This study determined the roles of OX40 and OX40L in women with recurrent spontaneous abortion (RSA). We compared the expression of OX40 and OX40L genes in peripheral blood mRNA levels and serum levels of OX40L in women with a history of RSA to the control group. In this case-control study, 40 women with a history of RSA (case group), and 40 others with no history of abortion (control group) were investigated. The expressions of OX40 mRNA and OX40L mRNA were determined in the two groups using the quantitative polymerase chain reaction. Also, the enzyme-linked immunosorbent assay was used to determine the levels of serum OX40L in the two groups. There were no significant differences in the maternal age of women in the two groups (30.1 ± 4.28 years in the case vs. 30.03 ± 4.23 years in the control group). There was no difference in terms of the levels of OX40 and OX40L mRNA between the groups ( p = 0.08 and p = 0.56, respectively). In addition, there was no significant correlation between the expression of OX40 and OX40L mRNA levels with age or the number of abortions. The correlation between OX40 and OX40L mRNA levels was not significant. RSA history group turned to show a higher level of serum OX40L than the control group ( p = 0.03). In conclusion, our findings demonstrated that the expression of OX40 mRNA and OX40L mRNA was similar between women with a history of RSA and the control group. The elevation of serum OX40L level may be considered as a risk factor for RSA.

The striatal dopamine (DA) deficiency is known as the main cause of the clinical picture of Parkinson's disease (PD). The disease is a progressive degeneration of dopaminergic neurons in the striatum. The treatment of PD is based on compensation for the brain's supply of DA lost by drug therapy, deep brain stimulation, surgery, gene and cell therapies. Clinical studies have focused on the utility of stem cell-based therapies in PD. Embryonic and mesenchymal stem cells (MSCs) are widely used. Recently, human adipose derived stem cells (hADSCs) have been considered as a suitable source of tissue for this purpose. In this project, hADSCs differentiated into dopaminergic neurons and the specificity of the cell preparations was examined. Human adipose tissues were collected from healthy volunteers undergoing liposuction and hADSCs were isolated by collagenase-based enzymatic method. Flow cytometry was performed using the surface cluster of differentiation (CD) markers to confirm the cell typical properties. Then hADSCs were differentiated to dopaminergic neurons in neurobasal medium in the presence of differentiation factors and confirmed by immunocytochemistry via neuronal and dopaminergic markers. The isolated hADSCs were cultured and identified by the expression of MSCs surface markers including CD90, and CD44. These cells did not express hematopoietic surface markers such as CD45 and CD14. Differentiated cells express neuronal marker NeuN and dopaminergic marker tyrosine hydroxylase (TH). It is concluded that hADSCs can be easily taken from the patient's own body and differentiated into dopaminergic cells having a lower risk of transplant rejection.

Statement of the Problem: Recently, zeolite has been regarded to improve the properties of dental materials such as mineral trioxide aggregate (MTA)., Purpose: The aim of the present study was to evaluate the effect of incorporating zeolite/silver/zinc (Ze/Ag/Zn) composite at 2 wt% to MTA powder on the push-out bond strength in simulated furcation perforations., Materials and Method: Furcal perforations, measuring 1.3 mm in diameter and 2 mm in height, were simulated in 40 human mandibular first molars. The samples were allocated to two groups (n=20) based on the material used for the repair of perforations. In the group 1, MTA and in the group 2, MTA plus Ze/Ag/Zn (2%) was used. The samples were incubated at 37°C for 1 week. Then the universal testing machine was employed to measure bond strength. The resistance of materials to dislodgment was recorded in MPa. Data were analyzed using t-test. Statistical significance was set at p < 0.05., Results: The push-out bond strength in the group 1 (6.40±1.98 MPa) was significantly higher than that in the group 2 (2.1±0.6 MPa) with p = 0.001., Conclusion: Under the limitations of the present study, it can be concluded that incorporation of Ze/Ag/Zn at 2 wt% to MTA powder had a negative effect on the push-out bond strength.

Research Question: Can a combination of time-lapse morphokinetic parameters and cumulus cell gene expression in polycystic ovary syndrome (PCOS) women be used to predict assisted reproductive treatment outcome?, Design: A total of 547 embryos from 100 intracytoplasmic sperm injection (ICSI) cycles were evaluated. Fifty women with PCOS and 50 women who were categorized as tubal factor infertility were recruited. Time-lapse records were annotated for time to pronuclear fading (tPNf), time to 2 to 8 cells (t2-t8), reverse cleavage, direct cleavage and also for the presence of multinucleation. Expression levels of three genes involved in mitotic divisions, diaphanous-related formin 2 (DIAPH2), nibrin (NBN) and NIMA-related protein kinase (NEK4), were measured in 100 associated cumulus cell samples using quantitative real-time polymerase chain reaction., Results: Expression of DIAPH2 and NBN was significantly higher in the embryos of PCOS patients that resulted in implantation, biochemical and clinical pregnancies as well as live birth compared with embryos that were negative for these outcomes (P <0.01). However, in the tubal factor group, NBN gene expression was significantly higher in embryos resulting in biochemical pregnancy, clinical pregnancy and live birth (P <0.01) only. Multivariate logistic regression analysis showed that tPNf together with DIAPH2 gene expression were independent prognostic factors of clinical pregnancy rate and live birth in both groups., Conclusions: Some time-lapse embryo parameters may be related to cumulus gene expression and clinical outcome. Introduction: This study was designed to determine the effect of Osteon II mineralized bone powder on the surface microhardness of two retrofilling materials: Mineral trioxide aggregate (MTA) and Biodentine (BD)., Methods and Materials: Each retrograde material was mixed and carried into 30 sterile custom-made plastic cylinders. Half of the samples in each group were exposed to Osteon II. All cylinders were submerged in simulated tissue fluid and incubated at 37 ° C and 100% relative humidity for 7 days. Surface microhardness values of each study group was attained using Vicker's microhardness test. The data were analyzed statistically using two-way ANOVA and independent t test at a significance level of 0.05., Results: In all the setting conditions, BD had significantly greater surface microhardness than MTA ( P <0.001). Surface microhardness of both materials was significantly reduced in the presence of osteon II ( P =0.006 for BD and P <0.001 for MTA)., Conclusion: Mineralized bone graft materials negatively affect surface microhardness of both MTA and BD. Introduction: The aim of this retrospective study was to analyze the frequency of C-shaped root canal configuration and characterize mandibular root canal morphology using cone-beam computed tomography (CBCT) with 3D images in an Iranian population., Methods and Materials: This study consisted of retrospective evaluation of CBCT images from 231 adult patients (153 with bilateral second mandibular molars). Two endodontists examined 384 mandibular second molars of a population in Tabriz, Iran to determine the presence of C-shaped canals and their anatomical characteristics. Root canal configurations were categorized at three different levels. Bilateral or unilateral occurrence of C-shaped canals and their relationship to gender, age and tooth position were examined and statistically analyzed using chi squared test and Fisher's exact test in SPSS 17. The significance level was set at 0.05., Results: Of 384 mandibular second molars examined, 82 (21.4%) molars from 58 patients had a C-shaped root canal configuration. The prevalence of bilateral C-shaped canals was 15.6% amongst 153 patients with bilateral mandibular second molars. There were no significant differences in the distribution of C-shaped canals with respect to gender or age ( P =0.06 and P =0.86, respectively). Only 4 teeth (4.9%) had the same root canal configuration from the orifice to the apex. In the remainder of the teeth, the cross-sectional root canal configuration changed at different levels of the root., Conclusion: There were significant variations in the number of roots and canal morphology in mandibular second molars, which should be considered during debridement and obturation of the root canal system., Competing Interests: ‘None declared’.

Introduction: Previous studies have reported that it is difficult to obtain proper anesthesia in mandibular molars with symptomatic irreversible pulpitis, and supplemental injections are usually unavoidable. The aim of the present study was to determine the anesthetic efficacy of articaine in mandibular first molars with symptomatic irreversible pulpitis with 3 injection methods: an inferior alveolar nerve block (IANB), an IANB with an intraligamentary injection, and an IANB with buccal infiltration before initiating the endodontic treatment., Methods: Ninety-six patients (54 women and 42 men) with a diagnosis of symptomatic irreversible pulpitis in mandibular first molars were selected and randomly assigned into 3 groups (n = 32) according to the injection method as follows: group 1, a conventional IANB injection; group 2, an IANB injection, and after profound lip numbness after the IANB (after 15 minutes), buccal infiltration was administered; and group 3, an IANB injection, and after profound lip numbness after the IANB (after 15 minutes), an intraligamentary injection was performed, and after 20 minutes from the IANB, the endodontic treatment was initiated. The anesthetic solution was articaine 4% with 1:100,000 epinephrine. Success was defined as no or mild pain on the basis of the visual analog scale recording upon access cavity preparation or initial instrumentation. Data were statistically analyzed using the chi-square and Mann-Whitney U tests, and P < .05 was set as significant., Results: The success rate for IANBs with an intraligamentary injection was 75%, and for IANBs with a buccal injection, it was 65.6%. For IANBs alone, the success rate was 28.1%., Conclusions: Considering the limitations of the present study, in can be concluded that the success rate of IANBs increased with intraligamentary injections and buccal infiltrations with articaine that were performed before initiating treatment. Background: HLA-G, a major histocompatibility complex of non-classical class Ib, plays a key role in the development of the primary tumors to metastatic stages. The aim of this study was to investigate HLA-G expression in oral squamous cell carcinomas and its relationship with clinicopathological factors. Methods: After immunohistochemical staining for HLA-G with 63 formalin fixed and paraffin embedded blocks (33 oral squamous cell carcinoma and 30 normal oral mucosa samples), staining intensity, percentage of stained cells and final immunoreactivity score were evaluated, along with other variables. Results: Staining intensity, percentage of stained cells and final immunoreactivity scores in oral squamous cell carcinomas were higher than those in normal oral mucosa (all P=0.001). The staining intensity in the parenchyma of squamous cell carcinoma cells was significantly associated with the clinical tumor stage (P=0.022) and the group with lymphatic metastasis exhibited a higher staining percentage (P=0.026). Staining intensity and immunoreactivity score (IRS) exhibited a significant but inverse correlation with survival rate (P=0.004 and P=0.018, respectively) and a significant direct relationship with clinical stage (P=0.001 and P=0.001). Conclusion: The results supported a role of HLA-G in development of oral squamous cell carcinomas and metastasis to lymph nodes. Background: The quality of oocyte is often considered as a limiting factor for fertility, especially IVF. Some mitochondrial mutations, particularly the 4977-bp deletion increase with the age. Thus, this mutation can serve as a marker for cell aging, which indicates the reduced quality of the oocytes for fertilization. It has been suggested that this can also be investigated in the blood cells of women with IVF failure., Objective: 1-Determination of the frequency of 4977-bp deletion in women with IVF failure, 2-Investigation of the relationship between 4977-bp deletion and the age of patients., Materials and Methods: Polymerase chain reaction was used to detect the 4977-bp deletion in blood samples of 52 IVF failure women and 52 women who had at least one healthy child. After polymerase chain reaction with deleted and wild-type primers, the products were examined using agarose gel electrophoresis., Results: 48.07% of women with IVF failure and 34.62% of healthy women had a mitochondrial 4977-bp deletion, with p=0.163 and OR: 1.749. Also, in association with the age of these patients and the frequency of 4977-bp mutation, p and OR were obtained 0.163 and 1.749, respectively and frequency of this mutation was higher in patients over 35 yr old compared to other subgroups (Patients ≥35: 57.69)., Conclusion: According to the findings of this study, there is no a significant relationship between the frequency of mitochondrial 4977-bp mutation and failure in IVF., Competing Interests: All the authors declare that there is no actual or potential conflict of interest.

Objectives: The presence of neurotrophic factors is critical for regeneration of neural lesions. Here, we transplanted combination of neurotrophic factor secreting cells (NTF-SCs) and human adipose derived stem cells (hADSCs) into a lysolecithin model of multiple sclerosis (MS) and determined the myelinization efficiency of these cells., Materials and Methods: In this experimental study, 50 adult rats were randomly divided into five groups: control, lysolecithin, vehicle, hADSCs transplantation and NTF-SCs/ hADSCs co-transplantation group. Focal demyelization was induced by lysolecithin injection into the spinal cord. In order to assess motor functions, all rats were scored weekly with a standard experimental autoimmune encephalomyelitis scoring scale before and after cell transplantation. Four weeks after cell transplantation, the extent of demyelination and remyelination were examined with Luxol Fast Blue (LFB) staining. Also, immunofluorescence method was used for evaluation of oligodendrocyte differentiation markers including; myelin basic protein (MBP) and Olig2 in the lesion area., Results: Histological study show somewhat remyelinzation in cell transplantation groups related to others. In addition, the immunofluorescence results indicated that the MBP and Olig2 positive labeled cells were significantly higher in co-cell transplantation group than hADSCs group (P<0.05). Also, outcome of motor functional test showed significant improvement function in cell transplantation groups, as compared to the others (P<0.01)., Conclusions: Our results indicated that the remyelinization process in co-cell transplantation group was better than other groups. Background: Multiple Sclerosis (MS) has been explained as an autoimmune mediated disorder in central nerve system. Since conventional therapies for MS are not able to stop or reverse the destruction of nerve tissue, stem cell-based therapy has been proposed for the treatment of MS. Astaxanthin (AST) is a red fat-soluble xanthophyll with neuroprotection activity. The aim of this study was evaluation of pre-inducer function of AST on differentiation of human Adipose-Derived Stem Cells (hADSCs) into oligodendrocyte precursor cells., Methods: After stem cell isolation, culture and characterization by flow cytometry, hanging drop technique was done for embryoid body formation. In the following, hADSCs were differentiated into oligodendrocyte cells in the presence of AST at various concentrations (1, 5, and 10 ng/ml ). Finally, immunocytochemistry and real-time PCR techniques were used for assessment of oligodendrocyte differentiation., Results: Flow cytometry results indicated that hADSCs were CD44, CD49-positive, but were negative for CD14, CD45 markers. In addition, immunocytochemistry results revealed that, in AST treated groups, the mean percentage of Olig 2 and A2B5 positive cells increased especially in 5 ng/ml AST treated group compared to control group (p<0.001). Moreover, real-time PCR analysis confirmed the results of immunocytochemistry., Conclusion: Since hADSCs have the potential to differentiate into multi lineage cells and due to important functions of AST in regulating various cellular processes, it seems that AST can be used as a promoter for oligodendrocyte differentiation of hADSCs for being used in cell transplantation in multiple sclerosis.

Background: Bariatric surgeries have turned to be a popular therapeutic option for morbid obesity nowadays. Gut microbiota is supposed to be responsible as a part of the bariatric surgeries success. In this systematic review, we detailed the human studies which investigated the effect of different bariatric surgeries on the composition of gut microbiota., Methods: We did a comprehensive search in PubMed, Web of Science, and Scopus for all clinical trials and longitudinal observational studies documented up to December 2015., Results: Our initial search yielded 1423 articles. After screening abstracts and full texts, 7 articles were included. In 6 studies, the type of surgical intervention was Roux-en-Y gastric bypass (RYGB) where one study assessed vertical banded gastroplasty (VBG), too. Only in one study the effect of laparoscopic sleeve gastrectomy (LSG) had been investigated. RYGB caused an increase in Proteobacteria and a decrease in Firmicutes. LSG led to less severe intestinal microbiota alteration compared to RYGB. Fecalibacterium prausnitzii species with anti-inflammatory properties increased after LSG. However, inconsistent alterations have been shown in abundance of Fecalibacterium species after RYGB., Conclusion: Weight loss after bariatric surgeries are associated with microbiota modifications caused by surgical procedures., (Copyright © 2018 Elsevier Ltd. Introduction: During periapical surgery, using of bone products in large endodontic lesions, is a treatment option that could affect the properties of the retro-filling endodontic material. The aim of present study was to evaluate the effect of Osteon II bone powder on the surface microhardness of calcium-enriched mixture (CEM) and mineral trioxide aggregate (MTA)., Methods and Materials: Each material was mixed and carried into 40 sterile custom-made plastic cylinders. Half of the samples in each group were exposed to Osteon II. All cylinders were submerged in simulated tissue fluid and incubated at 37 ° C and 100% relative humidity for 7 days. Surface microhardness values of each study group was attained using Vickers microhardness test. The data were analyzed statistically using two-way ANOVA and independent t -test at a significance level of 0.05., Results: The highest and lowest microhardness values were recorded in the MTA/without Osteon and MTA/with Osteon groups, respectively. Irrespective of the presence or absence of bone powder, the overall microhardness of CEM cement and MTA was not significantly different. In the MTA group, the presence of the powder resulted in a significant decrease ( P <0.05) of the microhardness; however, its effect on CEM cement was not significant ( P >0.05)., Conclusion: Under the limitations of the present in vitro study, the presence of Osteon bone powder had no negative effect on the microhardness of CEM cement, contrary to its effect on MTA., Competing Interests: ‘None declared’.

Objective: To determine the localization, expression, and function of Toll-like receptors (TLRs) in fallopian tube epithelial cells., Methods: The localization of TLRs in fallopian tube epithelial cells was investigated by immunostaining. Surprisingly, the intensity of staining was not equal in the secretory and ciliated cells. After primary cell culture of fallopian tube epithelial cells, ring cloning was used to isolate colonies of ciliated epithelial cells, distinct from non-ciliated epithelial cells. The expression of TLRs 1-10 was examined by quantitative real-time polymerase chain reaction, and protein localization was confirmed by immunostaining. The function of the TLRs was determined by interleukin (IL)-6 and IL-8 production in response to TLR2, TLR3, TLR5, TLR7, and TLR9 ligands., Results: Fallopian tube epithelial cells expressed TLRs 1-10 in a cell-type-specific manner. Exposing fallopian tube epithelial cells to TLR2, TLR3, TLR5, TLR7, and TLR9 agonists induced the secretion of proinflammatory cytokines such as IL-6 and IL-8., Conclusion: Our findings suggest that TLR expression in the fallopian tubes is cell-type-specific. Background: Stem cell-based therapy is a new method for the treatment of neurodegenerative diseases such as multiple sclerosis (MS). Human adipose-derived stem cells (hADSCs) are a kind of adult stem cells which have a higher frequency in the fat tissue and have the ability to differentiate into other cell types outside their lineage. Due to some serious adverse events of cell-based therapy such as tumorigenic potential, the aim of this study was to evaluate of hADSCs differentiation into oligodendrocytes as a valuable way for future cell transplantation. Methods: hADSC were isolated from lipoaspirate samples of human abdominal fat. After hADSC characterization via flow cytometry, the cells were induced to oligodendrocytes using a special differentiation medium. Finally, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), immunocytochemistry, and real-time polymerase chain reaction (RT-PCR) techniques were used for the evaluation of differentiated cells. Results: Flow cytometry indicated that hADSCs were CD105- and CD49-positive, but were negative for CD31 and CD45 markers. In addition, immunocytochemistry analysis revealed that a high percent of differentiated cells expressed oligodendrocyte progenitor cells markers [A2B5 and oligodendrocyte transcription factor (Olig2)] which were significantly higher than myelin basic protein (MBP) which is mature oligodendrocytes marker. Moreover, a very low percentage of differentiated cells expressed glial fibrillary acidic protein (GFAP) marker. Finally, real-time reverse transcription PCR analysis confirmed the results of immunocytochemistry. Conclusion: Since hADSCs have the potential to differentiate into multi-lineage cells and due to their additional characteristics such as immunomodulatory and neuroprotective properties, it seems that these cells may be an ideal cell source for oligodendrocytes differentiation.

Parkinson's disease (PD) is one of the most neurodegenerative disorders which can lead to severe neural disability and neurological defects. Cell-based therapy using fully differentiated cells is a new method for the treatment of this abnormal condition. In the present study, we investigated the effects of 6-bromoindirubin-3'-oxime (BIO) on dopaminergic differentiation of human immortalized RenVm cells in order to obtain a set of fully differentiated cells for transplantation in Parkinson's disease. To this end, the immortalized RenVm cells were induced to dopaminergic differentiation using a neuro basal medium supplemented with N2 and different concentrations (75, 150, 300, 600, and 1200 nM) of BIO for 4, 8, and 12 days. The efficiency of dopaminergic differentiation was determined using immunocytochemistry for tyrosine hydroxylase expressions. In addition, the expression of a β-catenin marker was measured using the western blot technique. The results of immunocytochemistry revealed that the mean percentage of Tuj1- and TH-positive sells in 150- and 300-nM-BIO-treated groups was significantly increased compared to that of other groups ( p  ≤ 0.01). In addition, the expression of the β-catenin marker was higher in these groups as compared with that of other groups. Introduction: This systematic review protocol aims to perform a protocol for assessing the effectiveness, safety, and mechanism of herbal medicines for the treatment of obesity and metabolic syndrome. Some systematic reviews conducted earlier have evaluated the efficacy of herbal medicine to treat obesity and metabolic syndrome. Owing to the huge burden imposed by obesity in the recent years, a need is felt for conducting new systematic reviews on the topic of obesity with a focus on randomized clinical trials, we felt the need., Methods and Analysis: All relevant clinical trials that examine the effectiveness of herbal medicines for the treatment of obesity and metabolic syndrome without restrictions on publication status will be applied.Four electronic databases will be searched: PubMed, Scopus, Web of Science, and the Cochrane Central Register of Controlled Trials. Introduction: The complexity of the root canal system presents a challenge for the practitioner. This systematic review evaluated the papers published in the field of root canal anatomy and configuration of the root canal system in permanent maxillary second molars., Methods and Materials: All articles related to the root morphology and root canal anatomy of the permanent maxillary second molars were collected by suitable keywords from PubMed database. The exhaustive search included all publications from 1981 to December 2015. The articles relevant to the study were evaluated and data was extracted. The author/year of publication, country, number of the evaluated teeth, type of study (method of the evaluation), number of roots and the canals, type of canals and the morphology of the apical foramen was noted., Results: The highest studied populations were in Brazil and United States. A total of 116 related papers were found, which had investigated 11945 teeth in total. Across all the studied populations, the three-rooted anatomy was most common, while the four-rooted anatomy had the lowest prevalence. The presence of the second mesiobuccal canal ranged from 11.53 % to 93.7%, where type II (2-1) configuration was the predominant type in Brazil and USA and types II and III (1-2-1) in Chinese populations. In 8.8-44% of cases, fusion was observed. The main reported cases were related to palatal root. The major method of anatomical investigation in case reports was periapical radiography, and the chief method in morphological studies was CBCT., Conclusion: The clinicians should be aware of normal morphology and anatomic variations to reduce the treatment failure.

Background: Stem cell-based therapy is a novel strategy for the treatment of neurodegenerative diseases. The transplantation of fully differentiated cells instead of stem cells in order to decrease serious adverse complications of stem cell therapy is a new idea. In this study, the effect of lithium chloride on dopaminergic differentiation of human immortalized RenVm cells was investigated in order to access a population of fully differentiated cells for transplantation in Parkinson disease., Methods: The immortalized RenVm cells were induced to dopaminergic differentiation using a neurobasal medium supplemented with N2 and different concentrations (1, 3, 6 mM ) of Lithium Chloride (LiCl) for 4, 8 and 12 days. The efficiency of dopaminergic differentiation was evaluated using immunocytochemistry and western blot techniques for tyrosine hydroxylase and β-catenin marker expression., Results: Our results indicated that LiCl can promote dopaminergic differentiation of RenVm cells in a dose-dependent manner., Conclusion: It can be concluded that LiCl is able to facilitate dopaminergic differentiation of cultured cells by affecting Wnt-frizzled signaling pathway.

Background: Tumor necrosis factor-alpha ( TNF -α) is a multifunctional cytokine that regulates different cellular activities related to spermatogenesis. Tumor necrosis factor-alpha receptor 1 ( TNFR1 ) mediates TNF -α activity and polymorphism in TNFR1 could lead to gene dysfunction and male infertility., Objective: The aim of this study is to determine the association of TNFR1 36 A/G polymorphism with the idiopathic azoospermia in Iranian population., Materials and Methods: This case-control study included 108 azoospermic and 119 fertile men. This research investigated the frequency of TNFR1 36 A/G polymorphism in cases who were idiopathic azoospermic men referred to Yazd Research and Clinical Center for Infertility, Iran in comparison with controls. polymerase chain reaction- restriction fragment length polymorphism (PCR-RFLP) method was used to investigate the polymorphism in both case and control groups. PCR fragments were digested by Mspa1I enzyme and products were appeared by gel electrophoresis. The abundance of A→G was calculated in the azoospermic and healthy men., Results: According to the present study, GG and AG genotypes frequency in the azoospermic men group were higher than the control group (OR= 2.298 (1.248-4.229), p=0.007), (OR=1.47 (0.869-2.498, p=0.149). Our findings also showed that G allele frequency in azoospermic men had significant difference compared to the control group (OR=2.302 (1.580-3.355), p<0.001)., Conclusion: It seems that the GG genotype and G allele have an association with increased risk of non-obstructive azoospermia., Competing Interests: The authors declare that there is no conflict of interest regarding this study.

Vicia sativa (common vetch), which contains β-cyanoalanine (BCA) and γ-glutamyl β-cyanoalanine (γGBCA) is used for adulteration of lentil and have neuro toxic effects on people and animals. Therefore the determination of BCA and γGBCA analytes in accurate and reliable manner has high importance for human and animal health. LC-IDMS/MS method has been developed and validated for quantitative analyses of β-cyanoalanine and 13 C labeled BCA is used as an internal standard in γGBCA analysis. In order to validate the method, linearity, recovery, precision (repeatability), intermediate precision, limit of detection, and limit of quantification parameters were investigated. The correlation coefficient was found to be greater than 0.99 for both analytes. The recoveriess were determined as 95.8% for BCA and 97.4% for γGBCA. The relative expanded measurement uncertainties of β-cyanoalanine and γ-glutamyl-β-cyanoalanine were obtained as 4.6% and 5.8%, respectively with the coverage factor, k, is 2 and at 95% confidence level., (Copyright © 2016 Elsevier Ltd. Multiple sclerosis (MS) is a chronic inflammatory disease characterized by central nervous system (CNS) lesions that can lead to severe physical or cognitive disability as well as neurological defects. Although the etiology and pathogenesis of MS remains unclear, the present documents illustrate that the cause of MS is multifactorial and include genetic predisposition together with environmental factors such as exposure to infectious agents, vitamin deficiencies, and smoking. These agents are able to trigger a cascade of events in the immune system which lead to neuronal cell death accompanied by nerve demyelination and neuronal dysfunction. Conventional therapies for MS are based on the use of anti-inflammatory and immunomodulatory drugs, but these treatments are not able to stop the destruction of nerve tissue. Thus, other strategies such as stem cell transplantation have been proposed for the treatment of MS. Overall, it is important that neurologists be aware of current information regarding the pathogenesis, etiology, diagnostic criteria, and treatment of MS. Thus, this issue has been discussed according to recent available information.

Objectives: Neurotrophic factors secreting cells (NTS-SCs) may be a superior cell source for cell-based therapy in neurodegenerative diseases. NTS-SCs are able to secrete some neurotrophic Such as nerve growth factor and glia-derived neurotrophic factor. Our primary aim was to assess transplantation of neurotrophic factor secreting cells derived from human adipose-derived stem cells (hADSCs) into the damaged spinal cord rats and determine the potential of these cells in remyelination., Materials and Methods: To this end, 40 adult male Wistar rats were categorized into four groups including; control, lysolecithin (Lysophosphatidylcholines or LPC), vehicle, and NTS-SCs transplan-tation. Local demyelination was induced using LPC injection into the lateral column of spinal cord. Seven days after the lysolecithin lesion, the cells transplantation was performed. The ultrastructure of myelinated fibers was examined with a transmission electron microscope to determine the extent of myelin destruction and remyelinization 4 weeks post cell transplantation. Moreover, the presence of oligodendrocyte in the lesion of spinal cord was assessed by immunohistochemistry procedure., Results: The results of current study indicated that in NTF-SCs transplantation group, the remyelination process and the mean of myelin sheath thickness as well as axonal diameters were significantly higher than other groups ( P <0.001). Furthermore, immunohistochemistry analysis revealed that in NTF-SCs transplantation group more than 10 percent of transplanted cells were positive for specific markers of oligodendrocyte cells., Conclusion: NTF-SCs transplantation represents a valuable option for cell-based therapy in the nervous tissue damages.

Background. of this study was to investigate the effect of different mixing methods (ultrasonic, amalgamator, and conventional) on the bacterial microleakage of white Portland cement (WPC) and white MTA (Tooth-colored Formula, Dentsply, Tulsa Dental, Tulsa, OK). Methods. A hundred human single-rooted permanent teeth were decoronated to obtain 14 mm of root length in all the samples. The root canals were cleaned, shaped and obturated. Three millimeters of each root apex were cut off and randomly divided into 6 groups of 15 each (3 groups for WMTA and 3 groups for WPC, each with 3 different mixing methods) and 2 positive and negative control groups (each containing 5 samples). Brain-heart infusion agar (BHI) suspension containing the bacterial species Enterococcus faecalis (ATCC 29212) was used for leakage assessment. Statistical analysis was carried out using descriptive statistics and Kaplan Mayer survival analysis with censored data and log rank test using SPSS 18. Statistical significance was set at P<0.05. Results. The survival means in PC for conventional method, amalgamator, and ultrasonic were 80.2±13.64, 78.5±13.46 and 84.667±11.42 days, with 49.13±12.96, 66±13.32 and 69.07±11.5 days for MTA, respectively. The log rank test showed no significant differences between the three methods in each material (P>0.05). Conclusion. Bacterial microleakage in the studied samples was not significantly different in terms of the type of the mixing method.

Several factors can affect performance of an ultrasound system such as quality of excitation signal and ultrasound transducer behaviour. Nonlinearity of piezoelectric ultrasound transducers is a key determinant in designing a proper driving power supply. Although, the nonlinearity of piezoelectric transducer impedance has been discussed in different literatures, the trend of the nonlinearity at different frequencies with respect to excitation voltage variations has not been clearly investigated in practice. In this paper, to demonstrate how the nonlinearity behaves, a sandwich piezoceramic transducer was excited at different frequencies. Different excitation signals were generated using a linear power amplifier and a multilevel converter within a range of 30-200V. Empirical relation was developed to express the resistance of the piezoelectric transducer as a nonlinear function of both excitation voltage and resonance frequency. The impedance measurements revealed that at higher voltage ranges, the piezoelectric transducer can be easily saturated. Also, it was shown that for the developed ultrasound system composed of two transducers (one transmitter and one receiver), the output voltage measured across receiver is a function of a voltage across the resistor in the RLC branches and is related to the resonance frequencies of the ultrasound transducer., (Copyright © 2016 Elsevier B.V. Objective: Process of angiogenesis is essential for successful gestation. Disruption in this pathway leads to various undesirable consequences in pregnancy such as recurrent spontaneous abortion (RSA). One of the most important genes involved in angiogenesis is kinase-insert domain-containing receptor (KDR). This study aimed to investigate the associations between two single-nucleotide polymorphisms (SNPs) of KDR gene, 1719A > T and 1192G > A, with idiopathic RSA in south-east Iran., Methods: A total of 230 women, including 110 women with a history of at least two consecutive spontaneous miscarriages and 120 healthy women were recruited in this study. Genomic DNA was extracted from peripheral blood samples of participants using the Salting out method. The KDR 1719A > T and 1192G > A polymorphisms were genotyped by the standard amplification refractory mutation system-polymerase chain reaction (ARMS-PCR) technique., Results: For the case group, frequencies of 2.73%, 30% and 67.27% were observed for AA, AT and TT genotypes in1719A > T SNP, respectively, and the genotype frequencies for controls were equal to AA = 3.33%, AT = 32.5% and TT = 64.17%. Distribution of genotypes in 1192G > A SNP in the case group was 79.1%, 19.1% and 1.8% for GG, AG and AA, respectively, whereas the corresponding values for the controls were GG  =  80%, AG  =  20% and AA  =  0. No significant difference was found between the case and control groups based on the frequency of KDR gene polymorphisms with the susceptibility to RSA., Conclusions: There is no association between these two SNPs of KDR gene and the susceptibility to RSA in women from south-east Iran.

Background: Calcium-enriched mixture (CEM) cement is used in the field of endodontics. It is similar to mineral trioxide aggregate in its main ingredients. The present study investigated the effect of different mixing methods on the bacterial microleakage of CEM cement., Methods: A total of 55 human single-rooted human permanent teeth were decoronated so that 14-mm-long samples were obtained and obturated with AH26 sealer and gutta-percha using lateral condensation technique. Three millimeters of the root end were cut off and randomly divided into 3 groups of 15 each (3 mixing methods of amalgamator, ultrasonic and conventional) and 2 negative and positive control groups (each containing 5 samples). BHI (brain-heart infusion agar) suspension containing Enterococcus faecalis was used for bacterial leakage assessment. Statistical analysis was carried out using descriptive statistics, Kaplan-Meier survival analysis with censored data and log rank test. Statistical significance was set at P<0.05., Results: The survival means for conventional, amalgamator and ultrasonic methods were 62.13±12.44, 68.87±12.79 and 77.53±12.52 days, respectively. The log rank test showed no significant differences between the groups., Conclusions: Based on the results of the present study it can be concluded that different mixing methods had no significant effect on the bacterial microleakage of CEM cement.

Attention-deficit hyperactivity disorder (ADHD) is a common psychiatric disorder among preschool children but the number of controlled clinical trials regarding psychopharmacological treatment in this age group is limited. The aim of this study was to compare methylphenidate with the combination of methylphenidate and risperidone in preschool children with ADHD. Forty-two preschool children, aged 3-6 years, diagnosed with ADHD by a child and adolescent psychiatrist according to the Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition-Text Revision criteria, were enrolled in a 6-week, single-blind clinical trial and administered with methylphenidate (5-30 mg/dl) or the combination of methylphenidate and risperidone (0.25-2 mg/dl) in Iran. Treatment outcomes were assessed using the Conners' Rating Scale and Clinical Global Impression (CGI) Scale at baseline and 3 and 6 weeks after starting the drugs administration. Side effects were rated by a checklist and body weight was measured at each visit. There were no significant differences between the two protocols in Parent Conners' Rating Scale scores ( P > 0.05) and CGI scores ( P > 0.05). Both groups showed a significant improvement in ADHD symptoms over the 6 weeks of treatment for Parent Conners' Rating Scale ( P < 0.001). The combination group used significantly lower doses of methylphenidate compared to the other group ( P = 0.002). The most common adverse effects were anorexia (21.7%) and daytime drowsiness (17.4%) in combination treatment group and insomnia (33.3%) and anorexia (25%) in methylphenidate group. Risperidone and methylphenidate may be effective and well tolerated in preschool children with ADHD, and adding risperidone to methylphenidate may decrease the occurrence of some side effects of methylphenidate such as insomnia and anorexia and lower the dose of methylphenidate may be needed to control symptoms.

Introduction: The aim of this study was to evaluate the effect of adding propylene glycol (PG) to mineral trioxide aggregate (MTA) liquid with volume ratio of 20% on the compressive strength (CS) of MTA in two time periods (4 and 21 days) after mixing., Methods and Materials: Four groups of steel cylinders ( n =15) with an internal diameter of 3 and a height of 6 mm were prepared and MTA (groups 1 and 2) and MTA+PG (80% MTA liquid+20% PG) (groups 3 and 4) were placed in to the cylinders. In groups 1 and 3 the CS was evaluated after 4 days and in groups 2 and 4 after 21 days. Data were calculated using the two-ways ANOVA. The level of significance was set at 0.05., Results: The highest (52.22±18.92 MPa) and lowest (4.5±0.67 MPa) of CS was obtained in 21-day MTA samples and 4-day MTA+PG specimen, respectively. The effect of time and PG were significant on the CS ( P <0.05). Mixing MTA with PG significantly reduced the CS; but passing the time from 4 to 21 days significantly increased the CS., Conclusion: Considering the limitations of this study, PG had a negative effect on CS of MTA.

Introduction: One of the most common complications of root canal treatment is postoperative pain. The aim of the present clinical trial was to compare the severity of postoperative pain after root canal preparation with RaCe rotary system and hand K-Flexofile., Methods and Materials: A total of 96 mandibular first and second molars were divided into two groups ( n =48) based on root canal preparation technique. The teeth in both groups underwent one-session root canal treatment and the severity of postoperative pain was evaluated using visual analog scale (VAS) at 4-, 8-, 12-, 24- and 48-h and 1-week intervals. In addition, the type and dosage of analgesics were recorded. Data were analyzed with repeated-measures ANOVA. Statistical significance was set at 0.05., Results: The difference between the two groups during this period and at subsequent intervals were not significant ( P >0.05). There were no significant differences between the two groups in type and the number of analgesics in pain-free subjects ( P =0.12 and P =0.61, respectively)., Conclusion: There were no statistically significant differences in pain severity between the two groups at any intervals.

Background: Human leukocyte antigen-E (HLA-E)is a non-classical major histocompatibility complex (MHC) class I antigens which expressed on extra villous cytotrophoblast, which interacts with NKG2A, is an inhibitory receptor on natural killer (NK) cells and leading to down regulation of immune response in the maternal-fetal interface and provides maternal immune tolerance of the fetus., Objective: This study was designated to investigate the gene frequencies of E0101 and E0103 in HLA-E gene in Iranian women with recurrent spontaneous abortion (RSA)., Materials and Methods: Amplification Refractory Mutation System (ARMS-PCR) technique was carried out to detect polymorphism in exon 3 of the HLA-E gene in women with RSA and controls (n=200). Differences between groups were analyzed by SPSS19 software using (2) test., Results: There was no significant difference in the allele frequencies of the HLA-E polymorphism between RSA and fertile controls but HLA-E 0101/0103 heterozygous genotype was found to be significantly higher in RSA group (p=0.006, OR=1.73), so this genotype might confer susceptibility to RSA., Conclusion: Our results suggest that HLA-E 0101/0103 heterozygous genotype leads to increase of RSA risk. It seems that by genotyping of HLA-E polymorphism, we can predict the risk of RSA in infertile women.

Human leukocyte antigen (HLA)-G is involved in immunoregulatory processes and particularly in pathogenesis of inflammatory disorders such as recurrent spontaneous abortions (RSA). The purpose of the current study was to examine whether two single nucleotide polymorphisms (SNPs) of HLA-G gene (rs1736936 and HLA-G*0105N) influence susceptibility to recurrent spontaneous abortion. Genomic DNA from 117 RSA patients and 117 normal fertile control individuals was isolated using the salted out method. The two single nucleotide polymorphisms in HLA-G gene were analyzed using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Differences between the two groups were analyzed by SPSS19 software using Chi-square test. The results revealed a significant increase in HLA-G*0105N allele in the proportion of whole group of RSA women compared with fertile controls (P value = 0.015), OR (95 % CI) = 2.054 (1.798-2.347), as well as an absence of homozygosity for HLA-G*0105N in the study population. No significant difference was observed between the RSA and the fertile groups in terms of alleles and genotypes frequency of rs1736936 (P value = 0.323), OR (95 CI %) = 1.056 (0.844-1.319). The presented data suggest that the investigated HLA-G*0105N allele is potentially associated with RSA through linkage disequilibrium with other genetic elements. Meanwhile, the rs1736936 SNP do not predispose to RSA in the study population.

Aims: The genetics of folate metabolism is one of the most significant mechanisms influencing fetal growth and may underlie some cases of unexplained recurrent miscarriage. Reduced folate carrier 1, encoded by the SLC19A1 gene, is a transporter of folate. Folate deficiency and elevated levels of homocysteine could be disadvantageous for the female reproductive system health. Thus, the balance between homocysteine and folate status can be used to measure the risk of recurrent pregnancy loss., Methods: The purpose of this study was to determine the association between -43T>C, 80G>A, and 696C>T polymorphisms of the SLC19A1 gene in 147 women who had unexplained recurrent miscarriage in comparison with 150 healthy women. Amplification refractory mutation system-polymerase chain reaction was used to genotype the molecular polymorphisms of this gene., Results: The results indicated that the -43T>C single nucleotide of the SLC19A1 gene was significantly associated with a risk of recurrent miscarriage in Iranian women (p < 0.05). No significant association was observed for the other two polymorphisms. The haplotype frequency distribution of -43C/80G/696C, -;43C/80G/696T, -43C/80G, and 80G/696T was significantly different in patients than controls, which may represent a novel risk factor for idiopathic recurrent pregnancy loss., Conclusions: Polymorphisms and haplotypes of the SLC19A1 gene can be considered risk factors for idiopathic recurrent pregnancy loss.

Background: The aim of this study was to assess plasma levels of endothelium- and heart-associated microRNAs (miRNAs) miR-126 and miR-499, respectively, using quantitative reverse transcriptase polymerase chain reaction., Methods: A two-step analysis was conducted on 75 patients undergoing off-pomp coronary artery bypass graft (CABG) surgery. Five biomarkers of inflammation and cardiac injury were assessed in addition to the above-mentioned miRNAs., Results: Plasma concentrations of miRNAs were found to be significantly correlated with plasma levels of cardiac troponin I (cTnI) (miR-499, r 0.49, p~0.002; miR-126, r = 0.30, p~0.001), indicating cardiac damage. Data analysis revealed that miR-499 had higher sensitivity and specificity for cardiac injury than miR-126, which reflects more endothelial activation. Interestingly, a strong correlation was observed between both miRNAs and uric acid (UA) levels with ventricular contractility measured as ejection fraction (EF) (miR-499/EF%, r = 0.58, p~0.004; UA/EF%, r = -0.6, p~0.006; UA/miR-499, r = -0.34; UA/miR-126, r = 0.5, p~0.01)., Conclusions: In patients undergoing CABG, circulating miR-126/499 is associated with presentation of traditional risk factors and reflects post-operative response to injury. Plasma pool of miRNAs likely reflects extracellular miRNAs which are proportional to intracellular miRNA levels. Introduction: The aim of this in vitro study was to evaluate the effect of calcium hydroxide (CH) on push-out bond strength of white mineral trioxide aggregate (WMTA) and calcium-enriched mixture (CEM) cement in simulated furcation perforations., Methods and Materials: Furcation perforations, measuring 1.3 mm in diameter and 2 mm in height, were created in 80 human mandibular first molars. The teeth were then divided into 4 groups (n=20). In groups 1 and 3 CH was placed in the perforation for one week, before placement of WMTA and CEM. In groups 2 and 4 perforations were repaired without placement of CH. In groups 1 and 2 the perforation sites were repaired with WMTA and CEM cement was used in groups 3 and 4. After 7 days, the push-out test was carried out using a universal testing machine. Data were analyzed with two-way ANOVA. The level of significance was set at 0.05., Results: The maximum and minimum bond strength values were recorded in the WMTA/CH (13.08±1.8 MPa) and CEM cement groups (8.03±0.98 MPa), respectively. There were significant differences in resistance to dislodgement between the WMTA/CH and other groups (P<0.05)., Conclusion: Placement of CH before placement of WMTA in furcation perforation improves the push-out bond strength of this material.

Introduction: The aim of this in vitro study was to evaluate the effect of MTA-based sealer (MTA Fillapex), eugenol-based sealer (Dorifill) and an epoxy resin sealer (AH Plus) on the bond strength of fiber posts cemented with a self-etch adhesive., Materials and Methods: The root canals of 72 maxillary incisors were prepared using the step-back technique after removing/cutting off the crowns. The samples were randomly divided to 4 groups (n=18). In group 1 (the controls) gutta-percha was used without sealer. In groups 2, 3 and 4, the canals were filled with gutta-percha using AH Plus, Dorifill and MTA Fillapex sealers, respectively, by cold lateral compaction technique. After post space preparation, the fiber posts were cemented in the root canals using self-etch adhesive. Then 1-mm-thick disks were prepared from the coronal thirds of all the root canals and subjected to a push-out test. Data were analyzed using the one-way ANOVA and post hoc Tukey's tests., Results: The maximum (4.45±0.09 MPa) and minimum (1.02±0.03 MPa) bond strength values were recorded in the control and Dorifill groups, respectively. The mean push-out bond strength values were similar for MTA Fillapex and AH Plus sealers (P>0.05). However these values were significantly higher than that of the Dorifill sealer (P<0.05)., Conclusion: Sealer type affected the bond strength of the fiber posts and MTA Fillapex decreased the dislodgment resistant of the fiber post.

Introduction: The aim of the present in vitro study was to compare the accuracy of cone-beam computed tomography (CBCT) in determining root canal morphology of mandibular first molars in comparison with staining and clearing technique., Methods and Materials: CBCT images were taken from 96 extracted human mandibular first molars and the teeth were then evaluated based on Vertucci's classification to determine the root canal morphology. Afterwards, access cavities were prepared and India ink was injected into the canals with an insulin syringe. The teeth were demineralized with 5% nitric acid. Finally, the cleared teeth were evaluated under a magnifying glass at 5× magnification to determine the root canal morphology. Data were analyzed using the SPSS software. The Fisher's exact test assessed the differences between the mesial and distal canals and the Cohen's kappa test was used to assess the level of agreement between the methods. Statistical significance was defined at 0.05., Results: The Kappa coefficient for agreement between the two methods evaluating canal types was 0.346 (95% CI: 0.247-0.445), which is considered a fair level of agreement based on classification of Koch and Landis. The agreement between CBCT and Vertucci's classification was 52.6% (95% CI: 45.54-59.66%), with a significantly higher agreement rate in the mesial canals (28.1%) compared to the distal canals (77.1%) (P<0.001)., Conclusion: Under the limitations of this study, clearing technique was more accurate than CBCT in providing accurate picture of the root canal anatomy of mandibular first molars.

Background: The aim of the present study was to compare the efficacy of Nd:YAG laser and K3 rotary system in the non-surgical root canal retreatment., Methods: Sixty-one rooted human teeth were divided into two groups of 30 after root canal therapy. In groups 1 and 2 root canal retreatments were carried out using K3 rotary system and Nd:YAG laser, respectively. CBCT images were used to evaluate canal transportation and root canal debridement at coronal, middle and apical canal cross-sections. In addition, the time necessary for canal debridement and the temperature on the external root surface were calculated. Mann-Whitney U test was used for statistical analysis of data with SPSS 17 after means ± SD were calculated. Shapiro-Wilk test was used to evaluate normal distribution of data. Statistical significance was defined at P<0.05., Results: The mean time necessary for the debridement of root canals in the laser group was significantly shorter than that in the K3 group (P=0.001). However, there were no significant differences in external root surface temperatures between the two groups (P=0.17). The coronal third of the canals was cleaner in the laser group significantly (p=0.01). Less canal transportation was seen in the laser group compared to the K3 group, significantly (P=0.04)., Conclusions: Nd:YAG is superior to K3 rotary system in the non-surgical root canal retreatment because it is faster and results in less canal transportation in the middle and apical thirds of the canals.

Purpose: To assess the potential of triblock copolymers based on poly(ethylene glycol) (PEG) and functionalized poly(ε-caprolactone) as temperature/pH responsive gels for controlled delivery of macromolecules., Methods: Poly(α-carboxylate-co-α-benzylcarboxylate-ε-caprolactone)-PEG-poly(α-carboxylate-co-α-benzylcarboxylate-ε-caprolactone) (PCBCL-PEG-PCBCL) was synthesized through ring opening polymerization of α-benzyl carboxylate-ε-caprolactone by PEG, followed by 30% debenzylation of the lateral blocks. The effect of Tris buffer and pH on the sol-gel transition temperature of PCBCL-PEG-PCBCL was assessed. The temperature/pH responsive release of tetramethylrhodamine-dextran (TMR-D) (10 and 40 kDa) from PCBCL-PEG-PCBCL was investigated., Results: Replacement of water with Tris buffer reduced PCBCL-PEG-PCBCL sol-gel transition temperature. Thermo-reversible hydrogels were only formed at pHs ≥ 5.0, but PCBCL-PEG-PCBCL transition temperature was not affected by pH above pH 5.0. In contrast to Pluronic F127 that released 100% of TMR-D within 2 h, PCBCL-PEG-PCBCL hydrogel controlled TMR-D release efficiently at pH = 7.4 and 37°C (~27 and 11% TMRD 10 and 40 kDa release within 150 h, respectively). At 50°C or pH = 9.0, TMR-D release was increased slightly, while at room temperature or pH = 5.0, no control over TMR-D release was observed by PCBCL-PEG-PCBCL hydrogel., Conclusion: PCBCL-PEG-PCBCL hydrogel provides depot release of macromolecules at physiological conditions. This release can be triggered through changes in the temperature or pH.

Aim: The aim of the present study was to compare the antibacterial properties of PLGA polymer containing ZnO, Ag and ZnO/Ag nanoparticles with those of 2.5% NaOCl in root canals contaminated with Enterococcus faecalis., Methods: A total of 100 maxillary central incisors were selected. After the crowns were removed to achieve 12-mm-length roots, the root canals were prepared with RaCe rotary system. The samples were sterilized; then 200 µL of E. faecalis suspension (ATCC 29212) was placed in each root canal. The samples were divided into 5 groups based on the antibacterial agent used. Group 1 served as the control group. In group 2 the polymer suspension containing ZnO nanoparticles, in group 3 the polymer solution containing ZnO/Ag nanoparticles, in group 4 the polymer solution containing Ag nanoparticles and in group 5, 2.5% NaOCl were used. After 24 hours, a solution was prepared using dentin chips from each root canal and placed on agar plates, followed by colony-forming units (CFU) count determination. Means ± standard deviations were calculated and Kruskal-Wallis test was used to evaluate the antibacterial properties of different irrigation agents. Mann-Whitney U test was used for two-by-two comparisons of the groups, with the level of significance being set at 0.05., Results: The maximum and minimum CFU counts were observed in the control and NaOCl groups, respectively. In the polymer groups, the maximum and minimum CFU counts were observed in the Ag and ZnO/Ag groups, respectively. There were significant differences in CFU counts between the study groups after application of irrigation solutions (P<0.05). Two-by-two comparisons of the groups using Mann-Whitney U test showed significant differences between all the study groups (P<0.05), except for groups ZnO and Ag (P=0.7)., Conclusion: Sodium hypochlorite solution was more effective than copolymer containing nanoparticles and of all the tested nanoparticles; ZnO/Ag nanoparticles exhibited the highest antibacterial activity.

Introduction: Flow rate (FR) and compressive strength (CS) are important properties of endodontic biomaterials that may be affected by various mixing methods. The aim of this experimental study was to evaluate the effect of different mixing methods on these properties of mineral trioxide aggregate (MTA) and calcium-enriched mixture (CEM) cement., Materials and Methods: Hand, amalgamator and ultrasonic techniques were used to mix both biomaterials. Then 0.5 mL of each mixture was placed on a glass slab to measure FR. The second glass slab (100 g) was placed on the samples and 180 sec after the initiation of mixing a 100-g force was applied on it for 10 min. After 10 min, the load was removed, and the minimum and maximum diameters of the sample disks were measured. To measure the CS, 6 sample of each group were placed in steel molds and were then stored in distilled water for 21 h and 21 days. Afterwards, the CS test was performed. Data were analyzed with multi-variant ANOVA and post hoc Tukey tests. The level of significance was set at 0.05., Results: There were significant differences in FR of MTA and CEM cement with different mixing techniques (P<0.05). In the MTA group, none of the mixing techniques exhibited a significant effect on CS (P>0.05); however, in CEM group the CS at 21-h and 21-day intervals was higher with the hand technique (P<0.05)., Conclusion: Mixing methods affected the flowability of both biomaterials and compressive strength of CEM cement.

Introduction: The aim of the present study was to evaluate the effect of different mixing techniques (conventional, amalgamator and ultrasonic mixing) on the physical properties the working time (WT), setting time (ST), dimensional changes (DC) and film thickness (FT)] of calcium-enriched mixture (CEM) cement and mineral trioxide aggregate (MTA)., Methods and Materials: The mentioned physical properties were determined using the ISO 6786:2001 specification. Six samples of each material were prepared for three mixing techniques (totally 36 samples). Data were analyzed using descriptive statistics, two-way ANOVA and Post Hoc Tukey's tests. The level of significance was defined at 0.05., Results: Irrespective of mixing technique, there was no significant difference between the WT and FT of the tested materials. Except for the DC of MTA and the FT of the all materials, other properties were significantly affected with mixing techniques (P<0.05). The ultrasonic technique decreased the ST of MTA and CEM cement and increased the WT of CEM cement (P<0.05)., Conclusion: The mixing technique of the materials had no significant effect on the dimensional changes of MTA and the film thickness of both materials.

Introduction: The aim of the present study was to evaluate whether adding 2% chlorhexidine (CHX) and 2.6% sodium hypochlorite (NaOCl) to calcium-enriched mixture (CEM) cement would affect its setting time (ST), or not., Methods and Materials: In this study, the setting time of CEM cement was evaluated in three groups (n=9) as follows: group 1; CEM cement, group 2; CEM cement+2% CHX and group 3; CEM cement+2.6% NaOCl. Then the mean values of ST were calculated and the Kolmogorov-Smirnov test was used to evaluate the normal distribution of data. The Kruskal-Wallis and Mann-Whitney U tests were used for statistical analysis. Statistical significance was set at 0.05., Results: The mean ST for groups 1, 2 and 3 were 105, 120 and 220 min, respectively. There was a significant increase in the duration of ST in group 3 (NaOCl) in comparison with the two other groups (P<0.05)., Conclusion: NaOCl significantly increased the ST of CEM cement, whereas chlorhexidine did not alter the ST.

Aim: The aim of the present study was to compare the apical microleakage of root canals obturated with AH Plus and MTA Fillapex® sealers., Methods: A total of 76 anterior teeth were randomly divided into 4 groups (N.=15) and in each group 2 positive and 2 negative control teeth. All the teeth were cleaned, shaped and obturated using lateral compaction technique with gutta-percha and AH Plus® sealer in groups 1 and 2 and MTA Fillapex® sealer in groups 3 and 4. In groups 1 and 3 post spaces were prepared immediately and groups 2 and 4 were stored for 1 week at 100% relative humidity and then post spaces were prepared. The teeth were connected to a bacterial microleakage evaluation system. Each 72 hours, E. faecalis bacteria were inoculated into the coronal thirds of the root canals. The study period was 90 days and the samples were evaluated on a daily basis, Meier-Kaplan technique was used for estimation of the mean time of microleakage to occur. Log-rank test was used for comparison of microleakage., Results: Contamination occurred in positive control groups on the first day of bacterial inoculation; however, the negative control groups did not exhibit any contamination up to the end of 90-day study period. the difference was not statistically significant between two materials (P>0.05)., Conclusion: There were no significant differences between AH Plus® and MTA Fillapex® sealers with the two immediate and delayed postspace preparation techniques.

Background: Cyclo-oxygenase-2 (COX-2) is an early response gene that is induced by growth factors, oncogenes and carcinogens and its expression is increased in various tumors. Increased expression of COX-2 plays a significant role in the development and growth of tumors by interfering in biological processes such as cell division, cellular immunity, cell adhesion, apoptosis, and angiogenesis. This study aimed to investigate the immunohistochemical expression of COX-2 in keratocystic odontogenic tumor (KOT) in comparison with ameloblastoma and dentigerous cyst with regards to different clinical behavior and histopathological features of these lesions., Materials and Methods: Paraffined blocks of 45 cases including 15 cases of dentigerous cyst, 15 cases of KOT and 15 cases of ameloblastoma were stained with immunohistochemical method for COX-2. Five high-power fields of each sample were evaluated to determine the percentage of stained cells and the intensity of staining. Degree of immunoreactivity was obtained from the sum of two. Statistical evaluation was performed by the Kruskal-Wallis and ANOVA Mann-Whitney test (P < 0.05)., Results: Overexpression of COX-2 in ameloblastoma and KOT was observed compared with dentigerous cyst (P < 0.001). However, no significant difference was observed between the expression of COX-2 in ameloblastoma and KOT (P = 0.148)., Conclusion: The COX-2 expression in odontogenic tumors such as ameloblastoma and cystic neoplasm with aggressive behavior such as KOT increases. However, it does not seem that COX-2 affects the development and growth of cysts with noninvasive behavior like dentigerous cyst.

Background: Recurrent pregnancy loss (RPL) has been defined as two or more miscarriages before 20(th) week of gestation. It seems that IL-27 may reduce inflammatory responses and affect the survival of the embryo during human pregnancy. IL-27 polymorphisms may influence RPL by altering the levels or the activity of gene product., Objective: We studied for the first time the association of IL-27 -964 A>G single nucleotide polymorphism (SNP) with RPL in Iranian women., Materials and Methods: A case-controlled study was performed on two groups consisting of 150 healthy women with at least one delivery (control group) and 150 women with two or more primary RPLs history (RPL group). The -964 A>G SNP in IL-27 gene was determined by PCR-RFLP technique. Genotype and allele frequencies were compared using (2) tests between two groups., Results: There was no difference between the two groups regarding age of women (29±4.4 [control] vs. 30.84±5.2 years [case]). In the RPL group, the genotype frequencies of -964 A>G polymorphism were AG (49.3%), AA (40%), and GG (10.7%), and in the control group, they were AG (43.3%), AA (48.7%), and GG (8%). There was no significant difference between the genotypes of AA, AG, and GG in two groups (p=0.23). As the frequency of allele A was 64.7% in the RPL group and 70.3% in the control group, the difference in frequency of allele A in -964 A>G between two groups was not significant (p=0.19)., Conclusion: Our findings indicate that SNP of -964 A>G in IL-27 gene is not a risk factor for RPL in Iranian women.

The main aim of this study was to evaluate the uptake of E6 mRNA antisense into cervical cancer cells, induced by human papilloma virus (HPV). In this study, the carrier of the antisense was tri-calcium phosphate nanoparticles (TCP NPs) conjugated with dioleoyl phosphatidyl ethanolamine (DOPE) and/or anti-E6 antibody. At first, TCP NPs were synthesized, coated with carboxy-polyethylene glycol, and then conjugated with anti-E6 antibody and/or DOPE by carbodiimide cross-linker. Then, a single stranded DNA, which was complementary (antisense) of E6 mRNA, was attached to each one. Finally, the uptake of conjugated and unconjugated TCP NPs into HelaS3 cells was separately evaluated by Fourier transform infrared spectroscopy, optical microscopy, and fluorescent microscopy. Also, the cytotoxicity of these carriers was measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay. Overall, 4 types of TCP NPs were used in this study, including 1) TCP NPs conjugated with DOPE (TCP NPs/DOPE), 2) TCP NPs conjugated with DOPE and antibody (TCP NPs/DOPE/Anti-E6 Ab), 3) TCP NPs conjugated with antibody (TCP NPs/Anti-E6 Ab), and 4) TCP NPs which not conjugated with DOPE and antibody (unconjugated TCP NPs). Uptake tests showed that although all types of TCP NPs could transfer antisense of E6 mRNA into HelaS3 cells, TCP NPs/DOPE and TCP NPs/DOPE/Anti-E6 Ab had more uptake than TCP NPs/Anti-E6 Ab and unconjugated TCP NPs. Moreover, MTT assay showed that TCP NPs/DOPE was more toxic than TCP NPs/DOPE/Anti-E6 Ab, TCP NPs/Anti-E6 Ab, and unconjugated TCP NPs. It can be concluded that TCP NPs/DOPE/Anti-E6 Ab is a good choice for oligonucleotide delivery, because of higher uptake and less toxicity, compared with other formulations., (Copyright © 2014 Elsevier B.V.
