202. EdgeShield: Enabling Collaborative DDoS Mitigation at the Edge

Abstract

Edge computing (EC) enables low-latency services by pushing computing resources to the network edge. Due to the geographic distribution and limited capacities of edge servers, EC systems face the challenge of edge distributed denial-of-service (DDoS) attacks. Existing systems designed to fight cloud DDoS attacks cannot mitigate edge DDoS attacks effectively due to new attack characteristics. In addition, those systems are typically activated upon detected attacks, which is not always realistic in EC systems. DDoS mitigation needs to be cohesively integrated with workload migration at the edge to ensure timely responses to edge DDoS attacks. In this paper, we present EdgeShield, a novel DDoS mitigation system that leverages edge servers’ computing resources collectively to defend against edge DDoS attacks without the need for attack detection. Aiming to maximize system throughput over time without causing significant service delays, EdgeShield monitors service delays and migrates workloads across an EC system with adaptive mitigation strategies. The experimental results show that EdgeShield significantly outperforms state-of-the-art solutions in both system throughput and service delays.

Abstract

Obesity has become an ongoing global crisis, since it increases the risks of cardiovascular disease, type 2 diabetes, fatty liver, cognitive decline, and some cancers. Adipose tissue is closely associated with the disorder of lipid metabolism. Several efforts have been made toward the modulation of lipid accumulation, but have been hindered by poor efficiency of cellular uptake, low safety, and uncertain effective dosage. Herein, we design an Fe3O4 microsphere-doped composite hydrogel (Fe3O4 microspheres @chitosan/ β -glycerophosphate/collagen), termed as Fe3O4@Gel, as the magnetocaloric agent for magnetic hyperthermia therapy (MHT), aiming to promote lipolysis in white adipocytes. The experimental results show that the obtained Fe3O4@Gel displays a series of advantages, such as fast sol–gel transition, high biocompatibility, and excellent magneto-thermal performance. MHT, which is realized by Fe3O4@Gel subjected to an alternating magnetic field, leads to reduced lipid accumulation, lower triglyceride content, and increased mitochondrial activity in white adipocytes. This work shows that Fe3O4@Gel-mediated MHT can effectively promote lipolysis in white adipocytes in vitro, which provides a potential approach to treat obesity and associated metabolic disorders. We propose three test criteria each of which is appropriate for testing, respectively, the equivalence hypotheses of symmetry, homogeneity, and independence, with multivariate data. All quantities have the common feature of involving weighted-type distances between characteristic functions and are convenient from the computational point of view if the weight function is properly chosen. The asymptotic behaviour of the tests under the null and alternative hypotheses is investigated. 221. Divergent Effects of Warmth and Competence Social Rejection: An Explanation Based on the Need-Threat Model.

Abstract

Based on the need-threat model, we hypothesized that "warmth rejection" threatens belongingness more than "competence rejection," whereas competence rejection threatens sense of efficacy more than warmth rejection. To restore threatened belongingness, warmth (vs. competence) rejection was predicted to result in higher affiliative responses. In contrast, to restore the threatened sense of efficacy, competence (vs. warmth) rejection would lead to higher self-focus. Across six studies, we found that the participants exhibited more affiliative responses after being rejected due to low warmth than due to low competence (Studies 1–6), whereas they became more self-focused after being rejected due to low competence than due to low warmth (Studies 3–6). Furthermore, the effect of warmth rejection on affiliation was mediated by perceived threat to belongingness (Studies 4–6), whereas the effect of competence rejection on self-focus was mediated by perceived threat to control and belongingness (Studies 4–6). Abstract

The use of β-lactam (BL) and β-lactamase inhibitor combination to overcome BL antibiotic resistance has been validated through clinically approved drug products. However, unmet medical needs still exist for the treatment of infections caused by Gram-negative (GN) bacteria expressing metallo-β-lactamases. Previously, we reported our effort to discover pan inhibitors of three main families in this class: IMP, VIM, and NDM. Herein, we describe our work to improve the GN coverage spectrum in combination with imipenem and relebactam. This was achieved through structure- and property-based optimization to tackle the GN cell penetration and efflux challenges. A significant discovery was made that inhibition of both VIM alleles, VIM-1 and VIM-2, is essential for broad GN coverage, especially against VIM-producing P. aeruginosa. In addition, pharmacokinetics and nonclinical safety profiles were investigated for select compounds. Key findings from this drug discovery campaign laid the foundation for further lead optimization toward identification of preclinical candidates.
