Neurotensin as Modulator of Basal Ganglia- Thalamocortical Motor Circuit – Emerging Evidence for Neurotensin NTS1 Receptor as a Potential Target in Parkinson's Disease

La scoperta dei polimeri conduttori ha offerto la possibilità di realizzare celle solari di plastica, flessibili e con bassi costi di produzione, che potranno cambiare gli scenari futuri in campo fotovoltaico. Lo studio alla scala atomica di questi materiali strutturalmente e chimicamente complessi, richiede lo sviluppo di modelli teorici e l’uso intensivo del supercalcolo, al fine di migliorarne le proprietà di fotoconversione e renderli tecnologicamente competitivi.

A seguito del recente credit crunch, le principali istituzioni finanziarie stanno abbandonando la prassi di aggiornare la propria posizione di rischio solo durante la notte, implementando soluzioni intra-day, che siano in grado non solo di valutare i rischi di liquidità, di mercato e di controparte per l’intera istituzione, ma anche di discriminare criticità legate a diverse classi di asset finanziari e/o associate al business di singole unità operative. Ottenere questo tipo di risultati, per di più in tempo reale, richiede la soluzione di complessi modelli numerici che elaborino grosse moli di dati di mercato; pertanto per gli operatori del settore è ormai imprescindibile il ricorso alle più sofisticate piattaforme di calcolo ad alte prestazioni oggi disponibili sul mercato, tanto che alcune importanti istituzioni finanziarie internazionali hanno installato grossi cluster per HPC, utilizzando anche la tecnologia delle GP-GPU.

The importance of using photovoltaics (PVs) to obtain clean and reliable energy hasbeen widely recognized. The scientific research on new PV materials is crucial to identifynew approaches to the concept of solar energy conversion and to increase the photonversionefficiency. Among the different approaches for PV, the quantum dot (QD) solarcell is one of the most promising. A single layer QD consists of a distribution ofnanocrystals embedded into an amorphous dielectric matrix. It is in principle possibleto affect the optoelectronic properties of QD layers by controlling the morphology ofthe nanograins. The complex atomic scale structure of such materials makes it quitedifficult to predict theoretically their optoelectronic properties. The theoretical challengeis two-fold; on the one hand, it is necessary to generate large scale atomistic models;on the other hand, the corresponding electronic properties must be computed over alarge number of atoms. While the first issue can be coped with large-scale moleculardynamics (MD) simulations, the second issue is a major computational bottleneck andit requires a class of advanced algorithms for the electronic structure calculations inwhich the computational workload scales linearly with the number of atoms. Within acollaboration among CASPUR and the SLACS (Sardinian Laboratory for Computationalmaterials science) we have implemented a new numerical method to investigate theoptoelectronic properties of silicon based QD layers. By using such a theoretical frameworkit is possible to assist the design of new efficient materials for PV.

In view of the ability of neurotensin (NT) to increase glutamate release, the role of NT receptor mechanisms in oxygen-glucose deprivation (OGD)-induced neuronal degeneration in cortical cultures has been evaluated by measuring lactate dehydrogenase (LDH) levels, mitochondrial dehydrogenase activity with 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide levels, and microtubule-associated protein 2 (MAP2) immunoreactivity. Apoptotic nerve cell death was analyzed measuring chromatin condensation with Hoechst 33258, annexin V staining, and caspase-3 activity. Furthermore, the involvement of glutamate excitotoxicity in the neurodegeneration-enhancing actions of NT was analyzed by measurement of extracellular glutamate levels. NT enhanced the OGD-induced increase of LDH, endogenous extracellular glutamate levels, and apoptotic nerve cell death. In addition, the peptide enhanced the OGD-induced loss of mitochondrial functionality and increase of MAP2 aggregations. These effects were blocked by the neurotensin receptor 1 (NTR1) antagonist SR48692. Unexpectedly, the antagonist at 100 nM counteracted not only the NT effects but also some OGD-induced biochemical and morphological alterations. These results suggest that NTR1 receptors may participate in neurodegenerative events induced by OGD in cortical cultures, used as an in vitro model of cortical ischemia. The NTR1 receptor antagonists could provide a new tool to explore the clinical possibilities and thus to move from chemical compound to effective drug.

It is proposed that the co-expression of statistically significant motifs among the sequences of a proteome is a phylogenetic trait. From the co-expression matrix of such motifs in a group of prokaryotic proteomes a suitable definition of a phylogenetic distance is introduced and the corresponding distance matrix between proteomes is constructed. From the distance matrix a phylogenetic tree is inferred, following a standard procedure. It compares well with a reference tree deduced from a distance matrix obtained from the alignment of ribosomal RNA sequences. Our results are consistent with the hypothesis that biological evolution manifests itself with a modulation of basic correlations between shared peptides of short length, present in protein sequences. Moreover, the simple procedure we propose reconfirms that it is possible, sampling entire proteomes, to average the effects of lateral gene transfer and infer reasonable phylogenies., Comment: Minor typos corrected, partly rewritten conclusions and bibliography added
