and stage of nephropathy. When renal tissue is stimulated or experiences significant damage, the expression of MCP-1 increases, demonstrating a direct correlation with the severity of renal injury.

ObjectiveThis study offers a bibliometric analysis of the current situation, hotspots, and cutting-edge domains of genetic factors of adolescent idiopathic scoliosis (AIS).MethodsAll publications related to genetic factors of AIS from January 1, 1992, to February 28, 2023, were searched from the Web of Science. CiteSpace software was employed for bibliometric analysis, collecting information about countries, institutions, authors, journals, and keywords of each article.ResultsA cumulative number of 308 articles have been ascertained. Since 2006, publications relating to genetic factors of AIS have significantly increased. China leads in both productivity and influence in this area, with the Chinese Academy of Medical Sciences being the most productive institution. The most prolific scholars in this field are Y. Qiu and Z. Z. Zhu. The publications that contributed the most were from Spine and European Spine Journal. The most prominent keywords in the genetic factors of AIS were “fibrillin gene”, “menarche”, “calmodulin”, “estrogen receptor gene”, “linkage analysis”, “disc degeneration”, “bone mineral density”, “melatonin signaling dysfunction”, “collagen gene”, “mesenchymal stem cell”, “LBX1”, “promoter polymorphism”, “Bone formation”, “cerebrospinal fluid flow” and “extracellular matrix”.ConclusionThis analysis provides the frontiers and trends of genetic factors in AIS, including relevant research, partners, institutions and countries.

Copper-doped Zinc Tellurium (ZnTe:Cu) films were deposited on borosilicate glass using magnetron co-sputtering technique. The influence of the substrate temperature on the structural, morphological, optical and electrical properties of ZnTe:Cu films was investigated by X-ray diffraction (XRD), Raman spectroscopy, X-ray photoelectron spectroscopy (XPS), atomic force microscopy (AFM), UV–Vis spectrophotometer and Hall effect measurement system. The results indicate that substrate temperature significantly affects the properties of the ZnTe:Cu films. When the substrate temperature increases from room temperature to 600 °C, the (111)-preferred orientation of ZnTe:Cu films is gradually replaced by the (220)-preferred orientation. At high substrate temperatures (≥500 °C), the CuxTe phase appears in the ZnTe:Cu films, resulting in higher carrier concentration (>1019 cm−3) and lower resistivity (
