Abstract

In view of the important roles of reactive oxygen species (ROS) and reactive sulfur species (RSS) in the complex signal transduction and oxidation pathways, fluorescent probes that are able to display distinct signals to hypochlorous acid and SO 2 derivatives are highly valuable. Herein, a novel dual-functional probe (DFP) as an efficient single fluorescent-molecule which can respond to HClO and HSO 3 − with two different sets of fluorescence signals was presented. The DFP displayed desired properties such as high specificity, suitable sensitivity, appreciable water solubility and stability. The sensing mechanism was confirmed by high-resolution mass spectroscopy analysis and 1 H NMR spectrometry analysis. Moreover, as a biocompatible molecule, the DFP has been successfully applied for the detection of HClO in living cells with a dual-channel mode. Therefore, the present work established a novel strategy for monitoring the multiple ROS and RSS species using a single fluorescent probe and the DFP is promising as a molecular tool to investigate the production and dynamics of HClO and HSO 3 − in the complex interaction networks of the living system.

Molecular targeting agent, sorafenib, is the main therapeutic option for antitumor therapy of advanced HCC, but it is currently too expensive for the public and its therapeutic effect does not satisfy initial expectation. Therefore, it is important to develop more effective molecular targeted therapeutic strategies for advanced HCC. Materials and methods: The antitumor effects of sorafenib or ARQ-197, an antagonist of c-MET (tyrosine-protein kinase Met or hepatocyte growth factor receptor), were examined by MTT or in murine tumor model. The effect of ARQ-197 on epithelial–mesenchymal transition (EMT) or multidrug resistance (MDR) was examined by quantitative real-time PCR for the expression of related genes. The clearance of sorafenib in HCC cells was detected by liquid chromatography-mass spectrometry/mass spectrometry. Results: ARQ-197 treatment enhanced the sensitivity of HCC cells to sorafenib. Mechanistic studies indicated that ARQ-197 inhibited the expression of EMT- and MDR-related genes. Moreover, ARQ-197 treatment decelerated the clearance of sorafenib in cultured HCC cells and subcutaneous HCC tumors in nude mice. Conclusion: In the present work, our data suggested that ARQ-197 decelerated the clearance of sorafenib in HCC cells and enhanced the antitumor effect of sorafenib. 225. Novel nanoparticle CS-C60-Fe3O4magnetically induces tissue-specific aggregation and enhances thermal ablation of hepatocellular carcinoma

Abstract

Metallofullerenes are an important type of metallic nanomaterial with promising applications in several medical fields. Thermal ablation, including radiofrequency ablation (RFA) and microwave ablation (MWA), is an important treatment strategy for advanced hepatocellular carcinoma (HCC). The thermal expansion of fullerenes makes them good adjuncts to thermal ablation treatment of HCC. In this study, we used an innovative method of emulsification and cross-linking to produce CS-C60-Fe3O4(Chitosan-C60-Fe3O4) nanoparticles, which have the advantages of uniform particle size and high bioavailability, as a kind of novel nano-pharmaceutical. The CS-C60-Fe3O4nanoparticles were prepared by the cross-linking reaction from chitosan–acetic acid solution, Fe3O4nanoparticles by Fe2SO4·7H2O and FeCl3·6H2O, and C60. The average particle size of CS-C60-Fe3O4was 194.3 nm. Because CS-C60-Fe3O4is magnetic, it can achieve specific and tissue aggregation in HCC tumor tissues. Moreover, compared with normal soluble C60(EL35-C60), CS-C60-Fe3O4prolonged the retention time of C60in the blood of mice. CS-C60-Fe3O4alone is not cytotoxic to cultured cells or tumor tissues, but when combined with thermal ablation strategies (RFA and MWA), it significantly upregulates the antitumor effects of thermal ablation on HCC tissues, that is, it acts as a sensitiser to thermal ablation. In the presence of thermal ablation, CS-C60-Fe3O4interfered with iron metabolism in HCC cells and induced ferroptosis of HCC cells in the tumor tissues. These results not only expand our understanding of metallofullerenes but also provide additional options for the treatment of advanced HCC.

Abstract

Background: The purpose of this study was to explore the efficacy and safety of high intensity focused ultrasound (HIFU) in gastric cancer with liver metastasis (GCLM) patients who were contraindicated for either hepatectomy or radiofrequency ablation (RFA).Methods: This is a prospective, observational study on GCLM patients with 1-3 liver metastases. The primary gastric lesions were thoroughly resected and any case that exhibited extra-hepatic metastasis was excluded. A 1:2:2 propensity score-matching analysis was performed using a logistic regression model on the HIFU group, best supportive care (BSC) group, and palliative chemotherapy (PC) group. The primary endpoints include progression-free survival (PFS) and overall survival (OS).Results: Forty patients were finally included, there were 8 cases in HIFU group, 16 cases in BSC group, and 16 cases in PC group. The median follow-up time for the entire cohort was 10 months. The median PFS was 16.5 months in HIFU group, 2 months in BSC group, and 5 months in PC group. The median OS was 27.5 months in the HIFU group, 7 months in the BSC group, and 11.5 months in the PC group. Additionally, no grade 3 or higher adverse events occurred in the HIFU group.Conclusion: The results of this study showed that HIFU treatment could improve the long-term prognosis of GCLM patients without a significant increase in the occurrence of adverse events. Compared with PC and BSC, HIFU is the preferred treatment option when GCLM patients without extra-hepatic metastasis are unable to undergo either surgery or RFA. [ABSTRACT FROM AUTHOR]

Abstract

ZLDI-8 is an A disintegrin and metalloproteinase domain 17 (ADAM17) inhibitor that suppresses the shedding of Notch1 to the Notch1 intracellular domain (NICD). In previous studies, we found that ZLDI-8 was able to sensitize HCC to sorafenib, but the mechanism of action remains unclear. The sensitizing effects of ZLDI-8 were tested both in vitro and in vivo. EMT-related factors, sorafenib sensitivity-related proteins and ECM-related gene expression were assessed using immunohistochemistry, RT PCR and Western blotting. Knockdown assays were conducted to determine the relationship between the Notch and Integrin pathways. CoIP assays, nuclear and cytoplasmic fractionation and immunofluorescence colocalization were applied to explore the interaction between the Notch and Integrin pathways. Appropriate statistical analysis methods were used to assess the significance of the experimental results and to ensure the scientific validity and reliability of the experimental design. We found that ECM- and EMT-related proteins were downregulated after ZLDI-8 treatment (P <0.05). ZLDI-8 significantly downregulated Integrinβ1 and Integrinβ3 in HCC in vitro and in vivo (P <0.05), possibly through Foxc2-dependent regulation. Mechanistically, interfering with the expression of both Integrin-linked kinase (ILK) and the NICD may downregulate the expression of proteins targeted by sorafenib, thereby sensitizing cells to sorafenib. The retroregulation of Integrinβ by ILK may occur through the interaction between the NICD and ILK and may be the result of the translocation of the complexus. Our study indicates that blocking the Notch pathway may affect Integrinβ through crosstalk between the Notch1 and Integrinβ/ILK signaling pathways, thus providing a potential therapeutic strategy for HCC. Abstract

Open cluster members are coeval and share the same initial bulk chemical compositions. Consequently, differences in surface abundances between members of a cluster that are at different evolutionary stages can be used to study the effects of mixing and internal chemical processing. We carry out an abundance analysis of seven elements (Li, O, Na, Mg, Al, Si, Fe) in 66 stars belonging to the open cluster M67, based on high resolution GALAH spectra, 1D MARCS model atmospheres, and, for the first time for a large sample of M67 stars, non-local thermodynamic equilibrium (non-LTE) radiative transfer. From the non-LTE analysis, we find a typical star-to-star scatter in the abundance ratios of around 0.05 dex; this scatter is slightly but systematically larger when LTE is assumed instead. We find trends in the abundance ratios with effective temperature, indicating systematic differences in the surface abundances between turn-off and giant stars; these trends are more pronounced when LTE is assumed. However, in the non-LTE analysis, most of the element trends have been flattened. Two species are exceptions to this behaviour, namely Al and Si, which both clearly display remaining trends in the non-LTE analysis.
