206. AKT overactivation can suppress DNA repair via p70S6 kinase-dependent downregulation of MRE11

Abstract

Deregulated AKT kinase activity due to PTEN deficiency in cancer cells contributes to oncogenesis by incompletely understood mechanisms. Here, we show that PTEN deletion in HCT116 and DLD1 colon carcinoma cells leads to suppression of CHK1 and CHK2 activation in response to irradiation, impaired G2 checkpoint proficiency and radiosensitization. These defects are associated with reduced expression of MRE11, RAD50 and NBS1, components of the apical MRE11/RAD50/NBS1 (MRN) DNA damage response complex. Consistent with reduced MRN complex function, PTEN-deficient cells fail to resect DNA double-strand breaks efficiently after irradiation and show greatly diminished proficiency for DNA repair via the error-free homologous recombination (HR) repair pathway. MRE11 is highly unstable in PTEN-deficient cells but stability can be significantly restored by inhibiting mTORC1 or p70S6 kinase (p70S6K), downstream kinases whose activities are stimulated by AKT, or by mutating a residue in MRE11 that we show is phosphorylated by p70S6K in vitro. In primary human fibroblasts, activated AKT suppresses MRN complex expression to escalate RAS-induced DNA damage and thereby reinforce oncogene-induced senescence. Taken together, our data demonstrate that deregulation of the PI3K-AKT/ mTORC1/ p70S6K pathways, an event frequently observed in cancer, exert profound effects on genome stability via MRE11 with potential implications for tumour initiation and therapy.

Contains fulltext : 96888.pdf (Publisher’s version ) (Open Access), EMPIRE10 (Evaluation of Methods for Pulmonary Image REgistration 2010) is a public platform for fair and meaningful comparison of registration algorithms which are applied to a database of intrapatient thoracic CT image pairs. Evaluation of nonrigid registration techniques is a nontrivial task. This is compounded by the fact that researchers typically test only on their own data, which varies widely. For this reason, reliable assessment and comparison of different registration algorithms has been virtually impossible in the past. In this work we present the results of the launch phase of EMPIRE10, which comprised the comprehensive evaluation and comparison of 20 individual algorithms from leading academic and industrial research groups. All algorithms are applied to the same set of 30 thoracic CT pairs. Algorithm settings and parameters are chosen by researchers expert in the configuration of their own method and the evaluation is independent, using the same criteria for all participants. All results are published on the EMPIRE10 website (http://empire10.isi.uu.nl). The challenge remains ongoing and open to new participants. Full results from 24 algorithms have been published at the time of writing. This paper details the organization of the challenge, the data and evaluation methods and the outcome of the initial launch with 20 algorithms. The gain in knowledge and future work are discussed.

Abstract

A DNA polymorphism is one of the most important factors determining individual variations in patients' responses to the same medicinal preparations. This work deals with assessment of associations between polymorphisms of 106 genes involved in key processes of cellular activity (xenobiotic metabolism, DNA repair, the cell cycle, and the regulation of apoptosis) and the outcomes of a cisplatin-based chemotherapy in a cohort of ovarian-cancer patients from Yakutia. The CDKN1B gene polymorphism (rs34330) has been found to be associated with the frequency of complete remissions. The allelic status of this locus was also shown to be significant for progression-free survival. The allelic status at the EPXH1 gene loci (rs2234922 and rs2260863) correlated with impairments in hearing, while the NBN gene polymorphism (rs1063045) was associated with severe emesis. A procedure was developed for the synthesis of gold and silver nanoparticles in the presence of polyols like ethylene glycol, diethylene glycol, triethylene glycol, and 1,2-propylene glycol. Spectral characteristics of the obtained nanocomposites were mesured and kinetics of the formation of the colloidal phase was studied. The influence of the polyol nature and concentration on the photoinitiated formation of silver and gold nanoparticles was studied. A pioneering procedure is presented for the photochemical synthesis of aqueous solutions of hollow bimetal silver-gold nanoparticles in the presence of poly- N-vinylpyrrolidone. The results were reported for the survey undertaken in Moscow and Krasnodar krai with the aim to elucidate the users' attitudes toward collection of spent household batteries. Their utilization is one of the most important problems of municipal solid waste management, whose solution involves organization of spent battery collection as the task of the worst complication. Based on the specially designed questionnaire, the users' willingness to participate in collection of spent batteries, as well as the preferred conditions for discarding spent and purchasing new batteries were determined.
