Abstract: International audience; Diversity of parasitoids species remains questioning in several ecosystems poorly surveyed. The present study is focused on dipteran parasitoids species infecting orthopteran in forest-steppe ecosystem of Mongolia. Among a total of 420 hosts from four species, 20 were found to be infected and 200 larvae of dipteran parasitoids recovered. Genetic studies on a subset of larvae shows the presence of up to seven taxa whose none is represented in databases (although 13 species were previously reported in Mongolia), indicating a large and unexplored diversity of dipteran parasitoid in Mongolia. Further studies will be focused in establishing of this diversity and the determination of the species.

Abstract: International audience; Reports of atypical human cases of Trypanosoma lewisi or T. lewisi-like and Trypanosoma evansi infections have increased in South-East Asia, urging to investigate the possible links between humans, animal reservoirs and habitats. We tested how habitat structure affects the infection by Trypanosoma species of common murine rodents, inhabiting human-dominated landscapes in South East Asia. For this, we used geo-referenced data of rodents investigated for Trypanosoma infection and land cover maps produced for seven study sites in Thailand, Cambodia and Lao PDR. High prevalence of infection by T. lewisi was observed in rodents living near human settlement and in areas with high cover of built-up habitat, while the infection of rodents by T. evansi was explained by increased landscape patchiness and high cover of rain-fed agriculture lands. These results suggest a likely role of wild rodents as reservoir and possible source of atypical human infection by animal trypanosomes.

Abstract: International audience; BACKGROUND:The process and effectiveness of knowledge translation (KT) interventions targeting policymakers are rarely reported. In Cambodia, a low-income country (LIC), an intervention aiming to provide evidence-based knowledge on pneumonia to health authorities was developed to help update pediatric and adult national clinical guidelines. Through a case study, we assessed the effectiveness of this KT intervention, with the goal of identifying the barriers to KT and suggest strategies to facilitate KT in similar settings.METHODS:An extensive search for all relevant sources of data documenting the processes of updating adult and pediatric pneumonia guidelines was done. Documents included among others, reports, meeting minutes, and email correspondences. The study was conducted in successive phases: an appraisal of the content of both adult and pediatric pneumonia guidelines; an appraisal of the quality of guidelines by independent experts, using the AGREE-II instrument; a description and modeling of the KT process within the guidelines updating system, using the Unified Modeling Language (UML) tools 2.2; and the listing of the barriers and facilitators to KT we identified during the study.RESULTS:The first appraisal showed that the integration of the KT key messages in pediatric and adult guidelines varied with a better efficiency in the pediatric guidelines. The overall AGREE-II quality assessments scored 37% and 44% for adult and pediatric guidelines, respectively. Scores were lowest for the domains of 'rigor of development' and 'editorial independence.' The UML analysis highlighted that time frames and constraints of the involved stakeholders greatly differed and that there were several missed opportunities to translate on evidence into the adult pneumonia guideline. Seventeen facilitating factors and 18 potential barriers to KT were identified. Main barriers were related to the absence of a clear mandate from the Ministry of Health for the researchers and to a lack of synchronization between knowledge production and policy-making.CONCLUSIONS:Study findings suggest that stakeholders, both researchers and policy makers planning to update clinical guidelines in LIC may need methodological support to overcome the expected barriers.

Abstract: Patient derived xenografts (PDXs) are increasingly appreciated models in cancer research, particularly for preclinical testing, as they reflect the patient's tumor biology more accurately than cancer cell lines. We have established a collection of 20 breast PDXs and characterized their biological and clinical features, as well as their genetic stability. While most PDXs originated from triple negative breast cancers (70%), our collection comprised five ER + cases (25%). Remarkably, the tumors that produced PDXs derived from a subset of aggressive breast cancers with a high proportion of grade 3 tumors and reduced recurrence‐free survival. Consistent with this, we found significant differences between the transcriptomic signatures of tumors that produced a PDX (Take) and those that did not (No Take). The PDXs faithfully recapitulate the histological features of their primary tumors, and retain an excellent conservation of molecular classification assignment and Copy Number Change (CNC). Furthermore, the CNC profiles of different PDXs established from the same tumor overlap significantly. However, a small fraction of CNCs in the primary tumor that correspond to oligoclonal events were gradually lost during sequential passaging, suggesting that the PDXs' genetic structure eventually stabilizes around a dominant clone present in the tumor of origin. Finally, de novo occurring genetic events covering up to 9% of the genome were found in only a minority of the PDXs, showing that PDXs have limited genetic instability. These data show that breast cancer PDXs represent a subset of aggressive tumors prone to relapse, and that despite of an excellent conservation of original features, they remain genetically dynamic elements., Highlights A collection of breast cancer PDXs is described.PDXs represent a subset of clinically aggressive breast tumors.Most PDXs derive from a dominant genetic subclone present in the tumor of origin.PDXs are genetically dynamic and susceptible to slow drift.

Abstract: PSA (Promastigote Surface Antigen) belongs to a family of membrane-bound and secreted proteins present in several Leishmania (L.) species. PSA is recognized by human Th1 cells and provides a high degree of protection in vaccinated mice. We evaluated humoral and cellular immune responses induced by a L. amazonensis PSA protein (LaPSA-38S) produced in a L. tarentolae expression system. This was done in individuals cured of cutaneous leishmaniasis due to L. major (CCLm) or L. braziliensis (CCLb) or visceral leishmaniasis due to L. donovani (CVLd) and in healthy individuals. Healthy individuals were subdivided into immune (HHR-Lm and HHR-Li: Healthy High Responders living in an endemic area for L. major or L. infantum infection) or non immune/naive individuals (HLR: Healthy Low Responders), depending on whether they produce high or low levels of IFN-γ in response to Leishmania soluble antigen. Low levels of total IgG antibodies to LaPSA-38S were detected in sera from the studied groups. Interestingly, LaPSA-38S induced specific and significant levels of IFN-γ, granzyme B and IL-10 in CCLm, HHR-Lm and HHR-Li groups, with HHR-Li group producing TNF-α in more. No significant cytokine response was observed in individuals immune to L. braziliensis or L. donovani infection. Phenotypic analysis showed a significant increase in CD4+ T cells producing IFN-γ after LaPSA-38S stimulation, in CCLm. A high positive correlation was observed between the percentage of IFN-γ-producing CD4+ T cells and the released IFN-γ. We showed that the LaPSA-38S protein was able to induce a mixed Th1 and Th2/Treg cytokine response in individuals with immunity to L. major or L. infantum infection indicating that it may be exploited as a vaccine candidate. We also showed, to our knowledge for the first time, the capacity of Leishmania PSA protein to induce granzyme B production in humans with immunity to L. major and L. infantum infection. Abstract: Understanding the adaptive response to environmental fluctuations represents a central issue in evolutionary biology. Population admixture between divergent ancestries has often been considered as an efficient short-term adaptation strategy. Cattle populations from the West African Bos taurus × Bos indicus hybrid zone represent a valuable resource to characterize the effect of such adaptive admixture at the genome level. We here provide a detailed assessment of the global and local genome ancestries of the Borgou breed, one of the most representative cattle of this hybrid zone. We analysed a large data set consisting of 38 100 SNPs genotyped on 203 Borgou and 591 individuals representative of all the different cattle ancestries. At the global genomic level, we show that Borgou is a stabilized admixed breed whose origin (c. 130 years ago) traces back to the great African rinderpest pandemic, several centuries after the last admixture events, the West African zebus originate from (c. 500 years ago). To identify footprints of adaptive admixture, we combined the identification of signatures of selection and the functional annotation of the underlying genes using systems biology tools. The detection of the SILV coat coloration gene likely under artificial selection may be viewed as a validation of our approach. Overall, our results suggest that the long-term presence of pathogens and the intermediate environmental conditions are the main acting selective pressures. Our analytical framework can be extended to other model or nonmodel species to understand the process that shapes the patterns of genetic variability in hybrid zones.

Abstract: In West Africa, Trypanosoma brucei gambiense, causing human African trypanosomiasis (HAT), is associated with a great diversity of infection outcomes. In addition to patients who can be diagnosed in the early hemolymphatic phase (stage 1) or meningoencephalitic phase (stage 2), a number of individuals can mount long-lasting specific serological responses while the results of microscopic investigations are negative (SERO TL+). Evidence is now increasing to indicate that these are asymptomatic subjects with low-grade parasitemia. The goal of our study was to investigate the type of immune response occurring in these “trypanotolerant” subjects. Cytokines levels were measured in healthy endemic controls (n = 40), stage 1 (n = 10), early stage 2 (n = 19), and late stage 2 patients (n = 23) and in a cohort of SERO TL+ individuals (n = 60) who were followed up for two years to assess the evolution of their parasitological and serological status. In contrast to HAT patients which T-cell responses appeared to be activated with increased levels of IL2, IL4, and IL10, SERO TL+ exhibited high levels of proinflammatory cytokines (IL6, IL8 and TNFα) and an almost absence of IL12p70. In SERO TL+, high levels of IL10 and low levels of TNFα were associated with an increased risk of developing HAT whereas high levels of IL8 predicted that serology would become negative. Further studies using high throughput technologies, hopefully will provide a more detailed view of the critical molecules or pathways underlying the trypanotolerant phenotype., Author Summary Whereas immunological mechanisms involved in the control of trypanosome infections have been extensively studied in animal models, knowledge of how Trypanosoma brucei gambiense interacts with its human hosts lags far behind. In this study we measured cytokine levels in sleeping sickness patients and individuals who were apparently able to control infection to subdetection levels over long periods of time or who were engaged in a process of self-cure as demonstrated by the disappearance of specific antibodies. In contrast to patients, trypanotolerant subjects were characterized by a strong inflammatory response with elevated levels of IL8, IL6, and TNFα. This study indicates that both protective immune responses and markers of disease development exist in human T. brucei. gambiense infection and constitute an important step forward to identify new diagnostic or therapeutic targets in the fight against sleeping sickness.

Abstract: Sodalis glossinidius, one of the three tsetse fly maternally inherited symbionts, was previously shown to favor fly infection by trypanosomes, the parasites causing human sleeping sickness. Among a population of flies taking a trypanosome-infected blood meal, only a few individuals will acquire the parasite; the others will escape infection and be considered as refractory to trypanosome infection. The aim of the work was to investigate whether fly refractoriness could be associated with specific Sodalis gene expression. The transcriptome of S. glossinidius harbored by flies that were fed either with a non-infected blood meal (control) or with a trypanosome-infected meal but that did not develop infection were analyzed, using microarray technology, and compared. The analysis using the microarray procedure yielded 17 genes that were found to have a significant differential expression between the two groups. Interestingly, all these genes were overexpressed in self-cured (refractory) flies. Further analysis of functional annotation of these genes indicated that most associated biological process terms were related to metabolic and biosynthetic processes as well as to oxido-reduction mechanisms. These results evidence the occurrence of molecular crosstalk between the different partners, induced by the passage of the trypanosomes through the fly's gut even though the parasites were unable to establish in the gut and to develop a permanent infection.

Abstract: Tsetse flies, such as Glossina palpalis gambiensis, are blood-feeding insects that could be subverted as hosts of the parasite Trypanosoma brucei gambiense: initiated in the tsetse fly mid gut, the developmental program of this parasite further proceeds in the salivary glands. The flies act as vectors of this human-invasive parasite when their salivary glands sustain the generation of metacyclic trypomastigotes, the exclusive morphotypes pre-programmed to further develop in the human individuals. Briefly, once the metacyclic trypomastigotes have been deposited in the skin of humans from whom the parasite-hosting tsetse flies are taking their blood meals, the complex developmental program of this Trypanosoma brucei subspecies can result in a severe disease named sleeping sickness. Unveiling the processes that could prevent, in tsetse flies, the developmental program of T. b. gambiense could contribute reducing the prevalence of the disease. Using a global approach, we were curious to extract transcriptional signatures of Sodalis glossinidius, a symbiont hosted by three distinct groups of tsetse flies. To meet this objective, the transcriptome of S. glossinidius from susceptible and refractory tsetse flies was analyzed at 3, 10 and 20 days after flies blood feed on T. b. gambiense-hosting mice. Within this temporal window, 176 trypanosome responsive genes were shown to interact in well-defined patterns making it possible to distinguish flies susceptible to the parasite infection from refractory flies. Among the modulated transcripts in the symbiont population of flies refractory to trypanosome infection many were shown to cluster within the following networks: "lysozyme activity, bacteriolytic enzyme, bacterial cytolysis, cell wall macromolecule catabolic process". These novel data are further delineated in the following questions: could the activation of prophage hosted by S. glossinidius lead to the release of bacterial agonists that trigger the tsetse fly immune system along a profile that no more allows the parasite developmental program? Abstract: International audience; Triatoma infestans, the major vector of Chagas disease south of the Amazon in South America, has a large distribution of wild populations, contrary to what has previously been stated. These populations have been suspected of being the source of reinfestation of human habitats and could impede the full success of vector control campaigns. This study examined gene flow between intra-peridomestic populations and wild populations collected in the surround areas in three Andean localities in Bolivia. The populations were defined according to temporal, ecological, and spatial criteria. After DNA extraction from the legs of each insect, the samples were analyzed using seven microsatellite markers. First, the analysis of molecular variance (AMOVA) detected an absence of differentiation between wild and intra-peridomestic populations, although strong structuring was observed between the populations within each environment. Then for some populations, the Bayesian method of assignment to inferred populations showed very similar assignment patterns of the members of wild or intra-peridomestic populations in each locality. Finally, the detection of the first-generation migrants within the different populations provided evidence of insect displacement from the wild to the intra-peridomestic environment. This result indicates that, after control campaigns in the Andes, controlling this new paradigm of vector transmission risk stemming from the invasion of human habitats by wild populations of T. infestans requires long-term maintenance of public monitoring to keep the risk at a minimal level. Since wild populations of T. infestans have also been detected elsewhere in Argentina, Paraguay, and Chile, there is an urgent need to take these populations into account in future monitoring of Chagas disease transmission.

Abstract: Ce numéro d’Innovations Agronomiques comprend les articles correspondant aux présentations du colloque « La diversité génétique, ressource pour l’agriculture de demain » qui s’est tenu à Paris le 18 juin 2013; The origin of the currently observed cattle species diversity is intimately related to human population history since first domestication events. While new genomic tools (medium to high density SNP chips, next generation sequencing) provide insights into this complex history through the genetic characterisation of cattle breeds, they also allow identifying footprints left in their genome by human (artificial selection pressures) and more generally by environmental constraints (natural selection pressures). [br/]After a brief report on current knowledge of possible origins of cattle diversity, based on the characterization of cattle breeds neutral genetic diversity, we illustrate, in this article, studies of natural and artificial selection footprints in bovine genome from analyses carried out in African and New World tropical cattle breeds and in high-producing dairy cattle.; L’origine de la grande diversité actuelle de l'espèce bovine est intimement liée à l’histoire des peuplements humains depuis les premiers événements de domestication. Si les nouveaux outils génomiques (puces de génotypage de SNP à moyenne et haute densité, séquençage à haut-débit) aident à retracer cette histoire complexe, à travers la caractérisation génétique fine des races actuelles, ils permettent également d'identifier dans le génome de ces races les empreintes laissées par les contraintes exercées par l'homme (pressions de sélection artificielle) et plus généralement par l'environnement (pressions de sélection naturelle). 213. Understanding the genetic, demographical and/or ecological processes at play in invasions: lessons from the southern cattle tick Rhipicephalus microplus (Acari: Ixodidae)

Abstract: The southern cattle tick, Rhipicephalus microplus, is the ixodid species causing the largest economic losses in tropical agrosystems because of its recurrent invasive success, explosive demography on bovine herds, vector competence for diverse pathogens and frequent development of acaricide resistance. Its ecology and the physiological bases of the acaricide resistances it developed, as well as alternative tick control measures, have been intensively studied for decades. By contrast, the tick population genetic structure and its remarkable ability to quickly adapt to new environments have not yet received much attention. We investigated such issues using population genetics analyses in the recently invaded island New Caledonia. In this paper we aim to describe some guidelines for acarologists willing to investigate the processes at play in Acari invasions. Particular emphasis is given to the accuracy of sampling designs and sampling scales for population genetics to be actually informative on the demographical processes of the species (i.e., its mating rules, the determinants of population limits, population sizes, the relationships between genetic exchanges and geographical distances and relevant ecological factors).

Abstract: Filariae are a leading cause of infections which are responsible for serious dermatological, ocular, and vascular lesions. Infective third stage larvae (L3) are transmitted through the bite of a haematophagous vector.Litomosoides sigmodontisis a well-established model of filariasis in the mouse, with the vector being the miteOrnithonyssus bacoti. The aim of the study was to analyse the filarial infection in mites to determine the consequences of filarial infection in the blood-feeding and the reproduction of mites as well as in the regulation of vector-induced inflammation in the mouse skin. Firstly, L3 are unevenly distributed throughout the host population and the majority of the population harbours a moderate infection (1 to 6 L3). Filarial infection does not significantly affect the probing delay for blood feeding. The number of released protonymphs is lower in infected mites but is not correlated with the L3 burden. Finally, induced excreted proteins from infected mites but not from uninfected mites stimulate TNF-αand the neutrophil-chemoattractant KC production by antigen-presenting cells (APCs). Altogether, these results describe the modification of the mite behavior under filarial infection and suggest that the immunomodulatory capacity of the mite may be modified by the presence of the parasite, hindering its defensive ability towards the vertebrate host.

Abstract: Chantier qualité GA; Background: The Senepol cattle breed (SEN) was created in the early XXth century from a presumed cross between a European (EUT) breed (Red Poll) and a West African taurine (AFT) breed (N'Dama). Well adapted to tropical conditions, it is also believed trypanotolerant according to its putative AFT ancestry. However, such origins needed to be verified to define relevant husbandry practices and the genetic background underlying such adaptation needed to be characterized. Methodology/Principal Findings: We genotyped 153 SEN individuals on 47,365 SNPs and combined the resulting data with those available on 18 other populations representative of EUT, AFT and Zebu (ZEB) cattle. We found on average 89% EUT, 10.4% ZEB and 0.6% AFT ancestries in the SEN genome. We further looked for footprints of recent selection using standard tests based on the extent of haplotype homozygosity. We underlined i) three footprints on chromosome (BTA) 01, two of which are within or close to the polled locus underlying the absence of horns and ii) one footprint on BTA20 within the slick hair coat locus, involved in thermotolerance. Annotation of these regions allowed us to propose three candidate genes to explain the observed signals (TIAM1, GRIK1 and RAI14). Conclusions/Significance: Our results do not support the accepted concept about the AFT origin of SEN breed. Initial AFT ancestry (if any) might have been counter-selected in early generations due to breeding objectives oriented in particular toward meat production and hornless phenotype. Therefore, SEN animals are likely susceptible to African trypanosomes which questions the importation of SEN within the West African tsetse belt, as promoted by some breeding societies. Besides, our results revealed that SEN breed is predominantly a EUT breed well adapted to tropical conditions and confirmed the importance in thermotolerance of the slick locus.

Abstract: International audience; Ixodes ricinus is a European tick that transmits numerous pathogenic agents, including the bacteria that cause Lyme disease (some genospecies of Borrelia burgdorferi sensu lato complex). This tick has been considered as a classic example of an extreme generalist vector. However, host-associations in such vector species are difficult to determine from field observations alone and recent work suggests that host specificity may be more frequent in ticks than previously thought. The presence of host-associated vector groups can significantly alter the circulation and evolutionary pathway of associated pathogens. In thispaper, we explicitly test for host-associated genetic structure in I. ricinus.We analyzed genetic variability at 11 microsatellite markers in a large sample of ticks collected directly from trapped wild animals (birds, rodents, lizards, wild boar and roe deer) at five sites in Western and Central Europe. We found significant levels of genetic structure both among host individuals and among host types within local populations, suggesting that host use is not random in I. ricinus. These results help explain previous patterns of structure found in off-host tick samples, along with epidemiological observations of Lyme disease

Abstract: In Cambodia, dengue virus (DENV) was first isolated in 1963 and has become endemic with peak epidemic during raining season. Since 2000, the Dengue National Control Program has reported from 10,000 to 40,000 cases per year with fatality rates ranging from 0.7 to 1.7. All four dengue serotypes are found circulating in Cambodia with alternative predominance of serotypes DENV-2 and DENV-3. The DENV-1 represents from 5% to 20% of all circulating viruses, depending upon the year. In this work, 79 clinical strains of DENV-1 were isolated between 2000 and 2009 and their genome fully sequenced. Four distinct lineages with different dynamics were identified. The main evolutionary drive was negative selective pressure but each lineage was characterized by the presence of specific mutations acquired through evolution. Coexistence, extinction and replacement of lineages occurred over the 10-year period. Lineages 1, 2 and 3 were all detected since 2000–2002 and disappeared in 2003, 2004–2005 and 2007, respectively. Lineages 1 and 2 displayed different dynamics. Lineage 1 was very diverse whereas lineage 2 was very homogeneous. Lineage 4 which derived from lineage 3 in 2003 remained the only one at the end of the sampling period in 2008–2009 owing to a selective sweep. The lineages dynamic of DENV-1 viruses and consequences for molecular epidemiology are discussed.

Abstract: Background African animal trypanosomosis is a major obstacle to the development of more efficient and sustainable livestock production systems in West Africa. Riverine tsetse species such as Glossina palpalis gambiensis Vanderplank and Glossina tachinoides Westwood are the major vectors. A wide variety of control tactics is available to manage these vectors, but their removal will in most cases only be sustainable if the control effort is targeting an entire tsetse population within a circumscribed area. Methodology/Principal Findings In the present study, genetic variation at microsatellite DNA loci was used to examine the population structure of G. p. gambiensis and G. tachinoides inhabiting four adjacent river basins in Burkina Faso, i.e. the Mouhoun, the Comoé, the Niger and the Sissili River Basins. Isolation by distance was significant for both species across river basins, and dispersal of G. tachinoides was ∼3 times higher than that of G. p. gambiensis. Thus, the data presented indicate that no strong barriers to gene flow exists between riverine tsetse populations in adjacent river basins, especially so for G. tachinoides. Conclusions/Significance Therefore, potential re-invasion of flies from adjacent river basins will have to be prevented by establishing buffer zones between the Mouhoun and the other river basin(s), in the framework of the PATTEC (Pan African Tsetse and Trypanosomosis Eradication Campaign) eradication project that is presently targeting the northern part of the Mouhoun River Basin. We argue that these genetic analyses should always be part of the baseline data collection before any tsetse control project is initiated., Author Summary Tsetse flies are insects that transmit trypanosomes to humans (sleeping sickness) and animals (nagana). Controlling these vectors is a very efficient way to control these diseases. In Burkina Faso, a tsetse eradication campaign is presently targeting the northern part of the Mouhoun River Basin. To attain this objective, the approach has to be area-wide, i.e. the control effort targets an entire pest population within a circumscribed area. To assess the level of this isolation, we studied the genetic structure of Glossina palpalis gambiensis and Glossina tachinoides populations in the target area and in the adjacent river basins of the Comoé, the Niger and the Sissili River Basins. Our results suggest an absence of strong genetic isolation of the target populations. We therefore recommend establishing permanent buffer zones between the Mouhoun and the other river basin(s) to prevent reinvasion. This kind of study may be extended to other areas on other tsetse species.

Abstract: International audience; Trypanosoma cruzi, the agent of Chagas disease, is usually subdivided into six discrete typing units (DTUs), TcI to TcVI, among which TcI and TcV are most common in human infections in Bolivia. Multilocus microsatellite typing (MLMT) was selected to further explore the structure of the natural populations belonging to these DTUs. The analysis showed that microsatellite clustering does not fully match the six DTUs, but it is relevant for the within DTUs analyses. Population genetics analysis was conducted on 11 relevant subsamples of stocks from Bolivia and Peru, belonging to TcI (6) and TcV (5), defined by four criterions: DTU, vector species, geographic origin, and date of isolation. Most TcV strains presented the same multilocus genotype over all subsamples with the puzzling characteristic that five loci were heterozygous and the other five homozygous. In TcI, four clusters were defined according to the vector species. Most of them appeared in agreement with clonal propagation (stocks isolated from Triatoma infestans and Triatoma sordida), while a few highly homozygous stocks (e.g. those isolated from Rhodnius stali) suggested that scarce sex events can occur. The poor role played by spatio-temporal factors in describing the observed genetic diversity suggested that ecology, in particular as regard to host played a significant role. These results highlight the extreme heterogeneity of T. cruzi and suggest that further population genetics surveys will need to target the most possible precise spatio-temporal and ecological scales.

Abstract: International audience; Scrub typhus is an acute infectious disease caused by an obligate intracellular bacterium Orientia tsutsugamushi following the bite of infected trombiculid mites of the genus Leptotrombidium. This zoonotic disease is a major cause of febrile illness in the Asia-Pacific region, with a large spectrum of clinical manifestations from unapparent or mild disease to fatal disease. O. tsutsugamushi is characterized by a very high genomic plasticity and a large number of antigenic variants amongst strains. The 56-kDa type specific antigen (TSA) gene, encoding the major antigenic protein, was used as reference to investigate the genetic relationships between the strains and to genotype O. tsutsugamushi isolates. The open reading frame of the 56-kDa TSA gene of 41 sequences (28 Cambodian and 13 Vietnamese strains) from patient samples were sequenced and used for genotyping. The 28 Cambodian isolates clustered into 5 major groups, including Karp (43.5%), JG-v (25%), Kato/TA716 (21.5%), TA763 (3.5%) and Gilliam (3.5%). Karp (77%), TA763 (15.5%) and JG-v (7.5%) strains were identified amongst the 13 Vietnamese isolates. This is the first countrywide genotyping description in Cambodia and in Central Vietnam. These results demonstrate the considerable diversity of genotypes in co-circulation in both countries. The genotyping result might raise awareness amongst Cambodian and Vietnamese clinicians of the high genetic diversity of circulating O. tsutsugamushi strains and provides unique and beneficial data for serological and molecular diagnosis of scrub typhus infections as well as raw materials for future studies and vaccine development.

Abstract: Warm-Blooded Reptiles?Existing reptiles are not thought to be endothermic, but what about extinct species? Three large extinct swimming reptiles, the ichthyosaurs, plesiosaurs, and mosasaurs, were active predators in the Mesozoic oceans.Bernardet al.(p.1379; see the Perspective byMotani) investigated their metabolism by analyzing the oxygen isotopes in their teeth, compared with fish in deposits from a variety of ocean environments. The data imply that the ichthyosaurs and plesiosaurs, which were both pursuit predators, probably controlled their own temperature. The data for the mosasaurs, which are thought to have hunted by ambush, are more equivocal.

Abstract: International audience; Human and animal African trypanosomoses, or sleeping sickness and Nagana, are neglected vector-borne parasitic diseases caused by protozoa belonging to the Trypanosoma genus. Advances in proteomics offer new tools to better understand host-vector-parasite crosstalks occurring during the complex parasitic developmental cycle, and to determine the outcome of both transmission and infection. In this review, we summarize proteomics studies performed on African trypanosomes and on the interactions with their vector and mammalian hosts. We discuss the contributions and pitfalls of using diverse proteomics tools, and argue about the interest of pathogenoproteomics, both to generate advances in basic research on the best knowledge and understanding of host-vector-pathogen interactions, and to lead to the concrete development of new tools to improve diagnosis and treatment management of trypanosomoses in the near future.

Abstract: International audience; Cowdriosis or heartwater is a major tick-borne disease on ruminants in Africa and the Caribbean. The causative agent is Ehrlichia ruminantium, an intracellular bacterium. Development of vaccines against heartwater has been hampered the limited efficiency of vaccine in the field, thought to be a consequence of the high genetic diversity of strains circulating in a same area. A sampling scheme was set to collect ticks over 2 years in a delimited area and well identified flock. Prevalence was low at about 3%. A set of 37 strains was considered for MLST analysis along with two reference strains, i.e. ERGA and ERWO, for which full-length genome was available, using a previously described scheme based on the genes gltA,groEL, lepA, lipA, lipB, secY, sodB and sucA. Two populations were identified both with limited genetic variability but with differing evolutionary patterns. Population 1 is in genomic stasis, in agreement with the paradigm for intracellular bacteria. The two reference strains, one from the Caribbean separated from West African strains three centuries ago and another one isolated in South Africa, belong to Population 1. Population 2 is on expansion following a recent clonal emergence from Population 1. The founder strain was identified as strain 395. Strain 623 displays a particularly high rate of mutations in groEL. Owing to the chaperone function of GroEL, this might indicate another clonal emergence under way. This work brings further insight in the genomic plasticity of E. ruminantium and its impact on vaccine strategy.

Abstract: Background Recent developments in high-throughput methods of analyzing transcriptomic profiles are promising for many areas of biology, including ecophysiology. However, although commercial microarrays are available for most common laboratory models, transcriptome analysis in non-traditional model species still remains a challenge. Indeed, the signal resulting from heterologous hybridization is low and difficult to interpret because of the weak complementarity between probe and target sequences, especially when no microarray dedicated to a genetically close species is available. Results We show here that transcriptome analysis in a species genetically distant from laboratory models is made possible by using MAXRS, a new method of analyzing heterologous hybridization on microarrays. This method takes advantage of the design of several commercial microarrays, with different probes targeting the same transcript. To illustrate and test this method, we analyzed the transcriptome of king penguin pectoralis muscle hybridized to Affymetrix chicken microarrays, two organisms separated by an evolutionary distance of approximately 100 million years. The differential gene expression observed between different physiological situations computed by MAXRS was confirmed by real-time PCR on 10 genes out of 11 tested. Conclusions MAXRS appears to be an appropriate method for gene expression analysis under heterologous hybridization conditions.

Both the World Health Organisation and African countries through the Pan African Tsetse and Trypanosomiasis Eradication Campaign (PATTEC) have recently asserted their determination to rid the sub-continent of these diseases, and it is increasingly recognised that vector control should play an important role. This review mainly focuses on population genetics of tsetse of the palpalis group, the main vectors of sleeping sickness, and reports recent results on tsetse population structure and on measures of gene flow between populations. Abstract: Trypanosomasecretome was shown to be involved in parasite virulence and is suspected of interfering in parasite life-cycle steps such as establishment in theGlossinamidgut, metacyclogenesis. Therefore, we attempted to identify the proteins secreted by procyclic strains ofT. brucei gambienseandT. brucei brucei, responsible for human and animal trypanosomiasis, respectively. Using mass spectrometry, 427 and 483 nonredundant proteins were characterized inT. brucei bruceiandT. brucei gambiensesecretomes, respectively; 35% and 42% of the corresponding secretome proteins were specifically secreted byT. brucei bruceiandT. brucei gambiense, respectively, while 279 proteins were common to both subspecies. The proteins were assigned to 12 functional classes. Special attention was paid to the most abundant proteases (14 families) because of their potential implication in the infection process and nutrient supply. The presence of proteins usually secreted via an exosome pathway suggests that this type of process is involved in trypanosome ESP secretion. The overall results provide leads for further research to develop novel tools for blocking trypanosome transmission.

Abstract: International audience; The West African trypanosomoses are mostly transmitted by riverine species of tsetse fly. In this study, we estimate the dispersal and population size of tsetse populations located along the Mouhoun river in Burkina Faso where tsetse habitats are experiencing increasing fragmentation caused by human encroachment. Dispersal estimated through direct (mark and recapture) and indirect (genetic isolation by distance) methods appeared consistent with one another. In these fragmented landscapes, tsetse flies displayed localized, small subpopulations with relatively short effective dispersal. We discuss how such information is crucial for designing optimal strategies for eliminating this threat. To estimate ecological parameters of wild animal populations, the genetic measures are both a cost- and time-effective alternative to mark-release-recapture. They can be applied to other vector-borne diseases of medical and/or economic importance.

Abstract: Background We undertook a population genetics analysis of the tsetse fly Glossina palpalis gambiensis, a major vector of sleeping sickness in West Africa, using microsatellite and mitochondrial DNA markers. Our aims were to estimate effective population size and the degree of isolation between coastal sites on the mainland of Guinea and Loos Islands. The sampling locations encompassed Dubréka, the area with the highest Human African Trypanosomosis (HAT) prevalence in West Africa, mangrove and savannah sites on the mainland, and two islands, Fotoba and Kassa, within the Loos archipelago. These data are discussed with respect to the feasibility and sustainability of control strategies in those sites currently experiencing, or at risk of, sleeping sickness. Principal Findings We found very low migration rates between sites except between those sampled around the Dubréka area that seems to contain a widely dispersed and panmictic population. In the Kassa island samples, various effective population size estimates all converged on surprisingly small values (10, Author Summary Guinea is the country with the highest prevalence of sleeping sickness in West Africa, and we undertook a population genetics analysis there of the most dangerous tsetse fly species of West Africa, Glossina palpalis gambiensis. We found very low migration rates of tsetse between sites except between those situated in the Dubréka area, which seems to contain a widely distributed panmictic tsetse population (i.e. a population where mating occurs at random). Effective population sizes on Loos islands estimated with various techniques all converged to surprisingly small values. These values might be explained by a recent decrease in tsetse numbers on Kassa Island due to bauxite mining activities. But on the other sites, other explanations have to be found, including possible variance in reproductive success. Our genetic results suggest that different control strategies should be advised on the mainland (reduction in tsetse densities, no elimination) compared to the islands (total elimination feasible). This approach could be extended to many areas where vector control of Human and Animal Trypanosomoses is contemplated.

Abstract: Animal trypanosomosis is a major constraint to livestock productivity in the tropics and has a significant impact on the life of millions of people globally (mainly in Africa, South America and south-east Asia). In Africa, the disease in livestock is caused mainly by Trypanosoma congolense, Trypanosoma vivax, Trypanosoma evansi and Trypanosoma brucei brucei. The extracellular position of trypanosomes in the bloodstream of their host requires consideration of both the parasite and its naturally excreted-secreted factors (secretome) in the course of pathophysiological processes. We therefore developed and standardised a method to produce purified proteomes and secretomes of African trypanosomes. In this study, two strains of T. congolense exhibiting opposite properties of both virulence and pathogenicity were further investigated through their secretome expression and its involvement in host-parasite interactions. We used a combined proteomic approach (one-dimensional SDS-PAGE and two-dimensional differential in-gel electrophoresis coupled to mass spectrometry) to characterise the whole and differentially expressed protein contents of secretomes. The molecular identification of differentially expressed trypanosome molecules and their correlation with either the virulence process or pathogenicity are discussed with regard to their potential as new diagnostic or therapeutic tools against animal trypanosomosis.

Abstract: Many scientists working on pathogens (viruses, bacteria, fungi, parasites) are betting heavily on data generated by longitudinal genomic-transcriptomic-proteomic studies to explain biochemical host-vector-pathogen interactions and thus to contribute to disease control. Availability of genome sequences of various organisms, from viruses to complex metazoans, led to the discovery of the functions of the genes themselves. The postgenomic era stimulated the development of proteomic and bioinformatics tools to identify the locations, functions, and interactions of the gene products in tissues and/or cells of living organisms. Because of the diversity of available methods and the level of integration they promote, proteomics tools are potentially able to resolve interesting issues specific not only to host-vector-pathogen interactions in cell immunobiology, but also to ecology and evolution, population biology, and adaptive processes. These new analytical tools, as all new tools, contain pitfalls directly related to experimental design, statistical treatment, and protein identification. Nevertheless, they offer the potency of building large protein-protein interaction networks for in silica, analysis of novel biological entities named "interactomes," a way of modeling host-vector-pathogen interactions to define new interference strategies.

Abstract: International audience; Animal trypanosomosis is one of the most severe constraints to agricultural development in sub-Saharan Africa and is also an important disease of livestock in Latin America and Asia. The causative agents are various species of protozoan parasites belonging to the genus Trypanosoma, among which T. congolense and T. evansi are the major pathogenic species. The extracellular position of trypanosomes obliges us to consider both the parasite and its excreted/secreted factors in the course of the physiopathologic process. The advent of proteomics led us to propose a comparative approach of the proteome (i.e., the whole parasite content) and the secretome (i.e., naturally excreted/secreted molecules) of T. congolense and T. evansi with particular attention to common and specific molecules between strains of differing virulence and pathogenicity. The molecular identification of differentially expressed trypanosome molecules correlated with either the virulence process or the pathogenicity will provide new potential molecular targets for improved field diagnosis and chemotherapy of animal trypanosomosis

Abstract: Resume Objectif Evaluer la situation de la maladie du sommeil dans le centre-ouest de la Cote d'Ivoire de 2000 a 2003, en tenant compte des evenements survenus depuis en septembre 2002. Methodes Enquete active realisee par des equipes medicales et detection passive des cas. Resultats Entre 2000 et 2003, 250 patients ont ete diagnostiques pour la maladie du sommeil. De prime abord la prevalence de la maladie du sommeil semblait avoir baisse depuis le debut de la guerre. Mais cette baisse apparente etait due a une faible couverture de la population. La participation dans l'enquete medicale etait differente selon le groupe ethnique, refletant les conflits entre les differentes communautes pour les terres. De tels conflits sont courants dans la zone mais ont ete exacerbes par la guerre. Conclusion L’evaluation de l'importance de la maladie du sommeil durant la guerre par enquete medicale seule est tres difficile. Mais la detection de la maladie du sommeil par surveillance passive a augmente. En un principio parecia que la prevalencia de la enfermedad habia caido desde el comienzo de la guerra. Pero esta caida aparente fue debida a una mala cobertura de la poblacion. La participacion en las encuestas medicas fue diferente dependiendo del grupo etnico, lo cual refleja el conflicto en el uso de la tierra entre comunidades etnicas. Estos conflictos son comunes en esta area, pero la guerra los ha exacerbado Conclusiones Valorar durante la guerra la importancia de la enfermedad del sueno solamente mediante encuestas medicas es muy dificil. La deteccion de la enfermedad del sueno por deteccion pasiva de casos, sin embargo, ha aumentado.

Abstract: International audience; Glossina palpalis is the main vector of human African trypanosomosis (HAT, or sleeping sickness) that dramatically affects human health in sub-Saharan Africa. Because of the implications of genetic structuring of vector populations for the design and efficacy of control campaigns, G. palpalis palpalis in the most active focus of sleeping sickness in Côte d'Ivoire was studied to determine whether this taxon is genetically structured. High and statistically significant levels of within population heterozygote deficiencies were found at each of the five microsatellite loci in two temporally separated samples. Neither null alleles, short allele dominance, nor trap locations could fully explain these deviations from random mating, but a clustering within each of the two samples into different genetic sub-populations (Wahlund effect) was strongly suggested. These different genetic groups, which could display differences in infection rates and trypanosome identity, were composed of small numbers of individuals that were captured together, leading to the observed Wahlund effect. Implications of this population structure on tsetse control are discussed.

Abstract: The clinical evolution of the chronic form of Human African Trypanosomiasis starts with the haematolymphatic or first stage (P1). The meningoencephalitic or second stage (P2) begins when trypanosomes reach the cerebrospinal fluid (CSF). The classical stage determination method is based on CSF cell count, CSF protein concentration and/or the presence of trypanosomes detected in CSF. However their cut-off values and the sensitivity of detection of trypanosomes in CSF remains doubtful while the appropriate treatment depends on this determination of disease stage. Thus, the classical stage determination is reconsidered using new serological tests, and results were compared to the clinical data. Thirty-eight patients were classified into 4 clinical groups according to the observed degree of severity of neuropsychiatric signs. Based on multivariate analysis to evaluate the relevance of the new serological tests as compared with clinical groups, we confirm that Latex IgM CSF, cheap and easy to perform under field conditions, may improve stage determination of the disease.African Journal of Biotechnology Vol. 4 (6), pp. 517-521, 2005

Abstract: Trypanosoma iax is a widespread hemoparasite in tropical areas and is pathogenic to ruminant domestic livestock as well as wild ruminants. The accurate identification of parasites in both hosts and vectors is crucial for epidemiological studies and disease control programs. We describe here the development of molecular markers specific for T. iax identification. These markers were used to identify mouthpart infections in field-collected tsetse flies from Cameroon. The markers target the genomic sequence of a species-specific antigen from the bloodstream stages. No cross amplification with other trypanosome species was observed, which makes the markers a reliable tool to detect T. iax infections, both in hosts and vectors. The PCR-amplified sequence contains a (CA)n microsatellite repeat for which 11 different alleles were identified. This microsatellite, which showed high polymorphism, provides a suitable marker for population genetic studies. © 2001 Elsevier Science B.V. All rights reserved.

Abstract: A putative proximal promoter was defined previously for the mouse glucagon receptor (GR) gene. In the present study, a distal promoter was characterized upstream from a novel non-coding exon revealed by the 5′-rapid amplification of cDNA ends from mouse liver tissue. The 5′-flanking region of the mouse GR gene was cloned up to 6 kb and the structural organization was compared to the 5′ untranslated region of the rat gene cloned up to 7 kb. The novel exon, separated by an intron of 3.8 kb from the first coding exon, displayed a high homology (80%) with the most distal of the two untranslated exons found in the 5′ region of the rat GR gene. The mouse distal promoter region, extending up to −1 kb from the novel exon, displayed 85% identity with the rat promoter. Both contain a highly GC-rich sequence with five putative binding sites for Sp1, but no consensus TATA or CAAT elements. To evaluate basal promoter activities, 5′-flanking sequences of mouse or rat GR genes were fused to a luciferase reporter gene and transiently expressed in a mouse and in a rat cell line, respectively or in rat hepatocytes. Both mouse and rat distal promoter regions directed a high level of reporter gene activity. Deletion of the Sp1 binding sites region or mutation of the second proximal Sp1 sequence markedly reduced the distal promoter activity of the reporter gene. The mouse proximal promoter activity was 2- to 3-fold less than the distal promoter, for which no functional counterpart was observed in the similar region of the rat gene.

Abstract: International audience; Members of the Rho/Rac/Cdc42Hs family of GTPases have been shown to participate in many aspects of the signaling of cell growth and differentiation. Although the biochemical properties of these GTPases have been extensively studied, very little is known about the structure of the corresponding genes. To gain insight on the evolution of the Rho family, we were interested in studying the genomic structure of several members. We report here the structure and the localization to 22q12 of the human Rac2 gene, as well as the localization to 17qter of Rac3, a new member closely related to Rac1 and Rac2. Unlike the structure of its closest relative ARH-G gene, which contains a single intron, Rac2 is made of at least 7 exons, spanning over 18 kb of DNA. Comparison of gene structure and exonic borders suggests that the emergence of the whole superfamily took place early during evolution. (C) 1997 Academic Press.

Abstract: Background The analysis of humoral responses directed against the saliva of blood-sucking arthropods was shown to provide epidemiological biomarkers of human exposure to vector-borne diseases. However, the use of whole saliva as antigen presents several limitations such as problems of mass production, reproducibility and specificity. The aim of this study was to design a specific biomarker of exposure to tsetse flies based on the in silico analysis of three Glossina salivary proteins (Ada, Ag5 and Tsgf1) previously shown to be specifically recognized by plasma from exposed individuals. Methodology/Principal Findings Synthetic peptides were designed by combining several linear epitope prediction methods and Blast analysis. The most specific peptides were then tested by indirect ELISA on a bank of 160 plasma samples from tsetse infested areas and tsetse free areas. Significantly higher anti- Tsgf118–43 responses were also observed in sleeping sickness patients as compared to uninfected controls suggesting that Tsgf118–43 may be used both to assess human tsetse contacts and the risk of infection by trypanosomes. This new sero-epidemiological tool could thus help National Control Programs to quickly map human exposure levels in order to better target vector control efforts and monitor vector control efficiency.

Abstract: International audience; BACKGROUND: Many tools used to analyze microarrays in different conditions have been described. However, the integration of deregulated genes within coherent metabolic pathways is lacking. Currently no objective selection criterion based on biological functions exists to determine a threshold demonstrating that a gene is indeed differentially expressed. METHODOLOGY/PRINCIPAL FINDINGS: To improve transcriptomic analysis of microarrays, we propose a new statistical approach that takes into account biological parameters. We present an iterative method to optimise the selection of differentially expressed genes in two experimental conditions. The stringency level of gene selection was associated simultaneously with the p-value of expression variation and the occurrence rate parameter associated with the percentage of donors whose transcriptomic profile is similar. Our method intertwines stringency level settings, biological data and a knowledge database to highlight molecular interactions using networks and pathways. Analysis performed during iterations helped us to select the optimal threshold required for the most pertinent selection of differentially expressed genes. CONCLUSIONS/SIGNIFICANCE: We have applied this approach to the well documented mechanism of human macrophage response to lipopolysaccharide stimulation. We thus verified that our method was able to determine with the highest degree of accuracy the best threshold for selecting genes that are truly differentially expressed.

Abstract: Background Human African trypanosomiasis is a lethal disease caused by the extracellular parasite Trypanosoma brucei. The proteins secreted by T. brucei inhibit the maturation of dendritic cells and their ability to induce lymphocytic allogenic responses. To better understand the pathogenic process, we combined different approaches to characterize these secreted proteins. Results Overall, 444 proteins were identified using mass spectrometry, the largest parasite secretome described to date. Functional analysis of these proteins revealed a strong bias toward folding and degradation processes and to a lesser extent toward nucleotide metabolism. These features were shared by different strains of T. brucei, but distinguished the secretome from published T. brucei whole proteome or glycosome. In addition, several proteins had not been previously described in Trypanosoma and some constitute novel potential therapeutic targets or diagnostic markers. Interestingly, a high proportion of these secreted proteins are known to have alternative roles once secreted. Furthermore, bioinformatic analysis showed that a significant proportion of proteins in the secretome lack transit peptide and are probably not secreted through the classical sorting pathway. Membrane vesicles from secretion buffer and infested rat serum were purified on sucrose gradient and electron microscopy pictures have shown 50- to 100-nm vesicles budding from the coated plasma membrane. Mass spectrometry confirmed the presence of Trypanosoma proteins in these microvesicles, showing that an active exocytosis might occur beyond the flagellar pocket. Conclusions This study brings out several unexpected features of the secreted proteins and opens novel perspectives concerning the survival strategy of Trypanosoma as well as possible ways to control the disease. In addition, concordant lines of evidence support the original hypothesis of the involvement of microvesicle-like bodies in the survival strategy allowing Trypanosoma to exchange proteins at least between parasites and/or to manipulate the host immune system.

Abstract: International audience; Tsetse flies (Diptera: Glossinidae) are the vectors of trypanosomes causing sleeping sickness in humans, and nagana (animal trypanosomosis) in domestic animals, in Subsaharan Africa. They have been described as being strictly hematophagous, and transmission of trypanosomes occurs when they feed on a human or an animal. There have been indications however in old papers that tsetse may have the ability to digest sugar. Here we show that hungry tsetse (Glossina palpalis gambiensis) in the lab do feed on water and on water with sugar when no blood is available, and we also show that wild tsetse have detectable sugar residues. We showed in laboratory conditions that at a low concentration (0.1%) or provided occasionally (0.1%, 0.5%, 1%), glucose had no significant impact on female longevity and fecundity. However, regular provision of water with 1% glucose increased the mortality and reduced the fecundity of female G. p. gambiensis. The proportion of wild tsetse caught by traps, which have detectable sugar residue in their midgut varied between 5 and 10% according to species (p < 10 (3)) and sex, with more females being found with sugar residues than males (p < 10 (3)). We also observed a higher frequency of sugar residues in the dry season than in the rainy season (p < 10(3)). The infection status did not affect the frequency of sugar residues found (p = 0.65), neither did age (p = 0.23). These observations represent a fundamental change in our knowledge of this insect vector. They open the way for further research on the field to know more on tsetse feeding behavior regarding other sources of meal than blood, in particular with plants, and may constitute future new means of controlling this vector of neglected tropical diseases.

Abstract: The distribution of neutral genetic variation in subdivided populations is driven by the interplay between genetic drift, migration, local extinction and colonization. The influence of environmental and demographic factors has also been increasingly examined in empirical studies, but generally focusing on a single species. An open question is whether these factors will similarly, or idiosyncratically, affect a guild of species occupying the same, though exhibiting different traits, mating systems and histories. We addressed it by comparing the population genetic structure of the four most common species of hermaphroditic freshwater snails in Guadeloupe (Lesser Antilles), which occupy a network of patchily distributed sites experiencing temporal variation in water availability. We analyzed microsatellite variability in 21 to 43 populations per species, and built predictions on how several environmental and demographic variables, quantified from a long-term annual survey, as well as species traits, may affect the distribution of genetic variation. These species displayed similarities, such as fairly high levels of variation, but with marked differences among sites, as well as strong genetic differentiation and limited isolation by distance, which can be explained by passive dispersal (strong role of site connectivity), extinction/colonization dynamics and variation in local population size. They also exhibit differences, largely due to the mating system with less genetic diversity and more genetic differentiation in the two selfing species when compared to the two outcrossing ones. These differences can also be attributed to interspecific interactions resulting from the ongoing invasion of Guadeloupe by one of the species studied, which affects the demography of other species, and, to a limited extent, to local environmental factors. Our comparative approach shows both differences and uniqueness in the way species occupy the same landscape, and provides a possible entry to interspecific interactions in community assembly.

Abstract: International audience; Context: Galactose binding protein (PpGalec) plays an important role in the specificity of Phlebotomus papatasi sandfly for Leishmania major. The molecular diversity of this ligand is currently unknown but might have some influence on the ability of PpGalec to efficiently recognize L. major in natural sandfly populations.Objective: To explore the molecular diversity of the P. papatasi Galectin gene (PpGalec) in natural sandfly population of Morocco.Results & Conclusions: Sequence variations of PpGalec was analyzed in 31 P. papatasi specimens collected from endemic and non-endemic zoonotic cutaneous leishmaniasis foci of Morocco. Among the 211 amino acid positions analyzed, 11 are subjected to mutation. Interestingly, we observe that one mutation directly affect an amino acid known to be involved in the substrate recognition by galectin. The repercussion of this polymorphism on the capacity of the galectin to efficiently bind the L. major Lipophosphoglycane (LPG) awaits further investigations.

Abstract: Trypanosoma brucei gambiense, an extracellular eukaryotic flagellate parasite, is the main causative agent of Human African Trypanosomiasis (HAT), also known as sleeping sickness. Trypanosomes have developped efficient mechanisms to escape the host immune response. New therapeutic options are needed for patients with HAT. Sterol 14α-demethylase (CYP51) is a potential drug target but its essentiality has not been studied in T. brucei. In a first study, we demonstrated its essentiality by RNA interference (CYP51RNAi) in vitro. CYP51RNAi induction caused morphological defects with multiflagellated cells, suggesting cytokinesis dysfunction. Additionally, the survival of CYP51RNAi infected-mice was improved, showing CYP51 RNAi effect on trypanosomal virulence. During infection with virulent strains, posaconazole-eflornithine and nifurtimox-eflornithine combinations showed similar improvement in mice survival. Thus, our results provide support for a CYP51 targeting based treatment in HAT. In a second work, we studied the innate host immune system characteristics in trypanosomiasis, as a severe immune dysregulation is present in HAT. To analyse the potential immunomodulatory activity of T. gambiense in human settings, we assess the effect of its secretome on dendritic cells (DCs) in vitro, using human monocyte-derived DCs. A significant inhibition of the LPS-induced maturation of DCs was observed with secretome. In line with this impairment, secretome down regulated cytokines production by LPS-activated DCs. TbKHC1, a kinesin heavy chain, is a component of the parasite secretome. We confirmed its role in parasitic escape to immune system by inducing arginase activity, in a murine model. Our results provide new information about the immune system characteristics during trypanosomiasis, which may help to uncover new therapeutic approachs in HAT., La Trypanosomose Humaine Africaine (THA) ou maladie du sommeil est une infection provoquée par un protozoaire du genre Trypanosoma. La recherche de nouvelles cibles thérapeutiques est nécessaire afin d’améliorer l’efficacité et la tolérance des traitements. Dans un premier travail, nous avons étudié l’importance de CYP51 (stérol 14α-déméthylase), une cible potentielle, par la technique d’interférence à ARN (RNAi). Nous avons démontré le caractère essentiel de cette enzyme pour le parasite, ainsi que les conséquences de sa déplétion sur la cytodiérèse. De plus, la survie de souris infectées par la souche CYP51RNAi induite était prolongée, montrant l’implication de CYP51 dans la virulence. La combinaison du posaconazole, un dérivé triazolé inhibant CYP51, à l’éflornithine a montré un effet similaire à la combinaison nifurtimox-éflornithine dans un modèle murin. Nos résultats soulignent l’intérêt potentiel d’un traitement ciblant CYP51 dans la trypanosomose. Du fait de l’importance de l’immunodépression dans la THA et de la capacité du trypanosome à échapper au système immunitaire de l’hôte, nous avons étudié, dans un deuxième travail, l’effet de T. gambiense et de son sécrétome (protéines excrétées/sécrétées) sur des cellules dendritiques humaines (DCs) in vitro. Nous avons ainsi montré une altération de la maturation des DCs induite par le LPS en présence du sécrétome. Nous avons également montré qu’une des protéines de ce sécrétome, TbKHC1, est exprimée par différentes espèces de trypanosomes. Elle est impliquée dans l’induction de l’arginase macrophagique chez la souris, un mécanisme d’échappement au système immunitaire. Ces travaux apportent des éléments pour une meilleure compréhension des phénomènes immunopathologiques rencontrés, dans la perspective de thérapeutiques ciblées et d’une approche vaccinale.

Abstract: [b]Background[/b]: Fragmentation of tsetse habitat in eastern Zambia is largely due to encroachments by subsistence farmers into new areas in search of new agricultural land. The impact of habitat fragmentation on tsetse populations is not clearly understood. This study was aimed at establishing the impact of habitat fragmentation on physiological and demographic parameters of tsetse flies in order to enhance the understanding of the relationship between fragmentation and African animal trypanosomosis (AAT) risk. [b]Methods[/b]: A longitudinal study was conducted to establish the age structure, abundance, proportion of females and trypanosome infection rate of Glossina morsitans morsitans Westwood (Diptera: Glossinidae) in areas of varying degrees of habitat fragmentation in Eastern Zambia. Black screen fly rounds were used to sample tsetse populations monthly for 1 year. Logistic regression was used to analyse age, proportion of females and infection rate data. [br/][b]Results[/b]: Flies got significantly older as fragmentation increased (p < 0.004). The proportion of old flies, i.e. above ovarian category four, increased significantly (P < 0.001) from 25.9 % (CI 21.4-31.1) at the least fragmented site (Lusandwa) to 74.2 % (CI 56.8-86.3) at the highly fragmented site (Chisulo). In the most fragmented area (Kasamanda), tsetse flies had almost disappeared. In the highly fragmented area a significantly higher trypanosome infection rate in tsetse (P < 0.001) than in areas with lower fragmentation was observed. Consequently a comparatively high trypanosomosis incidence rate in livestock was observed there despite lower tsetse density (p < 0.001). The overall proportion of captured female flies increased significantly (P < 0.005) as fragmentation reduced. The proportion increased from 0.135 (CI 0.10-0.18) to 0.285 (CI 0.26-0.31) at the highly and least fragmented sites, respectively. [br/][b]Conclusions[/b]: Habitat fragmentation creates conditions to which tsetse populations respond physiologically and demographically thereby affecting tsetse-trypanosome interactions and hence influencing trypanosomosis risk. Temperature rise due to fragmentation coupled with dominance of old flies in populations increases infection rate in tsetse and hence creates high risk of trypanosomosis in fragmented areas. Possibilities of how correlations between biological characteristics of populations and the degree of fragmentation can be used to structure populations based on their well-being, using integrated GIS and remote sensing techniques are discussed.
