Abstract

In this study, social force model (SFM) is extended by using a discretisation grid to permit pedestrians to change their desired speed directions dynamically. In reality, other pedestrians may obscure the visions of the behind pedestrians, so the behind pedestrians will be blocked if they insist to walk in the final desired directions calculated by the SFM. So, a dynamic destination choice model is established to provide pedestrians a series of available intermediate destinations. Based on the dynamic destination choice model, the authors use a discretisation grid to represent all the potential moving directions of pedestrians, and model the weight of every potential moving direction. The direction with the maximum weight is selected as the optimal route at that time step. Besides, pedestrians prefer to pass near to crowds with a low relative velocity and choose the low space occupancy route when several routes have the same pedestrian density. So, pedestrian speeds, space un-occupancy, route length, crowd density and object pedestrian ratio are used to develop the weight model. The modified SFM guarantees pedestrians to obtain an available and optimal route. Compared to other models, the proposed models can be used to reproduce the behavior of bidirectional pedestrians more really.

215. Structural insights into the potency and selectivity of covalent pan-FGFR inhibitors.

Abstract

FIIN-2, TAS-120 (Futibatinib) and PRN1371 are highly potent pan-FGFR covalent inhibitors targeting the p-loop cysteine of FGFR proteins, of which TAS-120 and PRN1371 are currently in clinical trials. It is critical to analyze their target selectivity and their abilities to overcome gatekeeper mutations. In this study, we demonstrate that FIIN-2 and TAS-120 form covalent adducts with SRC, while PRN1371 does not. FIIN-2 and TAS-120 inhibit SRC and YES activities, while PRN1371 does not. Moreover, FIIN-2, TAS-120 and PRN1371 exhibit different potencies against different FGFR gatekeeper mutants. In addition, the co-crystal structures of SRC/FIIN-2, SRC/TAS-120 and FGFR4/PRN1371 complexes reveal structural basis for kinase targeting and gatekeeper mutations. Taken together, our study not only provides insight into the potency and selectivity of covalent pan-FGFR inhibitors, but also sheds light on the development of next-generation FGFR covalent inhibitors with high potency, high selectivity, and stronger ability to overcome gatekeeper mutations. The human fibroblast growth factor receptors (FGFRs) are attractive targets for cancer therapy, but the structural basis for kinase targeting and gatekeeper mutations of covalent FGFR inhibitors are not fully understood. Abstract

Thallium (Tl) is an uncommon toxic element, with an even greater toxicity than that of As, Hg and Cd. Steel-making industry has been identified as an emerging new significant source of Tl contamination in China. This paper presents a pilot investigation of the contamination and geochemical transfer of Tl and associated metal(loid)s in river sediments affected by long-term waste discharge from the steel-making industry. The results uncovered an overall Tl contamination (1.96 ± 0.42 mg/kg) across a sediment profile of approximately 1.5 m in length, even 10 km downstream the steel plant. Highly elevated contents of Pb, Cu, Cd, Zn and Sb were found in the fluvial sediments, displaying strong positive correlations with Tl contents. Elevated levels of geochemically mobile Tl as well as Cd, Zn, Cu and Pb occurred in the fluvial sediments, signifying anthropogenic imprints from steel production activities at high temperature. Levels of contamination and ecological risk were calculated to be moderate to considerable for Tl, Cu, Zn and high to very high for Cd, Pb, Sb. The results highlight that there is a great challenge in view of potentially considerable Tl pollution due to continuous massive steel production in many other parts of China. It is high time to initiate process-based management of Tl contamination control for the ambient aquifer system in the steel-making area.

Abstract

Thallium (Tl) is a well-recognized hazardous heavy metal with very high toxicity. It is usually concentrated in sulfide minerals, such as pyrite (FeS2), sphalerite (ZnS), chalcopyrite (CuS) and galena (PbS). Here, this study was carried out to investigate the indigenous microbial communities via 16S rRNA gene sequence analysis in typical surface sediments with various levels of Tl pollution (1.8–16.1 mg/kg) due to acid mine drainage from an active Tl-containing pyrite mining site in South China. It was found with more than 50 phyla from the domain Bacteria and 1 phyla from the domain Archaea. Sequences assigned to the genera Ferroplasma, Leptospirillum, Ferrovum, Metallibacterium, Acidithiobacillus, and Sulfuriferula manifested high relative abundances in all sequencing libraries from the relatively high Tl contamination. Canonical correspondence analysis further uncovered that the overall microbial community in this area was dominantly structured by the geochemical fractionation of Tl and geochemical parameters such as pH and Eh. Spearman's rank correlation analysis indicated a strong positive correlation between acidophilic Fe-metabolizing species and Tltotal, Tloxi, and Tlres. Abstract

Thallium (Tl) is a trace metal of severe toxicity. Its health concerns via consumption of contaminated vegetables have often been overlooked or underestimated. This study was designed to gain insight into the actual level and distribution characteristics of Tl and metal (loid)s (Pb, Cd, Cr, Sb, Mn, Cu, Zn, Ni, and Co) in agricultural soils and common vegetables cultivated in different zones (upstream, midstream, and downstream) of a densely populated residential area in a typical mine city, which has been open-pit exploiting Tl-bearing pyrite minerals since 1960s. The results show that most of the agricultural soils exhibit contaminated levels of Tl, with Tl contents (upstream: 1.35–4.31 mg/kg, midstream: 2.43–5.19 mg/kg, and downstream: 0.65–2.33 mg/kg) mostly exceeding the maximum permissible level (MPL) for agricultural land use (1 mg/kg). Sequential extraction procedure indicates that even Tl is predominantly retained in the residual fraction, significant levels of Tl are still present in the geochemically mobile fractions. Besides, metals like Cu, Cd, Mn, and Co are mostly distributed in the labile fractions. Almost all metal (loid)s in edible parts of the vegetables exceed their corresponding MPL for consumption. The chronic daily intake (CDI) and hazard quotient (HQ) values calculated for inhabitants at different ages indicate non-negligible Tl risks via consumption of local vegetables, especially for children. Therefore, it is critical to establish effective measures for hazardous waste management and enforceable regulations in Tl-polluted area to mitigate potential severe impacts of Tl on human health through food chain. © 2019 Elsevier Ltd; Alarming levels of toxic thallium were widely detected in vegetables and soils in a typical pyrite mining city. © 2019 Elsevier Ltd

Abstract

Chronic myeloid leukemia (CML) is a hematologic malignancy originating from BCR-ABL oncogene-transformed hematopoietic stem cells (HSCs) known as leukemia stem cells (LSCs). Therefore, targeting LSCs is of primary importance to eradicate CML. The present study demonstrates that picropodophyllin (PPP) effectively induces apoptosis and inhibits colony formation in CML stem/progenitor cells as well as quiescent CML progenitors resistant to imatinib therapy, while sparing normal hematopoietic cells in vitro. Administration of PPP in vivo markedly diminishes CML stem/progenitor cells in a transgenic mouse model of CML by inhibition of cell proliferation and enhancement of apoptosis in LSK cells, and significantly improves survival of CML mice. Furthermore, PPP treatment preferentially leads to transcriptional activation of p53 in CML but not normal CD34+ cells, upregulation of p53 protein in LSCs-enriched Sca-1+ cells from CML mice, and increased phosphorylation of p53 and upregulation of Bax protein in Ku812 cells. These results suggest that the inhibitory effects of PPP on CML stem/progenitor cells are associated with selective activation of p53 pathway and propose that PPP is a potent agent that selectively targets CML LSCs, and may be of value in the CML therapy. • PPP induced apoptosis and suppressed CFC in CML stem/progenitor cells in vitro. • PPP diminished CML stem/progenitor cells and improved survival in a CML mouse model. • PPP preferentially activated p53 pathway in CML but not normal CD34+ cells. Abstract

Probabilistic topic models have been used to detect topic-based content presentations when facing a collection of documents. However, topic models capture the semantic information according to reasonable simplifying hypotheses, which ignore the worthwhile word-order information. This paper proposes an attention hierarchical topic modeling, which adopts attention mechanism to unify topic embedding and word embedding together into a framework to enhance the clustering effect of hierarchical Dirichlet process. Otherwise, the multi-information integration Chinese restaurant franchise is adopted to construct this model, which further combines timestamp, user, and topic label to optimize topic modeling. Abstract

Data stream has been widely used in lots of modern applications such as Social networks and the Internet of things. Aiming at the problem of Top-k dominating query in distributed data stream, a distributed Top-k query algorithm based on Spark Streaming framework is proposed. Based on partitioning, double pruning techniques are implemented on the data. Local and global pruning can significantly reduce the number of candidate sets, reduce the computational overhead and space costs, and improve the query efficiency. Experimental results show that the algorithm has good performance and scalability.
