Abstract

Legal information is one of the fundamental information in society, and a statue database serves as a social information infrastructure. Recently, there has been increasing social demands for compiling a database which consists of not only Japanese statutes but also their English translations. In this paper, we describe the problem of translating Japanese statues and show how to solve it by utilizing technologies developed for natural language processing. In particular, we show how to support both the compilation of standard bilingual dictionary and the unification of translation equivalents of legal technical terms in compliance with the dictionary by using word alignment.

Abstract

During the course of our studies of the development of fluorogenic reagents having a 4,7-disubstituted benzofurazan structure, we previously proposed 7-acetylamino-4-mercapto-2,1,3-benzoxadiazole (AABD-SH) as a fluorogenic reagent for carboxylic acids. Since then, progress has made it possible to estimate the fluorescence quantum yields of the 4,7-disubstituted benzofurazan compounds on the basis of the PM3 calculation of their [S.sub.1]-[T.sub.2] energies. Subsequently, a new fluorogenic reagent, 4-mercapto- 7-methylthio-2,1,3-benzoxadiazole (MTBD-SH) was designed and synthesized. In the presence of condensation reagents, triphenylphosphine (TPP) and 2,2'-dipyridyl disulfide (DPDS), MTBD-SH readily reacted with n-caprylic acid within 1 min at room temperature. The derivatives of five carboxylic acids (n-caprylic acid, n-capric acid, lauric acid, myristic acid, and palmitic acid) were well-separated on a reversed-phase column and were fluorimetzically detected at 519 nm with excitation at 391 nm. The detection limits (S/N = 3) were 2.4-5.0 fmol. Thus, MTBD-SH had properties that were considered to be superior. For carboxylic acids, it was superior not only to AABD-SH, but also to many other conventional reagents. The superiority was examined in terms of its reactivity and sensitivity and the avoidance of interfering peaks that were derived from the reagent itself or degradation products in the chromatogram.

Abstract

Using room-temperature parametric oscillation of a LiNb[O.sub.3] crystal pumped by a Q-switched Nd:YAG laser with a simple configuration, we have realized a widely tunable coherent terahertz- (THz-) wave source in the range between i and 3 THz. Inasmuch as the THz wave is affected by total internal reflection at the crystal edge, we used a Si prism coupler to couple out the THz wave. We introduce an arrayed Si-prism coupler that increases the efficiency and decreases the diffraction angle. By use of the arrayed-prism coupler, there is a sixfold increase in coupling efficiency and a 40% decrease in the far-field beam diameter, compared with the use of a single-prism coupler. We discuss the negative effect of the free carriers at the Si-prism surface that is excited by the scattered pump beam, and the positive effect of cavity rotation on the unidirectional radiation of the THz wave from a Si prism. Abstract: Aristolochic acid (AA) causes interstitial renal fibrosis, called aristolochic acid nephropathy (AAN). There is no specific indicator for diagnosing AAN, so this study aimed to investigate the biomarkers for AAN using a proteomics method. The C3H/He female mice were given ad libitum AA–distilled water (0.5 mg/kg/day) and distilled water for 56 days in the AA and normal groups, respectively. The AA‐induced proteins in the kidney were investigated using a proteomics study, including fluorogenic derivatization with 7‐chloro‐N‐[2‐(dimethylamino)ethyl]‐2,1,3‐benzoxadiazole‐4‐sulfonamide, followed by high‐performance liquid chromatography analysis and liquid chromatography tandem mass spectrometry with a MASCOT database searching system. There were two altered proteins, thrombospondin type 1 (TSP1) and G protein‐coupled receptor 87 (GPR87), in the kidney of AA‐group mice on day 56. GPR87, a tumorigenesis‐related protein, is reported for the first time in the current study. The renal interstitial fibrosis was certainly induced in the AA‐group mice under histological examination. Based on the results of histological examination and the proteomics study, this model might be applied to AAN studies in the future. TSP1 might be a novel biomarker for AAN, and the further role of GPR87 leading to AA‐induced tumorigenesis should be researched in future studies. [ABSTRACT FROM AUTHOR]

Abstract

Abstract: Biological functions in organisms are usually controlled by a set of interacting proteins, and identifying the proteins that interact is useful for understanding the mechanism of the functions. Immunoprecipitation is a method that utilizes the affinity of an antibody to isolate and identify the proteins that have interacted in a biological sample. In this study, the FD‐LC–MS/MS method, which involves fluorogenic derivatization followed by separation and quantification by HPLC and finally identification of proteins by HPLC–tandem mass spectrometry, was used to identify proteins in immunoprecipitated samples, using heat shock protein 90 (HSP90) as a model of an interacting protein in HepaRG cells. As a result, HSC70 protein, which was known to form a complex with HSP90, was isolated, together with three different types of HSP90‐beta. The results demonstrated that the proposed immunoaffinity–FD‐LC–MS/MS method could be useful for simultaneously detecting and identifying the proteins that interact with a certain protein. Abstract

AimsHuman epidermal growth factor receptor 2 (HER2)-targeted agents are an effective approach to treating patients with HER2-positive breast cancer. However, the lack of survival benefit in HER2-negative patients, as well as the toxic effects and high cost of the drugs, highlight the need for accurate and prompt assessment of HER2 status. Our aim was to evaluate the clinical utility of a novel reagent-saving immunohistochemistry method (AC-IHC) that saves HER2 antibody by taking advantage of the non-contact mixing effect in microdroplets subjected to an alternating current electric field.MethodsNinety-five specimens were used from patients diagnosed with primary breast cancers identified immunohistochemically as HER2 0/1+, 2+ or 3+ using ASCO/CAP guideline-certified standard IHC. The specimens were all tested using the conventional IHC method (1:50 antibody dilution) as well as AC-IHC (1:50 dilution) and reagent-saving AC-IHC (1:100 dilution).ResultsThe reagent-saving AC-IHC produced stable results with less non-specific staining using smaller amounts of labelled antibody. Moreover, the staining and accuracy of HER2 status evaluated with the reagent-saving AC-IHC method was equal to that achieved with standard IHC.ConclusionsThese results suggest reagent-saving AC-IHC could be used as a clinical tool for accurate and stable HER2 IHC, even when reagent concentrations vary.
