Recurrent neuroinflammation in relapsing-remitting MS (RRMS) is thought to lead to neurodegeneration, resulting in progressive disability. Repeated magnetic resonance imaging (MRI) of the brain provides non-invasive measures of atrophy over time, a key marker of neurodegeneration. This study investigates regional neurodegeneration of the brain in recently-diagnosed RRMS using volumetry and voxel-based morphometry (VBM). RRMS patients (N = 354) underwent 3T structural MRI <6 months after diagnosis and 1-year follow-up, as part of the Scottish multicentre 'FutureMS' study. MRI data were processed using FreeSurfer to derive volumetrics, and FSL for VBM (grey matter (GM) only), to establish regional patterns of change in GM and normal-appearing white matter (NAWM) over time throughout the brain. Volumetric analyses showed a decrease over time (q<0.05) in bilateral cortical GM and NAWM, cerebellar GM, brainstem, amygdala, basal ganglia, hippocampus, accumbens, thalamus and ventral diencephalon. Additionally, NAWM and GM volume decreased respectively in the following cortical regions, frontal: 14 out of 26 regions and 16/26; temporal: 18/18 and 15/18; parietal: 14/14 and 11/14; occipital: 7/8 and 8/8. Left GM and NAWM asymmetry was observed in the frontal lobe. GM VBM analysis showed three major clusters of decrease over time: 1) temporal and subcortical areas, 2) cerebellum, 3) anterior cingulum and supplementary motor cortex; and four smaller clusters within the occipital lobe. Widespread GM and NAWM atrophy was observed in this large recently-diagnosed RRMS cohort, particularly in the brainstem, cerebellar GM, and subcortical and occipital-temporal regions; indicative of neurodegeneration across tissue types, and in accord with limited previous studies in early disease. Volumetric and VBM results emphasise different features of longitudinal lobar and loco-regional change, however identify consistent atrophy patterns across individuals. Atrophy measures targeted to specific brain regions may provide improved markers of neurodegeneration, and potential future imaging stratifiers and endpoints for clinical decision making and therapeutic trials. Rationale: Despite the increased recognition of TBX4 (T-BOX transcription factor 4)-associated pulmonary arterial hypertension (PAH), genotype-phenotype associations are lacking and may provide important insights. Objectives: To compile and functionally characterize all TBX4 variants reported to date and undertake a comprehensive genotype-phenotype analysis. Methods: We assembled a multicenter cohort of 137 patients harboring monoallelic TBX4 variants and assessed the pathogenicity of missense variation (n = 42) using a novel luciferase reporter assay containing T-BOX binding motifs. We sought genotype-phenotype correlations and undertook a comparative analysis with patients with PAH with BMPR2 (Bone Morphogenetic Protein Receptor type 2) causal variants (n = 162) or no identified variants in PAH-associated genes (n = 741) genotyped via the National Institute for Health Research BioResource-Rare Diseases. Measurements and Main Results: Functional assessment of TBX4 missense variants led to the novel finding of gain-of-function effects associated with older age at diagnosis of lung disease compared with loss-of-function effects (P = 0.038). Variants located in the T-BOX and nuclear localization domains were associated with earlier presentation (P = 0.005) and increased incidence of interstitial lung disease (P = 0.003). Event-free survival (death or transplantation) was shorter in the T-BOX group (P = 0.022), although age had a significant effect in the hazard model (P = 0.0461). Carriers of TBX4 variants were diagnosed at a younger age (P < 0.001) and had worse baseline lung function (FEV1, FVC) (P = 0.009) than the BMPR2 and no identified causal variant groups. Conclusions: We demonstrated that TBX4 syndrome is not strictly the result of haploinsufficiency but can also be caused by gain of function. The pleiotropic effects of TBX4 in lung disease may be in part explained by the differential effect of pathogenic mutations located in critical protein domains.

The European Union’s 2020 and 2030 sustainable energy policies seek significant reductions in both energy consumption and carbon emissions. These policies demand a greater use of energy efficient technologies and a transition away from fossil fuels. This paper studies one such technology, an indoor climate control system with a reverse-flow enthalpy recovery ventilator, capable of recovering both sensible and latent heat. The thermal performance characteristics are established using an experimental facility and calculation methods defined by European Standard EN 13141-7:2010. This involves measurement of temperature, humidity, pressure and volumetric air flow rates over a range of operating conditions. Total thermal energy recovery rates ranged from 0.63 kW to 2.2 kW, with energy recovery efficiency of 72.8 % to 88.6 %. The recovery efficiency ratio, which reflects the capacity of the indoor climate control system to recover thermal energy relative to its power consumption ranged between 6.87 to 19.97. Due to the unique reverse-flow defrost function, the system demonstrates operation down to -7 °C without frost formation. These results highlight the potential that this system can make towards the EU goals of reducing energy consumption, operating costs and carbon emissions associated with indoor climate control.

- Abstract
Missense and truncating variants in the X-chromosome-linked CLCN4 gene, resulting in reduced or complete loss-of-function (LOF) of the encoded chloride/proton exchanger ClC-4, were recently demonstrated to cause a neurocognitive phenotype in both males and females. Through international clinical matchmaking and interrogation of public variant databases we assembled a database of 90 rare CLCN4 missense variants in 90 families: 41 unique and 18 recurrent variants in 49 families. For 43 families, including 22 males and 33 females, we collated detailed clinical and segregation data. To confirm causality of variants and to obtain insight into disease mechanisms, we investigated the effect on electrophysiological properties of 59 of the variants in Xenopus oocytes using extended voltage and pH ranges. Detailed analyses revealed new pathophysiological mechanisms: 25% (15/59) of variants demonstrated LOF, characterized by a "shift" of the voltage-dependent activation to more positive voltages, and nine variants resulted in a toxic gain-of-function, associated with a disrupted gate allowing inward transport at negative voltages. Functional results were not always in line with in silico pathogenicity scores, highlighting the complexity of pathogenicity assessment for accurate genetic counselling. The complex neurocognitive and psychiatric manifestations of this condition, and hitherto under-recognized impacts on growth, gastrointestinal function, and motor control are discussed. Including published cases, we summarize features in 122 individuals from 67 families with CLCN4-related neurodevelopmental condition and suggest future research directions with the aim of improving the integrated care for individuals with this diagnosis.

- Abstract
Fieldwork was integrated with remote sensing on satellite and drone derived 3D virtual outcrops to study the geometries of the Jurassic to Late Cretaceous sedimentary sequences belonging to the Organyà Basin, Catalunya, Spain. A total of 329 dips were measured in the field and 433 dips were extracted from the interpretation of virtual outcrops.

Diagnosis of genetic disorders is hampered by large numbers of variants of uncertain significance (VUSs) identified through next-generation sequencing. Many such variants may disrupt normal RNA splicing. We examined effects on splicing of a large cohort of clinically identified variants and compared performance of bioinformatic splicing prediction tools commonly used in diagnostic laboratories. Two hundred fifty-seven variants (coding and noncoding) were referred for analysis across three laboratories. Blood RNA samples underwent targeted reverse transcription polymerase chain reaction (RT-PCR) analysis with Sanger sequencing of PCR products and agarose gel electrophoresis. Seventeen samples also underwent transcriptome-wide RNA sequencing with targeted splicing analysis based on Sashimi plot visualization. Bioinformatic splicing predictions were obtained using Alamut, HSF 3.1, and SpliceAI software. Eighty-five variants (33%) were associated with abnormal splicing. The most frequent abnormality was upstream exon skipping (39/85 variants), which was most often associated with splice donor region variants. SpliceAI had greatest accuracy in predicting splicing abnormalities (0.91) and outperformed other tools in sensitivity and specificity. Splicing analysis of blood RNA identifies diagnostically important splicing abnormalities and clarifies functional effects of a significant proportion of VUSs. Bioinformatic predictions are improving but still make significant errors. RNA analysis should therefore be routinely considered in genetic disease diagnostics.

Trabajo presentado al 7th International Workshop on Occultations for Probing Atmosphere and Climate y al 9th Workshop of the International Radio Occultation Working Group (OPAC-IROWG), celebrados del 8 al 14 de septiembre de 2022 en Leibnitz, Austria., After more than 4 years on orbit, the Radio Occultations and Heavy Precipitation aboard PAZ satellite (ROHP-PAZ) experiment has already demonstrated the ability of polarimetric radio occultations (PRO) to detect precipitation. In fact, PRO have shown potential not only in rain detection, but also in precipitation characterization and in sensing the associated vertical cloud structures. PAZ PRO Δϕ observable profiles were made available in 2020 trough the ICE servers (https://paz.ice.csic.es), (https://genesis.jpl.nasa.gov). and more recently through the JPL A new re-processing of the PRO observations is being carried out with the aim to make it public during the second half of 2022. In addition to a better treatment of the rainy observations, the new re-processed profiles will come with an extensive collocation dataset that will allow the users to address scientific studies much more easily. These will take into account the limb-sounding geometry of the observations, performing the collocations directly into the RO rays obtained through a ray-tracer. These collocations include observations like the 30-minute geostationary 10.8 µm brightness temperature, GPM IMERG surface precipitation, microwave brightness temperatures from the numerous overpasses by the satellites in the GPM constellation, radar reflectivities from the GPM core satellite and the NEXRAD ground based weather radars, among others. Furthermore, the collocation algorithms are designed so that more external observations can be easily included. In addition to the exact collocations as described above, external databases are also checked so that coincidences with Tropical Cyclones, Mesoscale Convective Systems and other relevant precipitating systems are identified nearby PAZ observations. In this presentation, we will show a brief overview of the re-processing of the ROHP-PAZ data, with emphasis in the differences between the Δϕ profiles obtained from UCAR’s CDAAC excess phases and from those obtained from JPL excess phases. After that, examples of the coincident datasets will be presented. Results will include statistics gathered from the differentiation of different precipitation regimes (e.g. stratiform vs convective), identification and validation of cloud top height determination, and comparison with other relevant parameters obtained from the collocated observations., The ROHP-PAZ project is part of the Grant RTI2018-099008-B-C22 funded by the Spanish Ministry of Science and Innovation MCIN/AEI/10.13039/501100011033 and by “ERDF A way of making Europe” of the “European Union”.
