Abstract

Sixteen species, of which 13 are new, in the subgenus Hebridosimulium Grenier & Rageau are now known. Fiji has only Simulium laciniatum, which is widely distributed on five islands. Tahiti has only one species, the enigmatic S. anatolicum. The remaining species are in Vanuatu and known from nine islands, ranging the full length and breadth of the archipelago. Hebridosimulium species are divided among two species groups. The callipygium species group is characterized, in part, by a smooth posterior abdomen in the larvae, and all but one species possess dorsal tubercles on the larval abdomen. Simulium callipygium is the type species, with the tuberculate species in a subgroup comprised of S. banksi, S. johnfrumi, S. pankumuense, and S. tuberculum. The second species group has larvae with the posteroventral abdomen markedly expanded, and all species lack dorsal tubercles. Constituents of the steatopygium species group are S. anatolicum, S. jolyi, S. laciniatum, S. lucyae, S. paradisium, S. steatopygium, S. subparadisium, S. supercilium, and S. undulatum. Two other species of this group, S. adsonense and S. spiroi, of which larvae have markedly pigmented heads, are assigned to the adsonense species subgroup. A chromosome map for Hebridosimulium is established for the first time.

Abstract

Ochlerotatus (Finlaya) japonicus (Theobold) is newly established in Ontario, Canada. It was first discovered in 2001 during the province-wide West Nile virus mosquito surveillance program implemented by the Ontario Ministry of Health and Long Term Care. Although the numbers of adults trapped in CDC light traps were low in 2001 and 2002, they increased during the 2003 and 2004 surveillance seasons. Oc. japonicus larvae also have been collected in large numbers in the Niagara Peninsula in a variety of natural and artificial containers. The number of health units with records for Oc. japonicus has increased over the 4 yr of surveillance, illustrating the ability of this species to rapidly extend its range. As a potential arboviral bridge vector, its establishment in Ontario requires further study and should be considered a public health concern.

Abstract

The detection of West Nile virus (WNV) in mosquitoes by real-time RT-PCR provides valuable information on the epidemiology of the virus and identifies mosquito species that are potential vectors. Testing sets of pooled mosquitoes of the same species is logistically the easiest and most cost-effective approach for WNV testing; however, little information is available on how the results of small pooled sets relate to those of testing individual mosquitoes. During the 2002 outbreak, we compared pooled and individual samples of two mosquito species (Culex pipiens and Culex restuans) collected from three Health Unit regions in Ontario, Canada. Significantly more Cx. restuans were infected with WNV compared to Cx. pipiens. We show that with pool sizes of five individuals both MIR (minimum infection rates) and MLE (maximum likelihood estimation) values were acceptable in estimating infection rates.

Abstract

In situ hybridization was used to determine the physical location of the Hsp70 genes in salivary polytene chromosomes of Anopheles darlingi from Manaus and Macapa, Brazil, and to assess the usefulness of the Hsp70 locus as a genetic marker in A. darlingi populations. In both populations, the double markings corresponding to the Hsp70-12A and Hsp70-14A genes were located on the right arm of chromosome 2. The Hsp70 locus was considered to be an excellent marker for studying chromosomal evolution and relationships among A. darlingi populations.

Abstract

Simulium craigi Adler and Currie is a polymorphic species based on polytene chromosome banding patterns in the long arm of chromosome III (IIIL). Three cytotypes are described based on the predominant IIIL sequences. These correspond to three broad geographic areas: cytotype 'CC' from Pennsylvania; cytotype 'AF' from Ontario and Manitoba; and cytotype 'ACF/BCF' from New Hampshire. In the absence of sympatric populations, these cytotype differences are best explained by clinal variation within a single species. The relationship of S. craigi to other described members of the S. vernum group is discussed.

Abstract

This study, done on the Avalon Peninsula, Newfoundland, identified selected wild mammalian hosts and quantified the incidence of sugar feeding for several species (species complexes) of black flies. Female flies were taken from fox, snowshoe hare, lynx, caribou, and human baits. Simulium truncatum/venustum complex females took blood from fox, lynx, rabbit, and caribou and were frequently taken in sweep net collections around humans. Blood-fed females of the Simulium rostratum/verecundum complex were collected only from caribou, were rarely attracted to the other mammalian baits, and were not attracted to humans. Although most females had sugar-fed before seeking a host, significant interspecific differences in the incidence of fructose-positive females were found among the four species (species complexes) examined ( Prosimulium mixtum , 92.9%; S. truncatum/venustum complex, 87.4%; Stegopterna mutata (triploid cytospecies), 83.3%; Simulium vittatum complex, 67.4%). No significant differences were found in the proportion of sugar-fed flies among three habitats examined (forest, scrubland, and pasture) for either P. mixtum or the S. truncatum/venustum complex. However, the percentage of sugar-fed S. truncatum/venustum complex females was significantly higher in the evening than in the morning. Time of day did not influence the incidence of sugar-fed P. mixtum .

A modified ELISA, in which a biotinylated second antibody and a streptavidin-biotinylated peroxidase complex are used, has been developed to identify the blood meal sources of black flies. The modified assay is sensitive enough to identify correctly 100% of blood meals at 73 h postingestion and 80% of blood meals at 116 h postingestion in black flies held in the field at ambient temperatures.

Abstract

A new species of black fly, Stegopterna diplomutata n. sp. (Diptera: Simuliidae) is described and illustrated. This species is most closely related to Stegopterna mutata (Malloch, 1914), with which it has long been confused. The two species are most easily distinguished based on the presence or absence of males; St. diplomutata is a diploid bisexual species that possesses males, whereas St. mutata is a triploid parthenogenetic species that lacks males. The two species are otherwise not distinguishable except through examination of their larval polytene chromosomes. Observations are provided about the evolution of triploidy in Stegopterna.

Abstract The black fly subgenus Inseliellum is present on a series of archipelagos in the South Pacific. In this study, larval polytene chromosome maps of six Inseliellum species are presented. Chromosomal relationships among taxa were determined through shared fixed inversions or chromosomal landmark positioning. Three fixed inversions (IL-2, IIS-1, and IIIS-1) were shared among species, as was the position of the nucleolar organizer (NO) (IL or IIL). Comparisons to two previously studied species of Inseliellum are included to produce a cytological transformation series among eight taxa. The NO position defines two clades in the phylogeny of Inseliellum, herein named the NO-IL and NO-IIL clades. The utility of this cytological data set is discussed. [ABSTRACT FROM AUTHOR]

In situ hybridization was used to determine the physical location of the Hsp70 genes in salivary polytene chromosomes of Anopheles darlingi from Manaus and Macapá, Brazil, and to assess the usefulness of the Hsp70 locus as a genetic marker in A. darlingi populations. In both populations, the double markings corresponding to the Hsp70-12A and Hsp70-14A genes were located on the right arm of chromosome 2. Abstract

The polytene chromosomes of four closely related Eusimulium (Roubaud) morphospecies from Europe have been examined. These are Eusimulium vernum (Macquart), E. costatum (Friedrichs), E. cryophilum (Rubtzov), and E. carpathicum (Knoz). Interspecific comparisons indicate that vernum and costatum are homosequential in chromosomal banding pattern; however, costatum does differ from vernum in possessing a conspicuous chromocenter. Both species may have B chromosomes, but these are not identical. Three kinds of vernum populations are found: pure X0Y0 (with undifferentiated X and Y chromosomes), pure X0Y1 (with males heterozygous for IIS-1 ve), and mixed X0Y0/X0Y1 populations. Only two European vernum cytotypes are indicated. Eusimulium cryophilum and E. carpathicum share a suite of autosomal polymorphisms as well as a fixed paracentric (IL-2) and a fixed pericentric (IIIP-1) inversion with respect to the vernum "Knebworth" standard. They differ from one another by three fixed inversions (IIS-1, IIIS-3, and IIIS-4). It is argued that at least two siblings, cryophilum A and cryophilum B, exist within cryophilum s.l. Sections 86B and 91C in the long arm of chromosome III appear to be extremely labile for breakage and reunion phenomena because several tandem or near-tandem inversions occur in this area. A cytophylogeny of the taxa under study is presented.

To test whether Simulium furculatum (Shewell) belongs to the Simulium vernum (Macquart) species-group, a comparison was made of the polytene chromosomes of S. furculatum and the S. vernum "Knebworth" standard. Only two chromosome arms (of six) could be completely analysed. It is argued that S. furculatum does not belong to the S. vernum species-group. A complex sex-chromosome system (X1, X2, Y1) is found in both eastern and western Canada. Phylogenetically, the single Y chromosome is intermediate between the two X chromosomes. Intraspecific inversion polymorphisms, which serve to differentiate eastern from western populations, are also identified. Abstract

Background Several recent climate change reviews have stressed the possibility of some malaria vectors occupying regions of higher altitudes than previously recorded. Indeed, highland malaria has been observed in several African nations, possibly attributable to changes in land use, vector control and local climate. This study attempts to expand the current knowledge of the distribution of common Anopheles species in Ecuador, with particular attention to highland regions (> 500 m) of the Andes. Methods Extensive field collections of larvae were undertaken in 2008, 2009 and 2010 throughout all regions of Ecuador (except the lower-altitude Amazonian plain) and compared to historical distribution maps reproduced from the 1940s. Larvae were identified using both a morphological key and sequencing of the 800 bp region of the CO1 mitochondrial gene. In addition, spatial statistics (Getis-Ord Hotspot Analysis: Gi*) were used to determine high and low-density clusters of each species in Ecuador. Results Distributions have been updated for five species of Anopheles in Ecuador: Anopheles albimanus, Anopheles pseudopunctipennis, Anopheles punctimacula, Anopheles eiseni and Anopheles oswaldoi s.l.. Historical maps indicate that An. pseudopunctipennis used to be widespread in highland Andean valleys, while other species were completely restricted to lowland areas. By comparison, updated maps for the other four collected species show higher maximum elevations and/or more widespread distributions in highland regions than previously recorded. Gi* analysis determined some highland hot spots for An. albimanus, but only cold spots for all other species. Conclusions This study documents the establishment of multiple anopheline species in high altitude regions of Ecuador, often in areas where malaria eradication programs are not focused.

237. West Nile virus in Ontario, Canada: A twelve-year analysis of human case prevalence, mosquito surveillance, and climate data.

Abstract

West Nile Virus (WNV) first arrived in Ontario, Canada in 2001 and has since spread throughout most of the province, causing disease in humans. The provincial government established a province-wide surveillance program to monitor WNV transmission throughout the 36 regional health units. Here we have acquired records of WNV human and mosquito surveillance from 2002 to 2013 to describe seasonal and geographic trends in WNV activity in southern Ontario. Additionally, we obtained climate data from seven municipalities to investigate how temperature and precipitation affect WNV transmission dynamics. We identified a strong quadratic relationship between the number of confirmed human cases and positive Culex mosquito pools recorded at the end of each year (R2 = 0.9783, p < 0.001). Using Spearman rank correlation tests, we identified that the minimum infection rate of Culex pipiens/restuans pools are the strongest predictor of human cases at a 1 week lag period. We also identified positive correlations between minimum infection rates, temperature, vector abundance, and cumulative precipitation. Global Moran's I index indicates strong positive autocorrelation and clustering of positive Culex pool counts in southern Ontario. Local indicators of spatial association tests revealed a total of 44 high-high and 1 high-low trap locations (n = 680). In the current work we have identified when and where hot spots of WNV activity have occurred in southern Ontario. The municipalities surrounding the western shore of the Lake Ontario and Windsor-Essex County have the largest records of positive mosquitoes and human cases. We identified that positive mosquitoes are a strong indicator of human cases to follow in the coming weeks. An epidemic action threshold of cumulative positive Culex pools was established, allowing Ontario public health officials to predict an epidemic at epidemiological week 34 (rho = 0.90, p < 0.001). These data have the potential to contribute to more efficient larvicide programs and awareness campaigns for the public.

Abstract

Larval habitat for three highland Anopheles species: Anopheles albimanus Wiedemann, Anopheles pseudopunctipennis Theobald, and Anopheles punctimacula Dyar & Knab was related to human land uses, rivers, roads, and remotely sensed land cover classifications in the western Ecuadorian Andes. Of the five commonly-observed human land uses, cattle pasture (n=30) provided potentially suitable habitat for An. punctimacula and An. albimanus in less than 14% of sites, and was related in a Principal Components Analysis (PCA) to the presence of macrophyte vegetation, greater surface area, clarity and algae cover. Empty lots (n=30) were related in the PCA to incident sunlight and provided potential habitat for An. pseudopunctipennis and An. albimanus in less than 14% of sites. The other land uses surveyed (banana, sugarcane and mixed tree plantations; n=28, 21, 25, respectively) provided very little standing water that could potentially be used for larval habitat. River edges and eddies (n=41) were associated with greater clarity, depth, temperature and algae cover, which provide potentially suitable habitat for An. albimanus in 58% of sites and An. pseudopunctipennis in 29% of sites. Road-associated water bodies (n=38) provided potential habitat for An. punctimacula in 44% of sites and An. albimanus in 26% of sites surveyed. Species collection localities were compared to land cover classifications using Geographic Information Systems software. All three mosquito species were associated more often with the category "closed/open broadleaved evergreen and/or semi-deciduous forests" than expected (P ≤ 0.01 in all cases), given such a habitat's abundance. This study provides evidence that specific human land uses create habitat for potential malaria vectors in highland regions of the Andes.

Abstract

ChEMBL (https://www.ebi.ac.uk/chembl/) is a manually curated, high-quality, large-scale, open, FAIR and Global Core Biodata Resource of bioactive molecules with drug-like properties, previously described in the 2012, 2014, 2017 and 2019 Nucleic Acids Research Database Issues. Since its introduction in 2009, ChEMBL's content has changed dramatically in size and diversity of data types. Through incorporation of multiple new datasets from depositors since the 2019 update, ChEMBL now contains slightly more bioactivity data from deposited data vs data extracted from literature. In collaboration with the EUbOPEN consortium, chemical probe data is now regularly deposited into ChEMBL. Release 27 made curated data available for compounds screened for potential anti-SARS-CoV-2 activity from several large-scale drug repurposing screens. In addition, new patent bioactivity data have been added to the latest ChEMBL releases, and various new features have been incorporated, including a Natural Product likeness score, updated flags for Natural Products, a new flag for Chemical Probes, and the initial annotation of the action type for ∼270 000 bioactivity measurements., (© The Author(s) 2023. Abstract

We report the major highlights of the School of Cheminformatics in Latin America, Mexico City, November 24-25, 2022. Six lectures, one workshop, and one roundtable with four editors were presented during an online public event with speakers from academia, big pharma, and public research institutions. One thousand one hundred eighty-one students and academics from seventy-nine countries registered for the meeting. As part of the meeting, advances in enumeration and visualization of chemical space, applications in natural product-based drug discovery, drug discovery for neglected diseases, toxicity prediction, and general guidelines for data analysis were discussed. Experts from ChEMBL presented a workshop on how to use the resources of this major compounds database used in cheminformatics. The school also included a round table with editors of cheminformatics journals. ChEMBL is a large, open-access bioactivity database (https://www.ebi.ac.uk/chembl), previously described in the 2012, 2014 and 2017 Nucleic Acids Research Database Issues. In the last two years, several important improvements have been made to the database and are described here. These include more robust capture and representation of assay details; a new data deposition system, allowing updating of data sets and deposition of supplementary data; and a completely redesigned web interface, with enhanced search and filtering capabilities.

A short-cut review was carried out to establish whether intravenous tranexamic acid is beneficial in managing acute epistaxis. Seven papers were found in Medline, Embase and the Cochrane Library using the reported searches, but none presented any evidence to answer the clinical question. A short cut review was carried out to establish the diagnostic utility of Emergency Physician performed ultrasound in confirming or refuting the diagnosis of deep vein thrombosis compared with a radiology scan. A systematic review incorporating 16 studies and two studies published subsequently were found to be relevant to the question. Abstract

A short-cut review was carried out to establish whether the finding of a normal cranial CT scan, in a child with a history of minor head injury, a Glasgow coma scale (GCS) score of 15 and normal neurological examination could facilitate safe discharge from hospital. Ninety-eight articles were found using the reported search and a further two papers were found from the references of selected papers. A total of 11 trials were included with one prospective cohort study including 13,543 patients. The authors, date and country of publication, patient group studied, study type, relevant outcomes, results and study weaknesses of these papers are tabulated (table 3). It is concluded that in the absence of any coagulation problems, a child who presents following a minor head injury with a GCS of 15, normal neurological examination and a normal CT scan can be safely discharged to the care of a capable parent.

246. West Nile virus surveillance and diagnostics: A Canadian perspective.

Abstract

A surveillance program has been in place since 2000 to detect the presence of West Nile virus (WNV) in Canada. Serological assays are most appropriate when monitoring for human disease and undertaking case investigations. Genomic amplification procedures are more commonly used for testing animal and mosquito specimens collected as part of ongoing surveillance efforts. The incursion of WNV into this country was documented for the first time in 2001 when WNV was demonstrated in 12 Ontario health units during the late summer and fall. In 2002 WNV activity was documented by avian surveillance in Ontario by mid-May with subsequent expansion of the virus throughout Ontario and into Quebec, Manitoba, Saskatchewan and Nova Scotia. Human cases were recorded in both Ontario and Quebec in 2002 with approximately 800 to 1000 probable, confirmed and suspect cases detected. The possible recurrence and further spread of WNV to other parts of Canada in 2003 must be anticipated with potential risk to public health. The continued surveillance and monitoring for WNV-associated human illness is necessary and appropriate disease prevention measures need to be in place in 2003.
