Scanning Thermal Microscopy (SThM) has become an important measurement technique for characterizing the thermal properties of materials at the nanometer scale. This technique requires a SThM probe that combines an Atomic Force Microscopy (AFM) probe and a very sensitive resistive thermometer; the thermometer being located at the apex of the probe tip allows for the mapping of temperature or thermal properties of nanostructured materials with very high spatial resolution. The high interest of the SThM technique in the field of thermal nanoscience currently suffers from a low temperature sensitivity despite its high spatial resolution. To address this challenge, we developed a high vacuum-based AFM system hosting a highly sensitive niobium nitride (NbN) SThM probe to demonstrate its unique performance. As a proof of concept, we utilized this custom-built system to carry out thermal measurements using the 3ω method. By measuring the V3ω voltage on the NbN resistive thermometer under vacuum conditions, we were able to determine the SThM probe's thermal conductance and thermal time constant. Two phenomenological approaches are currently used in the study of the vitreous state. One is based on the concept of fictive temperature introduced by Tool [J. Res. Natl. Bur. Stand. 34, 199 (1945)] and recently revisited by Nieuwenhuizen [Phys. Rev. Lett. 80, 5580 (1998)]. The other is based on the thermodynamics of irreversible processes initiated by De Donder at the beginning of the last century [L’Affinité (Gauthier-Villars, Paris, 1927)] and recently used by Möller et al. for a thorough study of the glass transition [J. Chem. Phys. 125, 094505 (2006)]. This latter approach leads to the possibility of describing the glass transition by means of the freezing-in of one or more order parameters connected to the internal structural degrees of freedom involved in the vitrification process. In this paper, the equivalence of the two preceding approaches is demonstrated, not only for glasses but in a very general way for any system undergoing an irreversible transformation. This equivalence allows the definition of an effective temperature for all systems departed from equilibrium generating a positive amount of entropy. In fact, the initial fictive temperature concept of Tool leads to the generalization of the notion of temperature for systems out of thermodynamic equilibrium, for which glasses are just particular cases. Introduction Le récepteur nucléaire PPARalpha est un facteur de transcription. Il est exprimé dans le foie et les autres organes producteurs de glucose mais aussi dans les tissus adipeux et les muscles. De récents travaux indiquent un lien étroit entre la lipolyse adipocytaire et la néoglucogénèse hépatique. Ainsi, la production d'acétyl-coA par la beta-oxydation hépatique des acides gras d'origine adipocytaire jouerait un rôle déterminant dans l'insulino-résistance hépatique. Dans cette étude, nous avons testé l'hypothèse que l'activité hépatocytaire de PPARalpha joue un rôle décisif dans l'utilisation des acides gras adipocytaires in vivo. Matériels et Méthodes Pour cela nous avons créé et utilisé un nouveau modèle de souris transgénique présentant une déficience hépato-spécifique du récepteur PPARalpha, puis nous avons : – identifié les gènes spécifiquement régulés par PPARalpha dans les hépatocytes – étudié leur cinétique d'expression hépatique lors de la lipolyse adipocytaire – analysé la réponse hépatique à une activation pharmacologique de la lipolyse par le récepteur beta3-adrénergique – mesuré l'importance de l'activité hépatique de PPARalpha lors du diabète de type II. Résultats Nos résultats montrent que PPARalpha est un régulateur autonome des gènes impliqués dans le catabolisme des acides gras et dans la fonction mitochondriale hépatocytaire. L'absence du récepteur dans le foie se traduit par une augmentation des acides gras libres circulants, une absence de FGF21, une baisse des corps cétoniques et une hypoglycémie lors du jeûne. Les animaux présentant une déficience hépatique pour PPARalpha deviennent stéatotiques, sans diabète ni obésité. De plus, nous avons mis en évidence que la cinétique d'induction de la lipolyse adipocytaire est très fortement corrélée à l'activité hépatique de PPARalpha. En outre, l'activation pharmacologique de la lipolyse adipocytaire se traduit par une augmentation de l'activité de PPARalpha dans les hépatocytes. Finalement, nos données montrent que l'absence de PPAR-alpha hépatocytaire protège du diabète associé à l'obésité. Conclusions PPARalpha est un effecteur majeur du dialogue entre le tissu adipeux et le foie. Introduction La voie Wnt/β-caténine joue un rôle essentiel dans le foie contrôlant le métabolisme azoté et glucidique. Elle est activée par la liaison de ligands Wnts, qui subissent plusieurs modifications post-traductionnelles, notamment des acylations nécessaires à leur sécrétion et action sur leurs récepteurs. En particulier, ils sont palmitoléylés par la protéine Porcupine, une acyltransférase localisée dans le RE. Cependant, la régulation de cette voie par les hormones régulant le métabolisme glucidolipidique reste à déterminer. L’objectif de cette étude a donc été de déterminer l’influence des hormones régulant le métabolisme glucidolipidique, et notamment, l’insuline sur l’activation de la voie Wnt/β-caténine dans le foie. Matériels et méthodes L’activité de la voie Wnt/β-caténine a été étudiée par des approches de gène rapporteur, RT-qPCR et Western Blot, in vitro , dans des cultures primaires d’hépatocytes de souris et, in vivo , dans le foie de souris soumises à des manipulations nutritionnelles (jeune/réalimentation), des inhibiteurs pharmacologiques, ou géniques (souris invalidées pour le récepteur de l’insuline hépatique). Résultats Au cours de cette étude, nous montrons que la voie Wnt/β-caténine est activée par l’insuline dans le foie de souris renourries. Des analyses in vitro dans des cultures primaires d’hépatocytes et in vivo dans un modèle de souris invalidées pour le récepteur à l’insuline dans le foie, montrent qu’en absence d’un signal insulinique efficace, l’expression des gènes cibles de la voie Wnt/β-caténine est diminuée. Ce processus implique la voie PI3K/mTORC1 et l’induction de la lipogenèse. En effet, l’enzyme SCD1, qui désature le palmitate en palmitoléate, est nécessaire à l’activation de la voie Wnt/β-caténine. De plus ce processus implique également l’action de l’acyltransférase Porcupine qui acyle les ligands Wnt avec le palmitoléate. Conclusion En conclusion, l’insuline joue un rôle déterminant dans l’induction de la voie Wnt/β-caténine dans le foie des souris renourries. Les protéines SCD1 et Porcupine participent à ce nouveau mécanisme d’activation des ligands Wnt. Introduction Des données récentes indiquent que l’activation du récepteur des estrogènes α (ERα) dans les hépatocytes pourrait prévenir l’installation de pathologies dysmétaboliques du foie. Pour mieux caractériser l’intérêt thérapeutique de cette cible moléculaire, nous avons étudié le rôle du ERα hépatique dans les effets métaboliques exercés par le tamoxifène (TMX), un SERM ( Selective Estrogen Receptor Modulator ) activant spécifiquement la fonction de tran-sactivation ERαAF1. Matériels et méthodes Des souris femelles invalidées pour ERα, totalement (ERα-/-) ou spécifiquement dans les hépatocytes (ERα-Hep º), et des souris n’exprimant pas la fonction ERαAF1 (ERαAF1 º) ont été ovariectomisées, traitées par placebo ou TMX (1,2 mg/kg/j) en administration sous-cutanée continue pendant 12 semaines et simultanément soumises à un régime hyperli-pidique (45 % lipides). Résultats L’administration de TMX prévient la prise de poids, l’expansion des tissus adipeux, l’installation d’une insulinorésistance et d’une intolérance au glucose, ainsi que le développement d’une stéatose hépatique (contenu en triglycérides, histologie), induits par le régime hyperlipidique ( p < 0,01 vs placebo pour l’ensemble des paramètres). Cet effet protecteur est totalement aboli chez les souris ERα-/- et ERαAF1 º, confirmant l’implication spécifique de cette fonction de transactivation dans le mécanisme d’action du TMX. Comme attendu, le TMX perd également son action protectrice vis-à-vis de la stéatose hépatique chez les animaux ERα-Hep º. De plus, les effets de prévention de l’adiposité, de l’insulinorésistance et des troubles du métabolisme glucidique induits par le régime sont également totalement abolis dans ce modèle d’invalidation spécifique du ERα hépatocytaire. Conclusion Au-delà des effets de protection vis-à-vis de la stéatose hépatique, une stratégie de modulation sélective du ERα hépatocytaire privilégiant l’activation de la fonction de transactivation AF1 permet de prévenir l’installation d’une obésité et de troubles de l’homéostasie glucidique en réponse à un régime hyperlipidique. Nos résultats renforcent donc l’intérêt du ERα hépatique en tant que cible thérapeutique pour la prévention des pathologies dysmétaboliques. Introduction L’inhibition de la lipolyse adipocytaire améliore la sensibilité à l’insuline in vivo et in vitro . Au vu des connaissances actuelles, nous avons posé l’hypothèse que l’amélioration de la réponse à l’insuline observée lors d’une inhibition de la Lipase hormono-sensible (LHS) est dirigée par l’augmentation de la voie de la lipogenèse de novo (LDN), sous le contrôle du facteur de transcription ChREBP. Matériels et méthodes Dans des préadipocytes humains en culture (cellules hMADS), l’inhibition de l’expression de la LHS (siLHS), du facteur ChREBP (siChREBP) ou des deux (siLHS/siChREBP) est réalisée à l’aide de petits ARN interférents. Le métabolisme du glucose est évalué à l’aide de glucose radiomarqué. L’expression des différentes enzymes dirigeant la LDN a été mesurée par RT-qPCR. La composition intra-cellulaire en acides gras a été déterminée par chromatographie en phase gazeuse. Enfin, la transmission du signal insulinique a été évaluée par Western Blot. Résultats Les adipocytes siLHS présentent une amélioration du transport et de l’oxydation du glucose ainsi que de la LDN en réponse à l’insuline en parallèle à une forte augmentation de l’expression du facteur ChREBP. Ces cellules ont aussi une meilleure transmission du signal insulinique. La perte de ces effets dans les adipocytes siLHS/siChREBP suggère un rôle important de ce facteur dans les effets médiés par l’inhibition de la LHS. L’amélioration de la réponse à l’insuline des adipocytes siLHS pourrait s’expliquer par une augmentation de l’oléate au détriment du palmitoléate qui est partiellement perdue dans les cellules siLHS/siChREBP. Une expression différentielle de l’élongase ELOVL6 pourrait notamment expliquer ces différences. Conclusion Ces résultats mettent en évidence l’implication de la LDN, induite par l’augmentation du facteur ChREBP, dans l’amélioration de la sensibilité à l’insuline adipocytaire observée lors d’une inhibition de la LHS. This review aims to provide an update on our current knowledge of the various effects of pesticide cocktails. We have collected data from studies conducted in mammalian models in vitro and in vivo that was published between 2000 and 2014. All ecotoxicological studies were voluntarily excluded. Cocktail effects were classified according to how they had been classified by each author. The frequency of the various cocktail effects and the classes and chemical families of pesticides involved in the observed effects were assessed. When focusing on the function of pesticides (i.e. herbicide, insecticide or fungicide), 46% of the mixtures contained insecticides alone, 15% fungicides alone, and 4.5% herbicides alone. Mixtures with effects associated with neurotoxicity were mainly composed of insecticides, and most studies on the effects of fungicide mixtures (90%) were associated with effects on endocrine regulation and/or reproduction. Dose addition was observed with each kind of mixture except herbicide combinations. In contrast, synergic interactions or greater-than-additive effects were mainly reported for insecticide mixtures. There were few examples of potentiating and antagonistic interactions. We have identified chemical families of compounds specifically involved in synergy, addition, potentiation and antagonism, and those that do not interact when combined. The chemical families identified as being involved in synergy are in agreement with data from another recently published compilation of ecotoxicological studies. For most mixtures investigated, further validation data is still needed from experiments using other compounds and other experimental models but this update provides useful information to help in human health risk assessments. The Samtskhe-Javakheti volcanic plateau (Republic of Georgia) is the northernmost and youngest expression of the magmatism following the Arabia-Eurasia collision. Here, we present whole rock elemental and twenty-one new unspiked K/Ar and 40 Ar/ 39 Ar ages for the volcanic sequence well exposed east of the plateau. Based on our new radio-isotopic ages, we have identified three magmatic episodes. The oldest one (2.84–1.08 Ma), corresponding to the “Javakheti plateau s.s. ”, is mainly constituted of medium-K alkaline mafic lavas (basalt, basaltic trachyandesite) and of intermediate composition domes (dacite). The more recent volcanic activity has constructed an impressive N–S trending volcanic ridge (Samsari s.s. ) composed of evolved rocks (medium-K dacite to rhyolite). Within this ridge, two main periods of activity could be depicted: Middle Pleistocene (439–189 ka) and Late Pleistocene (90–13 ka). The youngest activity is restricted to the northern edge of this prominent magmatic structure and linked to the Tavkvetili volcano activity. According to these young ages, this area can be considered as a potential zone of volcanic hazards. The oldest volcanic activity shaping the Javakheti plateau is distributed between major strike slip faults in pull-apart position. The emplacement of the volcanism is controlled by a localized upper crustal extension. This is particularly outlined by the N–S linear array of domes that constitutes the Samsari ridge. This volcanic structure emplaced indeed on top of two major N–S faults that have probably played a key role to control the Middle to Late Pleistocene volcanism. The new 40 Ar/ 39 Ar ages date between 2.32 and 1.54 Ma the fauna assemblage of the Tsalka paleontological site. Rocks from the Samtskhe-Javakheti volcanic plateau derived from a low degree of melting of a metasomatized lithospheric mantle source (spinel facies). Except the obsidians from the Chickiani dome, they all derived from this source and evolved following a crystallization sequence involving mainly clinopyroxene, garnet, and/or amphibole. A crustal contamination component modified the composition of the youngest products (Samsari ridge rocks). According to the geochemical signature of these rocks, it seems that the magmatism does not fit with models involving asthenospheric upwelling in this region. [ABSTRACT FROM AUTHOR]

Abstract: Polytetrafluoroethylene can be considered as a model for calorimetric studies of complex systems with thermodynamics transitions at ambient temperature. This polymer exhibits two phase transitions of different nature at 292K and 303K. We show that sensitive ac-calorimetry measurements allow us to study the thermodynamic behaviour of polytetrafluoroethylene when it is brought out of thermodynamic equilibrium. Thanks to the thermal modelization of our calorimetric device, the frequency-dependent complex heat capacity of this polymer is extracted. The temperature and frequency variations of the real and imaginary parts of the complex heat capacity are obtained when polytetrafluoroethylene undergoes its first-order structural phase transition at 292K. The Teide and Pico Viejo stratocones and the Northwest and Northeast Rifts are products of the latest eruptive phase of the island of Tenerife, initiated with the lateral collapse of its northern flank that formed the Las Cañadas Caldera and the Icod--La Guancha Valley ca. 200 ka. The eruptive and structural evolution of this volcanic complex has been reconstructed after detailed geological mapping and radioisotopic dating of the significant eruptive events. A set of 54 new 14C and K/Ar ages provides precise age control of the recent eruptive history of Tenerife, particularly Teide Volcano, the third-highest volcanic feature on Earth (3718 m above sea level, >7 km high), and unique in terms of its intraplate setting. The development of the Teide--Pico Viejo Volcanoes may be related to the activity of the Northwest and Northeast Rifts. Volcanic and intrusive activity along both rift zones may have played an important role in activating the gravitational landslide and in the subsequent growth, nested within the collapse embayment, of an increasingly higher central volcano with progressively differentiated magmas. The coeval growth of the central volcano with sustained activity along the rifts led to a clear bimodal distribution in composition of eruptive products, with the basaltic eruptions in the distal part of the rifts and phonolitic and more explosive eruptions in the central area, where the differentiated stratocones developed. Current volcanic hazard in Tenerife is considered to be moderate, because eruptive frequency is low, explosivity is modest, and the eruptive activity of the Teide stratocone seems to have declined over the past 30 k.y., with only one eruption in this period (1150 yr B.P.).
