Background: The first step in developing new drugs is to find binding sites for a protein structure that can be used as a starting point to design new antagonists and inhibitors. The methods relying on convolutional neural network for the prediction of binding sites have attracted much attention. This study focuses on the use of optimized neural network for three-dimensional (3D) non-Euclidean data., Methods: A graph, which is made from 3D protein structure, is fed to the proposed GU-Net model based on graph convolutional operation. The features of each atom are considered as attributes of each node. The results of the proposed GU-Net are compared with a classifier based on random forest (RF). A new data exhibition is used as the input of RF classifier., Results: The performance of our model is also examined through extensive experiments on various datasets from other sources. Objective: This study aimed to find the common inborn errors of metabolism in Iranian patients with autism spectrum disorder., Methods: In this cross-sectional multicenter study, 105 children and adolescents with autism spectrum disorder from six centers in different cities of Iran were enrolled between August, 2019 and October, 2020. Metabolic screening, including measuring plasma levels of amino acids, acylcarnitines, creatine, and guani-dinoacetate, and urinary levels of organic acids, purines, and pyrimidines was performed. Other data, including age, parental consanguinity, history of seizure, developmental mile-stones, and physical examination, were also recorded., Results: An inborn error of metabolism was found in 13 (12.4%) patients. Five patients (4.8%) had cerebral creatine deficiency syndrome, 4 (3.8%) had arginine succinate aciduria, 2- methylbutyryl glycinuria, short-chain acyl-CoA dehydrogenase deficiency, and combined methylmalonic aciduria/malonic aciduria. There was a strong association between positive meta-bolic evaluation and parental consanguinity, history of seizures, microcephaly, and delayed development., Conclusions: Our results suggest that metabolic screening should be performed in the cases of autism associated with parental consanguinity, developmental delay, and a history of seizures. The assays to be considered as a screening panel include plasma or blood amino acids, acylcarnitines, creatine and guanidinoacetate, and urinary levels of organic acids.

Iron is one of the most important essential elements for cell function. However, iron overload can exert destructive effects on various tissues, especially the liver. The present study was designed to evaluate the effect of thymoquinone (TQ) on hepatotoxicity induced by iron-overload in in vitro and mouse model. After in vitro studies, thirty mice were divided into five groups, six each. Group 1 received normal saline. Group 2 received five doses of iron dextran (i.p; 100 mg/kg, one dose every 2 days). Group 3 received TQ (orally, 2 mg/kg/day). Groups 4 and 5 were administrated iron dextran saline (i.p; 100 mg/kg, one dose every 2 days) following treatment with 0.5 and 2 mg/kg/day of TQ, respectively. Based on the findings of the DPPH experiment, although TQ has significant anti-radical potential, at a safe dose of 15 × 10 +3  nM, it reduced the IC 50 of iron dextran on HepG2 cells by about 25%, in in vitro. Following administration of low-dose TQ (0.5 mg/kg), a significant improvement was observed in serum hepatic enzymes activity and hepatic lipid peroxidation compared to iron dextran. However, administration of TQ-high dose (2 mg/kg) led to decrease antioxidant defense alongside increased serum hepatic enzymes and pathological damages in iron dextran-treated animals. Due to the different efficacy of TQ in treatment groups, it seems that the TQ therapeutic index is low and does not have significant safety in the iron overload status., (© 2022. Objectives. Human error (HE) plays a crucial role in the occurrence of accidents in chemical process industries (CPIs). Emergency response team (ERT) members are predisposed to HEs due to the nature of their work. The HE potential is influenced by the performance shaping factors (PSFs). Managing PSFs can diminish the human error probability (HEP) and consequently increase the emergency response success chance. This article aimed to determine the PSFs for ERT members in CPIs. Methods . First, an initial list of PSFs was searched and classified within human reliability analysis methods and studies. Then, an expert panel of the emergency management system was utilized to identify, classify and weight the initial PSFs. The fuzzy Delphi method and content analysis technique were applied to summarize and categorize the PSFs. Results. The results of the study showed that 11 PSFs had greater impacts on the ERT members' error potential. Findings revealed that stress and physiological stressors, competency, and team and organization were the three most important PSFs. Conclusion. The most important and relevant PSFs can be effectively used in accurate HE assessment of ERT members in CPIs.

Background: Dermatophytes have the ability to invade the keratin layer of humans and cause infections. The aims of this study were the accurate identification of dermatophytes by Polymerase Chain Reaction-Restriction Fragment Length Polymorphism method and sequencing and comparison between the in vitro activities of newer and established antifungal agents against them., Methods: Clinical specimens of patients from five Iranian university laboratories were entered in this study. Samples were cultured on sabouraud dextrose agar medium. For molecular identification, extracted DNAs were amplified by the universal fungal primers ITS1 and ITS4, and digested with MvaI enzymes. The antifungal susceptibility test for each isolate to terbinafine, griseofulvin, caspofungin, fluconazole, itraconazole, luliconazole, and isavuconazole was performed, according to the microdilution CLSI M38-A2 and CLSI M61 standard methods., Results: Two hundred and seven fungi species similar to dermatophytes were isolated of which 198 (95.6%) were dermatophytes by molecular assay. The most commonly isolated were Trichophyton mentagrophytes (76/198), followed by Trichophyton interdigitale (57/198), Trichophyton rubrum (34/198), Trichophyton tonsurans (12/198), Microsporum canis (10/198), Trichophyton simii (3/198), Epidermophyton floccosum (3/198), Trichophyton violaceum (2/198), and Trichophyton benhamiae (1/198). The GM MIC and MIC 90 values for all the isolates were as follows: terbinafine (0.091 and 1 μg/ml), griseofulvin (1.01 and 4 μg/ml), caspofungin (0.06 and 4 μg/ml), fluconazole (16.52 and 32 μg/ml), itraconazole (0.861 and 8 μg/ml), isavuconazole (0.074 and 2 μg/ml), and luliconazole (0.018 and 0.25 μg/ml)., Conclusion: Trichophyton mentagrophytes, Trichophyton interdigitale, and Trichophyton rubrum were the most common fungal species isolated from the patients. luliconazole, terbinafine, and isavuconazole in vitro were revealed to be the most effective antifungal agents against all dermatophyte isolates., (© 2023. Protective gloves can affect hand performance indicators (HPIs) like manual dexterity and hand grip. The present study was conducted to comprehensively and comparatively investigate several types of protective gloves and HPI assessment tools. Seventeen healthy men participated in this study. Four types of protective gloves, two structural firefighting and two general protective gloves, were investigated using four different dexterity tests and the bulb dynamometer. Structural firefighting gloves were significantly different in terms of dexterity, while there was no significant difference between general protective gloves. In contrast, firefighting gloves were not significantly different in terms of hand grip strength, while general protective gloves were significantly different in this regard. Among the four investigated tests, the hand tool dexterity test had the highest discrimination power. The negative effects of structural firefighting gloves on HPIs were higher than those of general protective gloves. Background: Cytochrome P450 complex plays a key role in drug metabolism. CYP2B6 has an essential part in Cytochrome P450 complex metabolism. This study aims to determine the allelic distribution of CYP2B6 ∗ 2 and CYP2B6 ∗ 3 in three main Iranian ethnicities: Fars, Turk, and Kurd., Methods: The study was conducted on 174 unrelated healthy volunteers from three main Iranian ethnicities. After DNA extraction from peripheral blood samples, genotyping of CYP2B6 ∗ 2 and ∗ 3 was performed using tetra ARMS and ARMS PCR, respectively., Results: The average age of 174 cases was 40.69 ± 11.87 (mean ± SD) and 39.06 ± 11.63 (mean ± SD) for males and females. In the CYP2B6 ∗ 2 variant, the genotyping frequency of wild type (C/C), heterozygous (C/T), and homozygous mutant (T/T) was 8.7%, 86%, and 5.2%, respectively. The CYP2B6 ∗ 2 (c.64C > T) allele frequency was 48.2% (95% CI: (37.8-58.6)). In the CYP2B6 ∗ 3 variant, the frequency of wild type (C/C), heterozygous (C/T), and homozygous mutant (T/T) was 75.3%, 11%, and 13.6%, respectively. The CYP2B6 ∗ 3 (c.777C > A) allelic frequency was 19.1% (95% CI: (17.5-20.7))., Conclusion: Allelic distribution in three main Iranian ethnicities, i.e., Turk, Kurd, and Fars, is remarkably higher than that in other populations, even that in Southern Iran. High frequencies of CYP2B6 ∗ 2 and ∗ 3 in the Iranian population highly affect drug responsiveness. Background: Application of a Clinical Information System (CIS) like Electronic Patient Record (EPR), PACS system and CPOE has turned into one of the most important criteria of priorities of health care systems. The aims of the clinical information system include improving the physicians' efficiency level, integrating the caring process, and expanding the fuzzy quality of the services offered to patients. Achievement of these benefits in reality is not an easy task, and there are lots of plans in this field which are doomed to failure. About 50% of the implementation plans of clinical information systems in health care organizations have failed, and this trend is significantly affecting industrial countries. Proper implementation of hospital information systems lies in identifying and assessing the relationships among the most important risk factors of fuzzy. The present study aimed to provide an applicable model for identifying, ranking and evaluating the risk factors associated with projects of clinical information technology in hospitals of Shiraz University of Medical Sciences., Method: This is an applied study which evaluates the risk factors associated with implementation of clinical information technology projects in hospitals of Shiraz Medical Sciences University. The participants consisted of professionals and senior experts of clinical information technology. Fuzzy logic was used in this study. We also applied ANP-DEMATEL combined model with fuzzy procedure to provide the analytic model of the study., Results: According to the study findings, lack of top-executive supports, and unstable organizational environment were the two most important risk factors, while the main organizational factors and technology were also highly important. Inspired by natural resources, such as peptides and carbohydrates, glycopolypeptide biopolymer has recently emerged as a new form of biopolymer being recruited in various biomedical applications. Glycopolypeptides with well-defined secondary structures and pendant glycosides on the polypeptide backbone have sparked lots of research interest and they have an innate ability to self-assemble in diverse structures. The nanostructures of glycopolypeptides have also opened up new perspectives in biomedical applications due to their stable three-dimensional structures, high drug loading efficiency, excellent biocompatibility, and biodegradability. Although the development of glycopolypeptide-based nanocarriers is well-studied, their clinical translation is still limited. Aims: The relationship between circulating Lutein and zeaxanthin (L/Z) concentrations, and plasma lipoproteins has been indicated by observational studies. However, the beneficial impact of L/Z administration on dyslipidemia are unclear. This meta-analysis aimed to investigate the effect of oral intake of L/Z on circulating total cholesterol (TC), low-density lipoprotein-cholesterol (LDL-C), as well as high-density lipoprotein-cholesterol (HDL-C) levels., Methods: We electronically assessed all eligible interventional studies through different electronic databases, including PubMed, Scopus, ISI -Web of Science, and Cochrane library until Jun 2021. After identifying the quality of each included randomized controlled trials, they were evaluated by assessing the risk-difference between treatment and control groups by pooling available data on net change of serum LDL-C, HDL-C, and Cholesterol., Results: L/Z supplementation has null effect on circulating levels of TC (WMD: -3.82 95% CI: -13.83, 6.18; I-square: 85.2%), and LDL-C (WMD: -4.54; 95% CI: -11.5, 2.48; I-square: 83.9%). In contrast, L/Z treatment could significantly increase HDL-C levels in older adults (WMD: 4.06; 95% CI: 0.64, 7.48; I-square: 50.7%)., Conclusion: L/Z administration could be an effective treatment for improving circulating HDL-C concentration in elderly adults., (Copyright © 2022 Elsevier Inc. Paper-based analytical devices (PADs) have shown great promise for point-of-care testing and on-site detection of analytes with chemical, biochemical, and environmental importance owing to their low cost, convenience, scalability, portability, and biocompatibility. The World Health Organization stated that sensors should meet the ASSURED criteria (affordable, sensitive, specific, user-friendly, rapid and robust, equipment-free, and deliverable). Paper-based optical sensors meet most of these criteria, making them in high demand and applicable in remote areas. Optical PADs outputs are obtained by different means, such as dyes, nanostructures, redox agents, and pH indicators. The outstanding physical and chemical characteristics of nanostructures, their intense signals, and tunable optical properties make them ideal for many sensing platforms, including paper-based ones. This review focuses primarily on paper-based nanosensors using various nanostructures to fabricate and produce optical signals for visualization. We describe the fundamentals and state of the art of PADs and comprehensively explain the following topics: paper types as the substrate of PADs, PAD fabrication approaches, nanostructure stabilization on PADs, signal acquisition, data handling, interpretation of results, sensing mechanisms, and application areas. Epstein-Barr virus (EBV) is a gamma-herpesvirus associated with 10% of all gastric cancers (GCs) and 1.5% of all human cancers. EBV-associated GCs (EBVaGCs) are pathologically and clinically distinct entities from EBV-negative GCs (EBVnGCs), with EBVaGCs exhibiting differential molecular pathology, treatment response, and patient prognosis. However, the tumor immune landscape of EBVaGC has not been well explored. In this study, a systemic and comprehensive analysis of gene expression and immune landscape features was performed for both EBVaGC and EBVnGC. EBVaGCs exhibited many aspects of a T cell-inflamed phenotype, with greater T and NK cell infiltration, increased expression of immune checkpoint markers (BTLA, CD96, CTLA4, LAG3, PD1, TIGIT, and TIM3), and multiple T cell effector molecules in comparison with EBVnGCs. EBVaGCs also displayed a higher expression of anti-tumor immunity factors (PDL1, CD155, CEACAM1, galectin-9, and IDO1). Six EBV-encoded miRNAs (miR-BARTs 8-3p, 9-5p, 10-3p, 22, 5-5p, and 14-3p) were strongly negatively correlated with the expression of immune checkpoint receptors and multiple markers of anti-tumor immunity. These profound differences in the tumor immune landscape between EBVaGCs and EBVnGCs may help explain some of the observed differences in pathological and clinical outcomes, with an EBV-positive status possibly being a potential biomarker for the application of immunotherapy in GC.

C-reactive protein (CRP) is a plasma protein that is one of the most expressed proteins in acute phase inflammation cases. It is a well-known biomarker for inflammatory disorders. There is a significant correlation between increasing CRP concentration and the risk of being exposed to cardiovascular diseases (CVD) and sepsis; thus, monitoring and quantifying CRP levels in a simple, inexpensive, and quick manner can improve clinical diagnostics and help prevent major inflammatory conditions. Here a nanobiosensor was developed, benefiting from the LSPR property of gold-nanorod (GNR) to measure CRP concentration. Nanorods were fabricated using One-pot synthesis by trimethyl ammonium bromide (CTAB) as a surfactant. This method provides the advantage of both step and time reduction in synthesis and decreases the contamination probability of nanorods as the products. The nanorods were characterized using TEM with an average size of (24 ± 1 nm) × (5 ± 1 nm) and a typical aspect ratio of ∼4.9. The surface of the rods was modified with a specific aptamer for the target protein, and the LSPR shifts due to the gold nanorod's refractive index change as the result of protein interaction with the biosensor investigated using a 100-900 nm UV absorption device. The results indicated that the nanobiosensor could respond to different CRP concentrations within 30 min. The selectivity test has shown nonresponsive results of nanobiosensor to BSA and TNF-α proteins which are used to evaluate the biosensor behavior in non-target proteins. In clinical practice, bone defects that occur alongside tumors, infections, or other bone diseases present significant challenges in the orthopedic field. Although autologous and allogeneic grafts are introduced as common traditional remedies in this field, their applications have a series of limitations. Various approaches have been attempted to treat large and irregularly shaped bone defects; however, their success has been less than optimal due to a range of issues related to material and design. However, in recent years, additive manufacturing has emerged as a promising solution to the challenge of creating implants that can be perfectly tailored to fit individual defects during surgical procedures. By fabrication of constructs with specific designs using this technique, surgeons are able to achieve much better outcomes for patients. Polymers, ceramics, and metals have been used as biomaterials in Orthopedic Surgery fields. Polymeric scaffolds have been used successfully in total joint replacements, soft tissue reconstruction, joint fusion, and as fracture fixation devices. The use of polymeric biomaterials, either in the form of pre-made solid scaffolds or injectable pastes that can harden in situ, shows great promise as a substitute for commonly used autografts and allografts. As the frontline of our education system, teachers endure greater job-related stress than other professionals, even under the best of circumstances. While they were already exposed to certain stressors affecting their emotional health, the pandemic outbreak introduced new challenges putting teachers at risk of experiencing higher rates of emotional distress. This longitudinal study aimed to identify changes in teaching stressors and teachers' coping strategies in the period before the pandemic to the immediate outbreaks of COVID-19 in Fall 2020. In addition, we examined the correlation of teachers' coping approaches with stress, anxiety, and depression to understand if coping strategies correlate and predict such emotional distress. To this end, 376 English as a foreign language (EFL) teachers in Iran in the first wave and 351 in the second wave completed a battery of validated inventories capturing their emotional distress, teaching stressors, and coping approaches. The mean scores of the stressors and coping strategies across two waves indicated significant shifts toward using functional coping strategies upon experiencing new demands. Furthermore, we found that novice teachers experienced higher rates of stress and anxiety, which were positively associated with dysfunctional coping strategies. The results of the stepwise regression analysis with (dys)functional coping strategies indicated that coping mechanisms significantly account for 25% of variances in stress, anxiety, and depression. Background: Workplace inspections are applied to facilitate the adherence to the occupational health and safety regulations. The Iranian Ministry of Health introduced a new software system for tablets to inspect workplaces., Objectives: The aim of this study was to take measurements of the usability, mental workload, and mood of inspectors., Methods: Inspectors used both pen-and-paper and tablet methods to inspect the automotive industry in a mixed-method procedure. The NASA-TLX score, QUIS score, I-PANAS (SF) situation, inspection time, and number of errors were collected throughout the procedure. The differences were investigated using a paired sample and the Wilcoxon signed ranks test., Results: In terms of efficacy, using the tablet resulted in lower error rates, but it took longer to complete the inspection task (P < 0.001). Participants perceived a lower workload when inspecting with a tablet rather than the traditional method. (Mental Demand: p < 0.002, Performance: p < 0.009, Effort: p < 0.012, TLX: p < 0.002 based on various subcomponents of NASA-TLX). The newly introduced system's usability was insufficient., Conclusion: Although the use of tablets has improved safety inspections, ergonomic redesign of the system and consideration of a user-centered approach, as well as inspector training, can make the system more likely to succeed.

Problematic internet use (PIU) can cause mental and physical harm to individuals and may be an emerging factor contributing to unsafe work behavior. In this study, the relationship between PIU and safety behavior was investigated. Moreover, it was hypothesized that safety climate can moderate the relationship between PIU and safety behavior. Participants were employees from a mining industry in Iran. Three validated questionnaires were distributed to gather the required data regarding PIU, safety climate, and safety behavior. Hierarchical regression analysis was used to assess the moderating effect of safety climate on the relationship between PIU and safety behavior. The size of moderation effect was assessed using f2 index. Four hundred eighty-five employees participated in this study. The results demonstrated a negative relationship between PIU and safety behavior. The moderating effect of safety climate on the relationship between PIU and safety behavior was supported. The f2 index was 0.027 which demonstrates a small moderation effect. In conclusion, PIU has a significant negative effect on the safety behavior of employees. Background: In this study, the effects of methadone and naloxone on the expression of toll-like receptor 4 ( TLR4 ) gene have been evaluated in human non-small cell lung carcinoma A549 cell line migration using in-silico and in vitro techniques., Materials and Methods: Lung cancer A549 cell cultures were stimulated for 24 h with methadone (5, 10, and 20 μM) and naloxone (20 and 40 μM) concentrations. The level of TLR4 expression was determined by the quantitative real-time polymerase chain reaction. Migration of the A549 cells was investigated after a 4-h incubation period with methadone using the Boyden Chamber assay., Results: Migration rate of the A549 cells treated with 5 ( P < 0.05) and 20 ( P < 0.01) μM methadone was, respectively, increased and decreased with 20 μM naloxone ( P < 0.05). Furthermore, the TLR4 expression was enhanced with 5 ( P < 0.05) and 20 ( P < 0.01) μM methadone and decreased with 20 ( P < 0.05) and 40 μM naloxone ( P < 0.01). In addition, in silico docking analysis revealed docking of methadone to MD-2 and TLR4 ., Conclusion: According to the present DATA, methadone affects the TLR4 expression. It may however cause adverse consequences by increasing the TLR4 expression. In recent decades, the incidence of Candida infections has increased in immunocompromised patients. This multicenter study aimed to evaluate in vitro antifungal activities of 8 antifungal agents against the Candida species isolated from 10 university hospitals in Iran. During the period from Dec 2019 to Dec 2021, Candida species were collected from clinical samples of patients. The isolates were identified by PCR restriction fragment length polymorphism and sequencing methods. The antifungal susceptibility tests of each isolate to eight antifungal agents were performed according to the microdilution CLSI M27, M59, and M60 standard methods. A total of 598 Candida strains were isolated from clinical samples. The most commonly isolated Candida species was C. albicans, followed by C. glabrata, C. parapsilosis, Debaryomyces hansenii (Candida famata), C. tropicalis, Pichia kudriavzevii (Candida krusei), C. orthopsilosis, Meyerozyma guilliermondii (Candida guilliermondii), Kluyveromyces marxianus (Candida kefyr), and Clavispora lusitaniae (Candida lusitaniae). MIC 90 values in all Candida species were as follows: 0.25 μg/mL for caspofungin and voriconazole; 0.5 μg/mL for amphotericin B and isavuconazole; 2 μg/mL for itraconazole, luliconazole, and posaconazole; and 16 μg/mL for fluconazole. Although 30/285 C. albicans, 15/31 C. hansenii, 3/12 M. guilliermondii , 67/125 C. glabrata, 5/15 P. kudriavzevii , 6/60 C. parapsilosis, and 5/23 C. tropicalis isolates were multiazole resistant with resistance to 2 to 4 azoles, pan-azole resistance was not observed. According to our data, Candida albicans and C. glabrata were the most frequent species isolated from clinical samples in Iran. Caspofungin and voriconazole, with lower MIC 90 values, are the most effective than other antifungal agents for the treatment of Candida infections in this region. IMPORTANCE Candida species cause severe invasive infections of the heart, brain, eyes, bones, and other parts of the body. Knowledge of regional distributions of causative Candida agents and their antifungal susceptibility patterns can help to monitor resistance to antifungal agents of various species and support local and national surveillance programs. In the present study, C. albicans and C. glabrata were the most frequently isolated species from clinical samples in Iran. Increasing rates of non- albicans Candida isolates from the Iranian population should be looked at as alarming due to various levels of intrinsic MIC values or resistance to various antifungal drugs. Caspofungin and voriconazole are recommended over fluconazole for the treatment of Candida infections in the study region. However, amphotericin B and isavuconazole are also active against the most common Candida species isolated from patients. Pan azole-resistant Candida species were not observed in the present study.

Background: Fever is the most common reason for children's visits to medical centers. Its management is an essential duty of a pediatric nurse. The aim of this study was to determine the effect of body wash with Marshmallow plant on children's fever., Methods: This parallel clinical trial was performed on 92 children aged 6 months to 10 years with a tympanic temperature above 38.3 °C. Participants were randomly assigned to groups. Simultaneously with receiving acetaminophen, body wash was performed in the control group with lukewarm water and in the intervention group with white Marshmallow extract. The children's temperature; from the beginning of the study was checked and recorded every 15 min in the first hour and in the 4th and 6th hours. The time duration to resolve fever, the frequency of afebrile children at different times of the study, and the value of temperature reduction were primary outcomes. Heart rate, the need to administer the next dose of acetaminophen, and the time of fever recurrence were recorded as secondary outcomes., Results: The mean time duration to resolve fever in the intervention group was shorter than in the control group (B = 8.181, 95% CI 3.778-12.584, p < 0.001). The frequency of the children without fever was higher in the intervention group during different times of the study (p < 0.001). The mean value of temperature reduction in the intervention group was higher than the control group (B = -0.27 °C, 95% CI: -0.347 to -0.193, P < 0.001), although, after adjusting the effect of confounding variables it was not significant (P = 0.127). The mean of adjusted heart rate change (p = 0.771), the time of fever recurrence (P = 0.397), and the frequency of children requiring the next dose of acetaminophen (p = 0.397) did not show a significant difference between the groups., Conclusion: Body wash with Marshmallow extract reduced children's fever in a shorter period of time and to some extent a greater extent than the control group without side effects. Therefore, it can be used as an effective and safe complementary method to help reduce fever. Ongoing novel coronavirus (COVID-19) with high mortality is an infectious disease in the world which epidemic in 2019 with human-human transmission. According to the literature, S-protein is one of the main proteins of COVID-19 that bind to the human cell receptor angiotensin-converting enzyme 2 (ACE2). In this study, it was attempted to identify the main effective drugs approved that may be repurposed to the binding site of ACE2. High throughput virtual screening based on the docking study was performed to know which one of the small-molecules had a potential interaction with ACE2 structure. Forasmuch as investigating and identifying the best ACE2 inhibitors among more than 3,500 small-molecules is time-consuming, supercomputer was utilized to apply docking-based virtual screening. The outbreak of the COVID-19 pandemic has resulted in many emotional consequences for teachers, including feelings of isolation, loneliness, and hopelessness. However, evidence on the prevalence of hopelessness and the associated factors in teachers during the pandemic is limited. The purpose of this research was to examine the prevalence of hopelessness in public school teachers and identify risk factors associated with it. A sample of 168 teachers aged 25-49 years participated in the study by completing the Socio-Demographic Questionnaire, the Beck Hopelessness Scale (BHS), and the Multi-Dimensional Scale of Perceived Social Support (MSPSS). The results revealed a moderate level of experienced hopelessness in teachers. Hopelessness prevalence was also significantly different across teacher gender (males = 79%), age groups (>40 = 77%), socioeconomic status (poor socioeconomic status = 70%), educational level (high school teachers = 79%), professional experience (experienced teachers = 82%), and perceived social support (low perceived social support = 79%). The results of a logistic regression analysis confirmed the effects of these demographic and occupational attributes on hopelessness by explaining ~71% of the variance in hopelessness feelings. Higher odds ratios were associated with age, socioeconomic status, and perceived social support, signifying the prominence of these factors in predicting hopelessness. The last pandemic exposed critical gaps in monitoring and mitigating the spread of viral respiratory infections at the point-of-need. A cost-effective multiplexed fluidic device (NFluidEX), as a home-test kit analogous to a glucometer, that uses saliva and blood for parallel quantitative detection of viral infection and body's immune response in an automated manner within 11 min is proposed. The technology integrates a versatile biomimetic receptor based on molecularly imprinted polymers in a core-shell structure with nano gold electrodes, a multiplexed fluidic-impedimetric readout, built-in saliva collection/preparation, and smartphone-enabled data acquisition and interpretation. NFluidEX is validated with Influenza A H1N1 and SARS-CoV-2 (original strain and variants of concern), and achieves low detection limit in saliva and blood for the viral proteins and the anti-receptor binding domain (RBD) Immunoglobulin G (IgG) and Immunoglobulin M (IgM), respectively. It is demonstrated that nanoprotrusions of gold electrodes are essential for the fine templating of antibodies and spike proteins during molecular imprinting, and differentiation of IgG and IgM in whole blood. Background: Invasive aspergillosis is one of the important causes of infection in immunocompromised patients. This study aimed to evaluate the roles of biomarkers in the diagnosis of invasive aspergillosis and their relationship with antifungal stewardship programs., Methods: 190 sera from 52 immunocompromised patients and volunteer individuals were included in this study. 18 immunocompromised volunteers without IA and 34 patients with probable and proven aspergillosis according to the European Organization for Research and Treatment of Cancer and the Mycoses Study Group consensus definitions were entered in this study. The respective sera were evaluated for procalcitonin, soluble triggering receptor expressed on myeloid cells-1 (sTREM-1) levels; white blood cells count (WBC) count, C reactive protein (CRP), lactate dehydrogenase (LDH), and erythrocyte sedimentation rate (ESR) values. Demographic data and clinical characteristics of patients were extracted from their files., Results: The male-to-female ratio and mean age of patients were 22/12 and 38.9 years, respectively. The hematologic disorder was the most predisposing factor (29/34, 85.3%). Sensitivity of biomarkers for diagnosis of invasive aspergillosis was 70.6% (cut off value > 190 pg/mL for sTREM-1, 71% (cut off value > 260 pg/mL) for PCT, 85.3% (cut off value > 193 U/L) for LDH, 94.1% (cut off value > 8 mg/l) for CRP, 64.7% (cut off value < 5200 cells/ml) for WBC, and 85.3% (cut off value > 23 mm/h) for ESR. Twelve patients died, with significantly increased sTREM-1 levels and decreased WBC count in them., Conclusion: According to our data, evaluation of the biomarkers can help in the diagnosis, management, and prediction of the severity of Aspergillus infection, and the rational use of antifungal agents in immunocompromised patients., (© 2022. Endometriosis is a benign chronic disease in women that is characterized by the presence of active foci of the endometrium or endometrial tissue occurring outside of the uterus. The disease causes disabling symptoms such as pelvic pain and infertility, which negatively affect a patient's quality of life. In addition, endometriosis imposes an immense financial burden on the healthcare system. At present, laparoscopy is the gold standard for diagnosing the disease because other non-invasive diagnostic tests have less accuracy. In addition, other diagnostic tests have low accuracy. Therefore, there is an urgent need for the development of a highly sensitive, more specific, and non-invasive test for the early diagnosis of endometriosis. Numerous researchers have suggested miRNAs as potential biomarkers for endometriosis diagnosis due to their specificity and stability. However, the greatest prognostic force is the determination of several miRNAs, the expression of which varies in a given disease. Despite the identification of several miRNAs, the studies are investigatory in nature, and there is no consensus on them. In the present review, we first provide an introduction to the dysregulation of miRNAs in patients with endometriosis and the potential use of miRNAs as biomarkers in the detection of endometriosis. Then we will describe the role of the mir-200 family in endometriosis. Several studies have shown that the expression of the mir-200 family changes in endometriosis patients, suggesting that they could be used as a diagnostic biomarker and therapeutic target for endometriosis.

The present study aimed to investigate fatigue among firefighters and its relationship with their safety behavior. The moderation and mediation effects of perceived safety climate on this relationship were also investigated. Firefighters from 10 petrochemical companies in Iran were investigated. Moderation analysis was performed based on hierarchical regression analyses and mediation effects were studied using structural equation modeling. A total number of 261 firefighters participated in this study. The mean scores of general fatigue, physical fatigue, reduced activity, reduced motivation and mental fatigue were 10.5, 9.0, 8.2, 7.6 and 8.7, respectively. Fatigue had a negative significant effect on firefighters' safety behavior. Perceived safety climate had a mediation but not moderation effect on the relationship between fatigue and safety behavior of firefighters. Generally, the fatigue level among firefighters was fairly low, but had a significant negative effect on firefighter's safety behavior. Perceived safety climate can mediate such a negative effect.

Objectives. The hydrocarbon road tanker loading operation is vulnerable to human error. The present study aimed to develop a methodology for predicting human error probabilities (HEPs) in various subtasks of this operation. Methods. First, task analysis was performed using hierarchal task analysis. Then, HEP was calculated using a hybrid technique of fuzzy set theory (FST), Bayesian network (BN) and cognitive reliability and error analysis method (CREAM). FST was used for handling uncertainties regarding common performance conditions (CPCs) and the BN was employed for modeling the interrelationships among CPCs and HEPs. The weighted sum algorithm was used for quantifying conditional probability tables in the network. Results . Twenty-six subtasks were required for completing the road tanker loading operation. Investigating the internal parts of the tanker before the loading operation and attaching the ground rode clamp were the subtasks with highest HEPs. Working conditions and crew collaboration were the CPCs with the highest contribution to these errors. HEP was most sensitive to crew collaboration. Conclusion . Improving collaboration among the driver, site operators and control room operators, as well as increasing the knowledge of the road tanker driver regarding the hazards of incompatible chemicals, are the best practices for reducing HEPs in this operation.

The secretion efficiency of a heterologous protein in E. coli is mainly dictated by the N-terminal signal peptide fused to the desired protein. In this study, we aimed to select and introduce mutations into the - 1, - 2 and - 3 positions of the gIII signal peptide (originated from filamentous phage fd Gene III) fused to the N-terminus of the human growth hormone (hGH), and study its effect on the secretion efficiency of the recombinant hGH into the periplasmic space of E. coli Top10. Bioinformatics software such as SignalP-5.0 and PrediSi were employed to predict the effects of the mutations on the secretion efficiency of the recombinant hGH. Site-directed mutagenesis was applied to introduce the desired mutations into the C-terminus of the gIII signal peptide. The periplasmic expression and the secretion efficiency of the recombinant hGH using the native and mutant gIII signal peptides were compared in E. coli Top10 under the control of araBAD promoter. Our results from bioinformatics analysis indicated that the mutant gIII signal peptide was more potent than the native one for secretion of the recombinant hGH in E. coli. While our experimental results revealed that the mutation had no effect on hGH secretion. This result points to the importance of experimental validation of bioinformatics predictions., (© 2022. Background: Coronavirus infection has caused widespread concern among mothers and physicians about the health of pregnant women and infants., Objective: The aim of this study was to evaluate the clinical and prenatal findings of pregnant women with coronavirus disease-2019 (COVID-19) virus., Materials and Methods: The present study was a descriptive study that was conducted in 6 mother and child care centers. In this study, 81 pregnant women with COVID-19 admitted to centers in the period from March 2020-September 2020 were studied. Clinical and prenatal findings of the pregnant mothers were recorded using a data collection form with details of demographic characteristics and these were analyzed., Results: The gestational age of the affected women was between 4 and 40 wk. 48 deliveries were performed and 25% of deliveries were preterm. Coronavirus infection was the cause of termination of pregnancy in 4 cases. The most common symptoms of women when visiting the medical centers were: dry cough (58.0%), muscle pain and myalgia (56.8%) and fever (51.9%). The most common laboratory findings in the women were: increased C-reactive protein (67.90%), lymphopenia (18.51%), decreased white blood cells (27.16%), and increased liver enzymes (18.51%). Regarding the status of the newborns, out of the 33 neonates examined, 3 neonates were diagnosed with COVID-19., Conclusion: The most common symptoms of pregnant women with COVID-19 are similar to those of other adults. Breast cancer is the most common type of cancer in women and the second leading cause of cancer death in female. Triple-negative breast cancer has a more aggressive proliferation and a poorer clinical diagnosis than other breast cancers. The most common treatments for TNBC are chemotherapy, surgical removal, and radiation therapy, which impose many side effects and costs on patients. Nanobodies have superior advantages, which makes them attractive for use in therapeutic agents and diagnostic kits. There are numerous techniques suggested by investigators for early detection of breast cancer. Nevertheless, there are fewer molecular diagnostic methods in the case of TNBC due to the lack of expression of famous breast cancer antigens in TNBC. Although conventional antibodies have a high ability to detect tumor cell markers, their large size, instability, and costly production cause a lot of problems. Since the HER-2 do not express in TNBC diagnosis, the production of nanobodies for the diagnosis and treatment of cancer cells should be performed against other antigens expressed in TNBC. We herein fabricated a cancer nanotheranostics platform based on Graphene Oxide Quantum Dot-Chitosan-polyethylene glycol nanoconjugate (GOQD-CS-PEG), which were targeted with MUC-1 aptamer towards breast and colon tumors. The interaction between aptamer and MUC-1 receptor on the desired cells was investigated utilizing molecular docking. The process of curcumin release was investigated, as well as the potential of the produced nanocomposite in targeted drug delivery, specific detection, and photoluminescence imaging. The fluorescence intensity of GOQD-CS-PEG was reduced due to transferred energy between (cytosine-guanin) base pairs in the hairpin structure of the aptamer, resulting in an "on/off" photoluminescence bio-sensing. Interestingly, the integration of pH-responsive chitosan nanoparticles in the nanocomposite results in a smart nanocomposite capable of delivering more curcumin to desired tumor cells. When selectively binds to the MUC-1 receptor, the two strands of aptamer separate in acidic conditions, resulting in a sustained drug release and photoluminescence recovery. The cytotoxicity results also revealed that the nanocomposite was more toxic to MUC-1-overexpressed tumor cells than to negative control cell lines, confirming its selective targeting. As a result, the proposed nanocomposite could be used as an intelligent cancer nanotheranostic platform for tracing MUC-1-overexpressed tumor cells and targeting them with great efficiency and selectivity., (Copyright © 2022 Elsevier B.V. Layered metal chalcogenide materials are exceptionally appealing in optoelectronic devices thanks to their extraordinary optical properties. Recently, their application as flexible and wearable photodetectors have received a lot of attention. Herein, broadband and high-performance paper-based PDs were established in a very facile and inexpensive method by rubbing molybdenum disulfide and titanium trisulfide crystals on papers. Transferred layers were characterized by SEM, EDX mapping, and Raman analyses, and their optoelectronic properties were evaluated in a wavelength range of 405-810 nm. Although the highest and lowest photoresponsivities were respectively measured for TiS 3 (1.50 mA/W) and MoS 2 (1.13 μA/W) PDs, the TiS 3 -MoS 2 heterostructure not only had a significant photoresponsivity but also showed the highest on/off ratio (1.82) and fast response time (0.96 s) compared with two other PDs. This advantage is due to the band offset formation at the heterojunction, which efficiently separates the photogenerated electron-hole pairs within the heterostructure. Numerical simulation of the introduced PDs also confirmed the superiority of TiS 3 -MoS 2 heterostructure over the other two PDs and exhibited a good agreement with the experimental results. Finally, MoS 2 PD demonstrated very high flexibility under applied strain, but TiS 3 based PDs suffered from its fragility and experience a remarkable drain current reduction at strain larger than ± 0.33%. Background: Crohn's disease (CD) has a chronic course, which its recurrence varies widely among different patients. In this study we prospectively analyzed blood samples of 19 CD patients. Alteration in transcription of inflammatory and anti-inflammatory cytokines was analyzed compared with household members after three month follow up., Methods: CD patients were diagnosed based on clinical symptoms, endoscopic and histopathologic characteristics. Nineteen CD patients and their households were evaluated from Jun 2019 to Feb 2021 at Tehran university hospitals. CD activity score, biological, clinical and demographic data of the patients were recorded at two time point intervals. Bacteriological tests were done using aerobic and anaerobic blood cultures. To investigate transcriptional alterations, peripheral blood mononuclear cells (PBMCs) were isolated using Ficol centrifugation method and relative quantitative real-time PCR was done to determine the expression level of IFN-γ, TNF-α, IL10, and FOXP3 cytokines., Results: Our results showed a correlation between fecal calprotectin level (709.8 ± 554.6), C-reactive protein concentration (18.1 ± 15.9), and erythrocyte sedimentation rate (30.4 ± 17.9) with disease activity (Flare/remission). IL10 and Foxp3 anti-inflammatory gene's expression were significantly ( P = 0.003 for IL10 and P = 0.008 Foxp3) higher during the flare and remission in patients with active disease respectively. Bacteriological examination showed infection with Streptococcus spp. and Clostridium spp. in two CD patients during flares, which was correlated with upregulation and down-regulation of IL10, TNF-α, IFN-γ and FOXP3 proteins, respectively., Conclusion: Occurrence of bacteremia, and higher amount of CAP, CRP and ESR are correlated with higher level of transcription for inflammatory cytokines, which could effectively reflect the disease activity. Aspergillus species are a major cause of life-threatening invasive infections and noninvasive diseases. This study seeks to investigate the frequency of Aspergillus species among Iranian patients and their susceptibility to seven antifungals. In a cross-sectional study, 233 Aspergillus isolates were collected from 11 university hospitals in Iran between 2018 and 2021. Aspergillus isolates were identified based on colony morphology, microscopic characteristics, PCR-restriction fragment length polymorphism (RFLP), and sequencing of the beta-tubulin gene. The CLSI M38-A2 reference methodology was used for antifungal susceptibility testing of amphotericin B, voriconazole, posaconazole, itraconazole, luliconazole, isavuconazole, and caspofungin. Members of Aspergillus section Flavi (117/233, 50.2%), Aspergillus section Nigri (77/233, 33.1%), Aspergillus section Fumigati (21/233, 9%), Aspergillus section Terrei (14/233, 6%), Aspergillus pseudodeflectus (2/233, 0.85%), and Aspergillus melleus (2/233, 0.85%) were isolated from the samples. The lowest 0.25 MIC 90 values for all isolates tested were for luliconazole (0.016 μg/mL) and isavuconazole (0.250 μg/mL), and the highest value was observed for itraconazole (≥ 8μg/mL). The 90% minimum effective concentration (MEC 90 ) value for caspofungin was 0.125 μg/mL. MIC 90 values for voriconazole, amphotericin B, and posaconazole were 1, 2, and 2 μg/mL, respectively. The non-wild-type species were presented for amphotericin B (3%), voriconazole (1.3%), posaconazole (2.6%), luliconazole (1.3%), isavuconazole (1.7%), and caspofungin (4.7%). Positive correlations in the MIC values of azole antifungals were observed, and using one azole increases the MIC value rates of other ones. None of the species were pan-azole resistant. Species of Aspergillus section Flavi were the most common Aspergillus species isolated from Iranian samples. Luliconazole, caspofungin, and isavuconazole present the most effective antifungal agents for treatment of infection due to Aspergillus species. Susceptibility tests should be performed frequently in each region for the best management of patients. IMPORTANCE Aspergillus species are the leading cause of invasive aspergillosis in immunocompromised hosts. The susceptibility of Aspergillus species to antifungal agents might be different. Azole-resistant species have emerged worldwide. Performing susceptibility testing in each region can help in the best management of patients. Here, we show the epidemiology and distribution of Aspergillus species in Iran and their susceptibility patterns for seven antifungal agents. The significant points of the present study are that species of Aspergillus section Flavi are the most prevalent Aspergillus species isolated from 11 university hospitals. Luliconazole, caspofungin, and isavuconazole were effective antifungal agents against all Aspergillus species.

Background: This study aimed to assess the risk of work-related musculoskeletal disorders (WMSDs) using the path analysis models., Methods: This study was carried out on 350 office employees with good general health. All variables were collected using a questionnaire. Personality traits and mental workload of employees were evaluated using the NEO Personality Inventory and the NASA-task load index software, respectively. The individual and personality traits were used as predictor variables, and mental workload (MWL) and body posture scores as mediating variables of the musculoskeletal discomforts. The role of predictor and mediating variables on discomforts was explained based on the path analysis models., Results: The impact coefficient of MWL and posture on WMSDs was significant. The coefficient of the direct effect of body mass index (BMI) and gender on musculoskeletal disorders was significant and positive and the women have reported a higher rate of discomforts. The strongest positive impact of personality traits on MWL and posture was conscientiousness, followed by neuroticism and agreeableness. In return, the strongest negative impact was extroversion, followed by openness. The strongest positive impact of individual factors on MWL and posture was BMI, followed by work experience., Conclusion: Gender, BMI, neuroticism, extraversion, and conscientiousness can be strong predictors for musculoskeletal discomforts which can mediate the impact of body posture and mental workload (mediating factors) on musculoskeletal discomfort. Therefore, personality and individual traits can be strong alarming and indicators for risk identification and preventing musculoskeletal disorders when choosing people for a job or task., (© 2022. Background: The potential use of biomarkers in the diagnosis of fungal infections is a challenge. The aim of this study was to evaluate the role of a biomarker-guided antifungal stewardship program for hospitalized pediatrics suffering from invasive aspergillosis (IA)., Methods: Pediatric patients with suspected probable or proven IA were enrolled in this study. Demographic data were collected from their records. Clinical samples were examined by wet mount KOH smear, culture, galactomannan Ag test, and real-time PCR. Patients' sera were evaluated for procalcitonin (PCT) and soluble-triggering receptor expressed on myeloid cells -1 (sTREM-1) levels by ELISA Kits., Results: A total of 73 children were entered in this study with a mean age of 5 years and the male to female ratio 39/34. The most predisposing factors were hematologic disorders (71.2%). The area under the curves (95% confidence interval) for each biomarker were 0.9 (0.85% to 97%) for lactate de hydrogenase (LDH), 0.9 (0.85% to 0.94%) for C-reactive protein, 0.8 (0.75% to 0.84%) for PCT, 0.8 (0.73% to 0.85%) for erythrocyte sedimentation rate, 0.7 (0.6% to 0.8%) for sTREM-1, and 0.5 (0.45% to 0.58%) for white blood cell count. During the study period, 27.4% patients died. The LDH and sTREM-1 levels were significant increase in died patient (p < 0.05)., Conclusions: According to our data, evaluation of biomarkers along with radiologic and clinical signs and symptoms of pediatric patients can lead to proper antifungal therapy and decreased side effects, antifungal resistance, and cost. Congenital hypothyroidism (CH) occurs with a relatively alarming prevalence in infants, and if not diagnosed and treated in time, it can have devastating consequences for the development of the nervous system. CH is associated with genetic changes in several genes that encode transcription factors responsible for thyroid development, including mutations in the NK2 homeobox 1 (NKX2.1) gene, which encodes the thyroid transcription factor-1 (TTF-1). Although CH is frequently observed in pediatric populations, there is still a limited understanding of the genetic factors and molecular mechanisms contributing to this disease. The sequence of the NKX2.1 gene was investigated in 75 pediatric patients with CH by polymerase chain reaction (PCR), single-stranded conformation polymorphism (SSCP), and direct DNA sequencing. Four missense heterozygous variations were identified in exon 3 of the NKX2.1 gene, including three novel missense variations, namely c.708A>G, p.Gln202Arg; c.713T>G, p.Tyr204Asp; c.833T>G, p.Tyr244Asp, and a previously reported variant rs781133468 (c.772C>G, p.His223Gln). Importantly, these variations occur in highly conserved residues of the TTF-1 DNA-binding domain and were predicted by bioinformatics analysis to alter the protein structure, with a probable alteration in the protein function. These results indicate that nucleotide changes in the NKX2.1 gene may contribute to CH pathogenesis.

Background and Aim: Human-robot collaboration is the key component of the fourth industrial revolution concept. Workers' willingness to collaborate with industrial robots is a basic requirement for an efficient and effective interaction. The roles of human-robot trust and technology affinity as mediators in the relationship between robot usability and worker willingness were analyzed in this study. As other critical variables, the mediator roles of Age and STARA were also calculated., Materials and Methods: This study included 400 workers from a car company who interacted with industrial robots in their daily work activities. After examining the questionnaires' validity and reliability, the main variables were determined to be willingness to use robots and robot usability. AMOS software also considered human-robot trust and worker technology affinity as mediators. The bootstrapping method was used to evaluate indirect relationships. A set of goodness-of-fit indices were presented to determine the adequacy of the goodness of fit between the proposed model and the data., Results: Based on model fit indices, an overall satisfactory model fit was obtained for the direct/indirect relationship between robot usability and worker willingness to use it (with mediating role of human-robot trust). Workers' age and fear of Smart Technology, Artificial Intelligence, Robotics, and Algorithms (STARA) were identified as moderators in the relationship between usability and willingness., Conclusion: Attention to the robot usability and the role of workers' trust in robots appears to be required to ensure workers' willingness to use robots and the success of human-robot collaboration in future workplaces. The output beams generated by industrial electron accelerators should be swept over the product. It is necessary to determine the exact scan length in order to ensure that an efficient dose reaches the product and to avoid the loss of the beam in contact with the sides of the product. Another challenge associated with scanning beams is the non-uniformity of the dose across the entire product during irradiation. In this study, the use of beam correction magnets is proposed for correcting products that receive a non-uniform dose. These magnets correct beams that are not swept in parallel. In addition, we simulated the dose received by the products in two non-uniform and uniform modes (modified by beam correction magnets) using the Monte Carlo method, and compared the results to assess the importance of using an appropriate irradiation system. Among the proposed magnets, a modified sector magnet is recommended for correcting the beam path and reducing the side effect. Using this magnet for a long horn does not require a cooling system, but using it for a short horn increases the need for a water-cooling system., (Copyright © 2022 Elsevier Ltd. In order to decipher the complex molecular mechanisms that regulate the assembly and disassembly of actin filaments, it is a great asset to monitor individual reactions live in well-controlled conditions. To do so, live single-filament experiments have emerged over the past 20 years, mostly using total internal reflection fluorescence (TIRF) microscopy, and have provided a trove of key results. In 2011, in order to further expand the possibilities of these experiments and to avoid recurring problematic artifacts, we introduced simple microfluidics in these assays. This study details our basic protocol, where individual actin filaments are anchored by one end to the passivated coverslip surface, align with the flow, and can be successively exposed to different protein solutions. We also present the protocols for specific applications and explain how controlled mechanical forces can be applied, thanks to the viscous drag of the flowing solution. We highlight the technical caveats of these experiments and briefly present possible developments based on this technique. These protocols and explanations, along with today's availability of easy-to-use microfluidics equipment, should allow non-specialists to implement this assay in their labs.

Graphene-based supercapacitors demonstrate extraordinary energy storage capacity due to their layered structure, large effective surface area, high electrical conductivity and acceptable chemical stability. Herein, reduced graphene oxide (rGO)-based supercapacitors were introduced in a simple, and fast method with considerable performance. For this purpose, graphene oxide (GO) was synthesized by the modified Hummers' method and then easily reduced to desired patterns of rGO using a commercial LightScribe DVD drive. In order to increase the effective surface area, as well as the electrical conductivity of rGO layers, oxygen/sulfur hexafluoride plasma was applied to the rGO followed by laser irradiation. By performing such sequential processes, an rGO-based supercapacitor was introduced with a capacitance of about 10.2 F/cm 3 , which had high stability for more than 1000 consecutive charge-discharge cycles. The fabrication steps of the electrodes were investigated by different analyses such as SEM, TEM, Raman, surface resistance, BET, and XPS measurements. The study investigated the effect of buprenorphine (BUP) on oxidative indices and gene expression of apoptotic molecules in the hippocampus of neonates during the fetal stage. BUP (1 or 0.5 mg/kg) was subcutaneously administrated to pregnant rat dams. After parturition, the pups were maintained to the end of breastfeeding period, then hippocampi were assessed for oxidative stress indices [glutathione (GSH), thiobarbituric acid reactive substances (TBARS), superoxide dismutase (SOD), total antioxidant capacity (TAC)] and mRNA expression of apoptotic markers (Bax, Bcl2 and caspase 3). Our data indicated that BUP (0.5 mg/kg) administration during gestation significantly increased GSH and TAC concentrations in the hippocampus of pups versus control group (p < 0.05). BUP (0.5 and 1 mg/kg) administration significantly elevated the expression levels of Bcl2 in the hippocampus of neonates compared with controls. BUP injection (0.5 and 1 mg/kg) to pregnant rats markedly reduced the expression levels of caspase 3 in the hippocampus of neonates in BUP 0.5 group (p < 0.01) and BUP 1 group (p < 0.05) versus the controls. Purpose. This study aimed to analyze causal relationships among safety climate dimensions, safety performance dimensions and occupational accidents for nurses working in hospitals. Methods. Data were gathered from questionnaires filled in by nurses from three public hospitals. To analyze interactions among variables, a Bayesian network (BN) analysis was conducted. Using the function of BN 'belief updating', variables with the highest influences on occupational accidents were determined. Results . A total of 211 nurses participated in this study. They were mainly female and married. The results showed that 39.3% of nurses experienced occupational accidents during the last 12 months before the study. Less than half of the nurses had an acceptable safety performance. Safety participation had the highest influence on occupational accidents, followed by safety compliance. Reporting of errors had the highest score among the safety climate dimensions. Training of nurses was necessary for improving both safety compliance and safety participation, and thereby reducing occupational accidents. Conclusion. The effect of safety participation on occupational accidents among nurses was higher than that of safety compliance. Among the safety climate dimensions, supervisors' attitude to safety and safety training had the highest effects on both nurses' safety performance and occupational accidents.

Medication errors can endanger the health and safety of patients and need to be managed appropriately. This study aimed at developing a new and comprehensive method for estimating the probability of medication errors in hospitals. An extensive literature review was conducted to identify factors affecting medication errors. Success Likelihood Index Methodology was employed for calculating the probability of medication errors. For weighting and rating of factors, the Fuzzy multiple attributive group decision making methodology and Fuzzy analytical hierarchical process were used, respectively. A case study in an emergency department was conducted using the framework. A total number of 17 factors affecting medication error were identified. Workload, patient safety climate, and fatigue were the most important ones. The case study showed that subtasks requiring nurses to read the handwritten of other nurses and physicians are more prone to human error. As there is no specific method for assessing the risk of medication errors, the framework developed in this study can be very useful in this regard. Tight control of gene expression networks required for adipose tissue formation and plasticity is essential for adaptation to energy needs and environmental cues. However, the mechanisms that orchestrate the global and dramatic transcriptional changes leading to adipocyte differentiation remain to be fully unraveled. We investigated the regulation of nascent transcription by the sumoylation pathway during adipocyte differentiation using SLAMseq and ChIPseq. We discovered that the sumoylation pathway has a dual function in differentiation; it supports the initial downregulation of pre-adipocyte-specific genes, while it promotes the establishment of the mature adipocyte transcriptional program. By characterizing endogenous sumoylome dynamics in differentiating adipocytes by mass spectrometry, we found that sumoylation of specific transcription factors like PPARγ/RXR and their co-factors are associated with the transcription of adipogenic genes. Finally, using RXR as a model, we found that sumoylation may regulate adipogenic transcription by supporting the chromatin occurrence of transcription factors. Our data demonstrate that the sumoylation pathway supports the rewiring of transcriptional networks required for formation of functional adipocytes. Background: Betatrophin may be associated with metabolic diseases., Objective: To investigate the betatrophin level and its association with metabolic and inflammatory parameters in infertile women with polycystic ovary syndrome (PCOS) and other infertile women during the intrauterine insemination cycle., Materials and Methods: This case-control study was conducted with 90 infertile women (45 with PCOS and 45 without) chosen by convenience sampling, in the infertility clinic of Imam Khomeini Hospital in Tehran, Iran. Participants were interviewed to obtain their age, body mass index, and reproductive history. Fasting brachial venous blood samples were obtained on the 3 rd day of the menstrual cycle to measure the levels of betatrophin, fasting blood sugar, insulin, luteinizing hormone, follicle-stimulating hormone, low-density lipoprotein cholesterol, estradiol, and high-sensitivity C-reactive protein., Results: The results showed that the level of betatrophin in women with PCOS was significantly higher than in the control group (p = 0.05). Based on multiple linear regression analyses, the effects of metabolic and inflammatory parameters on betatrophin were not significant (p = 0.19). The results showed no significant difference between groups in folliculogenesis (p = 0.57)., Conclusion: According to the results, betatrophin levels were higher in infertile women with PCOS than in those without. The findings suggest that there may be an association between increased betatrophin and increased incidence of PCOS. Background: MicroRNAs have short sequences of 20 ~ 25-nucleotides which are similar among family members and play crucial regulatory roles in numerous biological processes, such as in cell development, metabolism, proliferation, differentiation, and apoptosis., Results: We reported a strategy for the construction of a dual-emission fluorescent sensor using carbon dots (CDs) and confirmed their applications for ratiometric microRNA-21 sensing and bioimaging of cancer cells in a microfluidic device. The composition of blue CDs (B-CDs) and yellow CDs (Y-CDs) depicts dual-emission behavior which is centered at 409 and 543 nm under an excitation wavelength of 360 nm. With increasing microRNA-21 concentration, the robust and specific binding of DNA probe functionalized B-CDs to complementary microRNA-21 target induced perturbations of probe structure and led to changing fluorescence intensity in both wavelengths. Consequently, the ratio of turn-on signal to turn-off signal is greatly altered. With monitoring of the inherent ratiometric fluorescence variation (ΔF 540nm /ΔF 410nm ), as-prepared BY-CDs were established as an efficient platform for ratiometric fluorescent microRNA-21 sensing, with a wide linear range of 0.15 fM to 2.46 pM and a detection limit of 50 aM., Conclusions: Furthermore, the proposed assay was applied for detecting microRNA-21 in dilute human serum samples with satisfactory recovery and also in MCF-7 cell lines in the range 3000 to 45,000 (cell mL -1 ) with a detection limit (3 cells in 10 μL), demonstrating the potential of the assay for clinic diagnosis of microRNA-associated disease. More importantly, the images revealed that MCF-7 cells well labeled with BY-CDs could exhibit the applicability of the proposed microfluidic system as an effective cell trapping device in bioimaging., (© 2022. Objective: The present study aimed to evaluate the antidiabetic effects of glycinium [(pyridine-2, 6-dicarboxylato) oxovanadate (V)] complex in type 2 diabetes rat model., Materials and Methods: Rats were allocated into 6 groups. Group I, nondiabetic rats; Group II, diabetic rats; Group III, diabetic rats receiving an intraperitoneal (i.p.) injection of metformin (45 mg/kg); Groups IV, V and VI were diabetic rats receiving i.p. injection of 5, 10, and 20 mg/kg of the complex for 3 weeks, respectively. Fasting blood glucose (FBG), insulin, liver enzymes, malondialdehyde (MDA), total antioxidant capacity (TAC), lipid profile, and HbA1c were measured., Results: AST, ALT and GGT activities and MDA levels were increased, while TAC was decreased in diabetic animals. Treatment of diabetic rats improved the HOMA-IR and returned HbA1c level to the normal value as well as elevated TAC and reduced MDA level., Conclusion: We found that the complex possesses antidiabetic properties in experimental diabetes.

Background and Purpose: In vitro development of functional gametes from pluripotent stem cells is a promising prospect to treat infertility. Mesenchymal stem cells with a high degree of plasticity and less tumorigenicity are a reliable source of stem cells for the generation of gametes. The present study aimed to compare the differentiation potential in the mesenchymal stem cells that are derived from bone marrow (BMDMSCs) and adipose tissue derived mesenchymal stem cells (AD-MSCs) into germ cells in a culture medium containing bone morphogenic protein-4 (BMP-4)., Experimental Approach: In this study, MSCs were isolated from both bone marrow and adipose tissue of murine samples. To further verify the nature of the harvested stem cells, their multipotency and surface marker were examined. The identified stem cells were cultured in a medium supplemented with 0 and 25 ng/mL of BMP-4 for 4 days. Flow cytometry analysis, immunofluorescence staining, and real RT-PCR were used to assess the expression levels in germ cell-specific biomarkers (Mvh, Dazl, Stra8, and Scp3)., Findings/results: CD44+, CD45-, CD31-, BMD-MSCs, and AD-MSCs showed to be capable of differentiating to osteo-adipogenic lineages. The flow cytometry, immunofluorescence, and RT-PCR results indicated that early germ cell markers (Mvh and Dazl) were expressed in both types of cells but they were significantly higher in BMD-MSCs than AD-MSCs., Conclusion and Implications: Based on our results, the addition of exogenous BMP4 to the culture medium could differentiate BMD-MSCs and AD-MSCs into primordial germ cells, but it is inadequate to further develop into late germ cells in vitro . Moreover, the results revealed that, although AD-MSCs were easier to collect and had faster growth and proliferation rates than BMD-MSCs, the BMD-MSCs were better capable of differentiation into primordial germ cells.
