Bread wheat is a major crop that has long been the focus of basic and breeding research. Assembly of its genome has been difficult because of its large size and allohexaploid nature (AABBDD genome). Following the first reported assembly of the genome of the experimental strain Chinese Spring (CS), the 10+ Wheat Genomes Project was launched to produce multiple assemblies of worldwide modern cultivars. The only Asian cultivar in the project is Norin 61, a representative Japanese cultivar adapted to grow across a broad latitudinal range, mostly characterized by a wet climate and a short growing season. Here, we characterize the key aspects of its chromosome-scale genome assembly spanning 15 Gb with a raw scaffold N50 of 22 Mb. Analysis of the repetitive elements identified chromosomal regions unique to Norin 61 that encompass a tandem array of the pathogenesis-related 13 family. We report novel copy-number variations in the B homeolog of the florigen gene FT1/VRN3, pseudogenization of its D homeolog and the association of its A homeologous alleles with the spring/winter growth habit. Furthermore, the Norin 61 genome carries typical East Asian functional variants different from CS, ranging from a single nucleotide to multi-Mb scale. Examples of such variation are the Fhb1 locus, which confers Fusarium head-blight resistance, Ppd-D1a, which confers early flowering, Glu-D1f for Asian noodle quality and Rht-D1b, which introduced semi-dwarfism during the green revolution. The adoption of Norin 61 as a reference assembly for functional and evolutionary studies will enable comprehensive characterization of the underexploited Asian bread wheat diversity.

Rationale Pediatric COVID-19 studies have been mostly restricted to case reports and small case series, which have prevented the identification of specific pediatric lung disease patterns in COVID-19. The overarching goal of this systematic review and meta-analysis is to provide the first comprehensive summary of the findings of published studies thus far describing COVID-19 lung imaging data in the pediatric population. Methods A systematic literature search of PubMed was performed to identify studies assessing lung-imaging features of COVID-19 pediatric patients (0-18 years). A single-arm meta-analysis was conducted to obtain the pooled prevalence and 95% confidence interval (95% CI). Results A total of 29 articles (n = 1026 children) based on chest computerized tomography (CT) images were included. The main results of this comprehensive analysis are as follows: (1) Over a third of pediatric patients with COVID-19 (35.7%, 95% CI: 27.5%-44%) had normal chest CT scans and only 27.7% (95% CI: 19.9%-35.6%) had bilateral lesions. (2) The most typical pediatric chest CT findings of COVID-19 were ground-glass opacities (GGO) (37.2%, 95% CI: 29.3%-45%) and the presence of consolidations or pneumonic infiltrates (22.3%, 95% CI: 17.8%-26.9%). (3) The lung imaging findings in children with COVID-19 were overall less frequent and less severe than in adult patients. (4) Typical lung imaging features of viral respiratory infections in the pediatric population such as increased perihilar markings and hyperinflation were not reported in children with COVID-19. Conclusion Chest CT manifestations in children with COVID-19 could potentially be used for early identification and prompt intervention in the pediatric population.

Aurora B kinase plays essential roles in mitosis. Its protein levels increase before the onset of mitosis and sharply decrease during mitosis exit. The latter decrease is due to a balance between the actions of the E3 ubiquitin ligase anaphase-promoting complex or cyclosome (activated by the Cdh1 adapter), and the deubiquitinating enzyme USP35. Aurora B also executes important functions in interphase. Abnormal modulation of Aurora B in interphase leads to cell cycle defects often linked to aberrant chromosomal condensation and segregation. Very little is however known about how Aurora B levels are regulated in interphase. Here we found that USP13-associates with and stabilizes Aurora B in cells, especially before their entry into mitosis. In order for USP13 to exert its stabilizing effect on Aurora B, their association is promoted by the Aurora B-mediated phosphorylation of USP13 at Serine 114. We also present evidence that USP13 instigates Aurora B deubiquitination and/or protect it from degradation in a non-catalytic manner. In addition, we report that genetic or chemical modulation of the cellular levels/activity of USP13 affects unperturbed cell-cycle progression. Overall our study unveils the molecular and cellular connections of the USP13-Aurora B axis, which potentially participates in the rewiring of the cell cycle happening in cancer cells.

Climate and land-use change have led to disturbance regimes in many ecosystems without a historical analog, leading to uncertainty about how species adapted to past conditions will respond to novel post-disturbance landscapes. We examined habitat selection by spotted owls in a post-fire landscape. We tested whether selection or avoidance of severely burned areas could be explained by patch size or configuration, and whether variation in selection among individuals could be explained by differences in habitat availability. We applied mixed-effects models to GPS data from 20 spotted owls in the Sierra Nevada, California, USA, with individual owls occupying home ranges spanning a broad range of post-fire conditions after the 2014 King Fire. Individual spotted owls whose home ranges experienced less severe fire ( 100-m into severe fire patches. Spotted owls avoided areas that had experienced post-fire salvage logging but the interpretation of this effect was nuanced. Owls also avoided areas that were classified as open and/or young forest prior to the fire. Our results support the hypothesis that spotted owls are adapted to historical fire regimes characterized by small severe fire patches in this region. Shifts in disturbance regimes that produce novel landscape patterns characterized by large, homogeneous patches of high-severity fire may negatively affect this species.

The design of computer-supported caregiving technology has generally not acknowledged the complexity and heterogeneity of the informal care provision to older adults. For instance, most Latin American older adults have not been capable to broadly embrace digital technology to interact with their family members. This attitude, complemented with a high commitment derived from a strong filial obligation, burdens those family members that are most engaged in caring for their older adults, indirectly producing tension within the family network. In order to better understand this scenario in Southern Cone families (i.e., a particular region within Latin America comprising Chile, Argentina, and Uruguay), we conducted a localized multi-method study involving shadowing, contextual inquiry, and semi-structured interviews with informal elderly caregivers. Our study results highlight design implications and structure the elderly caregiving ecosystem, providing means to support invisible work and its articulation in this highly collaborative scenario. In particular, (1) we define a set of roles that characterize the viewpoints and concerns of the different family members regarding informal elderly caregiving, and therefore sustain the articulation of the process; (2) we draft the main concerns of the involved stakeholders in the form of a caregiving matrix, which can be used for visualizing the current fulfillment of duties within the family network; and (3) we describe a set of typical caregiving scenarios aimed to inform the design of contextualized strategies for mediating the social interaction space of intergenerational families through computer-supported technology. By addressing the identified implications, HCI and CSCW researchers, designers, and practitioners would be able to better understand the complexity of informal elderly caregiving in the South of Latin America, and therefore identify plausible solutions that would improve user experience and the effectiveness of computer-supported mediation strategies this context.

Internal stochastic resonance (internal SR) is a phenomenon of non-linear systems in which the addition of a non-zero level of noise produces an enhancement in the coherence between two or more signals. In a previous study, we found that the simultaneous administration of multisensory visual and auditory noise augments global coherence in electroencephalographic (EEG) signals via this phenomenon. Here, we examined whether such global coherence can also be augmented with at least one noisy acoustic source. We performed experiments on healthy subjects and applied the following binaural and monaural noise-stimulation protocols. First, we administered to the left ear Gaussian noise of fixed intensity, while we delivered to the right ear a second Gaussian noise of variable intensity levels (binaural protocol). Second, we applied the Gaussian noise of the same variable intensity levels but only to one ear (monaural protocol). We performed a permutation test analysis, finding that during both noise protocols there was a significant enhancement in the global coherence in EEG signals via the occurrence of internal SR within central pathways of the auditory system.

Polycomb group (PcG) RING finger protein 5 (PCGF5) is a core component of the so-called Polycomb repressive complex 1.5 (PRC1.5), which is involved in epigenetic transcriptional repression. To explore the developmental function of Pcgf5, we generated Pcgf5 knockout (Pcgf5-/- ) mouse embryonic stem cell (mESC) lines with the help of CRISPR/Cas9 technology. We subjected the Pcgf5-/- and wild-type (WT) mESCs to a differentiation protocol toward mesodermal-cardiac cell types as aggregated embryoid bodies (EBs) and we found that knockout of Pcgf5 delayed the generation of the three germ layers, especially the ectoderm. Further, disruption of Pcgf5 impacted the epithelial-mesenchymal transition during EB morphogenesis and differentially affected the gene expression of essential developmental signaling pathways such as Nodal and Wnt. Finally, we also unveiled that loss of Pcgf5 induced the repression of genes involved in the Notch pathway, which may explain the enhancement of cardiomyocyte maturation and the dampening of ectodermal-neural differentiation observed in the Pcgf5-/- EBs.

Purpose This study aims to analyse the existing literature on continuous improvement (CI) initiatives and dynamic capabilities (DCs) development to explore the question whether CI initiatives foster development of DCs in organisations. Design/methodology/approach A systematic literature review (SLR) was undertaken. Four databases were included in the structured searches (EBSCOhost, Web of Knowledge, Scopus, ProQuest and Emerald Insight), 19 studies were finally included and analysed in detail. Findings The results obtained identify issues such as a growing trend in the publication of studies, the dominant position of the USA and the predominance of empirical papers. The literature was classified according to whether it presents CI as a DC in itself, as an enabler of DC or as a result of the DC. The main critical success factors to be implemented in CI initiatives (CII) were also identified, to enhance the development of DCs. Finally, based on the analysis of the specific DC literature, ten theoretical propositions for possible future research have been developed. Originality/value CII such as Lean Management, Six Sigma and Total Quality Management have been widely implemented in organisations. Despite their reputation, the effects of these initiatives on long-term benefits remain debated, this motivates the SLR of CII and DC. The DC theory tackles the question of how firms can sustain their advantage and profits in the long term, making this perspective ideal for tackling controversy on the benefits of CII.

Chronic granulomatous disease (CGD) is an inborn error of immunity caused by defects in the phagocytic nicotinamide adenine dinucleotide phosphate oxidase complex, leading to increased susceptibility to infection and inflammatory autoimmune diseases. Up to 50% of patients have gastrointestinal (GI) involvement and meet diagnostic criteria for inflammatory bowel disease (CGD-IBD).We analyzed patients with CGD from the US Immunodeficiency Network (USIDNET) registry to determine whether IBD changes the presentation, treatment, and outcomes of patients with CGD.A retrospective evaluation of CGD cases from the USIDNET registry was completed. CGD-IBD was defined as the presence of any major physician-reported inflammatory, noninfectious GI disease manifestation. Demographic information, conditions, infections, antimicrobial therapies, immunomodulator use, and hematopoietic stem cell transplantation data were analyzed.Of 194 patients with a diagnosis of CGD, 96 met criteria for IBD and 98 were categorized in the non-IBD group. Patients with CGD-IBD had an increased rate of infection compared with the non-IBD group (0.66 vs 0.36 infections/patient/year). Enteric organism infections were more common in patients with IBD. Immunomodulators were used at a significantly higher percentage in patients with IBD compared with patients without IBD (80% vs 56%, P.001). Of the entire CGD cohort, 17 patients died (8.8%), with no significant difference between patients with IBD and patients without IBD (P = 1.00).Infectious events, enteric organism infections, and use of immunomodulatory drugs were higher in patients with IBD than patients without IBD; however, mortality was not increased. Patients with CGD and concurrent IBD are at increased risk for disease complications, supporting the importance of early recognition, diagnosis, and treatment.

To assess the feasibility of using high-temperature superconductors for the beam screens of future circular colliders, we have undertaken a study of the power dependence of the microwave surface resistance in state-of-the-art REBCO coated conductors at about 8 GHz and 50 K. We have employed a dielectric resonator to produce radio-frequency (RF) electromagnetic fields on the surface of the coated conductors having amplitudes similar to those generated by proton bunches circulating in the vacuum chamber of the proposed future circular collider Hadron-Hadron (FCC-hh) at CERN We show that surface resistances in REBCO coated conductors without artificial pinning centers are more affected by a RF magnetic field than those containing nano-inclusions. Despite that, at 8 GHz, 50 K, and 9 T, most REBCO coated conductors studied outperform copper in terms of surface resistance, with the best sample having a 2.3 mΩ surface resistance while being subject to an RF field 2.5 times stronger than that in the FCC-hh. We also extrapolate the measured data to 16 T and 1 GHz, the actual FCC-hh dipole magnetic field, and the mid-beam frequency spectrum, demonstrating the possibility of lowering the surface resistance of the vacuum chamber by up to two orders of magnitude compared to copper. Haloscope experiments directly search for these particles in strong magnetic fields with RF cavities as detectors. The Relic Axion Detector Exploratory Setup (RADES) at CERN in particular is searching for axion dark matter in a mass range above 30 $\mu$eV. The figure of merit of our detector depends linearly on the quality factor of the cavity and therefore we are researching the possibility of coating our cavities with different superconducting materials to increase the quality factor. Since the experiment operates in strong magnetic fields of 11 T and more, superconductors with high critical magnetic fields are necessary. Suitable materials for this application are for example REBa$_2$Cu$_3$O$_{7-x}$, Nb$_3$Sn or NbN. We designed a microwave cavity which resonates at around 9~GHz, with a geometry optimized to facilitate superconducting coating and designed to fit in the bore of available high-field accelerator magnets at CERN. Several prototypes of this cavity were coated with different superconducting materials, employing different coating techniques. Supporting asymmetric social interaction is a key requirement for developers of ad hoc social applications, who need to keep up with the pace of the evolution of social media technology without leaving digital orphans on the road. This asymmetry becomes more evident in intergenerational social communication and it has been recognized as an open problem given the lack of interoperability among the social media platforms. The quick evolution and diversification of these platforms help increase this asymmetry. On the one hand, this situation impacts the developers of ad hoc social applications, who have to keep updating their systems. On the other hand, the end-users of these applications are affected as well, given that they depend on the developers to continue interacting with such platforms. This manuscript presents social message translator (SMT), a software system that allows addressing the interaction asymmetry between ad hoc social applications and regular social media platforms. The message translation mechanisms of SMT, its security, and the usability and extensibility of its services were evaluated through three empirical studies. The obtained results are highly positive, showing that SMT goes one step forward in the development of socio-domestic computing systems to support asymmetric social interaction.

The cell division cycle is a process that is exquisitely controlled by a complex interplay between E3 ubiquitin ligases and deubiquitinating enzymes (DUBs). We have previously reported that the DUB USP13 regulates Aurora B levels along the cell cycle. That observation prompted us to explore any possible connection between USP13 and the APC/CWe performed immunoprecipitation assays followed by western-blotting to assess the interaction between USP13 and CDH1. The cellular effects of USP13 gain or loss of function were analyzed by transfection of FLAG-tagged USP13 plasmid or small interfering RNAs and short hairpin RNAs directed against USP13. The levels of CDH1 and other proteins were quantified in cell extracts by western-blotting.We found that USP13 binds to the APC/C adaptor CDH1. In addition, we report for the first time that USP13 controls CDH1 protein levels in cells: overexpression of USP13 increased CDH1 levels, whereas depletion of USP13 decreased CDH1 levels.We unveil the existing interplay between USP13 and CDH1: USP13 is capable of stabilizing CDH1 levels. Systemic autoinflammatory diseases (SAIDs) are characterized by unprovoked exaggerated inflammation on a continuum from benign recurrent oral ulceration to life-threatening strokes or amyloidosis, with renal failure as a potential sequela. The ability to discriminate these diagnoses rests on the genetic and mechanistic defect of each disorder, considering potential overlapping autoinflammation, autoimmunity, and immune deficiency. A comprehensive and strategic genetic investigation influences management as well as the consequential expected prognoses in these subsets of rare diseases. The ever-expanding therapeutic armamentarium reflects international collaborations, which will hasten genetic discovery and consensus-driven treatment.

BackgroundLoss of secretory immunoglobulin A (SIgA) is common in COPD small airways and likely contributes to disease progression. We hypothesized loss of SIgA results from reduced expression of pIgR, a chaperone protein needed for SIgA transcytosis, in the COPD small airway epithelium.MethodspIgR-expressing cells were defined and quantified at single-cell resolution in human airways using RNA in-situ hybridization, immunostaining, and single-cell RNA sequencing. Complementary studies in mice utilized immunostaining, primary murine tracheal epithelial cell (MTEC) culture, and transgenic mice with secretory or ciliated cell-specific knockout of pIgR. SIgA degradation by human neutrophil elastase or secreted bacterial proteases from non-typeable Haemophilus influenzae (NTHi) was evaluated in vitro.ResultsWe found that secretory cells are the predominant cell type responsible for pIgR expression in human and murine airways. Loss of SIgA in small airways was not associated with a reduction in secretory cells but rather a reduction in pIgR protein expression despite intact PIGR mRNA expression. Neutrophil elastase and NTHi-secreted proteases are both capable of degrading SIgA in vitro and may also contribute to a deficient SIgA immunobarrier in COPD.InterpretationLoss of the SIgA immunobarrier in small airways of patients severe COPD is complex and likely results from both pIgR-dependent defects in IgA transcytosis and SIgA degradation.Key MessagesWhat is the key question? Localized SIgA deficiency in small airways is an established driver of COPD pathogenesis, but the mechanism of loss remains unclear. We hypothesized loss of SIgA is due to reduced numbers of pIgR-expressing cells in SIgA-deficient small airways.What is the bottom line? pIgR is primarily expressed by secretory cells in human and murine airways. Although numbers of secretory cells are similar between SIgA-deficient and SIgA-replete airways in COPD, there is reduced expression of pIgR protein, but not mRNA, in SIgA-deficient airways. Additionally, host and bacterial proteases degrade SIgA in vitro, suggesting loss of SIgA may relate to both impaired transcytosis and increased degradation.Why read on? This study highlights the complexity of SIgA immunobarrier maintenance and suggests that strategies aimed at restoring the SIgA immunobarrier will need to account for both impaired transcytosis and degradation by host and/or bacterial proteases.

Heat stress affects oocyte developmental competence and is a major cause of reduced fertility in heat stressed cattle. Negative effects of heat stress on the oocyte have been observed at morphological, biochemical and developmental levels. However, the mechanisms by which heat stress affects the oocyte at the transcriptional and epigenetic levels remain to be further elucidated. Here we aimed to investigate the effect of heat stress on oocyte quality, transcriptomic profiles and DNA methylation of oocytes collected through the transition from spring to summer under Louisiana conditions. Summer season resulted in a lower number of high quality oocytes obtained compared to the spring season. There was no difference in in vitro maturation rates of oocytes collected during spring as compared to summer. RNA sequencing analysis showed that a total of 211 and 92 genes were differentially expressed as a result of heat stress in GV and MII oocytes, respectively. Five common genes (E2F8, GATAD2B, BHLHE41, FBXO44, and RAB39B) were significantly affected by heat in both GV and MII oocytes. A number of pathways were also influenced by heat stress including glucocorticoid biosynthesis, apoptosis signaling, and HIPPO signaling in GV oocytes, and Oct4 pluripotency, Wnt/beta-catenin signaling, and melatonin degradation I in MII oocytes. In addition, fluorescent immunocytochemistry analysis showed no difference in global levels of DNA methylation and DNA hydroxymethylation at either the GV or MII stage between spring and summer oocytes. The results of this study contribute to a better understanding of the effect of heat stress on the molecular mechanisms altered in bovine oocytes.

Previous initiatives have identified the potential of the Latin American region for CSCW research. However, CSCW work in the region is still disconnected, produced by small, isolated groups that often have a hard time learning about each other and finding opportunities to do research together. This workshop is an initiative of the SIGCHI Latin America Committee to bring these groups together to discuss their research, the challenges they perceive, and the possible initiatives that could be conducted to strengthen the region.

Pressure retarded osmosis (PRO) power units, which produce electrical energy from salinity gradient sources located at coastlines, are a technology still in the process of maturation, however, there is an expectation that this technology will need to be integrated into electrical distribution networks. Such integration will drive changes in the electric response of the distribution systems which may lead to harmful operating conditions. Power flow analysis is a tool used to reveal the steady-state operating condition of distribution systems and which could be extended to study and address the integration of PRO power units. To the best of the authors’ knowledge, such extension of power flow analysis has not yet been addressed in the literature. Accordingly, this paper comprehensively provides a model to evaluate the electric current and complex power produced by PRO power units. This model is directly embedded in the forward-backward sweep (FBS) method, extending the power flow analysis of electric distribution systems in this way so as to consider the integration of PRO power units. The resulting approach permits revealing of the steady-state operating response of distribution systems and the effects that may be driven by the integration of PRO power units, as corroborated through numerical results on a 14-node test distribution system.

Study Objectives Several birth cohorts have defined the pivotal role of early lower respiratory tract infections (LRTI) in the inception of pediatric respiratory conditions. However, the association between early LRTI and the development of obstructive sleep apnea (OSA) in children has not been established. Methods To investigate whether early LRTIs increase the risk of pediatric OSA, we analyzed clinical data in children followed during the first 5 years in the Boston Birth Cohort (n = 3114). Kaplan–Meier survival estimates and Cox proportional hazards models adjusted by pertinent covariates were used to evaluate the risk of OSA by the age of 5 years between children with LRTI during the first 2 years of life in comparison to those without LRTI during this period. Results Early life LRTI increased the risk of pediatric OSA independently of other pertinent covariates and risk factors (hazard ratio, 1.53; 95% CI, 1.15 to 2.05). Importantly, the association between LRTI and pediatric OSA was limited to LRTIs occurring during the first 2 years of life. Complementarily to this finding, we observed that children who had severe respiratory syncytial virus bronchiolitis during infancy had two times higher odds of OSA at 5 years in comparison with children without this exposure (odds ratio, 2.09; 95% CI, 1.12 to 3.88). Conclusions Children with severe LRTIs in early life have significantly increased risk of developing OSA during the first 5 years of life. Our results offer a new paradigm for investigating novel mechanisms and interventions targeting the early pathogenesis of OSA in the pediatric population.

Fertilization in angiosperms involves the germination of pollen on the stigma, followed by the extrusion of a pollen tube that elongates through the style and delivers two sperm cells to the embryo sac. Sexual selection could occur throughout this process when male gametophytes compete for fertilization. The strength of sexual selection during pollen competition should be affected by the number of genotypes deposited on the stigma. As increased self-fertilization reduces the number of mating partners, and the genetic diversity and heterozygosity of populations, it should thereby reduce the intensity of sexual selection during pollen competition. Despite the prevalence of mating system shifts, few studies have directly compared the molecular signatures of sexual selection during pollen competition in populations with different mating systems. Here we analyzed whole-genome sequences from natural populations of Arabis alpina, a species showing mating system variation across its distribution, to test whether shifts from cross- to self-fertilization result in molecular signatures consistent with sexual selection on genes involved in pollen competition. We found evidence for efficient purifying selection on genes expressed in vegetative pollen, and overall weaker selection on sperm-expressed genes. This pattern was robust when controlling for gene expression level and specificity. In agreement with the expectation that sexual selection intensifies under cross-fertilization, we found that the efficacy of purifying selection on male gametophyte-expressed genes was significantly stronger in genetically more diverse and outbred populations. Our results show that intra-sexual competition shapes the evolution of pollen-expressed genes, and that its strength fades with increasing self-fertilization rates.

Precision-cut lung slices (PCLS) are increasingly utilized for ex vivo disease modeling, but a high-resolution characterization of cellular phenotype stability in PCLS has not been reported. Comparing the single-cell transcriptomic profile of human PCLS after five days of culture to freshly isolated human lung tissue, we found striking changes in endothelial cell and alveolar epithelial cell programs, reflecting both injury and pathways activated in static culture, while immune cell frequencies and programs remained largely intact and similar to the native lung. These cellular dynamics should be considered when utilizing PCLS as a model of the human lung.

The present work shows the implementation of the Response Surface Methodology (RSM), fed by an experimental Central Composite Design (CCD) to find the conditions that allow maximizing the inhibition of the microorganism Staphylococcus aureus with nanoparticles of TiO2 silanized with 3-Aminopropyltriethoxysilane (APTES) and doped with Ag. In addition, Poly(lactic) acid composites were prepared with these Ag/TiO2 nanoparticles with the aim to confer their antimicrobial effect. The independent variables considered were pH, AgNO3/TiO2 ratio (% w/w), and TiO2 nanoparticles concentration (g/250 mL), and as the variable of response, the length of the diameter of the halo or zone of inhibition presented by the microorganism (mm). Statistical analysis found that maximization of S. aureus inhibition occurs at intermediate levels with a value of 10 for pH and 5 g of TiO2 solids, while for the concentration of AgNO3 high levels are required, greater than 10% w/w. Likewise, the statistical significance was determined using the Student's t-test and the p-value; it was found that the significant effect corresponds to the concentration of AgNO3, so a second experimental CCD design equirradial with two factors was considered, estimating AgNO3 concentration and TiO2 amount, the pH at constant 10 value. The second experimental design indicated that maximization in S. aureus inhibition occurs at an AgNO3 concentration between 20-25% w/w with high amounts of TiO2 solids (7-8 g), with a resulting zone of inhibition between 26-28 mm. The quadratic model obtained, which represents the relationship between the length of the zone of inhibition with the variables considered, shows an adjustment of experimental data with a coefficient of determination (R2) of 0.82.

Patients with chronic lung disease (CLD) have an increased risk for severe coronavirus disease-19 (COVID-19) and poor outcomes. Here, we analyze the transcriptomes of 611,398 single cells isolated from healthy and CLD lungs to identify molecular characteristics of lung cells that may account for worse COVID-19 outcomes in patients with chronic lung diseases. We observe a similar cellular distribution and relative expression of SARS-CoV-2 entry factors in control and CLD lungs. CLD AT2 cells express higher levels of genes linked directly to the efficiency of viral replication and the innate immune response. Additionally, we identify basal differences in inflammatory gene expression programs that highlight how CLD alters the inflammatory microenvironment encountered upon viral exposure to the peripheral lung. Over the past decade, "omics" approaches have advanced our understanding of the molecular programming of the airways in humans. Several studies have identified potential molecular mechanisms that contribute to early life epigenetic reprogramming, including DNA methylation, histone modifications, microRNAs, and the homeostasis of the respiratory mucosa (epithelial function and microbiota). Current evidence supports the notion that early infancy is characterized by heightened susceptibility to airway genetic reprogramming in response to the first exposures in life, some of which can have life-long consequences. Here, we summarize and analyze the latest insights from studies that support a novel epigenetic paradigm centered on human maturational and developmental programs including three cardinal elements: genes, environment, and developmental timing. The combination of these factors is likely responsible for the functional trajectory of the respiratory system at the molecular, functional, and clinical levels.

Background: Tick-borne diseases (TBD) and dirofilariasis are currently not under surveillance in most Latin American countries, and there is a significant lack of studies describing the current situation in most endemic areas, including Colombia. Seroprevalence studies are crucial for understanding the epidemiology of these vector-borne diseases.Methods: A serosurvey for TBD and dirofilariasis amongst 100 dogs was carried out in the municipality of Pereira, located in the Coffee-Triangle region, Colombia. Samples were tested using a rapid assay test system (SNAP® 4Dx®); based on an enzyme immunoassay technique‚ screening for antibodies to Anaplasma phagocytophilum/platys (sensitivity 99.1%)‚ Borrelia burgdorferi s.l. (98.8%), and Ehrlichia canis/ewingii (96.2%) by using specific antigens, and checking for Dirofilaria immitis antigen based on specific antibodies (99.2%). Bivariate analyses were performed on Stata®14, significant pResults: Global seroprevalence to the selected vector-borne pathogens was 74% (95%CI 65-83%). The highest seroprevalence was found for Ehrlichia canis/ewingii (74%), followed by Anaplasma phagocytophilum/platys (16%). Seropositivity for Borrelia spp. and Dirofilaria spp. was 0%. All Anaplasma spp. seropositive dogs showed co-detection of Ehrlichia spp. (16%). Seroprevalence was significantly higher among dogs from families of lower socioeconomic status / level (I, 86%), followed by level II (74%), and III (36%) (p=0.001); no seropositive dogs were found in for levels IV and V (higher socioeconomic status). All dogs exhibiting anorexia (12%) were invariably seropositive (100%) (p=0.029). Seroprevalence was higher amongst those showing mucocutaneous paleness (95%) compared to those without paleness (68%) (p=0.013) (OR=9.3; 95%CI 1.18-72.9). There was high variability in seroprevalence through the studied areas, ranging from 0% (La Libertad Park) up to Combia, Cesar Nader, Las Brisas and Saturno localities (100%) (p=0.033). Conclusions: Given the high seroprevalence obtained in an area with documented presence of ticks, there is a potential risk for zoonotic transmission to humans. Further seroprevalence studies in humans are needed to assess the prevalence of infections. Poverty is highly associated with these tick-borne pathogens in Pereira, as shown in the present study.

The extent to which wildfire adversely affects spotted owls (Strix occidentalis) is a key consideration for ecosystem restoration efforts in seasonally dry forests of the western United States. Recently, Jones et al. (2016) demonstrated that the 2014 King Fire (a “megafire”) adversely affected a population of individually-marked California spotted owls (S. o. occidentalis) monitored as part of a long-term demographic study in the Sierra Nevada, California, USA because territory occupancy declined substantially at territories burned at high-severity and GPS-tagged spotted owls avoided large patches of high-severity fire. Hanson et al. (2018) attempted to reassess changes in territory occupancy of the Jones et al. (2016) study population and claimed that occupancy declined as a result of post-fire salvage logging not fire per se and suggested that the avoidance of GPS-marked owls from areas that burned at high-severity was due to post-fire logging rather than a response to high-severity fire. Here, we demonstrate that Hanson et al. (2018) used erroneous data, inadequate statistical analyses and faulty inferences to reach their conclusion that the King Fire did not affect spotted owls and, more broadly, that large, high-severity fires do not pose risks to spotted owls in western North American dry forest ecosystems. We also provide further evidence indicating that the King Fire exerted a clear and significant negative effect on our marked study population of spotted owls. Collectively, the additional evidence presented here and in Jones et al. (2016) suggests that large, high-severity fires can pose a threat to spotted owls and that restoration of natural low- to mixed-severity frequent fire regimes would likely benefit both old-forest species and dry forest ecosystems in this era of climate change. Meeting these dual objectives of species conservation and forest restoration will be complex but it is made more challenging by faulty science that does not acknowledge the full range of wildfire effects on spotted owls.

An important challenge in the wireless power transfer is the sizing of the coils for transmission and energy reception. The output power in a wireless power transfer system can be determined by analytical formulation; however, the effects of using some geometry or material for the transmitter and receiver coil assembly are not easily quantifiable by the analytical method. This work quantifies by numerical simulation the impact on output power changing the separation between the transmitter coil and receiver, materials, geometries and number of turns. The quantities obtained allowed to determine a model for coupling coefficient to compute optimal frequency for the power transfer. The analysis and evaluation of these parameters were developed through an electromagnetic simulator with finite element; the work considers planar circular and square coils. Experimental results were obtained in order to validate the proposed model to compute the wireless power transferred. The document contributes to identify the important parameters to be controlled or modified in the coils with the purpose to optimize the power transmission for a wireless system.
