Plant microRNAs (miRNAs) associate with ARGONAUTE1 (AGO1) to direct post-transcriptional gene silencing and regulate numerous biological processes. Although AGO1 predominantly binds miRNAs in vivo, it also associates with endogenous small interfering RNAs (siRNAs). It is unclear whether the miRNA/siRNA balance affects miRNA activities. Here we report that FIERY1 (FRY1), which is involved in 5'-3' RNA degradation, regulates miRNA abundance and function by suppressing the biogenesis of ribosomal RNA-derived siRNAs (risiRNAs). In mutants of FRY1 and the nuclear 5'-3' exonuclease genes XRN2 and XRN3, we find that a large number of 21-nt risiRNAs are generated through an endogenous siRNA biogenesis pathway. The production of risiRNAs correlates with pre-rRNA processing defects in these mutants. We also show that these risiRNAs are loaded into AGO1, causing reduced loading of miRNAs. This study reveals a previously unknown link between rRNA processing and miRNA accumulation.

The TRNT1 gene encodes tRNA nucleotidyltransferase 1, which catalyzes the addition of cytosine-cytosine-adenosine (CCA) to the ends of cytoplasmic and mitochondrial tRNAs. The most common clinical phenotype associated with TRNT1 is autosomal recessive sideroblastic anemia with B-cell immunodeficiency, periodic fever, and developmental delay (SIFD). Muscle involvement has rarely been reported in TRNT1-related disorders. Here we report a Chinese patient with incomplete SIFD and hyperCKemia, and explored the skeletal muscle pathological changes. The patient was a 3-year-old boy with sensorineural hearing loss, sideroblastic anemia, and developmental delay since infancy. At the age of 11 months, significantly increased levels of creatine kinase were noted, accompanied by mild muscle weakness. Whole-exome sequencing revealed compound heterozygous variants of the TRNT1 gene, c.443C > T (p.Ala148Val) and c.692C > G (p.Ala231Gly), in the patient. Western blot showed a decreased expression of TRNT1 and cytochrome c oxidase subunit IV (COX IV) in the skeletal muscle of the patient. Electron microscopy observation of skeletal muscle pathology revealed abnormal mitochondria of various sizes and shapes, supporting a diagnosis of mitochondrial myopathy. The present case indicates that in addition to the classic SIFD phenotype, TRNT1 mutations can cause mitochondrial myopathy, a rare clinical phenotype of TRNT1-related disorders.

Adopting a walkthrough of the game application, participant observation of online fan communities, and a narrative analysis of storylines, this study seeks to understand how local female-orientated love simulation games re-calibrate Western feminist ideologies with/through constructions of romantic love. This study argues that with the development and backlash of feminist ideologies and neo-liberal economic restructuring in post-socialist China, female-oriented love simulation games now present what I coin “Platformed Post-Feminist Negotiation” (hereafter PPN). First, the PRP transplants Western post-feminism to veil the latent and toxic consumerism with the rhetoric of women’s empowerment. Second, the PPN inherits Chinese feminism’s dynamic negotiation with Confucian ideals and the conservative state ideology. While compromising with dominant ideologies to escape from China’s stringent digital censorship, game developers have aligned with players to carry out micro, covert, and indirect resistance to protecting female sexual freedom. Third, the PPN refers to how digital games are platformed as technical tools to manufacture a variant post-feminism that can be defined as empowering women, ignoring structural gender inequalities, and negotiating with state censorship. This study contributes to Chinese digital games’ post-feminist narrative patterns and how contemporary post-feminism appropriates technological affordances in an increasingly platformed society. The quantum uncertainty relation characterizes the restriction of the quantum coherence on different measurement bases. The skew information of coherence is a coherence measure both operationally and informatically. We study the quantum uncertainty relation for the skew information of coherence under any two orthonormal bases. In qubit and qutrit systems, we derive the quantum uncertainty relations with the lower bounds in the factorized forms by the purity of quantum states and the incompatibility between two orthonormal bases. Especially the lower bound in qubit systems is strictly positive for non-maximally mixed states and incompatible orthonormal bases. These quantum uncertainty relations reveal the intrinsic relation among the coherence, the purity and the incompatibility between two orthonormal bases quantitatively. The advantages of these quantum uncertainty relations are illustrated by some specific examples. This method we developed here is beneficial to some other quantum uncertainty relations.
