The goal of this study is the selective oxyfunctionalization of steroids under mild and environmentally friendly conditions using fungal enzymes. With this purpose, peroxygenases from three basidiomycete species were tested for the hydroxylation of a variety of steroidal compounds, using H2O2 as the only cosubstrate. Two of them are wild-type enzymes from Agrocybe aegerita and Marasmius rotula, and the third one is a recombinant enzyme from Coprinopsis cinerea. The enzymatic reactions on free and esterified sterols, steroid hydrocarbons, and ketones were monitored by gas chromatography, and the products were identified by mass spectrometry. Hydroxylation at the side chain over the steroidal rings was preferred, with the 25-hydroxyderivatives predominating. Interestingly, antiviral and other biological activities of 25-hydroxycholesterol have been reported recently (M. Blanc et al., Immunity 38:106–118, 2013, http://dx.doi.org/10.1016/j.immuni.2012.11.004). However, hydroxylation in the ring moiety and terminal hydroxylation at the side chain also was observed in some steroids, the former favored by the absence of oxygenated groups at C-3 and by the presence of conjugated double bonds in the rings. To understand the yield and selectivity differences between the different steroids, a computational study was performed using Protein Energy Landscape Exploration (PELE) software for dynamic ligand diffusion. These simulations showed that the active-site geometry and hydrophobicity favors the entrance of the steroid side chain, while the entrance of the ring is energetically penalized. Hydroxylation of vitamin D by Agrocybe aegerita and Coprinopsis cinerea peroxygenases was investigated in a combined experimental and computational study. 25-Monohydroxylated vitamin D3 (cholecalciferol) and D2 (ergocalciferol), compounds of high interest in human health and animal feeding, can be obtained through reaction with both fungal enzymes. Differences in conversion rates and regioselectivity were nevertheless observed, and, to rationalize the results, diffusion of D2 and D3 on the molecular structure of the two enzymes was performed with PELE software. In good agreement with experimental conversion yields, simulations indicate more favorable energy profiles for the substrates’ entrance in C. cinerea than for A. aegerita enzyme. Furthermore, GC-MS analyses show that while a full regioselective conversion into the active C25 form is catalyzed by C. cinerea peroxygenase for D2 and D3, A. aegerita yielded a mixture of the hydroxylated D3 products. From the molecular simulations, relative distance distributions between the haem compound I oxygen and H24/H25 atoms (hydrogens on C24 and C25 respectively) were plotted. Results show large populations for O-H25 distances below 3 Å for D2 and D3 in C. cinerea in accord with the high reactivity observed for this enzyme. In A. aegerita, however, cholecalciferol has similar populations (below 3 Å) for O-H25 and O-H24 which can justify the small degree of hydroxylation observed in C24. There is an increasing interest in enzymes that catalyze the hydroxylation of naphthalene under mild conditions and with minimal requirements. To address this challenge, an extracellular fungal aromatic peroxygenase with mono(per)oxygenase activity was engineered to convert naphthalene selectively into 1‐naphthol. Mutant libraries constructed by random mutagenesis and DNA recombination were screened for peroxygenase activity on naphthalene together with quenching of the undesired peroxidative activity on 1‐naphthol (one‐electron oxidation). The resulting double mutant (G241D‐R257K) obtained from this process was characterized biochemically and computationally. The conformational changes produced by directed evolution improved the substrate's catalytic position. Powered exclusively by catalytic concentrations of H2O2, this soluble and stable biocatalyst has a total turnover number of 50 000, with high regioselectivity (97 %) and reduced peroxidative activity.

Selective oxygenations of aliphatic compounds are among the most challenging reactions in organic chemistry for the regio and/or stereo specific synthesis of pharmaceuticals and fine chemicals. Recently, a new peroxidase type, which shares the active-site architecture and reaction mechanism of cytochromes P450, but has the advantage of being activated directly by H2O2, was isolated from Agrocybe aegerita (Ullrich, 2004), and later identified in a variety of sequenced basidiomycete genomes, including that of Coprinopsis cinerea (Floudas, 2012). The A. aegerita heme-thiolate (HTP) enzyme has been the most widely investigated basidiomycete peroxygenase, and recent studies have shown that the C. cinerea enzyme has comparative advantages related to its high conversion yield/selectivity for some hydroxylation reactions, and its production as a recombinant protein in an industrial expression host (Babot, 2013; 2014). 25-monohydroxylated vitamin D3 (cholecalciferol) and D2 (ergocalciferol), compounds of high interest in human health and animal feeding, can be obtained through reaction with both A. aegerita and C. cinerea enzymes. Unspecific peroxygenase (UPO) represents a new type of heme-thiolate enzyme with self-sufficient mono(per)oxygenase activity and many potential applications in organic synthesis. With a view to taking advantage of these properties, we subjected the Agrocybe aegerita UPO1-encoding gene to directed evolution in Saccharomyces cerevisiae. To promote functional expression, several different signal peptides were fused to the mature protein, and the resulting products were tested. Over 9,000 clones were screened using an ad hoc dual-colorimetric assay that assessed both peroxidative and oxygen transfer activities. After 5 generations of directed evolution combined with hybrid approaches, 9 mutations were introduced that resulted in a 3,250-fold total activity improvement with no alteration in protein stability. A breakdown between secretion and catalytic activity was performed by replacing the native signal peptide of the original parental type with that of the evolved mutant; the evolved leader increased functional expression 27-fold, whereas an 18-fold improvement in the kcat/Km value for oxygen transfer activity was obtained. The evolved UPO1 was active and highly stable in the presence of organic cosolvents. Mutations in the hydrophobic core of the signal peptide contributed to enhance functional expression up to 8 mg/liter, while catalytic efficiencies for peroxidative and oxygen transfer reactions were increased by several mutations in the vicinity of the heme access channel. Overall, the directed-evolution platform described is a valuable point of departure for the development of customized UPOs with improved features and for the study of structure-function relationships. © 2014, American Society for Microbiology.

Die enzymatischen Transformationen mit der pilzlichen Peroxygenase aus Agrocybe aegerita haben gezeigt, dass das Enzym insgesamt über ein sehr breites Substratspektrum bezüglich der Flavonoide verfügt. Die Flavonoide werden mittels AaeAPO regioselektiv in 6-Position hydroxyliert. Der Reaktionsmechanismus der AaeAPO bei Flavonoiden läuft über eine Epoxidstufe ab, wobei der eingefügte Sauerstoff bei Hydroxylierungen dem Wasserstoffperoxid entstammt (Hinweis auf eine echte Peroxygenase-Reaktion). Die Enzymproduktion der Agrocybe aegerita wird durch den Extraktzusatz (aus jeweils Aronia melanocarpa, Fragaria ananassa, Trifolium pratense und Bellis perennis) im Fall der Laccase stimuliert. Die AaeAPO -Aktivität wird dagegen gehemmt. Für Bjerkandera adusta kann eine moderate Stimulierung der MnP-Bildung nach der Extraktzugabe beobachtet werden. Der positive bzw. negative Einfluss auf die Enzymproduktion ist bei Agrocybe aegerita sowohl auf Flavonoide als auch auf das Lösungsmittel Ethanol zurückzuführen.

Unspecific peroxygenase (EC 1.11.2.1) represents a group of secreted hemethiolate proteins that are capable of catalyzing the selective mono-oxygenation of diverse organic compounds using only H2O2 as a cosubstrate. In this study, the peroxygenase from Agrocybe aegerita (AaeUPO) was found to catalyze the hydroxylation of various linear (e.g n-hexane), branched (e.g. 2,3-dimethylbutane) and cyclic alkanes (e.g. cyclohexane). The size of n-alkane substrates converted by AaeUPO ranged from gaseous propane (C3) to n-hexadecane (C16). They were mono-hydroxylated mainly at the C2 and C3 position, rather than at the terminal carbon, and the corresponding ketones were formed as a result of overoxidation. In addition, a number of alkenes were epoxidized by AaeUPO, including linear terminal (e.g. 1-heptene), branched (2-methyl-2-butene) and cyclic alkenes (e.g. cyclopentene), as well as linear and cyclic dienes (buta-1,3-diene, cyclohexa-1,4-diene). Furthermore, the conversion of terminal alkynes (e.g. 1- octyne) gave the corresponding 1-alkyn-3-ol in low yield. Some of the reactions proceeded with complete regioselectivity and - in the case of linear alkanes, terminal linear alkenes and alkynes - with moderate to high stereoselectivity. The conversion of n-octane gave (R)-3-octanol with 99% enantiomeric excess (ee) and the preponderance of the (S)-enantiomer reached up to 72% ee of the epoxide product for the conversion of 1-heptene. Catalytic efficiencies (kcat/ Km) determined for the hydroxylation and respectively epoxidation of the model compounds cyclohexane and 2-methyl-2-butene were 2.0 × 103 M-1 s-1 and 2.5 × 105 M−1 s−1. The results obtained in the deuterium isotope effect experiment with semideuterated n-hexane and the radical clock experiment with norcarane clearly demonstrated that the hydroxylation of alkanes proceeds via hydrogen abstraction, the formation of a substrate radical and a subsequent oxygen rebound mechanism. B. 1-Hepten), verzweigter (z. B. 2-Methyl-2-Buten) und zyklischer Alkene (z. B. Cyclopenten) sowie linearer und zyklischer Diene (1,3-Butadien, 1,4-Cyclohexadien) durch die AaeUPO epoxidiert. Die Umsetzung terminaler Alkine (z. B. 1-Octin) führte zur Entstehung der jeweiligen 1-Alkin-3-ole. Manche dieser Reaktionen verliefen ausgeprägt regioselektiv und, im Falle der linearen Alkane sowie der linearen terminalen Alkene und Alkine, mit mittlerer bis hoher Stereoselektivität. So ergab beispielsweise die Umsetzung von n-Octan einen Enantiomerenüberschuss größer 99% für (R)-3-Octanol; die Epoxidierung von 1-Hepten lieferte einen Enatiomeerenüberschuss (ee) von bis zu 72% für das (S)-Enantiomer. Die katalytischen Effizienzen, die für die Hydroxylierung bzw. Epoxidierung der Modellverbindungen Cyclohexan und 2-Methyl-2-Buten ermittelt wurden, betragen 2.0 × 103 M-1 s-1 und 2.5 × 105 M−1 s−1. Over the recent years, increasing scientific attention has been paid to pharmaceuticals, other drugs and their metabolites. These substances are of particular interest because of their physiological, toxicological and ecotoxicological effects in the human body and respectively in the environment. Cytochrome P450 enzymes (P450s) play a key role in the conversion and detoxification of bioactive compounds including many pharmaceuticals and drugs. Most of these enzymes belong to the monooxygenases; they are intracellular and rather unstable biocatalysts that are difficult to purify and require expensive, complex cofactors, which alltogether hampers their use in isolated form. The investigations carried out here with fungal peroxygenases have shown that this enzyme sub-subclass (EC 1.11.2.x) has a promising potential for oxyfunctionalizations and can catalyze a variety of reactions typical for P450s. Peroxygenases are extracellular, i.e. secreted fungal enzymes with high stability, which merely need peroxide for function. Results obtained with the unspecific/aromatic peroxygenases (APOs) of Agrocybe aegerita, Coprinellus radians and Marasmius rotula have demonstrated that APOs catalyze numerous H2O2-dependent monooxygenations of pharmaceuticals and psychoactive drugs. Among them are i) the monooxygenation of aromatic compounds, ii) the benzylic hydroxylation of toluene derivatives, iii) the O-dealkylation of different ether structures including the scission of benzodioxoles (O-demethylenation) and esters as well as iv) the N-dealkylation of secondary and tertiary amines. The peroxygenases studied considerably differ in their substrate spectrum and the preferred positions of oxidation. This finding opens the possibility to develop in the future an “enzymatic toolbox“ on the basis of fungal peroxygenases for the oxyfunctionalization of pharmaceutically relevant compounds. Diese Substanzen sind aufgrund ihrer physiologischen und toxikologischen sowie ökotoxikologischen Wirkungen im menschlichen Körper bzw. in der Umwelt von besonderem Interesse. Cytochrom-P450-Enzyme (P450s) spielen eine Schlüsselrolle bei der Umsetzung und Detoxifizierung bioaktiver Substanzen, darunter vieler Pharmazeutika und Drogen. Es handelt sich bei diesen Enzymen in erster Linie um Monooxygenasen, die intrazellulär lokalisiert und relativ instabil sind; sie benötigen komplexe, teure Kofaktoren und sind nur unter hohem Aufwand zu reinigen, was ihre Anwendung in isolierter Form insgesamt erschwert. Die hier durchgeführten Untersuchungen zu pilzlichen Peroxygenasen haben gezeigt, dass diese Enzymsubklasse (EC 1.11.2.x) ein hohes Oxyfunktionalisierungspotenzial besitzt und eine Vielzahl P450-typischer Reaktionen zu katalysieren vermag. Peroxygenasen sind extrazelluläre, d.h. sekretierte Pilzenzyme, die eine hohe Stabilität aufweisen und lediglich ein Peroxid als Kosubstrat benötigen. Die unter Verwendung der unspezifischen/aromatischen Peroxygenasen (APOs) von Agrocybe aegerita, Coprinellus radians und Marasmius rotula gewonnenen Ergebnisse belegen, dass APOs verschiedene H2O2-abhängige Monooxygenierungen von Pharmazeutika und psychoaktiven Substanzen realisieren. Dazu gehören i) die Monooxygenierung von Aromaten, ii) die benzylische Hydroxylierung von Toluolderivaten, iii) die O-Dealkylierung verschiedener Etherstrukturen einschließlich der Spaltung von Benzodioxolen (O-Demethylenierung) und Estern sowie iv) die N-Dealkylierung von sekundären und tertiären Aminen. Die untersuchten Peroxygenasen wiesen teilweise deutliche Unterschiede im Substratspektrum und den präferierten Oxidationspositionen auf. In der vorliegenden Arbeit werden für ein stark anthropogen beeinflusstes Einzugsgebiet die komplexen Zusammenhänge zwischen Witterung, Landnutzung, Erosion, Gerinneabfluss, Stofffrachten und Makrozoobenthos analysiert. Dafür wurde eine Messstation eingerichtet, an der Fließgeschwindigkeit, Trübung, pH Wert und Leitfähigkeit kontinuierlich gemessen wurden. Mit Hilfe eines durchflussgesteuerten Probennehmers wurden Wasserproben entnommen und im Labor auf verschiedene Inhaltsstoffe (Nährstoffe, Schwermetalle) analysiert sowie die Konzentration von Feinsediment bestimmt. Es konnte eine Beziehung zwischen Trübung und Sedimentkonzentration abgeleitet werden. Die chemischen Analysen zeigten einen komplexen Zusammenhang zwischen der Erosion von landwirtschaftlichen Flächen, dem Stoffeintrag von versiegelten Arealen (Verkehrsflächen, Dächer) aber auch witterungs- und vegetationsbedingte saisonale Effekte. Ein Vergleich der Eigenschaften der Schwebstoffe/Feinsediment im Landwasser mit denen von Ackerboden (Maisfelder) zeigte, dass diese die „chemische Handschrift“ der Ackerböden trugen. Mit Hilfe der Modelle EROSION 3D und ICECREAM wurden die abgetragenen Bodenmengen für das Einzugsgebiet bzw. für ausgewählte Hänge quantifiziert. EROSION 3D überschätzt den Bodenaustrag aus dem Untersuchungsgebiet deutlich. Das Modell ist ungeeignet, um für das Einzugsgebiet reale Abtragswerte mit den zur Verfügung stehenden flächenhaften Eingangsdaten zu berechnen. Bereits für Einzelhänge werden zu große Bodenabträge berechnet. Verstärkend kommt hinzu, dass auf Grund der Modellannahme jeder Partikel, der das Gewässer erreicht auch aus dem Einzugsgebiet heraustransportiert wird. Das Modell kann empfohlen werden, um erosionsgefährdete Flächen auszuweisen. Das Wechselspiel Boden-Pflanze und die damit verbundenen Wassergehaltsänderungen im Boden bestimmen die Funktionalität einer Wasserhaushaltsschicht. Untersuchungen an der Lysimeterstation Bautzen/Nadelwitz machen deutlich, dass vielfältige Faktoren (z. B. nFK, Durchwurzelungstiefe) die Mächtigkeit einer Wasserhaushaltsschicht bestimmen. Der Bodeneinbau mit einem Verdichtungsgrad von 90% - 92% Proctordichte gewährleistet ein setzungsstabiles Korngerüst unter Gewährleistung der geforderten nFK und LK. Durch die Erweiterung des Feinbodenartendreiecks mit den Ebenen „Durchwurzelung“, „nutzbare Feldkapazität“ und „Standsicherheit“ wird eine fachübergreifende Eignungsbeurteilung von Böden ermöglicht. Die wasserhaushaltspezifische Anwendung des Dreiecks der Phasenzusammensetzung nach RUDERT erlaubt die Ausweisung von Schwankungsbreiten bzgl. der Einbaukenngrößen, ohne Abweichungen von den geforderten bodenphysikalischen Anforderungen an eine Wasserhaushaltsschicht hinnehmen zu müssen.

Litter-decay fungi have recently been shown to secrete heme-thiolate peroxygenases that oxidize various organic chemicals, but little is known about the physiological role or the mechanism of these enzymes. The aromatic peroxygenase of Agrocybe aegerita (AaeAPO) was purified and catalytically characterized. An overall reaction mechanism was proposed. The results show that AaeAPO catalyzed diverse H2O2-dependent monooxygenations (two-electron oxidations) including (a) the cleavage of aliphatic and aromatic ethers, (b) the regio- and enantioselective hydroxylation of aromatic compounds, (c) the stepwise oxygenation of benzylic compounds, (d) the N-dealkylation of secondary amines and (e) the dehalogenation of halogenated aliphatic compounds as well as typical peroxidase reactions (suggested to involve one-electron oxidation) such as (f) oxidation and polymerization of phenols and (g) halogenations. The enzyme failed to oxidize polymers such as polyethylene glycol (PEG). Mechanistic studies with several model substrates provided information about the reaction cycle of AaeAPO: (1) stoichiometry of tetrahydrofuran cleavage showed that the reaction was a two-electron oxidation that generated one aldehyde group and one alcohol group, yielding the ring-opened product 4-hydroxybutanal, (2) steady-state kinetics results with methyl 3,4-dimethoxybenzyl ether, which was oxidized to 3,4-dimethoxybenzaldehyde, gave parallel double reciprocal plots suggestive of a ping-pong mechanism, (3) the cleavage of methyl 4-nitrobenzyl ether, the hydroxylation of aromatics such as diclofenac and nitrophenol and the oxygenation of benzylic compounds, resulted in incorporation of 18O into the reaction product in the presence of H218O2, and (4) the demethylation of 1-methoxy-4-trideuteromethoxybenzene showed an distinct observed intramolecular deuterium isotope effect. These results support a mechanism similar to that envisaged for the peroxygenase activity of P450s in which the enzyme heme is, Die Peroxygenase des Südlichen Ackerling (Agrocybe aegerita, AaeAPO) wurde gereinigt, ihr Katalysepotential ermittelt und ein allgemeiner Reaktionsmechanismus postuliert. Die AaeAPO katalysiert sowohl H2O2-abhängige Monooxygenierungen (Zwei-Elektron Oxidationen) wie (a) die Spaltung aliphatischer und aromatischer Ether, (b) die regio- und enantioselektive Hydroxylierung von Aromaten, (c) die schrittweise Monooxygenierung von Toluolderivaten, (d) die N-Dealkylierung sekundärer Amine und (e) die Dehalogenierung chlorierter Aliphaten als auch typische Reaktionen bekannter Peroxidasen (vermutlich Ein-Elektron-Oxidation) unter anderem (f) die Oxidation/ Polymerisierung von Phenolen und (g) die Halogenierung von Aromaten. Polymere Verbindungen wie Polyethylenglycol (PEG) werden nicht oxidiert. Mechanistische Untersuchungen zur Etherspaltung am Beispiel der AaeAPO haben Einblick in den generellen Reaktionsmechanismus dieses neuen Enzymtyps ermöglicht: (1) die Stöchiometrie der Spaltung von Tetrahydrofuran entspricht der einer zwei-Elektron-Oxidation, (2) die Spaltung von Methyl-3,4-Dimethoxybenzylether zu 4-Dimethoxybenzaldehyd und Methanol ergaben parallele Verläufe für die ermittelten Ausgleichsgeraden in der doppelt reziproken Darstellung, was einem „Ping-Pong“-Reaktionsmechanismus entspricht (3) die Monooxygenierungen haben stets den Einbau eines aus dem Peroxid (H2O2) stammenden Sauerstoffatoms in das Produkt zur Folge, (4) die O-Dealkylierung von 1-Methoxy-4-Trideuterummethoxybenzol zeigt einen ausgeprägten Deuterium Isotopen Effekt, was auf die primäre Abspaltung eines Wasserstoffatoms vom Substratmolekül hindeutet. Demnach verläuft die Peroxygenase-katalysierte Monooxygenierung über Wasserstoffabstraktion und eine unmittelbar anschließende Sauerstoffrückbindung (hydrogen abstraction - oxygen rebound mechanism). A-grocybe aegerita peroxidase/peroxygenase (AaP) is an extracellular fungal biocatalyst that selectively hydroxylates the aromatic ring of naphthalene. Under alkaline conditions, the reaction proceeds via the formation of an intermediary product with a molecular mass of 144 and a characteristic UV absorption spectrum (A (max) 210, 267, and 303 nm). The compound was semistable at pH 9 but spontaneously hydrolyzed under acidic conditions (pH <7) into 1-naphthol as major product and traces of 2-naphthol. Based on these findings and literature data, we propose naphthalene 1,2-oxide as the primary product of AaP-catalyzed oxygenation of naphthalene. Using (18)O-labeled hydrogen peroxide, the origin of the oxygen atom transferred to naphthalene was proved to be the peroxide that acts both as oxidant (primary electron acceptor) and oxygen source.
