Abstract

Mestrado em Microbiologia A resistência aos antimicrobianos é um problema crescente no contexto clínico. É consequência de uma terapia inadequada e resulta das dificuldades crescentes no tratamento de infecções bacterianas. Actualmente é a grande ameaça no público saudável e tem aumentado significativamente em infecções adquiridas na comunidade, como por exemplo infecções urinárias. E. coli é o principal microrganismo responsável por infecções urinárias em ambulatório e apresenta uma percentagem significativa de resistências ao Sulfametoxazol + Trimetoprim (SxT). A eficácia antimicrobiana e o baixo custo fizeram com que se tornassem antibióticos de eleição para o tratamento de infecções urinárias. Contudo, por esse motivo, têm-se verificado um aumento da taxa de resistência a estes compostos. O presente estudo foi efectuado, durante um período de 3 meses, em isolados de E. coli recolhidos de pacientes de ambulatório da região centro de Portugal, Aveiro, com suspeita de ITU. Tendo em conta o perfil de antibióticos testado verificou-se maior percentagem de resistência à amoxicilina, cefatrizina, norfloxacina e SxT. Este último grupo está fortemente relacionado com terapias empíricas, tendo por isso sido o seleccionado para o presente estudo. São conhecidos três genes, sul1, sul2 e sul3, implicados na resistência às Sulfonamidas. De um total de 547 isolados de E. coli foram seleccionados 25 isolados resistentes ao SxT. Destes, 19 foram positivos na pesquisa do gene de integrase de classe 1, 13 para a pesquisa de sul1, 16 para a pesquisa de sul2 e 2 para a pesquisa de sul3. Variantes do qgene dfr foram predominantes nas zonas variáveis amplificadas. Em conclusão, a percentagem de resistência ao SxT em estirpes de E.coli não é a mais elevada contudo a localização de genes de resistência em elementos genéticos móveis pode indicar uma forte disseminação da resistência e um aumento no fracasso dos tratamentos com este grupo de antibióticos. ABSTRACT: Antimicrobial resistance is a major problem in the clinical context. It results from inadequate therapy and results in difficulties to treat bacterial infrctions. Nowadays, it represents a significant threat for the general healthy public as infections are increasing significantly among the community, with urinary infections as an example of that. Escherichia coli is the main microorganism responsible for urinary infections in the community and shows a significant percentage of resistance to Sulfamethoxazole + Trimethoprim (SxT). Due to its antimicrobial efficacy and low cost, SxT became the leading antibiotic for treatment of urinary tract infections. However, the resistance rates among the isolates have increased over the last decades. During a three month study performed at the community level, results showed that Escherichia coli is the most common microorganism isolated from patients with UTI. Antibiotic resistance profile revealed a high rate of resistance to Amoxicilin, Cefatrizin, Norfloxacin and SxT. This last group is strongly related to the empirical therapy, therefore isolates resistant to SxT were selected for further studies. Three genes are known to be involved in Sulfonamides resistance: sul1, sul2 and sul3. From a total of 547 E. coli isolates collected from a health care unit from individuals from Aveiro, Central Portugal, with suspected ITU, 25 E.coli resistant to SxT were selected for this study. 19 of these were positive for the presence of the gene int1, indicating the presence of class 1 integron(s); 13 were positive for the presence of sul1, 16 were positive for sul2 and 2 for the sul3 gene. Variants of the dfr were predominant in the amplified variable regions. Results showed that the main resistance mechanism to Sulfonamides seems to be related to the presence of class 1 integrons. To conclude, the percentage of resistance to SxT in the E.coli strains is not very high; the resistance genes are located in mobile genetic elements providing the mobilization of these genes and, therefore, contributing to the failure of the treatments with this group of antibiotics.

216. The role of *language in early childhood mathematics

Abstract

Math literacy is imperative to succeed in society. Experience is key for acquiring math literacy. A preschooler's world is full of mathematical experiences. Children are continually counting, sorting and comparing as they play. As children are engaged in these activities they are using language as a tool to express their mathematical thinking. If teachers are aware of these teachable moments and help children bridge their daily experiences to mathematical concepts, math literacy may be enhanced. This study described the interactions between teachers and preschoolers, determining the extent to which teachers scaffold children's everyday language into expressions of mathematical concepts. Of primary concern were the teachers' responsive interactions to children's expressions of an implicit mathematical utterance made while engaged in block play. The parallel mixed methods research design consisted of two strands. Strand 1 of the study focused on preschoolers' use of everyday language and the teachers' responses after a child made a mathematical utterance. Twelve teachers and 60 students were observed and videotaped while engaged in block play. Each teacher worked with five children for 20 minutes, yielding 240 minutes of observation. Interaction analysis was used to deductively analyze the recorded observations and field notes. Using a priori codes for the five mathematical concepts, it was found children produced 2,831 mathematical utterances. Teachers ignored 60% of these utterances and responded to, but did not mediate 30% of them. Only 10% of the mathematical utterances were mediated to a mathematical concept. Strand 2 focused on the teacher's view of the role of language in early childhood mathematics. The 12 teachers who had been observed as part of the first strand of the study were interviewed. Abstract

The adhesion molecule CD44 and the membrane-type matrix metalloproteinase MT1-MMP act coordinately in tumor cells to promote cell invasion through a yet unclear mechanism. We are interested in studying the interplay between CD44 and MT1-MMP in carcinoma cells embedded in HA containing three-dimensional collagen I matrices (3D HA-Col I) by time-lapse confocal microscopy imaging. Here we report the in vivo interaction between CD44 and MT1-MMP, revealed by fluorescence resonance energy transfer (FRET) microscopy. MT1-MMP interacts with CD44 preferentially at the trailing edge of the invading tumor cells during rear retraction and on membrane fragments released during the invasion process. A fluorescent biosensor designed to monitor the proteolytic processing of CD44 by live cell imaging demonstrates that cleavage of the CD44 extracellular domain is enriched in the retracting rear ends of invasive tumor cells. Invasion assays showed that MT1-MMP mediates CD44-dependent tumor-cell invasion, whereas CD44 is not essential for MT1-MMP-mediated invasion of 3D HA-Col I matrices. Theconventional water network synthesis approach greatly simplifieswastewater treatment units by using fixed recoveries, creating a gapfor their applicability to industrial processes. This work describesa unifying approach combining various technologies capable of removingall the major types of contaminants through the use of more realisticmodels. The following improvements are made over the typical superstructure-basedwater network models. First, unit-specific shortcut models are developedin place of the fixed contaminant removal model to describe contaminantmass transfer in wastewater treatment units. Shortcut wastewater treatmentcost functions are also incorporated into the model. In addition,uncertainty in mass load of contaminants is considered to accountfor the range of operating conditions. Furthermore, the superstructureis modified to accommodate realistic potential structures. We presenta modified Lagrangean-based decomposition algorithm in order to solvethe resulting nonconvex mixed-integer nonlinear programming (MINLP)problem efficiently. Abstract

Introduction: Recent systematic reviews highlighted increasing use of cadaveric models in the surgical training, but reports on the characteristics of the models and their impact on training are lacking, as well as standardized recommendations on how to ensure the quality of surgical studies. The aim of our survey was to provide an easy guideline that would improve the quality of the studies involving cadavers for surgical training and research.Methods: After accurate literature review regarding surgical training on cadaveric models, a draft of the CACTUS guidelines involving 10 different items was drawn. Afterwards, the items were improved by questionnaire uploaded and spread to the experts in the field via Google form. The guideline was then reviewed following participants feedback, ergo, items that scored between 7 and 9 on nine-score Likert scale by 70% of respondents, and between 1 and 3 by fewer than 15% of respondents, were included in the proposed guideline, while items that scored between 1 and 3 by 70% of respondents, and between 7 and 9 by 15% or more of respondents were not. The process proceeded with Delphi rounds until the agreement for all items was unanimous.Results: In total, 42 participants agreed to participate and 30 (71.4%) of them completed the Delphi survey. Unanimous agreement was almost always immediate concerning approval and ethical use of cadaver and providing brief outcome statement in terms of satisfaction in the use of the cadaver model through a short questionnaire. Other items were subjected to the minor adjustments.Conclusion: 'CACTUS' is a consensus-based guideline in the area of surgical training, simulation and anatomical studies and we believe that it will provide a useful guide to those writing manuscripts involving human cadavers. Abstract

The regulator of angiogenesis most extensively studied is VEGF. VEGF mRNA in plasma from patients with colorectal cancer was analyzed as a possible surrogate marker of tumor angiogenesis. VEGF mRNA was measured by quantitative PCR in plasma, tumors and circulating tumor cells from colorectal cancer patients. Circulating VEGF protein was analyzed by ELISA. Microvessel density was determined. Levels of VEGF mRNA and protein in plasma were higher in patients than in controls. VEGF mRNA was overexpressed in tumors with respect to normal tissues. Levels of VEGF protein were associated with VEGF mRNA in plasma, but no associations with tumor samples were found. A trend to statistical significance was shown between high VEGF mRNA and vascular invasion. MVD was not related to VEGF mRNA in plasma. Thus, VEGF mRNA could be a marker similar to VEGF protein in plasma. [ABSTRACT FROM AUTHOR]
