Abstract: Humans and many other animals use acoustical signals to mediate social interactions with conspecifics. The evolution of sound-based communication is still poorly understood and its neural correlates have only recently begun to be investigated. In the present study, we applied functional MRI to humans and macaque monkeys listening to identical stimuli in order to compare the cortical networks involved in the processing of vocalizations. At the first stages of auditory processing, both species showed similar fMRI activity maps within and around the lateral sulcus (the Sylvian fissure in humans). Monkeys showed remarkably similar responses to monkey calls and to human vocal sounds (speech or otherwise), mainly in the lateral sulcus and the adjacent superior temporal gyrus (STG). In contrast, a preference for human vocalizations and especially for speech was observed in the human STG and superior temporal sulcus (STS). The STS and Broca''s region were especially responsive to intelligible utterances. The evolution of the language faculty in humans appears to have recruited most of the STS. It may be that in monkeys, a much simpler repertoire of vocalizations requires less involvement of this temporal territory. [Copyright &y& Elsevier]

In vivo optical imaging is being conducted in a variety of medical applications, including optical breast cancer imaging, functional brain imaging, endoscopy, exercise medicine, and monitoring the photodynamic therapy and progress of neoadjuvant chemotherapy. In the past three decades, in vivo diffuse optical breast cancer imaging has shown promising results in cancer detection, and monitoring the progress of neoadjuvant chemotherapy. The use of near infrared spectroscopy for functional brain imaging has been growing rapidly. In fluorescence imaging, the difference between autofluorescence of cancer lesions compared to normal tissues were used in endoscopy to distinguish malignant lesions from normal tissue or inflammation and in determining the boarders of cancer lesions in surgery. Recent advances in drugs targeting specific tumor receptors, such as AntiBodies (MAB), has created a new demand for developing non-invasive in vivo imaging techniques for detection of cancer biomarkers, and for monitoring their down regulations during therapy. Targeted treatments, combined with new imaging techniques, are expected to potentially result in new imaging and treatment paradigms in cancer therapy. Similar approaches can potentially be applied for the characterization of other disease-related biomarkers. In this chapter, we provide a review of diffuse optical and fluorescence imaging techniques with their application in functional brain imaging and cancer diagnosis. [ABSTRACT FROM AUTHOR]

Abstract: This letter shows a novel computer aided diagnosis (CAD) system for the early diagnosis of Alzheimer’s Disease (AD). The proposed method evaluates the reliability of association rules (ARs) aiming to discover interesting associations between attributes in functional brain imaging, i.e. single photon emission computed tomography (SPECT) and positron emission tomography (PET). AR mining firstly requires a masking process for reducing the computational cost, which is based on Fisher discriminant ratio (FDR), in order to identify “transactions” or relationships among discriminant brain areas. Once the activation map is achieved by means of activation estimation (AE), the resulting regions of interest (ROIs) are subjected to AR discovery with a specified minimum support and confidence. Finally, the proposed CAD system performs image classification by evaluating the number of previously mined rules from controls that are verified by each subject. Several experiments were carried out on two different image modalities (SPECT and PET) in order to highlight the generalization ability of the proposed method. The AR-based method yields an accuracy up to 92.78% (with 87.5% sensitivity and 100% specificity) and 91.33% (with 82.67% sensitivity and 100% specificity) for SPECT and PET, respectively, thus outperforming recently developed methods for early diagnosis of AD. Abstract: The study aims to investigate the effect of cerebral ischemia or hypoperfusion in the evaluation of neural activity with blood-oxygen-level dependent (BOLD) functional magnetic resonance imaging (fMRI), and to examine whether the severity of the compromised hemodynamic status in patients with major cerebral artery diseases could, conversely, be assessed with the use of neural activity as endogenous vasodilator. 28 neurological impairment-free patients with anterior-circulation-territory ischemia performed a bimanual hand-grasping task. Magnitude and temporal shift of evoked BOLD response, baseline cerebral blood flow (CBF) and its increment, and the severity of hemodynamic impairment stratified by blood flow pattern were evaluated. For fMRI data, both conventional analysis with a canonical HRF and an HRF-model-free analysis were performed. The severity of hemodynamic impairment was significantly correlated (p<0.0001) with baseline CBF, CBF increment, and magnitude and delay of BOLD response. BOLD response delay was also significantly correlated (p<0.0001) with baseline CBF, CBF increment, and response magnitude. In 10 out of 45 ischemic motor cortices, conventional analysis completely failed to detect areas of activation that were demonstrated by HRF-model-free analysis. These data suggest that delay and reduced magnitude of BOLD response can be an indicator of the severity of compromised hemodynamic status, and that reduced regional baseline CBF and its increment underlie impaired BOLD response, which necessitates an alternative approach to conventional analysis with any single HRF. Abstract: Placebo analgesia is a psychosocial context effect that is rarely studied in visceral pain. Patients with irritable bowel syndrome (IBS) exhibit visceral hyperalgesia and heightened affective/cognitive brain region activation during visceral stimuli. Psychological factors alter the pain and brain activation pattern, and these changes are more pronounced in IBS patients. Expectation constitutes the major neuropsychological mechanism in the placebo effect. This study confirmed the heightened affective/cognitive brain responses in IBS patients during visceral placebo analgesia using a placebo model with expectation, which was enhanced by suggestion and conditioning. Seventeen IBS patients and 17 age-/sex-matched controls were enrolled. Psychophysical inventories (Hospital Anxiety and Depression Scale [HADS], visual analogue scale, and short-form McGill questionnaire) were completed. Brain activity during placebo intervention and anticipation was assessed in response to rectal distension using 3T-functional magnetic resonance imaging. Suggestion-/conditioning-enhanced placebo was used to convince controls/patients of the efficacy of a newly developed intravenous drug (saline, in actuality) for the relief of rectal distension-induced visceral pain. A comparable visceral placebo analgesia was observed in IBS patients and control subjects. IBS patients demonstrated a higher HADS-anxiety score, which was predictive of a weak placebo effect. Suggestion-/conditioning-enhanced placebo evoked more activity in affective/cognitive brain regions (insula, midcingulate cortex, and ventrolateral prefrontal cortex [VLPFC]) in IBS patients than in healthy controls. VLPFC was also more active during anticipation in IBS patients. In conclusion, IBS patients and control subjects achieved comparable placebo analgesia during experimentally induced rectal pain. Abstract: Background: Mitochondrial myopathy, encephalopathy, lactic acidosis and stroke-like episodes (MELAS) is the most common type of mitochondrial disease and is characterized by stroke-like episodes (SEs), myopathy, lactic acidosis, diabetes mellitus, hearing-loss and cardiomyopathy. The causal hypotheses for SEs in MELAS presented to date are angiopathy, cytopathy and neuronal hyperexcitability. L-arginine (Arg) has been applied for the therapy in MELAS patients. Scope of review: We will introduce novel in vivo functional brain imaging techniques such as MRI and PET, and discuss the pathogenesis of SEs in MELAS patients. We will further describe here our clinical experience with L-arg therapy and discuss the dual pharmaceutical effects of this drug on MELAS. Major conclusions: Administration of L-arg to MELAS patients has been successful in reducing neurological symptoms due to acute strokes and preventing recurrences of SEs in the chronic phase. L-Arg has dual pharmaceutical effects on both angiopathy and cytopathy in MELAS. General significance: In vivo functional brain imaging promotes a better understanding of the pathogenesis and potential therapies for MELAS patients. Abstract: The N-methyl-d-aspartate receptor (NMDAR) plays a critical role in glutamatergically mediated neurotransmission, and thereby affects a wide range of brain circuits and important behavioral functions, not limited to learning and memory. While glutamate is classically considered to be the brain''s principal excitatory neurotransmitter, there is also evidence the NMDAR plays a “functionally inhibitory” role, not in that it directly hyperpolarizes neurons but rather it suppresses circuit level neural activity, including through activation of GABAergic interneurons. This paper reviews data on the NMDAR “suppression” hypothesis (while also examining circuit level activation), with a focus on the following 6 lines of evidence: (1) epilepsy studies, (2) neurotoxicity studies, (3) mouse knockout studies of particular receptor subunits, (4) electrophysiological studies, (5) 2-deoxyglucose studies, and (6) functional brain imaging studies. For many of these lines of evidence, the review focuses on data from two well-characterized NMDAR antagonists, ketamine and phencyclidine. Also, evidence regarding the NMDAR and schizophrenia, including the psychotomimetic properties of ketamine and phencyclidine, cuts across several of the lines of evidence. The data suggest the NMDAR participates in activation, as well as widespread suppression, of circuit level neural activity, where the suppression may be particularly prominent in limbic circuits. Abstract: Patients with central pain (CP) typically have chronic pain within an area of reduced pain and temperature sensation, suggesting an impairment of endogenous pain modulation mechanisms. We tested the hypothesis that some brain structures normally activated by cutaneous heat stimulation would be hyperresponsive among patients with CP but not among patients with a central nervous system lesion causing a loss of heat or nociceptive sensation with no pain (NP). We used H2 15O positron emission tomography to measure, in 15 healthy control participants, 10 NP patients, and 10 CP patients, increases in regional cerebral blood flow among volumes of interest (VOI) from the resting (no stimulus) condition during bilateral contact heat stimulation at heat detection, heat pain threshold, and heat pain tolerance levels. Both patient groups had a reduced perception of heat intensity and unpleasantness on the clinically affected side and a bilateral impairment of heat detection. Compared with the HC group, both NP and CP patients had more hyperactive and hypoactive VOI in the resting state and more hyperresponsive and hyporesponsive VOI during heat stimulation. Compared with NP patients, CP patients had more hyperresponsive VOI in the intralaminar thalamus and sensory–motor cortex during heat stimulation. Our results show that focal CNS lesions produce bilateral sensory deficits and widespread changes in the nociceptive excitability of the brain. The increased nociceptive excitability within the intralaminar thalamus and sensory–motor cortex of our sample of CP patients suggests an underlying pathophysiology for the pain in some central pain syndromes. This paper introduces the idea of drawing upon the cognitive neuroscience literature to inform IS research (herein termed "NeuroIS"). Recent advances in cognitive neuroscience are uncovering the neural bases of cognitive, emotional, and social processes, and they offer new insights into the complex interplay between IT and information processing, decision making, and behavior among people, organizations, and markets. The paper reviews the emerging cognitive neuroscience literature to propose a set of seven opportunities that IS researchers can use to inform IS phenomena, namely (1) localizing the neural correlates of IS constructs, (2) capturing hidden mental processes, (3) complementing existing sources of IS data with brain data, (4) identifying antecedents of IS constructs, (5) testing consequences of IS constructs, (6) inferring the temporal ordering among IS constructs, and (7) challenging assumptions and enhancing IS theories. The paper proposes a framework for exploring the potential of cognitive neuroscience for IS research and offers examples of potentially fertile intersections of cognitive neuroscience and IS research in the domains of design science and human-computer interaction. This is followed by an example NeuroIS study in the context of e-commerce adoption using fMRI, which spawns interesting new insights. The challenges of using functional neuroimaging tools are also discussed. Conclusion. We have demonstrated differences in cortical activation with language-related visual stimuli in patients who were profoundly deafened due to genetic mutations in GJB2 and SLC26A4. The differences in cortical processing patterns between these two cases may have been influenced by the differing clinical courses and pathogenesis of hearing loss due to genetic mutations. Our results suggest the importance of hearing during early childhood for the development of a normal cortical language network. Objectives. To investigate the cortical activation with language-related visual stimuli in patients who were profoundly deafened due to genetic mutations in GJB2 and SLC26A4. Methods: The cortical activity of two adult patients with known genetic mutations ( GJB2, SLC26A4) was evaluated with fluorodeoxyglucose-positron emission tomography (FDG-PET) with a visual language task and compared with that of normal-hearing controls. Results: A patient with a GJB2 mutation showed activation in the right auditory association area [BA21, BA22], and the left auditory association area [BA42] even with visual language task; in contrast, a patient with an SLC26A4 mutation showed no significant activation in the corresponding area. Spatiotemporal brain activation profiles were obtained from 27 middle school students experiencing difficulties in reading comprehension as well as word-level skills (RD) and 23 age- and IQ-matched non-reading impaired students during performance of an oral pseudoword reading task using Magnetoencephalography (MEG). Based on their scores on standardized reading fluency tests 1 year later, students with RD who showed significant improvement were classified as Adequate Responders (AR) whereas those not demonstrating such gains were classified as Inadequate Responders (IR). At baseline, activation profiles of the AR group featured increased activity in the left supramarginal and angular gyri, as well as in the superior and middle temporal gyri, bilaterally compared to IR. The degree of activity in these regions was a significant predictor of the amount of subsequent gains in reading fluency. These results extend previous functional brain imaging findings of beginning readers, suggesting that recruitment of brain areas that typically serve as key components of the brain circuit for reading is an important factor in determining response to intervention in older struggling readers. The differential diagnosis between disorders of consciousness (DoC), on the one hand, and conditions of severe immobility with aphasia, on the other hand, is extremely difficult. A patient is described whose morphology of brain lesions permitted both kinds of disorder. A battery of functional magnetic resonance imaging (fMRI) examinations was applied indicating that severe DoC was implausible. The following clinical course confirmed this conclusion, resulting in a severe tetraparesis/aphasia, however with clear awareness. The data suggest that fMRI can be very useful in differentiating between severe motor disorders, including a locked-in syndrome, and DoC such as the vegetative state and the minimally conscious state. Rationale: Working memory dysfunction is frequently observed in schizophrenia. The neural mechanisms underlying this dysfunction remain unclear, with functional neuroimaging studies reporting increased, decreased or unchanged activation compared to controls. Objectives: We investigated the neural correlates of spatial working memory in schizophrenia with particular consideration of effects of antipsychotic treatment and relation to performance levels in the patient group. Method: We used functional magnetic resonance imaging and studied the blood-oxygen-level-dependent (BOLD) response of 45 schizophrenia outpatients and 19 healthy controls during a parametric spatial n-back task. Results: Performance in both groups deteriorated with increasing memory load (0-back, 1-back, 2-back), but the two groups did not significantly differ in performance overall or as a function of load. Patients produced stronger BOLD signal in occipital and lateral prefrontal cortex during task performance than controls. This difference increased with increasing working memory load in the prefrontal areas. We also found that in patients with good task performance, the BOLD response in left prefrontal cortex showed a stronger parametric increase with working memory load than in patients with poor performance. Second-generation antipsychotics were independently associated with left prefrontal BOLD increase in response to working memory load, whereas first-generation antipsychotics were associated with BOLD decrease with increasing load in this area. Conclusions: Together, these findings suggest that in schizophrenia patients, normal working memory task performance may be achieved through compensatory neural activity, especially in well-performing patients and in those treated with second-generation antipsychotics. Abstract: When several scattered grating elements are arranged in such a way that their directions of motion are consistent with a common path, observers perceive them as belonging to a globally coherent moving object. Here we investigated how this coherence changes the representation of motion signals in human visual cortex using functional magnetic resonance imaging (fMRI) and multivariate voxel pattern decoding, which have the potential to reveal how well a stimulus is encoded in different contexts. Only during globally coherent motion was it possible to reliably distinguish fMRI signals evoked by different directions of motion in early visual cortex. This effect was specific to the retinotopic representation of the visual field quadrant in V1 traversed by the coherent element path and could not simply be attributed to a general increase in signal strength. Decoding was more reliable for cortical areas corresponding to the lower visual field. Because some previous studies observed poorer speed discrimination when motion was grouped, we also conducted behavioural experiments to investigate this with our stimuli, but did not reveal a consistent relationship between coherence and perceived speed. Taken together, these data show that neuronal populations in early visual cortex represent information that could be used for interpreting motion signals as unified objects. [Copyright &y& Elsevier]

Older adults, compared to younger adults, focus on emotional well-being. While the lifespan trajectory of emotional processing and its regulation has been characterized behaviorally, few studies have investigated the underlying neural mechanisms. Here, older adults (range: 59–73 years) and younger adults (range: 19–33 years) participated in a cognitive reappraisal task during functional magnetic resonance imaging (fMRI) scanning. On each trial, participants viewed positive, negative or neutral pictures and either naturally experienced the image (‘Experience’ condition) or attempted to detach themselves from the image (‘Reappraise’ condition). Across both age groups, cognitive reappraisal activated prefrontal regions similar to those reported in prior studies of emotion regulation, while emotional experience activated the bilateral amygdala. Psychophysiological interaction analyses revealed that the left inferior frontal gyrus (IFG) and amygdala demonstrated greater inverse connectivity during the ‘Reappraise’ condition relative to the ‘Experience’ condition. The only regions exhibiting significant age differences were the left IFG and the left superior temporal gyrus, for which greater regulation-related activation was observed in younger adults. Controlling for age, increased performance on measures of cognition predicted greater regulation-related decreases in amygdala activation. Thus, while older and younger adults use similar brain structures for emotion regulation and experience, the functional efficacy of those structures depends on underlying cognitive ability. Abstract: Background: A consistent brain activity pattern has been identified in major depression across many resting positron emission tomography (PET) studies. This dysfunctional pattern seems to be normalized by antidepressant treatment. The aim of this meta-analysis was to identify more clearly the pattern associated with clinical improvement of depression following an antidepressant drug treatment, in emotional activation studies using functional magnetic resonance imaging (fMRI). Methods: A quantitative Activation Likelihood Estimation (ALE) meta-analysis was performed across 9 emotional activation fMRI and PET studies (126 patients) using the Activation Likelihood Estimation technique. Results: Following the antidepressant drug treatment, the activation of dorsolateral, dorsomedial and ventrolateral prefrontal cortices was increased whereas the activation of the amygdala, hippocampus, parahippocampal region, ventral anterior cingulate cortex, orbitofrontal cortex, and insula was decreased. Additionally, there was a decreased activation in the anterior (BA 32) and posterior cingulate cortices, as well as in the precuneus and inferior parietal lobule, which could reflect a restored deactivation of the default mode network. Limitations: The small number of emotional activation studies, using heterogeneous tasks, included in the ALE analysis. Conclusions: The activation of several brain regions involved in major depression, in response to emotional stimuli, was normalized after antidepressant treatment. To refine our knowledge of antidepressants'' effect on the neural bases of emotional processing in major depression, neuroimaging studies should use consistent emotional tasks related to depressive symptoms and that involve the default mode network, such as self-referential processing tasks. Abstract: This work proposes to use magnetoencephalography (MEG) and electroencephalography (EEG) source imaging to provide cinematic representations of the temporal dynamics of cortical activations. Cortical activation maps, seen as images of the active brain, are scalar maps defined at the vertices of a triangulated cortical surface. They can be computed from M/EEG data using a linear inverse solver every millisecond. Taking as input these activation maps and exploiting both the graph structure of the cortical mesh and the high sampling rate of M/EEG recordings, neural activations are tracked over time using an efficient graph cut based algorithm. The method estimates the spatiotemporal support of the active brain regions. It consists in computing a minimum cut on a particularly designed weighted graph imposing spatiotemporal regularity constraints on the activation patterns. Each node of the graph is assigned a label (active or non active). The method works globally on the full time-period of interest, can cope with spatially extended active regions and allows the active domain to exhibit topology changes over time. The algorithm is illustrated and validated on synthetic data. Results of the method are provided on two MEG cognitive experiments in the visual and somatosensory cortices, demonstrating the ability of the algorithm to handle various types of data. Recent advances in the development of high-throughput genotyping platforms allow for the unbiased identification of genes and genomic sequences related to heritable traits. In this study, we analyzed human short-term memory, which refers to the ability to remember information over a brief period of time and which has been found disturbed in many neuropsychiatric conditions, including schizophrenia and depression. We performed a genome-wide survey at 909 622 polymorphic loci and report six genetic variations significantly associated with human short-term memory performance after genome-wide correction for multiple comparisons. A polymorphism within SCN1A (encoding the α subunit of the type I voltage-gated sodium channel) was replicated in three independent populations of 1699 individuals. Functional magnetic resonance imaging during an n-back working memory task detected SCN1A allele-dependent activation differences in brain regions typically involved in working memory processes. BackgroundCognitive behavioral therapy (CBT) with exposure and response prevention (ERP) is the psychotherapeutic treatment of choice for obsessive–compulsive disorder (OCD). However, little is known about the impact of CBT on frontostriatal dysfunctioning, known to be the neuronal correlate of OCD.MethodA probabilistic reversal learning (RL) task probing adaptive strategy switching capabilities was used in 10 unmedicated patients with OCD and 10 healthy controls during an event-related functional magnetic resonance imaging (fMRI) experiment. Patients were scanned before and after intensive CBT, controls twice at comparable intervals.ResultsStrategy change within the RL task involved activity in a broad frontal network in patients and controls. No significant differences between the groups or in group by time interactions were detected in a whole-brain analysis corrected for multiple comparisons. However, a reanalysis with a more lenient threshold revealed decreased responsiveness of the orbitofrontal cortex and right putamen during strategy change before treatment in patients compared with healthy subjects. A group by time effect was found in the caudate nucleus, demonstrating increased activity for patients over the course of time. Patients with greater clinical improvement, reflected by greater reductions in Yale–Brown Obsessive Compulsive Scale (YBOCS) scores, showed more stable activation in the pallidum.ConclusionsAlthough these findings are preliminary and need to be replicated in larger samples, they indicate a possible influence of psychotherapy on brain activity in core regions that have been shown to be directly involved both in acquisition of behavioral rules and stereotypes and in the pathophysiology of OCD, the caudate nucleus and the pallidum. Abstract: This is a revision of guidelines, originally published in 2004, for the assessment of patients with neuropathic pain. Neuropathic pain is defined as pain arising as a direct consequence of a lesion or disease affecting the somatosensory system either at peripheral or central level. Screening questionnaires are suitable for identifying potential patients with neuropathic pain, but further validation of them is needed for epidemiological purposes. Clinical examination, including accurate sensory examination, is the basis of neuropathic pain diagnosis. For more accurate sensory profiling, quantitative sensory testing is recommended for selected cases in clinic, including the diagnosis of small fiber neuropathies and for research purposes. Measurement of trigeminal reflexes mediated by A-beta fibers can be used to differentiate symptomatic trigeminal neuralgia from classical trigeminal neuralgia. Measurement of laser-evoked potentials is useful for assessing function of the A-delta fiber pathways in patients with neuropathic pain. Functional brain imaging is not currently useful for individual patients in clinical practice, but is an interesting research tool. Skin biopsy to measure the intraepidermal nerve fiber density should be performed in patients with clinical signs of small fiber dysfunction. The intensity of pain and treatment effect (both in clinic and trials) should be assessed with numerical rating scale or visual analog scale. For future neuropathic pain trials, pain relief scales, patient and clinician global impression of change, the proportion of responders (50% and 30% pain relief), validated neuropathic pain quality measures and assessment of sleep, mood, functional capacity and quality of life are recommended. A robust modification of echo-planar imaging dubbed double-shot echo-planar imaging with center-out trajectories and intrinsic navigation (DEPICTING) is proposed, which permits imaging at ultra-short echo time. The k-space data is sampled by two center-out trajectories with a minimal delay achieving a temporal efficiency similar to conventional single-shot echo-planar imaging. Intersegment phase and intensity imperfections are corrected by exploiting the intrinsic navigator information from both central lines, which are subsequently averaged for image reconstruction. Phase errors induced by inhomogeneities of the main magnetic field are corrected in k-space, recovering the superior point-spread function achieved with center-out trajectories. Abstract: Hypnosis modulates pain perception but the associated brain mechanisms in chronic pain conditions are poorly understood. Brain activity evoked by painful repetitive pin-prick stimulation of the left mental nerve region was investigated with use of fMRI in 19 patients with painful temporomandibular disorders (TMD) during hypnotic hypoalgesia and hyperalgesia and a control condition. Pain intensity and unpleasantness of the painful stimulation was scored on a 0–10 Numerical Rating Scale (NRS). NRS pain and unpleasantness scores during hypnotic hypoalgesia were significantly lower than in the control condition and significantly higher in the hypnotic hyperalgesia condition. In the control condition, painful stimulation caused significant activation of right posterior insula, primary somatosensory cortex (SI), BA21, and BA6, and left BA40 and BA4. Painful stimulation during hypnotic hyperalgesia was associated with increased activity in right posterior insula and BA6 and left BA40 whereas hypnotic hypoalgesia only was associated with activity in right posterior insula. Unexpectedly, direct contrasts between control and hypnotic hyperalgesia conditions revealed significant decreases in S1 during hyperalgesia. Direct contrasts between control and hypnotic hypoalgesia conditions demonstrated significant decreases in right posterior insula and BA21, as well as left BA40 during hypoalgesia. These findings are the first to describe hypnotic modulation of brain activity associated with nociceptive processing in chronic TMD pain patients and demonstrate that hypnotic hypoalgesia is associated with a pronounced suppression of cortical activity and a disconnection between patient-based scores and cortical activity in S1 during hypnotic hyperalgesia. Abstract: We have previously used fluorodeoxyglucose (FDG) autoradiography to detect the pattern of metabolic declines in two different transgenic mouse models of fibrillar beta-amyloid pathology in Alzheimer''s disease (AD), including the PDAPP mouse, which overexpresses a mutant form of human APP, and the PSAPP mouse, which overexpresses mutant forms of the human APP and PS1 genes. In this study, we used the same approach to study a triple-transgenic (3xTG) model of AD, which overexpresses human APP, PS1 and tau mutations, and progressively develops amyloid plaques, neurofibrillary tangles, and synaptic dysfunction. Densitometric measurements from 55 brain regions were characterized and compared in 2, 12, and 18month-old 3xTG and wildtype control mice (n=12/group). By 18months of age, the 3xTG mice had significant reductions in FDG uptake in every measured brain region, including cortical and subcortical gray matter, cerebellar and brainstem regions. However, regional differences in normalized FDG uptake were apparent in the 2- and 12-month-old 3xTG mice, in a brain network pattern reminiscent of our previous analyses in the other mouse models. This prominently included the posterior cingulate/retrosplenial cortex, as in each previously-analyzed model. Overall, our analyses highlight consistencies in brain glucose uptake abnormalities across multiple mouse models of amyloid-associated pathophysiology. These mouse brain regional changes are homologous to alterations seen in PET scans from human AD patients and could thus be useful biomarkers for early testing of novel interventions. Fowler’s syndrome—chronic urinary retention and impaired bladder filling sensation without the presence of neurological disease—is a common cause of urinary retention in young women that responds well to treatment with sacral neuromodulation. The mechanisms of disease and therapy have remained difficult to understand, but two recent brain imaging studies—reviewed in this article—have helped dispel the obscurity. Despite different methodology, the two studies concur that brain responses to bladder filling are abnormal and are partially normalized after treatment, with restoration of voiding ability and sensation. The abnormal responses suggest exaggerated pro-continence behavior due to excessive afferent signals from an overactive urethra that inhibit bladder afferent signals at the sacral level. Historically, vision research in China was one of a few distinct research programs within the Chinese Academy of Sciences (CAS). With improved funding opportunities and research environment in neuroscience, vision research at several research institutes within the academy has made significant progress not only in the quantity of publications, but also in the quality of the work. Based on our own expertise, this review is mainly focused on the findings that have advanced the understanding of visual processing in the central visual pathway, visual perceptual learning, visual development and eye diseases. Abstract: A decade has passed since near infrared spectroscopy (NIRS) was first applied to functional brain imaging in infants. As part of the team that published the first functional near infrared spectroscopy (fNIRS) infant study in 1998, we have continued to develop and refine both the technology and methods associated with these measurements. The increasing international interest that this technology is generating among neurodevelopmental researchers and the recent technical developments in biomedical optics have prompted us to compile this review of the challenges that have been overcome in this field, and the practicalities of performing fNIRS in infants. We highlight the increasingly diverse and ambitious studies that have been undertaken and review the technological and methodological advances that have been made in the study design, optical probe development, and interpretation and analyses of the haemodynamic response. A strong emphasis is placed on the potential of the technology and future prospects of fNIRS in the field of developmental neuroscience. [Copyright &y& Elsevier]

Abstract: Deception involves complex neural processes and correlates in the brain. Functional brain imaging techniques have been used to study and understand brain mechanisms during deception. In this study, we utilized functional near-infrared spectroscopy (fNIRS) to investigate hemodynamic responses to deception in the prefrontal cortex (PFC) at the individual level. The protocol involved a mock theft scenario that was previously used in a functional MRI (fMRI) study of detecting deception. Subjects (N =11) were instructed to steal a ring or a watch and then conceal the item that they stole. Participants then responded to visually presented questions regarding which item they took. While the subjects were answering the questions, their PFC activity was measured using fNIRS. The brain activity associated with deceptive responses demonstrated significant changes in hemoglobin concentrations with respect to the baseline, while the response of truth telling was not statistically different from baseline. The regions of greater activation induced by deception identified by fNIRS were approximately consistent with those reported by the previous fMRI study using a similar protocol. This study demonstrates that fNIRS is a promising new technique to understand hemodynamic and neural correlates of deception and thus to detect deception with the added advantages of being compact, technically easier to implement, and inexpensive compared to functional MRI. Abstract: There is increasing evidence for a key role of psychological processes and their central nervous system substrates in functional gastrointestinal disorders, although the exact nature of the relationship remains only partially understood. However, progress in this key area of psychosomatic medicine is rapidly being made. In this review article, we will give an overview of recent advances in understanding the complex mechanisms by which psychological processes and functional gastrointestinal disorder symptoms reciprocally influence each other. Various lines of evidence from different branches of biomedical and psychological science will be discussed, particularly epidemiology, patho- and psychophysiology and functional brain imaging, focusing on the most recent and novel findings. Abstract: Patients with chronic pain disorders often show somatosensory disturbances that are considered to be functional. This paper aims at a more precise clinical description and at a documentation of functional neuroimaging correlates of this phenomenon. We examined 30 consecutive patients with unilaterally accentuated chronic pain not explained by persistent peripheral tissue damage and ipsilateral somatosensory disturbances including upper and lower extremities and trunk. The patients were assessed clinically and with conventional brain CT or MRI scan. In the last 11 patients functional neuroimaging was carried out (18-fluordeoxyglucose positron emission tomography=FDG-PET). Depressive symptoms were assessed with the Hamilton depression scale (HAMD-17) and pain intensity was rated with a visual analogue scale for pain (VAS). All patients suffered from mild to moderate depressive symptoms. All patients had experienced a prolonged antecedent phase of severe emotional distress; most of them remembered a “trigger episode of somatic pain” on the affected side. Somatosensory deficits were a replicable hyposensitivity to touch and heat perception of nondermatomal distribution. Conventional imaging procedures (brain CT or MRI scans) showed no structural changes. However, in 11 patients functional imaging with FDG-PET showed a significant hypometabolic pattern of changes in cortical and subcortical areas, mainly in the post-central gyrus, posterior insula, putamen, and anterior cingulate cortex. In summary, pain-related nondermatomal somatosensory deficits (NDSDs) are a phenomenon involving biological as well as psychosocial factors with replicable neuroperceptive clinical findings and a complex neurodysfunctional pattern in the FDG-PET. Introduction: Functional gastrointestinal disorders (FGIDs) are a highly prevalent group of heterogeneous disorders whose diagnostic criteria are symptom based in the absence of a demonstrable structural or biochemical abnormality. Chronic abdominal pain or discomfort is a defining characteristic of these disorders and a proportion of patients may display heightened pain sensitivity to experimental visceral stimulation, termed visceral pain hypersensitivity (VPH). Gastric distension activates a cerebral network including brainstem, thalamus, insula, perigenual anterior cingulate, cerebellum, ventrolateral prefrontal cortex and potentially somatosensory regions. Cortical deactivations during gastric distension have hardly been reported. To describe brain areas of decreased activity during gastric fundus distension compared to baseline, using data from our previously published study (Gastroenterology, 128, 2005 and 564). H215O-brain positron emission tomography was performed in 11 healthy volunteers during five conditions (random order): (C1) no distension (baseline); isobaric distension to individual thresholds for (C2) first, (C3) marked, (C4) unpleasant sensation and (C5) sham distension. Subtraction analyses were performed (in SPM2) to determine deactivated areas during distension compared to baseline, with a threshold of Puncorrected_voxel_level < 0.001 and Pcorrected_cluster_level < 0.05. Baseline–maximal distension (C1–C4) yielded significant deactivations in: (i) bilateral occipital, lateral parietal and temporal cortex as well as medial parietal lobe (posterior cingulate and precuneus) and medial temporal lobe (hippocampus and amygdala), (ii) right dorsolateral and dorso- and ventromedial PFC, (iii) left subgenual ACC and bilateral caudate head. Intragastric pressure and epigastric sensation score correlated negatively with brain activity in similar regions. The right hippocampus/amygdala deactivation was specific to sham. Gastric fundus distension in health is associated with extensive cortical deactivations, besides the activations described before. Whether this represents task-independent suspension of ‘default mode’ activity (as described in various cognitive tasks) or an visceral pain/interoception-specific process remains to be elucidated. Abstract: We review research from our laboratory that attempts to pull apart the functional and neural mechanisms of visual search using converging, inter-disciplinary evidence from experimental studies with normal participants, neuropsychological studies with brain lesioned patients, functional brain imaging and computational modelling. The work suggests that search is determined by excitatory mechanisms that support the selection of target stimuli, and inhibitory mechanisms that suppress irrelevant distractors. These mechanisms operate through separable though overlapping neural circuits which can be functionally decomposed by imposing model-based analyses on brain imaging data. The chapter highlights the need for inter-disciplinary research for understanding complex cognitive processes at several levels. [Copyright &y& Elsevier]

Using positron emission tomography (PET), we explored the neural correlates of an executive function, dual tasking, in patients with amnestic mild cognitive impairment (aMCI) and in elderly controls. The experiment employed simple auditory and visual tasks that were presented both in isolation and simultaneously to create a task condition requiring enhanced attentional control. Behaviorally, both groups performed well, albeit the patients made more errors on the visual task. The PET analysis focused at prefrontal regions where group differences in task-related activation patterns were expected. During dual task performance, the patients showed attenuated activity in the left inferior frontal region when compared to the controls. Abstract: To investigate maturational plasticity of fluid cognition systems, functional brain imaging was undertaken in healthy 8–19 year old participants while completing visuospatial relational reasoning problems similar to Raven’s matrices and current elementary grade math textbooks. Analyses revealed that visuospatial relational reasoning across this developmental age range recruited activations in the superior parietal cortices most prominently, the dorsolateral prefrontal, occipital–temporal, and premotor/supplementary cortices, the basal ganglia, and insula. There were comparable activity volumes in left and right hemispheres for nearly all of these regions. Regression analyses indicated increasing activity predominantly in the superior parietal lobes with developmental age. In contrast, multiple anterior neural systems showed significantly less activity with age, including dorsolateral and ventrolateral prefrontal, paracentral, and insula cortices bilaterally, basal ganglia, and particularly large clusters in the midline anterior cingulate/medial frontal cortex, left middle cingulate/supplementary motor cortex, left insula-putamen, and left caudate. Findings suggest that neuromaturational changes associated with visuospatial relational reasoning shift from a more widespread fronto-cingulate-striatal pattern in childhood to predominant parieto-frontal activation pattern in late adolescence. Abstract: The goal of the study was to investigate the neural circuit recruited by adult readers during performance of a lexical decision task by assessing the relative timing of neurophysiological activity in the brain regions that comprise this circuit. The time course of regional activation associated with lexical decision was studied in 17 adult volunteers using magnetoencephalography. Following activity in mesial occipital cortices, activation progressed to lateral and ventral occipito-temporal regions (often encompassing the posterior portion of the middle temporal gyrus), followed by activity in the superior temporal gyri (STGp), motor/premotor cortices, and the inferior frontal gyrus. The latency of STGp activation relative to the latency of the motor response to the word stimuli did not support critical involvement of this area in lexical decision. Timing, word length, and word frequency effects found for activity in each area are discussed in relation to the purported roles of each region into the brain circuit for reading. Abstract: To gain insight into memory disturbances in Complex Posttraumatic Stress Disorder (Complex PTSD), we investigated declarative memory function and medial temporal lobe activity in patients and healthy non-traumatized controls. A case-control study was performed in nine patients with Complex PTSD and nine controls. All respondents performed a declarative memory task with neutral and emotional, negative words during functional magnetic resonance imaging. Memory performance of neutral words was impaired in Complex PTSD with a relative conservation of recall of negative words. Deep encoding of later remembered negative words, as well as correct recognition of negative words and false alarms, was associated with an enhanced Blood Oxygenation Level Dependent (BOLD) response in the left hippocampus extending into the parahippocampal gyrus of Complex PTSD patients compared with controls. Post-hoc volumetric comparisons did not reveal significant anatomical differences in the medial temporal lobe between Complex PTSD patients and controls. We conclude that in Complex PTSD preferential recall of negative words is associated with increased activation in the left hippocampus and parahippocampal gyrus during both successful and false recall. Elucidation of biochemical, physiological, and cellular contributions to metabolic images of brain is important for interpretation of images of brain activation and disease. Discordant brain images obtained with [14C]deoxyglucose and [1- or 6-14C]glucose were previously ascribed to increased glycolysis and rapid [14C]lactate release from tissue, but direct proof of [14C]lactate release from activated brain structures is lacking. Analysis of factors contributing to images of focal metabolic activity evoked by monotonic acoustic stimulation of conscious rats reveals that labeled metabolites of [1- or 6-14C]glucose are quickly released from activated cells as a result of decarboxylation reactions, spreading via gap junctions, and efflux via lactate transporters. Label release from activated tissue accounts for most of the additional [14C]glucose consumed during activation compared to rest. Metabolism of [3,4-14C]glucose generates about four times more [14C]lactate compared to 14CO2 in extracellular fluid, suggesting that most lactate is not locally oxidized. In brain slices, direct assays of lactate uptake from extracellular fluid demonstrate that astrocytes have faster influx and higher transport capacity than neurons. Also, lactate transfer from a single astrocyte to other gap junction-coupled astrocytes exceeds astrocyte-to-neuron lactate shuttling. Astrocytes and neurons have excess capacities for glycolysis, and oxidative metabolism in both cell types rises during sensory stimulation. The energetics of brain activation is quite complex, and the proportion of glucose consumed by astrocytes and neurons, lactate generation by either cell type, and the contributions of both cell types to brain images during brain activation are likely to vary with the stimulus paradigm and activated pathways. Beyond the reading-related deficits typical of developmental dyslexia (DD), recent evidence suggests that individuals afflicted with this condition also show difficulties in motor sequence learning. To date, however, little is known with respect to the characteristics of the learning impairments, nor to the neural correlates associated with this type of procedural deficit in DD patients. Here, we first summarize the results of the few behavioral and brain imaging studies that have investigated the effects of DD on motor sequence learning. To help guide research in this field, we then discuss relevant psychophysical and neuroimaging work conducted in healthy volunteers in relation to three different conceptual perspectives: when, how, and what. More specifically, we examine the cognitive boundaries that affect performance across the different stages of learning (i.e., “when”), the different cognitive processes (i.e., “how”) under which learning occurs, and the mental representations (i.e., “what”) that are elicited when acquiring this type of skilled behavior. Cognitive neuroscience studies of Attention Deficit Hyperactivity Disorder (ADHD) suggest multiple loci of pathology with respect to both cognitive domains and neural circuitry. Cognitive deficits extend beyond executive functioning to include spatial, temporal, and lower-level "nonexecutive" functions. Atypical functional anatomy extends beyond frontostriatal circuits to include posterior cortices, limbic regions, and the cerebellum. Pathophysiology includes dopaminergic as well as noradrenergic neurotransmitter systems. Summary: Background: Whilst acute loss of ovarian function is associated with memory deficits, the biological basis of this is poorly understood. We have previously reported that acute loss of function during Gonadotropin Hormone Releasing Hormone agonists (GnRHa) treatment is associated with impaired verbal memory and a disruption of corresponding left inferior frontal gyrus (LIFG) during the encoding stage. In the current study, we provide a critical extension to this work by determining whether this memory deficit is reversible following normalization of ovarian function. To do this we carried out a further imaging study using the same verbal memory recognition task after cessation of GnRHa-induced ovarian suppression. Method: We used event-related fMRI to study verbal episodic memory performance and brain activation at the LIFG in 13 healthy pre-menopausal women pre-, during, and post-acute ovarian hormone suppression using GnRHa. Results: Following resolution of acute GnRHa-induced ovarian suppression, verbal recognition scores returned to their initial levels and this restoration was associated with a restored level of left frontal activation during successful encoding of words. Conclusions: Our findings suggest that the memory deficits associated with acute ovarian suppression are reversed following resolution of normal ovarian function and are associated with reversible attenuation of LIFG activation during encoding. These findings lend further support to the hypothesis that memory difficulties reported by some women following acute ovarian hormone withdrawal are reversible and may have a clear neurobiological basis. Abstract: Mental imagery (MI) is the mental rehearsal of movements without overt execution. Brain imaging techniques have made it possible to identify the brain regions that are activated during MI and, for voluntary motor tasks involving hand and finger movements, to make direct comparison with those areas activated during actual movement. However, the fact that brain activation differs for different types of imagery (visual or kinetic) and depends on the skill level of the individual (e.g., novice or elite athlete) raises a number of important methodological issues for the design of brain imaging protocols to study MI. These include instructing the subject concerning the type of imagery to use, objective measurement of skill level, the design of motor tasks sufficiently difficult to produce a range of skill levels, the effect of different environments on skill level (including the imaging device), and so on. It is suggested that MI is more about the neurobiology of the development of motor skills that have already been learned, but not perfected, than it is about learning motor skills de novo. A task set is a configuration of cognitive processes that is actively maintained for subsequent task performance. Single-unit and brain-imaging studies have identified the neural correlates for task sets in the prefrontal cortex. Here I examine whether the neural data obtained thus far are sufficient to explain the behaviors that have been illustrated within the conceptual framework of task sets. I first discuss the selectivity of neural activity in representing a specific task. I then discuss the competitions between neural representations of task sets during task switch. Finally I discuss how, in neural terms, a task set is implemented to facilitate task performance. The processes of representing, updating, and implementing task sets occur in parallel at multiple levels of brain organization. Abstract: Smooth pursuit eye movements (SP) are driven by moving objects. The pursuit system processes the visual input signals and transforms this information into an oculomotor output signal. Despite the object''s movement on the retina and the eyes’ movement in the head, we are able to locate the object in space implying coordinate transformations from retinal to head and space coordinates. To test for the visual and oculomotor components of SP and the possible transformation sites, we investigated three experimental conditions: (I) fixation of a stationary target with a second target moving across the retina (visual), (II) pursuit of the moving target with the second target moving in phase (oculomotor), (III) pursuit of the moving target with the second target remaining stationary (visuo-oculomotor). Precise eye movement data were simultaneously measured with the fMRI data. Visual components of activation during SP were located in the motion-sensitive, temporo-parieto-occipital region MT+ and the right posterior parietal cortex (PPC). Motor components comprised more widespread activation in these regions and additional activations in the frontal and supplementary eye fields (FEF, SEF), the cingulate gyrus and precuneus. The combined visuo-oculomotor stimulus revealed additional activation in the putamen. Possible transformation sites were found in MT+ and PPC. The MT+ activation evoked by the motion of a single visual dot was very localized, while the activation of the same single dot motion driving the eye was rather extended across MT+. The eye movement information appeared to be dispersed across the visual map of MT+. This could be interpreted as a transfer of the one-dimensional eye movement information into the two-dimensional visual map. Potentially, the dispersed information could be used to remap MT+ to space coordinates rather than retinal coordinates and to provide the basis for a motor output control. The article addresses the potential impact of functional brain imaging (functional magnetic resonance imaging and positron-emission tomography) on surrogate end-of-life decision-making in light of varying state-law definitions of consciousness, some of which define awareness behaviorally and others functionally. The article concludes that, in light of admonitions by neuroscientists that functional brain imaging cannot yet replace behavioral evaluation to determine the existence of consciousness, state legislatures, courts and drafters of written advance healthcare directives should consider treating behavior, not function, as the touchstone for end-of-life decision-making.
