Shone's complex is a rare syndrome characterized by congenital left heart defects that can differ among the patients.To use echocardiography in the diagnosis of Shone's complex and analyze the causes of missed diagnosis and misdiagnosis.This was a retrospective study of patients who underwent echocardiography and repair surgery from February 14, 2008, to November 22, 2019. The patients were followed once a year at the outpatient clinic after surgery.Sixty-six patients were included. The patients were 2.7 (0.8-5.6) years of age, and 54.5% were male. Ten (15.2%) had a history of heart surgery. The most common heart defect was the Annulo-Leaflet mitral ring (ALMR) (50/66, 75.8%), followed by coarctation of the aorta (CoA) (43/66, 65.2%). The patients had a variety of combinations of defects. Only two (3.0%) patients had all four defects. None of the patients had a family history of congenital heart disease. The preoperative echocardiographic findings were examined against the intraoperative findings. Echocardiography missed an ALMR in 31 patients (47.0%), a parachute mitral valve (PMV) in one patient (1.5%), subaortic stenosis in one patient (1.5%), and CoA in two patients (3.0%).Echocardiography is an effective method to diagnose the Shone's complex. Due to this disease's complexity and interindividual variability, Improving the understanding of the disease can reduce misdiagnosis and missed diagnosis.

With the continuous development of computer technology, analysis techniques based on various types of sports data sets are also evolving. One typical representative is image-based motion recognition technology, which enables video action recognition with a certain degree of feasibility. In basketball technical action videos, technical action has obvious characteristics. The athletes in the footage in sports videos are relatively fixed, and the scenes are relatively homogeneous, so technical action analysis of basketball technical action videos has certain advantages. However, there are many challenges in basketball technical action recognition, mainly including the fact that basketball techniques are numerous and complex. To address the above issues, this research proposes a 3D convolutional neural network framework that two different resolution image inputs are performed on the basketball technical action dataset. The experimental results show that the algorithmic process designed in this study is effective for action recognition on the basketball technical action dataset.

The Internet and 5G era makes e-learning a vital part of modern education, and extensive evidence has shown that peer teaching and flipped classroom contribute to increased success in medical teaching. However, the applicability of these pedagogies in laboratory courses remains largely unexplored. This study aimed to evaluate the academic performance, proficiency in procedural skills, and perception of nursing students in physiology laboratory classes delivered with nontraditional classroom (NTC) pedagogies comprising the combination of e-learning, peer teaching, and flipped classroom. Each class was subdivided into two equal halves by successive student identification (ID) number and randomly assigned to the control or NTC group. Compared to the control class, NTC teaching significantly enhanced mean score of six preclass tests (67.77 ± 9.83 vs. 62.94 ± 9.70), with “B” graders increased obviously, suggesting that preclass e-learning was more effective than textbook-based preview, especially for general grasp of the topic. Similarly, average scores on postclass quizzes in the NTC group were improved (79.40 ± 9.12 vs. 74.43 ± 8.88). Lesser time cost and higher success rates were observed in trachea, artery, and heart catheterizations in the NTC group, although no disparities were found in ureteral intubation. The majority (∼74%) of students supported the reform and shared positive experiences with NTC methodology. They reported that virtual experiments and self-paced procedural skill videos affected pre- and in-class learning outcomes most, respectively. These findings indicated that NTC pedagogy was workable to improve students’ subject scores and proficiency in complicated and direct-viewing procedural skills and was favorable to students.

Chlorogenic acid (CGA) displays cognition-improving properties, but the underlying mechanisms remain unclear. Herein, CGA supplementation (150 mg/kg body weight) for 14 weeks significantly prevented obesity and insulin resistance, cognitive-behavioral disturbances, and synaptic dysfunction induced by a high-fat and high-fructose diet (HFFD). Moreover, CGA supplementation enhanced the expression of genes enriched in the neuroactive ligand-receptor interaction pathway and reduced inflammatory factor expressions. Furthermore, CGA treatment increased gut microbiota diversity and the level of bacterial genera producing SCFAs. CGA also decreased the concentration of energy metabolism substrates, while it increased phosphorylcholine. Finally, we observed a significant correlation among synaptic transmission genes, gut microbiota, and neurotransmission in the CGA supplementation group by targeted multiomics analysis. Together, our results supported that the alteration of gut microbiota and metabolite composition is the underlying mechanism of CGA improving cognitive function. CGA is also a promising intervention strategy to prevent HFFD-induced cognitive impairment.

Hydroxylammonium nitrate(HAN)-based thrusters have been continuously researched as a replacement for conventional hydrazine thrusters. At present, catalytic ignition starting is the most commonly used starting method for hydroxylammonium nitrate-based thrusters, and it is also the most reliable and effective starting method. During the catalytic ignition and start-up process of hydroxylammonium nitrate-based thruster, the catalytic decomposition reaction of hydroxylammonium nitrate always affects the state of the thruster. It is worth noting that the catalytic bed is the key structure of the hydroxylammonium nitrate-based thruster to achieve catalytic ignition. It is necessary to optimize the performance of the hydroxylammonium nitrate-based thruster by optimizing the structure of the catalytic bed. However, there are some limitations to study the working process of hydroxylammonium nitrate-based thruster by experiment. Therefore, the working process of 150N-class hydroxylammonium nitrate-based thruster by numerical simulation was studied in this paper. Then, under the condition that the inlet mass flow rate of the monopropellant was consistent, simulation calculations were carried out on hydroxylammonium nitrate-based thrusters with different catalytic bed structures based on the studied simulation method. Finally, with thrust as the evaluation parameter, the structural parameters of the catalytic bed of the hydroxylammonium nitrate-based thruster were optimized based on the response surface method. After optimized calculation, the hydroxylammonium nitrate-based thruster with a catalytic bed length of 72.43mm, a catalytic bed diameter of 74mm, and a catalytic bed porosity of 0.6 has better performance, which provides a reference for the design of hydroxylammonium nitrate-based thrusters.

The design of an efficient absorbent is the premise for recovery and resource utilization of hydrogen chloride (HCl) from its industrial tail gases. Herein, a series of 1-butyl-3-methylimidazolium chloride (BmimCl) based-deep eutectic solvents (DESs) were designed and the solubility behavior for HCl was studied in terms of their structure, basicity, free volume, intermolecular interaction energy, and absorption enthalpy. The relationship between the interaction energy and the phase change in the HCl dissolution process was explored in detail. BmimCl-TAA (thioacetamide) (1 : 1) shows high reversible solubility due to its high free volume, suitable absorption enthalpy, and closer H-bonding (HB) interactions between BmimCl and TAA or HCl. The dissolution mechanism of HCl and the dynamic evolution of the HB network were verified by FT-IR and NMR spectra and quantum chemical calculations. The results show that it is the competitive HB interaction that promotes the dissolution of HCl, reduces the absorption enthalpy, and renders a reversible absorption. Compared with BmimCl, the absorption enthalpy of HCl in BmimCl-TAA (1 : 1) is reduced by 25% and the reversible solubility increased 150%. To study the effects of different diluents on the propagation characteristics of H2/CO/air mixture turbulent premixed flames, a series of experiments were carried out in a turbulent premixed flame experimental system. The effects of turbulence intensity (0.49–1.31 m/s), dilution gas content (10%, 20%, and 30%), hydrogen fraction (50%, 70%, and 90%), and equivalence ratio (0.6, 0.8, and 1.0) on the turbulent premixed flame were studied. The results show that with the increase in hydrogen fraction or turbulence intensity or equivalence ratio, the ST and ut increase at the same radius. Compared with N2 dilution, CO2 dilution showed a more obvious inhibition effect on ST. With the increase of Ka, ST;35mm/u’ gradually decreased, and the extent of ST;35mm/u’ decrease gradually became smaller. As the intensity of turbulence increases or the hydrogen fraction increases, the slope of ST,35mm/u’ with Da/Le gradually decreases. In the turbulence intensity range of this experiment, the ut,35mm/μl under nitrogen dilution condition has a larger floating range. The growth rate of ut,35mm/μl at a low equivalence ratio is significantly higher than that at a high equivalence ratio.

Vanillin is one of the most commonly used natural-occurring flavors in the world. This study successfully constructed an efficient whole-cell catalytic system for vanillin biosynthesis from ferulic acid by regulating feruloyl-CoA synthetase (FCS) and enoyl-CoA hydratase (ECH). First, we constructed an efficient cell-free catalytic system with FCS-Str (fcs from Streptomyces sp. V-1) and ECH-Str (ech from Streptomyces sp. V-1) combination at 1:1. The efficient cell-free catalytic system provided necessary strategies for optimizing the whole-cell catalytic system. Then, we constructed the recombinant Escherichia coli by heterologously expressing the fcs-Str and ech-Str combination. Moreover, E. coli JM109 was a better recombinant Escherichia coli than E. coli BL21 with higher vanillin production. Finally, we first adjusted the ratio of FCS and ECH in E. coli JM109 to 1:1 using two copies of fcs-Str. For higher vanillin production, we further optimized the induction conditions of E. coli JM109 to increase the amount of FCS and ECH. The optimized E. coli JM109-FE-F constructed in this study has the highest vanillin synthesis rate of converting 20 mM ferulic acid to 15 mM vanillin in 6 h among all of the E. coli catalytic systems. Our study made a significant contribution to the construction of the vanillin biosynthesis system and provided a valuable strategy for increasing vanillin production. KEY POINTS: • The efficient cell-free vanillin biosynthesis system was constructed by FCS-Str and ECH-Str combination at 1:1. • Escherichia coli JM109 was determined as a better recombinant Escherichia coli than E. coli BL21 with higher vanillin production. • Escherichia coli JM109-FE-F with two copies of fcs-Str and one copy of ech-Str has the highest catalytic efficiency for vanillin production.

A new cytochalasin dimer, verruculoid A (1), three new cytochalasin derivatives, including 12-nor-cytochalasin F (2), 22-methoxycytochalasin B6 (3), and 19-hydroxycytochalasin B (4), and 20-deoxycytochalasin B (5), a synthetic product obtained as a natural product for the first time, together with four known analogues (6-9), were isolated and identified from the culture extract of Curvularia verruculosa CS-129, an endozoic fungus obtained from the inner fresh tissue of the deep-sea squat lobster Shinkaia crosnieri, which was collected from the cold seep area of the South China Sea. Structurally, verruculoid A (1) represents the first cytochalasin homodimer containing a thioether bridge, while 12-nor-cytochalasin F (2) is the first 12-nor-cytochalasin derivative. Their structures were elucidated by detailed interpretation of the NMR spectroscopic and mass spectrometric data. X-ray crystallographic analysis and ECD calculations confirmed their structures and absolute configurations. Compound 1 displayed activity against the human pathogenic bacterium Escherichia coli (MIC = 2 μg/mL), while compounds 4, 8, and 9 showed cytotoxicity against three tumor cell lines (HCT-116, HepG-2, and MCF-7) with IC50 values from 5.2 to 12 μM. The structure-activity relationship was briefly discussed.

To achieve security and privacy for data stored on the cloud, we need the ability to secure data in compute. Equality comparisons, “ $x=y, x\ne y$ x = y , x ≠ y ”, have been widely studied with many proposals but there is much room for improvement for order comparisons, “ $x y \text{ and } x \geq y$ x y , x ≤ y , x > y and x ≥ y ”. Most protocols for order comparisons have some limitation, either leaking some information about the data or requiring several rounds of communication between client and server. In addition, little work has been done on retrieving with compound conditions, mixing several equality and order comparisons. Fully homomorphic encryption (FHE) promises the ability to compute arbitrary functions on encrypted data without sacrificing privacy and without communication, but its potential has yet to be fulfilled. Particularly, private comparisons for database queries using FHE are expensive to compute. In this article, we design an efficient private database query (PDQ) protocol which supports compound conditions with equality and order comparisons. To this end, we first present a private comparison algorithm on encrypted integers using FHE, which scales efficiently for the length of input integers, by applying techniques from finite field theory. Then, we consider a scenario for PDQ protocols, querying for values based on a conjunction of one order and four equality conditions on key columns. The proposed algorithm and protocol are implemented and tested to determine their performance in practice. The proposed comparison algorithm takes about $25.259$ 25 . 259 seconds to compare 697 pairs of 64-bit integers using Brakerski-Gentry-Vaikuntanathan's leveled FHE scheme with single instruction multiple data (SIMD) techniques at more than 138 bits of security. This yields an amortized rate of just 36 milliseconds per comparison. On top of that, we show that our techniques achieve an efficient PDQ protocol for one order and four equality comparisons, achieving an amortized time and communication cost of 57 milliseconds and 448 bytes per database element.

Flow cytometry assay and RT-PCR were utilized to evaluate the CD44+CD133+ stem cell population, the expression of inflammatory cytokines and series tumor stemness biomarkers. Meanwhile, Western blot was used to analyze the alteration of cell signaling associated-molecules by treatment with TSA, an original antifungal antibiotic and a panel inhibitor of histone deacetylase.Results: Our study found that LPS promoted the migration, invasion and stem-like tumoroshpere forming in multiple PCa cell lines including DU145, PC3, 22RV1, LNCaP. LPS also enriched CD44+CD133+ stem cell population and increased the expression of series tumor stemness biomarkers (e.g., CD44, CD133, SOX-2, α-intergrin, Nestin, etc.). TSA was found to prevent tumor cell migration, invasion and tumorosphere forming in DU145 and PC3 cells with increasing tumor suppressive Maspin and reducing both phosphorylation of Stat3 (p-Stat3) and pro-oncogene c-Myc expression in LPS-treated DU145 cells. Furthermore, blocking Stat3 signaling pathway by treatment with TSA and/or small molecule compound Stattic of an p-Stat3 inhibitor effectively abrogated LPS-induced tumorosphere forming with decrease of IL-6, IL-8 and stemness biomarkers CD44, SOX-2 expression.Conclusion: Our data demonstrated that the inflammatory agent of bacterial LPS augmented malignant transformation and promoted the cancerous stemness in PCa epithelial cells. TSA could prevent, at least in part, the LPS-induced malignant transformation by targeting p-Stat3/c-Myc signaling pathway and reducing inflammatory IL-6, IL-8. BackgroundTo enrich the probiotic lactic acid bacteria (LAB) strains and expand the commercialization of new fermented juice products, we have identified two LAB strains with excellent potential in fermenting apple juice from pickles.MethodsThe two strains were morphologically, physiologically, and genetically characterized. The strains’ fermentation performance and alterations in volatile aroma components of apple juice and ability to survive in a simulated gastrointestinal environment were evaluated.ResultsTwo strains were identified as Lacticaseibacillus paracasei (WFC 414) and Lactiplantibacillus plantarum (WFC 502). The growth of WFC 414 and WFC 502 in apple juice for 48 h reached 8.81 and 9.33 log CFU/mL, respectively. Furthermore, 92% and 95% survival rates were achieved in 2 h simulated gastric juice, and 80.7 and 83.6% survival rates in 4 h simulated intestinal juice. During the fermentation, WFC 414 and WFC 502 reduced the soluble sugars and total polyphenols in apple juice, and consumed malic acid to produce large amounts of lactic acid (3.48 and 5.94 mg/mL). In addition, the esters and aldehydes were reduced, and the production of alcohols, acids and ketones was elevated in the apple juice fermented by both strains.ConclusionThese results show that WFC 414 and WFC 502 have great potential applications in the fermented fruit juice industry.
