Abstract

Genes specifically expressed in neurons contain members with extended long introns. Longer genes present a problem with respect to fulfilment of gene length transcription, and evidence suggests that dysregulation of long genes is a mechanism underlying neurodegenerative and psychiatric disorders. Here, we report the discovery that RNA-binding protein Sfpq is a critical factor for maintaining transcriptional elongation of long genes. We demonstrate that Sfpq co-transcriptionally binds to long introns and is required for sustaining long-gene transcription by RNA polymerase II through mediating the interaction of cyclin-dependent kinase 9 with the elongation complex. Phenotypically, Sfpq disruption caused neuronal apoptosis in developing mouse brains. Expression analysis of Sfpq-regulated genes revealed specific downregulation of developmentally essential neuronal genes longer than 100 kb in Sfpq-disrupted brains; those genes are enriched in associations with neurodegenerative and psychiatric diseases. Abstract

The kinetics of the glass transition and the crystallization behavior of Ti–Cu–Ni–Sn–M (M=Zr,Mo,Ta) glassy alloys were investigated by differential scanning calorimetry and x-ray diffraction. The glass-forming ability (GFA) of the glasses was analyzed in terms of the fragility concept and the width of the supercooled liquid region. The Ti-base glasses studied were found to have lower GFA than Zr, Pd, La, Fe, Mg-base bulk metallic glasses (BMGs), but to be comparative GFA to Ni- and Au-base BMGs. Higher Mo content or higher Ti–Ta content in the alloys lowers the GFA. The chemical complexity and multiphase crystallization requiring long-range atomic diffusion may contribute to the thermal stability of the supercooled liquid against crystallization. The products of crystallization of the glasses include bcc β-Ti(M) solid solution and Ti[sub 2]Ni and Ti[sub 2]Cu intermetallics. © 2004 American Institute of Physics. [ABSTRACT FROM AUTHOR]

Abstract

Measurements of midrapidity charged particle multiplicity distributions, $dN_{\rm ch}/d\eta$, and midrapidity transverse-energy distributions, $dE_T/d\eta$, are presented for a variety of collision systems and energies. Included are distributions for Au$+$Au collisions at $\sqrt{s_{_{NN}}}=200$, 130, 62.4, 39, 27, 19.6, 14.5, and 7.7 GeV, Cu$+$Cu collisions at $\sqrt{s_{_{NN}}}=200$ and 62.4 GeV, Cu$+$Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV, U$+$U collisions at $\sqrt{s_{_{NN}}}=193$ GeV, $d$$+$Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV, $^{3}$He$+$Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV, and $p$$+$$p$ collisions at $\sqrt{s_{_{NN}}}=200$ GeV. Centrality-dependent distributions at midrapidity are presented in terms of the number of nucleon participants, $N_{\rm part}$, and the number of constituent quark participants, $N_{q{\rm p}}$. For all $A$$+$$A$ collisions down to $\sqrt{s_{_{NN}}}=7.7$ GeV, it is observed that the midrapidity data are better described by scaling with $N_{q{\rm p}}$ than scaling with $N_{\rm part}$. 245. Reduction of Phosphate Application in Paddy Rice Cultivation for More Sustainable and Environmentally Sound Agriculture

Abstract

Proceedings of Shinshu University International Symposium 2007 “Sustainable Agriculture and Environment:Asian Network”, Heavy application of phosphate （P2O5） in paddy rice production has been continued since 1960s in Japan. Although yield was increased and stabilized during that period, recent environmental concerns require minimization of P2O5 application. Six-year experiment on the effect of reduced P2O5 application on growth and yield of paddy rice was carried out in Andosols area, where heavy P2O5 application had been continued. The experiment showed that growth and yield were not significantly influenced by reduced P2O5 application and that reduction by 25-50% compared to the present recommended application amount was safely practicable. Input and loss of available P2O5 in the soil was estimated to balance when P2O5 application is 4.6 kg/10 a. Since available P2O5 has been accumulated in the soil, drastic reduction of P2O5 application over the long term is possible without causing significant decline in yield and its stability by making most of the accumulated P2O5 stock., Article, 信州大学農学部紀要. 44(1-2): 53-56 (2008)

Abstract

SR proteins are non-snRNP splicing factors harbouring a domain rich in Arg-Ser repeats, which are extensively phosphorylated by several kinases. We performed a comparative study of different SR kinases, including SRPK, Clk, PRP4 and DYRK, and found that only Clks efficiently altered 5' splice site selection of Adenovirus E1A. The phosphorylation state of SR proteins was examined using a phospho-SR specific antibody mAb1H4 and a 75 kDa protein was most evidently hyperphosphorylated by Clks. Administration of TG003, a specific inhibitor for the Clk family members, specifically and rapidly induced dephosphorylation of 75 kDa SR protein. Imaging with mRFP-SRp75 in living cells revealed that its nuclear distribution was rapidly altered upon inhibition of the Clk activity by TG003. Co-transfection experiments demonstrated that HA-tagged SRp75 was hyperphosphorylated by Clk family members, but not by other SR kinases. These results indicate that Clks specifically hyperphosphorylate SRp75. Furthermore, SRp75 over-expression promoted the selection of 12S 5' splice site in E1A pre-mRNA, which is stimulated by co-expression of Clks. These results suggest that the specific combination of SR protein and SR kinase plays a distinct role in alternative splicing through dynamic balance of phosphorylation.
