Background Studies have shown racial differences in cancer outcomes. We investigate whether survival differences existed in Hispanic patients with glioblastoma (GBM) compared with other ethnicities from our modern radiotherapy series, because no study to date has focused on outcomes in this group after radiation therapy. Methods We retrospectively evaluated 428 patients diagnosed with GBM from 1996 to 2014 at our institution, divided into 4 groups based on self-report: white, black, Hispanic, and Asian/Indian. The primary outcome was overall survival. We analyzed differences in prognostic factors among the whole cohort compared with the Hispanic cohort alone. Results Baseline characteristics of the 4 racial groups were comparable. With a median follow-up of 387 days, no survival differences were seen by Kaplan-Meier analysis. Median overall survival for Hispanic patients was 355 days versus 450 days for the entire cohort. Factors significant for patient outcomes in the entire cohort differed slightly from those specific to Hispanic patients. Low Karnofsky Performance Status was significant on multivariate analysis in the whole population, but not in Hispanic patients. Extent of resection, recursive partitioning analysis class, and radiation therapy total dose were significant on multivariate analysis in both the whole population and Hispanic patients. Conclusions We found that Hispanic patients with GBM had no difference in survival compared with other ethnicities in our cohort. Differences exist in factors associated with outcomes on single and multivariate analysis for Hispanic patients with GBM compared with the entire cohort. Additional studies focusing on Hispanic patients will aid in more personalized treatment approaches in this group.

We detect the kinematic Sunyaev-Zel'dovich (kSZ) effect with a statistical significance of $4.2 \sigma$ by combining a cluster catalogue derived from the first year data of the Dark Energy Survey (DES) with CMB temperature maps from the South Pole Telescope Sunyaev-Zel'dovich (SPT-SZ) Survey. This measurement is performed with a differential statistic that isolates the pairwise kSZ signal, providing the first detection of the large-scale, pairwise motion of clusters using redshifts derived from photometric data. By fitting the pairwise kSZ signal to a theoretical template we measure the average central optical depth of the cluster sample, $\bar{\tau}_e = (3.75 \pm 0.89)\cdot 10^{-3}$. We compare the extracted signal to realistic simulations and find good agreement with respect to the signal-to-noise, the constraint on $\bar{\tau}_e$, and the corresponding gas fraction. High-precision measurements of the pairwise kSZ signal with future data will be able to place constraints on the baryonic physics of galaxy clusters, and could be used to probe gravity on scales $ \gtrsim 100$ Mpc., Comment: 23 pages, 14 figures; matches version published in MNRAS

The Dark Energy Survey (DES) is a next generation optical survey aimed at measuring the expansion history of the universe using four probes: weak gravitational lensing, galaxy cluster counts, baryon acoustic oscillations, and Type Ia supernovae. To perform the survey, the DES Collaboration is building the Dark Energy Camera (DECam), a 3 square degree, 570 Megapixel CCD camera which will be mounted at the Blanco 4-meter telescope at the Cerro Tololo Inter- American Observatory. DES will survey 5000 square degrees of the southern galactic cap in 5 filters (g, r, i, z, Y). DECam will be comprised of 74 250 micron thick fully depleted CCDs: 62 2k x 4k CCDs for imaging and 12 2k x 2k CCDs for guiding and focus. Construction of DECam is nearing completion. In order to verify that the camera meets technical specifications for DES and to reduce the time required to commission the instrument, we have constructed a full sized telescope simulator and performed full system testing and integration prior to shipping. To complete this comprehensive test phase we have simulated a DES observing run in which we have collected 4 nights worth of data. We present the first results of a survey for high redshift, z $\ge$ 6, quasars using izY multi-colour photometric observations from the Dark Energy Survey (DES). Here we report the discovery and spectroscopic confirmation of the $\rm z_{AB}, Y_{AB}$ = 20.2, 20.2 (M$_{1450}$ = $-$26.5) quasar DES J0454$-$4448 with an emission line redshift of z = 6.10$\pm$0.03 and a HI near zone size of 4.6 $\pm$ 1.7 Mpc.The quasar was selected as an i-band drop out with i$-$z = 2.46 and z$_{AB} < 21.5$ from an area of $\rm \sim$300 deg$^2$. It is the brightest of our 43 candidates and was identified for follow-up spectroscopically solely based on the DES i$-$z and z$-$Y colours. The quasar is detected by WISE and has $W1_{AB} = 19.68$. The discovery of one spectroscopically confirmed quasar with 5.7 $$ 50-100 new quasars with z $>$ 6 including 3-10 with z $>$ 7 dramatically increasing the numbers of quasars currently known that are suitable for detailed studies including determination of the neutral HI fraction of the intergalactic medium (IGM) during the epoch of Hydrogen reionization., Comment: 9 pages, 13 figures, this is a pre-copyedited, author-produced version of an article accepted for publication in MNRAS following peer review

Knowledge of the scatter in the mass-observable relation is a key ingredient for a cosmological analysis based on galaxy clusters in a photometric survey. In this paper we aim to quantify the capability of the correlation function of galaxy cluster to constrain the intrinsic scatter $\sigma_{\ln M}$. We demonstrate how the linear bias measured in the correlation function of clusters can be used to determine the value of this parameter. The new method is tested in simulations of a $5, 000 deg^2$ optical survey up to $z \sim 1$, similar to the ongoing Dark Energy Survey (DES). Our results show that our method works better at lower scatter values. We can measured the intrinsic scatter $\sigma_{ln M} = 0.1$ with a standard deviation of $\sigma(\sigma_{ln M}) \sim 0.03$ using this technique. However, the expected intrinsic scatter of the DES RedMaPPer cluster catalog $\sigma_{ln M} \sim 0.2$ cannot be recovered with this method at suitable accuracy and precision because the area coverage is insufficient. For future photometric surveys with a larger area such as LSST and Euclid, the statistical errors will be reduced. Therefore, we forecast higher precision to measure the intrinsic scatter including the value mention before. We conclude that this method can be used as an internal consistency check method on their simplifying assumptions and complementary to cross-calibration techniques in multi-wavelength cluster observations, Comment: Accepted for publication on ApJ

BACKGROUND: Adjuvant hypofractionated radiotherapy (HRT) for elderly patients with newly diagnosed glioblastoma (GBM) is reasonable compared to standard fractionation radiotherapy (SRT). Comparison of HRT in the presence of temozolomide (TMZ) vs SRT with TMZ is unclear. We examined HRT for GBM with TMZ in comparison to SRT with TMZ. METHODS: Retrospective analysis of patients > 60 years-old who received SRT or HRT of newly diagnosed GBM in the post-operative setting from 1994 to 2014. Inclusion criteria included SRT (60 Gy/30 fractions or 59.4 Gy/33 fractions) vs HRT of 40 Gy/15 fractions. RESULTS: 158 patients received SRT vs 26 with HRT. Median survival receiving SRT versus HRT was 430 and 475 days (p = 0.550). 95% of the SRT patients received TMZ vs 100% with HRT. Patients receiving HRT were older (median: 72 vs 66 years). All HRT patients were treated with IMRT technique vs SRT. Multivariate Cox regression showed decreased survival in age > 70 (Hazard ratio [HR] 1.84; 95% confidence interval [CI] 1.23-2.75), lower Karnofsky performance status (HR 5.25; 95% CI 1.73-15.99), biopsy vs. surgical resection (HR 4.18; 95% CI 1.49-11.77), RT planning technique 3D- or 2D-planning vs. IMRT (HR 1.91; 95% CI 1.32-2.76 and HR 3.40; 95% CI 1.60-7.22, respectively). IMRT based planning showed no difference between HRT vs SRT. For patients receiving TMZ, there was no survival difference between HRT and SRT. CONCLUSIONS: Elderly GBM patients receiving HRT or SRT had similar overall survival. Subset analysis patients receiving concurrent TMZ showed no difference in survival between HRT and SRT.

AMG 386 is an investigational first-in-class peptide-Fc fusion protein (peptibody) that inhibits angiogenesis by preventing the interaction of angiopoietin-1 (Ang1) and Ang2 with their receptor, Tie2. Although the therapeutic value of blocking Ang2 has been shown in several models of tumorigenesis and angiogenesis, the potential benefit of Ang1 antagonism is less clear. To investigate the consequences of Ang1 neutralization, we have developed potent and selective peptibodies that inhibit the interaction between Ang1 and its receptor, Tie2. Although selective Ang1 antagonism has no independent effect in models of angiogenesis-associated diseases (cancer and diabetic retinopathy), it induces ovarian atrophy in normal juvenile rats and inhibits ovarian follicular angiogenesis in a hormone-induced ovulation model. Surprisingly, the activity of Ang1 inhibitors seems to be unmasked in some disease models when combined with Ang2 inhibitors, even in the context of concurrent vascular endothelial growth factor inhibition. Dual inhibition of Ang1 and Ang2 using AMG 386 or a combination of Ang1- and Ang2-selective peptibodies cooperatively suppresses tumor xenograft growth and ovarian follicular angiogenesis; however, Ang1 inhibition fails to augment the suppressive effect of Ang2 inhibition on tumor endothelial cell proliferation, corneal angiogenesis, and oxygen-induced retinal angiogenesis. In no case was Ang1 inhibition shown to (a) confer superior activity to Ang2 inhibition or dual Ang1/2 inhibition or (b) antagonize the efficacy of Ang2 inhibition. These results imply that Ang1 plays a context-dependent role in promoting postnatal angiogenesis and that dual Ang1/2 inhibition is superior to selective Ang2 inhibition for suppression of angiogenesis in some postnatal settings. We have previously shown N-arylnaphthamides can be potent inhibitors of vascular endothelial growth factor receptors (VEGFRs). N-Alkyl and N-unsubstituted naphthamides were prepared and found to yield nanomolar inhibitors of VEGFR-2 (KDR) with an improved selectivity profile against a panel of tyrosine and serine/threonine kinases. The inhibitory activity of this series was retained at the cellular level. Naphthamides 3, 20, and 22 exhibited good pharmacokinetics following oral dosing and showed potent inhibition of VEGF-induced angiogenesis in the rat corneal model. Once-daily oral administration of 22 for 14 days led to 85% inhibition of established HT29 colon cancer and Calu-6 lung cancer xenografts at doses of 10 and 20 mg/kg, respectively.

Angiogenesis is vital for solid tumor growth, and its prevention is a proven strategy for the treatment of disease states such as cancer. The vascular endothelial growth factor (VEGF) pathway provides several opportunities by which small molecules can act as inhibitors of endothelial proliferation and migration. Critical to these processes is signaling through VEGFR-2 or the kinase insert domain receptor (KDR) upon stimulation by its ligand VEGF. Herein, we report the discovery of 2,3-dihydro-1,4-benzoxazines as inhibitors of intrinsic KDR activity (IC 50 < 0.1 microM) and human umbilical vein endothelial cell (HUVEC) proliferation with IC 50 < 0.1 microM. More specifically, compound 16 was identified as a potent (KDR: < 1 nM and HUVEC: 4 nM) and selective inhibitor that exhibited efficacy in angiogenic in vivo models. In addition, this series of molecules is typically well-absorbed orally, further demonstrating the 2,3-dihydro-1,4-benzoxazine moiety as a promising platform for generating kinase-based antiangiogenic therapeutic agents.

Inhibition of the VEGF signaling pathway has become a valuable approach in the treatment of cancers. Guided by X-ray crystallography and molecular modeling, a series of 2-aminobenzimidazoles and 2-aminobenzoxazoles were identified as potent inhibitors of VEGFR-2 (KDR) in both enzymatic and HUVEC cellular proliferation assays. In this report we describe the synthesis and structure-activity relationship of a series of 2-aminobenzimidazoles and benzoxazoles, culminating in the identification of benzoxazole 22 as a potent and selective VEGFR-2 inhibitor displaying a good pharmacokinetic profile. Compound 22 demonstrated efficacy in both the murine matrigel model for vascular permeability (79% inhibition observed at 100 mg/kg) and the rat corneal angiogenesis model (ED(50) = 16.3 mg/kg).

The Dark Energy Camera is a new imager with a 2.2-degree diameter field of view mounted at the prime focus of the Victor M. Blanco 4-meter telescope on Cerro Tololo near La Serena, Chile. The camera was designed and constructed by the Dark Energy Survey Collaboration, and meets or exceeds the stringent requirements designed for the wide-field and supernova surveys for which the collaboration uses it. The camera consists of a five element optical corrector, seven filters, a shutter with a 60 cm aperture, and a CCD focal plane of 250 micron thick fully-depleted CCDs cooled inside a vacuum Dewar. The 570 Mpixel focal plane comprises 62 2kx4k CCDs for imaging and 12 2kx2k CCDs for guiding and focus. The CCDs have 15 microns x15 microns pixels with a plate scale of 0.263 arc sec per pixel. A hexapod system provides state-of-the-art focus and alignment capability. The camera is read out in 20 seconds with 6-9 electrons readout noise. Intermittent, normobaric hypoxia confers robust cardioprotection against ischemia-induced myocardial infarction and lethal ventricular arrhythmias. δ-Opioid receptor (DOR) signaling and reactive oxygen species (ROS) have been implicated in cardioprotective phenomena, but their roles in intermittent hypoxia are unknown. This study examined the contributions of DOR and ROS in mediating intermittent hypoxia-induced cardioprotection. Mongrel dogs completed a 20 day program consisting of 5-8 daily, 5-10 min cycles of moderate, normobaric hypoxia (FIO2 0.095-0.10), with intervening 4 min room air exposures. Subsets of dogs received the DOR antagonist naltrindole (200 μg/kg, sc) or antioxidant N-acetylcysteine (250 mg/kg, po) before each hypoxia session. Twenty-four hours after the last session, the left anterior descending coronary artery was occluded for 60 min and then reperfused for 5 h. Arrhythmias detected by electrocardiography were scored according to the Lambeth II conventions. Left ventricles were sectioned and stained with 2,3,5-triphenyl-tetrazolium-chloride, and infarct sizes were expressed as percentages of the area at risk (IS/AAR). Intermittent hypoxia sharply decreased IS/AAR from 41 ± 5 % (n = 12) to 1.8 ± 0.9 % (n = 9; P < 0.001) and arrhythmia score from 4.1 ± 0.3 to 0.7 ± 0.2 (P < 0.001) vs. non-hypoxic controls. Naltrindole (n = 6) abrogated the cardioprotection with IS/AAR 35 ± 5 % and arrhythmia score 3.7 ± 0.7 (P < 0.001 vs. untreated intermittent hypoxia). N-acetylcysteine (n = 6) interfered to a similar degree, with IS/AAR 42 ± 3 % and arrhythmia score 4.7 ± 0.3 (P < 0.001 vs. untreated intermittent hypoxia). Without the intervening reoxygenations, hypoxia (n = 4) was not cardioprotective (IS/AAR 50 ± 8 %; arrhythmia score 4.5 ± 0.5; P < 0.001 vs. intermittent hypoxia). Thus DOR, ROS and cyclic reoxygenation were obligatory participants in the gradually evolving cardioprotection produced by intermittent hypoxia.

The standard of care for patients with newly diagnosed glioblastoma (GBM) is maximal safe resection followed by adjuvant radiation therapy (RT) and temozolomide (TMZ). To investigate whether the timing of adjuvant RT after surgery affected outcome in patients with GBM. We retrospectively reviewed all patients with a diagnosis of GBM at our institution. A total of 447 patients were included in our analysis. Patients were divided into 3 equal groups based on the interval between surgery and RT. The primary outcome was overall survival (OS). Patients who began RT less than 21 days after surgery tended to be older, have a lower a Karnofsky Performance Status score, and higher recursive partitioning analysis class. These patients were more likely to have undergone biopsy only and received 3-dimensional conformal RT or 2-dimensional RT. The median OS for patients who started RT less than 21 days after surgery, between 21 and 32 days after surgery, and more than 32 days after surgery was 374, 465, and 478 days, respectively (P = .004). On multivariate Cox regression analysis, Karnofsky Performance Status score lower than 70, undergoing biopsy only, recursive partitioning analysis classes IV and V/VI, use of less than 36 Gy RT, and lack of TMZ chemotherapy were predictors of worse OS. The interval between surgery and RT was not significantly associated with OS on multivariate analysis. Patients who begin RT less than 21 days after surgery tend to have worse prognostic factors than those who begin RT later. When accounting for significant covariates, the effect of timing between surgery and RT is not significant. In this article a stochastic model for point events expected in the output image of scientific CCDs is derived. This kind of events are typical in CCD systems dedicated to particle detection, such as DAMIC and CONNIE. The model includes the randomness of the charge movement in the silicon and the noise at the readout stage providing the base for an optimal detection technique.

Weak gravitational lensing allows one to reconstruct the spatial distribution of the projected mass density across the sky. These "mass maps" provide a powerful tool for studying cosmology as they probe both luminous and dark matter. In this paper, we present a weak lensing mass map reconstructed from shear measurements in a 139 deg2 area from the Dark Energy Survey (DES) Science Verification (SV) data overlapping with the South Pole Telescope survey. We compare the distribution of mass with that of the foreground distribution of galaxies and clusters. The overdensities in the reconstructed map correlate well with the distribution of optically detected clusters. Cross-correlating the mass map with the foreground galaxies from the same DES SV data gives results consistent with mock catalogs that include the primary sources of statistical uncertainties in the galaxy, lensing, and photo-z catalogs. The statistical significance of the cross-correlation is at the 6.8 sigma level with 20 arcminute smoothing. A major goal of this study is to investigate systematic effects arising from a variety of sources, including PSF and photo-z uncertainties. We make maps derived from twenty variables that may characterize systematics and find the principal components. We find that the contribution of systematics to the lensing mass maps is generally within measurement uncertainties. We test and validate our results with mock catalogs from N-body simulations. In this work, we analyze less than 3% of the final area that will be mapped by the DES; the tools and analysis techniques developed in this paper can be applied to forthcoming larger datasets from the survey.

We present Magellan/M2FS, VLT/GIRAFFE, and Gemini South/GMOS spectroscopy of the newly discovered Milky Way satellite Reticulum II. Based on the spectra of 25 Ret II member stars selected from Dark Energy Survey imaging, we measure a mean heliocentric velocity of 62.8 +/- 0.5 km/s and a velocity dispersion of 3.3 +/- 0.7 km/s. The mass-to-light ratio of Ret II within its half-light radius is 470 +/- 210 Msun/Lsun, demonstrating that it is a strongly dark matter-dominated system. Despite its spatial proximity to the Magellanic Clouds, the radial velocity of Ret II differs from that of the LMC and SMC by 199 and 83 km/s, respectively, suggesting that it is not gravitationally bound to the Magellanic system. The likely member stars of Ret II span 1.3 dex in metallicity, with a dispersion of 0.28 +/- 0.09 dex, and we identify several extremely metal-poor stars with [Fe/H] < -3. In combination with its luminosity, size, and ellipticity, these results confirm that Ret II is an ultra-faint dwarf galaxy. With a mean metallicity of [Fe/H] = -2.65 +/- 0.07, Ret II matches Segue~1 as the most metal-poor galaxy known. Although Ret II is the third-closest dwarf galaxy to the Milky Way, the line-of-sight integral of the dark matter density squared is log J = 18.8 +/- 0.6 Gev^2/cm^5 within 0.2 degrees, indicating that the predicted gamma-ray flux from dark matter annihilation in Ret II is lower than that of several other dwarf galaxies., 14 pages, 6 figures (4 in color), 2 tables. The Dark Energy Survey (DES) is currently undertaking an observational program imaging $1/4$ of the southern hemisphere sky with unprecedented photometric accuracy. In the process of observing millions of faint stars and galaxies to constrain the parameters of the dark energy equation of state, the DES will obtain pre-discovery images of the regions surrounding an estimated 100 gamma-ray bursts (GRBs) over five years. Once GRBs are detected by, e.g., the Swift satellite, the DES data will be extremely useful for follow-up observations by the transient astronomy community. We describe a recently-commissioned suite of software that listens continuously for automated notices of GRB activity, collates useful information from archival DES data, and disseminates relevant data products back to the community in near-real-time. Of particular importance are the opportunities that non-public DES data provide for relative photometry of the optical counterparts of GRBs, as well as for identifying key characteristics (e.g., photometric redshifts) of potential GRB host galaxies. We provide the functional details of the DESAlert software as it presently operates, as well as the data products that it produces, and we show sample results from the application of DESAlert to several previously-detected GRBs., 14 pages, 6 figures, 2 tables. In this article the feasibility of using charge coupled devices (CCD) to detect low-energy neutrinos through their coherent scattering with nuclei is analyzed. The detection of neutrinos through this standard model process has been elusive because of the small energy deposited in such interaction. Typical particle detectors have thresholds of a few keV, and most of the energy deposition expected from coherent scattering is well below this level. The CCD detectors discussed in this paper can operate at a threshold of approximately 30 eV, making them ideal for observing this signal. On a CCD array of 500 g located next to a power nuclear reactor the number of coherent scattering events expected is about 3000 events/year. Our results shows that a detection with a confidence level of 99% can be reached within 16 days of continuous operation; with the current 52 g detector prototype this time lapse extends to five months. We present measurements of radioactive contamination in the high-resistivity silicon charge-coupled devices (CCDs) used by the DAMIC experiment to search for dark matter particles. Novel analysis methods, which exploit the unique spatial resolution of CCDs, were developed to identify $\alpha$ and $\beta$ particles. Uranium and thorium contamination in the CCD bulk was measured through $\alpha$ spectroscopy, with an upper limit on the $^{238}$U ($^{232}$Th) decay rate of 5 (15) kg$^{-1}$ d$^{-1}$ at 95% CL. We also searched for pairs of spatially correlated electron tracks separated in time by up to tens of days, as expected from $^{32}$Si-$^{32}$P or $^{210}$Pb-$^{210}$Bi sequences of $\beta$ decays. The decay rate of $^{32}$Si was found to be $80^{+110}_{-65}$ kg$^{-1}$ d$^{-1}$ (95% CI). An upper limit of $\sim$35 kg$^{-1}$ d$^{-1}$ (95% CL) on the $^{210}$Pb decay rate was obtained independently by $\alpha$ spectroscopy and the $\beta$ decay sequence search. The Dark Energy Camera has captured a large set of images as part of Science Verification (SV) for the Dark Energy Survey. The SV footprint covers a lar ge portion of the outer Large Magellanic Cloud (LMC), providing photometry 1.5 magnitudes fainter than the main sequence turn-off of the oldest LMC stel lar population. We derive geometrical and structural parameters for various stellar populations in the LMC disk. For the distribution of all LMC stars, we find an inclination of $i=-38.14^{\circ}\pm0.08^{\circ}$ (near side in the North) and a position angle for the line of nodes of $��_0=129.51^{\circ}\pm0.17^{\circ}$. We find that stars younger than $\sim 4$ Gyr are more centrally concentrated than older stars. Fitting a projected exponential disk shows that the scale radius of the old populations is $R_{>4 Gyr}=1.41\pm0.01$ kpc, while the younger population has $R_{, 18 pages, 13 figures, 3 tables; accepted for publication in MNRAS

In order to detect Dark Matter (DM), scientists in the Dark Matter In CCDs (DAMIC) Collaboration have set an experimental array of Charge-Coupled Devices (CCDs) in a nickel mine underground, and have developed all analysis tools to discern any known trace of conventional matter from what they expect to find in case a DM particle crosses the CCDs. In order to calibrate the signals from the CCDs, they have also designed experiments to quantify neutron-silicon interactions, assuming that neutrons can mimic DM interactions in the CCDs. Here we present preliminary results from the analysis of data obtained in these experiments.

We measure the weak-lensing masses and galaxy distributions of four massive galaxy clusters observed during the Science Verification phase of the Dark Energy Survey. This pathfinder study is meant to 1) validate the DECam imager for the task of measuring weak-lensing shapes, and 2) utilize DECam's large field of view to map out the clusters and their environments over 90 arcmin. We conduct a series of rigorous tests on astrometry, photometry, image quality, PSF modeling, and shear measurement accuracy to single out flaws in the data and also to identify the optimal data processing steps and parameters. We find Science Verification data from DECam to be suitable for the lensing analysis described in this paper. The PSF is generally well-behaved, but the modeling is rendered difficult by a flux-dependent PSF width and ellipticity. We employ photometric redshifts to distinguish between foreground and background galaxies, and a red-sequence cluster finder to provide cluster richness estimates and cluster-galaxy distributions. By fitting NFW profiles to the clusters in this study, we determine weak-lensing masses that are in agreement with previous work. For Abell 3261, we provide the first estimates of redshift, weak-lensing mass, and richness. In addition, the cluster-galaxy distributions indicate the presence of filamentary structures attached to 1E 0657-56 and RXC J2248.7-4431, stretching out as far as 1 degree (approximately 20 Mpc), showcasing the potential of DECam and DES for detailed studies of degree-scale features on the sky., Comment: accepted by MNRAS; high-resolution versions of figures can be downloaded from https://www.physics.ohio-state.edu/~melchior.12/research/des_sv_clusters.html

The growth of solid tumors is dependent on the continued stimulation of endothelial cell proliferation and migration resulting in angiogenesis. The angiogenic process is controlled by a variety of factors of which the vascular endothelial growth factor (VEGF) pathway and its receptors play a pivotal role. Small-molecule inhibitors of VEGF receptors (VEGFR) have been shown to inhibit angiogenesis and tumor growth in preclinical models and in clinical trials. A novel nicotinamide, AMG 706, was identified as a potent, orally bioavailable inhibitor of the VEGFR1/Flt1, VEGFR2/kinase domain receptor/Flk-1, VEGFR3/Flt4, platelet-derived growth factor receptor, and Kit receptors in preclinical models. AMG 706 inhibited human endothelial cell proliferation induced by VEGF, but not by basic fibroblast growth factor in vitro, as well as vascular permeability induced by VEGF in mice. Oral administration of AMG 706 potently inhibited VEGF-induced angiogenesis in the rat corneal model and induced regression of established A431 xenografts. AMG 706 was well tolerated and had no significant effects on body weight or on the general health of the animals. Histologic analysis of tumor xenografts from AMG 706–treated animals revealed an increase in endothelial apoptosis and a reduction in blood vessel area that preceded an increase in tumor cell apoptosis. In summary, AMG 706 is an orally bioavailable, well-tolerated multikinase inhibitor that is presently under clinical investigation for the treatment of human malignancies. We present the measurement of R = B(t -> Wb)/B(t -> Wq) in pp collisions at root s = 1.96 TeV, using 230 pb(-1) of data collected by the DO experiment at the Fermilab Tevatron Collider. We fit simultaneously R and the number (N-tt) of selected top quark pairs (tt), to the number of identified b-quark jets in events with one electron or one muon, three or more jets, and high transverse energy imbalance. To improve sensitivity, kinematical properties of events with no identified b-quark jets are included in the fit. We measure R = 1.03(-0.17)(0.19) (stat + syst), in good agreement with the standard model. We set lower limits of R > 0.61 and vertical bar V-tb vertical bar > 0.78 at 95% confidence level.

The results of a search for squarks and gluinos using data from p p ¯ collisions recorded at a center-of-mass energy of 1.96 TeV by the DO detector at the Fermilab Tevatron Collider are reported. The topologies analyzed consist of acoplanar-jet and multijet events with large missing transverse energy. No evidence for the production of squarks or gluinos was found in a data sample of 310 pb−1. Lower limits of 325 and 241 GeV were derived at the 95% C.L. on the squark and gluino masses, respectively, within the framework of minimal supergravity with tan β = 3 , A 0 = 0 , and μ 0 .

We report on a search for the pair production of second generation scalar leptoquarks (LQ_2) in ppbar collisions at the center-of-mass energy sqrt(s) = 1.96 TeV, using data corresponding to an integrated luminosity of 294 +- 19 pb^-1 recorded with the D0 detector. No evidence for a leptoquark signal in the LQ_2 LQ_2 -> muq muq channel has been observed, and upper bounds on the product of cross section times branching fraction were set. This yields lower mass limits of m(LQ_2) > 247 GeV for beta = Br(LQ_2->muq) = 1 and m(LQ_2) > 182 GeV for beta = 1/2. Combining these limits with previous D0 results, the lower limits on the mass of a second generation scalar leptoquark are m(LQ_2) > 251 GeV and m(LQ_2) > 204 GeV for beta = 1 and beta = 1/2, respectively.

We present a measurement of the top quark pair ($t\bar{t}$) production cross section ($\sigma_{t\bar{t}}$) in $p\bar{p}$ collisions at $\sqrt{s}=1.96$ TeV using 230 pb$^{-1}$ of data collected by the D0 experiment at the Fermilab Tevatron Collider. We select events with one charged lepton (electron or muon), missing transverse energy, and jets in the final state. We employ lifetime-based b-jet identification techniques to further enhance the $t\bar{t}$ purity of the selected sample. For a top quark mass of 175 GeV, we measure $\sigma_{t\bar{t}}=8.6^{+1.6}_{-1.5}(stat.+syst.)\pm 0.6(lumi.)$ pb, in agreement with the standard model expectation

We present a search for electroweak production of single top quarks in the s-channel and t-channel using neural networks for signal-background separation. We have analyzed 230 pb(-1) of data collected with the DO detector at the Fermilab Tevatron Collider at a center-of-mass energy of 1.96 TeV and find no evidence for a single top quark signal. Objective To assess the capacities of the cytokine inhibitors interleukin-1 receptor antagonist (IL-1Ra; anakinra) and PEGylated soluble tumor necrosis factor receptor I (PEG sTNFRI; pegsunercept) to suppress neovascularization. Methods A corneal angiogenesis assay was performed by implanting nylon discs impregnated with an angiogenic stimulator (basic fibroblast growth factor or vascular endothelial growth factor) into one cornea of female Sprague-Dawley rats. Animals were treated with IL-1Ra or PEG sTNFRI for 7 days, after which new vessels were quantified. In a parallel study, male Lewis rats with mycobacteria-induced adjuvant-induced arthritis were treated with IL-1Ra or PEG sTNFRI for 7 days beginning at disease onset, after which scores for inflammation and bone erosion as well as capillary counts were acquired from sections of arthritic hind paws. Results Treatment with IL-1Ra yielded a dose-dependent reduction in growth factor–induced corneal angiogenesis, while PEG sTNFRI did not. IL-1Ra, but not PEG sTNFRI, significantly reduced the number of capillaries in arthritic paws, even though both anticytokines reduced inflammation and bone erosion to a similar degree. Conclusion These data support a major role for IL-1, but not TNFα, in angiogenesis and suggest that an additional antiarthritic mechanism afforded by IL-1 inhibitors, but not anti-TNF agents, is the suppression of the angiogenic component of pannus.

Talimogene laherparepvec is an oncolytic immunotherapy based on a modified herpes simplex virus type 1 (HSV-1) designed to kill cancer cells through two mechanisms: a) direct viral-mediated lysis and b) stimulation of a tumor antigen-specific adaptive immune response. We developed a mouse model of metastatic melanoma using the B16F10 cell line to study local and systemic responses following treatment with talimogene laherparepvec alone or in combination with CTLA-4 blockade. OncoVEXmGM-CSF, an HSV-1 modified similarly to talimogene laherparepvec (murine GM-CSF is expressed instead of human GM-CSF) was used in these studies. B16F10 cells have been reported to be resistant to HSV-1 infection due to a lack of requisite entry receptors. To overcome resistance, we transduced B16F10 cells with a lentiviral vector expressing mouse nectin1 (mNectin1) or eGFP as a control. Expression was confirmed by ddPCR and sensitivity to OncoVEXmGM-CSF evaluated in an in vitro viability assay. B16F10-mNectin1 cells were highly sensitive to OncoVEXmGM-CSF with a multiplicity of infection (MOI) IC50 of 0.001 (B16F10-eGFP were insensitive at MOI of 100). In vivo, B16F10-mNectin1 and B16F10-eGFP cells showed similar growth when injected subcutaneously. OncoVEXmGM-CSF treatment of B16F10-mNectin1 tumors (intratumoral, 5x106 PFU/dose, 3X) caused a significant (p In conclusion, OncoVEXmGM-CSF in combination with CTLA-4 blockade significantly reduced systemic tumor burden and prolonged median overall survival in a B16F10 mouse model of metastatic melanoma. This article describes the design features and the first test measurements obtained during the installation of a novel high resolution 2D neutron detection technique. The technique proposed in this work consists of a boron layer (enriched in 10B) placed on a scientific Charge Coupled Device (CCD). After the nuclear reaction 10B(n,α)7Li, the CCD detects the emitted charge particles thus obtaining information on the neutron absorption position. The above-mentioned ionizing particles, with energies in the range 0.5-5.5MeV, produce a plasma effect in the CCD which is recorded as a circular spot. This characteristic circular shape, as well as the relationship observed between the spot diameter and the charge collected, is used for the event recognition, allowing the discrimination of undesirable gamma events. We present the first results recently obtained with this technique, which has the potential to perform neutron tomography investigations with a spatial resolution better than that previously achieved. Numerical simulations indicate that the spatial resolution of this technique will be about 15 μm, and the intrinsic detection efficiency for thermal neutrons will be about 3%. The Dark Energy Survey (DES) is a next generation optical survey aimed at understanding the accelerating expansion of the universe using four complementary methods: weak gravitational lensing, galaxy cluster counts, baryon acoustic oscillations, and Type Ia supernovae. To perform the 5000 sq-degree wide field and 30 sq-degree supernova surveys, the DES Collaboration built the Dark Energy Camera (DECam), a 3 square-degree, 570-Megapixel CCD camera that was installed at the prime focus of the Blanco 4-meter telescope at the Cerro Tololo Inter-American Observatory (CTIO). DES started its first observing season on August 31, 2013 and observed for 105 nights through mid-February 2014. This paper describes DES “Year 1” (Y1), the strategy and goals for the first year's data, provides an outline of the operations procedures, lists the efficiency of survey operations and the causes of lost observing time, provides details about the quality of the first year's data, and hints at the “Year 2” plan and outlook.

We introduce the fully-depleted charge-coupled device (CCD) as a particle detector. We demonstrate its low energy threshold operation, capable of detecting ionizing energy depositions in a single pixel down to 50 eVee. We present results of energy calibrations from 0.3 keVee to 60 keVee, showing that the CCD is a fully active detector with uniform energy response throughout the silicon target, good resolution (Fano ~0.16), and remarkable linear response to electron energy depositions. We show the capability of the CCD to localize the depth of particle interactions within the silicon target. We discuss the mode of operation and unique imaging capabilities of the CCD, and how they may be exploited to characterize and suppress backgrounds. We present the first results from the deployment of 250 um thick CCDs in SNOLAB, a prototype for the upcoming DAMIC100. We present results from a study of the photometric redshift performance of the Dark Energy Survey (DES), using the early data from a Science Verification (SV) period of observations in late 2012 and early 2013 that provided science-quality images for almost 200 sq.~deg.~at the nominal depth of the survey. We assess the photometric redshift performance using about 15000 galaxies with spectroscopic redshifts available from other surveys. These galaxies are used, in different configurations, as a calibration sample, and photo-$z$'s are obtained and studied using most of the existing photo-$z$ codes. A weighting method in a multi-dimensional color-magnitude space is applied to the spectroscopic sample in order to evaluate the photo-$z$ performance with sets that mimic the full DES photometric sample, which is on average significantly deeper than the calibration sample due to the limited depth of spectroscopic surveys. Empirical photo-$z$ methods using, for instance, Artificial Neural Networks or Random Forests, yield the best performance in the tests, achieving core photo-$z$ resolutions $\sigma_{68} \sim 0.08$. Moreover, the results from most of the codes, including template fitting methods, comfortably meet the DES requirements on photo-$z$ performance, therefore, providing an excellent precedent for future DES data sets., Comment: Published in MNRAS. The Dark Energy Camera, the Imager and its cooling system was installed onto the Blanco 4m telescope at the Cerro Tololo Inter-American Observatory in Chile in September 2012. The imager cooling system is a LN2 two-phase closed loop cryogenic cooling system. The cryogenic circulation processing is located off the telescope. Liquid nitrogen vacuum jacketed transfer lines are run up the outside of the telescope truss tubes to the imager inside the prime focus cage. The design of the cooling system along with commissioning experiences and initial cooling system performance is described. The LN2 cooling system with the DES imager was initially operated at Fermilab for testing, then shipped and tested in the Blanco Coude room. Now the imager is operating inside the prime focus cage. It is shown that the cooling performance sufficiently cools the imager in a closed loop mode, which can operate for extended time periods without maintenance or LN2 fills.

Talimogene laherparepvec, is a modified oncolytic herpes simplex virus type-1 (HSV-1) designed to selectively replicate in tumors and to initiate a systemic immune response to target cancer cells. Intralesional administration of talimogene laherparepvec is intended to result in oncolysis within injected tumors, with lytic cell destruction promoting the local release of progeny virus and tumor derived antigens. GM-CSF, a product of the viral transgene, is also produced locally and is designed to recruit and stimulate antigen presenting cells to further enhance systemic antitumor immune response. While available evidence suggests that talimogene laherparepvec can induce effects in distant tumors via an immune-mediated effect, additional mechanistic evidence is being sought to help better understand the putative systemic effect. For preclinical mechanism of action studies we employed OncoVEXmGM-CSF, an HSV-1 virus modified in the same manner as talimogene laherparepvec except that murine GM-CSF is expressed. We utilized this virus to further dissect the underlying innate and adaptive immunity following viral administration in syngeneic tumor models. Intratumoral administration of OncoVEXmGM-CSF into established murine tumors induced regressions in the injected lesion but also resulted in significant anti-tumor effects in contralateral (uninjected) lesions. We have previously demonstrated by immunohistochemistry that the systemic effects observed in the contralateral lesions are not driven by systemic spread of the virus, but instead correlate with the infiltration of CD3+ T cell populations. Here we report the full phenotypic characterization of immune infiltrates by flow cytometry in both the injected and contralateral lesions. In a time-course fashion, this characterization revealed the initial infiltration of the injected tumor by innate effector cells and a concomitant induction of a potent type I interferon response. This in turn drives the local release of proinflammatory cytokines and chemokines, resulting in the recruitment of adaptive immune subsets to the injected tumor and enhancing systemic anti-tumor reactivity. Additionally, both the primary oncolytic replication of the virus and the activation of type I interferon pathway are directly correlated with STING activity. In conclusion, in vitro and in vivo observations suggest that intratumoral injection of OncoVEXmGM-CSF activates multiple immune-mediated mechanisms of action leading to T-cell activation and induction of anti-tumor immunity in mice. Background: Talimogene laherparepvec is an oncolytic immunotherapy based on a modified herpes simplex virus type 1 (HSV-1), designed to kill neoplastic cells through two mechanisms: a) direct lysis and b) stimulation of a tumor antigen-specific adaptive immune response by way of GM-CSF expression. In order to support mechanism of action studies in a mouse melanoma model relevant to talimogene laherparepvec's approved U.S. indication, we expressed the HSV-1 entry receptor Nectin1 on B16F10 melanoma cells. B16F10 cells have previously been shown to be resistant to HSV-1 due to lack of entry receptor expression (Miller et al., Mol Ther, 2001). The goal of our work was to determine if mNectin1 expression in B16F10 melanoma cells renders them sensitive to talimogene laherparepvec treatment. For preclinical studies we used OncoVEXmGM-CSF, an HSV-1 modified similarly to talimogene laherparepvec except that murine GM-CSF is expressed instead of human GM-CSF. Herein we describe studies to assess the efficacy of OncoVEXmGM-CSF in a novel B16F10-mNectin1 syngeneic melanoma model. Methods: Parental B16F10 cells were transduced with lentivirus expressing either mNectin1, or eGFP as a control. In vitro sensitivity was determined by plating both cell types in 96-well plates, infecting with serially diluted OncoVEXmGM-CSF and then measuring ATP 72 hours later. For efficacy studies, B16F10 or B16F10-mNectin1 cells were injected subcutaneously on the right flank of female C57BL/6 mice. On day 10, mice were randomized into treatment groups based on tumor volume (n = 10/group). Talimogene laherparepvec was delivered intratumorally (5×106 PFU/dose,) every 3 days during the first week. Tumor growth and body weight were measured twice per week throughout the experiment. Survival analysis was performed using a Kaplan-Meier estimator. Results: B16F10 cells were insensitive to OncoVEXmGM-CSF up to 100 MOI (multiplicity of infection). In contrast, two other syngeneic cell lines, A20 and CT-26, showed MOI IC50 of 1 and 20. B16F10 cells expressing mNectin1 showed high sensitivity to OncoVEXmGM-CSF with an MOI IC50 of 0.01. In contrast, B16F10 cells expressing eGFP were insensitive up to an MOI of 100. In vivo, B16F10 mNectin1 cells showed a similar growth pattern as the parental B16F10 when injected subcutaneously. We report the discovery of an excess of main sequence turn-off stars in the direction of the constellations of Eridanus and Phoenix from the first year data of the Dark Energy Survey (DES). The Eridanus-Phoenix (EriPhe) overdensity is centered around l~285 deg and b~-60 deg and spans at least 30 deg in longitude and 10 deg in latitude. The Poisson significance of the detection is at least 9 sigma. The stellar population in the overdense region is similar in brightness and color to that of the nearby globular cluster NGC 1261, indicating that the heliocentric distance of EriPhe is about d~16 kpc. The extent of EriPhe in projection is therefore at least ~4 kpc by ~3 kpc. On the sky, this overdensity is located between NGC 1261 and a new stellar stream discovered by DES at a similar heliocentric distance, the so-called Phoenix Stream. Given their similar distance and proximity to each other, it is possible that these three structures may be kinematically associated. Alternatively, the EriPhe overdensity is morphologically similar to the Virgo overdensity and the Hercules-Aquila cloud, which also lie at a similar Galactocentric distance. These three overdensities lie along a polar plane separated by ~120 deg and may share a common origin. Here we report on the results of our theoretical study of 1H-MoS2 clusters grown on graphene oxide. First, we studied graphene cluster, graphene oxide (1,2) and (1,3) ether-like monolayers with a Stone-Wales defect. Secondly, a 1H-MoS2 nanoparticle was grown over the defect creating a sandwich like structure. For all considered structures energy bands as well as total and partial density of states (DOS) were calculated in order to inquire about their electronic behaviour. It has been found that for graphene oxide (1,2) ether-like with 1H-MoS2cluster located over the defect and either terminating as Mo- or S-edges, it behaves like a semi metal, while for graphene oxide (1,3) ether-like it yields semiconductor behaviour, respectively. Our main concern was the sandwich, in which total and partial DOS indicate a strong hybridisation of bands formed by C p-, S p-, Mo d- especially for graphene oxide (1,3) Mo- or S-edges.

Scientific CCD detectors are typically readout using the Correlated Double Sampling (CDS) technique. At low pixel rates, noise of ~2e- RMS is typically achieved. The limitation for reaching lower noise comes from the 1/f component on the output of the CCD, and this noise cannot be eliminated using CDS. A new readout technique based on a digital filter is presented here for suppressing the 1/f. Using this new technique a noise of 0.4e- is achieved.

The Dark Energy Survey CCD imager was constructed at the Fermi National Accelerator Laboratory and delivered to the Cerro Tololo Inter-American Observatory in Chile for installation onto the Blanco 4m telescope. Several efforts are described relating to preparation of the instrument for transport, development and testing of a shipping crate designed to minimize transportation loads transmitted to the camera, and inspection of the imager upon arrival at the observatory. Transportation loads were monitored and are described. For installation of the imager at the telescope prime focus, where it mates with its previously-installed optical corrector, specialized tooling was developed to safely lift, support, and position the vessel. The installation and removal processes were tested on the Telescope Simulator mockup at FNAL, thus minimizing technical and schedule risk for the work performed at CTIO. Final installation of the imager is scheduled for August 2012.

The Dark Energy Camera and its cooling system has been shipped to Cerro Tololo Inter-American Observatory in Chile for installation onto the Blanco 4m telescope. Along with the camera, the cooling system has been installed in the Coude room at the Blanco Telescope. Final installation of the cooling system and operations on the telescope is planned for the middle of 2012. Initial commissioning experiences and cooling system performance is described.

Scientific CCDs designed in thick high resistivity silicon (Si) are excellent detectors for astronomy, high energy and nuclear physics, and instrumentation. Many applications can benefit from CCDs ultra low noise readout systems. The present work shows how sub electron noise CCD images can be achieved using digital signal processing techniques. These techniques allow 0. 4 electrons of noise at readout bandwidths of up to 10 Kpixels per second while keeping the full CCD spatial resolution and signal dynamic range. © 2012 Springer Science+Business Media B.V. The readout noise for Charge-Coupled Devices (CCDs) has been the main limitation when using these detectors for measuring small amplitude signals. A scientific CCD fabricated on a high-resistivity silicon substrate utilizing a floating gate amplifier with the capability of multiple sampling of the charge signal is described in this paper. The Skipper CCD architecture and its advantages for low noise applications are discussed. A technique for obtaining sub-electron readout noise levels is presented, and its noise and signal characteristics are derived. We demonstrate that with this procedure a very low readout noise of 0.2e − RMS can be achieved. The contribution of other noise sources (output stage, vertical and horizontal charge transfer inefficiency, and dark current noise) are also considered. The optimum number of samples for achieving the total lowest possible noise level is obtained. This technique is applied to an X-rays experiment using a 55Fe source. Fil: Fernández Moroni, Guillermo. Purpose To determine whether systemic treatment with AMG 386, a selective angiopoietin 1/2-neutralizing peptibody, inhibits neovascular processes in animal models of ocular disease. Methods AMG 386 was tested in a laser-induced choroidal neovascularization (CNV) model in monkeys using fluorescein angiography. The biodistribution of (125)I-AMG 386 was determined in cynomolgus monkeys by whole-body autoradiography and radioanalysis of ocular tissues. A murine retinopathy of prematurity (ROP) model was used to examine the effect of AMG 386 on established and newly formed retinal vessels, either as a single agent or when combined with VEGF inhibition.AMG 386 pharmacokinetics were evaluated in each model. Results In the CNV model, AMG 386 significantly decreased fluorescent angiographic leakage and reduced fibroplasia, indicating an impaired healing response consistent with angiogenesis blockade. Radiolabeled AMG 386 was widely distributed across ocular tissues, with highest concentrations in the choroid, cornea, retinal pigmented epithelium, iris/ciliary body, and sclera. In the ROP model, AMG 386 prevented pathologic retinal angiogenesis when administered from P8 to P16 but transiently impeded regression of these abnormal vessels when administered from P17 to P23. Combining AMG 386 with VEGF inhibition led to cooperative prevention of retinal angiogenesis in this model. No AMG 386-related ocular toxicities occurred, and no treatment-related clinical observations were made in any of the studies. Conclusions In this study, AMG 386 inhibited angiogenesis in animal models of CNV and ROP, supporting investigation of AMG 386 for the treatment of ocular neovascular diseases in the clinical setting.
