Abstract

The $(n,m)^{\th}$ KdV hierarchy is a restriction of the KP hierarchy to a submanifold of pseudo-differential operators in a radio form. Explicit formula of the restricted Hamiltonian structure of KP is given which provides a new, more constructive proof of the isomorphism between the associated $W(n,m)$-algebra to $W_{n+m}\oplus W_m\oplus U(1)$ algebra, and the Hamiltonian property of the $(n,m)^{\th}$ KdV hierarchy as well as its Lax-Manakov triad representation. Similarly the Hamiltonian property for a version of modified $n^{\th}$ KdV and the isomorphism between $W_n$-algebra to $W_l\oplus W_m\oplus U(1)$ algebra are shown, where $l+m=n$. 209. RNA demethylase ALKBH5 promotes tumorigenesis in multiple myeloma via TRAF1-mediated activation of NF-κB and MAPK signaling pathways

Abstract

N6-methyladenosine (m6A), an internal modification in mRNA, plays a critical role in regulating gene expression. Dysregulation of m6A modifiers promotes oncogenesis through enzymatic functions that disrupt the balance between the deposition and removal of m6A modification on critical transcripts. However, the roles of mRNA m6A in multiple myeloma (MM) are poorly understood. The present study showed that RNA demethylase ALKBH5 was overexpressed in MM and associated with a poor prognosis in MM patients. Knocking down ALKBH5induced apoptosis and inhibited the growth of MM cells in vitro. Xenograft models and gene set enrichment analysis with patient transcriptome datasets also supported the oncogenic role of ALKBH5 in MM. Mechanistic studies showed that ALKBH5 exerted tumorigenic effects in myeloma in an m6A-dependent manner, and TNF receptor-associated factor 1 (TRAF1) was a critical target of ALKBH5. Specifically, ALKBH5 regulated TRAF1 expression via decreasing m6A abundance in the 3'-untranslated region (3'-UTR) of TRAF1 transcripts and enhancing TRAF1mRNA stability. As a result, ALKBH5 promoted MM cell growth and survival through TRAF1-mediated activation of NF-κB and MAPK signaling pathways. Collectively, our data demonstrated that ALKBH5 played a critical role in MM tumorigenesis and suggested that ALKBH5 could be a novel therapeutic target in MM.

Abstract

BACKGROUND: Non-invasive fractional flow reserve (FFR) has been increasingly used in the clinical workflow to assist clinical decision-making for percutaneous coronary intervention (PCI). This clinical study evaluates the diagnostic accuracy of coronary stenosis assessed by a non-invasive FFR analysis method (termed AccuFFRangio) based on invasive coronary angiography (ICA). It is a blinded, self-controlled, retrospective, and dual-center clinical investigation study. METHODS: Coronary angiography data and the related information of 320 patients with 320 vessels were collected, and AccuFFRangio was used to assess the FFR for these patients. Compared with the wire-measured FFR values, we evaluated AccuFFRangio performance by its accuracy, sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV). RESULTS: The diagnostic accuracy, sensitivity, specificity, PPV, and NPV for AccuFFRangio in identifying hemodynamically significant coronary stenosis were 93.3%, 92.6%, 93.5%, 84.3%, and 97.1%, respectively. 226. The distortion study of rogue waves of the generalized nonlinear Schrödinger equation under the third-order dispersion perturbation.

Abstract

We have investigated the robustness of the rogue wave solutions of two reductions of the generalized nonlinear Schrödinger equation with the third-order dispersion perturbation term. The two reductions are the nonlinear Schrödinger (NLS) equation and the second-type derivative nonlinear Schrödinger (DNLSII) equation. The perturbed equations have practical physical application value. However, they are non-integrable so their exact rogue wave solutions can hardly be obtained by analytical methods. In this paper, we use numerical methods to simulate the perturbed rogue wave solutions and use the quantitative analysis method to assess the robustness of the rogue wave solutions. Two criteria c and r are defined based on the definition of rogue waves in ocean science to analyze the distortion degree of rogue waves quantitatively. The numerical simulation results and the values of these criteria show that the rogue wave solutions of these two reductions are robust under the third-order dispersion perturbation, while the rogue wave solution of the DNLSII equation is more sensitive to the perturbation than that of the NLS equation. Recently, a new (3 + 1) -dimensional integrable KP equation is derived from a specific reduction of the (4 + 2) -dimensional KP equation in Fokas et al. (Fractal Fract 6:425, 2022). We proposed in current paper the Wronskian and Grammian solutions of this (3 + 1) -dimensional integrable KP equation on the basis of Pl u ¨ cker relation and the Jacobi identity for determinants. Furthermore, three kinds of semi-rational solutions of this equation, namely (i) a hybrid of one lump and one soliton, (ii) a hybrid of multiple lumps and one soliton, and (iii) a hybrid of multiple lumps and multiple solitons are proposed and their novel dynamics are discussed.
