Abstract Three recent approvals and over 100 ongoing clinical trials make adeno‐associated virus (AAV)‐based vectors the leading gene delivery vehicles in gene therapy. Pharmaceutical companies are investing in this small and nonpathogenic gene shuttle to increase the therapeutic portfolios within the coming years. This prospect of marking a new era in gene therapy has fostered both investigations of the fundamental AAV biology as well as engineering studies to enhance delivery vehicles. Driven by the high clinical potential, a new generation of synthetic‐biologically engineered AAV vectors is on the rise. Concepts from synthetic biology enable the control and fine‐tuning of vector function at different stages of cellular transduction and gene expression. It is anticipated that the emerging field of synthetic‐biologically engineered AAV vectors can shape future gene therapeutic approaches and thus the design of tomorrow's gene delivery vectors. This review describes and discusses the recent trends in capsid and vector genome engineering, with particular emphasis on synthetic‐biological approaches.

In this work, morphological, electrical, and dielectric performance of nylon copolymer (PA6, 66)/ethylene propylene diene rubber (EPDM) blends were systematically studied with reference to blend ratio and compatibilisation. As the concentration of PA6, 66 and the percentage of compatibiliser in the blend increases the resistivity values decreases. The existence of two phases, PA6, 66 and EPDM with different conductivity and interfacial polarization are responsible for the increase in the dielectric properties of the blends. Compatibilisation of the blends improved the dielectric constant of the blend system. Addition of 2.5% of compatibiliser gave the highest value of dielectric constant. At a high concentration of EPM-g-MA, the polarity of the compatibilised blends was found to be increased, which resulted in the substantial increase in the value of loss and dissipation factor. The dielectric values of the blends were correlated with blend phase morphology. The adoption of sovereign blue bonds by the Republic of Seychelles, hereafter referred to as Seychelles, focuses on resource sustainability and illustrates options for island countries to use their ocean resources for years into the future. The fishing industry is one of the main pillars of Seychelles’ economy and is of crucial importance for domestic food- and employment-security. In order to promote long-term ecological sustainability and economic viability of domestic fisheries, accurate and long-term baseline information is required. Such baseline data were derived here with a reconstruction of the Seychelles’ domestic fisheries catches and fishing effort within its Exclusive Economic Zone (EEZ) from 1950 to 2017, coupled with resulting Catch Per Unit Effort data (CPUE). The total reconstructed domestic catch was approximately 1.5 times larger than the baseline as reported by the United Nations Food and Agriculture Organization (FAO) on behalf of Seychelles from 1950 to 2017 after adjustment for fully domestic catches within the EEZ. Domestic catches (i.e., excluding the large-scale industrial pelagic catches) increased by over 500% throughout the time period, growing from 1,900 t⋅year−1 in the 1950s to around 11,200 t in 2017. The major targeted taxa were jacks (Carangidae), tuna-like fishes (Scombridae) and snappers (Lutjanidae). Total fishing effort in the form of fishing capacity grew from 21,500 kWdays in 1950 to over 3.4 million kWdays in 2017. The resultant artisanal CPUE displayed a declining trend over time, suggesting a potential decline in relative abundance of fish populations within the Seychelles EEZ or targeted fishing areas.

Somatic cell nuclear transfer and transcription-factor-based reprogramming revert adult cells to an embryonic state, and yield pluripotent stem cells that can generate all tissues. Through different mechanisms and kinetics, these two reprogramming methods reset genomic methylation, an epigenetic modification of DNA that influences gene expression, leading us to hypothesize that the resulting pluripotent stem cells might have different properties. Here we observe that low-passage induced pluripotent stem cells (iPSCs) derived by factor-based reprogramming of adult murine tissues harbour residual DNA methylation signatures characteristic of their somatic tissue of origin, which favours their differentiation along lineages related to the donor cell, while restricting alternative cell fates. Such an 'epigenetic memory' of the donor tissue could be reset by differentiation and serial reprogramming, or by treatment of iPSCs with chromatin-modifying drugs. In contrast, the differentiation and methylation of nuclear-transfer-derived pluripotent stem cells were more similar to classical embryonic stem cells than were iPSCs. Our data indicate that nuclear transfer is more effective at establishing the ground state of pluripotency than factor-based reprogramming, which can leave an epigenetic memory of the tissue of origin that may influence efforts at directed differentiation for applications in disease modelling or treatment.

Here, we provide data on the mathematical model-guided implementation of a biomaterials-based positive feedback loop for the detection of proteolytic activities.
