This paper presents the results of simultaneous high time-resolution measurements of biogenic aerosol (methane sulfonic acid (MSA), non-sea salt sulfate nssSO42-) with its gaseous precursor dimethylsulfide (DMS), performed at the Italian coastal base Mario Zucchelli Station (MZS) in Terra Nova Bay (MZS) during two summer campaigns (2018–2019 and 2019–2020). Data on atmospheric DMS concentration are scarce, especially in Antarctica. The DMS maximum at MZS occurs in December, one month earlier than at other Antarctic stations. The maximum of DMS concentration is connected with the phytoplanktonic senescent phase following the bloom of Phaeocystis antarctica that occurs in the polynya when sea ice opens up. The second plankton bloom occurs in January and, despite the high dimethylsufoniopropionate (DMSP) concentration in seawater, atmospheric DMS remains low, probably due to its fast biological turnover in seawater in this period. The intensity and timing of the DMS evolution during the two years suggest that only the portion of the polynya close to the sampling site produces a discernible effect on the measured DMS. The closeness to the DMS source area and the occurrence of air masses containing DMS and freshly formed oxidation products allow us to study the kinetic of biogenic aerosol formation and the reliable derivation of the branch ratio between MSA and nssSO42- from DMS oxidation that is estimated to be 0.84±0.06. Conversely, for aged air masses with low DMS content, an enrichment of nssSO42- with respect to MSA, is observed. We estimate that the mean contribution of freshly formed biogenic aerosol to PM10 is 17 % with a maximum of 56 %. The high contribution of biogenic aerosol to the total PM10 mass in summer in this area highlights the dominant role of the polynya on biogenic aerosol formation. Finally, due to the regional and year-to-year variability of DMS and related biogenic aerosol formation, we stress the need for long-term measurements of seawater and atmospheric DMS and biogenic aerosol along the Antarctic coast and in the Southern Ocean.

Abstract Targeting necroptosis is considered a promising therapeutic strategy in cancer, including Glioblastoma Multiforme (GBM), one of the most lethal brain tumors. Necroptosis is a mechanism of programmed cell death overcoming the apoptosis resistance mechanism underlying GBM tumorigenesis and malignant progression. N6-isopentenyladenosine (iPA), adenosine modified with isoprenoid derivative, displays antitumor activity in different cancer models. In previous studies, we demonstrated that iPA interferes with EGFR signaling reducing glioma cell viability. Here, we show that iPA induces necroptosis in glioblastoma cell lines and in primary cells established from tumor explants, without affecting the viability of non-cancerous brain cell lines, (Normal Human Astrocyte). The activation of RIP1, RIP3, and MLKL and the upregulation of necrosome formation were increased upon iPA treatment while caspase-3, caspase-8, and PARP were not activated in GBM cells. Co-treatment with specific necroptosis inhibitor necrostatin-1 (Nec-1) or Necrosulfonamide (NSA) prevented cell death caused by iPA treatment while the general caspase inhibitor Z-VAD-fluoromethylketone (z-VAD-fmk) did not elicit any effect, suggesting that this molecule induces caspase-independent necroptosis. These results suggest that iPA treatment can be able to bypass the apoptosis resistance mechanism in glioblastoma thereby offering higher therapeutic efficacy.

Abstract Long-term exposure to air pollution has been associated with the development of some inflammatory processes related to skin. The goal of modern medicine is the development of new products with antiflammatory action deriving from natural sources to improve environmental and economic sustainability. In this study, two different humic acids (HA) were isolated from from lignite (HA-LIG) and composted artichoke wastes (HA-CYN) and characterized by infrared spectrometry, NMR spectroscopy, thermochemolysis-GC/MS, and high-performance size-exclusion chromatography (HPSEC), while their antiflammatory activity was evaluated on HaCaT cells. Spectroscopic results showing the predominance of apolar aliphatic and aromatic components in HA-LIG, whereas HA-CYN revealed a presence of polysaccharides and polyphenolic lignin residues. The HA application on human keratinocyte pre-treated with Urban Dust revealed a general increase of viability suggesting a protective effect of humic matter due to the content of aromatic, phenolic and lignin components. Conversely, the gene expression of IL-6 and IL-1β cytokines indicated a significant decrease after application of HA-LIG, thus exhibiting a greater antiflammatory power than HA-CYN. The specific combination of HA protective hydrophobic components, viable conformational arrangements, and content of bioactive molecules, suggests an innovative applicability of humic matter in dermatology as skin protectors from environmental irritants and as antiflammatory agents.

From the recent awareness of the booming number of space debris and their derived worldwide re-entry event threat originating from the use of high survivability components, complementary mitigation measures must be taken for future orbital elements. In this direction, the implementation of a design for demise at an early stage of spacecraft conception allows a progressive and effective solution. As part of a collaborative effort launched by École Polytechnique Fédérale de Lausanne and the European Space Agency, we focus on the evaluation of polymer composite bolts as the main structural satellite panel fastening systems to improve the overall spacecraft demisability during its destructive re-entry into the atmosphere as compared to baseline critical systems, while maintaining equivalent mission-relevant properties. Two carbon fiber-reinforced polyetheretherketone (PEEK) designs were compared to a stainless steel baseline in terms of static properties at room temperature, dynamic properties over a temperature range, as well as demise capability by static re-entry simulation testing. Natural killer (NK) cells have recently gained popularity as an alternative for cancer immunotherapy. Adoptive cell transfer employing NK cells offers a safer therapeutic option compared to T-cell-based therapies, due to their significantly lower toxicity and the availability of diverse autologous and allogeneic NK cell sources. However, several challenges are associated with NK cell therapies, including limited in vivo persistence, the immunosuppressive and hostile tumor microenvironment (TME), and the lack of effective treatments for solid tumors. To address these limitations, the modification of NK cells to stably produce cytokines has been proposed as a strategy to enhance their persistence and proliferation. Additionally, the overexpression of activating receptors and the blockade of inhibitory receptors can restore the NK cell functions hindered by the TME. To further improve tumor infiltration and the elimination of solid tumors, innovative approaches focusing on the enhancement of NK cell chemotaxis through the overexpression of chemotactic receptors have been introduced. This review highlights the latest advancements in preclinical and clinical studies investigating the engineering of activating, inhibitory, and chemotactic NK cell receptors; discusses recent progress in cytokine manipulation; and explores the potential of combining the chimeric antigen receptor (CAR) technology with NK cell receptors engineering. Currently, the biomimetic approach of drawing inspiration from nature has frequently been employed in designing drug nanocarriers (NCs) of actively target various diseases, ranging from cancer to neuronal and inflammation pathologies. The cell-membrane coating can confer upon the inner nanomaterials a biological identity and the functions exhibited by the cells from which the membrane is derived. Monocyte- and macrophage-membrane-coated nanomaterials have emerged as an ideal delivery system to target inflamed vasculature. Herein, we developed two biomimetic NCs using a human-derived leukaemia monocytic cell line (THP-1), either undifferentiated or differentiated by phorbol 12-myristate 13-acetate (PMA) into adherent macrophage-like cells as membrane sources for NC coating. We employed a secondary oil-in-water nano-emulsion (SNE) as the inner core, which served as an optimal NC for high payloads of lipophilic compounds. Two different biomimetic systems were produced, combining the biomimetic features of biological membranes with the physicochemical and nano-sized characteristics of SNEs. These systems were named Monocyte NEsoSome (M-NEsoSome) and Macrophage NEsoSome (M0-NEsoSome). Their uptake ability was investigated in tumour necrosis factor alfa (TNFα)-treated human umbilical vein endothelial cells (HUVECs), selected as a model of inflamed endothelial cells. The M0 membrane coating demonstrated accelerated internalisation compared with the monocyte coating and notably surpassed the uptake rate of bare NCs. In conclusion, M0-NEsoSome NCs could be a therapeutic system for targeting inflamed endothelial cells and potentially delivering anti-inflammatory drugs in vascular inflammation. The disruptive deployment of collaborative robots, named cobots, in Industry 5.0 has brought attention to the safety and ergonomic aspects of industrial human–robot interaction (HRI) tasks. In particular, the study of the operator’s mental workload in HRI activities has been the research object of a new branch of ergonomics, called neuroergonomics, to improve the operator’s wellbeing and the efficiency of the system. This study shows the development of a combinative assessment for the evaluation of mental workload in a comparative analysis of two assembly task scenarios, without and with robot interaction. The evaluation of mental workload is achieved through a combination of subjective (NASA TLX) and real-time objective measurements. This latter measurement is found using an innovative electroencephalogram (EEG) device and the characterization of the cognitive workload through the brainwave power ratio β/α, defined after the pre-processing phase of EEG data. Finally, observational analyses are considered regarding the task performance of the two scenarios. The statistical analyses show how significantly the mental workload diminution and a higher level of performance, as the number of components assembled correctly by the participants, are achieved in the scenario with the robot.

We searched the Gaia DR3 database for ultramassive white dwarfs with kinematics consistent with having escaped the nearby Hyades open cluster, identifying three such candidates. Two of these candidates have masses estimated from Gaia photometry of approximately 1.1 solar masses; their status as products of single-stellar evolution that have escaped the cluster was deemed too questionable for immediate follow-up analysis. The remaining candidate has an expected mass >1.3 solar masses, significantly reducing the probability of it being an interloper. Analysis of follow-up Gemini GMOS spectroscopy for this source reveals a nonmagnetized hydrogen atmosphere white dwarf with a mass and age consistent with having formed from a single star. Assuming a single-stellar-evolution formation channel, we estimate a 97.8% chance that the candidate is a true escapee from the Hyades. With a determined mass of 1.317 solar masses, this is potentially the most massive known single-evolution white dwarf and is by far the most massive with a strong association with an open cluster.

We report the discovery of ZTF J0127+5258, a compact mass-transferring binary with an orbital period of 13.7 minutes. The system contains a white dwarf accretor, which likely originated as a post–common envelope carbon–oxygen (CO) white dwarf, and a warm donor ( T _eff,donor = 16,400 ± 1000 K). The donor probably formed during a common envelope phase between the CO white dwarf and an evolving giant that left behind a helium star or white dwarf in a close orbit with the CO white dwarf. We measure gravitational wave–driven orbital inspiral with ∼51 σ significance, which yields a joint constraint on the component masses and mass transfer rate. While the accretion disk in the system is dominated by ionized helium emission, the donor exhibits a mixture of hydrogen and helium absorption lines. Phase-resolved spectroscopy yields a donor radial velocity semiamplitude of 771 ± 27 km s ^−1 , and high-speed photometry reveals that the system is eclipsing. We detect a Chandra X-ray counterpart with L _X ∼ 3 × 10 ^31 erg s ^−1 . Depending on the mass transfer rate, the system will likely either evolve into a stably mass-transferring helium cataclysmic variable, merge to become an R CrB star, or explode as a Type Ia supernova in the next million years. We predict that the Laser Space Interferometer Antenna (LISA) will detect the source with a signal-to-noise ratio of 24 ± 6 after 4 yr of observations. The system is the first LISA-loud mass-transferring binary with an intrinsically luminous donor, a class of sources that provide the opportunity to leverage the synergy between optical and infrared time domain surveys, X-ray facilities, and gravitational-wave observatories to probe general relativity, accretion physics, and binary evolution.

The structural connectivity from the primary olfactory cortex to the main secondary olfactory areas was previously reported as relatively increased in the medial orbitofrontal cortex in a cohort of 27 recently SARS-CoV-2-infected (COV+) subjects, of which 23/27 had clinically confirmed olfactory loss, compared to 18 control (COV-) normosmic subjects, who were not previously infected. To complement this finding, here we report the outcome of an identical high angular resolution diffusion MRI analysis on follow-up data sets collected in 18/27 COV+ subjects (10 males, mean age±SD: 38.7±8.1 years) and 10/18 COV- subjects (5 males, mean age±SD: 33.1±3.6 years) from the previous samples who repeated both the olfactory functional assessment and the MRI examination after ~1 year. By comparing the newly derived subgroups, we observed that the increase in the structural connectivity index of the medial orbitofrontal cortex was not significant at follow-up, despite 10/18 COV+ subjects were still found hyposmic after ~1 year from SARS-CoV-2 infection. We concluded that the relative hyperconnectivity of the olfactory cortex to the medial orbitofrontal cortex could be, at least in some cases, an acute or reversible phenomenon linked to the recent SARS-CoV-2 infection with associated olfactory loss.

Magnetars are the most strongly magnetized neutron stars, and one of the most promising targets for X-ray polarimetric measurements. We present here the first Imaging X-ray Polarimetry Explorer observation of the magnetar 1RXS J170849.0-400910, jointly analyzed with a new Swift observation and archival NICER data. The total (energy- and phase-integrated) emission in the 2–8 keV energy range is linerarly polarized, at a ∼35% level. The phase-averaged polarization signal shows a marked increase with energy, ranging from ∼20% at 2–3 keV up to ∼80% at 6–8 keV, while the polarization angle remains constant. This indicates that radiation is mostly polarized in a single direction. The spectrum is well reproduced by a combination of either two thermal (blackbody) components or a blackbody and a power law. Both the polarization degree and angle also show a variation with the spin phase, and the former is almost anticorrelated with the source counts in the 2–8 and 2–4 keV bands. We discuss the possible implications and interpretations, based on a joint analysis of the spectral, polarization, and pulsation properties of the source. A scenario in which the surface temperature is not homogeneous, with a hotter cap covered by a gaseous atmosphere and a warmer region in a condensed state, provides a satisfactory description of both the phase- and energy-dependent spectro-polarimetric data. The (comparatively) small size of the two emitting regions, required to explain the observed pulsations, does not allow to reach a robust conclusion about the presence of vacuum birefringence effects.

So far, the majority of the studies on the role of exclusive enteral nutrition (EEN) in CD are conducted in the pediatric population and have highlighted the efficacy of EEN for achieving mucosal healing. This implicates that a similar approach would be beneficial in adult patients. However, the evidence for EEN in adults is heterogeneous, with meta-analyses reporting it as inferior to steroids while growing data demonstrate improvement in complicated CD. Currently, EEN is less used in adult patients with IBD. Indeed, the lack of palatability of enteral formula leads to difficulties in acceptance and compliance. The search for more tolerable and still effective diets has become an intense area of research aiming to explore the potential role of diet to control inflammation in patients with CD. Thus, this narrative review provides the state-of-the-art on the use of EEN treatment in CD and highlights the perceived barriers to its implementation in adult CD patients.Keywords: Crohn’s disease, exclusive enteral nutrition, barriers, diet, nutritional interventions
