The purpose of this study is to quantify the effect of various design and operating parameters on smoking room performance. Twenty-eight experiments were conducted in a simulated smoking room with a smoking machine and an automatic door opener. Measurements were made of air flows, pressures, temperatures, two particle-phase ETS tracers, two gas-phase ETS tracers, and sulfur hexafluoride. Quantification of leakage flows, the effect of these leaks on smoking room performance and non-smoker exposure, and the relative importance of each leakage mechanism are presented. The results indicate that the first priority for an effective smoking room is to depressurize it with respect to adjoining non-smoking areas. Another important ETS leakage mechanism is the pumping action of the smoking room door. Substituting a sliding door for a standard swing-type door reduced this source of ETS leakage significantly. Measured results correlated well with model predictions (R2 = 0.82-0.99).

Functional symbiosis is considered one of the successful mechanisms by which plants that inhabit extreme environment improve their ability to tolerate different types of stress. One of the most conspicuous type of symbiosis is the endophyticism. This interaction has been noted to play a role in the adaptation of the native vascular plant Colobanthus quitensis to the stressful environments of Antarctica, characterized by low temperatures and extreme aridity. Projections of climate change for this ecosystem indicate that abiotic conditions will be less limiting due to an increase in temperature and water availability in the soil. Due to this decrease in stress induced by the climate change, it has been suggested that the positive role of fungal endophytes on performance of C. quitensis plants would decrease. In this study, we evaluated the role of endophytic fungi on osmoprotective molecules (sugar production, proline, oxidative stress) and gene expression (CqNCED1, CqABCG25, and CqRD22) as well as physiological traits (stomatal opening, net photosynthesis, and stomatal conductance) in individuals of C. quitensis. Individual plants of C. quitensis with (E+) and without (E−) endophytic fungi were exposed to simulated conditions of increased water availability (W+), having the current limiting water condition (W−) in Antarctica as control. The results reveal an endophyte-mediated lower oxidative stress, higher production of sugars and proline in plants. In addition, E+ plants showed differential expressions in genes related with drought stress response, which was more evident in W− than in W+. These parameters corresponded with increased physiological mechanisms such as higher net photosynthesis, stomatal opening and conductance under presence of endophytes (E+) as well as the projected water condition (W+) for Antarctica. These results suggest that the presence of fungal endophytes plays a positive role in favoring tolerance to drought in C. quitensis. However, this positive role would be diminished if the stress factor is relaxed, suggesting that the role of endophytes could be less important under a future scenario of climate change in Antarctica with higher soil water availability.

OBJECTIVES:The study attempted to determine the incidence of HIV among pregnant women in Kabarole District, Uganda, and to identify socio-demographic and behavioral risk factors for seroconversion during pregnancy. METHODS:We carried out a retrospective cohort study among women for whom a documented HIV-negative test result from the first pregnancy trimester could be confirmed using available records, and who were HIV-retested in the third trimester or during delivery. In total, 1610 pregnant women from three different healthcare settings took part in the study. We captured the results of repeated HIV tests and conducted semi-structured interviews to explore participants' socio-demographic characteristics and sexual risk behavior. For HIV incidence rates, we calculated the number of seroconversions per 100 person-years. We used Fisher's exact test to test for potential associations. Penalized maximum likelihood logistic regression and Poisson regression were applied to adjust for potential confounders. RESULTS:The overall HIV incidence rate among participants was 2.9/100 women-years. Among socio-demographic characteristics, the multivariable analysis showed a significant association of marital status with HIV incidence in pregnancy (IRR 8.78, 95%CI [1.13-68.33]). Risky sexual behaviors including higher number of sexual partners in pregnancy (IRR 2.78 [1.30-5.94]), unprotected sex with unknown persons (IRR 14.25 [4.52-44.93]), alcohol abuse (IRR 12.08 [4.18-34.90]) and sex under the influence of drugs or alcohol (IRR 6.33 [1.36-29.49]) were significantly associated with seroconversion in pregnancy (similar results in logistic regression). CONCLUSIONS:HIV incidence was three times higher among our pregnant study population compared to the general female population in Uganda. This underlines the importance of HIV prevention and repeat testing during pregnancy. Identified risk groups should be considered for pre-exposure prophylaxis.

Glucose-dependent insulinotropic polypeptide (GIP) is an incretin hormone involved in regulating glucose and lipid metabolism. GIP receptor (GIPR) antagonism is believed to offer therapeutic potential for various metabolic diseases. Pharmacological intervention of GIPR, however, has limited success due to lack of effective antagonistic reagents. Previously we reported the discovery of two mouse anti-murine GIPR monoclonal antibodies (mAbs) with distinctive properties in rodent models. Here, we report the detailed structural and biochemical characterization of these two antibodies, mAb1 and mAb2. In vitro and in vivo characterizations demonstrated mAb2 is a full GIPR antagonistic antibody and mAb1 is a non-neutralizing GIPR binder. To understand the molecular basis of these two antibodies, we determined the co-crystal structures of GIPR extracellular domain in complex with mAb1 and with mAb2 at resolutions of 2.1 and 2.6 Å, respectively. While the non-neutralizing mAb1 binds to GIPR without competing with the ligand peptide, mAb2 not only partially occludes the ligand peptide binding, but also recognizes the GIPR C-terminal stalk region in a helical conformation that acts as a molecular mimic of the ligand peptide and locks GIPR in a novel auto-inhibited state. Furthermore, administration of mAb2 in diet-induced obesity mice for 7 weeks leads to both reduction in body weight gain and improvement of metabolic profiles. In contrast, mAb1 has no effect on body weight or other metabolic improvement. Together, our studies reveal the unique molecular mechanism of action underlying the superior antagonistic activity of mAb2 and signify the promising therapeutic potential of effective GIPR antagonism for the treatment of metabolic disorders.

Background Plants possess a sophisticated immune system to defend from herbivores. These defence responses are regulated by plant hormones including salicylic acid (SA) and jasmonic acid (JA). Sometimes, plant defences can be complemented by the presence of symbiotic microorganisms. A remarkable example of this are grasses establishing symbiotic associations with Epichloë fungal endophytes. We studied the level of resistance provided by the grass’ defence hormones, and that provided by Epichloë fungal endophytes, against an introduced herbivore aphid. These fungi protect their hosts against herbivores by producing bioactive alkaloids. We hypothesized that either the presence of fungal endophytes or the induction of the plant salicylic acid (SA) defence pathway would enhance the level of resistance of the grass to the aphid. Methods Lolium multiflorum plants, with and without the fungal endophyte Epichloë occultans, were subjected to an exogenous application of SA followed by a challenge with the aphid, Sipha maydis. Results Our results indicate that neither the presence of E. occultans nor the induction of the plant’s SA pathway regulate S. maydis populations. However, endophyte-symbiotic plants may have been more tolerant to the aphid feeding because these plants produced more aboveground biomass. We suggest that this insect insensitivity could be explained by a combination between the ineffectiveness of the specific alkaloids produced by E. occultans in controlling S. maydis aphids and the capacity of this herbivore to deal with hormone-dependent defences of L. multiflorum.

This article discusses a surge in imported cases of American cutaneous leishmaniasis (ACL) from Mexico in 2023. ACL is caused by Leishmania (L.) mexicana and is endemic to the Yucatan peninsula in Mexico. The infection is transmitted through the bite of female sandflies, and most cases occur in men engaged in occupational activities in sylvatic areas. The article presents a case series of six patients diagnosed with imported ACL in Germany and France, highlighting the increase in cases compared to previous years. The rise in cases is likely linked to environmental changes, including the construction and deforestation associated with the 'Tren Maya' project in the Yucatan peninsula. We present the realization and analysis of the microwave performances of interdigited varactors integrating thin ferroelectric layers of barium and strontium titanate (BaxSr1−xTiO3). Devices based on ferroelectric films of different compositions (x = 0.8 and x = 0.5) have been characterized in the millimeter-wave domain, from 200 MHz to 110 GHz. By applying different bias voltages, the tunability of the capacitance can reach up to 40% for the Ba0.8Sr0.2TiO3 composition, under relatively low applied electric fields of about 167 kV/cm. These promising characteristics allow the integration of the varactor devices in tunable antennas for a large frequency domain, from the microwaves to the millimeter waves range.

León es uno de los departamentos de Nicaragua con mayor importancia y que cuenta con muchos valores patrimoniales de la nación. A raíz de los eventos sísmicos del 2014, el CNU convocó a las universidades (UNI, UNAN, UCA) a evaluar el daño de varios de estos bienes, por lo cual se conformaron equipos de trabajos con especialistas y estudiantes. Evaluando la documentación histórica, el reconocimiento en campo para el desarrollo del diagnóstico, y utilizando la técnica de semaforización (3 niveles de daños), se logró evidenciar en corto tiempo los edificios más deteriorados a causa de los sismos y construir los expedientes actualizados, con información de Planimetría de los bienes inmuebles.
