O universo transmídia do seriado True Blood (Tese, 2017).pdf: 2791143 bytes, checksum: 2c6efa12b3b0b48b290ae865ed0a5651 (MD5) CAPES Esta tese de doutorado almejou explorar, tanto teórica quanto metodologicamente, um fenômeno da comunicação contemporânea: os processos de transmidiação de seriados ficcionais no contexto da criação e do consumo no mercado televisivo estadunidense. Chamamos de processos de transmidiação as estratégias e práticas criativas que fazem um conteúdo monomidiático, o seriado televisivo no nosso estudo de caso, percorrer diversas mídias e plataformas comunicacionais. Trata-se da criação de artefatos que se vinculam a um texto de referência, fazendo-o migrar de mídias enquanto simultaneamente alarga seu escopo de significações. O nosso objetivo central foi o de analisar o que chamamos de universo transmídia de True Blood: os textos, paratextos e extensões ficcionais, alocados em múltiplas mídias e plataformas, que se utilizam de diversos formatos e linguagens, e produzidos tanto pelas organizações empresariais quanto pelas organizações dos fãs. Estas peças, arguimos, compõem o complexo emaranhado comunicacional que se encontra em circulação cultural e que se refere a uma obra específica, o seriado True Blood. A análise buscou explorar as estratégias de criação de extensões a partir de suas relações com o texto principal, sendo dividida conforme as duas macro-categorias de extensões transmídia (as paratextuais e as ficcionais) e examinando organicamente as criações oriundas de ambas as instâncias produtivas. As extensões paratextuais foram analisadas conforme as categorias de: antecipação, recuperação, remixagem e contexto de produção. As extensões ficcionais foram analisadas a partir das estratégias de alargamento do mundo ficcional do texto principal, que são: continuação de histórias, construção de mundo, construção de personagens, backstories (histórias do passado), histórias alternativas, e, por fim, jogos. Ao fim, foi possível identificar as semelhanças e diferenças dos produtos oriundos das empresas e dos fãs, compreendendo as finalidades estratégicas que elas cumprem no ínterim do universo transmídia de True Blood. This doctoral dissertation aimed to explore, both theoretically and methodologically, a phenomenon of contemporary communication: the transmedialisation processes around television fictions within the context of creation and consumption in the American television market. We refer to transmedialisation processes as the creative strategies and practices that make a single-media content – the television series in our case study – travel across various media and platforms. It refers to the creation of artifacts that link themselves to a reference text, making it migrate to another media while simultaneously broaden its scope of meanings. Our main goal was to analyze what we called the transmedia universe of True Blood: the texts, paratextos and fictional extensions, placed in multiple media and platforms, using various formats and languages, and being produced by both the media industry organizations and fans organizations. We argue that these pieces compose the complex communicational entanglement which is in broad cultural circulation and refers to an specific artwork, the television series True Blood. The analysis aimed to explore the creation strategies of transmedia extensions through their relations to the main text, divided by two major categories of extensions (paratextual and fictional) and examining organically the creations that came from both productive instances. The paratextual extensions are analyzed according the categories of: anticipation, recuperation, remixing, and production context. The fictional extensions are analyzed through their strategies of enlarging the storyworld of the main text, which are: continuation of stories, worldbuilding, characters construction, backstories, alternate stories, and games. By the end, it was possible to identify the similarities and differences of products originated from the industries and the fans, understanding the strategical purposes that they fulfill inside the transmedia universe of True Blood.

Para tal, foram realizadas revisões de literatura acerca dos processos de transmidiação, das especificidades da narrativa transmídia na televisão contemporânea, e das estratégias de serialidade na ficção seriada televisiva. Partimos do pressuposto que a narrativa transmídia representa, contemporaneamente, a escalada de um modo de entretenimento calcado na constante expansão das narrativas, na convergência midiática e no público participativo disposto a migrar entre mídias para consumir suas histórias favoritas. O corpus de análise foi composto pelas quatro primeiras temporadas do seriado televisivo (HBO, 2008-2011; atualmente na quinta temporada e sem previsão de término) e pelas extensões transmídia identificadas no decorrer da pesquisa. A análise foi realizada em torno de dois eixos: a) a análise da narrativa empreendida a partir da organização das tramas principais do seriado televisivo em associação com as extensões transmídia, que proporcionam a expansão da narrativa; b) a análise das funções desempenhadas pelas extensões transmídia, que operam estratégias de construção de mundo, de construção de personagens e de multiplicidade. A abordagem analítica empregada levou em consideração a organização narrativa complexa de episódios e temporadas do seriado televisivo e as estratégias de serialidade que caracterizam True Blood, observados, principalmente, nos ganchos e na continuidade absoluta das tramas e episódios. Os resultados obtidos indicaram que as extensões transmídia de True Blood ampliam o escopo de significações da narrativa principal – o seriado televisivo – de modo a compor um mundo ficcional cujas partes se encontram em plataformas de mídia distintas. As peças que compõem o universo transmídia de True Blood são: os episódios e temporadas do seriado televisivo, sites e blogs diversos, um blog de personagem, perfis em redes sociais, um canal de YouTube, mini-episódios online, depoimentos com a perspectiva de personagens, uma campanha viral com características de ARG (jogo de realidade alternativa), uma campanha de marketing viral, revistas em quadrinhos online e impressas, e uma coleção de romances.

Muitas telenovelas estão baseadas na estratégia maniqueísta de opor as forças do Bem e do Mal, gerando os conflitos que fazem com que o público torça pelas personagens, e alguns desses vilões entraram para a história da telenovela nacional e são lembrados até hoje por suas maldades. Nazaré Tedesco, a vilã principal de Senhora do Destino, destaca-se nesse cenário pela construção envolvente na disputa de Nazaré e Maria do Carmo, personagem principal da novela, pelo reconhecimento de Isabel/Lindalva como sua mãe. Foram definidos conceitos de construção de telenovela e de personagem, analisando a trajetória do autor roteirista Aguinaldo Silva com especial atenção para os seus vilões, para, então, voltar-se para a análise interna de Senhora do Destino. A partir dessa análise, foi possível destrinchar mais detalhadamente as estratégias a que Aguinaldo Silva recorre na construção de seus vilões e das quais lançou mão para criar Nazaré Tedesco, percebendo não só as recorrências em seus trabalhos, como sua busca por inovação.

Ao encontrar, na crÃtica à minissérie, uma tendência a considerá-la pouco próxima ao estilo de Eça de Queirós, este trabalho procurou questionar a posição dessa crÃtica, analisando o modo como foi possÃvel à equipe de realizadores a criação de estratégias para a composição de programas de efeitos que aproximassem a minissérie ao texto queirosiano, sem deixar de imprimir suas marcas no produto realizado. As bases conceituais que nortearam esta investigação estão fundamentadas na ideia da adaptação como tradução ou transposição entre sistemas semióticos, desenvolvida por Plaza; na ideia de dialogismo e intertextualidade em Bakhtin; e na teoria da adaptação proposta por Hutcheon. A teoria assinada por Bourdieu possibilita o entendimento e a análise do contexto de produção do campo literário e do televisivo para compreender como a perspectiva autoral de Maria Adelaide Amaral e de Luiz Fernando Carvalho colaborou em uma adaptação de Eça de Queirós para a televisão brasileira, caracterizada por uma perspectiva autoral dos adaptadores em que procuravam um resultado em que os traços do estilo de Eça pudessem ser reconhecidos. Os resultados da pesquisa demonstram que a adaptação operada por Maria Adelaide Amaral e Luiz Fernando Carvalho, autores da minissérie, apresentou aproximações com o estilo de Eça de Queirós no que se refere à condução da narrativa, ao modo como a história foi contada, ao cuidado com a música, à apresentação dos presságios e indÃcios. Neste caso, o objetivo dos adaptadores configurou-se em um grau elevado de aproximação, em que foram resguardados os traços do escritor português, atitude que colocou em risco a recepção da minissérie pelo público. O objeto de análise desta tese foi constituÃdo pelos 42 capÃtulos da minissérie Os Maias, exibida de terça a sexta-feira, pela Rede Globo de Televisão, de 09 de janeiro de 2001 a 23 de março de 2001, à s 23h. A articulação entre a matriz metodológica que orienta os trabalhos do Laboratório de Análise FÃlmica â a Poética do Filme e as proposições advindas da análise que examina as instâncias de produção, fruição e consumo de teleficção â orientada por Bourdieu â fomentaram a análise que observou os efeitos sensoriais, sentimentais e cognitivos que a obra pode produzir sobre o apreciador, nesse caso, a minissérie fruto da adaptação de romances para a televisão brasileira. No. of bitstreams: 1 AmandaAouadAlmeida.pdf: 12312077 bytes, checksum: 23f841a11fd7bd85aaea8508bd987469 (MD5) Este presente trabalho apresenta resultados de uma pesquisa sobre a estratégia de criação da narrativa policial na telenovela Força de um Desejo (Globo, 1999), de autoria de Gilberto Braga e Alcides Nogueira com direção geral de Mauro Mendonça Filho. Percebe-se que diversas telenovelas utilizam a estratégia da pergunta “quem matou?”, gerando curiosidade e audiência, porém, em sua maioria, a resposta frustra o público por revelar o nome do suspeito mais óbvio, ou de alguém que jamais poderia ser suspeito na visão do espectador. A telenovela Força de um Desejo, no entanto, destacava-se nesse cenário pelo cuidado na construção e resolução desse mistério, que possuía implicações desde o início da trama e, por isso, foi objeto de investigação. Foram definidos conceitos de construção de uma telenovela e de narrativas policiais, analisando a trajetória do roteirista / autor Gilberto Braga e suas oito tramas de assassinato, que geraram um mistério só revelado no último capítulo, para, enfim, debruçar-se sobre a telenovela Força de um Desejo com uma análise interna detalhada para comprovar a eficácia de sua construção. 218. Assessing the performance of AI chatbots in answering patients' common questions about low back pain.

- Abstract
Objectives: The aim of this study was to assess the accuracy and readability of the answers generated by large language model (LLM)-chatbots to common patient questions about low back pain (LBP)., Methods: This cross-sectional study analysed responses to 30 LBP-related questions, covering self-management, risk factors and treatment. The questions were developed by experienced clinicians and researchers and were piloted with a group of consumer representatives with lived experience of LBP. The inquiries were inputted in prompt form into ChatGPT 3.5, Bing, Bard (Gemini) and ChatGPT 4.0. Responses were evaluated in relation to their accuracy, readability and presence of disclaimers about health advice. The accuracy was assessed by comparing the recommendations generated with the main guidelines for LBP. The responses were analysed by two independent reviewers and classified as accurate, inaccurate or unclear. Readability was measured with the Flesch Reading Ease Score (FRES)., Results: Out of 120 responses yielding 1069 recommendations, 55.8% were accurate, 42.1% inaccurate and 1.9% unclear. Treatment and self-management domains showed the highest accuracy while risk factors had the most inaccuracies. Overall, LLM-chatbots provided answers that were 'reasonably difficult' to read, with a mean (SD) FRES score of 50.94 (3.06). Disclaimer about health advice was present around 70%-100% of the responses produced., Conclusions: The use of LLM-chatbots as tools for patient education and counselling in LBP shows promising but variable results. These chatbots generally provide moderately accurate recommendations. However, the accuracy may vary depending on the topic of each question. - Abstract
Background: Given the rapid advances in communication technology and the need that emerged from the COVID-19 pandemic, telehealth initiatives have been widely used worldwide. This masterclass aims to provide an overview of telerehabilitation for musculoskeletal conditions, synthesizing the different terminologies used to describe telehealth and telerehabilitation, its effectiveness and how to use it in clinical practice, barriers and facilitators for the implementation in health services, and discuss the need of a curriculum education for the near future., Main Body: Telerehabilitation refers to the use of information and communication technologies provided by any healthcare professionals for rehabilitation services. Telerehabilitation is a safe and effective option in the management of musculoskeletal conditions in different models of delivery. There are many technologies, with different costs and benefits, synchronous and asynchronous, that can be used for telerehabilitation: telephone, email, mobile health, messaging, web-based systems and videoconferences applications. To ensure a better practice of telerehabilitation, the clinician should certify safety and access, and appropriateness of environment, communication, technology, assessment, and therapeutic prescription. Despite the positive effect of telerehabilitation in musculoskeletal disorders, a suboptimal telerehabilitation implementation may have happened due to the COVID-19 pandemic, especially in countries where telehealth was not a reality, and clinicians lacked training and guidance. This emphasizes the need to identify the necessary curriculum content to guide future clinicians in their skills and knowledge for telerehabilitation. There are some challenges and barriers that must be carefully accounted for to contribute to a health service that is inclusive and relevant to health professionals and end users., Conclusions: Telerehabilitation can promote patient engagement in health care and plays an important role in improving health outcomes in patients with musculoskeletal conditions. Digital health technologies can also offer new opportunities to educate patients and facilitate the process of behavior change to a healthy lifestyle. Currently, the main needs in telerehabilitation are the inclusion of it in health curriculums in higher education and the development of cost-effectiveness and implementation trials, especially in low- and middle-income countries where access, investments and digital health literacy are limited., (© 2022. Rationale: Sarcoidosis is a multisystemic inflammatory disease characterized by the formation of granulomas in response to persistent stimuli. The long pentraxin PTX3 (pentraxin 3) has emerged as a component of humoral innate immunity with essential functions in the resolution of inflammation, but its role during granuloma formation is unknown. Objectives: To evaluate PTX3 as a modulator of pathogenic signals involved in granuloma formation and inflammation in sarcoidosis. Methods: Peripheral blood mononuclear cells obtained from patients with sarcoidosis harboring loss-of-function genetic variants and gene-deleted mice were used to assess the role of PTX3 in experimental models of granuloma formation in vitro and in vivo . The identified mechanisms of granulomatous inflammation were further evaluated in tissue and BAL samples and correlated with the disease course. Measurements and Main Results: We have identified a molecular link between PTX3 deficiency and the pathogenic amplification of complement activation to promote granuloma formation. Mechanistically, PTX3 deficiency licensed the complement component C5a-mediated activation of the metabolic checkpoint kinase mTORC1 (mammalian target of rapamycin complex 1) and the reprogramming of macrophages toward increased glycolysis to foster their proliferation and aggregation. This process sustained the further recruitment of granuloma-promoting immune cells and the associated proinflammatory microenvironment and influenced the clinical course of the disease. Conclusions: Our results identify PTX3 as a pivotal molecule that regulates complement-mediated signaling cues in macrophages to restrain granulomatous inflammation and highlight the therapeutic potential of this signaling axis in targeting granuloma formation in sarcoidosis.

- Abstract
Immune checkpoint inhibitors were approved for advanced nonsmall cell lung cancer (NSCLC) treatment. Despite improved survival, not all patients benefit from these agents. Here, the prognostic impact of pretreatment modified Glasgow Prognostic Score (mGPS) and neutrophil-to-lymphocyte ratio (NLR) was assessed. From 77 patients included, 83.2% received at least one prior systemic therapy. Immune-related adverse events (irAE) occurred in 20 patients. A lower mGPS was associated with higher median overall survival (OS), and a lower Eastern Cooperative Oncology Group (ECOG), irAE and fewer metastatic sites with better survival. A trend towards greater OS and progression-free survival (PFS) was stated among patients with NLR <5. mGPS 0 was associated with better survival; ≥3 metastatic sites with worse PFS and OS; ECOG >2 with worse OS and irAE with better survival. Pretreatment mGPS seems to be useful for predicting survival among advanced NSCLC patients treated with anti-programmed cell death 1 drugs, with ECOG performance status, irAE occurrence, and number of metastatic sites acting as survival predictors., (Copyright © 2021 Wolters Kluwer Health, Inc. 222. Resistance Profile of Osimertinib in Pre-treated Patients With EGFR T790M-Mutated Non-small Cell Lung Cancer.

Background: Osimertinib efficacy in pre-treated patients with epidermal growth factor receptor (EGFR) T790M-mutated non-small cell lung cancer (NSCLC) has been demonstrated in clinical trials, but real-world data, particularly regarding resistance profile, remains limited. This study aims to analyze the resistance mechanisms acquired after treatment with Osimertinib. Methods: Clinical outcomes and molecular results from re-biopsies at the time of osimertinib progression of EGFR T790M-mutated NSCLC patient were analyzed. Results: Twenty-one patients with stage IV adenocarcinoma were included [median 69 years; 57.1% female; 85.7% never-smokers; 23.8% ECOG performance status (PS) ≥2]. Median PFS and OS were 13.4 (95% CI: 8.0-18.9) and 26.4 (95% IC: 8.9-43.8) months, respectively. At the time of analysis, 10 patients had tumor progression (47.6%). T790M loss occurred in 50%, being associated with earlier progression (median PFS 8.1 vs. 21.4 months, p = 0.011). Diverse molecular alterations were identified, including C797S mutation ( n = 1), PIK3CA mutation ( n = 2), MET amplification ( n = 1), CTNNB1 mutation ( n = 1), and DCTN1-ALK fusion ( n = 1). Histological transformation into small cell carcinoma occurred in one patient. Purpose: Pain is common in youngsters. No matter its nature, youngsters' consideration of pain is complex since its communications vary as a function of several components. The aim of this study is to explore the youngsters' perception linked to their experience of physiological pain at the home/family setting, as well as their experience with the utilisation of over-the-counter pain medication as needed., Design and Methods: A qualitative exploratory study was undertaken with 22 youngsters aged 5-17 years old. Semidirected interviews led to the understanding of these components by means of the pain experiences. Thematic analysis allowed the detection of the themes and subthemes emerging from the verbatim collected with the participants., Results: The way pain is described is influenced by the child's development, previous experiences, and the projection of having pain. The pain communication is influenced by the severity perceived, the beliefs of the youngster experiencing pain, the comparison of the pain communication with his brothers and sisters, as well as the anticipated consequences of expressing his pain. The choice of behaviour towards pain is influenced by self-management through nonpharmacological management, with medicines if needed, and by family modelization., Practice Implications: This study confirms that previous pain experiences, beliefs related to pain tolerance and intended reactions of parents exert influence not only on the communication of pain, but also on youngsters' behaviour towards pain. 226. Adult Congenital Heart Disease Outpatient Clinic. Descriptive Analysis of A 12-Year Experience in Brazil.

- Abstract
Objective: Description of adult congenital heart disease (CHD) outpatient characteristics has not been reported and several aspects regarding these patients require attention. We describe the 12-year experience of a Brazilian unit., Methods: The main characteristics of 1168 patients were reviewed annotating for each patient age, gender, city of residence, main diagnosis, functional class at last examination, defect complexity and in-hospital referral pattern., Results: Increasing workload was documented. Among the CHD patients, 663 (57%) were between 14 and 30 years old and 920 (79%) lived in the referral region. Referrals were made by hospital cardiologists for 611 (52%) patients, while 519 (45%) were referred by pediatric cardiologists. Regarding CHD severity, 637 (55%) had a defect of mild complexity. Of the patients analyzed, 616 (53%) had undergone an intervention, mainly atrial septal defect (ASD) closure, correction of tetralogy of Fallot, ventricular septal defect (VSD) closure and relief of coarctation of the aorta (CoAo). The main diagnosis of the 552 (47%) patients not submitted to an intervention were ASD, VSD, aortic stenosis, complex CHD and pulmonary stenosis. Regarding functional class, 1016 (87%) were in class I and 280 (24%) were lost to follow-up. Seventy-three patients had died, mainly due to cardiac death., Conclusion: In a unit were complex pediatric congenital heart surgery started twenty years ago, an increasing adult CHD workload was documented. Referral came predominantly from cities around the unit, most patients had low complexity defects and were in functional class I, a significant loss of follow-up was documented, and the death of patients was mainly due to the heart defect.

228. Diagnostic yield and safety of transbronchial cryobiopsy in sarcoidosis.

- Abstract
Introduction: Transbronchial lung cryobiopsy (TBLC) is an endoscopic technique proven to be useful in diagnostic approach to interstitial lung disease (ILD), but its role in sarcoidosis is not fully established. The aim of the present study was to assess the diagnostic yield of TBLC in sarcoidosis and its safety profile., Methods: Retrospective analysis of patients, evaluated in a tertiary hospital ILD outpatient clinic, who underwent TBLC in the diagnostic work-up. TBLC was performed in accordance with the 2018 expert statement from the Cryobiopsy Working Group., Results: 32 patients were included (mean±sd age 47.7±12.6 years, 59.4% male) and divided into three groups: highly likely sarcoidosis (n=21), possible sarcoidosis (n=6) and unlikely sarcoidosis (n=5). A mean of 2.8±0.8 TBLCs were performed. The definitive diagnosis was established by TBLC in 20 out of 27 patients with suspected sarcoidosis. Two patients were diagnosed with sarcoidosis by other methods performed afterwards. TBLC leaded to other diagnosis as well, such as fungal infection (n=1), hypersensitivity pneumonitis (n=1) and silicosis (n=3), making the diagnostic yield for suspected sarcoidosis of TBLC of 92.6%. TBLC was also able to show compatible histological features in five patients whom sarcoidosis was not previously considered. The complications reported overall were pneumothorax in five (15.6%) patients and moderate bleeding in one (3.1%) case., Conclusion: In this cohort, TBLC was a safe, reliable and useful procedure in sarcoidosis diagnosis. 230. Targeted Gene Next-Generation Sequencing Panel in Patients with Advanced Lung Adenocarcinoma: Paving the Way for Clinical Implementation.

- Abstract
Identification of targetable molecular changes is essential for selecting appropriate treatment in patients with advanced lung adenocarcinoma. Methods : In this study, a Sanger sequencing plus Fluorescence In Situ Hybridization (FISH) sequential approach was compared with a Next-Generation Sequencing (NGS)-based approach for the detection of actionable genomic mutations in an experimental cohort (EC) of 117 patients with advanced lung adenocarcinoma. Its applicability was assessed in small biopsies and cytology specimens previously tested for epidermal growth factor receptor (EGFR) and anaplastic lymphoma kinase (ALK) mutational status, comparing the molecular changes identified and the impact on clinical outcomes. Subsequently, an NGS-based approach was applied and tested in an implementation cohort (IC) in clinical practice. Using Sanger and FISH, patients were classified as EGFR-mutated ( n = 22, 18.8%), ALK-mutated ( n = 9, 7.7%), and unclassifiable (UC) ( n = 86, 73.5%). Retesting the EC with NGS led to the identification of at least one gene variant in 56 (47.9%) patients, totaling 68 variants among all samples. Still, in the EC, combining NGS plus FISH for ALK, patients were classified as 23 (19.7%) EGFR; 20 (17.1%) KRAS; five (4.3%) B-Raf proto-oncogene (BRAF); one (0.9%) Erb-B2 Receptor Tyrosine Kinase 2 (ERBB2); one (0.9%) STK11; one (0.9%) TP53, and nine (7.7%) ALK mutated. Only 57 (48.7%) remained genomically UC, reducing the UC rate by 24.8%. Fourteen (12.0%) patients presented synchronous alterations. Concordance between NGS and Sanger for EGFR status was very high (κ = 0.972; 99.1%). In the IC, a combined DNA and RNA NGS panel was used in 123 patients. Genomic variants were found in 79 (64.2%). In addition, eight (6.3%) EML4-ALK, four (3.1%), KIF5B-RET, four (3.1%) CD74-ROS1, one (0.8%) TPM3-NTRK translocations and three (2.4%) exon 14 skipping MET Proto-Oncogene (MET) mutations were detected, and 36% were treatable alterations. Conclusions : This study supports the use of NGS as the first-line test for genomic profiling of patients with advanced lung adenocarcinoma.

- Abstract
Interest in the quality of life of people with mental disorders living in therapeutic residential care facilities is an important indicator for the evaluation of therapeutic interventions in the area of health. Physical activity can contribute to a good quality of life. This study evaluated the quality of life and levels of physical activity of people living in therapeutic residential care facilities in the metropolitan area of Porto Alegre. This case series study (n = 68) used SF-36, EuroQol and IPAQ and social-demographic questions. The SF-36 results showed that the domain of general health received the lowest scores (57.47 ± 14.27). The highest scores were in terms of social aspects (77.39 ± 20.21) and physical aspects (77.57 ± 39.71). When using EuroQol, at least one problem (mild or extreme) in at least one dimension was evident among 82% of the residents. The physical activity levels showed that most of the participants were insufficiently active (48.5%) and 14.7% were sedentary. The domains of pain and mobility suggested that the residents were not being encouraged enough to perform physical activities. Knowledge about the perceptions of those living in therapeutic residential care facilities is critical in order to establish effective public policies.

Migrant workers usually show higher rates of work-related health problems than natives. However, little information is available about their exposure to occupational risks. We describe self-reported working exposure in Spanish and foreign-born workers. A cross-sectional survey was conducted as part of the ITSAL Project. Data on sociodemographic and self-reported occupational exposure in 1,841 foreign-born and 509 Spanish workers were collected through face-to-face interviews. Prevalence and adjusted odds ratios-aOR- (by age, education, type of contract) were calculated. Foreign-born men in non-services sectors and those in manual occupations perceived exposure to occupational risks with lower prevalence than Spanish workers. Foreign-born women reported higher prevalence of exposure than Spanish female workers. By occupation, foreign-born female workers were more likely than Spanish workers to report working many hours/day (aOR2.68; 95 % CI 1.06-6.78) and exposure to extreme temperatures (aOR2.19; 95 % CI 1.10-4.38). Some groups of migrant workers may need increased protection regarding some occupational exposures.

- Abstract
Pulmonary arterial hypertension (PAH) is an obstructive vasculopathy characterized by enhanced pulmonary artery smooth muscle cell (PASMC) proliferation and suppressed apoptosis. This phenotype is sustained by the activation of the Src/signal transducer and activator of transcription 3 (STAT3) axis, maintained by a positive feedback loop involving miR-204 and followed by an aberrant expression/activation of its downstream targets such as Pim1 and nuclear factor of activated T-cells (NFATc2). Dehydroepiandrosterone (DHEA) is a steroid hormone shown to reverse vascular remodeling in systemic vessels. Since STAT3 has been described as modulated by DHEA, we hypothesized that DHEA reverses human pulmonary hypertension by inhibiting Src/STAT3 constitutive activation. Using PASMCs isolated from patients with PAH (n = 3), we demonstrated that DHEA decreases both Src and STAT3 activation (Western blot and nuclear translocation assay), resulting in a significant reduction of Pim1, NFATc2 expression/activation (quantitative RT-PCR and Western blot), as well as Survivin and upregulation of bone morphogenetic protein receptor 2 (BMPR2) and miR-204. Src/STAT3 axis inhibition by DHEA is associated with 1) mitochondrial membrane potential (tetramethylrhodamine methyl-ester perchlorate; n = 150; P < 0.05) depolarization increasing apoptosis by 25% (terminal deoxynucleotidyl transferase dUTP-mediated nick-end labeling; n = 150; P < 0.05); and 2) decreased intracellular Ca(2+) concentration (fluo-3 AM; n = 150; P < 0.05) and proliferation by 30% (PCNA). Finally, in vivo similarly to STAT3 inhibition DHEA improves experimental PAH (monocrotaline rats) by decreasing mean PA pressure and right ventricle hypertrophy. These effects were associated with the inhibition of Src, STAT3, Pim1, NFATc2, and Survivin and the upregulation of BMPR2 and miR-204. We demonstrated that DHEA reverses pulmonary hypertension in part by inhibiting the Src/STAT3.

- Abstract
Dehydroepiandrosterone (DHEA) is an endogenous steroid hormone involved in a number of biological actions in humans and rodents, but its effects on renal tissue have not yet been fully understood. The aim of this study is to assess the effect of DHEA treatment on diabetic rats, mainly in relation to renal function and metabolism. Diabetic rats were treated with subcutaneous injections of a 10mg/kg dose of DHEA diluted in oil. Plasma glucose and creatinine, in addition to urine creatinine, were quantified espectophotometrically. Glucose uptake and oxidation were quantified using radioactive glucose, the urinary Transforming Growth Factor β(1) (TGF-β(1)) was assessed by enzyme immunoassay, and the total glutathione in the renal tissue was also measured. The diabetic rats displayed higher levels of glycemia, and DHEA treatment reduced hyperglycemia. Plasmatic creatinine levels were higher in the diabetic rats treated with DHEA, while creatinine clearance was lower. Glucose uptake and oxidation were lower in the renal medulla of the diabetic rats treated with DHEA, and urinary TGF-β(1), as well as total gluthatione levels, were higher in the diabetic rats treated with DHEA. DHEA treatment was not beneficial to renal tissue, since it reduced the glomerular filtration rate and renal medulla metabolism, while increasing the urinary excretion of TGF-β(1) and the compensatory response by the glutathione system, probably due to a mechanism involving a pro-oxidant action or a pro-fibrotic effect of this androgen or its derivatives. - Abstract
This study examined, in young and old (3 and 24 month-old, respectively) healthy Wistar rats, the in vivo effect of DHEA (10 mg/kg body weight) administered subcutaneously for 5 weeks. Reduced (GSH) and oxidized (GSSG) glutathione levels, glucose-6-phosphate dehydrogenase (G6PDH), glutathione peroxidase (GPx), glutathione-S-transferase (GST) and thioredoxin (Trx) reductase activities, hydrogen peroxide steady-state concentration and Nrf2, GST, Trx-1, Akt and p-Akt expressions were assessed in heart tissue. DHEA treatment significantly increased GST activity in 3 and 24 month-old treated groups. The aging factor diminished hydrogen peroxide concentration and Nrf2 expression, independently of treatment. However, the aging process increased GST, Akt and p-Akt expressions in both 24 month-old groups. The aged group responded differently to DHEA respective to GSSG content, GPx activity and p-Akt concentration. Further studies are needed to form conclusions about the efficacy and safety of DHEA replacement in the elderly, and to better understand DHEA's net effect on oxidative stress parameters and its modulation of signaling cascades., (Copyright © 2010 Elsevier Inc. - Abstract
Ageing is an inevitable biological process characterized by a general decline in various physiological functions. DHEA and DHEAS levels are maximal between the second and third life decades, then start to decline 2% per year, leaving a residual of 10-20% of the peak production by the eighth decade. Erythrocytes are exposed to frequent oxidative stress due to the oxygen radicals continuously generated by haemoglobin auto-oxidation. We investigated DHEA chronic (10 mg/kg, subcutaneously, for 5 weeks) effects over oxidative stress markers in erythrocytes of male Wistar rats of 3, 13 and 18 month-old. In the 13 month-old group, we found increased lipid peroxidation (LPO), superoxide dismutase (SOD), glutathione-S-transferase and catalase activities when compared to the other age groups. DHEA produced a marked increase in LPO of 13 month-old group when compared to its control. DHEA exerted this pro-oxidant effects in all ages studied, especially in age 13 month-old. It seems that at 13 month-old there would be an important depletion of some specific anti-oxidant in order to determine such susceptibility to DHEA effects. Since this approach allows a minimally invasive assessment, it would be useful as a routine method in human clinical studies investigating DHEA effects during the ageing process., (2009 John Wiley & Sons, Ltd.)

- Abstract
The secretion of DHEA-synthesized mainly in the adrenal cortex-increases in the postnatal aging, peaks in the twenties and decreases with age afterwards. Exogenous DHEA can exert a dual effect depending on dose and on tissue. Akt is a serine/threonine kinase whose activity has been seen as an interventional approach for cardiomyopathic damage resulting from aging changes. In order to evaluate DHEA effects over myocardial Akt protein expression associated to oxidative stress markers during aging, male Wistar rats (3 and 18 months) were assigned into two groups: control or DHEA (10mg/kg, subcutaneously, for 5 weeks). In the aged group, we found increased lipid peroxidation and glutathione-S-transferase activity. DHEA produced an increase in p-Akt protein expression and a decrease in SOD activity in both ages. Akt pathway activation might be related to changes in oxidative stress parameters according to age.

- Abstract
Early-life environmental events can induce profound long-lasting changes in several behavioral and neuroendocrine systems. The neonatal handling procedure, which involves repeated brief maternal separations followed by experimental manipulations, reduces stress responses and sexual behavior in adult rats. The purpose of this study was to analyze the effects of neonatal handling on social behaviors of male and female rats in adulthood, as manifest by the results of social memory and social interaction tests. The number of oxytocin (OT) and vasopressin (VP) neurons in the paraventricular (PVN) and supraoptic (SON) nuclei of hypothalamus were also analyzed. The results did not demonstrate impairment of social memory. Notwithstanding, handling did reduce social investigative interaction and increase aggressive behavior in males, but did not do so in females. Furthermore, in both males and females, handling was linked with reduced number of OT-neurons in the parvocellular region of the PVN, while no differences were detected in the magnocellular PVN or the SON. On the other hand, handled males exhibited increased number of VP-neurons in the magnocellular zone of the PVN. We may conclude that the repeated brief maternal separations can reduce affiliative social behavior in adult male rats. Moreover, the disruption of the mother-infant relationship caused by the handling procedure induced long-lasting morphological changes in critical neuroendocrine areas that are involved in social bonding in mammals.

Objective: To describe the activities of safety representatives in workplaces in Spain., Methods: A specific questionnaire was applied to a representative sample of safety representatives. Activities developed during the past year, presented in a closed ended list, were categorised into three groups: information and advising; participation in occupational health management; and pressure on or negotiation with employers. Personal phone interviews were conducted from September to December 2004. A sample of 1201 interviews was attained. Crude and multivariate analyses were carried out., Results: Spanish safety representatives were mostly men (76%), aged 26 to 45 years (62%), with fixed contracts (94%), and more than 10 years in their company (57%). On a comparable 0-10 scale, the mean (95% confidence interval (CI)) number of activities relating to information, management, and negotiation developed during the previous year were, respectively, 6.8 (6.7 to 6.9), 4.5 (4.4 to 4.7), and 4.0 (3.8 to 4.1). In multivariate analysis, workplace size (>30 workers), industrial sector, training, and support from the labour inspectorate were the factors most consistently associated with safety representatives' activity. Additionally, support from the employer was associated with participation in occupational health management (odds ratio = 2.38 (95% CI, 1.73 to 3.29))., Conclusions: Safety representatives in Spain have a variety of activities, mostly in the category of information and advising. These are necessary but not sufficient for real participation of workers in decisions concerning their health and safety. More participation of safety representatives in occupational health management at workplaces seems to be needed, and factors associated with this participation reinforced.

- Abstract
Myocardial activity and gene expression of antioxidant defenses and oxidative damage were examined in an experimental model of pressure overload hypertrophy. Male Wistar rats were divided into abdominal aortic-banded or sham-operated groups. After 30 days, arterial pressure and heart rate were measured. Heart, lung, and liver were extracted and weighted to evaluate cardiac hypertrophy and pulmonary and hepatic congestion. Heart homogenates were prepared to quantify lipid peroxidation (LPO); the activities of superoxide dismutase (SOD), catalase (CAT), glutathione-S-transferase (GST), glutathione peroxidase (GPx) and glutathione reductase (GR); and Cu-Zn SOD and GST concentrations. Total glutathione (GSH) myocardial content was also measured. Arterial pressure (142 +/- 17 mmHg) and cardiac hypertrophy index (3.4 +/- 0.45 mg/g) were significantly increased (by 38% and 22%, respectively, p<0.0001) in the aortic-banded group. LPO was enhanced by 55% in the aortic-banded group (11891 +/- 766 cps/mg protein, p<0.001) compared with that in the controls. SOD activity and concentration were higher (40% and 38%, 15.15 +/- 1.03 U/mg protein, 49.187 pixels, respectively, p<0.05) in the aortic-banded group than in the controls. Aortic-banding induced a decrease by 28% in GST (48 +/- 10 pmol/min/mg protein, p<0.005), by 36% in GPx (38.2 +/- 9.5 nmol/min/mg protein, p<0.005), by 31% in GR activities (1.55 +/- 0.23 nmol/mg protein, p<0.0005), and by 43% in GSH content (0.13 +/- 0.02 nmol/mg protein, p<0.005). In conclusion, in this model it was observed that myocardial oxidative stress induces alterations in antioxidant enzyme activities and protein expression. The follow up of these parameters could afford an early therapeutical window to avoid heart failure progression.
