THIS PAPER ILLUSTRATES THE VALUE of strategic alignment as a model to navigate the current Australian education landscape, highlighting numerous changes to national overseeing bodies and their mandates. More importantly, it provides a critical review of recent government agency changes and policy agendas, making overt the existence of complex and incoherent ideas concerning the notion of early childhood. Based on the current analysis, it is concluded that no common conceptualisation exists at the national level of what constitutes early childhood and by default, early childhood education. This is a disturbing state of affairs, which makes the work of responsible statutory agencies, overseeing national bodies and state departments overseeing the implementation of new national policy directives very difficult. more...

Learning design research (LDR) is establishing itself as a separate and specialised field of educational research. Worldwide, technology-mediated learning experiences in higher and further education are on the increase. LDR investigates their success in providing effective outcomes-based and personalised learning experiences. This paper reports on a meta-ethnographic review study of a subsection of current LDR to document current conceptualisations, preferred methodologies and possible themes and struggles. The findings suggest that LDR often uses an ad hock approach to Learning Design conceptualisation. The future of assessment faces major challenges including the use of IT to facilitate formative assessment that is important for improving learners’ development, motivation and engagement in learning. In many countries, in recent years, a renewed focus on assessments to support learning has been pushing against the burgeoning of testing for accountability, which in some countries, renders effective formative assessment practices almost impossible. Moreover, a systematic review by Harlen and Deakin Crick (2002) revealed that a strong focus on summative assessment for accountability can reduce motivation and disengage many learners. At the same time use of IT‐enabled assessments has been increasing rapidly, as they offer promise of cheaper ways of delivering and marking assessments as well as access to vast amounts of assessment data from which a wide range of judgements might be made about students, teachers, schools and education systems (Gibson & Webb, 2015). These opportunities also extend to assessment of complex collaborative work (Webb & Gibson, 2015). Current opportunities for using IT, including for harnessing the data that is being collected automatically, for formative assessment are underexplored and less well understood than those for summative assessments. Opportunities for learning with IT and perhaps with less teacher input are increasing but this depends on students developing as autonomous or independent learners. Research in formative assessment including effective feedback has emphasised the value of peer assessment practices for developing self‐assessment capabilities and hence independent learners (Black, Harrison, Lee, Marshall, & William, 2003). This paper explores the use of transmedia pedagogy in a massive open online course (MOOC) environment for work integrated learning (WIL). Using a communication course as a case study, the paper seeks to understand how the use of transmedia pedagogy and a MOOC environment may create a democratic space for discovery learning and how it can improve connectedness between students, their peers and their supervisors (university and workplace). This conceptual paper forms the foundation of a longitudinal research project that seeks to understand to what extent transmedia pedagogy can enhance student connectedness and learning outcomes in a work integrated learning (WIL) MOOC environment. Transmedia pedagogy is relatively new and has been noted to create memorable learning through its narrative focus (Raybourn, 2014). Students have an opportunity to become active participants in their learning, and part of the educational environment that sees curriculum content created, shared and re-purposed by students as well as their educators. A MOOC environment, together with transmedia pedagogy may be able to bridge the gap of the connection between students and their resources, their educators and their peers. In theory, the transmedia pedagogy MOOC environment may provide opportunities for the creation of a holistic, democratic learning experience, which becomes particularly important for WIL courses where students do not attend classes. The MOOC re-imagined as a community encourages dialogue, interaction and the meeting of the personal and social aspects of learning (Kop, 2011). The synergistic relationship between learning design and learning analytics has the potential for improving learning and teaching in near real-time. The potential for integrating the newly available and dynamic information from ongoing analysis into learning design requires new perspectives on learning and teaching data processing and analysis as well as advanced theories, methods, and tools for supporting dynamic learning design processes. Three perspectives of learning analytics design provide summative, real-time, and predictive insights. In a case study with 3,550 users, the navigation sequence and network graph analysis demonstrate the potential of learning analytics design. The study aims to demonstrate how the analysis of navigation patterns and network graph analysis could inform the learning design of self-guided digital learning experiences. Even with open-ended freedom, only 608 sequences were evidenced by learners out of a potential number of hundreds of millions of sequences. The aim of the study was to explore the tensions, as experienced by student teachers, in a teacher education program that is based on psychodynamic theory. Previous research shows that students have had orientation problems in innovative university studies because it involves students having to take steps toward their discomfort zone. It was therefore considered important to study the mismatch or match of the training model and student experiences. The data were gathered through student interviews. The data analysis utilized a case study methodology and proceeded from thematic network analysis toward a deeper interpretation of the data. The analysis shows students struggle to understand the meaning and practices of psychodynamic-oriented pedagogy. The analysis also shows that students were unable and unwilling to share their learning experiences with students outside the new program. © 2017 IEEE. This paper reports on an investigation into perceptions of international university lecturers and their concerns about students' social and academic motivation for learning in online learning environments. The study results suggest that pedagogic lurking is a widespread phenomenon, with some perceiving it as problematic and others as a step towards more active participation. Most interestingly, there does seem to be a lack of appetite for learning analytics software that would allow the close monitoring of student learning engagement and progress. Az Opus et Educatio 2. évfolyam 3. számában jelent meg – amely az alábbi linken érhető el: http://opuseteducatio.hu/index.php/opusHU/article/view/60/83) - „A diákok mindennapjai, a tanulói munkaterhek vizsgálata az érettségi előtti és utáni 2-2 évfolyamon” című publikációm. Itt megvizsgáltam a diákok mindennapjait 455 diák bevonásával, 4 régió 6 városában. Kérdőívemben felmértem az iskolában és az iskolán kívül a tanulás érdekében fordított időt - a közlekedésre, a szakkörön, emelt szintű oktatáson, a korrepetáláson töltött időt, és még a délutáni tanulást is – az érettségi előtti 11-12. évfolyamon, valamint az érettségi utáni 13-14. ún. szakképző évfolyamokon.Jelenlegi tanulmányom a fenti téma folytatása, mélyebb kutatása a „diákmunkaidő” területén, mégpedig a 12. évfolyamon belül az átlagos tanulmányi eredménytől felfelé és lefelé eltérő képességű közgazdasági szakmacsoportos tanulók esetében. Ezen belül az általuk az iskolában és az iskolán kívül a tanulásra fordított időt hétköznap, hétvégeken, tanórákon valamint tanórán kívül elemzem. Ezután megvizsgálom, hogy ezzel a tanulásra fordított idővel hogyan teljesítettek az országos versenyeken, a tanév végén a közép- és emelt szintű érettségi vizsgákon, és azt, hogy az elért eredményekkel milyen oktatási intézményekben tudták felsőfokú tanulmányaikat folytatni. Az eredmény megdöbbentő, hogy mennyi tanulást, lemondást, kitartást vár el az élet már 18-19 évesen az átlagtól felfelé eltérő képességű diákoktól, akik meg szeretnék valósítani az álmaikat, vágyaikat. De az átlagtól lefelé eltérő képességű tanulók esetében is elgondolkodtató a kutatásom eredménye.***My publication titled “The everyday lives of students – an examination of the pupils’ school burdens on the two grades before and after their school-leaving exam” was published in Opus et Educatio Vol. 2. With the involvement of 455 pupils of six cities in four regions I surveyed pupils’ everyday lives. With my query I investigated the time spent on learning within and outside the school – also the time spent on transportation, study circles, advanced-level education, tuition and even studying at home – on the grades 11-12 preceding the school-leaving exam and grades 13-14 following it (the so-called vocational training grades). This paper is a continuation of the topic described above, it is a deeper research in the field of “pupils’ working hours” in the case of the pupils studying economic trade groups and with learning results deviating from the average upwards or downwards. I analyse the time they spend on learning inside and outside school, on weekdays, weekends, classroom lessons and outside those. Afterwards, I examine how they performed at national competitions, at the basic and advanced level school-leaving exams and which institutions of higher education they were accepted at. The consequences are astonishing – how much studying, resignation and persistence life requires of the 18-19-year-old students whose abilities deviate from the average upwards and who want to realize their dreams and wills. The Larnaca Declaration on Learning Design arose from a 2012 meeting of experts in Larnaca, Cyprus who sought to provide a new theoretical foundation for the field of Learning Design, based on a synthesis of research and practice in the field to date. It begins by acknowledging the vast benefits that would arise from wider sharing of effective teaching practices, and it uses an analogy from the history of music notation to identify the need for a representational framework for describing teaching and learning ideas. It provides a range of examples of learning designs and different representational systems to illustrate the need for a “Learning Design Framework” (LD-F). It then acknowledges the wider educational context and its impact on design decisions by educators, and provides a “Learning Design Conceptual Map” (LD-CM) to identify contextual components and their interactions in design decisions. It concludes by discussing the philosophical and practical challenges of identifying and sharing effective teaching and learning ideas under the heading “Learning Design Practice” (LD-P). more...

PRINS, a noncoding RNA identified earlier by our research group, contributes to psoriasis susceptibility and cellular stress response. We have now studied the cellular and histological distribution of PRINS by using in situ hybridization and demonstrated variable expressions in different human tissues and a consistent staining pattern in epidermal keratinocytes and in vitro cultured keratinocytes. To identify the cellular function(s) of PRINS, we searched for a direct interacting partner(s) of this stress-induced molecule. In HaCaT and NHEK cell lysates, the protein proved to be nucleophosmin (NPM) protein as a potential physical interactor with PRINS. Immunohistochemical experiments revealed an elevated expression of NPM in the dividing cells of the basal layers of psoriatic involved skin samples as compared with healthy and psoriatic uninvolved samples. Others have previously shown that NPM is a ubiquitously expressed nucleolar phosphoprotein which shuttles to the nucleoplasm after UV-B irradiation in fibroblasts and cancer cells. We detected a similar translocation of NPM in UV-B-irradiated cultured keratinocytes. The gene-specific silencing of PRINS resulted in the retention of NPM in the nucleolus of UV-B-irradiated keratinocytes; suggesting that PRINS may play a role in the NPM-mediated cellular stress response in the skin. more...

221. Decongestion improves cell-mediated immunity in postmastectomy arm lymphoedema: a pilot study

Background Chronic lymphoedematous limbs have an increased propensity for infections and primary or secondary malignant tumours. It has been attributed to suppressed delayed-type hypersensitivity measured in lymphoedemas related to Stewart–Treves syndrome, Kaposi’s sarcoma or breast cancer treatment. Cell-mediated immunity is an effective defence mechanism against bacteria, fungi, viruses and tumour cells. Objective We aimed to examine whether decongestive lymphoedema therapy could improve cell-mediated immunity in breast cancer treatment-related lymphoedema (BCRL). Methods Eight women with unilateral BCRL were included in this study. At baseline, tuberculin skin test (TST) was performed on the volar surfaces of the forearms of the affected and non-affected sides using 0.5, 1 and 5 tuberculin units in the form of three consecutive injections with 3-cm spaces in-between, and arm volumes were measured using the Kuhnke’s disc model. Decongestive lymphatic therapy was given to swollen arms in 10 consecutive working days. At the end of intensive decongestion, TST on affected side and bilateral volumetry were repeated. The global call for teacher quality improvement and numerous accounts of resistance to education reform at all levels of the education system brings to the forefront the tension between rhetoric and reality. This case study reports on a failed innovation attempt, which was based on the need for a signature pedagogy in Australian teacher education that better prepares beginning teachers for the demands of flexible, student-centred learning design. To assist teacher education students’ development of deep learning engagement, which is a pre-condition for the acquisition of 21st century knowledge, skills and learning attitudes, we need to better understand resistance behaviour. The reported reserach illustrates how the learning-centric teaching design was unable to engage ‘consumer students’ in deep learning experiences due to heightened negative emotion experienced by a great number of students. The provision of this illustrative practical example of innovation failure has the potential to make apparent how students’ ‘out-of-comfort-zone’ behaviour and resistance to change from transmission education practices to social constructivist approaches will need to be managed. Abstract: This paper examines the use of photography in Raymond Carver, both in the short story, “Viewfinder,” and in the biographical photo-essay, Carver Country, as a metaphor for the political implications of the author's aesthetic. Photodynamic therapy is based on the selective accumulation of a photosensitizer in tumors, followed by destruction of the target tissue by a light source. Protoporphyrin IX, a well-known photosensitizer, was recently reported as an endogenous substrate for the multidrug transporter ABCG2. We investigated the role of ABCG2 protein in the porphyrin extrusion ability of keratinocytes, with regard to the impact of the specific inhibition of ABCG2 by a non-toxic fumitremorgin C analog, Ko-134, on photodynamic therapy efficacy. We studied the level of porphyrin accumulation in response to delta-aminolevulinic acid pretreatment in proliferating and highly differentiated HaCaT keratinocytes. An in vitro model of photodynamic therapy on HaCaT cells was established with a therapeutically approved narrow-bandwidth red-light source. The porphyrin extrusion ability of HaCaT cells proved to correlate with their ABCG2 expression which was higher in proliferating cells than in differentiated cells. Moreover, the specific inhibition of ABCG2 by Ko-134 enhanced the sensitivity of keratinocytes to photodynamic therapy in vitro. These results suggest that ABCG2 may serve as a target molecule via which to improve the photodynamic therapy of skin lesions: its inhibition by the non-toxic Ko-134 is a promising therapeutic modality. more...

A semester-long trial was conducted in which library pod/vodcasts were provided to first-year teacher education students. Psoriasis Susceptibility-Related RNA Gene Induced by Stress (PRINS) is a non-coding RNA overexpressed in lesional and non-lesional psoriatic epidermis and induced by stress. Its function in healthy and psoriatic skin is still not known. Here, we report that PRINS regulates G1P3, a gene with anti-apoptotic effects in keratinocytes. siRNA-mediated inhibition of PRINS gene resulted in altered cell morphology and gene expression alterations, as demonstrated in a microarray experiment. One of the genes regulated by PRINS ncRNA was G1P3, an interferon-inducible gene with anti-apoptotic effects in cancer cells. Interestingly, we found that G1P3 was 400-fold upregulated in hyperproliferative lesional and ninefold upregulated in non-lesional psoriatic epidermis compared to healthy epidermis. In vitro, G1P3 protein levels were highest in proliferating keratinocytes and siRNA-mediated downregulation of G1P3 resulted in increased cell apoptosis. These data indicate that G1P3 inhibits spontaneous keratinocyte apoptosis and hence its high expression in psoriatic skin may contribute to the development of psoriatic lesions. In this paper, the five interlocking de Bono LAMS sequences are introduced as a new form of generic template design. This transdisciplinary knowledge-mobilising strategy is based on Edward de Bono's attention-directing ideas and thinking skills, commonly known as the CoRT tools. The development of the de Bono LAMS sequence series is an important milestone, signifying the current paradigmatic shift in higher education, from a student-consumer paradigm to a student-producer paradigm. Surpassing surface and shallow knowledge stages requires the use of multidisciplinary and generic knowledge in new and unfamiliar situations. The proportion of T lymphocytes in the peripheral blood, their phytohae-magglutinin reactivity, and the leycocyte migration inhibitory effect of purified tuberculin protein were found to be equal in 25 recurrent corneal herpes simplex patients and 17 healthy individuals who had never suffered from clinically manifest herpes simplex virus (HSV) infection. Conversely, the HSV vaccine inhibited the leucocyte migration in the healthy group, but not in the recurrent herpes patients when the clinical symptoms were manifest. In 14 recurrent corneal herpes simplex patients, the examinations were repeated after a 3-month symptom-free period, and in these periods the patients corresponded equally to the healthy group in every respect. more...

Summary: The synergism between the polymorphonuclear granulocytes and ketoconazole was studied in vitro. The phagocyte index proved to be 87.3 + 8.1. while the Candida killing index was 20.8 + 4.8. Treatment of granulocytes with ketoconazole did not influence their activity. However, the treatment of Candida albicans with ketoconazole led to unchanged phagocytosis, but a significantly higher killer activity. The results indicate that not the immunomodulating effect of the imidazole derivative, but its antimycotic effect, is responsible for the synergism observed between ketoconazole and the host defence mechanisms. Zusammenfassung: Der Synergismus zwischen polymorphonuklearen Granulozyten und Ketoconazol wurde in der vorliegenden Studie in vitro untersucht. Die Bestimmung des Phagozytosenindex ergab einen Wert von 87,3 + 8,1. und beim Candida-Abtotungsindex 20,8 + 4,8. Die Behandlung der Granulozyten mit Ketoconazol lies die Aktivitat dieser Zellen unbeeinflust. Wird der Candida-Suspension Ketoconazol zugesetzt, beeinflust es die Phagozytose der Candida-Zellen nicht, sondern erhoht signifikant die candidacide Aktivitat. Summary: In 18 patients suffering from diabetes mellitus and in 15 healthy volunteers the Candida killing activity of polymorphonuclear leukocytes was tested. Granulocytes from diabetes patients exhibited a significantly depressed Candida killing activity as compared with that of healthy volunteers. Sera from 8 patients with diabetes mellitus failed to influence the Candida killing activity of granulocytes from healthy persons. Further in diabetes mellitus proportion of polymorphonuclear leukocytes capable of forming rosettes with complement-coated zymosan beads (C 3 rosette-forming cells) did not differ from that in the healthy group. The depressed intracellular yeast killing, which seems to be independent of serum factors, may obviously play a significant role in the higher susceptibility of diabetics to Candida infections. Zusammenfassung: Wir untersuchten bei 18 Patienten mit Diabetes mellitus und bei 15 gesunden Vergleichspersonen die Candida-abtotende Aktivitat von polymorphkernigen Leukozyten. Die Granulozyten von diabetischen Patienten zeigten eine signifikant verminderte Abtotungs-Aktivitat im Vergleich zu der von gesunden Kontrollpersonen. Ein gepooltes Serum von 8 Patienten mit Diabetes mellitus hatte keinen Einflus auf die Candida-abtotende Aktivitat von Granulozyten gesunder Personen. Bei Diabetikern und Gesunden gibt es keinen Unterschied hinsichtlich des Anteils der Granulozyten, die in der Lage sind, Rosetten mit komplement-beladenen Zymosanpartikeln (C 3 Rosetten-bildende Zellen) zu bilden. Die verminderte intra-zellulare Hefezell-Abtotung, die vermutlich unabhangig von Serumfaktoren ist, spielt eine bedeutsame Rolle bei der hoheren Empfanglichkeit von Diabetikern fur Candida-Infektionen. more...

Background Malignant melanoma is an increasing public health problem worldwide; accordingly, identification of the constitutional and environmental factors which contribute to the development of the disease, and hence identification of the individuals at high risk of melanoma, is an indispensable step in all primary prevention efforts. Objectives This paper aims to assess the prevalence of different pigmented lesions among schoolchildren and to investigate their relationship with phenotypic pigmentary characteristics, sun exposure and other factors. Patients/methods A cross-sectional study was performed in two secondary schools in Szeged, Hungary. A total of 1320 schoolchildren, aged 14 to 18 years, underwent a whole-body skin examination. A standardized questionnaire was used to collect data on phenotypic, sun exposure and other variables. Results One to 10 common melanocytic naevi were found in 27% of the participants, and the naevus numbers were in the range of 10–100 in 67%; 5.4% of them had more than 100 common melanocytic naevi. The prevalence of clinically atypical naevi was 24.3%. Statistically significant associations were found between the number of pigmented lesions and gender, hair colour, eye colour, skin phototype, a history of severe painful sunburns and a family history of a large number of melanocytic naevi. Conclusion Our study population displayed a markedly high prevalence of clinically atypical melanocytic naevi. Moreover, a considerable proportion of the investigated individuals had multiple common melanocytic naevi. Since the presence of a large number of melanocytic naevi is a strong predictor for future melanoma development, health educational programmes on melanoma prevention should be aimed at young age groups. more...

Syndecan 4 (SDC4), a heparan sulfate proteoglycan, and neuropilin 1 (NRP1), a transmembrane receptor, are both involved in normal wound healing, but little is known about their possible role in venous leg ulcer pathogenesis. We aimed to investigate whether there are any expression abnormalities and/or gene polymorphisms of SDC4 and NRP1 associated with venous leg ulcer. SDC4 showed significantly lower mRNA and protein expression in the uninvolved dermis of venous leg ulcer patients (n=15) compared with controls (n=15; p=0.0136), while NRP1 showed no expression abnormalities. None of the examined SDC4 and NRP1 polymorphisms showed a difference in their allelic distribution between leg ulcer patients (n=92) and controls (n=92). We hypothesize that SDC4 may play an essential role not only in the inflammation and tissue formation phases of normal wound healing, but its expression abnormalities observed in the uninvolved dermis of venous leg ulcer patients may contribute to venous leg ulcer development. more...

In this study, we show that the G0-G1/S phase of HaCaT keratinocyte cell cycle is characterized by D1-type cyclin expression, whereas during the repeated rapid turnover of highly proliferating cells, the expression of cyclins D2 and D3 dominates. Knocking down cyclin D1 mRNA resulted in no change of cell proliferation and morphology, indicating that D2 and D3 cyclins could substitute for D1 in driving the cell cycle. Increased numbers of cyclin D1-expressing keratinocytes were found in the basal layers of the lesional psoriatic epidermis compared to both normal and non-lesional epidermis without increased expression of cyclin D1 mRNA, suggesting a possible dysfunction in the degradation of cyclin D1 protein. We also detected a significant increase in cyclin D2 and D3 mRNA expressions in psoriatic epidermis compared to normal epidermis with no difference in protein expressions. Blocking alpha5-integrin function by a neutralizing antibody in HaCaT keratinocytes downregulated the expression of cyclin D1 mRNA without affecting the expressions of cyclin D2 and D3 indicating a regulatory role for alpha5-integrin in the expression of cyclin D1. Our data suggest a possible role for D-type cyclins in the excessive basal-cell proliferation and perturbed keratinocyte differentiation in the psoriatic epidermis. more...

SUMMARY. The aim of our study was to investigate the incidence of duodeno-gastroesophageal reflux-induced malignant transformation in a series of duodeno-esophageal anastomosis operations in rats. This surgical method provides a model for reflux-induced esophageal pathologies, without carcinogen administration. The study design included the follow-up of 31 cases. Thirty weeks of duodeno-gastroesophageal reflux disease significantly increased the risk of the development of Barrett's esophagus, and reflux-induced esophageal adenocarcinoma formation was evident in four animals. In one of these particular cases, a superficial squamous cell cancer was noted in close vicinity to the adenocarcinoma formation. For further analysis, a detailed immunohistochemical staining protocol was used. The immunophenotypes revealed cyclin D1 expression (nuclear positivity in 35% of all the squamous cells), p53 protein accumulation (50% nuclear positivity), with a low expression of cox-2, and negative c-erbB2 staining in the squamous carcinoma cells. The specialized intestinal metaplasia and mucinous adenocarcinoma cells exhibited exclusively diffuse cox-2 positivity (90% of all glandular cells) and weak focal c-erbB2 (5%) staining, without cyclin D1 expression or p53 protein accumulation. Real-time polymerase chain reaction was applied to quantify the abundance of p53, cyclin D1 and cox-2 mRNAs in this biopsy. The most dramatic changes were observed in the level of expression of cyclin D1 (a 9.08-fold expression as compared with the non-treated esophagus samples), while the p53 and cox-2 gene expressions were increased by 1.61 and 2.45-fold, respectively, relative to the non-treated samples. The results afford evidence of the simultaneous activation of more than one possible carcinogenetic pathway in experimental gastroesophageal reflux disease. The P48T germ line mutation of p16 was detected in a Hungarian multiple primary melanoma patient (deceased at the age of 39) with no affected family members. Genetic analysis of the patient and his family revealed that the patient was homozygous for the mutation, whereas his parents (father currently aged 69 and mother 63), who are free from any malignancies and atypical moles, are both heterozygous for the mutation. Our data suggest that the P48T mutation of p16 is a strong melanoma-predisposing factor, but the fact that the heterozygous mutant parents have not yet exhibited melanoma or atypical moles indicates that the penetrance of this allele might depend on modifying factors. The rare P48T germ line mutation of p16 has been reported previously in only four independent studies, all in patients with Italian ancestry. Here, we first report the inheritance of the rare P48T mutation of CDKN2A in a Hungarian family with a homozygous multiple primary melanoma member and unaffected heterozygous family members. The question of whether the mutation detected in Hungary is the result of an independent event, or migration of the founder mutation occurred at some time in the past, necessitates further investigations. more...

In this paper, I discuss some research findings regarding the characteristics that democratic schools appear to have in common. These commonalities seem to have contributed to their status as being seen as reputable democratic schools. For the purposes of the reported study, schools that were diverse in their philosophical approaches to education and socio‐economic composition were selected as case‐study schools. A specific selection criterion was that these schools had a reputation for nurturing the critical capabilities of students with an explicit ‘citizenship framework’. Students were not seen as ‘objects to be acted upon’, but rather were trusted to be subjects of rights and responsibilities within the school community in some form or other. Ultraviolet B (UV-B) radiation is a modality widely used for the treatment of different skin diseases. One of the major mechanisms of UV-B immunosuppression in this treatment modality is thought to be an apoptosis-inducing effect on T cells infiltrating the skin. We examined the T-cell apoptosis-induction capacities of four different UV light sources, with and without UV filters. The xenon chloride (XeCl) laser proved to be the strongest apoptosis inducer. The use of a phtalic acid filter eliminated UV radiation almost completely below 300 nm, which resulted in a severe decrease in the apoptosis-inducing capacity of different UV-B sources. Using the results of the measurements with polychromatic UV light sources, the wavelength dependence of UV-B light for the induction of T-cell apoptosis was also determined. The regression line of the action spectrum demonstrated a continuous decrease from 290 to 311 nm. The apoptosis-inducing capacity of the XeCl laser was almost four times higher than the calculated value according to the action spectrum, which might be attributed to the high irradiance of the laser as compared with nonlaser light sources. more...

We describe a novel chemical mitogen-free in vitro culture technique for obtaining pure melanocyte cultures using normal human adult epidermis as a source. The culture medium consists equal parts of the commercially available Keratinocyte Basal and AIM-V media (both from Gibco), as basal medium, which is supplemented with fetal bovine serum, bovine pituitary extract and recombinant human epidermal growth factor (EGF). Melanocytes harvested from human adult skin proliferate extensively and can be passaged serially up to 10-15 times using this medium. We have verified the identity of the cultured cells by tyrosinase mRNA expression and TRP-1 protein staining. Moreover, we showed that autologous human serum alone, without additional supplements is able to provide sufficient growth support for the cultured cells in the basal medium, making this culture technique suitable for autologous melanocyte transplantation. In this culture system normal human adult melanocytes expressed both EGF receptor (EGFR) mRNA and protein and EGF showed a dose dependent mitogenic effect on the cells. The primary objective of this open-label study was to assess the long-term safety of tacrolimus ointment. The primary end-point was the incidence of adverse events. Secondary end-points included the Eczema Area and Severity Index and a modified version of this index. A total of 466 children with atopic dermatitis, aged 2-15 years, applied 0.03% or 0.1% tacrolimus ointment twice daily for up to 29.5 months. Skin burning and pruritus were the most common application site events; their prevalence decreased over time. There was no increase in viral infections or other adverse events over time. Laboratory profiles were consistent with those reported in atopic populations. Substantial improvement in all efficacy end-points was observed by week 2 and maintained throughout the study. Long-term treatment with tacrolimus ointment is safe and effective in these patients with atopic dermatitis. more...

Education faces many challenges in the changing modern world. Learners are changing in their approaches to education – they use digital technologies, they multi-task, they collaborate and they are becoming less patient with teacher-centric styles of education. \ud \ud Educators face many changes – such as expectations of adopting innovative teaching approaches, alignment of teaching to external standards, growing requirements for professional development and difficulties in balancing a complex range of demands from different stakeholders. \ud \ud Government and educational institutions also face many changes, such as the rise of the knowledge economy and the need for different kinds of graduates, a shift from knowledge scarcity to abundance, and the impact of technology – especially the internet via open sharing of educational resources and massive open online courses (MOOCs).\ud \ud In the context of these changes, effective teaching and learning in the classroom (and beyond) remains central. How can educators become more effective in their preparation and facilitation of teaching and learning activities? How can educators be exposed to new teaching ideas that take them beyond their traditional approaches? How can technology assist educators without undermining them? How can learners be better prepared for the world that awaits them?\ud \ud This paper describes how the new field of Learning Design contributes to the central challenge of improving teaching and learning. Learning Design can assist educators to describe effective teaching ideas so that they can be shared with, and adapted by, other educators. While the field has primarily focussed on higher education and K-12 schools to date, it also has implications for vocational and professional training. This paper describes how ongoing work to develop a descriptive language for teaching and learning activities (often including the use of technology) is changing the way educators think about planning and facilitating educational activities. The ultimate goal of Learning Design is to convey great teaching ideas among educators in order to improve student learning.\ud \ud The paper begins with this Introduction, followed by an analogy from music to provide a context for Part 1, which considers the possibility of educational notation. Part 2 describes how this possibility is being realised in the field of Learning Design, illustrated with an example based on a Role Play. Part 3 considers current definitional challenges in Learning Design and its provocative aspiration towards pedagogical neutrality. Part 4 provides a wider conceptual map of education for exploring the place of Learning Design, including more examples of current Learning Design approaches, and how the map can be used to analyse pedagogical theories. Part 5 returns to the relationship between Learning Design and pedagogical theories, and the central question of effective teaching and learning approaches. The Conclusion offers a new synthesis of the ideas discussed in this paper as a foundation for the future of Learning Design, and the Epilogue returns to the music analogy to reflect on the future prospects of this synthesis.\ud \ud While the concepts discussed in this paper have potentially far-reaching implications for many aspects of education, this paper is written primarily for those with an interest in Learning Design and in pedagogical theories.
