- Abstract
Non-coding genetic variants play an important role in driving susceptibility to complex diseases but their characterization remains challenging. Here, we employed a novel approach to interrogate the genetic risk of such polymorphisms in a more systematic way by targeting specific regulatory regions relevant for the phenotype studied. We applied this method to meningococcal disease susceptibility, using the DNA binding pattern of RELA - a NF-kB subunit, master regulator of the response to infection - under bacterial stimuli in nasopharyngeal epithelial cells. We designed a custom panel to cover these RELA binding sites and used it for targeted sequencing in cases and controls. Variant calling and association analysis were performed followed by validation of candidate polymorphisms by genotyping in three independent cohorts. We identified two new polymorphisms, rs4823231 and rs11913168, showing signs of association with meningococcal disease susceptibility. In addition, using our genomic data as well as publicly available resources, we found evidences for these SNPs to have potential regulatory effects on ATXN10 and LIF genes respectively. The variants and related candidate genes are relevant for infectious diseases and may have important contribution for meningococcal disease pathology. Finally, we described a novel genetic association approach that could be applied to other phenotypes.

- Abstract
Fever is the most common reason that children present to Emergency Departments. Clinical signs and symptoms suggestive of bacterial infection are often non-specific, and there is no definitive test for the accurate diagnosis of infection. The 'omics' approaches to identifying biomarkers from the host-response to bacterial infection are promising. In this study, lipidomic analysis was carried out with plasma samples obtained from febrile children with confirmed bacterial infection (n = 20) and confirmed viral infection (n = 20). We show for the first time that bacterial and viral infection produces distinct profile in the host lipidome. Some species of glycerophosphoinositol, sphingomyelin, lysophosphatidylcholine and cholesterol sulfate were higher in the confirmed virus infected group, while some species of fatty acids, glycerophosphocholine, glycerophosphoserine, lactosylceramide and bilirubin were lower in the confirmed virus infected group when compared with confirmed bacterial infected group. A combination of three lipids achieved an area under the receiver operating characteristic (ROC) curve of 0.911 (95% CI 0.81 to 0.98). This pilot study demonstrates the potential of metabolic biomarkers to assist clinicians in distinguishing bacterial from viral infection in febrile children, to facilitate effective clinical management and to the limit inappropriate use of antibiotics.

Aim: The purpose of this study is to review the indications, techniques, advantages, and outcomes of perichondrial cutaneous grafts (PCCG). Background The PCCG is a composite graft utilized in the reconstruction of select head and neck defects. This graft, much studied and popularized by Dr Frederick Stucker, has several intrinsic benefits due to its size, thickness, contour, and unique perichondrial layer. Results The PCCG is primarily utilized in the reconstruction of nasal and lower eyelid defects. This graft has been demonstrated to yield superior results to a full-thickness skin graft (FTSG) in these cosmetically sensitive locations and has the convenience of a single-stage procedure that may be performed under local anesthesia in contrast to a paramedian forehead flap (PMFF). Animal studies have shown less contraction in comparison to FTSGs. Large case series have reported low rates of graft loss and good esthetic outcomes. Conclusion The PCCG is an option to consider in the reconstruction of select head and neck defects, as it tends to offer excellent cosmetic results and minimal morbidity. Clinical significance The PCCG has several advantages over a FTSG for moderate to large defects of the face, and is an excellent reconstructive option for nasal defects in which a PMFF may not be a viable option. This minireview provides an overview of known and potential gender differences in physiological responses to spaceflight. The paper covers cardiovascular and exercise physiology, barophysiology and decompression sickness, renal stone risk, immunology, neurovestibular and sensorimotor function, nutrition, pharmacotherapeutics, and reproduction. Potential health and functional impacts associated with the various physiological changes during spaceflight are discussed, and areas needing additional research are highlighted. Historically, studies of physiological responses to microgravity have not been aimed at examining gender-specific differences in the astronaut population. Insufficient data exist in most of the discipline areas at this time to draw valid conclusions about gender-specific differences in astronauts, in part due to the small ratio of women to men. The only astronaut health issue for which a large enough data set exists to allow valid conclusions to be drawn about gender differences is orthostatic intolerance following shuttle missions, in which women have a significantly higher incidence of presyncope during stand tests than do men. The most common observation across disciplines is that individual differences in physiological responses within genders are usually as large as, or larger than, differences between genders. Individual characteristics usually outweigh gender differences per se.

- Abstract
The structure and organization of a species genome at a karyotypic level, and in interphase nuclei, have broad functional significance. Although regular sized chromosomes are studied extensively in this regard, microchromosomes, which are present in many terrestrial vertebrates, remain poorly explored. Birds have more cytologically indistinguishable microchromosomes (~ 30 pairs) than other vertebrates; however, the degree to which genome organization patterns at a karyotypic and interphase level differ between species is unknown. In species where microchromosomes have fused to other chromosomes, they retain genomic features such as gene density and GC content; however, the extent to which they retain a central nuclear position has not been investigated. In studying 22 avian species from 10 orders, we established that, other than in species where microchromosomal fusion is obvious (Falconiformes and Psittaciformes), there was no evidence of microchromosomal rearrangement, suggesting an evolutionarily stable avian genome (karyotypic) organization. Moreover, in species where microchromosomal fusion has occurred, they retain a central nuclear location, suggesting that the nuclear position of microchromosomes is a function of their genomic features rather than their physical size.

The Palaeolithic record of Greece remains highly fragmented and discontinuous in both space and time. Nevertheless, new surveys and excavations, along with the revisiting of known sites or old collections, and the conduction of lithic and faunal laboratory analyses, have altogether enriched the Greek Palaeolithic dataset with important new evidence and novel interpretations. The goal of this paper is threefold: 1) to critically review the most important aspects of the Greek Pleistocene archaeological record, from the Lower to the Upper Palaeolithic; 2) to provide a synthesis of current knowledge about the Palaeolithic of Greece and in the framework of broader discussions in human evolution research; and 3) to put in prospect the Greek record by addressing a research agenda for the future. The review of the evidence shows that Palaeolithic research in Greece has expanded its focus not only geographically but also temporally: it now includes investigations at previously under-studied areas, such as the insular settings of the Aegean and Ionian Seas, as well as formerly overlooked targets, such as Lower Palaeolithic open-air sites. The synthesis and discussion which follows offers a state-of-the-art perspective on how the primary Palaeolithic data can be assessed within local or regional geomorphic, paleoenvironmental and chronological contexts; here, our focus is on spatio-temporal discontinuities, trends in subsistence strategies and lithic technology, as well as potentially emerging biogeographical patterns. Finally, we highlight the complex topography and mosaic landscapes of the Greek peninsula in order to address two major themes for a future research agenda: the potential role of Greece as a glacial refugium, and how the Greek record could contribute to our knowledge of early hominin mobility patterns.

- Abstract
Robots that reside inside the body to restore or enhance biological function have long been a staple\ud of science fiction. Creating such robotic implants poses challenges both in signaling between the\ud implant and the biological host as well as in implant design. To investigate these challenges, we\ud created a robotic implant to perform in vivo tissue regeneration via mechano-stimulation. The\ud robot is designed to induce lengthening of tubular organs, such as the esophagus and intestines, by\ud computer-controlled application of traction forces. Esophageal testing in swine demonstrates that\ud the applied forces can induce cell proliferation and lengthening of the organ without a reduction\ud in diameter, while the animal is awake, mobile and able to eat normally. Such robots can serve as\ud research tools for studying mechanotransduction-based signaling and can also be employed\ud clinically for conditions such as long-gap esophageal atresia and short bowel syndrome.
