Abstract

The endoplasmic reticulum (ER)-to-Golgi transport is critical to protein secretion and intracellular sorting. Here, we report a highly elongated tubular ER-Golgi intermediate compartment (t-ERGIC) that selectively expedites the ER-to-Golgi transport for soluble cargoes of the receptor SURF4. Lacking the canonical ERGIC marker ERGIC-53 yet positive for the small GTPases Rab1A/B, the t-ERGIC is further marked by its extraordinarily elongated and thinned shape. With its large surface-to-volume ratio, high intracellular traveling speeds, and ER-Golgi recycling capabilities, the t-ERGIC accelerates the trafficking of SURF4-bound cargoes. The biogenesis and cargo selectivity of t-ERGIC both depend on SURF4, which recognizes the N terminus of soluble cargoes and co-clusters with the selected cargoes to expand the ER-exit site. In the steady state, the t-ERGIC-mediated fast ER-to-Golgi transport is antagonized by the KDEL-mediated ER retrieval. Together, our results argue that specific cargo-receptor interactions give rise to distinct transport carriers that regulate the trafficking kinetics.

Abstract

Privacy-preserving data mining has become an important topic. People have built several multi-party-computation (MPC)-based frameworks to provide theoretically guaranteed privacy, the poor performance of real-world algorithms have always been a challenge. Using Principal Component Analysis (PCA) as an example, we show that by considering the unique performance characters of the MPC platform, we can design highly effective algorithm-level optimizations, such as replacing expensive operators and batching up. We achieve about 200$\times$ performance boost over existing privacy-preserving PCA algorithms with the same level of privacy guarantee. Also, using real-world datasets, we show that by combining multi-party data, we can achieve better training results., Comment: 11 pages, 3 figures, 5 tables

Abstract

To develop highly sensitive, stable and repeatable surface-enhanced Raman scattering (SERS) substrates is crucial for analytical detection, which is a challenge for traditional metallic structures. Herein, by taking advantage of the high surface activity of 1T' transition metal telluride, we have fabricated high-density gold nanoparticles (AuNPs) that are spontaneously in-situ prepared on the 1T' MoTe2 atomic layers via a facile method, forming a plasmonic-2D material hybrid SERS substrate. This AuNP formation is unique to the 1T' phase, which is repressed in 2H MoTe2 with less surface activity. The hybrid structure generates coupling effects of electromagnetic and chemical enhancements, as well as excellent molecule adsorption, leading to the ultrasensitive (4*10^-17 M) and reproducible detection. Additionally, the immense fluorescence and photobleaching phenomena are mostly avoided. Flexible SERS tapes have been demonstrated in practical applications. Our approach facilitates the ultrasensitive SERS detection by a facile method, as well as the better mechanistic understanding of SERS beyond plasmonic effects.

218. Insights into the Electron-Electron Interaction from Quantum Monte Carlo Calculations

Abstract

The effective electron-electron interaction in the electron gas depends on both the density and spin local field factors. Variational Diagrammatic Quantum Monte Carlo calculations of the spin local field factor are reported and used to quantitatively present the full spin-dependent, electron-electron interaction. Together with the charge local field factor from previous Diffusion Quantum Monte Carlo calculations, we obtain the complete form of the effective electron-electron interaction in the uniform three-dimensional electron gas. Very simple quadratic formulas are presented for the local field factors that quantitatively produce all of the response functions of the electron gas at metallic densities. Exchange and correlation become increasingly important at low densities. At the compressibility divergence at rs = 5.25, both the direct (screened Coulomb) term and the charge-dependent exchange term in the electron-electron interaction at q=0 are separately divergent. However, due to large cancellations, their difference is finite, well behaved, and much smaller than either term separately. As a result, the spin contribution to the electron-electron interaction becomes an important factor. The static electron-electron interaction is repulsive as a function of density but is less repulsive for electrons with parallel spins. The effect of allowing a deformable, rather than rigid, positive background is shown to be as quantitatively important as exchange and correlation. As a simple concrete example, the electron-electron interaction is calculated using the measured bulk modulus of the alkali metals with a linear phonon dispersion. The net electron-electron interaction in lithium is attractive for wave vectors $0-2k_F$, which suggests superconductivity, and is mostly repulsive for the other alkali metals., Comment: 16 pages, 20 figures
