Insights into the diets of early societies can be gained, indirectly, from the cultural evidence of artefacts related to food procurement, preparation and consumption and human skeletal remains. However, more direct evidence for dietary constituents derives from the identification of intact plant and animal remains collected during the excavations but also from the exam of the amorphous remains of foodstuff associated with artefacts. Organic residues adhering to the surface or absorbed into the porous fabric of an unglazed cooking vessel should provide important information both about the usage of the vessel and dietary practices. This contribute deals with the results of a combined analytical research via 1 H- 1 H NMR 2D-TOCSY, ATR FT-IR and SEM-EDX on organic residues on pottery from two Sicilian prehistoric sites of Monte Kronio and Sant'Ippolito. The nuclear magnetic resonance (NMR)-based metabolomic approach was used as analytical methodology to study the urine samples of chronic inflammatory rheumatic disease (CIRD) patients. The urine samples of CIRD patients were compared to the ones of both healthy subjects and patients with multiple sclerosis (MS), another immuno-mediated disease. Urine samples collected from 39 CIRD patients, 25 healthy subjects, and 26 MS patients were analyzed using H NMR spectroscopy, and the NMR spectra were examined using partial least squares-discriminant analysis (PLS-DA). PLS-DA models were validated by a double cross-validation procedure and randomization tests. Clear discriminations between CIRD patients and healthy controls (average diagnostic accuracy 83.5 ± 1.9%) as well as between CIRD patients and MS patients (diagnostic accuracy 81.1 ± 1.9%) were obtained. Leucine, alanine, 3-hydroxyisobutyric acid, hippuric acid, citric acid, 3-hydroxyisovaleric acid, and creatinine contributed to the discrimination; all of them being in a lower concentration in CIRD patients as compared to controls or to MS patients. The application of NMR metabolomics to study these still poorly understood diseases can be useful to better clarify the pathologic mechanisms; moreover, as a holistic approach, it allowed the detection of, by means of anomalous metabolic traits, the presence of other pathologies or pharmaceutical treatments not directly connected to CIRDs, giving comprehensive information on the general health state of individuals. Arundo donax exhibits rapid growth and requires little nutrient input, making it an ideal perennial biomass crop species. However, this growth is accompanied by high rates of water use, potentially restricting the use of A. donax in rain-fed marginal lands. Here, we investigated the physiological and morphological responses to drought in two ecotypes of A. donax from contrasting habitats: one from an arid environment in Morocco, and the second from a warm humid sub-Mediterranean climate in central Italy. Prolonged drought resulted in identical reductions in leaf-level photosynthesis ( PN) and stomatal conductance ( Gs) in the two ecotypes. However, water deficit induced an increase in xylem vessel diameter in the Moroccan plants, improving the movement of water along the stem, but also likely reducing the resistance to embolism. In contrast, the Italian ecotype reduced xylem vessel area, thus increasing resistance to water transport and xylem embolism. The increased xylem vessel size and associated vulnerability to embolism in the Moroccan plants may have contributed to an increase in the loss of leaf numbers, but also to higher relative water content ( RWC) in the remaining leaves in comparison to the Italian ecotype, where a greater number of leaves persisted. Despite the Moroccan plants possessing stems with a lower basal area than their Italian counterparts, both ecotypes exhibited identical leaf to supporting stem area ratios under both control and water deficit conditions. This may account for the similarities observed in leaf area measures of PN and Gs in this and previous studies of different A. donax ecotypes. Selection of A. donax ecotypes on the basis of xylem responses to drought may facilitate the development of varieties suited to arid environments prone to severe drought and wetter habitats where prolonged droughts occur less frequently. The effectiveness and the mechanism of copper stain removal from stones by agar gels was systematically studied using marble laboratory specimens, stained and cleaned in well-controlled and reproducible conditions. The same cleaning procedure was also applied on the marble base of Napoleon's statue by A. Canova. The water release from agar gels to stones was investigated by capillarity absorption and unilateral Nuclear Magnetic Resonance. The cleaning by different agar gel formulations (pure and added with chemicals) was studied both on the stone substrate (optical microscopy and colour measurements) and in the gels (inductively coupled plasma-atomic emission spectrometry, electron paramagnetic resonance spectroscopy). Among the considered cleaning systems, the most effective ones for copper removal were agar gels 3% containing additives, with no significant difference among the used additives. However, agar gels with additives host copper in different ways: in gels added with ethylenediamine tetraacetic acid (EDTA), all copper centers are coordinated, while copper centers are also dispersed in water within gels added with ammonium citrate tribasic (TAC). Isotope Ratio Mass Spectrometry (IRMS), 1 H Nuclear Magnetic Resonance ( 1 H NMR), conventional chemical analysis and chemometric elaboration were used to assess quality and to define and confirm the geographical origin of 177 Italian PDO (Protected Denomination of Origin) olive oils and 86 samples imported from Tunisia. Italian olive oils were richer in squalene and unsaturated fatty acids, whereas Tunisian olive oils showed higher δ 18 O, δ 2 H, linoleic acid, saturated fatty acids β-sitosterol, sn -1 and 3 diglyceride values. Furthermore, all the Tunisian samples imported were of poor quality, with a K 232 and/or acidity values above the limits established for extra virgin olive oils. By combining isotopic composition with 1 H NMR data using a multivariate statistical approach, a statistical model able to discriminate olive oil from Italy and those imported from Tunisia was obtained, with an optimal differentiation ability arriving at around 98%. A multi-analytical study was carried out on materials, techniques, and state of conservation of a set of detached Nubian mural paintings, originally belonging to the 10th century pictorial cycle of the church of Sonqi Tino, Sudan (70 km south of the Egypt–Sudan border). These paintings represent one of the very few examples of Christian Nubian pictorial cycles preserved from the wide defacement experienced by these artifacts with the building up of the Aswan Dam. Non-invasive NMR depth profiling was used to preliminary study the hydrogen rich layers of the detached mural paintings. By this technique, it was possible to perform a virtual coring with a reconstruction of the complex series of layers that characterize the investigated artifacts. Then, micro-sampling and cross-section examination by SEM-EDX, micro-Raman, micro-FTIR, and 27 Al and 29 Si MAS NMR, allowed us to shed light on materials and technology exploited by the Medieval Nubian painters, with the identification of pigments and composition of the original primer, obtaining also information on layering of materials used by conservators during the detachment of the paintings and their transfer to a new support. The combination of preliminary non-invasive investigation with a set of micro-invasive analytical techniques allowed us to set up a protocol for a satisfactory decoding of the multilayered complex systems that characterize the detached paintings. A novel sub-2 μm chiral stationary phase (CSP) was prepared immobilizing vancomycin onto 1.8 μm diol hydride-based silica particles. The CSP was packed into fused silica capillaries of 75 μm i.d. with a length of 11 cm and evaluated by means of nano-liquid chromatography (nano-LC) using model compounds of both pharmaceutical and environmental interest (some non-steroidal anti-inflammatory drugs, β-blockers and herbicides). The study of the effect of the linear velocity of the mobile phase on chromatographic efficiency showed good enantioresolutions up to a value of 5.11 at the optimal linear velocity with efficiencies in terms of number of plates per meter in the range 51,650–68,330. The results were compared with the ones obtained employing 5 μm vancomycin modified diol-silica particles packed in capillaries of the same i.d. For the acidic analytes the sub-2 μm CSP showed better performances, the baseline chiral separation of several studied compounds occurred in an analysis time of less than 3 min. Column-to-column packing reproducibility ( n = 3) expressed as relative standard deviation was in the range 2.2–5.8% and 0.5–7.7% for retention times and peak areas, respectively. This paper reports our investigation regarding the thermal properties of new polymer-silica hybrid materials obtained by radiation grafting. The polymer poly(dimethylsiloxane),bis(3-aminopropyl)terminated is γ-grafted on a silica gel surface. The thermal behaviour of γ-grafted hybrid materials reveals remarkable differences compared to the thermal behaviour of physically adsorbed polymers. These differences allow us to assess the ability of γ-rays to produce a polymer chemically bonded on a silica surface. Abstract: We have developed conceptually new stationary phases containing two bidentate urea-type functions suitable for the separation of a wide variety of polar compounds by hydrophilic interaction chromatography (HILIC) through a facile one-pot two-step procedure with the aim of obtaining high hydrolytic stability in a variety of elution conditions. The preparation of the new phases involves a first reaction of 1,2-ethylendiamine with (3-isocyanatopropyl)triethoxysilane to give an intermediate bis-urea with two pendant triethoxysilane functions, followed by anchoring on the silica surface. Two stationary phases were prepared, namely an urea-type stationary phase (USP-HILIC) and an urea-type phase bearing free amino groups (USP-HILIC-NH2), whereas silanization with 1,2-bis(trichlorosilyl)ethane yielded USP-HILIC-sil and USP-HILIC-NH2-sil phases, respectively. The silanization step aimed at forming a hydrophilic stable coating through cross-linking between adjacent silanols which prevents silica dissolution at alkaline pH. A full chemical characterization of the new materials has been obtained through solid-state NMR (both 29Si and 13C CPMAS) spectroscopy. A major application field of the bidentate urea-type stationary phase with free amino groups USP-HILIC-NH2-sil was sugars analysis, usually hampered by α/β anomer peak splitting and instability of the stationary phases under conditions normally employed to suppress it. Complex mixtures of mono-, di- and oligosaccharides were successfully resolved under mild chromatographic conditions, which also allowed an easy interface with mass spectrometry. The potential of such materials was shown in the separation of other highly polar compounds, including polyols, hydroxybenzoic acids, nucleobases, and vitamins. [Copyright &y& Elsevier]

The solid state and solution structure of 2,3-dicyano-5,6-di(2-thienyl)-1,4-pyrazine, [(CN)2Th2Pyz], and its PdII derivative, [(CN)2Th2Pyz(PdCl2)2]·H2O, formed by reaction of [(CN)2Th2Pyz] with [(C6H5CN)2PdCl2] were characterized by X-ray, UV-visible, ¹H and 13C NMR, and extended X-ray absorption fine structure (EXAFS) spectral measurements. The X-ray crystal structure of [(CN)2Th2Pyz] shows the presence of one thienyl ring positioned orthogonal to the rest of the molecule, with the two vicinal thienyl rings lying orthogonal to each other in a rare arrangement. NMR studies of [(CN)2Th2Pyz] in the solid state and in solutions of dimethylformamide or dimethyl sulfoxide confirm a nonequivalence of the thienyl rings in the solid state and also in solution. EXAFS results indicate that two distinct PdII coordination sites are formed at the di(2-thienyl)pyrazino moiety of [(CN)2Th2Pyz(PdCl2)2]·H2O, with identical Pd-Npyz (2.03(3) Å) and Pd-Cl (2.36(3) Å) bond lengths but with different Pd-S1 (2.25(4) Å) and Pd-S2 (3.21(5) Å) bond distances in an overall asymmetric molecular framework. Density functional theory (DFT) and time-dependent DFT (TDDFT) theoretical studies also provide information about the structure and spectral behavior of the precursor and its metalated PdII derivative. ¹H/13C NMR and UV-visible spectral measurements were also carried out on two heteropentametallic porphyrazine macrocycles which were prepared by a reaction of PdCl2 with [Th8TPyzPzM] where Th8TPyzPz = tetrakis-2,3-[5,6-di-(2-thienyl)-pyrazino]porphyrazinato dianion and M = MgII(H2O) or ZnII. Spectroscopic data on the newly synthesized [(PdCl2)4Th8TPyzPzM] compounds suggest that the binding of PdCl2 involves coordination sites of the type S2(th)PdCl2 with the two thienyl rings of each di(2-thienyl)pyrazino fragment bound to PdII in an equivalent manner ("th-th" coordination). Abstract: Hydrogels have been widely used in tissue engineering as a support for tissue formation and/or to deliver drug locally. A novel procedure for the in situ rapid chemical gelation of aqueous solutions of hyaluronan (HA) was employed. HA was functionalised with an arm bearing a terminal azido group (HAAA). When HAAA was mixed with a series of dialkyne reagents of different length, a 1,3-dipolar cycloaddition (“click-chemistry”) reaction took place in the presence of catalytic amount of Cu(I) resulting in fast gelation at room temperature. The resulting gels were characterised in terms of degree of cross-linking by 1H HR-MAS NMR. The kinetic of gelation and the determination of elastic moduli as well as the degree of swelling and the controlled release of a model drug, were studied as a function of chemical nature of the dialkyne group, catalyst concentration, HAAA concentration and temperature. All these variables allowed the swelling ratio and the extent of release of a drug, doxorubicin, entrapped within the gel, to be modulated. In all cases the kinetic of release reached the stationary state within 150h. The height of the plateau was dependent on the overall (chemical and topological) degree of cross-linking. Abstract: Grafting of methyl methacrylate and ethyl acrylate onto cellulose chains of textiles with oxidized sites has been carried out. To this aim, carboxyl and carbonyl groups have been introduced by oxidation of the cellulose and used as photosensitive agents, allowing the formation of radical sites. The copolymerization has been started irradiating the substrate with ultraviolet light for a short time. With the grafting of acrylic monomers the mechanical strength and the thermal stability of cellulose based textiles may be improved, as well as the resistance to chemical and biological agents. The method has been successfully tested performing the grafting on a naturally aged cotton sample. It is worth to note that the grafting does not affect the aspect of the material. All samples have been characterized with 13C solid state NMR spectroscopy. A semi-quantitative evaluation of the grafting has been performed either studying the cross-polarization dynamic process, or by direct integration of resonances in 13C single pulse excitation spectra. A high-field NMR study of the polysaccharidic fraction extracted from Lentinula edodes mycelium grown on olive mill waste waters is reported. Diffusion-ordered NMR spectroscopy (DOSY) was applied to the polysaccharidic fraction. The results showed the presence of two polysaccharides of different sizes, whose structures were revealed using one- and two-dimensional NMR techniques. Owing to the amphiphilic nature of their constituent molecules, binary mixtures of pure liquid surfactants are usually characterized by enhanced nano-segregation and thus can exhibit interesting transport properties and complex macroscopic behavior. In this ambit it was recently shown by Turco Liveri et al. (J. Mol. Liq. 263 (2018) 274–281) at room temperature that mixtures of short aliphatic chains compounds, such as dibutyl phosphate (DBP) and n -propylamine (PA) liquids, due to their ability to allow for phosphate-to-amine proton transfer, display ionic liquid–like behavior with composition-dependent enhanced conductivity, viscosity, and magnetically-induced birefringence. To understand the molecular mechanisms at the basis of this behavior, in the present study a combination of nuclear magnetic resonance (NMR) and dielectric spectroscopy investigations has been carried out for the same materials for different amine molar ratios. It was found that at certain compositions all studied dynamical processes (conformational changes, local hopping of "free" protons among neighboring polar headgroups, long-range charge migration) exhibit significant deviations from ideal mixing behavior. The worldwide-cultivated chicory (Cichorium intybus L.) produces food and beneficial compounds, and young pre-flowering inflorescence stems are newly marketed vegetables. These sink-organs undergo growth by metabolizing sugars of leaf origin; the carbohydrate content and sweetness are crucial aspects for consumers' nutrition and acceptance. NMR profiling of 31 hydrosoluble phytochemicals showed that stem contents varied as influenced by genotype, environment and interaction, and that higher sucrose levels were associated with the sweeter of two landraces. Integrative analyses of metabolic and transcriptomic profile variations allowed the dissection of sucrose pathway. Overall, 427 and 23 unigenes respectively fell into the categories of sucrose metabolism and sugar carriers. Among 10 differentially expressed genes, the 11474/sucrose synthase , 53458/fructokinase , 9306 and 17035/hexokinases , and 20171/SWEET-type genes significantly associated to sugar content variation, and deduced proteins were characterised in silico. Correlation analyses encompassing sugar level variation, expressions of the former genes and of computationally assigned transcription factors (10938/NAC , 14712/bHLH , 40133/TALE and 17846/MIKC) revealed a gene network. The latter was minimally affected by the environment and accomplished with markers, representing a resource for biological studies and breeding. Integrative analysis combining metabolic and gene transcription profiles were used to give insights into chicory stem sugar metabolism of two landraces grown in different sites in order to define a gene network subtending sugar content differences. Lignin was isolated from wood wastes comprising Iroko sawdust (IR) and mixed sawdust from Iroko and Norway spruce (IRNS), furnished by a local wood houses producer. The respective acidolysis lignin fractions were structurally characterized using pyrolysis (Py)-GCMS, two-dimensional heteronuclear single quantum correlation nuclear magnetic resonance (2D HSQC NMR), Fourier-transform infrared FTIR and ultraviolet-visible (UV-VIS) spectroscopies, size exclusion chromatography, and standard wet-chemistry methods for Klason lignin and polysaccharides determination. The isolated lignin fractions were subsequently used for the preparation of lignin nanoparticles (LNPs) using a non-solvent method. LNPs were then used for wood surface treatment using a dip-coating technique. The coated wood samples were analyzed by colorimetry and scanning electron microscopy (SEM) before and after artificial weathering experiments in a UV chamber to investigate the UV protection potential of the LNPs coatings. Wood samples dip-coated with LNPs showed promising surface modifications resembling a sort of film of fused LNPs. Coatings made from IR-LNPs and IRNS-LNPs performed significantly better in artificial weathering experiments than uncoated reference samples. [ABSTRACT FROM AUTHOR]

In the wide panorama of diacetylenic lipids, the photoresponsive conjugated 1,3-diyne function is usually encased into the hydrocarbon chain of the amphiphile at a variable distance from the headgroup. Therefore, the polydiacetylene network obtained by polymerization upon UV irradiation of the corresponding liposomes, exploited as sensing function, is embedded in the hydrophobic region of liposomes. Structurally related cationic diacetylenic amphiphiles featuring the conjugated triple bonds proximate to charged nitrogen were synthesized and evaluated in their ability to polymerize under aggregative conditions. The occurrence of polymerization only in certain aggregating conditions was rationalized by nuclear magnetic resonance (NMR) and Langmuir trough experiments.

The chemical composition of the inflorescences from four Cannabis sativa L. monoecious cultivars (Ferimon, Uso-31, Felina 32 and Fedora 17), recently introduced in the Lazio Region, was monitored over the season from June to September giving indications on their sensorial, pharmaceutical/nutraceutical proprieties. Both untargeted (NMR) and targeted (GC/MS, UHPLC, HPLC-PDA/FD and spectrophotometry) analyses were carried out to identify and quantify compounds of different classes (sugars, organic acids, amino acids, cannabinoids, terpenoids, phenols, tannins, flavonoids and biogenic amines). All cultivars in each harvesting period showed a THC content below the Italian legal limit, although in general THC content increased over the season. Citric acid, malic acid and glucose showed the highest content in the late flowering period, whereas the content of proline drastically decreased after June in all cultivars. Neophytadiene, nerolidol and chlorogenic acid were quantified only in Felina 32 cultivar, characterized also by a very high content of flavonoids, whereas alloaromadendrene and trans -cinnamic acid were detected only in Uso-31 cultivar. Naringenin and naringin were present only in Fedora 17 and Ferimon cultivars, respectively. Moreover, Ferimon had the highest concentration of biogenic amines, especially in July and August. Cadaverine was present in all cultivars but only in September. These results suggest that the chemical composition of Cannabis sativa L. inflorescences depends on the cultivar and on the harvesting period. Producers can use this information as a guide to obtain inflorescences with peculiar chemical characteristics according to the specific use.

Celery is a widely used vegetable known for its peculiar sensorial and nutritional properties. Here, the white celery (Apium graveolens L.) "sedano bianco di Sperlonga" PGI ecotype was investigated to obtain the metabolic profile of its edible parts (blade leaves and petioles) also related to quality, freshness and biological properties. A multi-methodological approach, including NMR, MS, HPLC-PDA, GC-MS and spectrophotometric analyses, was proposed to analyse celery extracts. Sugars, polyalcohols, amino acids, organic acids, phenols, sterols, fatty acids, phthalides, chlorophylls, tannins and flavonoids were detected in different concentrations in blade leaf and petiole extracts, indicating celery parts as nutraceutical sources. The presence of some phenols in celery extracts was here reported for the first time. Low contents of biogenic amines and mycotoxins confirmed celery quality and freshness. The study focuses on the understanding, at molecular level, the mechanism of interaction between protein and flavonoids. Collagen and catechin interactions were investigated by NMR in solution and solid state. The effect of catechin on the stability of collagen to oxidation was also explored. Collagen was treated with two concentrations of catechin solutions. Oxidation was carried out by incubation of collagen solution with three oxidation systems: Fe(II)/H 2 O 2 , Cu(II)/H 2 O 2 , and NaOCl/H 2 O 2 . The effects of oxidation systems were evaluated by high resolution 1 D and 2 D proton spectroscopy and solid state NMR ( 13 C CP MAS) experiments. Interactions between collagen and catechin preferentially occur between catechin B ring and the amino acids Pro and Hyp of collagen. Results showed that both iron and copper oxidation systems were able to interact with collagen by site specific attack. Moreover, catechin protects collagen proline from oxidation by metal/H 2 O 2 systems, preventing copper and iron approach to collagene molecule;this behaviour was more evident for the copper/H 2 O 2 system.

Extra-virgin olive oil (383 samples; EVOOs) of three consecutive harvesting years from nine Italian regions were collected and submitted to an ¹H NMR-chemometric protocol to characterize the samples according to their origin (geographical area and variety). A more complete assignment of the olive oil ¹H spectrum in CDCl₃ and DMSO d ₆ was reported identifying 24-methylencycolartanol. A single classification model provided the discrimination of EVOOs among the three geographical macro-areas (North, Islands, Center-South), whereas a hierarchical approach based on breaking the overall classification problem into a series of smaller linear discriminant analysis (LDA) sub-models was tested to differentiate olive oils according to their geographical regions. Actinidia deliciosa cv. Hayward fruit is renowned for its micro- and macronutrients, which vary in their levels during berry physiological development and postharvest processing. In this context, we have recently described metabolic pathways/molecular effectors in fruit outer endocarp characterizing the different stages of berry physiological maturation. Here, we report on the kiwifruit postharvest phase through an integrated approach consisting of pomological analysis combined with NMR/LC-UV/ESI-IT-MS n - and 2D-DIGE/nanoLC-ESI-LIT-MS/MS-based proteometabolomic measurements. Kiwifruit samples stored under conventional, cold-based postharvest conditions not involving the use of dedicated chemicals were sampled at four stages (from fruit harvest to pre-commercialization) and analyzed in comparison for pomological features, and outer endocarp metabolite and protein content. About 42 metabolites were quantified, together with corresponding proteomic changes. Proteomics showed that proteins associated with disease/defense, energy, protein destination/storage, cell structure and metabolism functions were affected at precise fruit postharvest times, providing a justification to corresponding pomological/metabolite content characteristics. Bioinformatic analysis of variably represented proteins revealed a central network of interacting species, modulating metabolite level variations during postharvest fruit storage. Kiwifruit allergens were also quantified, demonstrating in some cases their highest levels at the fruit pre-commercialization stage. By lining up kiwifruit postharvest processing to a proteometabolomic depiction, this study integrates previous observations on metabolite and protein content in postharvest berries treated with specific chemical additives, and provides a reference framework for further studies on the optimization of fruit storage before its commercialization.

The adsorption behavior of neodymium (Nd 3+ ) and yttrium (Y 3+ ) cations on synthetic FAU zeolite X in its sodium form (NaX) has been investigated by means of macroscopic (adsorption isotherm determination and thermal analysis) and microscopic measurements (including solid-state NMR spectroscopy and X-ray powder diffraction). The multidisciplinary study reveals some unexpected features. Firstly, adsorption constants of cations are not correlated to their ionic radii or hydration enthalpy. The adsorption constant of Y 3+ on NaX was indeed about twice that of Nd 3+ , which is the opposite of what could be expected based on the size of the cations. In addition, adsorption was accompanied by partial dealumination of the zeolite framework. The extent of dealumination changed depending on the exchanged cations, with the extent being more significant on the Nd-exchanged zeolite than on the Y-exchanged one. The most interesting finding of this study, however, is the presence of supramolecular clusters composed of water, Nd 3+ , residual sodium ions, and extra-framework aluminum at the interface of Nd-exchanged zeolite. A multi-methodological approach was applied to study red sweet peppers (Capsicum annuum L.) ecotype "Cornetto di Pontecorvo" grown in a greenhouse or in open field. This approach includes morphological analysis, chemical composition determination, and biological activity evaluation of different extracts from pepper fruits. Untargeted analyses, namely NMR spectroscopy and mass spectrometry, allowed the comprehensive pepper metabolite profile of pepper pulp, peel and seeds hydroalcoholic and organic extracts to be determined, showing the presence of sugars, organic acids, amino acids and other secondary metabolites. Targeted analyses, such as HPLC-PDA, HPLC-TLC and spectrophotometric analyses allowed polyphenols, tannins, flavonoids and pigments content to be determined. Samples quality and freshness were verified by the low content of biogenic amines and mycotoxins, as determined using HPLC-FLD and HPLC-MS, respectively. An analytical approach including Panel Test, Isotope Ratio Mass Spectrometry (IRMS) and Nuclear Magnetic Resonance (NMR) spectroscopy was proposed to characterize Italian " Colline Pontine " PDO olive oils (40 samples) of two consecutive crop years. Our approach has evidenced the high quality of these olive oils. Only 6 of 40 olive oils samples were defined as " defective " by the official Panel Test due to the detection of negative sensory attributes. The low variability of isotopic data monitored by IRMS confirmed that the olive oil samples all came from a limited geographical area. NMR spectra did not evidence any chemical composition anomaly in the investigated samples. In order to assess the influence of harvesting year over the olive oil chemical composition, the NMR analysis was extended to other 22 olive oil samples of a third harvesting year. NMR data were submitted to two different statistical methods, namely, analysis of variance (ANOVA) and principal component analysis (PCA) allowing olive oils of three consecutive harvesting years to be grouped.

Capillary methacrylate-based monoliths were prepared for the high performance liquid chromatography (HPLC) separation of both small molecules and large biomolecules. An efficient grafting from/to synthetic approach was adopted introducing a network of activated sites in the inner wall surface using the new silanization agent (N-trimethoxysilylpropyl)-polyethylenimine. Copolymerization of lauryl methacrylate monomer and 1,6-hexanediol dimethacrylate cross-linker in the presence of porogenic solvents was obtained under continuous γ-ray exposure with high conversion yield. The morphology and porous structure of the resulting monoliths have been investigated by Scanning Electron Microscopy (SEM) and 1 H NMR cryoporosimetry. By chromatographic investigation, the new capillary columns attested high kinetic performance (with efficiency larger than 100,000 theoretical plate/m for small molecules at optimum mobile phase linear velocity of about 0.5mm/s) and also excellent mechanical stability and repeatability. An NMR and chemometric analytical approach to classify beers according to their brand identity was developed within the European TRACE project (FP6-2003-FOOD-2-A, contract number: 0060942). Rochefort 8 Trappist beers (47 samples), other Trappist beers (76 samples) and non-Trappist beers (110 samples) were analyzed by 1 H NMR spectroscopy. Selected NMR signals were measured and used to build classification models. Three different classification problems were identified, namely Trappist versus non-Trappist, Rochefort versus Non-Rochefort, and Rochefort 8 versus non-Rochefort 8. In all the three cases, both a discriminant and a modeling approaches were followed, using partial least squares discriminant analysis (PLS-DA) and soft independent modeling of class analogies (SIMCA), respectively, leading to very high classification accuracy as evaluated by external validation. Information regarding chemical composition was also obtained: Trappist beers contain a higher amount of formic and pyruvic acids and a lower amount of acetic acid and alanine with respect to non-Trappist ones. Rochefort beers turned out to have also a higher content of propanol and isopentanol with respect to non-Rochefort samples. Purpose: To synthesize a new polymeric prodrug based on α,β-poly(N-2-hydroxyethyl)(2-aminoethylcarbamate)-d,l-aspartamide copolymer bearing amine groups in the side chain (PHEA-EDA), covalently linked to the anticancer drug doxorubicin and to test its potential application in anticancer therapy., Methods: The drug was previously derivatized with a biocompatible and hydrophilic linker, leading to a doxorubicin derivative highly reactive with amino groups of PHEA-EDA. The PHEA-EDA-DOXO prodrug was characterized in terms of chemical stability. The pharmacokinetics, biodistribution and cytotoxicity of the product was investigated in vitro and in vivo on human breast cancer MCF-7 and T47D cell lines and NOD-SCID mice bearing a MCF-7 human breast carcinoma xenograft. Data collected were compared to those obtained using free doxorubicin., Results: The final polymeric product is water soluble and easily hydrolysable in vivo, due to the presence of ester and amide bonds along the spacer between the drug and the polymeric backbone. In vitro tests showed a retarded cytotoxic effect on tumor cells, whereas a significant improvement of the in vivo antitumor activity of PHEA-EDA-DOXO and a survival advantage of the treated NOD-SCID mice was evidenced, compared to that of free doxorubicin., Conclusions: The features of the PHEA-EDA-DOXO provide a potential protection of the drug from the plasmatic enzymatic degradation and clearance, an improvement of the blood pharmacokinetic parameters and a suitable body biodistribution. The data collected support the promising rationale of the proposed macromolecular prodrug PHEA-EDA-DOXO for further potential development and application in the treatment of solid cancer diseases.

NMR stratigraphy was used to investigate in situ, non-destructively and non-invasively, the stratigraphy of hydrogen-rich layers of an ancient Nubian detached mural painting. Because of the detachment procedure, a complex multi-layered artefact was obtained, where, besides layers of the original mural painting, also the materials used during the procedure all became constitutive parts of the artefact. NMR measurements in situ enabled monitoring of the state of conservation of the artefact and planning of minimum representative sampling to validate results obtained in situ by solid-state NMR analysis of the samples. This analysis enabled chemical characterization of all organic materials. Use of reference compounds and prepared specimens assisted data interpretation.

The aerobic metabolism of a mixture of acetate and starch was studied with main emphasis on their interaction and the effect on their storage as PHB and glycogen, respectively. Pulse feeding strongly increased the storage of both substrates; however, the presence of starch decreased PHB storage whereas the presence of acetate did not affect glycogen storage. Indeed, 13C NMR isotopomer analysis suggested an increase of acetate utilization towards TCA cycle, due to an increased request of ATP production for glycogen biosynthesis regulated by ADP-GlcPPase. This in turn influenced the partition flux for pyruvate synthesis between TCA cataplerosis and glyoxylate shunt. The corresponding reduction of PHB synthesis was in agreement with the competition for HS-CoA between KGDH activity and acetyl-CoA for PHB synthesis pathway. As a practical consequence, bioprocesses for PHA production from volatile fatty acids could be negatively affected from other carbon sources, such as unfermented carbohydrates., (Copyright © 2012 Elsevier Ltd. Hydrophobic treatment is one of the most important interventions usually carried out in the conservation of stone artifacts and monuments. The analytical study reported in this paper was aimed at answering general questions such as the penetration depth of a hydrophobic treatment into a porous material, its capability to impair the water absorption, how the presence of a treatment may change the open porosity available to the water, and how a treatment may affect the diffusion of water inside a porous structure. Also, inhomogeneities in treated stones due to sharp variations of the amount of the absorbed product in the porous material were evidenced and scaled. The results of this fully non-invasive analytical study were rationalized in terms of new parameters obtained by a suitable process of nuclear magnetic resonance data. These analytical parameters reported here for the first time, namely the hydrophobic efficiency, the penetration depth, and angles describing changes in slope in depth profiles, gave important information in assessing the performance of a treatment.

The system described in this paper was obtained by soaking calcium alginate (CaAlg) microspheres in a water solution of poly-[(3-acrylamidopropyl)-trimethylammonium chloride-b-N-isopropylacrylamide] [poly(AMPTMA-b-NIPAAM)], a new block co-polymer recently synthesized by atom transfer radical polymerization (ATRP). The block co-polymer is characterized by a lower critical solution temperature (LCST) of 41 degrees C in aqueous 0.1 M NaCl solution, and can be anchored on the CaAlg microspheres by means of polyion interactions. Polycations (permanently positively charged blocks) and polyanions (free alginate carboxylic groups) interact, leading to microspheres with thermosensitive properties. As an effect of interaction with the microspheres the LCST of the co-polymer is lowered to 36-38 degrees C. In this temperature range a colloidal water suspension of the microspheres collapses, forming macroscopic aggregates. The new system shows, at human body temperature, an improved ability to carry and deliver both hydrophobic and hydrophilic molecules in comparison with unmodified CaAlg microspheres. The release properties of the microspheres loaded with different model drugs can be appropriately modulated by the amount of the poly(AMPTMA-b-NIPAAM). Furthermore, the microspheres show the interesting capability of retaining the activity of a loaded enzyme (horseradish peroxidase), used as a model protein. Organically modified alpha-layered zirconium phosphate samples (ZrP(C(12))(x)) containing dodecyl groups bonded to the alpha-layers through P-O-C bonds have been prepared by reaction of 1,2-epoxydodecane solutions in tetrahydrofuran (THF) with gels of partially exfoliated zirconium phosphate in THF. Two dimensional correlation solid state NMR experiments for (1)H-(13)C and (1)H-(31)P nuclei have been used to prove the formation of P-O-C bonds arising from nucleophilic attack of POH mainly to carbon 1 and, to a lesser extent, to carbon 2 of epoxydodecane. ZrP(C(12))(x) samples with x in the range from approximately 0.5 to approximately 2.0 are thermally stable up to at least 200 degrees C, and their interlayer distance increases continuously with x from approximately 20 to approximately 35 A. On the basis of structural considerations, it has been suggested that samples with low x values could intercalate aliphatic polymers. Accordingly, preliminary results have shown that molten polyethylene is intercalated in ZrP(C(12))(0.49) and ZrP(C(12))(0.73). These materials can therefore be regarded as filler candidates for polymer matrixes.

As is well known, the deterioration of wall paintings due to the capillary rise of water through the walls is a very widespread problem. In this paper, a study of microclimate monitoring, unilateral nuclear magnetic resonance (NMR), and evanescent-field dielectrometry (EFD) was applied to map non-destructively, in situ, and in a quantitative way the distribution of the moisture in an ancient deteriorated wall painting of the eleventh century. Both unilateral NMR and EFD are quite new, fully portable, and non-destructive techniques, and their combination is absolutely new. The approach reported here is proposed as a new analytical protocol to afford the problem of mapping, non-destructively, the moisture in a deteriorated wall painting in a hypogeous building such as that of the second level of St. Clement Basilica, Rome (Italy), where the use of IR thermography is impaired due to the environmental conditions, and the gravimetric tests are forbidden due to the preciousness of the artifact. The moisture distribution was mapped at different depths, from the very first layers of the painted film to a depth of 2 cm. It has also been shown how the map obtained in the first layers of the artwork is affected by the environmental conditions typical of a hypogeous building, whereas the maps obtained at higher depths are representative of the moisture due to the capillary rise of water from the ground. The quantitative analysis of the moisture was performed by calibrating NMR and EFD signals with purposely prepared specimens. This study may be applied before and after performing any intervention aimed at restoring and improving the state of conservation of this type of artwork and reducing the dampness or extracting salts (driven by the variation of moisture content) and monitoring the effectiveness of the performed interventions during the time. This protocol is applicable to any type of porous material.

An integrated study of microclimate monitoring, IR thermography (IRT), gravimetric tests and portable unilateral nuclear magnetic resonance (NMR) was applied in the framework of planning emergency intervention on a very deteriorated wall painting in San Rocco church, Cornaredo (Milan, Italy). The IRT investigation supported by gravimetric tests showed that the worst damage, due to water infiltration, was localized on the wall painting of the northern wall. Unilateral NMR, a new non-destructive technique which measures the hydrogen signal of the moisture and that was applied directly to the wall, allowed a detailed map of the distribution of the moisture in the plaster underlying the wall panting to be obtained. With a proper calibration of the integral of the recorded signal with suitable specimens, each area of the map corresponded to an accurate amount of moisture. IRT, gravimetric tests and unilateral NMR applied to investigate the northern wall painting showed the presence of two wet areas separated by a dry area. The moisture found in the lower area was ascribed to the occurrence of rising damp at the bottom of the wall due to the slope of the garden soil towards the northern exterior. The moisture found in the upper area was ascribed to condensation phenomena associated with the presence of a considerable amount of soluble, hygroscopic salts. In the framework of this integrated study, IRT investigation and gravimetric methods validated portable unilateral NMR as a new analytical tool for measuring in situ and without any sampling of the distribution and amount of moisture in wall paintings.

A portable unilateral nuclear magnetic resonance (NMR) instrument was used to detect in field conditions the water status of leaves of herbaceous crops (Zea mays, Phaseolus vulgaris), mesophyllous trees (Populus nigra), and natural Mediterranean vegetation characterized by water-spending shrubs (Cistus incanus) and water-saving sclerophyllous trees (Quercus ilex). A good relationship was observed between NMR signal, leaf relative water content, and leaf transpiration in herbaceous leaves undergoing fast dehydration or slowly developing a drought stress. A relationship was also observed between NMR signal and water potential of Populus leaves during the development of a water stress and when leaves recovered from the stress. In the natural vegetation, the relationship between NMR signal and water status was found in Cistus, the species characterized by high transpiration rates, when measured during a drought stress period and after a rainfall. In the case of the sclerophyllous Quercus, the NMR signal, the relative water content, and the transpiration rate did not change at different leaf water status, possibly because a large amount of water is compartmentalized in cellular structures and macromolecules. The good association between NMR signal and relative water content was lost in leaves exposed for 24 h to dehydration or to an osmotic stress caused by polyethylene glycol feeding. At this time, the transverse relaxation time became longer than in leaves maintained under optimal water conditions, and two indicators of membrane damage, the ion leakage and the emission of products of membrane lipoxygenation [(Z)-3-hexenal, (Z)-3-hexenol, and (E)-2-hexenol], increased. These results taken all together give information on the physiological state of a leaf under a developing stress and show the usefulness of the NMR instrumentation for screening vegetation health and fitness in natural and cultivated conditions. It is concluded that the portable unilateral NMR instrument may be usefully employed in field conditions to monitor nondestructively the water status of plants and to assist agricultural practices, such as irrigation scheduling, to minimize stomatal closure and the consequent limitation to plant production.

pi-Conjugated low molecular weight polymers characterized by regio- and stereoregular alternation of phenylene and ( E)-1-en-3-yne moieties have been synthesized by polyaddition of 1,4-diethynylbenzene or of 2,5-diethynyl-1,4-alkoxybenzene monomers, employing the commercially available di-micro-chlorobis[( p-cymene)chlororuthenium(II)] complex as the metal catalyst source, under homogeneous, atom-economical, amine- and phosphine-free conditions. Bulk materials of poly( p-phenyleneethynylenevinylene) derivatives are obtained with yields larger than 80%, from which polymers readily soluble in chlorinated solvents and in tetrahydrofuran are extracted in 60-75% yields. The polymers with average degrees of polymerization in the range n AV = 4-8 display optical properties in solution similar to those of the higher molecular weights analogues.
