Abstract

Intravascular brachytherapy is a novel therapy for preventing the restenosis of coronary artery by use of low-dose irradiation. JAERI and Kyoto University have been developing sup 1 sup 9 sup 2 Ir radiation sources by the cooperative research project entitled as 'The research on safety and effectiveness of the intravascular brachytherapy for preventing restenosis of the coronary artery disease' since 1998. The radiation source was introduced into the stenosis through a catheter (a guide-tube to insert directly into vascular) to irradiate the diseased part. Ten sup 1 sup 9 sup 2 Ir seed sources (phi 0.4 mm x 2.5 mm) were positioned between nylon spacers (phi 0.3 mm x 1.0 mm) in a flexible covering tube and the tube was plugged with a core-wire; the tube was shrunk to fix the inside materials and the size is 0.46 mm in diameter and 3 m in length. The physically optimal design was determined to insert the radiation source easily into vascular and to get the dose uniformity in the diseased part. The production method of the radiation source, which is practical to use in the clinics was also established.

Abstract

Topological photonics emerged as a novel route to engineer the flow of light. Topologically protected photonic edge modes, which are supported at the perimeters of topologically nontrivial insulating bulk structures, are of particular interest as they may enable low-loss optical waveguides immune to structural disorder. Very recently, there has been a sharp rise of interest in introducing gain materials into such topological photonic structures, primarily aiming at revolutionizing semiconductor lasers with the aid of physical mechanisms existing in topological physics. Examples of remarkable realizations are topological lasers with unidirectional light output under time-reversal symmetry breaking and topologically protected polariton and micro/nanocavity lasers. Moreover, the introduction of gain and loss provides a fascinating playground to explore novel topological phases, which are in close relevance to non-Hermitian and parity-time symmetric quantum physics and are, in general, difficult to access using fermionic condensed matter systems. Here, we review the cutting-edge research on active topological photonics, in which optical gain plays a pivotal role. We discuss recent realizations of topological lasers of various kinds, together with the underlying physics explaining the emergence of topological edge modes. In such demonstrations, the optical modes of the topological lasers are determined by the dielectric structures and support lasing oscillation with the help of optical gain. We also address recent research on topological photonic systems in which gain and loss, themselves, essentially influence topological properties of the bulk systems. We believe that active topological photonics provides powerful means to advance micro/nanophotonics systems for diverse applications and topological physics, itself, as well.

208. A functional SNP upstream of the beta-2 adrenergic receptor gene (ADRB2) is associated with obesity in Oceanic populations

Abstract

OBJECTIVE:Obesity is a growing health concern in the Oceanic populations. To investigate the genetic factors associated with adult obesity in the Oceanic populations, the association of single nucleotide polymorphisms (SNPs) of the beta-2 adrenergic receptor (ADRB2) gene with obesity was examined in 694 adults living in Tonga and Solomon Islands. RESULTS:A screening for variation in 16 Oceanic subjects detected 17 SNPs in the entire region of ADRB2, of which nine SNPs including two non-synonymous ones, rs1042713 (Arg16Gly) and rs1042714 (Gln27Glu), were further genotyped for all subjects. The rs34623097-A allele, at a SNP located upstream of ADRB2, showed the strongest association with risk for obesity in a logistic regression analysis adjusted for age, sex, and population (P=5.6 × 10^<−4>, odds ratio [OR]=2.5, 95% confidence interval [CI]=1.5–4.2). The 27Glu was also significantly associated with obesity in the single-point association analysis (P=0.013, OR=2.0, 95%CI=1.2–3.4); however, this association was no longer significant after adjustment for rs34623097 since these SNPs were in linkage disequilibrium with each other. A copy of the obesity-risk allele, rs34623097-A, led to a 1.6 kg/m^2 increase in body mass index (BMI; defined as weight in kilograms divided by height in meters squared) (P=0.0019). A luciferase reporter assay indicated that rs34623097-A reduced the transcriptional activity of the luciferase reporter gene by approximately 10% compared with rs34623097-G. An electrophoretic mobility shift assay demonstrated that rs34623097 modulated the binding affinity with nuclear factors. An evolutionary analysis implies that a G>A mutation at rs34623097 occurred in the Neandertal genome and then the rs34623097-A allele flowed into the ancestors of present-day humans. CONCLUSION:The present results suggest that rs34623097-A, which would lead to lower expression of ADRB2, contributes to the onset of obesity in the Oceanic populations.

Abstract

Acute lesions in the dorsal skin topically applied with T-2 toxin (10 p1 of 0.5 mg/ml-solution to lcm2) were examined in Wistar-derived hypotrichotic WBNIILA-Ht rats up to 24 hours after treatment (24HAT). In the epidermis, depression of basal cell proliferating activity was detected at 3HAT by immunostaining for proliferating cell nuclear antigen (PCNA), and the percentage of PCNA-positive basal cells decreased thereafter. At 12HAT, in addition to intracytoplasmic edema of spinous cells, acidophilic degeneration of basal cells characterized by shrinkage of cell body with acidophilic cytoplasm and pyknotic or karyorrhectic nuclei became prominent. Most of these nuclei were positive for TUNEL which is a widely used immunostaining for the in situ detection of fragmented DNA, i.e. apoptosis, and the percentage of TUNELpositive basal cells increased thereafter. The nuclei of these basal cells also showed ultrastructural changes characteristic for apoptosis. On the other hand, in the dermis, infiltration of inflammatory cells including mast cells started at 3HAT and increased thereafter. In addition, capillary and small vessel endothelial degeneration developed at 6HAT and progressed thereafter. These results suggest that T-2 toxin directly affects the epidermis and produces apoptosis in basal cells.

Abstract

A prediction method of the critical current-bending strain relation of the Bi2223 composite tape, which was bent at room temperature and cooled down to 77 K for measurement of critical current, was presented. The present method consisted of (a) measurement of residual strain of Bi2223 filaments at room temperature in the sample length direction by the X-ray diffraction method, (b) estimation of tensile fracture strain of the filaments from the analysis of the load-strain curve at room temperature, (c) measurement of geometrical factors such as the thickness of the sample and shape of the core by observation of the cross-section with optical microscope, and (d) calculation of the critical current as a function of bending strain by using the measured parameters mentioned in (a)-(c). The predicted variation of critical current with bending strain by the present approach described well the experimental one.

Abstract

Mechanical and electromagnetic stresses are exerted on Bi2223/Ag/Ag alloy superconducting composite tapes during fabrication/winding and operation, which cause reduction in critical current when the Bi2223 filaments are damaged. In the damage process, the thermally induced residual strain and fracture strain of the Bi2223 filaments play a dominant role. The aim of the present work was to propose a comprehensive method for estimation of these strain values and a quantitative description method of the relation of critical current to the applied bending/tensile strain, and to examine the accuracy of the method in comparison with the experimental results. The residual strain of Bi2223 filaments in the composite tape was measured by the x-ray diffraction method. The measured residual strain value was used for analysis of the load-strain curve, from which the intrinsic fracture strain of filaments was estimated. The relation of critical current to applied bending/tensile strain was predicted by the proposed calculation procedure, in which the estimated strain values were input. The predicted critical current-applied strain relation agreed well with the experimental results, suggesting that the present method is a useful tool for prediction/description of tensile/bending applied strain dependence of critical current of multifilamentary-type conductors.

Abstract

The detailed histology of the dorsal skin of Wistar-derived hypotrichotic WBNIILA-Ht rats (HtRs) was examined in their developing stage (at 3 and 7 weeks of age) using Wistar rats (WRs) as controls. As a result, except for the existence of time-lag in the hair follicular cycle and some quantitative differences, there were no essential qualitative differences between the two strains. The follicular epithelial cell number and the amounts of tonofilaments and trichohyalin granules were less in HtRs than in WRs in the same phase of hair follicular cycle. In addition, the diameter of hair shaft was significantly larger in WRs than in HtRs at 7 weeks of age. The number of proliferating cell nuclear antigen (PCNA)-positive epidermal basal cells decreased in HtRs and increased in WRs from 3 to 7 weeks of age, respectively. Apoptosis was sparsely observed in the sebaceous gland epithelia1 cells, keratinizing portion of hair cuticules and inner root sheath in both strains, but the frequency of apoptosis in the hair follicle was somewhat higher in 7-week-old HtRs. HtRs are considered to be useful experimental animals for dermatotoxicological studies.

Abstract

Background The quail and chicken major histocompatibility complex (Mhc) genomic regions have a similar overall organization but differ markedly in that the quail has an expanded number of duplicated class I, class IIB, natural killer (NK)-receptor-like, lectin-like and BG genes. Therefore, the elucidation of genetic factors that contribute to the greater Mhc diversity in the quail would help to establish it as a model experimental animal in the investigation of avian Mhc associated diseases. Aims and approaches The main aim here was to characterize the genetic and genomic features of the transcribed major quail MhcIIB (CojaIIB) region that is located between the Tapasin and BRD2 genes, and to compare our findings to the available information for the chicken MhcIIB (BLB). We used four approaches in the study of the quail MhcIIB region, (1) haplotype analyses with polymorphic loci, (2) cloning and sequencing of the RT-PCR CojaIIB products from individuals with different haplotypes, (3) genomic sequencing of the CojaIIB region from the individuals with the different haplotypes, and (4) phylogenetic and duplication analysis to explain the variability of the region between the quail and the chicken. Results Our results show that the Tapasin-BRD2 segment of the quail Mhc is highly variable in length and in gene transcription intensity and content. Haplotypic sequences were found to vary in length between 4 to 11 kb. Tapasin-BRD2 segments contain one or two major transcribed CojaIIBs that were probably generated by segmental duplications involving c-type lectin-like genes and NK receptor-like genes, gene fusions between two CojaIIBs and transpositions between the major and minor CojaIIB segments. The relative evolutionary speed for generating the MhcIIBs genomic structures from the ancestral BLB2 was estimated to be two times faster in the quail than in the chicken after their separation from a common ancestor.
