The aims of the present study were to reveal the prevalence of the TERT C228T mutation in pediatric papillary thyroid carcinoma (PPTC) and to further investigate the role of the TERT C228T mutation in PPTC. We also tested another TERT mutation, TERT C250T, although this was not detected in PPTC patients. In this study, 48 patients with PPTC (41 with classic PPTC) were enrolled. DNA was extracted from PPTC tissues and TERT C228T mutation analysis was performed. Chi-squared analysis, Fisher's exact test, and a t-test were applied to test the significance of differences. The TERT C228T mutation presented in 13 (27.1%) of the 48 PPTC patients and 10 (24.4%) of the 41 classical PPTC patients. There were significant differences between PPTC patients with the TERT C228T mutation and those without in terms of modified radical neck dissection, multifocality, capsular invasion, extrathyroidal invasion, and American Joint Committee on Cancer (AJCC) tumor stage (P<0.05). In classical PPTC, there were additional significant differences in other clinic-pathological features, such as AJCC nodal stage (P=0.009) and American Thyroid Association (ATA) PPTC stage (P=0.021) between patients with and without the TERT C228T mutation. Water imbalances remain the primary problem from the past to the coming future in karst area of South China. A well-admissive method to cope with drought and flood problems is under searching. Rainwater harvesting by water tanks has being an approved way to alleviate water shortage, but with some defects such as bad water quality, unstable sources, and limited volume. To find out its running status, 50 water tanks in a part region of Fengshan County were surveyed. One of them was chosen for online-monitoring and 25 water tanks were sampled for chemical analysis. The results show that there were three typical problems during its running. First, water tanks usually get dry in rainless months, especially in extremely drought events. Second, water quality declines once water has stored for about 100 days with no new supplement. The deterioration of water quality is related to the long-preserved time rather than exogenetic pollution. Third, the situation becomes more serious under the pressure of climate change and the rocky desertification. Therefore, a new rainwater harvesting strategy is proposed for improving this situation. The conception site model (CSM) in hill slope for tank location selection is the critical point, since water harvesting for tanks depends on slope characters. As a CSM example, the hillslope hydrology related to the overland flow, the resurgence of flow in epikarst, and vadose flow at Yaji Karst Experiment Site were investigated to illuminate the complexity of hydrological process and diversity of flow pattern. Then, it is suggested to make full use of different flows from slopes, especially to increase the availability of vadose flow. Finally, besides drinking purpose, water tanks should be extensively used for irrigation and reforestation to obtain economic return and support its maintenance. [ABSTRACT FROM AUTHOR]

The paper chose a typical karst aquiferous system in southwest China to study karst water cycle process to help the sustainable development and utilization of karst water resources. Different water bodies' water chemistries and isotopes' characteristics and their implications are analyzed based on the conceptions of transformations among "precipitation, vadose zone water flow, karst conduit water flow, and surface water flow" and the "recharge, runoff, and drainage". Hydrological information can be obtained by analyzing water chemistries, δD and δ18O values, deuterium excess values, regression equations, influencing factors of all water bodies and precipitation in rainy season and dry season. Topography, geomorphology, and developed characteristics of karst areas affect circulation process of karst water resource, water chemistries and isotopes along the main conduit having a great fluctuation affected by changes of water dynamic conditions. Ca2+ and HCO3− are the main cation and anion in the karst aquiferous system, concentrations of Cl− and NO3− respond to the influence of human activities significantly, the dilution affection of precipitation recharging groundwater on the concentration of Ca2+ and Mg2+ is biggest, and affection of human activities on water bodies in rainy season is higher than dry season. δD and δ18O values have the increasing trend along the underground river flow path for the water–rock interaction; d values are higher and have more obvious fluctuating status along the main flow path in dry season for the long non-successive residence time. δD, δ18O and d values' dynamic variation characteristics of cave drip water, epikarst springs, karst conduit water, and surface water in different seasons are mainly affected by recharging sources and evapotranspiration, the hydrological environment and fractionation, the developed location and evaporation, condensation and evaporation, respectively. Dynamic variation characteristics of water chemistries, isotopes, d values and their implications in the karst water cycle can help decision makers to take effective measures to protect and utilize karst water resources. Abstract A kind of silicone rubber (SR)/paraffin (Pa)@silicon dioxide (SiO 2) composite form-stable phase change material (PCM) was developed in this paper. Pa@SiO 2 was obtained by choosing Pa as PCM core microencapsulated in SiO 2 shell based on tetraethoxysilane (TEOS) and γ-aminopropyl triethoxysilane (APTES) as precursors, then Pa@SiO 2 microencapsules were embedded in SR matrix to fabricate SR/Pa@SiO 2 composites. The SiO 2 shell can provide a barrier for the melted Pa and meanwhile have reinforcing effect on SR as inorganic filler. The in-situ modification of SiO 2 shell using APTES owing to the amino groups can not only prevent agglomeration, but also increase interfacial bonding between Pa@SiO 2 with SR. The thermal and mechanical properties of the form-stable PCMs were characterized by differential scanning calorimetry (DSC) and mechanical tests. The results showed that the thermal and mechanical properties of SR/Pa@SiO 2 composites were better than those of SR/Pa and SR/Pa/SiO 2. Leakage test was conducted by heating the prepared composites over the melting temperature of Pa. The SR/Pa@SiO 2 composites were found as form-stable PCM with low leakage rate. The phase change latent heat of the SR/Pa@SiO 2 composites was up to 92.39 J g−1 and the composites can be used for thermal energy storage. High-throughput next generation sequencing (NGS) is a shotgun approach applied in a parallel fashion by which the genome is fragmented and sequenced through small pieces and then analyzed either by aligning to a known reference genome or by de novo assembly without reference genome. This technology has led researchers to conduct an explosion of sequencing related projects in multidisciplinary fields of science. However, due to the limitations of sequencing-based chemistry, length of sequencing reads and the complexity of genes, it is difficult to determine the sequences of some portions of the human genome, leaving gaps in genomic data that frustrate further analysis. Particularly, some complex genes are difficult to be accurately sequenced or mapped because they contain high GC-content and/or low complexity regions, and complicated pseudogenes, such as the genes encoding xenobiotic metabolizing enzymes and transporters (XMETs). The genetic variants in XMET genes are critical to predicate inter-individual variability in drug efficacy, drug safety and susceptibility to environmental toxicity. We summarized and discussed challenges, wet-lab methods, and bioinformatics algorithms in sequencing "complex" XMET genes, which may provide insightful information in the application of NGS technology for implementation in toxicogenomics and pharmacogenomics. Graphene encapsulating paraffin (paraffin@graphene) microcapsules were fabricated by electrostatic interaction-based self-assembly. An aqueous dispersion of graphene sheets charged with cation, were mixed with a water-based emulsion containning negatively charged paraffin droplet spheres to form self-assembled microcapsules. The morphology of the microcapsules was characterized by scanning electron microscope (SEM). Results show that the microcapsules with a well-defined spherical structure were prepared successfully. Differential scanning calorimeter (DSC) results indicate that the phase change latent heat are all above 200 J g−1. With a graphene mass fraction of 8 wt%, the thermal conductivity of the fabricated composites can reach 1.73 W m−1 K−1. Attributing to the interlocking of graphene with each other, the microcapsules enable lock the paraffin in the shell thus successfully avoiding its leakage during phase change process. Abstract The bovine IFN-ω (BoIFN-ω) multigene family is located on chromosome 8, which has 14 potential functional genes and 10 pseudogenes. After aligning 14 BoIFN-ω subtypes and assigning the most frequently occurring amino acids in each position, one artificial consensus BoIFN-ω (CoBoIFN-ω) gene was designed, optimized and synthesized. Then, CoBoIFN-ω was expressed in Pichia pastoris , which was demonstrated to have 3.94-fold and 14.3-fold higher antiviral activity against VSV on MDBK cells than that of BoIFN-ω24 and BoIFN-ω3, respectively. Besides this, CoBoIFN-ω was confirmed to have antiviral activity against VSV on BL, BT, PK-15 cells, and against BEV, BHV-1, BPIV3 on MDBK cells. Additionally, CoBoIFN-ω could bind with bovine type I IFN receptors, and then activate the promoters of NF-κB, ISRE and BoIFN-β, and induce the transcription of ISGs and expression of Mx1 and NF-κB p65, which suggested CoBoIFN-ω exerts antiviral activity via activation of the JAK-STAT signaling pathway. Overall, this research on CoBoIFN-ω not only extends and improves consensus IFN research, but also reveals that CoBoIFN-ω has the potential to be used in the therapy of bovine viral diseases. Abstract IFN-kappa (IFN-κ) is a type I IFN expressed by keratinocytes, monocytes and dendritic cells with important roles during the innate immune response period. This research was conducted to elaborate the evolution and characteristics of IFN-κ in poultry. Chicken IFN-κ is located on the sex-determining Z chromosome, which is greatly different from mammals. Poultry IFN-κ cluster together in a species-specific manner through positive selection pressure and share only 19–33% homology with mammalian IFN-κ and poultry other type I IFN. Both chicken and duck IFN-κ was constitutively expressed in spleen, skin, lung, and peripheral blood mononuclear cells (PBMC), as well as being significantly induced after treatment with virus in PBMC. Biologically, poultry IFN-κ has antiviral activity against VSV in chicken embryonic fibroblasts and duck embryonic fibroblasts (CEF and DEF) cells, and induces the expression of IFN stimulated genes (ISGs). After treatment with JAK1 inhibitor, the ISGs expression can be down-regulated. Overall, our research on poultry IFN-κ not only enriches the knowledge about IFN-κ but also facilitates further research on the role of type I IFNs in antiviral defense responses in poultry. Paraffin, due to its linear chain and saturated hydrocarbons with low thermal conductivity, is difficult to transfer energy effectively. Using amphiphilicity of graphene oxide (GO), Pickering emulsion of paraffin@GO was obtained and then paraffin@graphene microencapsulated phase change materials (MEPCMs) were achieved by chemical reduction through adding hydrazine hydrate. Thermally conductive PCMs with segregated structure were constructed by hot compression of paraffin@graphene microencapsules. Scanning electronic microscopy, differential scanning calorimetry and thermal conductivity test were used to characterize microstructure and thermal properties of MEPCMs. Meanwhile, the effect of graphene on the phase change latent heat and phase transition temperature was investigated. Results indicated that GO did not react with paraffin during the process of chemical reduction and the obtained MEPCMs were regular spheres. These MEPCMs had paraffin content of 99% or even more. Graphene working as shell materials increased the phase change latent heat of paraffin from 227.6 to 232.4 J/g, without affecting the phase transition temperature. The hot-compress molding makes graphene shell form segregated structure with more thermal pathways, further enhancing thermal conductivity. Acetaminophen (APAP) overdose is the leading cause of acute liver failure. Yet the mechanisms underlying adaptive tolerance toward APAP-induced liver injury are not fully understood. To better understand molecular mechanisms contributing to adaptive tolerance to APAP is an underpinning foundation for APAP-related precision medicine. In the current study, the mRNA and microRNA (miRNA) expression profiles derived from next generation sequencing data for APAP-treated (5 and 10 mM) HepaRG cells and controls were analyzed systematically. Putative miRNAs targeting key dysregulated genes involved in APAP hepatotoxicity were selected using in silico prediction algorithms, un-biased gene ontology, and network analyses. Luciferase reporter assays, RNA electrophoresis mobility shift assays, and miRNA pull-down assays were performed to investigate the role of miRNAs affecting the expression of dysregulated genes. Levels of selected miRNAs were measured in serum samples obtained from children with APAP overdose (58.6–559.4 mg/kg) and from healthy controls. As results, 2758 differentially expressed genes and 47 miRNAs were identified. Four of these miRNAs (hsa-miR-224-5p, hsa-miR-320a, hsa-miR-449a, and hsa-miR-877-5p) suppressed drug metabolizing enzyme (DME) levels involved in APAP-induced liver injury by downregulating HNF1A, HNF4A and NR1I2 expression. Exogenous transfection of these miRNAs into HepaRG cells effectively rescued them from APAP toxicity, as indicated by decreased alanine aminotransferase levels. Importantly, hsa-miR-320a and hsa-miR-877-5p levels were significantly elevated in serum samples obtained from children with APAP overdose compared to health controls. Collectively, these data indicate that hsa-miR-224-5p, hsa-miR-320a, hsa-miR-449a, and hsa-miR-877-5p suppress DME expression involved in APAP-induced hepatotoxicity and they contribute to an adaptive response in hepatocytes.
