It is known that miR-381 plays a therapeutic role in a variety of cancers, but the regulatory mechanism of miR-381 in the treatment of lung cancer remains unclear. This study is aimed at exploring the expression level and mechanism of miR-381 in lung cancer. In this experiment, quantitative real-time PCR (qRT-PCR), western blot, and other methods were used to detect the expression of miR-381 and ubiquitin-specific protease 39 (USP39) in lung cancer tissues. The target genes of miR-381 were predicted by bioinformatics techniques, and the targeting relationship between miR-381 and USP39 was verified by the dual-luciferase reporting method. The expression levels of miR-381 and USP39 were adjusted to verify the effect of miR-381 on the expression of USP39 gene. The effect of miR-381 expression on proliferation of lung cancer cells was verified by cell proliferation and invasion experiments. miR-381 was downregulated in non-small-cell lung cancer tissues and cell lines, while USP39 was upregulated. The dual-luciferase reporter gene assay showed that miR-381 and USP39 had targeted binding sites. After transfection with miR-381 mimics, USP39 expression was significantly decreased, cell proliferation decreased, and apoptosis increased. After transfection with miR-381 inhibitor, USP39 expression was significantly increased, cell proliferation increased, and cell apoptosis decreased. Overexpression of USP39 significantly increased the invasion ability and cell survival curve ( p < 0.05 ). In conclusion, overexpression of miR-381 can regulate the expression of USP39, inhibit the proliferation and invasion of cancer cells, and induce apoptosis of cancer cells. This may provide a new perspective and strategy for targeted therapy of non-small-cell lung cancer.

Jute fibers are commonly used as reinforcement in composite materials manufacture due to its excellent mechanical properties. In this study, alkali (NaOH) and silane coupling agent KH570 treatment were conducted on component jute nonwovens to improve the bonding strength of jute/PLA composite interface. The jute/PLA composites were prepared from treated jute nonwovens and PLA films by molding method. The results show that alkali treatment can significantly improve the tensile, flexural and dynamic thermomechanical properties of jute/PLA composites. The composites treated with 1% NaOH exhibits the maximum tensile strength while the composites treated with 5% NaOH presents the maximum flexural strength and the minimum loss in the dynamic thermo-mechanical process. Moreover, the tensile and flexural strength of the composite treated with the combination of 5% NaOH-5% KH570 is significantly stronger than that when they act alone. This research can provide an optimal treatment strategy for the jute/PLA degradable composites with higher mechanical and thermos-mechanical tolerance.

They include the properties of electromagnetic form factors, impact parameter space GPDs, gravitational form factors and associated pressure profiles, and the character and consequences of all-orders evolution. Continuum Schwinger function methods for the strong-interaction bound-state problem are used to arrive at a unified set of parameter-free predictions for the semileptonic K → π , D → π , K and D s → K , B ( s ) → π ( K ) transition form factors and the associated branching fractions. The form factors are a leading source of uncertainty in all such calculations: our results agree quantitatively with available data and provide benchmarks for the hitherto unmeasured D s → K 0 , B ¯ s → K + form factors. The analysis delivers a value of | V c s | = 0.974 ( 10 ) and also predictions for all branching fraction ratios in the pseudoscalar meson sector that can be used to test lepton flavour universality. Quantitative comparisons are provided between extant theory and the recent measurement of B B s 0 → K − μ + ν μ . Here, further, refined measurements would be useful in moving toward a more accurate value of | V u b | .

This study was designed to explore the impact of Lycium barbarum polysaccharide (LBP) on inflammation and gut microbiota in mice with allergic asthma. Mice were divided into four groups: control group, OVA (ovalbumin) group, Con+LBP group, OVA+LBP group. After 28 days of LBP intervention, mice were euthanized and associated indications were investigated. Histopathological examination demonstrated that LBP reduced lung injury. The results of our current study provide evidence that supplementation with LBP in asthmatic mice decreases TNF, IL-4, IL-6, MCP-1, and IL-17A in plasma and bronchoalveolar lavage fluid (BALF). Sequencing and analysis of gut microbiota indicated that compared with the OVA group, Lactobacillus and Bifidobacterium were increased, but Firmicutes, Actinobacteria, Alistipes, and Clostridiales were decreased in the OVA+LBP group. We also found that gut microbiota were related to inflammation-related factors. Therefore, we speculate that LBP may improve allergic asthma by altering gut microbiota and inhibiting inflammation in mice.

Herein, a facial stratagem for the large-scale synthesis of NiO/C nanosheets used as magnetic adsorbents is proposed by using nanoscale coordination polymers (nanoCPs) as precursors. The NiO nanoparticles (NPs) scattered on the NiO/C nanosheets have an untrasmall size (~3 nm) as well as uniform dispersion, since the Ni2+ ions are preassembled into the nanoCPs precursors by coordination bonds and transformed to NiO NPs in situ under the restriction of carbon layers. Thanks to the small size and spatially uniform distribution of NiO NPs, fully exposed carbon matrix containing part of graphene structures, and the high specific surface area of the special nanosheet structure, the magnetic NiO/C nanosheets have an a great breakthrough in the adsorption performance toward Congo Red (CR) among many reported NiO-based materials. The maximum adsorption capacity for CR is up to 2000 mg/g, and the adsorption kinetics and thermodynamics are also systematically studied. Moreover, the NiO/C nanosheets can be separated from water and reused simply by an external magnetic field. The NiO/C nanosheets with low cost and extraordinary adsorption ability have great potential in water pollution treatment area.

Based on green chemistry, 3D hierarchical ZnO microflowers (MFs) constructed from layered 2D nanosheets have been designed as carriers to uniformly load PdO/Pd nanoparticles by the calcination of the precursor (ZnO/Pd(II) microflowers) in air. The ZnO/Pd(II) microflowers are prepared by dipping the 3D ZnO MFs in an ethanolic solution of PdCl2 without involving additional agents. The as-prepared 3D hierarchical ZnO microflowers are a stable support to enhance the dispersibility of PdO/Pd nanoparticles to achieve better catalytic performance for the reduction of 4-nitrophenol (4-NP) to 4-aminophenol (4-AP). In addition, the synergistic effect between the PdO and Pd nanoparticles is another major driver that promotes the catalytic process. Over 98% of 4-NP is reduced within 7 min. Furthermore, structural analysis of the ZnO/PdO/Pd microflowers can provide useful information for understanding structure-performance relationships.

Patients with schizophrenia (SCZ) consistently show prospective memory (PM) impairments, and the technique of implementation intentions has been shown to improve PM performance in these patients. PM is considered to have prospective and retrospective components. However, it remains unclear which component of PM is impaired in patients with SCZ and which component(s) is facilitated by implementation intentions (II). The present study aimed to examine these two issues. Forty-two patients with SCZ and 42 matched healthy controls were randomly assigned to an II group or a typical instruction group. All participants were administered a color-matching PM task. Results showed that, using a multinomial-modeling approach, patients with SCZ exhibited impairment in the retrospective component of PM. In addition, while II improved the prospective PM component in healthy controls, both prospective and retrospective PM components in patients with SCZ were improved. Together, our results shed light on the mechanism of PM impairment in SCZ patients and the mechanism of II in improving PM performance.

Rationale: Malignant ascites caused by cancer cells results in poor prognosis and short average survival time. No effective treatment is currently available for malignant ascites. In this study, the effects of lentinan (LNT)-functionalized selenium nanoparticles (Selene) on malignant ascites were evaluated. Furthermore, the mechanism of Selene targeting mitochondria of tumor cells were also investigated. Methods: Selene were synthesized and characterized by TEM, AFM and particle size analysis. The OVCAR-3 and EAC cells induced ascites models were used to evaluate the effects of Selene on malignant ascites. Proteomic analysis, immunofluorescence, TEM and ICP-MS were used to determine the location of Selene in tumor cells. Mitochondrial membrane potential, ROS, ATP content, and caspase-1/3 activity were detected to evaluate the effect of Selene on mitochondrial function and cell apoptosis. Immunofluorescence, Co-IP, pull-down, duolink, Western blot, and FPLC were used to investigate the pathway of Selene targeting mitochondria. Results: Selene could effectively inhibit ascites induced by OVCAR-3 and EAC cells. Selene was mainly located in the mitochondria of tumor cells and induced apoptosis of tumor cells. The LNT in Selene was involved in caveolae-mediated endocytosis through the interaction between toll-like receptor-4 (TLR4) and caveolin 1 (CAV1). Furthermore, the Selene in the endocytic vesicles could enter the mitochondria via the mitochondrial membrane fusion pathway, which was mediated by TLR4/TNF receptor associated factor 3 (TRAF3)/mitofusin-1 (MFN1) protein complex. Conclusion: Selene is a candidate anticancer drug for the treatment of malignant ascites. And TLR4/TRAF3/MFN1 may be a specific nano-drug delivery pathway that could target the mitochondria.

Background A previous study by our team reported the prevalence of primary headache disorders and factors associated with headache among nurses in three hospitals in North China. The aim of this cross-sectional survey was to learn more about how medical nurses in South China were affected by headache. Additionally, we determined the prevalence of headache and measured the impact of headache among doctors in mainland China for the first time. Methods Stratified random cluster sampling was used to select 280 physicians and 365 nurses from various departments in four hospitals in Sanya, which is one of southernmost cities in China. Information was collected on demographic data, occupational factors and headache characteristics by using a structured questionnaire. Results Among 645 medical staff, 548 (85%) responded (doctors = 240, nurses = 308). Among the medical staff, the 1-year prevalence of primary headache disorders was 50%, with 25.9% experiencing migraine and 24.1% experiencing tension-type headache (TTH). The prevalence of migraine in female doctors was higher than that in female nurses, although this difference was not significant (32.4% vs. 29.8%, P = 0.628). Multivariate analysis showed that being female and working in other specialties (Emergency Department & Radiology Department) remained independent risk factors for migraine in doctors (OR 2.314 and 3.223). In nurses, being married was a risk factor for migraine (OR 3.728), and job titles remained an independent risk factor for migraine and TTH (OR 2.294 and 4.695). Working more than 6 night-shifts per month was associated with an increased prevalence of migraine and TTH in doctors; the same was true in nurses for migraine, but not for TTH. Conclusion The prevalence of primary headache disorders in both nurses and doctors is higher than that in the general population in South China. Our study shows that occupation, geography and sex may play an important role. Further, female doctors are more susceptible than female nurses to migraine. The risk factors relevant to headache that were found in this study should provide an important reference for promoting occupational health in medical staff, especially female doctors in China.

A novel supercapacitor material was synthesized from SiO2-coated Co3O4 and CuO nanoparticles. These nanocapsules provided large specific surface area, fast electron transport, good mechanical stability and high electrochemical activity. The specific capacitance at 1 A g−1 was 835.4 F g−1, and it had excellent cycling performance.

We designed a facile and green self-assembly route for the large-scale production of bimetallic coordination polymer–Zn(MAA)2/Pd(II) nanoribbons by employing two terminal units with distinct hard–soft properties of the smallest semi-rigid methacrylate anion (MAA−) to combine with two different metal ions. The COO− (chemically hard base) served as the initial unit, which was applied to coordinate with the Zn2+ ions for constructing the host network of this Janus CPs by facilely adjusting the proportion of ethanol/water to control the degree of solvation. After soaking in an ethanol solution of PdCl2, the alkene units (chemically soft base) act as secondary donors to load the soft noble metal Pd2+ ions and form Zn(MAA)2/Pd(II) nanoribbons, which are ideal precursors of ZnO/PdO nanoribbons. By pyrolyzing the dual metal ion equipped Zn(MAA)2/Pd(II) precursors, ZnO nanoribbon supported PdO nanoparticles (NPs) with a well-defined structure were facilely fabricated on a large scale. Furthermore, the as-obtained ZnO/PdO nanoribbons exhibit efficient catalytic performance for the reduction of 4-NP, which can convert 99.3% of the substrates within 6 min. This superior catalytic performance may be attributed to the well-dispersed PdO nanoparticles as well as the absence of additional agents introduced a green synthetic method.

This paper focuses on the social function of painful experience as revealed by recent studies on social decision-making. Observing others suffering from physical pain evokes empathic reactions that can lead to prosocial behavior (e.g., helping others at a cost to oneself), which might be regarded as the social value of pain derived from evolution. Feelings of guilt may also be elicited when one takes responsibility for another’s pain. These social emotions play a significant role in various cognitive processes and may affect behavioral preferences. In addition, the influence of others’ pain on decision-making is highly sensitive to social context. Combining neuroimaging techniques with a novel decision paradigm, we found that when asking participants to trade-off personal benefits against providing help to other people, verbally describing the causal relationship between their decision and other people’s pain (i.e., framing) significantly changed participants’ preferences. This social framing effect was associated with neural activation in the temporoparietal junction (TPJ), which is a brain area that is important in social cognition and in social emotions. Further, transcranial direct current stimulation (tDCS) on this region successfully modulated the magnitude of the social framing effect. These findings add to the knowledge about the role of perception of others’ pain in our social life.

Involuntary mental time travel (MTT) refers to the phenomenon of mentally reliving past experiences or pre-living possible future events in an involuntary form. Few studies have explored involuntary MTT in individuals with schizotypal personality features. The present study aimed to first explore the psychometric properties of the Involuntary Autobiographic Memory Inventory (IAMI) in a Chinese sample (Study 1), and then to explore whether individuals with schizotypal personality features experience involuntary MTT more frequently than individuals without schizotypal features. Moreover, the study explored whether the aberrant frequency of involuntary MTT is correlated with positive schizotypal features (Study 2). The results showed that the IAMI had good structural validity and reliability in a Chinese sample. Individuals with schizotypal traits reported a significantly higher frequency, less positive emotion, and stronger emotional intensity for both involuntary memories and future thoughts compared with individuals without schizotypal features. Further analyses in individuals with schizotypal personality features showed that the frequencies of both involuntary memories and future thoughts were significantly correlated with positive schizotypal traits. These results have potential theoretical and clinical implications for a comprehensive understanding of involuntary MTT among individuals with schizotypal personality features.

Cystic echinococcosis (CE), a zoonotic disease caused by Echinococcus granulosus at the larval stage, predominantly develops in the liver and lungs of intermediate hosts and eventually results in organ malfunction or even death. The interaction between E. granulosus and human body is incompletely understood. Exosomes are nanosized particles ubiquitously present in human body fluids. Exosomes carry biomolecules that facilitate communication between cells. To the best of our knowledge, the role of exosomes in patients with CE is not reported. Here, we isolated exosomes from the sera of patients with CE (CE-exo) and healthy donors and subjected them to liquid chromatography-tandem mass spectrometry analysis. Proteomic analysis identified 49 proteins specifically expressed in CE-exo, including 4 proteins of parasitic origin. The most valuable parasitic proteins included tubulin alpha-1C chain and histone H4. And 8 proteins were differentially regulated in CE-exo (fold changegt;1.5), as analyzed with bioinformatic methods such as annotation and functional enrichment analyses. These findings may improve our understanding about the interaction between E. granulosus and human body, and may contribute to the diagnosis and prevention of CE.

Interleukin-17A (IL-17A) and interleukin-17F (IL-17F) as two potent proinflammatory cytokines and the signature cytokines of Th17 cells play important roles in human autoimmune diseases, inflammation and host defenses. In this study, rhesus macaque IL-17A (rhIL-17A) and IL-17F (rhIL-17F) were cloned and expressed, and their biological activities and in vivo distribution were examined. The resulting data showed that both the rhIL-17A and rhIL-17F genes were consisted of three exons and two introns. RhIL-17A and rhIL-17F shared 96.8% and 93.9% amino acid sequence identity with human IL-17A (huIL-17A) and IL-17F (huIL-17F) respectively and the sequences also shared one N-glycosylation site and six conserved cysteine residues with huIL-17A and huIL-17F. IL-17A and IL-17F transcripts were highly expressed in lymphoid tissues and the intestinal tract of rhesus macaques. Functionally, recombinant rhIL-17A and rhIL-17F showed similar effect on Act1 levels and NF-κB phosphorylation compared with that of commercial human IL-17A and IL-17F. Moreover, the antibacterial proteins (such as β-defensin 2, S100A8, S100A9, RegIIIα and Muc1) and the tight junction associated genes (including CLDN1, CLDN4, OCLN, and ZO1) expressed by Caco-2 cells were largely enhanced after treatment with rhIL-17A and rhIL-17F. Meanwhile, purified rhIL-17A and rhIL-17F could also induce the expression of IL-6 and TNF-α by THP-1 cells. These data indicated that rhesus macaque IL-17A and IL-17F are highly similar to that of humans in both structure and function. Studies on rhIL-17A/rhIL-17F are promising approach to contribute to the understanding of human IL-17A and IL-17F-related intestinal diseases.

BACKGROUND: In the healthy body, the fascial system maintains elasticity and coordination of movements. If these functions are destroyed, facial paraly appears. Myofascial induction therapy (MIT), a manual physical therapy method that focuses on restoring altered fascial tissue, is prevalently and widely used in clinical treatment. OBJECTIVE: The study aimed to observe the application of MIT in the rehabilitation of patients with acute facial palsy. METHODS: Sixty-eight patients with acute facial palsy were divided into control group and manual treatment group. The control group received drug treatments, such as prednisone, methylcobalamin, and vitamin B1, and instrumental physical therapy, such as semiconductor laser, shortwave therapy, and facial muscle training. In addition to these treatments, the manual treatment group received MIT. Both groups were treated for 4 weeks. The patients were assessed using the following methods: the House-Brackmann facial nerve function evaluation, Sunnybrook facial grading system, facial nerve electrophysiological examination compound muscle action potential (CMAP) amplitude, and blink reflex (BR) R1 latency. Ferroptosis is a newly defined form of regulated cell death, which is involved in various pathophysiological conditions. However, the role of ferroptosis in male infertility remains unclear. In this study, 42 asthenozoospermic and 45 normozoospermic individuals participated. To investigate the ferroptosis level in the two groups, the levels of reactive oxygen species (ROS), malondialdehyde (MDA), and iron were measured, and mitochondrial membrane potential (MMP) was detected as an indicator of mitochondrial injuries. Compared with the normozoospermic group, ROS (p 0.05), MDA (p 0.001), and iron (p 0.001) of the asthenozoospermic group were significantly increased. However, the asthenozoospermia group had a decreased MMP level (p 0.05). In addition, the expression levels of GSH-dependent peroxidase 4 (GPX4) (p 0.001) and solute carrier family 7 member 11 (SLC7A11) (p 0.05) were also reduced in asthenozoospermic individuals. In asthenozoospermic samples, a significantly high positive correlation was observed between GPX4 mRNA levels and progressive motility (r = 0.397, p = 0.009) and total motility (r = 0.389, p = 0.011), while a negative correlation was observed between GPX4 and iron concentration (r = - 0.276, p = 0.077). The function of ferroptosis in asthenozoospermic males has never been studied before. In our study, we concluded that GPX4 and SLC7A11 expression levels in asthenozoospermia patients were related to increased ferroptosis and impaired sperm function, revealing novel molecular insights into the complex systems involved in male infertility.

Sex differences in behaviour and cognition have been widely observed, however, little is known about such differences in maintaining a balanced time perspective or their potential underlying neural substrates. To answer the above questions, two studies were conducted. In Study 1, time perspective was assessed in 1,913 college students, including 771 males and 1,092 females, and demonstrated that females had a significantly more balanced time perspective than males. In Study 2, 58 males and 47 females underwent assessment of time perspective and structural brain imaging. Voxel-based morphometry analysis and cortical thickness analysis were used to analyse the structural imaging data. Results showed that compared with males, females demonstrated a more balanced time perspective, which primarily related to lower grey matter volume in left precuneus, right cerebellum, right putamen and left supplementary motor area. Analysis of cortical thickness failed to reveal any significant sex differences. Furthermore, the sex difference in grey matter volume of left precuneus, right cerebellum, right putamen and left supplementary motor area could account for the difference in balanced time perspective between males and females. The findings deepen our understanding of sex differences in human cognition and their potential neural signature, and may inform tailored interventions to support a balanced time perspective in daily life.

Schizotypal traits have been found to be negatively associated with satisfaction with life but the underlying mechanisms are not well understood. This study investigated the association between schizotypal traits and satisfaction with life and explored the mediating role of trait anxiety and mind wandering in the relationship between those two variables in a sample of Chinese young adults. One hundred and two individuals with high schizotypal traits and 104 individuals with low schizotypal traits were screened using the Schizotypal Personality Questionnaire. They completed a series of questionnaires including the Satisfaction with Life Scale, Trait Anxiety Inventory, and Mind Wandering Questionnaire. Results showed that: First, the high schizotypal trait group showed lower satisfaction with life, and higher trait anxiety and mind wandering frequency than the low schizotypal trait group. Second, the high schizotypal trait group, trait anxiety, and mind wandering negatively predicted satisfaction with life. Third, mediation analyses showed that all indirect effect paths in the mediation model were significant, that is, trait anxiety and mind wandering alone and together mediated the relationship between schizotypal trait group and satisfaction with life. In conclusion, high schizotypal trait is a risk factor for low satisfaction with life. The association between schizotypal traits and satisfaction with life was mediated by the combination of trait anxiety and mind wandering. This study has implications for improving life satisfaction in individuals with high schizotypal traits.

With the progress of precision medicine treatment in pancreatic ductal adenocarcinoma (PDAC), individualized cancer-related examination and prediction is of great importance in this high malignant tumor, and antibody-dependent cell phagocytosis (ADCP) with changed pathways highly enrolled in the carcinogenesis of PDAC. High-throughput data of pancreatic ductal adenocarcinoma were downloaded and 160 differentially expressed ADCP-related genes (ARGs) were obtained. Secondly, GO and KEGG enrichment analyses show that ADCP is a pivotal biologic process in pancreatic carcinogenesis. Next, CALB2, NLGN2, NCAPG and SERTAD2 are identified through multivariate Cox regression. These 4 genes are confirmed with significant prognostic value in PDAC. Then, a risk score formula is constructed and tested in PDAC samples. Finally, the correlation between these 4 genes and M2 macrophage polarization was screened. Some pivotal differentially expressed ADCP-related genes and biologic processes, four pivotal subgroup was among identified in the protein-protein network, and hub genes was found in these sub group. Then, an ADCP-related formula was set: CALB2* 0.355526 + NLGN2* -0.86862 + NCAPG* 0.932348 + SERTAD2* 1.153568. Additionally, the significant correlation between M2 macrophage-infiltration and the expression of each genes in PDAC samples was identified. Finally, the somatic mutation landscape and sensitive chemotherapy drug between high risk group and low risk group was explored. This study provides a potential prognostic signature for predicting prognosis of PDAC patients and molecular insights of ADCP in PDAC, and the formula focusing on the prognosis of PDAC can be effective. These findings will contribute to the precision medicine of pancreatic ductal adenocarcinoma treatment.

Formononetin is an isoflavone compound found in many traditional Chinese medicines that has a wide range of pharmacological activities. It is critical to develop a sensitive, accurate, and rapid determination method for in-depth formononetin research. A molecularly imprinted electrochemical sensor was constructed in this study. After the flexible electrode (ITO-PET) was modified with nitrogen-doped graphene (NG), the molecularly imprinted polymer (MIP) was electropolymerized on the surface using o-phenylenediamine (OPD) as the functional monomer and formononetin as the template. Under optimized experimental conditions, the MIP sensor detected formononetin selectively in 0.1 M phosphate buffer solution (PBS) with the linear range of 3 ~ 120 μM, a detection limit as low as 1.14 μM (S/N = 3), and good anti-interference ability and reproducibility. The analytical performance of the proposed MIP/NG/ITO-PET was evaluated for the detection of formononetin in real samples such as methanol extract of Radix Astragali and mouse plasma with good accuracy and precision.

Autism spectrum disorders (ASD) are associated with cognitive dysfunctions, including mental time travel (MTT). However, findings on diminished MTT ability may be confounded by a number of factors, including the individuals' language ability, factors related to the MTT task and the demographic factors of participants. The present study provided a meta-analysis of MTT ability in people with ASD. The results showed significant overall reductions in MTT ability in people with ASD. Moderator analyses revealed that the variables examined did not explain the reduction in MTT ability. These findings suggest that MTT ability is diminished in people with ASD and that the degree of this diminishment may not depend on the characteristics of measures or demographic variables of people with ASD.

Repulsive guidance molecule A (RGMa) is a potent inhibitor of axonal growth and a regulator of neuronal cell death. It is up-regulated following neuronal injury and accumulates in chronic neurodegenerative diseases. Neutralizing RGMa has the potential to promote neuroregeneration and neuroprotection. Previously we reported that a rat anti-N terminal RGMa (N-RGMa) antibody r5F9 and its humanized version h5F9 (ABT-207) promote neuroprotection and neuroregeneration in preclinical neurodegenerative disease models. However, due to its cross-reactivity to RGMc/hemojuvelin, ABT-207 causes iron accumulation in vivo, which could present a safety liability. Here we report the generation and characterization of a novel RGMa-selective anti-N-RGMa antibody elezanumab, which is currently under Phase 2 clinical evaluation in multiple disease indications. Elezanumab, a human monoclonal antibody generated by in vitro PROfusion mRNA display technology, competes with ABT-207 in binding to N-RGMa but lacks RGMc cross-reactivity with no impact on iron metabolism. It neutralizes repulsive activity of soluble RGMa in vitro and blocks membrane RGMa mediated BMP signaling. In the optic nerve crush and optic neuritis models, elezanumab promotes axonal regeneration and prevents retinal nerve fiber layer degeneration. In the spinal targeted experimental autoimmune encephalomyelitis (EAE) model, elezanumab promotes axonal regeneration and remyelination, decreases inflammatory lesion area and improves functional recovery. Finally, in the mouse cuprizone model, elezanumab reduces demyelination, which is consistent with its inhibitory effect on BMP signaling. Taken together, these preclinical data demonstrate that elezanumab has neuroregenerative and neuroprotective activities without impact on iron metabolism, thus providing a compelling rationale for its clinical development in neurodegenerative diseases.

The aim of this study was to assess the effects of two rigid taping methods, tibial internal rotation taping and external rotation taping, on knee pain during three clinical function tests in individuals with patellofemoral pain syndrome.Repeated measures study.Fifteen participants (male, height 174.9 ± 5.8 cm, weight 68.3 ± 10.2 kg) with unilateral patellofemoral pain syndrome were treated by applying internal rotation taping and external rotation taping, in randomized order. While taped, participants were asked to perform three clinical tests in random order: single-leg squat, double-leg squat, and quadriceps maximum isometric contraction. Pain was scored using a Visual Analog Scale.Compared with no taping, both internal rotation taping and external rotation taping significantly improved pain during each of the three clinical tests (F = 224.10, p 0.001, partial η² = 0.94). Furthermore, there was a tape direction effect, where external rotation taping was more effective than internal rotation taping (F = 4.90, p = 0.044, partial η² = 0.26). For each test, the amount of pain improvement after taping was positively correlated with initial pain level (0.539 ≤ rho ≤ 0.921, all p 0.05).Both internal rotation taping and external rotation taping were effective in improving pain in individuals with patellofemoral pain syndrome, and external rotation taping was more effective than internal rotation taping. The significant correlations observed between initial pain levels and pain improvement after taping suggest that those with greater patellofemoral pain achieve greater relief from tibial rotation taping.

Endoplasmic reticulum stress (ERS) occurred in S63del mutant CMT1B mice model, and few drugs has been studied. Mesencephalic astrocyte-derived neurotrophic factor (MANF) can inhibit ERS. This study aimed at investigating the effect of MANF on ERS of RT4-D6P2T schwannoma cells with S63del MPZ Mutation.Experimental grouping: blank control group, blank control + MANF group, lentivirus group, lentivirus + MANF group, S63del MPZ group, S63del MPZ + MANF group. CCK8 and Annexin-FITC/PI were used to detect cell proliferation and apoptosis. JC-1 was used to detect ΔΨm. MANF, GRP78 and CHOP mRNA and protein were detected by using RT-qPCR, western blotting and immunofluorescence. ER-Tracker and mito-tracker were used to observe the morphology of endoplasmic reticulum (ER) and mitochondria.Cell proliferation decreased (ERS occurred in RT4-D6P2T cells with S63del MPZ mutation, and MANF exerted protective effect in RT4-D6P2T cells with S63del MPZ mutation.

A number of studies have demonstrated that decremental response to low frequency repetitive nerve stimulation (LF-RNS) is frequently observed in amyotrophic lateral sclerosis (ALS). However, due to the small sample size involved in previous studies, large discrepancies exist about the positivity rates of LF-RNS tests and factors affecting decremental response. This retrospective study of 449 cases, the largest sample size ever reported, shows that the overall positivity rate of LF-RNS is 49.9%. 3Hz RNS delivered to the accessory nerve has the best sensitivity and highest positivity rate. It obviously increases in response to upper limb onset, disease progression rate < 0.5 score/month, definite ALS and electromyography positive(EMG(+)) in sternocleidomastoid muscle. There is a linear increase in the decrement percentage of CMAP amplitude at 3Hz RNS delivered to the accessory nerve in response to longer disease duration, longer MUP duration and greater MUP duration increment percentage. These findings substantially advance the understanding of RNS results in ALS patients and effectively instruct clinical application.

Previous behavioral studies have shown that sharing painful experiences can strengthen social bonds and promote mutual prosociality, yet the neural mechanisms underlying this phenomenon remain unclear. We hypothesized that sharing a painful experience induces brain-to-brain synchronization and mutual empathy for each other's pain between pain-takers and pain-observers, which then leads to enhanced social bonding. To test this hypothesis, we adopted an electroencephalographic (EEG) hyper-scanning technique to assess neuronal and behavioral activity during a Pain-Sharing task in which high- or low-intensity pain stimulation was randomly delivered to one participant of a dyad on different experimental trials. Single-brain analysis showed that sensorimotor α-oscillation power was suppressed more when expecting high-intensity pain than when expecting low-intensity pain similarly for self-directed or partner-directed pain. Dual-brain analysis revealed that expecting high-intensity pain induced greater brain-to-brain synchronization of sensorimotor α-oscillation phases between pain-takers and pain-observers than did expecting low-intensity pain. Mediation analysis further revealed that brain-to-brain synchronization of sensorimotor α-oscillations mediated the effects of pain-stimulation intensity on mutual affective sharing for partner-directed pain. This mutual affective empathy during the task predicted the social bonding, as indexed by prosocial inclinations measured after the task. These results support the hypothesis that sharing a painful experience triggers emotional resonance between pairs of individuals through brain-to-brain synchronization of neuronal α-oscillations recorded over the sensorimotor cortex, and this emotional resonance further strengthens social bonds and motivates prosocial behavior within pairs of individuals.

Beginning with precise data on the ratio of structure functions in deep inelastic scattering (DIS) from $^3$He and $^3$H, collected on the domain $0.19 \leq x_B \leq 0.83$, where $x_B$ is the Bjorken scaling variable, we employ a robust method for extrapolating such data to arrive at a model-independent result for the $x_B=1$ value of the ratio of neutron and proton structure functions. Combining this with information obtained in analyses of DIS from nuclei, corrected for target-structure dependence, we arrive at a prediction for the proton's valence-quark ratio: $\left. d_v/u_v \right|_{x_B\to 1} = 0.230 (57)$. With the aim of extracting the pion charge radius, we analyse extant precise pion+electron elastic scattering data on $Q^2 \in [0.015,0.144]\,$GeV$^2$ using a method based on interpolation via continued fractions augmented by statistical sampling. The scheme avoids any assumptions on the form of function used for the representation of data and subsequent extrapolation onto $Q^2\simeq 0$. Combining results obtained from the two available data sets, we obtain $r_\pi = 0.640(7)\,$fm, a value $2.4\,\sigma$ below today's commonly quoted average. The tension may be relieved by collection and similar analysis of new precise data that densely cover a domain which reaches well below $Q^2 = 0.015\,$GeV$^2$. Considering available kaon+electron elastic scattering data sets, our analysis reveals that they contain insufficient information to extract an objective result for the charged-kaon radius, $r_K$. New data with much improved precision, low-$Q^2$ reach and coverage are necessary before a sound result for $r_K$ can be recorded., Comment: 5 pages, 4 figures

A sensitive and turn-on fluorescence nanoprobe based on core-shell Ag@Au nanoparticles (Ag@AuNPs) as a fluorescence receptor and red emissive graphene quantum dots (GQDs) as a donor was fabricated. They were conjugated together through π-π stacking between the GQDs and single-strand DNA modified at the Ag@AuNPs surface. The absorption spectrum of the receptor significantly overlapped with the donor emission spectrum, leading to a strong Forster resonance energy transfer (FRET) and thus a dramatic quenching. The sensing mechanism relies on fluorescence recovery following DNA cleavage by •OH produced from Fenton-like reaction between the peroxidase-like Ag nanocore and H2O2. The red emissive feature (Ex/Em, 520 nm/560 nm) provides low background in physiological samples. The •OH production, great spectrum overlapping, and red emission together contributes to good sensitivity and living cell imaging capability. The fluorescence assay (intensity at 560 nm) achieves a low detection limit of 0.49 μM H2O2 and a wide linear range from 5 to 200 μM, superior to most of the reported fluorescent probes. The RSD value for 100 μM H2O2 was 1.4%. The nanoprobe exhibits excellent anti-interferences and shows low cytotoxicity. The recovery of 100 μM standard H2O2 in a cancer cell lysate was 85.8%. Most satisfactorily, it can realize monitoring and imaging H2O2 in living cells. This study not only presents a sensitive H2O2 probe but also provides a platform for detecting other types of reactive oxygen species.

Carcinoembryonic antigen (CEA) is an important malign tumor marker. In this study, a simple, label-free and antibody-free aptasensor was fabricated based on a multifunctional dendrimer-like DNA nanoassembly. The DNA nanoassembly was embedded with multiple G-quadruplex DNAzyme motifs and a hanging CEA aptamer motif. It was prepared from short DNA sequences by autonomous-assembly. The aptasensor was prepared simply by self-assembly of a capture DNA (cpDNA) on a gold electrode, followed by hybridization with a CEA aptamer (AptGAC-P). CEA as a model target was detected through competitive binding of CEA with AptGAC-P, exposing cpDNA to bind with the DNA nanoassembly. The detection process only contains 2 incubation steps. The high load of G-quadruplex DNAzyme motifs and their catalytic activity resulted in an amplified and label-free differential pulse voltammetry (DPV) electrochemical signal. The peak current correlated linearly with the CEA concentration, with a linear range of 2–45 ng mL−1, and an LOD value of 0.24 ng mL−1. The aptasensor showed high specificity and reproducibility, and retained 96.5% of detection signal intensities after 31 days of storage. The recovery rates for spiked CEA in human serum were within 100 ± 5%, and the coincidence rates for clinical human serum samples with ELISA kits were 80.7–111%. Conceivably, possessing simplicity, sensitivity, reproducibility, storage stability, and accuracy, the aptasensor should be a very prominent and applicable tool for clinical CEA detection and cancer diagnosis, and is promisingly applicable as a platform for detecting other targets of interests.

Enduring pain would change individuals' behavioral preferences and neural responses in multiple decision-making tasks. Yet few studies have investigated how money's casual association with painful experience would modify people's decisions with it. It is an important and common social situation. The present study investigated how money's association with pain influences the way people make monetary decisions. Participants gambled with money that they earned in four different ways: enduring pain (Pain), randomly assigned (Random), non-painful effort task (Effort), and observing negative images (NO). Results revealed two different patterns. In the Random and Pain conditions, participants were not sensitive to the gambling risk such that they more randomly chose high- and low-risk options; the differences in FNR amplitude triggered by high- and low-risk choices were comparable on the neural level. In contrast, in the Effort and NO conditions, participants showed higher sensitivity to the magnitude and larger differences in FNR amplitudes between high- and low-risk choices. These findings suggested that pain cannot increase the subjective value of monetary gain like other non-painful efforts can do and monetary rewards may not be the optimal way to compensate for the physical suffering or loss in society.

To address the issue of poor corrosion resistance of the Mg–Li alloy, electroless Ni–P plating was used to create a protective coating. However, there were significant differences between the upper and lower surface coatings, which were summarized as follows: (1) compared with the lower surface, the longitudinal differences between different areas of the upper surface coating were larger; and (2) the denseness of the upper surface coating was insufficient in areas where the insoluble phase was concentrated, resulting in significantly lower corrosion resistance of the upper surface coating than the lower surface. Resolving these differences could compensate for the defects of the upper surface coating so as to improve the overall corrosion resistance of the material. Therefore, in this paper, the deposition process of Ni–P was observed and speculated, and the reasons for these differences were analyzed in combination with experimental phenomena. Based on these, two optimization measures were proposed. The SEM observation results showed that the differences between the upper and lower surface coatings were significantly reduced after optimization. The results of potentiodynamic polarization tests and EIS tests showed that the optimized upper surface coating had good corrosion resistance similar to the lower surface coating.

Agricultural soils, which are closely linked to human health via food supply, have been recognized as an important reservoir for antibiotic resistance genes (ARGs). However, there is still a lack of knowledge regarding the role of soil aggregates in shaping ARG profile. In this study, we collected soils from long-term experimental farmland plots receiving inorganic and/or organic fertilizers and examined the patterns of antibiotic resistome distribution among differently sized soil aggregates using high-throughput quantitative polymerase chain reaction (HT-qPCR). Our results showed that the distribution of soil ARGs could be affected by manure application and aggregate size individually but not interactively. More diverse and abundant ARGs were found in the manured soils, compared to the non-manured soils. The aggregate size fraction of53 μm exhibited the highest diversity and abundance of ARGs. Variation partitioning analysis revealed that soil traits, mobile genetic elements, and bacterial community collectively contributed to the variation of soil antibiotic resistance. The knowledge about aggregation stratification of soil ARGs obtained in this study is fundamental and essential to understanding the fate of soil ARGs at the microscale.
