Conselho Nacional de Desenvolvimento Científico e Tecnológico O principal objetivo deste trabalho foi caracterizar, fenotipicamente, linhagens de feijão do cruzamento Ouro Negro x Pérola, quanto à reação a Colletotrichum lindemuthianum (raças 73, 81 e 89) e Uromyces appendiculatus (mistura das raças 15, 35, 45, 49, 50 e 59) e avaliar, em campo, o desempenho das linhagens que mostraram-se resistentes. Paralelamente, objetivou-se também avaliar a eficiência dos marcadores moleculares SCARF10 1050 (relacionado à resistência à antracnose e à ferrugem) e OPX11 550 (relacionado à resistência à ferrugem), identificados e mapeados em outras populações, com vistas à seleção assistida na população Ouro Negro x Pérola. Inicialmente, 400 linhagens (progênies F 7:8 ) do cruzamento Ouro Negro x Pérola foram inoculadas com a raça 89 de C. lindemuthianum. Dentre as linhagens resistentes, 68 com grãos de boa aceitação comercial foram inoculadas com as raças 73 e 81 de C. lindemuthianum e com a referida mistura de raças de U. appendiculatus. No experimento de campo, foram avaliadas 75 linhagens, que, sob inoculação artificial, mostraram-se resistentes à raça 89 de C. lindemuthianum. As testemunhas, constituídas pelos genitores (Pérola e Ouro Negro), o cultivar Talismã e as linhagens Vi 4899, VC4, e Rudá-Piramidado foram, também, avaliadas em campo. utilizou-se um látice quadrado triplo, com parcelas constituídas de duas linhas com 2m de comprimento cada uma. Na avaliação da eficiência dos marcadores moleculares, foram caracterizadas 150 linhagens do cruzamento Ouro Negro x Pérola, fenotipicamente, por meio de inoculação artificial de C. lindemuthianum (raça 89). Paralelamente, uma planta de cada linhagem foi genotipada, utilizando-se os marcadores SCARF10 1050 e OPX11 550 . A partir da inoculação artificial, foram identificadas 42 linhagens resistentes às raças de C. lindemuthianum e de U. appendiculatus, utilizadas no estudo. Considerando os resultados da caracterização fenotípica e da avaliação de campo, foram identificadas 10 linhagens que apresentaram grãos de bom aspecto comercial, iguais ou superiores àqueles do cultivar Pérola quanto à produtividade de grãos. Estas linhgens mostraram-se resistentes à antracnose e ferrugem, além de, moderadamente resistentes à mancha- angular. Das 150 linhagens avaliadas por meio de inoculação artificial, 61 mostraram-se resistentes às raças 73, 81 e 89 de C. lindemuthianum, sendo que, 42 destas foram resistentes a uma mistura de raças de U. appendiculatus. Vinte e cinco linhagens apresentaram as bandas correspondentes aos marcadores SCARF10 1050 e OPX11 550 . A eficiência de seleção para resistência à antracnose, com base na genotipagem de 150 linhagens utilizando o marcador SCARF10 1050 foi de 78%. Considerando que este marcador encontra-se a uma distância de 19,6 cM do gene de resistência, esperava-se uma eficiência de 72%, portanto, próxima àquela obtida no presente estudo. Para o marcador OPX11 550 , verificou-se uma eficiência de 85%. Esta maior eficiência era esperada, em razão de este marcador localizar-se mais próximo (11,6 cM) dos genes ou do bloco gênico, os quais conferem resistência à antracnose e ferrugem, presentes no cultivar Ouro Negro. Além disso, foi observado que o marcador OPX11 550 também pode ser utilizado no monitoramento do gene de resistência à antracnose. Embora estivessem a uma distância relativamente grande dos genes de resistência, esses marcadores proporcionaram boa eficiência de seleção. The main objective of this study was to phenotypically characterize the common bean lines from the crossing Ouro Negro x Pérola for the reaction to Colletotrichum lindemuthianum (races 73, 81 and 89) and Uromyces appendiculatus (a mixture of races 15, 35, 45, 49, 50 and 59), as well as to evaluate the performance of the lines showing to be resistant ones. Another target was to evaluate the efficiency of the molecular markers SCARF10 1050 (related to the resistance against anthracnose and rust) and OPX11 550 (associated to the resistance against rust), which were identified and mapped in other populations and targeted to the assisted selection in the population Ouro Negro x Pérola. Initially, all 400 lines (progenies F 7:8 ) from the Ouro Negro x Pérola crossing were inoculated with the race 89 of C. lindemuthianum. Sixty eight lines of the resistant ones, from which the common beans are commercially well accepted, were inoculated with the races 73 and 81 from C. lindemuthianum and with the mixture of races from U. appendiculatus. In the field experiment, the evaluations were performed for 75 lines, which under artificial inoculation showed to be resistant to the race 89 of C. lindemuthianum. The controls constituted by genitors (Pérola and Ouro Negro), the Talisman cv. and the lines Vi 4899, VC4, and Rudá-Piramidado were also inoculated. The triple square lattice design was used with the plots consisting into two rows with 2m each one. When evaluating the efficiency of the molecular markers, 150 lines from the crossing Ouro Negro x Pérola were phenotypically characterized, by artificial inoculation of C. lindemuthianum (race 89). At the same time, one plant of each line was genotyped, by using the markers SCARF10 1050 and OPX11 550 . From the artificial inoculation, 42 lines which were resistant to either races of C. lindemuthianum and U. appendiculatus were identified. Taking into account the results of both the phenotypic characterization and field evaluation, ten 10 lines were identified which were provided with commercially good beans, that were equal or superior to the Pérola cv. for bean productivity. These lines showed to be resistant to either anthracnose and rust, and moderately resistant to the angular leaf spot. From those 150 lines evaluated by artificial inoculation, 61 were resistant to the races 73, 81 and 89 of C. lindemuthianum, and 42 lines from these ones showed to be resistant to the mixture of the U. appendiculatus races. Twenty-five lines exhibited the bands corresponding to the markers SCARF10 1050 and OPX 11550 . The selection efficiency for the resistance to anthracnose, based upon genotyping of 150 lines, using the marker SCARF10 1050 was 78%. By considering this marker is at 19.6 cM distance from the resistance gene, an efficiency of 72% was expected, therefore the obtained value was close to the expected one. An efficiency of 85% was found for the marker OPX11 550 . This higher efficiency was expected because this marker is located near (11.6 cM) the genes or the gene block that confers resistance to both anthracnose and rust in Ouro Negro cv. Moreover, it was shown what this marker can be made useful, in the targeted of the resistance gene to anthracnose. These markers provided good selection efficiency, although they were at a relatively long distance from the resistance genes.

Conselho Nacional de Desenvolvimento Científico e Tecnológico O objetivo deste trabalho foi investigar o fluxo gênico, estimado pela taxa de fecundação cruzada entre cultivares de feijão, pertencente ao pool gênico mesoamericano, por meio de um esquema especialmente delineado para esse fim. O experimento foi realizado em duas etapas. A primeira etapa foi a instalação simultânea no campo do cultivar fonte de pólen (Diamante Negro) de flor violeta e do receptor (Talismã) de flor branca. O cultivar fonte foi semeado no centro, e ao seu redor foi semeado, em quadrados concêntricos, o cultivar receptor de pólen. Na maturação foram amostradas fileiras de tamanho fixo em distâncias variadas da fonte; e, em cada direção, essas constituíram as fontes de sementes a serem plantadas na etapa seguinte do experimento. Na segunda etapa do experimento, foram semeadas as sementes amostradas na etapa anterior. No florescimento dessas plantas avaliou-se a porcentagem de fecundação cruzada no feijoeiro, por meio de verificação da cor da flor. As flores violetas indicam ocorrência de cruzamentos naturais. A maior freqüência de híbridos naturais ocorreu à distância da fonte de 0,5 m (0,136%) e 1,0 m (0,11%). Essa freqüência foi reduzida a praticamente 0% a 3,25 m da fonte. The objective of this work was to investigate gene flow, esteem for the rate of out crossed between varieties cultivated of beans, pertaining to gene pool Mesoamerican, by means of a project especially delineated for this purpose. The experiment was accomplished in two stages. The first stage was the simultaneous installation in the field of variety cultivated source of pollen (Diamante Negro) of violet flower and the receiver (Talismã) of white flower. The source was sown in the center and around they had been sown, in concentrical squares, the variety cultivated pollen receiver. In the maturation rows of fixed size had been sampled at varied distance of the source and in each direction, these had constituted the sources of seeds to be planted in the next stage to the experiment. In the second stage of the experiment, had been sown the sampled seeds of the previous stage, and in the flowering of these plants it was evaluated percentage of out crossed in the common bean by means of verification of the color of the flower. The violet flowers indicate occurrence of natural crossings. The biggest frequency of natural hybrids occurred at a distance of the source of 0.5 m (0.136%) and 1.0 m (0.11%). This frequency was reduced practically to 0% at 3.25 m of the source.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Dois ensaios de valor de cultivo e uso (VCU), um com feijão do grupo preto e outro do grupo carioca, foram conduzidos nas safras das “águas”, da “seca” e do “inverno”, nos municípios de Viçosa, Ponte Nova, Coimbra, Leopoldina, Florestal e Capinópolis, em Minas Gerais, nos anos de 2002 e 2003, com o objetivo de avaliar a interação genótipos por ambientes e investigar a eficiência de diferentes métodos para a obtenção de estimativas dos parâmetros adaptabilidade e estabilidade de 20 linhagens avaliadas em 14 ambientes (combinação de safras, anos e locais), bem como a estratificação dos ambientes, com base na produtividade de grãos. As linhagens, de diferentes instituições de origem, incluídas nestes ensaios apresentaram expressiva variabilidade genética, bem como os ambientes de cada ensaio apresentaram variação suficiente para discriminar o potencial destas linhagens, que também apresentaram comportamento diferenciado ao longo dos ambientes em função da ocorrência de interação genótipos por ambientes. Tanto a performance, em relação à produtividade, de algumas linhagens do tipo preto quanto carioca apresentou relação inversa às suas sensibilidades à ferrugem. Os subgrupos formados pela estratificação ambiental, baseada no algoritmo proposto por Lin (1982), apresentaram, em sua maioria, ambientes referentes à safra de inverno, indicando, neste caso, pequeno efeito de locais sobre o comportamento das linhagens nesta safra. A decomposição da interação nas suas frações componentes simples e complexa, pela metodologia de CRUZ e CASTOLDI (1991), não foi eficiente em aprimorar o agrupamento com base no método tradicional de estratificação. A metodologia de estratificação ambiental de MURAKAMI (2001) foi mais eficiente em apontar a similaridade entre os ambientes do que aquela baseada no princípio de interação genótipos x ambientes não significativa, indicando seu potencial neste tipo de estudo. Para a recomendação de cultivares destacou-se o método de CARNEIRO (1998), baseado no trapézio quadrático ponderado pelo CV, em razão da unicidade do parâmetro MAEC, que englobou os conceitos de adaptação, adaptabilidade e estabilidade. Além da recomendação das linhagens para o grupo de ambientes em avaliação, o método permitiu avaliar o comportamento das linhagens em estudo. As linhagens Ouro negro e VI 5700P, de grãos pretos, e OP-NS-331, VI 4899C e VI 0669C, de grãos carioca, apresentaram adaptabilidade geral. Linhagens Valente e VP1, de grãos pretos, e OP-S-82, VC2 e OP-S-16, de grãos carioca, apresentaram adaptabilidade específica às condições favoráveis, ou seja, responsivas à melhoria das condições ambientais. Já para adaptabilidade específica às condições desfavoráveis destacaram-se as linhagens VI 5500P e VP5, de grãos pretos, e VC4, VC5 e VI 4599C, de grãos carioca, sendo, portanto, mais indicadas para os pequenos agricultores. Two studies of cultivation value and use (VCU), using black and carioca beans were carried in different sowing times: "waters", "drought" and "winter", in farm zone of the cities: Viçosa, Ponte Nova, Coimbra, Leopoldina, Florestal and Capinópolis, in Minas Gerais State, during years 2002 and 2003. The objective was evaluate genotypes x environment interaction and investigate the efficiency of different methods to estimate parameters of adaptability and stability of 20 lines evalueted in 14 ambients (combination of sowing times, years and places), as well as environmental stratification, based in grains yield. The lines from different origin, used in these experiments presented genetic variability, as well as the environmental component of each experiments, presented enough variation to discriminate the lines potential. The lines also presented behavior differentiated along the environmental as function of genotypes x environments interaction. Performance related to the productivity, of some lines of black bean as carioca beans presented inverse relationship to their sensibilities to rust. Subgroups formed by the environmental stratification, based on the algorithm proposed by Lin (1982), presented majority ambient regarding the winter sowing dates, indicating, in this case, little effect of places on lines behavior in this harvest. The decomposition of interaction in their components fractions simple and complex, by CRUZ and CASTOLDI (1991), methodology was not efficient to improve grouping based in traditional method of stratification. The methodology of environmental stratification proposed by MURAKAMI (2001) was more efficient to showing similarity among environments than the methodology based in the principle of genotypes x environments interaction no significant, indicating in potential for use in studies like this. For cultivate recommendation stood out the CARNEIRO (1998) methods, based on the quadratic trapezium meditated by CV, in reason of unique parameter MAEC, that included the adaptation, adaptability and stability concepts. Besides, the recommendation of the lines for group of environments evaluates, the method allowed to evaluate the behavior of lines studied. Lines Ouro Negro and VI 5700P, of black grains, and OP-NS-331, VI 4899C and VI 0669C of carioca grains, presented general adaptability. Lines Valente and VP1, of black grains, and OP-S-82, VC2 and OP-S-16, of carioca grains, presented specific adaptability to favorable conditions, in other words, responsiveness to improvement environmental conditions. To specific adaptability in unfavorable conditions pointed out lines VI 5500P and VP5, of black grains, and VC4, VC5 and VI 4599C, of carioca grains, being, therefore, more suitable for smaller farmers.

Conselho Nacional de Desenvolvimento Científico e Tecnológico O feijão é um dos constituintes básicos da dieta do brasileiro, além de ser um produto agrícola de grande valor econômico-social. Neste sentido, para atender as exigências dos consumidores, os programas de melhoramento começam a voltar atenção para características de qualidade do grão. Este trabalho faz parte do programa de melhoramento de qualidade do feijoeiro desenvolvido pelo BIOAGRO/UFV em parceria com a Embrapa - Arroz e Feijão e visa o aumento do teor de proteínas nos grãos do feijoeiro por meio de retrocruzamentos assistidos por marcadores moleculares. Foram utilizadas as variedades Baliza, Laranja, Gaurama e PVA 109 como fontes doadoras de genes para alto teor de proteína e a linhagem CNFC 7827 como genitor recorrente. Para atingir este objetivo as seguintes etapas foram realizadas: (a) realização de cruzamentos entre genótipos de alto teor de proteína com uma linhagem elite; (b) análise do teor de proteína de cerca de 400 sementes segregantes (F 2 ) de cada cruzamento por método não destrutivo; (c) seleção das sementes com maior teor de proteína de cada cruzamento (intensidade de seleção de 10%); (d) realização de fingerprinting das plantas F 2 selecionadas com marcadores RAPD; (e) retrocruzamentos de todas as plantas F 2 selecionadas com o genitor recorrente; (f) confirmação do teor de proteína pelo método Kjeldahl nas sementes F 2:3 ; (g) seleção das plantas RC 1 F 1 com base nos resultados de distância genética determinada com marcadores moleculares e o teor de proteína das plantas F 2 selecionadas e confirmadas na geração F 2:3 . Todas as populações tiveram herdabilidades altas variando de 58 a 77 %, o que sugere que são adequadas para obter alta eficiência de seleção ou estudos de mapeamento para a característica teor de proteína. Os ganhos gerais variaram e - 0,95 a 4,26%, este resultado indica que a seleção em F 2 tem baixa eficiência quando utilizada isoladamente. Entretanto, a ampla variação do teor de proteína em progênies F 2:3 e as porcentagens de ganho observadas (13,66 a 25,84% considerando as cinco progênies de maiores teores) demonstram que o processo de transferência de genes para alto teor protéico por meio de retrocruzamentos é eficiente quando baseado na pré-seleção de sementes F 2 e posterior seleção das melhores famílias F 2:3 . A seleção em geração F 2 com auxílio de micro análise não destrutiva foi pouco eficiente para a população derivada do cruzamento Laranja x CNFC 7827. Fatores como o efeito de dominância e interações genótipo x ambiente diferenciadas causaram dificuldades na seleção dos genótipos superiores. Em vista deste resultado, modificações no método de seleção utilizado podem ser propostas visando aumentar sua eficiência. Os ganhos de seleção para teor de proteína com o auxílio de marcadores moleculares variaram entre 9,92 a 15,65%. As plantas F 2 das diferentes populações apresentaram distância genética em relação ao progenitor recorrente bastante variável. Foi possível obter ganhos de similaridade em relação à média de similaridade encontrada nas populações F 2 variando de 12 a 16%. Com a utilização indivíduos selecionados de alto teor de proteína mais próximos ao genitor recorrente o número de retrocruzamentos necessários para recuperar o genoma do mesmo deve ser menor. The common bean is one of the most important component of the brazilian diet, in addition, is an agricultural product of great economic and social importance. In this sense, to attend the consumers exigencies, the breeding programs start to give attention for quality traits of the grain. This work is part of the quality bean breeding program being developed by BIOAGRO/UFV in partnership with Embrapa - Rice and Bean center and aims to increase the common bean protein content by means of backcrosses assisted by molecular markers. There were used the varieties Baliza, Laranja, Gaurama and PVA as gene donors for high protein content and the CNFC 7827 as recurrent genitor. To achieve this goal, specific steps were reached: (a) crossings high protein content genotypes with an elite cultivar (b) analysis of protein content of 400 F 2 seeds from each cross by using non-destructive method; (c) selection of seeds with high protein content from each cross (selection intensity of 10%); (d) fingerprinting the F 2 selected plants with RAPD markers; (e) backcrosses the selected F 2 plants with the recurrent genitor; (f) confirmation the protein content in the F 2:3 seeds by the Kjeldahl method; (g) selection and planting the RC1F1 based on genetic distances, obtained with molecular markers, and protein content of the F 2 plants selected and confirmed in the F2:3. All the populations had a high heritability varying from 58 to 77%, which suggested that they are adequate for selection or protein mapping loci. The general gains varied from - 0.95 to 4.26%, which indicates that the selection in F 2 has low efficiency when used isolated. However, the wide variation of protein content in F 2:3 and the gains observed (13,66 to 25,84% considering the five F 2:3 of high protein contents) demonstrate that the genes transfer process for high protein content by backcrosses is efficient when based on pre-selection of F 2 seeds and posterior selection of the best F 2:3 families . The selection gains for protein using molecular markers varied from 15.65 to 9.92%. The F 2 plants of the different populations had genetic distances regarding the recurrent genitor highly variable. It was possible to obtain similarity gains, regarding the similarity average found in the F 2 populations, varying from 12 to 16%. With the utilization of selected individuals of high protein content and genetically closer to the recurrent progenitor the number of backcrosses necessary to recover the genome of the recurrent should be smaller.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Este trabalho teve por objetivo estudar as variabilidades espacial e temporal do teor de água de um Argissolo Vermelho-Amarelo irrigado por pivô central, bem como avaliar o efeito do teor de água no solo sobre a produtividade do feijoeiro, em plantios direto e convencional de feijão. Inicialmente, foi feita a caracterização da cobertura vegetal da área sob plantio direto, bem quanto a análise de textura do solo, totalizando 30 e 60 pontos amostrais nos plantios direto e convencional, respectivamente, mediante o uso de malha de 15,00 por 15,00 m. Para a determinação do teor de água no solo, foram feitas amostragens de três em três dias, em uma malha de 10,00 por 10,00 m, com o total de 69 e 136 pontos nos plantios direto e convencional, respectivamente, totalizando 5.330 pontos amostrados em 26 dias. A produtividade do feijão foi avaliada em uma malha de 20,00 por 20,00 m, num total de 13 e 26 pontos nos plantios direto e convencional, respectivamente. As análises das frações granulométricas do solo permitiram caracterizá-lo, nos dois sistemas de plantio, como de textura argilosa. O alcance encontrado foi de 87,60; 80,80; e 87,30 m, em argila, silte e areia, respectivamente, no plantio convencional; e 64,66; 44,40; e 227,70 m, em argila, silte e areia, respectivamente, no plantio direto. Verificou-se que os modelos ajustados e selecionados de semivariogramas, nos dois sistemas de plantio, com exceção da variável areia, no plantio direto, foram os mesmos, evidenciando que a estrutura de variação no espaço é similar em todas as granulometrias, com forte grau de dependência espacial. Para a análise do teor de água no solo, foram consideradas as seguintes fases de desenvolvimento da cultura do feijão: V1 – Emergência, V3 – Primeira folha trifoliolada, R6 – Floração, R8 – Enchimento das vagens e R9 – Maturação fisiológica. O melhor ajuste dos modelos aos semivariogramas foi o esférico, em ambos os sistemas de plantio, à exceção da fase R6, do plantio direto, que apresentou o modelo exponencial. Em uma análise conjunta dos alcances de todas as fases de desenvolvimento da cultura, o alcance variou de 68,50 a 78,30 m no plantio convencional e de 58,20 a 210,90 m no plantio direto, permitindo classificar como forte o grau de dependência espacial. Descartando-se a fase V1, os alcances variaram nas demais fases de 68,50 a 78,30 m no plantio convencional e de 100,00 a 210,90 m no plantio direto, indicando que inferências a respeito dessa variável devem considerar, além do sistema de manejo empregado, a variação espacial desse atributo nas fases de desenvolvimento da cultura. A análise da produtividade apresentou melhor ajuste do modelo ao semivariograma, nos plantios convencional e direto; o modelo esférico, com alcance da ordem de 45,70 e 56,60 m, respectivamente, permitiu classificar como forte o grau de dependência espacial. A correlação entre a produtividade e o teor de água no solo nas fases R6 e R8, nos dois sistemas de plantio, apresentou-se baixa. Em face das metodologias empregadas neste trabalho, os resultados permitiram as seguintes conclusões: a) a dependência espacial das variáveis textura, teor de água no solo e produtividade ocorreu nos dois sistemas de plantio; b) identificou-se uma estabilidade na variabilidade temporal do teor de água no solo, nos dois sistemas de plantio; e c) a correlação entre a produtividade e o teor de água no solo, nos dois sistemas de plantio, foi baixa, verificando-se que a água não foi o fator limitante da produtividade. The objective of this work was to study the space-time variabilities of water content in a red-yellow argissol center pivot irrigated, as well as to evaluate the effect of water content on the soil and yield in conventional and no- tillage bean cultivation. Initially, the vegetable cover characterization of the area in no-tillage system was carried out, as well as the soil texture analysis, totaling 30 and 60 sample locations in no-tillage and conventional systems, respectively, on a 15 m square grid. To determine soil water content, samplings were collected every three days, on a 10 m square grid, yielding a total of 69 and 136 locations in no-tillage and conventional systems, respectively, totaling 5,330 locations sampled in 26 days. Bean yield was evaluated on a 20 m square grid, yielding a total of 13 and 26 locations in no-tillage and conventional systems, respectively. The granular fraction analyses classified the soil as clay in the two cultivating systems. The range found was 87.6; 80.8; and 87.3 m, for clay, silt and sand, respectively, for conventional system and 64.66; 44.4; and 227.7 m, for clay, silt and sand, respectively, for no-tillage system. It was verified that the fitted and selected semivariogram models in the two cultivating systems, except for the variable sand in no-tillage, were the same, showing that the structure of variation in the space is similar in all the granulometries, with high level of spatial dependence. For the soil water content analysis, the following stages of bean crop development were considered: V1 – Emerge \nce, V3 – First trifoliate leaf, R6 – Flowering, R8 – Pod-fill and R9 – Physiological maturation. The best fit to the semivariograms was the spherical model in both cultivating systems, except stage R6 in no-tillage that fitted to a exponential model. A grouped analysis showed that the ranges of all stages of crop development varied from 68.5 to 78.3 m in no-tillage and from 58.2 to 210.9 m in the conventional system, which is classified as high level of spatial dependence. Stage V1 apart, the ranges in the other stages varied from 68.5 to 78.3 m in the conventional system and from 100 to 210,9 m in no-tillage, indicating that inferences regarding this variable should consider, besides cultivation system, the spatial variation of this attribute in the stages of development. Yield analysis gave the spherical model the best fit to the semivariogram in the conventional and no-tillage systems; reaching 45.7 and 56.6 m, respectively, classifying spatial dependence as high level. The correlation between yield and soil water content in stages R6 and R8 was low in the two systems. The results obtained permitted the following conclusions: a) spatial dependence of variables texture, soil water content and yield occurred in both systems; b) it was verified a stability in time variability of soil water content in the two systems; c) and correlation between yield and soil water content was low in the two systems, showing that the water was not the yield limiting factor.

Conselho Nacional de Desenvolvimento Científico e Tecnológico O cultivar de feijão preto Diamante Negro além da boa aceitação comercial, apresenta resistência ao crestamento-bacteriano-comum e ao mosaico comum. No Programa de Melhoramento do Feijoeiro do BIOAGRO/UFV, foi produzida uma isolinha de grãos tipo carioca (Rudá “R”) contendo simultaneamente genes de resistência à Colletotrichum lindemuthianum (genes Co-6 e Co-4), à Uromyces appendiculatus (genes Ur-?) e à Phaeoisariopsis griseola (gene Phg –1), com o auxílio de marcadores moleculares ligados a cada um desses genes. Os objetivos deste trabalho foram: caracterizar o cultivar Diamante Negro quanto à reação às principais raças de C. lindemuthianum (antracnose), U. appendiculatus (ferrugem) e P. griseola (mancha-angular) e também quanto ao seu fingerprint molecular por meio de marcadores do DNA, e transferir genes de resistência já piramidados na isolinha Rudá “R”, para esse cultivar. O Diamante Negro foi inoculado com 10 patótipos de C. lindemuthianum, 10 de U. appendiculatus e seis de P. griseola. No resultado da avaliação, o Diamante Negro mostrou-se suscetível a três patótipos de C. lindemuthianum (65, 81 e 89) e resistente, mas segregante, a quatro (55, 87, 95 e 342). Com relação a U. appendiculatus, comportou-se como suscetível a todos os patótipos. Com relação a P. griseola, foi suscetivel a quatro patótipos (31.17, 63.19, 63.23 e 63.55). Para sua caracterização molecular frente a outros cultivares do tipo preto e ao cultivar Rudá, pela técnica de RAPD, foram utilizados 78 primers, gerando 146 bandas polimórficas que permitiram o cálculo das distâncias genéticas e análise de agrupamento. A maior distância observada em relação ao Diamante Negro foi com o cultivar andino Preto 60 dias (26,79%), a menor com o Ouro Negro (6,42%) e o Rudá mostrou-se a uma distância de 28,11%. Três grupos foram formados: um contendo o Preto 60 dias (andino), um contendo o Rudá (mesoamericano-carioca) e um contendo os demais cultivares (mesoamericanos-pretos). Para a introgressão dos genes de resistência, cruzamentos entre Diamante Negro e o Rudá “R” foram realizados e as plantas F 1 foram retrocruzadas com o Diamante Negro gerando 394 plantas. Com inoculações seriadas com os três patógenos, foi possível selecionar 32 plantas resistentes às três doenças. O DNA destas 32 plantas foi amplificado com os marcadores SCAR Y20 830a (Co-4), SCAR H13 490a (Phg-1), SCAR AZ20 940a (Co-6) e SCAR F10 1050a e SCAR BA08 560a (Ur-?). Com o resultado desta amplificação foi possível selecionar 21 plantas que possuíam pelo menos quatro marcas, sendo que quatro destas plantas (75, 93, 141 e 270) apresentaram as cinco marcas. As distâncias genéticas relativas entre estas plantas RC 1 F 1 e o Diamante Negro variaram de 37 a 65%. Todas as 21 plantas RC 1 F 1 foram conduzidas para o segundo retrocruzamento, gerando 231 plantas RC 2 F 1 . A amplificação do DNA destas plantas RC 2 F 1 com os marcadores SCAR, permitiu selecionar 18 plantas com pelo menos quatro marcas. Seis destas (75.8, 93.8, 141.2, 141.4, 141.6 e 141.7) apresentaram as cinco marcas. As distâncias genéticas relativas entre as plantas selecionadas e o Diamante Negro foram de 32 a 48%. As 18 plantas selecionadas foram usadas para o terceiro retrocruzamento. O objetivo final deste trabalho é o de selecionar linhagens de grãos pretos com resistência à antracnose, ferrugem, mancha-angular, crestamento- bacteriano-comum e mosaico comum, com bom potencial produtivo. Diamante Negro, a black-seeded common bean cultivar, is not only outstanding because of its excellent commercial acceptability, but also because it is resistant to common bean leaf blight and to common bean mosaic virus. Using molecular marker assisted selection, the common bean breeding program of the Universidade Federal de Viçosa (BIOAGRO) developed an isoline with carioca type grains (Rudá “R”) containing genes for resistance to Colletotrichum lindemuthianum (genes Co-6 and Co-4), to Uromyces appendiculatus (genes Ur-?), and to Phaeoisariopsis griseola (gene Phg –1). The objectives of this study were: a) to characterize the cultivar Diamante Negro in relation to its reaction to the main C. lindemuthianum (anthracnose), U. appendiculatus (rust), and P. griseola (angular leaf spot) races; b) to determine its molecular fingerprint using DNA markers; and c) to transfer the resistance genes present in Rudá “R” to this cultivar. Diamante Negro was inoculated with 10 pathotypes of C. lindemuthianum, 10 of U. appendiculatus, and six of P. griseola. Diamante Negro was susceptible to three pathotypes of C. lindemuthianum (65, 81, and 89) and resistant, though segregant, to four other (55, 87, 95, and 342). The cultivar was susceptible to all pathotypes of U. appendiculatus and to four pathotypes of P. griseola (31.17, 63.19, 63.23, and 63.55). The genetic distances between Diamante Negro and xother black-seeded cultivars and cultivar Rudá were determined. Seventy- eight RAPD primers produced 146 polymorphic bands. Andean cultivar Preto 60 dias presented the greatest distance in relation to Diamante Negro (26.79%), Ouro Negro, the shortest distance (6.42%), and Rudá a distance of 28.11%. Based on the genetic distances, three groups were formed: one containing Preto 60 dias (andean), the other containing Rudá (“carioca-type”, mesoamerican) and the last one including all the other cultivars (black-seeded, mesoamerican). For the introgression of the resistance genes, the F 1 plants of the cross Diamante Negro x Rudá “R” were backcrossed with Diamante Negro, producing 394 plants. By serial inoculations with the three pathogens, 32 plants resistant to the three diseases were selected. The DNA of these 32 plants was amplified with the markers 1), and SCAR SCAR Y20 830a (Co-4), F10 1050a and SCAR SCAR AZ20 940a (Co-6), SCAR H13 490a (Phg- BA08 560a (Ur-?). Twenty-one plants were selected harboring at least four markers of the markers. Four of these plants presented all five markers. The relative genetic distances between these RC 1 F 1 plants and Diamante Negro varied from 37 to 65%. All 21 RC 1 F 1 plants were included in the second backcrossing, giving rise to 231 RC 2 F 1 plants. The DNA amplification of these plants with the SCAR markers led to the selection of 18 plants with at least four markers. Six among them presented the five markers. Relative genetic distances between the selected plants and Diamante Negro ranged between 32 and 48%. The 18 selected plants were used for a third backcross. The ultimate goal of this work is to select black-seeded lines with resistance to anthracnose, rust, angular lea spot, common leaf blight and common mosaic with a promising yield potential.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior O sucesso na condução de um programa de melhoramento de plantas autógamas é dependente de decisões acertadas nas suas etapas iniciais. Um questionamento freqüente é como os genitores deverão ser cruzados para obtenção das populações segregantes. Visto que, na cultura do feijoeiro, existem poucos estudos a esse respeito, o presente estudo teve como objetivos avaliar o comportamento de populações segregantes de feijoeiro oriundas de híbridos simples e duplos, predizer o potencial dessas populações com vista à extração de linhagens com produtividade superior a cultivar Valente e avaliar o efeito da interação genótipos x safras na predição do potencial genético de populações segregantes de feijão. Assim, foram cruzados 40 genitores obtendo-se, inicialmente, 20 híbridos simples, posteriormente, estes foram combinados num esquema de dialelo circulante obtendo-se, 20 híbridos duplos. Os experimentos foram conduzidos na estação experimental de Coimbra/UFV, Minas Gerais. As 40 populações segregantes oram avaliadas nas gerações F 4 , F 5 e F 6 , juntamente com nove testemunhas, nas safras da seca, do inverno e das águas, respectivamente, visando estimar, entre outros efeitos, o efeito da interação populações segregantes safras. O delineamento experimental utilizado foi o látice triplo 7 x 7 com parcelas de quatro linhas de quatro metros. Utilizando dados de produtividade de grãos (gramas/planta) e a variância dentro das populações foi estimada a probabilidade de uma dada população produzir linhagens que supere, em 20%, a produtividade da cultivar Valente segundo a metodologia de JINKS & POONI (1976). Tanto com base nas produtividades de grãos (kg/ha) como na predição do potencial das populações em gerar linhagens promissoras não foram observadas diferenças entre as populações oriundas de híbridos simples e aquelas populações oriundas de híbridos duplos, indicando que o número de genitores envolvidos no cruzamento não foi determinante no desempenho final das populações segregantes. Foi constatado efeito significativo da interação populações segregantes x safras, cuja natureza foi predominantemente complexa, advertindo sobre a necessidade de condução e avaliação das populações segregantes nas diferentes safras da cultura do feijoeiro. The success in conducting a breeding program of self-pollinated plants depends on the decisions made in its initial steps. A frequent question is how the parents will have to be crossed in order to obtain segregant populations. Since, in common bean, few works exist that can help this decision, this study had as goals to evaluate the performance of segregant populations derived from single-cross and double-cross hybrids, to predict the potencial of that populations for lines extraction with surpass grain yield of Valente cultivar and evalute the effect of segregant populations x sowing dates interaction in the prediction of the genetic potencial of common bean segregant populations. Thus, 40 parents were crossed obtaining 20 single-cross hybrids. These hybrids were intercrossed according to the cycle diallel procedure obtaining, in that way, 20 double-cross hybrids. The experiment was conducted in Coimbra/ UFV experimental station, Minas Gerais. The populations were evaluated in the F 4 , F 5 and F 6 generation, with nine checks, the sowing dates being in dry, winter and wet seasons of 2002, respectively, aiming to study the effects of the interaction of segregant populations x sowing dates. The experimental design used was 7 x 7 triple lattice with plots of four lines, 4m long. Grain yield data (grams/plant) and genetic variance within the populations were used to estimate the probability of a given population to produce lines that could surpass grain yield of Valente cultivar by 20 percent, according to JINKS & POONI (1976) methodology. Based in average grain yield (Kg/ha) and also in the prediction of the populations potential of producing promising lines, any difference between the populations from single-cross hybrids and double-cross hybrids wasn t observed, indicating that the number of parents involved in the crossing to obtain the segregant populations wasn t determinant in the final performance of the segregant populations. The segregant populations x sowing dates interaction was significative, its nature was predominantly complex, warning about the necessity of the conduction and evaluation of segregant populations in the different sowing dates of the common bean culture.
