Coffee melanoidins are generated by the Maillard reaction during the thermal processes occurring in the journey of coffee from the plant to the cup (during drying and roasting). Melanoidins, the brown pigments formed as the end products of this reaction, have been reported in cascara, silverskin, spent coffee grounds, and coffee brew. The latter is one of the main natural sources of melanoidins of the daily diet worldwide. However, their presence in coffee by-products has been recently described. These complex macromolecules possess multiple health-promoting properties, such as antioxidant, anti-inflammatory, dietary fiber effect, and prebiotic capacity, which make them very interesting from a nutritional point of view. In addition, they have a great impact on the sensory profile of foods and their acceptance by the consumers. The present study is a descriptive, narrative, mini-review about the nature, structure, digestibility, properties (sensory, nutritional, and health-promoting), safety and regulatory status of melanoidins from the coffee brew and its by-products with a special emphasis on the latter.

Aim: Mediterranean terrestrial ecosystems serve as reference laboratories for the investigation of global change because of their transitional climate, the high spatiotemporal variability of their environmental conditions, a rich and unique biodiversity and a wide range of socio-economic conditions. As scientific development and environmental pressures increase, it is increasingly necessary to evaluate recent progress and to challenge research priorities in the face of global change. Location: Mediterranean terrestrial ecosystems. Methods: This article revisits the research priorities proposed in a 1998 assessment. Results: A new set of research priorities is proposed: (1) to establish the role of the landscape mosaic on fire-spread; (2) to further research the combined effect of different drivers on pest expansion; (3) to address the interaction between drivers of global change and recent forest management practices; (4) to obtain more realistic information on the impacts of global change and ecosystem services; (5) to assess forest mortality events associated with climatic extremes; (6) to focus global change research on identifying and managing vulnerable areas; (7) to use the functional traits concept to study resilience after disturbance; (8) to study the relationship between genotypic and phenotypic diversity as a source of forest resilience; (9) to understand the balance between C storage and water resources; (10) to analyse the interplay between landscape-scale processes and biodiversity conservation; (11) to refine models by including interactions between drivers and socio-economic contexts; (12) to understand forest-atmosphere feedbacks; (13) to represent key mechanisms linking plant hydraulics with landscape hydrology. Main conclusions: (1) The interactive nature of different global change drivers remains poorly understood. (2) There is a critical need for the rapid development of regional- and global-scale models that are more tightly connected with large-scale experiments, data networks and management practice. (3) More attention should be directed to drought-related forest decline and the current relevance of historical land use.

Introduction: Fanconi anemia (FA) is an inherited disorder characterized by bone marrow failure, congenital malformations, and predisposition to malignancies. Alterations in hematopoietic stem cells (HSC) have been reported, but little is known regarding the bone marrow (BM) stroma. Thus, the characterization of Mesenchymal Stromal Cells (MSC) would help to elucidate their involvement in the BM failure. Methods: We characterized MSCs of 28 FA patients (FA-MSC) before and after treatment (hematopoietic stem cell transplantation, HSCT; or gene therapy, GT). Phenotypic and functional properties were analyzed and compared with MSCs expanded from 26 healthy donors (HD-MSCs). FA-MSCs were genetically characterized through, mitomycin C-test and chimerism analysis. Furthermore, RNA-seq profiling was used to identify dysregulated metabolic pathways. Results: Overall, FA-MSC had the same phenotypic and functional characteristics as HD-MSC. Of note, MSC-GT had a lower clonogenic efficiency. These findings were not confirmed in the whole FA patients' cohort. Transcriptomic profiling identified dysregulation in HSC self-maintenance pathways in FA-MSC (HOX), and was confirmed by real-time quantitative polymerase chain reaction (RT-qPCR). Discussion: Our study provides a comprehensive characterization of FA-MSCs, including for the first time MSC-GT and constitutes the largest series published to date. Interestingly, transcript profiling revealed dysregulation of metabolic pathways related to HSC self-maintenance. Taken together, our results or findings provide new insights into the pathophysiology of the disease, although whether these niche defects are involved in the hematopoietic defects seen of FA deserves further investigation. This study presents U-Pb zircon ages and Lu-Hf zircon isotope data for Cretaceous-Paleocene plutonic rocks along a W-E transect in northwestern Mexico. These data are combined with tectonic reconstruction that restores Late Cenozoic extensional deformation and shows the position of magmatism at 36 Ma. Zircon U-Pb ages results span from 142 to 58 Ma and demonstrate that the continental arc migrated northeastward at 1–2.5 km/Myr. These rates are slower than previously interpreted, but consistent with landward arc migration rates observed in the Andes. Weighted mean initial epsilon hafnium (εHf(t)) values of plutonic rocks along the transect range from + 8.8 to −9.1. The heterogeneity in the zircon εHf(t) is spatially related to the pre-Cretaceous basement provinces that the intrusive rocks were emplaced into. Zircon εHf(t) values of western Baja California display positive values ranging from + 8.8 to + 2.6 suggesting they were formed from a moderately depleted mantle and were emplaced into the Guerrero-Alisitos-Vizcaino terrane. Zircon εHf(t) values in the eastern part of Baja California and most of Sonora are heterogeneous ranging between −0.7 and −9.1 and may be formed from a relatively slightly more evolved mantle source and end up more evolved after crustal assimilation of metasediments. Zircon εHf(t) values ranging from + 8.7 to + 2.9 in Chihuahua are consistent with a depleted-mantle derived melt and assimilation of Grenville lithospheric province. Our results highlight how Hf isotopic signatures help to constrain the pre-Cretaceous basement configuration in northwestern Mexico despite the few exposed outcrops along the transect. The digital maturity of universities has been a key element in coping with the forced situation brought about by COVID-19 and in subsequent years. This research paper presents a case study of a private Spanish university that aims to analyze the challenges and opportunities that have arisen after the pandemic in terms of resource management and training for teaching innovation and digitization. The nature of this study is qualitative, in which nine interviews were conducted with managers from different faculties and those responsible for Information and Communication Technologies. After analyzing the data with Atlas.ti, four categories were established (technological resources and devices, challenges, opportunities, and others). The study concludes by discussing the commitment to online training, the popularization of videoconferencing tools for teaching sessions, meetings, or synchronous tutorials, and the acceleration of digital transformation, among others. This letter to the editor discusses the case of an 8-month-old female who presented with severe thrombocytopenia. The patient had developed spontaneous bruising and bleeding of the mucous membranes, and initial tests revealed severe thrombocytopenia, mild neutropenia, and anemia. Despite initial treatment with platelet transfusion and immunoglobulin therapy, the patient's condition did not improve. A bone marrow study was conducted, and the patient was eventually diagnosed with autoimmune myelofibrosis (AIMF). Treatment with sirolimus resulted in the resolution of thrombocytopenia and normalization of blood cell counts. The authors emphasize the importance of an appropriate diagnostic work-up for immune cytopenias in children and suggest that sirolimus may be a promising therapeutic approach for similar cases of AIMF in pediatric patients. Abstract Background Pathogenic variants (PVs) of BRCA genes entail a lifetime risk of developing breast cancer in 50–85% of carriers. Their prevalence in different populations has been previously reported. However, there is scarce information regarding the most common PVs of these genes in Latin-Americans. This study identified BRCA1 and BRCA2 PV frequency in a high-risk female population from Northeastern Mexico and determined the association of these mutations with the patients’ clinical and pathological characteristics. Methods Women were divided into three groups: aged ≤ 40 years at diagnosis and/or risk factors for hereditary breast cancer (n = 101), aged > 50 years with sporadic breast cancer (n = 22), and healthy women (n = 72). Their DNA was obtained from peripheral blood samples and the variants were examined by next-generation sequencing with Ion AmpliSeq BRCA1 and BRCA2 Panel using next-generation sequencing. Results PVs were detected in 13.8% group 1 patients (BRCA1, 12 patients; BRCA2, 2 patients). Only two patients in group 2 and none in group 3 exhibited BRCA1 PVs. Variants of uncertain significance were reported in 15.8% patients (n = 16). In group 1, patients with the triple-negative subtype, PV frequency was 40% (12/30). Breast cancer prevalence in young women examined in this study was higher than that reported by the National Cancer Institute Surveillance, Epidemiology (15.5% vs. 5.5%, respectively). Conclusions The detected BRCA1 and BRCA2 PV frequency was similar to that reported in other populations. Our results indicate that clinical data should be evaluated before genetic testing and highly recommend genetic testing in patients with the triple-negative subtype and other clinical aspects.

O artigo apresenta uma pesquisa que se realizou a partir de um projeto constituído por aulas planejadas e desenvolvidas com a utilização do Facebook como espaço de aprendizagem da Língua Estrangeira. A experiência foi realizada em um curso de Língua Inglesa e selecionou um grupo de vinte estudantes para participarem do estudo. O trabalho foi motivado pela seguinte pergunta: É possível desenvolver na aula de língua estrangeira um processo de interação e aprendizagem significativo através da rede social Facebook? Utilizou-se nesta pesquisa a abordagem qualitativa aliada à pesquisa-ação. Espera-se, portanto, com a apresentação desta pesquisa, incentivar a reflexão sobre a prática pedagógica com o uso das redes sociais e de outros espaços interativos da Web para o Ensino de Línguas Estrangeiras.

Honey consumption and imports have increased in recent years, and it is considered by consumers to be a healthy alternative to more commonly used sweeteners. Honey contains a mixture of polyphenols and antioxidant compounds, and the botanical origin and geographical area of collection play an important role on its chemical composition. The present study investigated the physicochemical properties, total phenolic content and antioxidant capacity of Spanish thyme honey and chestnut honey, and their mixtures with royal jelly (2% and 10%) and propolis (2% and 10%). The analysis of the physicochemical parameters of both honey samples showed values within the established limits. Propolis showed the highest value of total phenolic content (17.21–266.83 mg GAE/100 g) and antioxidant capacity (DPPH, ORAC and ABTS assays; 0.63–24.10 µg eq. Tx/g, 1.61–40.82 µg eq. Tx/g and 1.89–68.54 µg eq. Tx/g, respectively), and significantly reduced ROS production in human hepatoma cells. In addition, mixtures of honey with 10% of propolis improved the results obtained with natural honey, increasing the value of total phenolic content and antioxidant capacity. A significant positive correlation was observed between total phenolic compounds and antioxidant capacity. Therefore, the antioxidant capacity could be attributed to the phenolic compounds present in the samples, at least partially. In conclusion, our results indicated that thyme and chestnut honey supplemented with propolis can be an excellent natural source of antioxidants and could be incorporated as a potential food ingredient with biological properties of technological interest, added as a preservative. Moreover, these mixtures could be used as natural sweeteners enriched in antioxidants and other bioactive compounds.

The concentration of CO2 is relatively large in poultry farms and high accumulations of this gas reduce animal welfare. Good control of its concentration is crucial for the health of the animals. The vocalizations of the chickens can show their level of well-being linked to the presence of carbon dioxide. An audio recording system was implemented and audio raw data was processed to extract acoustical features from four cycles of forty days, three of them from the same farm. This research aims to find the most relevant acoustic features extracted from the broiler’s calls that are related to the CO2 concentration and that could help to automate procedures. The results are encouraging since MFCC 6, 9, 4 and 3 are the most important features that relate the vocalizations of the chickens to the gas concentration, furthermore there is a clear and more similar representativeness trend during birds’ life period from day 15 to day 40.

Many species cannot either migrate or adapt at the rate of temperature increases due to climate warming. Therefore, they need active conservation strategies to avoid extinction. Facilitated adaptation actions, such assisted gene flow, aim at the increase of the evolutionary resilience of species affected by global change. In elevational gradients, marginal populations at the lower elevation edges are experiencing earlier snowmelt and higher temperatures, which force them to adapt to the new conditions by modifying their phenology. In this context, advancing the onset of flowering and seed germination times are crucial to ensure reproductive success and increase seedling survival prior to summer drought. Assisted gene flow may bring adaptive alleles and increase genetic diversity that can help throughout ontogeny. The main aim of this work is to assess the effects that different gene flow treatments could have on the desired trait changes in marginal populations. Accordingly, we established a common garden experiment in which we assayed four different gene flow treatments between Silene ciliata Pourr. (Caryophyllaceae) populations located in similar and different elevation edges, belonging to the same and different mountains. As a control treatment, within-population crosses of low elevation edge populations were performed. The resulting seeds were sown and the germination and flowering onset dates of the resulting plants recorded, as well as the seedling survival. Gene flow between populations falling on the same mountain and same elevation and gene flow from high-elevation populations from a different mountain to low-elevation populations advanced seed germination time with respect to control crosses. No significant effects of gene flow on seedling survival were found. All the gene flow treatments delayed the onset of flowering with respect to control crosses and this effect was more pronounced in among-mountain gene flows. The results of this study highlight two important issues that should be thoroughly studied before attempting to apply assisted gene flow in practical conservation situations. Firstly, among-populations gene flow can trigger different responses in crucial traits throughout the ontogeny of plant species. Secondly, the population provenance of gene flow is determinant and plays a significant role on the effects of gene flow.

Abstract Restoration of wild plant populations in a changing environment is not a simple task, and it becomes increasingly complex and challenging in the context of climate change. In the present study, a spatiotemporal approach was used to delimit provisional seed transfer zones (STZs), for restoration activities involving two wild perennial grasses in Argentina. Ecogeographical land characterization maps were built specifically for the two species of Trichloris genus for the present bioclimatic conditions to define the different STZs; then, they were projected for two future climate scenarios (2050 and 2070). This allowed us to identify populations occurring in cells that underwent changes in ecogeographical categories, potential future beneficiaries of germplasm, and populations that could be used as corresponding source populations under the present conditions. Finally, the conservation status of populations was surveyed and a germplasm collection for future restoration activities was proposed. For the 2050 scenario, for Trichloris crinita, 32 potential beneficiary populations belonging to nine changing categories were identified, whereas for Trichloris pluriflora, four changing categories determined seven beneficiary populations. The number of changing categories and, consequently, the beneficiary populations increased in the 2070 scenario in both species. Several potential seed source populations are already stored in Argentine germplasm banks, but there is little representation of potential beneficiary populations. Spatial locations of still non‐conserved populations, both seed sources and beneficiaries, were prioritized for future germplasm collecting missions. This novel approach used for Trichloris spp. can also be adopted for other species for collection and/or restoration purposes in the light of climate change.

El S de la provincia de Santa Fe y el NO de la provincia de Buenos Aires forman parte del Mar de Arena Pampeano (Sistema Eólico Pampeano), que ha sufrido una secuencia de episodios áridos y húmedos a lo largo de los últimos períodos del Pleistoceno. El paisaje actual resulta fundamentalmente de la influencia del período húmedo del Estadio Isotópico 3 (EIO 3; 64-36 ka) y de la actividad eólica durante un clima seco del Holoceno tardío (3,5-1,4 ka). La secuencia de eventos sedimentológicos y geomorfológicos fue la siguiente: Durante el período húmedo del EIO 3 se desarrolló una red fluvial de llanura, que actualmente forma la alta cuenca del Río Salado de Buenos Aires, labrada sobre sedimentos de la Fm Carcarañá. Posteriormente se depositó la Formación Teodelina, por sedimentación eólica. Esta tiene 10 a 12 m de espesor típico y unos 5 m en áreas sometidas a erosión; está compuesta por limo grueso arenoso y arena fina limosa, con modas principales entre 125-250 y 53-62 ?m, color 10YR 6/4 (marrón amarillento) y similares. La composición mineralógica de la fracción entre 53 y 62 ?m está dominada por vidrio volcánico, con feldespato, cuarzo y alteritas como accesorios. La redondez y esfericidad de los granos varían entre bajas y moderadas, con dos poblaciones. En el Pleistoceno final ocurrió un episodio eólico predominantemente erosivo, dominado por vientos del oeste que labraron cientos de depresiones de miles de metros de extensión. Ocurrió una sedimentación discontinua de loess arenoso de hasta 2 m de espesor con granulometría bimodal. El período Hypsithermal del Holoceno Medio (8,5-3,5 ka) estuvo caracterizado por un clima cálido y húmedo con régimen údico, que generó un suelo en los terrenos loéssicos y produjo el ascenso del nivel del agua en las lagunas y pantanos. El Holoceno tardío, entre 3,5 y 1,4 ka, estuvo dominado por un clima seco, con formación de campos de dunas denominadas aquí Formación San Gregorio. Está formada por arena muy fina a fina, suelta, masiva, color marrón amarillento, y compuesta por trizas vítreas, alteritas y feldespatos como componentes principales y cuarzo como componente secundario; los minerales pesados son de procedencia serrana. La época actual está caracterizada por un exceso de agua en el paisaje; el proceso sedimentológico más relevante es la movilización de grandes volúmenes de sales disueltas, particularmente cloruros y sulfatos.
