Bromodomain-containing proteins are known readers of histone acetylation that regulate chromatin structure and transcription. Although the functions of bromodomain-containing proteins in development, homeostasis, and disease states have been well studied, their role in self-renewal of hematopoietic stem and progenitor cells (HSPCs) remains poorly understood. Here, we performed a chemical screen using nine bromodomain inhibitors and found that the bromodomain and PHD finger-containing protein 1 (Brpf1) inhibitor OF-1 enhanced the expansion of Lin−Sca-1+c-Kit+ HSPCs ex vivo without skewing their lineage differentiation potential. Importantly, our results also revealed distinct functions of Brpf1 isoforms in HSPCs. Brpf1b promoted the expansion of HSPCs. By contrast, Brpf1a is the most abundant isoform in adult HSPCs but enhanced HSPC quiescence and decreased the HSPC expansion. Furthermore, inhibition of Brpf1a by OF-1 promoted histone acetylation and chromatin accessibility leading to increased expression of self-renewal-related genes (e.g. Mn1). The phenotypes produced by OF-1 treatment can be rescued by suppression of Mn1 in HSPCs. A manganese dioxide (MnO2) film is electrodeposited onto a gold-coated quartz crystal electrode by a galvanostatic method. The scanning electron microscopy (SEM) image shows that the electrodeposited MnO2 film consists of nanorods with diameters of 15–20 nm. The cyclic voltammetric (CV) and electrochemical quartz crystal microbalance (EQCM) results demonstrate that the MnO2 electrodes exhibit similar specific capacitance (SC) and mass-to-charge ratios (MCR) in those electrolytes that contain the same cation but different anions. However, the SC and MCR are different in those electrolytes that contain the same anion but different cations. The SC of MnO2 electrodes increases with a decrease of the cationic radius and an increase of the cationic charge. The smaller the cationic radius, the smaller the deviation of the MCR from the equivalent weight (EW) of the cation. The results illustrate that the pseudocapacitive charge storage mechanism of MnO2 involves the cation in the electrolyte and its intercalation/deintercalation, which are affected by the charge number and the cationic radius. Introduction: Present study was to evaluate whether increased arterial stiffness was associated with target organ damage in pre-hypertensive subjects.Material and Methods: Pre-hypertensive subjects enrolled and echocardiography was used to evaluate left ventricular hypertrophy (LVH) and the first morning urine was collected to evaluate albumin and creatinine ratio (ALB/Cr ratio). Carotid-femoral pulse wave velocity (cf-PWV) was measured.Results: A total of 420 subjects were recruited and mean age was 42.6 years. Mean systolic/diastolic blood pressure (SBP/DBP) were 130 ±9 mm Hg and 85 ±4 mm Hg. The prevalence of albuminuria and left ventricular hypertrophy was 8.6 % and 11.7 %. Mean cf-PWV was 9.2 ±1.0 m/s, with arterial stiffness prevalence was 8.8%. Subjects with arterial stiffness had higher cf-PWV value (10.6 ±0.4 m/s vs. 8.7 ±0.6 m/s, p < 0.05), and ALB/Cr ratio (28.3 ±13.2 µg/mg vs. 23.1 ± 11.4 µg/mg, p < 0.05). Overall, with multivariate regression analysis, aging and arterial stiffness were significantly associated with pre-hypertension. With stepwise adjusted for potential covariates including age, male gender, fasting plasma glucose, presence of current cigarette smoking, dyslipidemia, statins and SBP, increased cf-PWV remained independently associated with left ventricular hypertrophy and albuminuria, with an increased odds of 41 % and 24 % (p < 0.05), respectively.Conclusions: In pre-hypertensive subjects, arterial stiffness is independently associated with LVH and albuminuria and cf-PWV may be a useful marker to identify target organ damage in pre-hypertensive subjects. Wine frying status is one of the most important processes in the brewing of Chinese yellow rice wine (CYW) and has a great influence on the quality and flavor of CYW. The present work determined the ethyl carbamate (EC) concentration by gas chromatography-mass spectrometry (GC-MS) and aroma compounds by headspace solid-phase microextraction GC-MS (HS-SPME-GC-MS) during wine frying status in CYW. In addition, effects of wine frying conditions on the changes of EC and aroma compounds were also investigated. Results indicated that EC concentration increased along with the increase of wine frying temperature and time. Pearson test indicated that EC had highly significant correlation ( p < 0.01) with temperature and time. Esters, alcohols, and ethers consisted of the most aroma components and constituted the main flavor of CYW. Considering EC concentration, main flavor, unique ingredients, and proportions of volatile aroma compounds, 70 °C and 11-20 min are the most appropriate temperature and time for wine frying of CYW. ABSTRACT Keratin particles with microscale are prepared by ball mill and its influences on the chitosan membrane is evaluated. Composite membranes with various content of keratin are fabricated, and their physical and chemical properties such as morphology, wettability, crystallization, thermal stability, tensile strength, and break elongation are investigated. Optical microscope and situ topographic scan mode of nano-test system are used to examine the dispersion and aggregation of keratin on the surface of chitosan membrane. The result of contact angle (CA) and mechanical testing show that the incorporation of keratin particles decrease the CA from 98.1° to 58.2°. Tensile strength and break elongation of the composite membrane reaches a maximum of 65 ± 8 MPa and 15% when the keratin content is 6%, an increase of 80% and 88% compared with the pristine chitosan membrane. Both the increase in tensile strength and break elongation is result of the incorporation of keratin particles known for their excellent compatibility between keratin and chitosan matrix. These kind of composite material combines the antibacterial properties of chitosan with cell culture preference of keratin which may have potential biomedical application. © 2016 Wiley Periodicals, Inc. The three-dimensional distribution of Bi atoms in a GaAs 1− x Bi x /GaAs superlattice grown by metalorganic vapor phase epitaxy (MOVPE) was studied using atom probe tomography (APT). The Bi distribution in the growth direction deduced from APT agreed quantitatively with the complex Bi concentration profile that was discovered using high-angle annular dark-field scanning transmission electron microscopy in a previous study. More importantly, APT revealed the Bi atom distribution in the growth planes at near atomic resolution. Bi nearest neighbor distribution and concentration frequency distribution analysis of the APT data indicated a statistically random distribution of Bi atoms in 1–2 nm thick layers oriented perpendicular to the growth direction. These results provide evidence that Bi is incorporated homogeneously across the growth front even when the concentration profile in the growth direction is complex. They also suggest that MOVPE growth conditions can promote uniform Bi distribution within GaAs 1− x Bi x layers, opening a path for application of these materials in the optoelectronic devices for which they show much promise. The constant emergence of variants of concern (VOCs) challenges the effectiveness of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) vaccines over time. This is most concerning in clinically vulnerable groups, such as older adults. This study aimed to determine whether the novel adjuvant MF59-like adjuvant can improve cross-immunity against VOCs in aged animals. We compared the humoral and cellular immune responses of Alum and MF59-like adjuvant-formulated inactivated coronavirus disease 2019 (COVID-19) vaccines against prototype and SARS-CoV-2 variants in 18-month-old mice. Our results showed that two doses of the MF59-like adjuvant inactivated vaccines induced more robust binding and pseudo-neutralizing antibodies (Nabs) against the SARS-CoV-2 prototype and VOCs compared to the Alum-adjuvant and reduced Omicron variant escapes from Nabs in aged mice. The humoral immune responses of inactivated vaccines were much lower against VOCs than the prototype with or without adjuvants; however, T cell responses against VOCs were not affected. In addition, Alum and MF59-like adjuvanted vaccines induced Th1-biased immune responses with increased interferon-gamma and interleukin (IL)-2 secreting cells, and hardly detectable IL-4 and IL-5. Furthermore, the MF59-like adjuvant vaccine produced 1.9–2.0 times higher cross-reactive T cell responses against the SARS-CoV-2 prototype and VOCs than the Alum adjuvant.
