Release of individuals is an effective conservation approach to protect endangered species. To save this small isolated giant panda population in Liziping Nature Reserve, a few giant pandas have been released to this population. Here we assess genetic diversity and future changes in the population using noninvasive genetic sampling after releasing giant pandas. In this study, a total of 28 giant pandas (including 4 released individuals) were identified in the Liziping, China. Compared with other giant panda populations, this population has medium-level genetic diversity; however, a Bayesian-coalescent method clearly detected, quantified, and dated a recent decrease in population size. The predictions for genetic diversity and survival of the population in the next 100 years indicate that this population has a high risk of extinction. We show that released giant pandas can preserve genetic diversity and improve the probability of survival in this small isolated giant panda population. To promote the recovery of this population, we suggest that panda release should be continued and this population will need to release 10 males and 20 females in the future. [ABSTRACT FROM AUTHOR]

Climate change is one of the most pervasive threats to biodiversity globally, yet the influence of climate relative to other drivers of species depletion and range contraction remain difficult to disentangle. Here, we examine climatic and non-climatic correlates of giant panda (Ailuropoda melanoleuca) distribution using a large-scale 30 year dataset to evaluate whether a changing climate has already influenced panda distribution. We document several climatic patterns, including increasing temperatures, and alterations to seasonal temperature and precipitation. We found that while climatic factors were the most influential predictors of panda distribution, their importance diminished over time, while landscape variables have become relatively more influential. We conclude that the panda’s distribution has been influenced by changing climate, but conservation intervention to manage habitat is working to increasingly offset these negative consequences. Giant pandas (Ailuropoda melanoleuca) were historically hunted using dogs and are currently threatened by free-roaming dogs and their associated diseases. To better understand the spatial magnitude of this threat, we used a GIS approach to investigate edge effects of dogs on giant panda habitat. We first examined two nature reserves with contrasting free-roaming dog populations: Liziping, with many dogs (~0.44/km2), and Daxiangling, with few dogs (~0.14/km2). Spatial analysis indicated that giant pandas at Liziping (but not Daxiangling) showed a shift in habitat use away from populated areas consistent with a risk response to the foray distance of free-roaming dogs (10.9 km path-distance). Most giant panda locations (86%) from the 2014 census in Liziping were clustered around remote "dog-free zones." Expanding this analysis across the entire giant panda range revealed that 40% of panda habitat is within the foray distance of dogs. Our assessment will inform dog control programs including monitoring, education, veterinary care, and other measures. We recommend that reserves designated for the release of translocated pandas receive priority consideration for dog control efforts. Only by understanding and managing complex interactions between humans, domestic animals, and wild animals can we sustain natural systems in a world increasingly dominated by humans. Sensitivity to bitter tastes provides animals with an important means of interacting with their environment and thus, influences their dietary preferences. Genetic variants encoding functionally distinct receptor types contribute to variation in bitter taste sensitivity. Our previous study showed that two nonsynonymous sites, A52V and Q296H, in the TAS2R20 gene are directionally selected in giant pandas from the Qinling Mountains, which are speculated to be the causative base‐pair changes of Qinling pandas for the higher preference for bamboo leaves in comparison with other pandas. Here, we used functional expression in engineered cells to identify agonists of pTAS2R20 (i.e., giant panda's TAS2R20) and interrogated the differences in perception in the in vitro responses of pTAS2R20 variants to the agonists. Our results show that pTAS2R20 is specifically activated by quercitrin and that pTAS2R20 variants exhibit differences in the sensitivity of their response to the agonist. Compared with pTAS2R20 in pandas from other areas, the receptor variant with A52V and Q296H, which is most commonly found in Qinling pandas, confers a significantly decreased sensitivity to quercitrin. We subsequently quantified the quercitrin content of the leaves of bamboo distributed in the Qinling Mountains, which was found to be significantly higher than that of the leaves of bamboo from panda habitats in other areas. Our results suggest that the decreased sensitivity to quercitrin in Qinling pandas results in higher‐quercitrin‐containing bamboo leaves to be tasting less bitter to them and thus, influences their dietary preference. This study illustrates the genetic adaptation of Qinling pandas to their environments and provides a fine example of the functional effects of directional selection in the giant panda. Paradoxically, despite the growth in protected areas globally, many species remain threatened and continue to decline. Attempts to conserve species in suboptimal habitats (i.e., as refugee species) may in part explain this Protected Area Paradox. Refugee species yield poor conservation outcomes as they suffer lower densities and fitness. We suggest that the giant panda may serve as an iconic example, reflecting the contraction and shift in the giant panda's range, diet and habitat use over the past 3,500 years, coinciding with increasing human pressure, and now maintained by conservation efforts, this due to shifted baselines. The global bias of protected area location to less productive habitats indicates that this problem may be widespread. We urgently need efforts to identify victims of refugee species status to allow improved conservation management globally, reducing the paradoxical outcomes of our conservation efforts. Sperm cryopreservation is an essential approach for assisted reproduction and genetic resources conservation in captive giant pandas. Cryopreservation, however, leads to a significant decrease in sperm quality and, consequently, a low fertilization rate. Therefore, it is mandatory to disclose more suitable and efficient freezing strategies for sperm cryopreservation. In the present study, we compared for the first time the performance of two commercial freeze extender (INRA96 versus TEST) freezing methods on post-thawed semen quality. Semen cryopreserved with the INRA96 showed better total motility (73.00 ± 4.84% vs 57.56 ± 3.60%, P < 0.001), membrane integrity (60.92 ± 2.27% vs 40.53 ± 2.97%, P < 0.001) and acrosome integrity (90.39 ± 2.74% vs 84.26 ± 4.27%, P < 0.05) than stored with TEST. There was no significant difference in DNA integrity after thawing between the two extenders (95.69 ± 3.60% vs 94.26 ± 4.84%). Although several mammals impregnate their fur with environmental odors, a phenomenon termed scent anointing or rubbing, the functional relevance of this behavior often is unclear. One theory is that scent anointing could be a form of scent matching with environmental odors to signal competitiveness and home range occupation. In this study we presented giant pandas with a range of odors to determine whether scent matching could provide a functional explanation for scent anointing in this species. We found that only a musk-based perfume elicited significantly more scent-anointing and scent-marking behavior than control. Males were also significantly more likely to scent-anoint and scent-mark than females. A preference for anointing, but not scent marking, when presented with peppermint (an insecticide) also was revealed. Our results suggest that giant pandas differentially scent-anoint with foreign odors to signal home range occupation, and possibly to repel ectoparasites. We also highlight how chemical signaling of resource-holding potential is likely to play an important role in determining competitive interactions between adult male giant pandas. As a highly endangered species, the giant panda (panda) has attracted significant attention in the past decades. Considerable efforts have been put on panda conservation and reproduction, offering the promising outcome of maintaining the population size of pandas. To evaluate the effectiveness of conservation and management strategies, recognizing individual pandas is critical. However, it remains a challenging task because the existing methods, such as traditional tracking method, discrimination method based on footprint identification, and molecular biology method, are invasive, inaccurate, expensive, or challenging to perform. The advances of imaging technologies have led to the wide applications of digital images and videos in panda conservation and management, which makes it possible for individual panda recognition in a noninvasive manner by using image‐based panda face recognition method.In recent years, deep learning has achieved great success in the field of computer vision and pattern recognition. For panda face recognition, a fully automatic deep learning algorithm which consists of a sequence of deep neural networks (DNNs) used for panda face detection, segmentation, alignment, and identity prediction is developed in this study. To develop and evaluate the algorithm, the largest panda image dataset containing 6,441 images from 218 different pandas, which is 39.78% of captive pandas in the world, is established.The algorithm achieved 96.27% accuracy in panda recognition and 100% accuracy in detection.This study shows that panda faces can be used for panda recognition. It enables the use of the cameras installed in their habitat for monitoring their population and behavior. The giant panda is one of the most endangered mammals in the world, and many studies have revealed their evolutionary adaptation to the local environment (e.g., dietary cellulose and cyanide) on the evidences from population genetics and their gut microbiome. Here, based on the results of our analysis of the giant panda gut microbiome, we concluded that instability and resilience are the two primary characteristics of the giant panda gut microbiome. The giant panda (Ailuropoda melanoleuca) is one of the world's most endangered mammals and remains threatened as a result of intense environmental and anthropogenic pressure. The transformation and specialization of the giant panda's diet into a herbivorous diet have resulted in unique adaptabilities in many aspects of their biology, physiology and behavior. However, little is known about their adaptability at the molecular level. Through comparative analysis of the giant panda's genome with those of nine other mammalian species, we found some genetic characteristics of the giant panda that can be associated with adaptive changes for effective digestion of plant material. We also found that giant pandas have similar genetic characteristics to carnivores in terms of olfactory perception but have similar genetic characteristics to herbivores in terms of immunity and hydrolytic enzyme activity. Through the analysis of gene family expansion, 3752 gene families were found, which were enriched in functions such as digestion. A total of 93 genes under positive selection were screened out and gene enrichment identified these genes for the following processes: negative regulation of cellular metabolic process, negative regulation of nitrogen compound metabolic process, negative regulation of macromolecule metabolic process and negative regulation of metabolic process. Combined with the KEGG pathway, it was found that genes such as CREB3L1, CYP450 2S1, HSD11B2, LRPAP1 play a key role in digestion. Mammalian neonates are born at a wide range of maturity levels. Altricial newborns are born with limited sensory agency and require extensive parental care. In contrast, precocial neonates are relatively mature physically and often capable of independent function shortly after birth. In extant mammals, placental newborns vary from altricial to precocial, while marsupials and monotremes are all extremely altricial at birth. Bears (family Ursidae) have one of the lowest neonatal−maternal mass ratios in placental mammals, and are thought to also have the most altricial placental newborns. In particular, giant pandas (Ailuropoda melanoleuca) are thought to be exceptionally altricial at birth, and possibly marsupial‐like. Here we used micro‐computer (micro‐computed) tomography scanning to visualize the skeletal anatomy of ursid neonates and compare their skeletal maturity with the neonates of other caniform outgroups. Specifically, we asked whether ursid neonates have exceptionally altricial skeletons at birth compared with other caniform neonates. We found that most bear neonates are similar to outgroup neonates in levels of skeletal ossification, with little variation in degree of ossification between ursine bears neonates (i.e. bears of the subfamily Ursinae). Perinatal giant pandas, however, have skeletal maturity levels most similar to a 42−45‐day‐old beagle fetus (~70% of total beagle gestation period). No bear exhibits the skeletal heterochronies seen in marsupial development. With regards to skeletal development, ursine bears are not exceptionally altricial relative to other caniform outgroups, but characterized largely by the drastic difference between newborn and adult body sizes. A review on the existing hypotheses for ursids' unique reproductive strategy suggests that the extremely small neonatal−maternal mass ratio of ursids may be related to the recent evolution of large adult body size, while life history characteristics retained an ancestral condition. A relatively short post‐implantation gestation time may be the proximal mechanism behind the giant panda neonates' small size relative to maternal size and altricial skeletal development at birth. The potential threat of domestic dogs to wildlife habitat in China is not widely recognized, despite their large population, lack of regulations regarding their control, and threat they pose to native species. In a case study in 2017, we surveyed villages surrounding Liziping Nature Reserve, the primary site for the release of captive‐born giant pandas (Ailuropoda melanoleuca) into the wild. We conducted surveys of dog owners to assess the population size, demographics, free‐roaming status, and vaccination and sterilization history of their dogs. We collected blood and fecal samples to assess the prevalence of viral and parasite disease threats. At least 370 owned dogs lived near the core giant panda habitat; 64% were free‐roaming, 21% had positive antibody titers for ≥1 of the 4 viruses we tested (canine distemper, parvovirus, rotavirus, rabies), and 67% were positive for gastrointestinal parasites. The high proportion of free‐roaming dogs, uninhibited access to the reserve, and high prevalence of infectious diseases indicate that dogs pose a serious threat to wildlife within Liziping. Flagship species have been used widely as umbrella species (i.e., species with large home range whose protection often provides protection for sympatric species) in the management of China's nature reserves. This conflation of flagship and umbrella species is best represented by the giant panda (Ailuropoda melanoleuca) and other large, endangered mammals designated as conservation targets in site selection and planning of reserves. Few empirical studies have tested the effectiveness of flagship species as surrogates for a broader range of sympatric species. Using extensive camera‐trap data, we examined the effectiveness of management zones designated to protect flagship (target) species in conserving sympatric species in 4 wildlife reserves (Gutianshan, Changqing, Laohegou, and Wolong). We tested whether the progression from peripheral to core zones was associated with an increasing habitat association for both target and sympatric species. The distribution patterns of the study species across the zones in each reserve indicated a disparity between management zones and the species' habitat requirements. Management zone was included in the final model for all target species, and most of them had higher occurrence in core zones relative to less‐protected zones, but zone was not a predictor for most of the sympatric species. When management zone was associated with the occurrence of sympatric species, threatened species generally had higher detections in core zones, whereas common species had higher detections outside of the core zone. Our results suggested that reserve planning based on flagship species does not adequately protect sympatric species due to their specialized habitat requirements. We recommend re‐examining the effectiveness of management zoning and urge a multispecies and reserve‐wide monitoring plan to improve protection of China's wildlife. Reintroduction is a key approach in the conservation of endangered species. In recent decades, many reintroduction projects have been conducted for conservation purposes, but the rate of success has been low. Given the important role of gut microbiota in health and diseases, we questioned whether gut microbiota would play a crucial role in giant panda's wild‐training process. The wild procedure is when captive‐born babies live with their mothers in a wilderness enclosure and learn wilderness survival skills from their mothers. During the wild‐training process, the baby pandas undergo wilderness survival tests and regular physical examinations. Based on their performance through these tests, the top subjects (age 2–3 years old) are released into the wild while the others are translocated to captivity. After release, we tracked one released panda (Zhangxiang) and collected its fecal samples for 5 months (January 16, 2013 to March 29 2014). Here, we analyzed the Illumina HiSeq sequencing data (V4 region of 16S rRNA gene) from captive pandas (n = 24), wild‐training baby pandas (n = 8) of which 6 were released and 2 were unreleased, wild‐training mother pandas (n = 8), one released panda (Zhangxiang), and wild giant pandas (n = 18). Our results showed that the gut microbiota of wild‐training pandas is significantly different from that of wild pandas but similar to that of captive ones. The gut microbiota of the released panda Zhangxiang gradually changed to become similar to those of wild pandas after release. In addition, we identified several bacteria that were enriched in the released baby pandas before release, compared with the unreleased baby pandas. These bacteria include several known gut‐health related beneficial taxa such as Roseburia, Coprococcus, Sutterella, Dorea, and Ruminococcus. Objectives: Accessory digits have evolved independently within several mammalian lineages. Most notable among these is the pseudothumb of the giant panda, which has long been considered one of the most extraordinary examples of contingent evolution. To date, no primate has been documented to possess such an adaptation. Here, we investigate the presence of this structure within the aye‐aye (Daubentonia madagascariensis), a species renowned for several other specialized morphological adaptations in the hand, including a morphologically unique third digit. Materials and Methods: We combine physical dissection techniques with digital imaging processes across a sample of seven individuals (six adults and one immature individual) to describe and visualize the anatomy of the wrist and hand within the aye‐aye. Results: A distinct pseudothumb, which consists of both a bony component (an expanded radial sesamoid) and a dense cartilaginous extension (the "prepollex") was observed in all specimens. We demonstrate that this pseudodigit receives muscular attachments from three muscles, which collectively have the potential to enable abduction, adduction, and opposition. Finally, we demonstrate that the pseudothumb possesses its own distinct pad within the palm, complete with independent dermatoglyphs. Discussion: Pseudothumbs have been suggested to improve palmar dexterity in taxa with overly ‐generalized first digits (e.g., pandas) and to widen the hand for digging (e.g., some fossorial moles), but the aye‐aye's pseudothumb represents what we believe is a heretofore unrecognized third functional role: its accessory digit compensates for overspecialization of its fingers for non‐gripping functions (in this case, the aye‐aye's unique "tap foraging" practices). We report influenza A(H1N1)pdm09 virus infection in a captive giant panda in Hong Kong. The viral load peaked on day 1 and became undetectable on day 5, and an antibody response developed. Genome analysis showed 99.3%-99.9% nucleotide identity between the virus and influenza A(H1N1)pdm09 virus circulating in Hong Kong. [ABSTRACT FROM AUTHOR]

Documenting the different social and behavioural contexts that vocalisations are produced in remains an important step towards understanding the functional relevance of specific call types in a given species' vocal repertoire. In this study we investigated whether seasonal differences and the presence or absence of male and female conspecifics influence the production of male giant panda vocal signals. To this end, captive male giant pandas were observed during and outside of the breeding season in three social contexts: only male conspecific neighbours, only female conspecific neighbours, and a context with no neighbours. We found that males were more likely to bleat, chirp, honk and moan during the breeding season, and showed a tendency to growl more outside of the reproductive period. The contextual analysis revealed that bleats were more likely to be produced by males when opposite-sexed conspecifics are in close attendance during the breeding season. Conversely, males were more likely to chirp when neighboured by males than females or no neighbours. In addition, males were more likely to honk in the absence of neighbouring conspecifics during the breeding season, raising the possibility that these calls function to signal location and gain the attention of potential mates. Moans were produced more often when male giant pandas had male than female neighbours during the breeding season, which may reflect mild aggression towards these same-sexed rivals, whereas the production of barks and growls did not vary according to season or the sex of conspecific neighbours. Our findings underscore the importance of male giant panda bleats for coordinating reproduction and promoting contact with potential mating partners in this non-gregarious species, and yield fresh insights into the function of male honks that warrant further investigation. Bears are a group of carnivores with diverse diets and complicated dental structure. Several large rearrangements of dental structures are known in different lineages of bears, making the homology of dental structures between the different bears difficult to evaluate. By tracing the evolutionary processes of the two lineages of bears with the most complicated dental structure, i.e., the giant panda lineage (Ailuropodinae) and cave bear lineage (Ursinae), we were able to clarify the homology of dental structures of the two subfamilies. We define a new assemblage of dental nomenclature (based mainly on the homology to the giant panda) that can be very useful to infer the evolution of fossil bears. Medicinal herb collection has historical and cultural roots in many rural communities in developing countries. Areas where herb collection occurs may overlap with biodiversity hotspots and crucial habitat of endangered and threatened species. However, impacts of such practices on wildlife are unknown and possibly underestimated, perhaps due to the elusive nature of such activities. We examined this phenomenon in Wolong Nature Reserve, China, a protected area in the South‐Central China biodiversity hotspot that also supports a community of Tibetan, Qiang and Han people who use herb collection as a supplementary source of livelihood. We adopted a participatory approach in which we engaged local people in outlining spatial and temporal dynamics of medicinal herb collection practices. We found that the overall spatial extent of herb collection increased in the past two decades. We then overlaid herb collection maps with localities of giant panda (Ailuropoda melanoleuca) feces collected over two time points in the reserve. Using a Bayesian parameter estimation, we found evidence for declined giant panda occurrence in the areas most recently impacted by emerging medicinal herb collection. Our methodology demonstrates the potential power of integrating participatory approaches with quantitative methods for processes like herb collection that may be difficult to examine empirically. We discuss future directions for improving explanatory power and addressing uncertainty in this type of mixed‐method, interdisciplinary research. Signal detection theory predicts that animals should select scent‐marking sites in a way that maximizes their probability of detection by target receivers. Many studies have been conducted with a focus on signaling behavior and function. Yet, the role of the environment in structuring scent‐mark patterns is poorly known. We studied the giant panda, a solitary species that relies heavily on chemosignaling for communication through depositing urine and anogential gland secretions (AGS) on trees, to determine how environmental factors influence scent deposition strategies. Using a combination of sign surveys, camera trapping, and gas chromatography mass spectrometry analysis, we conducted a comparative study of giant panda scent marking in two reserves, Wanglang and Foping, in China. The two reserves differed with regard to prevalence of tree species that differed in bark roughness, diameter at breast height (DBH), and moss status. For urine‐marking pandas selected large, mossy trees with very rough bark trees, whereas AGS marking was used preferentially on rough‐barked moss‐free trees in both reserves. Both AGS and urine contained more volatile than nonvolatile constituents and urine had higher relative abundance of volatiles than AGS, indicating urine likely has a larger signal range and shorter signal persistence than AGS. Urine marking was significantly more prevalent in Wanglang, which contains more rough‐barked, moss covered, and higher DBH trees favored for urine marking, than in Foping. Thus, habitat features appear to constrain pandas' use of different types of marking, potentially altering how the communication system functions in different ecological contexts. For the time being, there are only about 2500 giant pandas in the world. Therefore, we still have to do much more efforts to protect the giant pandas. In captive wildlife, the cataract incidence of mammalian always increases with age. Currently, in China, the proportion of elderly giant pandas who suffering from cataract has reached 20%. The eye disorder thus has a strong influence on the physical health and life quality of the elderly giant pandas. To discover the genes associated with the pathogenesis of cataract in the elderly giant panda and achieve the goal of early assessment and diagnosis of cataract in giant pandas during aging, we performed whole genome methylation sequencing in 3 giant pandas with cataract and 3 healthy giant pandas using methylation-dependent restriction-site associated DNA sequencing (MethylRAD). In the present study, we obtained 3.62M reads, on average, for each sample, and identified 116 and 242 differentially methylated genes (DMGs) between the two groups under the context of CCGG and CCWGG on genome, respectively. Further KEGG and GO enrichment analyses determined a total of 110 DMGs that are involved in the biological functions associated with pathogenesis of cataract. The Sthenurinae were a diverse subfamily of short-faced kangaroos that arose in the Miocene and diversified during the Pliocene and Pleistocene. Many species possessed skull morphologies that were relatively structurally reinforced with bone, suggesting that they were adapted to incorporate particularly resistant foods into their diets. However, the functional roles of many unique, robust features of the sthenurine cranium are not yet clearly defined. Here, the finite element method is applied to conduct a comprehensive analysis of unilateral biting along the cheek tooth battery of a well-represented sthenurine, Simosthenurus occidentalis. The results are compared with those of an extant species considered to be of most similar ecology and cranial proportions to this species, the koala (Phascolarctos cinereus). The simulations reveal that the cranium of S. occidentalis could produce and withstand comparatively high forces during unilateral biting. Its greatly expanded zygomatic arches potentially housed enlarged zygomaticomandibularis muscles, shown here to reduce the risk of dislocation of the temporomandibular joint during biting with the rear of a broad, extensive cheek tooth row. This may also be a function of the zygomaticomandibularis in the giant panda (Ailuropoda melanoleuca), another species known to exhibit an enlarged zygomatic arch and hypertrophy of this muscle. Furthermore, the expanded frontal plates of the S. occidentalis cranium form broad arches of bone with the braincase and deepened maxillae that each extend from the anterior tooth rows to their opposing jaw joints. These arches are demonstrated here to be a key feature in resisting high torsional forces during unilateral premolar biting on large, resistant food items. This supports the notion that S. occidentalis fed thick, lignified vegetation directly to the cheek teeth in a similar manner to that described for the giant panda when crushing mature bamboo culms. Livestock grazing occurs in many protected areas for wildlife and has become a threat to wildlife worldwide. Livestock grazing within protected areas causes negative effects to rare wildlife (e.g., giant panda [Ailuropoda melanoleuca]) and their habitat. We used the 2,000‐km2 Wolong National Nature Reserve, Sichuan Province, southwestern China, to document the effects of livestock on the giant panda and its habitat. We monitored arrow bamboo (Bashania fangiana), wildlife sign (i.e., feces and tracks), and characteristics of plant communities in intact habitat (IH; limited livestock grazing) and disturbed habitat (DH; with grazing disturbance) to assess the effects of livestock grazing and the responses of giant pandas and sympatric species across spatial and temporal scales. Bamboo coverage and the height and basal diameter of bamboo in IH were greater than those in DH, whereas the number of herbaceous species and herbaceous coverage in IH were lower than those in DH. Wildlife signs in IH were greater than those in DH; specifically, giant panda and red panda (Ailurus fulgens) signs were greater, whereas signs of sambar (Rusa unicolor) and tufted deer (Elaphodus cephalophus) in IH were similar to those in DH. Livestock grazing reduced bamboo, which may threaten the long‐term survival of the giant panda. Our results have implications for understanding and management of livestock grazing in the Wolong National Nature Reserve and elsewhere. © 2019 The Wildlife Society. While the relatively stable land use and land cover (LULC) patterns is an important feature of protected areas (PAs), the influence of this feature on future species distribution and the effectiveness of the PAs has rarely been explored. Here, we assessed the role of land use patterns within PAs on the projected range of the giant panda (Ailuropoda melanoleuca) by comparing projections inside and outside of PAs for four model configurations: (1) only climate covariates, (2) climate and dynamic land use covariates, (3) climate and static land use covariates and (4) climate and hybrid dynamic-static land use covariates. Our objectives were twofold: to understand the role of protected status on projected panda habitat suitability and evaluate the relative efficacy of different climate modeling approaches. The climate and land use change scenarios used in the models include two shared socio-economic pathways (SSPs) scenarios: SSP126 [an optimistic scenario] and SSP585 [a pessimistic scenario]. We found that models including land-use covariates performed significantly better than climate-only models and that these projected more suitable habitat than climate-only models. Static land-use models projected more suitable habitat than both the dynamic and hybrid models under SSP126, while these models did not differ under SSP585. China's panda reserve system was projected to effectively maintain suitable habitat inside PAs. Panda dispersal ability also significantly impacted outcomes, with most models assuming unlimited dispersal forecasting range expansion and models assuming zero dispersal consistently forecasting range contraction. Our findings highlight that policies targeting improved land-use practices should be an effective means for offsetting some of the negative effects of climate change on pandas. In recent years, the continuous development of metagenomics has revealed that in addition to the digestive tract, some viruses are also common in mammalian blood. To explore and monitor potential novel viruses, in April 2015, a blood sample was collected from a healthy captive giant panda at the Chengdu Research Base of Giant Panda Breeding in Sichuan Province, China. The genomes of 25 different anelloviruses containing the complete ORF1 region have been identified. The BLASTp results showed that the amino acid sequence identity of these viruses with the best match in GenBank ranged from 27.15% to 41.29%. Based on phylogenetic analysis and SDT (Species Demarcation Tool) analysis of the complete ORF1 regions of these 25 viruses, these sequences were deduced to represent one or several novel virus genera or species. The role of climate change in the evolution and diversification of hominoids remains a hotly debated issue. Stable isotope analyses of fossil mammals that coexisted with the hominoids can provide insights into hominoid palaeoenvironments and shed light on this debate. Here, we report results of stable carbon and oxygen isotope analyses of tooth enamel samples from a variety of Pleistocene mammals including pandas, deer, elephants, pigs, rhinos, and bovids from two hominoid fossil localities (Yugong Cave and Baxian Cave) in South China. Enamel δ13C values indicate that most of the mammals living in the study area during the late Middle Pleistocene had C 3 -based diets but a small number of individuals consumed some C 4 grasses. This indicates the presence of C 4 plants in the region during the late Middle Pleistocene, most likely in patches of open areas in a predominantly forested environment. However, during the early Late Pleistocene, all of the mammals examined had C 3 -based diets, except one bovid and one panda that may have ingested small amounts of C 4 plants. This indicates a dense forested environment with little C 4 grasses during the early Late Pleistocene. Like the Early Pleistocene pygmy panda (Ailuropoda microta) from Yanliang Cave, the late Middle Pleistocene Ailuropoda baconi from Yugong Cave and the early Late Pleistocene Ailuropoda melanoleuca from Baxian Cave had higher mean diet-δ13C values than other co-occurring herbivores, indicating they preferred relatively open forest habitats and had more restricted diets compared to other mammals. The reconstructed mean paleo-meteoric water δ18O w values are lower than the annual average δ18O w value of modern precipitation in the region, suggesting that the climatic conditions during the times when these Pleistocene mammals were alive were colder and/or wetter than today. In addition, δ18O values of the obligate drinkers (pigs, rhinos, bovids) display an overall decreasing trend, accompanied by increased range of δ18O variations, from the Early Pleistocene to the early Late Pleistocene. This suggests that the regional climate became colder and/or wetter, with increased seasonality, from the Early Pleistocene to the early Late Pleistocene, likely related to intensified glaciation. The change in climate to colder conditions may be responsible for the extinction of the Gigantopithecus in this region. • δ13C and δ18O of enamel from Pleistocene mammals in S. Abstract The giant panda (Ailuropoda melanoleuca) is one of the most endangered animals in the world and is recognized worldwide as a symbol for conservation. The Qinling subspecies of giant panda (Ailuropoda melanoleuca qinlingensis) is highly endangered; fewer than 350 individuals still inhabit the Qinling Mountains. Last year, China announced the establishment of the first Giant Panda National Park (GPNP) with a goal of restoring and connecting fragmented habitats; the proposal ignored the environmental pollution caused by economic development in panda habitats. The spatial distribution of heavy metals (Cd, Pb, Hg, Cu, Zn, Mn, Cr, Ni and As) was analyzed in giant panda feces, soil, bamboo, and water in four of GPNP's functional areas at different altitudes and latitudes. Heavy metal pollution decreased with anthropogenic influences, from outside the park through the buffer and into the core area. Cu, Mn, Ni and Zn accumulated from natural sources; As, Hg and Cr were associated with fuel combustion; and Pb and Cd were associated with traffic and agriculture sources. The presence of heavy metals at high altitudes and latitudes in the proposed GPNP is due to emissions from Xi'an and other upwind industrial cities. We conclude that reducing emissions and heavy metal input should be included in the design of the GPNP. Policy interventions should consider functional zones planning, wind direction, reducing mining, and the abandonment of existing roads and farmland within the GPNP to reduce other direct human impacts on the Qinling panda. Competition often occurs between two or more sympatric species that use similar ecological niches. During competition, a superior species may exclude the competitor from parts of its fundamental niche or make it go extinct. Determining the potential competition between two sympatric species including an endangered one has important implications for conservation management. We evaluated potential food competition between the wild boar and the giant panda in a key national nature reserve established primarily for the giant panda protection. We monitored foraging plots for 9 years, conducted food macronutrient analysis, and combined our analysis with long-term population monitoring results for two species. The wild boar population increased dramatically in the past 18 years, benefiting from conservation policies of the Chinese government, whereas the giant panda population decreased. We found evidence for competition for bamboo shoots, an important seasonally limiting resource. The wild boar had a higher utilization rate at foraging plots than giant panda, which also avoided plots used by wild boar. This study indicates key seasonal food competition may exist between wild boar and giant panda. This competition for a key food resource may have negative impacts on giant panda populations, particularly under the substantial increase of the wild boar, yet this possibility has not figured prominently in conservation planning and policy for panda reserves. We suggest long-term monitoring of this competitive relationship across reserves to determine when and where management intervention is needed. The IUCN Red List has downgraded several species from "endangered" to "vulnerable" that still have largely unknown extinction risks. We consider one of those downgraded species, the giant panda, a bamboo specialist. Massive bamboo flowering could be a natural disaster for giant pandas. Using scenario analysis, we explored possible impacts of the next bamboo flowering in the Qinling and Minshan Mountains that are home to most giant pandas. Our results showed that the Qinling Mountains could experience large-scale bamboo flowering leading to a high risk of widespread food shortages for the giant pandas by 2020. The Minshan Mountains could similarly experience a large-scale bamboo flowering with a high risk for giant pandas between 2020 and 2030 without suitable alternative habitat in the surrounding areas. These scenarios highlight thus-far unforeseen dangers of conserving giant pandas in a fragmented habitat. We recommend advance measures to protect giant panda from severe population crashes when flowering happens. The melanocortin-3 receptor (MC3R) is a member of the G protein-coupled receptor superfamily that plays a critical role in controlling energy balance and metabolism. Although pharmacological characterization of MC3R has been reported previously in several other species, there is no report on the MC3R from giant panda (Ailuropoda melanoleuca). This ancient species is known as a 'living fossil' and is among the most endangered animals in the world. Giant panda survive on a specialized diet of bamboo despite possessing a typical carnivorous digestive system. We report herein the molecular cloning and pharmacological characterization of amMC3R. Homology and phylogenetic analysis showed that amMC3R was highly homologous (>85%) to several other mammalian MC3Rs. Using human MC3R (hMC3R) as a control, the binding of five agonists, [Nle4, D-Phe7]-α-melanocyte stimulating hormone (NDP-MSH), α-, β-, γ-, and D-Trp8-γ-MSH, was investigated, as well as Gs-cAMP and pERK1/2 signaling. The results showed that amMC3R bound NDP- and D-Trp8-γ-MSH with the highest affinity, followed by α-, β-, and γ-MSH, with the same rank order as hMC3R. When stimulated with agonists, amMC3R displayed increased intracellular cAMP and activation of pERK1/2. These data suggest that the cloned amMC3R was a functional receptor. The behavioural adaptation (movement and habitat utilization) of translocated organisms to a new environment after their release is crucial in translocation programs because it may affect survival. Therefore, identifying the factors determining habitat selection by the relocated animals is essential to improving the planning and the outcome of such programs. Using the data from three relocated giant pandas (Ailuropoda melanoleuca), we studied the habitat utilization, release-site fidelity, and interaction with resident giant pandas. The results showed that the quality of habitat used by the relocated giant pandas was significantly higher than the average habitat quality of the research region, and was significantly lower than that used by resident giant pandas. This suggested that the released three giant pandas had habitat selection abilities. The three released giant pandas gradually moved away from their release sites and did not exhibit site fidelity. In the first six months, the giant pandas stayed within about 3 km of their release sites, where habitat was good quality but overlapped with the distribution of resident giant pandas. Translocation is believed to be one of the important methods to protect small and isolated populations of giant pandas (Ailuropoda melanoleuca) from local extinction. Dynamic monitoring of the giant pandas' health statuses after release allows timely rescue measures or termination of the release procedure when necessary, thereby reducing the risk of death. However, it is difficult to diagnose the health of a giant panda after translocation by routine veterinary examinations. Based on data collected from GPS tracking collars, we analyzed the behaviours of six giant pandas after release. We compared the behavioural differences between the giant panda Xuexue (XU), which died due to disease, and four healthy giant pandas after release into the wild to identify a new method to assess the health statuses of giant pandas after release. Our results showed that daily activity levels of the healthy giant pandas were higher than XU after released into the wild; the healthy giant pandas were diurnal, but XU tended to be nocturnal. All healthy giant pandas and XU were not active in the crepuscular phase. These results suggest that daily activity levels and circadian rhythms can be used to assess the health status of giant pandas during the translocation project. [ABSTRACT FROM AUTHOR]

Deninger's bears (Ursus deningeri) have been studied less frequently than Ursus spelaeus s.l. Our objective is to present, for the first time, an analysis of the skull shape of U. deningeri. Bear crania and mandibles were digitised with a Microscribe or CT-scanned and the surface models subsequently landmarked. The landmarks were chosen based on a compromise between functional morphology and sample size. Results show that U. deningeri and U. spelaeus mandibles display very similar morphologies and allometric trajectories, both to each other and to Ailuropoda melanoleuca. It is inferred that masticatory adaptations to a herbivorous diet were already present in the Middle Pleistocene. U. deningeri displays a cranial morphology that is similar to that of U. spelaeus when comparing all species, but U. deningeri has a relatively narrower and dorsoventrally lower zygomatic arch than U. spelaeus, although the masticatory signal is less strong in the skull. We observe intraspecific differences between different populations of U. deningeri, which could parallel the genetic diversity found in U. spelaeus. The intraspecific differences found within U. deningeri may be temporal and/or geographical in nature and could be related to the evolution of the Late Pleistocene cave bear, but this hypothesis remains to be tested. In this study, the GnRHR gene polymorphism of the giant panda was detected, and its effects on litter size were analyzed. Thirty-eight giant pandas that were fed under similar conditions were randomly chosen, and polymorphism in the giant panda GnRHR gene coding region was detected by PCR-SSCP. The association between the GnRHR genotypes and litter size traits was carried out with the GLM procedure using the SAS software. The PCR-SSCP test results showed that the T516121C SNP locus existed in the GnRHR gene exon 1, the C504498T SNP locus existed in exon 2, but no SNP locus was found in exon 3. The T516121C and C504498T loci belong to the non-synonymous mutations. The T516121C substitution led to one amino acid mutation (Cys>Arg), as did the C504498T substitution (Ala>Val). In the group of 38 pandas, there were three genotypes at the T516121C and C504498T loci. The chi-square test showed that the T516121C and C504498T loci fit into the Hardy Weinberg equilibrium in the giant panda population (P>0.05). Furthermore, the correlation analysis results indicated that the different genotypes of the C504498T locus in the panda GnRHR gene had significant effects on the average litter size for second births (P=0.05). The average litter size for second births of giant pandas with the CC genotype was higher than that the size of the DD genotype (P=0.05). The first births of the giant pandas with the AA genotype had the maximum average litter size, and there was no significant correlation between the T516121C loci with litter size traits of giant panda. Therefore, these results suggest that the C504498T base mutation in the exon 2 of GnRHR could influence the litter size of giant panda. In other words, GnRHR could influence fertility in giant panda.
