Hypersplenism (HS) is a concomitant symptom of liver or blood disease. Not only does the treatment of HS face challenges, but the transcriptome of individual cells is also unknown. Here, the transcriptional profiles of 43,037 cells from four HS tissues and one control tissue were generated by the single-cell RNA sequencing and nine major cell types, including T-cells, B-cells, NK cells, hematopoietic stem cells, neutrophil cells, mast cells, endothelial cells, erythrocytes, and dendritic cells were identified. Strikingly, the main features were the lack of CCL5+ B-cells in HS and the presence of SESN1+ B cells in HS with hepatocellular carcinoma (HS-HCC). In cell-cell interaction analysis, CD74-COPA and CD94-HLA-E in HS were found to be up-regulated. We further explored HS-specifically enriched genes (such as FKBP5, ADAR, and RPS4Y1) and found that FKBP5 was highly expressed in HCC-HS, leading to immunosuppression. Taken together, this research provides new insights into the genetic characteristics of HS via comprehensive single-cell transcriptome analysis.

[Purpose/Significance] Based on the moderating effect of regional innovation cooperation network's clustering coefficient and average shortest path length, the paper analyzes the impact of enterprise centrality in network on technological innovation performance, in order to provide reference to enterprise innovation management. [Method/Process] Using patent data to construct patent cooperation networks, this paper builds a theoretical model measuring technological innovation performance of an enterprise, with degree centrality and betweenness centrality of the enterprise in the network as independent variables, with the clustering coefficient and average shortest path length of patent cooperation network as a moderator variable. [Results/Conclusions] The empirical study in the field of integrated circuit technology in the Yangtze River Delta is carried out and the stepwise multiple linear regression results show the following conclusions. The density of cooperation network, the accumulation of technological innovation, the number of R&D personnel, and the intensity of R&D investment in the province where the firm is located have a significant positive impact on the firm's technological innovation performance. Compared with enterprises and research institutes, when enterprises cooperate with universities, their technological innovation performance is more favorable. The degree centrality and the betweenness centrality of a firm in the cooperation network have a significant positive impact on the firm's technological innovation performance. The average shortest path length has a significant moderating effect on the relationship between intermediary centrality and firms' technological innovation performance. The longer the average shortest path length is, the more obvious the positive effect of intermediary centrality on firms' technological innovation performance is. Through the empirical analysis, the paper reveals the influence mechanism of regional innovation cooperation network on firms' technological innovation performance. The position (center, intermediary) of firms in the cooperation network has a significant influence on firms' technological innovation performance, and the structural attribute of the cooperation network has a significant moderating effect on this effect. Based on the analysis results, countermeasures and suggestions are put forward to improve the technological innovation performance of enterprises.

Influenza A virus outbreaks pose a significant burden to global health. However, detecting and subtyping the virus is challenging due to its high mutation rate, and it is difficult to distinguish viable viruses that cause infection from clinical samples. Here is a novel PMAxx-LAMP assay for the classification of viable and non-viable Influenza A virus together with differentiation between H1N1 and H3N2 subtypes. Our loop-mediated isothermal amplification (LAMP) primers displayed limits of detection of 102 copies/μL for both subtypes, high specificity, reproducibility, and good linearity with correlation coefficients of 0.9762 and 0.9901 for H1N1 and H3N2, respectively. PMAxx effectively distinguished between viable and non-viable viruses at an optimal concentration of 10 μM. In the LAMP method, the clinical specificity and sensitivity for the H1N1 subtype were both 100%, while for the H3N2 subtype, they were 94.44 and 92.31%, respectively. With the PMAxx-LAMP assay, the clinical specificity and sensitivity were 68% and 65% for the H1N1 subtype, and 81% and 69% for the H3N2 subtype, respectively. The reduced clinical specificity and sensitivity observed in the PMAxx-LAMP assay can be attributed to the presence of non-viable viruses, which are labeled with PMAxx and subsequently eliminated from positive samples. The PMAxx-LAMP offers potential benefits in determining the stage of infection, enabling early screening of patients, and shortening the window period for virus detection. The simplicity and efficiency of this assay make it suitable for use in resource limited settings and point-of-care (POCT) applications. [ABSTRACT FROM AUTHOR]
