A method has been devised for observing both circadian and noncircadian rhythms in a single wild type strain of Neurospora crassa. This method allows a direct comparison of the properties of the two types of rhythm. The circadian rhythm of conidiation always entrains to a light-dark cycle, damps out in constant light, and has a temperature-compensated period length. The noncircadian rhythm of hyphal branching, expressed by the same strain under different environmental conditions, does not entrain to a light-dark cycle, persists in constant light, and its period length is temperature-dependent. These results suggest that the two rhythms have different underlying mechanisms and demonstrate that the differences in the rhythms previously observed in different strains (patch, band, and clock) are not due to genetic differences between these strains but rather are inherent properties of the rhythms themselves.

In vitro, high density monolayer cultures of vascular smooth muscle cells can be induced to form multicellular nodules. The nodular cells appear to be morphologically differentiated smooth muscle cells. Sodium dodecyl sulfate (NaDodSO4)-polyacrylamide gel electrophoresis was used to compare the proteins synthesized and secreted by monolayer and nodular cultures of smooth muscle cells. Although most proteins appeared to be similar, the nodular cultures contained a unique heparin binding protein of Mr = 38,000 (38kD protein) (Millis, A.J.T., Hoyle, M., Reich, E., and Mann, D.M., 1985, J. Biol. Chem., 260:3754-3761). The 38kD protein was glycosylated and its apparent molecular weight was shifted to Mr = 32,500 after synthesis in the presence of tunicamycin or digestion with endoglycosidase F. The production of 38kD protein by nodular cell cultures did not appear to result from the degradation of a high molecular weight precursor in nodular conditioned medium. Further, it was not detected in monolayer cell conditioned medium that had been incubated with nodular cells. Finally, its synthesis was not induced in monolayer cell cultures that had been labeled in nodular cell conditioned medium. The 38kD protein appears to be uniquely associated with nodular cultures of smooth muscle cells.

The relationship of indoleacetic acid oxidase activity to peroxidase activity is complicated by numerous multiple forms of this enzyme system. It is not known if all isozymes of this complex system contain both types of activity. Isozyme analysis of commercial horseradish peroxidase and leaf extracts of yellow birch (Betula alleghaniensis) by isoelectric focusing in polyacrylamide gels was used to examine this problem. Horseradish and birch exhibited 20 and 13 peroxidase isozymes, respectively, by staining with benzidine or scopoletin. Guaiacol was less sensitive. Indoleacetic acid oxidase staining (dimethylaminocinnamaldehyde) generally showed fewer bands, and left doubt as to the residence of both types of activity on all isozymes. Elution of the isozymes from the gels and wet assays verified that all peroxidase isozymes contained indoleacetic acid oxidase activity as well. Estimation of oxidase to peroxidase ratios for the major bands indicated small differences in this parameter. A unique isozyme for one or the other type of activity was not found.

A cell surface associated "glucose regulated protein" has been described on nontransformed human fibroblasts. To examine the distribution of that protein on human fibroblasts specific antisera were used. The antisera was used in conjunction with indirect immunofluorescence and revealed that the glucose regulated protein was present as fibers on spread cells. Further, the antisera was used in complement mediated cytotoxicity assays to examine cells during specific stages of the mitotic cell cycle. Fibroblasts were synchronized by serum starvation, hydroxyurea inhibition, or colcemid inhibition followed by mitotic selection. The results demonstrated that the glucose regulated protein was maximally displayed during the G1 phase of the cell cycle and minimally displayed during the S and M phases.

A fourth mutant of Neurospora crassa, designated frq-4, has been isolated in which the period length of the circadian conidiation rhythm is shortened to 19. +/- 0.3 hours. This mutant is tightly linked to the three previously isolated frq mutants, and all four map to the right arm of linkage group VII about 10 map units from the centromere. Complementation tests suggest, but do not prove, that all four mutations are allelic, since each of the four mutants is co-dominant with the frq+ allel--i.e., heterokaryons have period lengths intermediate between the mutant and wild-type--and since heterokaryons between pairs of mutants also have period lengths intermediate between those of the two mutants.

Systemic and coronary hemodynamic effects of prenalterol, a beta-1 receptor agonist, were determined in patients with chronic congestive heart failure, initially after intravenous administration (10 patients) and then after oral administration (eight patients). Cardiac index increased by 33 percent and 30 percent after intravenous and oral prenalterol, respectively. The increase in stroke volume index and stroke work index and decrease in pulmonary capillary wedge pressure and systemic vascular resistance were not significant. Myocardial oxygen consumption and coronary sinus blood flow increased in the majority of patients, although these changes were not statistically significant. There were no significant changes in transmyocardial norepinephrine or epinephrine balance. The systemic and coronary hemodynamic effects of both intravenous and oral prenalterol were similar. Major side effects included sudden death (two patients) and hypotension and bradycardia (three patients) during oral prenalterol treatment. It is concluded that improved left ventricular function following both intravenous and oral prenalterol may be associated with increased myocardial oxygen consumption, and serious adverse effects may occur during prenalterol therapy.

Several improved techniques for isoelectric focusing of isoenzymes in polyacrylamide gel slabs were developed. Using these techniques, three commercial sources of horseradish peroxidase were each examined with three commercial sources of carrier ampholytes to determine their respective isoenzyme profiles.A much higher degree of isoenzyme resolution was obtained than reported previously. A composite of all of the various tests gave a total of 42 peroxidase isoenzymes. Commercial sources of horseradish peroxidase showed many similarities in their isoenzyme patterns, but their differences were sufficient to recognize each source easily. Isoenzyme patterns for all three sources spanned the entire pH range with all three sources of wide range carrier ampholytes. Only a few isoenzymes stained darkly. Most isoenzymes stained moderately to lightly, but all were well resolved. Gels stained for indoleacetic acid oxidase activity showed the same pattern as gels stained for peroxidase activity. This was true for all three commerial sources of the enzyme. The isoenzyme patterns obtained in each run were entirely reproducible, and linear pH gradients were obtained in all cases. Limitations in the pH range of the wide range carrier ampholytes relative to the isoelectric points of the extreme anodic and cathodic isoenzymes led to the adoption of a modified definition of focusing time. In addition, the labeling of isoenzymes commonly used in electrophoresis was adapted for labeling isoenzymes resolved by isoelectric focusing.

Comparison of the proteins secreted by early and late passage cultures of human fibroblasts reveals enhanced production of three proteins (Mr = 55,000, 58,000, and 61,000) in the late passage culture conditioned medium. The 55,000 and 58,000 Da proteins react with anticollagenase antibodies and are identified as procollagenase (Sottile, J. M. and Millis, A. J. T., J. Cell Biol. 106: 1518a, 1987). The production of immunoreactive collagenase was assayed using immunoprecipitation and immunofluorescence. Immunofluorescence microscopy revealed that each culture was heterogenous. The percentage of collagenase positive cells ranged from 1% (early passage) to 35% (late passage). Late passage cultures also secreted higher levels of immunoprecipitable collagenase into the culture medium than early passage cultures. High levels of collagenase production were also observed in fibroblasts cultured from donors with Werner's syndrome and from an aged donor. Media conditioned by the growth of human fibroblasts contained collagenase stimulating activity. In the presence of conditioned medium, the percentage of collagenase positive cells was increased to 10-20% (early passage) and 85% (late passage), indicating that each culture contains both responding and non-responding cells.

Three mutants of Neurospora crassa have been isolated which have altered period lengths of their circadian rhythm of conidiation. The strains, designated "frequency" (frq), were obtained after mutagenesis of the band (bd) strain with N-methyl-N'-nitro-N-nitrosoguanidine. In continuous darkness at 25 degrees bd has a period length of 21.6 +/- 0.5 hours; under the same conditions the period length of frq-1 is 16.5 +/- 0.5 hours; frq-2, 19.3 +/- 0.4 hours; and frq-3, 24.0 +/- 0.4 hours. Each of the mutants segregates as a single nuclear gene. All three mutants appear very tightly linked to each other, but it has not yet been determined whether the mutants are allelic. No major changes in the responses to light and temperature have been observed in any of the mutants. It is suggested that these mutants represent alterations in the basic timing mechanism of the circadian clock of Neurospora.
