Diagnosis, prognostic assessment, and monitoring disease activity in patients with large vessel vasculitis (LVV) can be challenging. Early recognition of LVV and treatment adaptation is essential because vascular complications (aneurysm, dilatations, ischemic complications) or treatment related side effects can occur frequently in these patients. 18-fluorodeoxyglucose positron emission tomography/computed tomography (2-[18F]FDG-PET/CT) is increasingly used to diagnose, follow, and evaluate treatment response in LVV. In this review, we aimed to summarize the current evidence on the value of 2-[18F]FDG-PET/CT for diagnosis, follow, and treatment monitoring in LVV.

IntroductionSex differences in cognition of HIV positive (HIV) patients are controversial. We aimed to investigate the relationship between cognition, HIV status, and sex, in a highly homogenous cohort of young Romanians parenterally infected during early childhood.MethodsIn total, 250 HIV participants were compared with age-matched HIV negative (HIV) controls (n = 72) in a cross-sectional study. After standardized neurocognitive, psychological testing and medical evaluation, linear regression was used to assess the effect of sex and HIV on neurocognitive outcomes.ResultsStudy participants were on average 23 years old with balanced sex distribution (% women = 52% vs. 43%). In this article, I am detailing an application of White’s perspective about the ‘conjunction’ between: (a) logical sentences, (b) moral sentences and (c) descriptive sentences. I examine several options from the main storyline of the role-playing video game Cyberpunk 2077. I am approaching the video game’s content from the standpoint of Morton White’s holistic pragmatism (i.e. ‘corporatism’). I assert that a ‘narrow’interpretation is necessary, to admit a moral objection.

Given paper is presented as a theoretical-applied study that falls within the scientific dimensions of the theory and methodology of didactic evaluation. The author uses descriptive and analytical research methods; comes with the literature review relevant to the stated topic; describes a formative assessment tool based on applying criteria and descriptors that could be implemented in the educational activities specific to the seminar classes within the university study programs with specializations in the field of Computer Science and ICT, but not only. The research's aim is to provide the university teachers and, not only, some methodological guidelines that would support them in developing and implementing a personal, descriptive tool for measuring the activity of the learner in the stated didactical circumstances.

In 2015– 2017 the Prut-Dniester expedition from the Institute for the History of Material Culture of the Russian Academy of Sciences carried out field research at a number of Paleolithic localities in the middle reaches of the Prut and Dniester in the north-west of Moldova. This paper presents the results of new research at the Middle Paleolithic open air site of Teţcani X and the cave site of Buzdujeni I. The cultural layer of Teţcani X yielded a representative collection of flint artifacts. The flaking technology is oriented on the production of points, which were struck from unifacial single platform cores. The tool set is typical of Mousterian, no bifaces are present. The cave of Buzdujeni I was nearly completely excavated by N. A. Chetraru in the 1970’ s, but remained largely unpublished. The re-analysis of the old collection together with the materials obtained by us in 2017 challenges the once popular idea that the industry of Buzdujeni I belongs to Denticulate Mousterian, highlighting instead its Micoquian affinities such as presence of various bifacial tools. The available biostratigraphic and radiometric data, including newly obtained radiocarbon dates, appears to suggest that both sites can be broadly dated to the first half of MIS-3 or to MIS-4, with Teţcani X being probably somewhat older than Buzdujeni I

Mesenchymal stromal cells (MSC) are nonhematopoietic cells with fibroblast-like morphology and multipotent capacity that are widely used in pre-clinical and clinical investigations. Unfortunately, the efficiency of MSC treatment is hindered by the poor survival rate after transplantation at the damaged tissue. The goal of this study was to investigate the fate of MSC exposed to various stimuli mimicking the in vivo microenvironment post transplantation. To this aim, murine bone marrow–derived MSC were stimulated with IFNgama and TNFalpha under low oxygen (hypoxia) or atmospheric (normoxia) conditions for 24 to 72 hours, in order to better mimic an ischemic injury. The results showed that MSC pre-stimulation with TNFalpha and IFNgama enhanced immunosuppressive pathways by over-expression of NOS2, IDO, COX2 and production of NO. However, MSC viability was affected by these two cytokines in dose-dependent and time-dependent manners. Besides, priming with TNFalpha and/or IFNgama under low oxygen concentrations revealed that significantly increased cell mortality rate and decreased NO production. Our data suggest that both hypoxia and inflammation could impact the cell survival after transplantation and reinforces the necessity of further investigations to better understand MSC behavior after transplantation in order to identify the MSC-based strategies with the highest therapeutic potential.

Sleepiness, fatigue, and stress in drivers are the leading causes of car crashes. In the late two decades, there is an endeavor to monitor vital signs, stress levels, and fatigue using adapted sensors supported by technological advances. To the best of our knowledge, this systematic review is the first to investigate the role of HRV measurement for sleepiness, fatigue, and stress level monitoring in car drivers. A search was performed in PubMed, Embase, and Cochrane databases using prespecified keywords. Studies were considered for inclusion if they reported original data regarding the association between different HRV measurements and drivers’ sleepiness, fatigue, or stress levels. Of the retrieved 749 citations, 19 studies were finally included. The sensibility and specificity of HRV significantly varied across studies, respectively 47.1%–95% and 74.6%–98%. Accuracy was also different, ranging from 56.6% to 95%. Nevertheless, in real-world conditions, confounding factors could affect sympathovagal tone and HRV. Multiple HRV parameters measurement rather than one parameter approach seems to be the optimal strategy for evaluating the vigilance state in drivers that it would be possible to achieve a good performance. As all studies were observational, data should be confirmed in randomized controlled trials. In conclusion, HRV represents a potentially valuable marker for sleepiness, fatigue, and stress monitoring in car drivers. HRV measurements could be implemented in future clinical models and sensors to detect early sleepiness and fatigue and prevent car crashes. More studies with larger populations are needed to support this evidence.

Industrial mixers are equipment used to mechanically combine different types of materials in order to obtain homogeneous mixtures. In concrete industry production, mixers play a crucial role by facilitating the efficient and consistent blending of various constituents to create high-quality concrete. Because the mixers in the concrete industry work in conditions characterized by abrasive and erosive loadings, the authors of this paper tried to establish a dependence between the quality of the material from which the mixing elements are made and their wear resistance. Three types of cast irons alloyed with chromium, specific to the construction of mixing blades, were used in this research. The working environment was a mixture of crushed mineral aggregates corresponding to the granulometric class 4–8 mm. The tests were carried out on an experimental stand designed and built by the authors of this paper. The stand reproduces on a scale of 1:2 a drum made up of a double-axis horizontal mixer. The stand had the possibility to change the value of the attack angle of the mixing blades, corresponding to the following values: 30, 45, and 60 degrees. The results of the tests established the dependence between the type of material and the wear rate of the blades as well as the influence exerted by the angle of attack on the wear of the mixing blades. It was shown that when the inclination angle of the blade relative to the shaft axis increases, the cumulative mass loss decreases, with values between 43% and 55.83%, as a function of the quality of blade material.

Extremely low-frequency electromagnetic stimulation (ELF-EMS) was demonstrated to be significantly beneficial in rodent models of permanent stroke. The mechanism involved enhanced cerebrovascular perfusion and endothelial cell nitric oxide production. However, the possible effect on the neuroinflammatory response and its efficacy in reperfusion stroke models remains unclear. To evaluate ELF-EMS effectiveness and possible immunomodulatory response, we studied neurological outcome, behavior, neuronal survival, and glial reactivity in a rodent model of global transient stroke treated with 13.5 mT/60 Hz. Next, we studied microglial cells migration and, in organotypic hippocampal brain slices, we assessed neuronal survival and microglia reactivity. ELF-EMS improved the neurological score and behavior in the ischemia-reperfusion model. It also improved neuronal survival and decreased glia reactivity in the hippocampus, with microglia showing the first signs of treatment effect. In vitro ELF-EMS decreased (Lipopolysaccharide) LPS and ATP-induced microglia migration in both scratch and transwell assay. Additionally, in hippocampal brain slices, reduced microglial reactivity, improved neuronal survival, and modulation of inflammation-related markers was observed. Our study is the first to show that an EMF treatment has a direct impact on microglial migration. Furthermore, ELF-EMS has beneficial effects in an ischemia/reperfusion model, which indicates that this treatment has clinical potential as a new treatment against ischemic stroke.

Topical antiseptics are essential in wound treatment, and adipose-derived stem cells (ADSCs) have recently been proven to facilitate healing. However, the impact of antiseptics on ADSCs has not been fully elucidated, especially in relation to other relevant cell types present in the wound microenvironment, e.g., fibroblasts. This study evaluated the effects of chlorhexidine and povidone–iodine on four cellular constructs in 2D and 3D in vitro culture systems. Cell constructs were treated with two concentrations of each antiseptic, after which cell migration activity, α-SMA, and Ki67 marker expressions were assessed and compared. Both tested concentrations of povidone–iodine impaired migration and sprouting compared to chlorhexidine, which had minimal effects when used in low concentrations. The gap in the wound healing assay did not close after 24 h of povidone–iodine treatment, although, at the lower concentration, cells started to migrate in a single-cell movement pattern. Similarly, in 3D culture systems, sprouting with reduced spike formation was observed at high povidone–iodine concentrations. Both antiseptics modulated α-SMA and Ki67 marker expressions at 5 days following treatment. Although both antiseptics had cytotoxic effects dependent on drug concentration and cell type, povidone–iodine contributed more substantially to the healing process than chlorhexidine, acting especially on fibroblasts.

The most recent assessment (2008) of the IUCN Red List of Threatened Species classified Romanichthys valsanicola as critically endangered (CR). In December 2022, an expert team from the National Institute for Research and Development in Environmental Protection Bucharest investigated the presence of the species in historical locations and in other potential sites. The authors correlated public data to the actual habitat area to calculate the potential species density in relation to the specific territorial behavior. The ecological carrying capacity was represented by the consensus between the behavioral density limitations and the trophic limitations of the actual species potential habitat. Both trophic availability and populational density present encouragingly high values for the sculpin perch in the Valsan River, indicating that the natural habitat could host a considerably higher number of individuals.

This study aimed to compare the outcomes and cost differences between primary lung cancer (PLC) and second primary lung cancer (SPLC) patients who underwent video-assisted thoracoscopic surgery (VATS). This was a retrospective analysis of 124 patients with lung cancer stages I, II, and III who underwent VATS between January 2018 and January 2023. The patients were divided into two groups based on their cancer status that was matched by age and gender: the PLC group (n = 62) and the SPLC group (n = 62). The results showed that there was no significant difference in the clinical characteristics between the 2 groups, except for the Charlson Comorbidity Index (CCI), with a score above 3 in 62.9% of PLC patients and 80.6% among SPLC patients (p = 0.028). Regarding the surgical outcomes, the operative time for the VATS intervention was significantly higher in the SPLC group, with a median of 300 min, compared with 260 min in the PLC group (p = 0.001), varying by the cancer staging as well. The average duration of hospitalization was significantly longer before and after surgery among patients with SPLC (6.1 days after surgery), compared with 4.2 days after surgery in the PLC group (0.006). Regarding the cost analysis, the total hospitalization cost was significantly higher in the SPLC group (15,400 RON vs. 12,800 RON; p = 0.007). Lastly, there was a significant difference in the survival probability between the two patient groups (log-rank p-value = 0.038). The 2-year survival was 41.9% among PLC patients and only 24.2% among those with SPLC. At the 5-year follow-up, there were only 1.6% survivors in the SPLC group, compared with 11.3% in the PLC group (p-value = 0.028). In conclusion, this study found that VATS is a safe and effective surgical approach for both PLC and SPLC patients. However, SPLC patients have a higher VATS operating time and require more healthcare resources than PLC patients, resulting in higher hospitalization costs. These findings suggest that careful pre-operative evaluation and individualized surgical planning are necessary to optimize the outcomes and cost-effectiveness of VATS for lung cancer patients. Nevertheless, the 5-year survival remains very low and concerning.

This article outlines a capacity review of Romania’s national road infrastructure and road safety in general. Romania’s road fatality rate per 100,000 population has improved overall from a 2008 high of around 15 to the current 2019 value of 9.6. However, the rate has flat-lined with no real improvement for the last decade, stalling at around 9.7 over the period 2011- 2019 and around double the EU rate. Moreover, Romania’s total annual number of road deaths has remained at an average of around 1900 fatalities per annum over this period. Romania has been the worst performing country in the European Union (EU) in recent years, and one of the worst performing countries compared to Organisation for Economic Co-operation and Development (OECD) nations in terms of road safety. The review performed in 2016 found inadequate political leadership and commitment to effective actions to reduce road fatalities, fragmented government road safety activities across a number of regulatory entities, speed limits set at levels that exceed internationally accepted survivable limits, weak traffic law enforcement including a lack of speed enforcement cameras resulting in a failure of drivers to comply with speed limits, and a lack of structured programs to implement human error tolerant road infrastructure constructed according to Safe System principles. A series of recommendations from the capacity review were adopted (as described here) since 2016, although much remains to improve road safety in Romania.

Abstract Mesenchymal stromal cells (MSC) are attractive tools for cell-based therapy, yet the mechanisms underlying their migration and survival post-transplantation are unclear. Accumulating evidence indicates that MSC apoptosis modulates both innate and adaptive immune responses which impact on MSC therapeutic effects. Using a dual tracking system, namely the Luciferase expression and VivoTrack680 labelling, and in vivo optical imaging, we investigated the survival and migration of MSC transplanted by various routes (intravenous, subcutaneous, intrapancreatic and intrasplenic) in order to identify the best delivery approach that provides an accumulation of therapeutic cells to the injured pancreas in the non-obese diabetic (NOD) mouse. The results showed that transplanted MSC had limited migration capacity, irrespective of the administration route, and were short-lived with almost total disappearance at 7 days after transplantation. Within one day after transplantation, cells activated hypoxia signalling pathways, followed by Caspase 3-mediated apoptosis. These were subsequently followed by local recruitment of immune cells at the transplantation site, and the engulfment of apoptotic MSC by macrophages. Our results argue for a “hit and die” mechanism of transplanted MSC. Further investigations will elucidate the molecular crosstalk between the inoculated and the host-immune cells.

Abstract Background Mesenchymal stem/stromal cells (MSC) represent adult cells with multipotent capacity. Besides their capacity to differentiate into multiple lineages in vitro and in vivo, increasing evidence points towards the immunomodulatory capacity of these cells, as an important feature for their therapeutic power. Although not included in the minimal criteria established by the International Society for Cellular Therapy as a defining MSC attribute, demonstration of the immunomodulatory capacity of MSC can be useful for the characterization of these cells before being considered MSC. Methods Here we present a simple and reliable protocol by which the immunosuppressive effect of mouse bone marrow-derived MSC can be evaluated in vitro. It is based on the measuring of the proliferation of activated T cells cultured in direct contact with irradiated MSC. Results Our results showed that mouse MSC have a dose-dependent inhibitory effect on activated T cell proliferation, which can be quantified as a percentage of maximum proliferation. Our data shows that batch-to-batch variability can be determined within one or multiple experiments, by extracting the area under curve of T cell proliferation plotted against the absolute number of MSC in co-culture. Conclusions The validation of the immunosupressive capacity of MSC could be added to the characterization of the cells before being used in various MSC-based approaches to treat immunological diseases. Our results showed that mouse MSC have a dose-dependent inhibitory effect on activated T cell proliferation. The immunosuppressive properties of MSC vary between batches, but not between different passages of the same batch.

Cryptocurrencies exist in databases with special rules that condition how they are offered to different people. Cryptocurrencies are decentralized and managed by a computer network, not by a particular company. They arose from the need to use an alternative currency, which does not require the presence of a managing authority, which operates beyond the political climate and which is easy to transfer.

Left ventricular (LV) aneurysm following acute myocardial infarction (MI) represents a less common complication, but with worse clinical outcomes. Ventricular surgical reconstruction is not always the intervention of choice due to high surgical risk. There were proposed less invasive LV aneurysm exclusion techniques such as the less invasive ventricular enhancement (LIVE) procedure. Our paper represents the first systematic approach to investigate the efficacy and safety of LIVE procedure using Revivent TCTM anchor system for LV aneurysm exclusion. Studies were considered if they reported original data regarding LIVE procedure’s efficacy and/or safety using the Revivent TCTM system in patients with LV aneurysms. Five studies met the inclusion criteria. The procedure is associated with a reduction in LV volumes and an improvement in LV ejection fraction (LVEF). The means of preoperative LVEF varied between 22.8% and 35.6%, while postoperative LVEF means ranged between 34% and 45.9% (P < 0.005) across studies. All included papers reported a significant difference between preoperative and postoperative LV end-systolic volume index (P ≤ 0.001) and LV end-diastolic volume index (P ≤ 0.001). Three out of four studies achieved statistical significance (P ≤ 0.001) when comparing preoperative (means range: 2.6–3.4) and postoperative (means range: 1.4–1.9) New York Heart Association (NYHA) class. One study reported a survival rate of 90.6 (95% CI, 84.6–97.0) at 12 months following the procedure. LIVE appears to be a promising and appropriate treatment strategy for a complex condition, which could extend the indication of LV aneurysm exclusion in the future.

Generalized arterial calcification of infancy (GACI) is an extremely rare autosomal recessive condition characterized by the storage of calcium at the level of internal elastic membrane of arteries. The main consequences are intimal fibrous thickening and arterial occlusion. We present the case of a preterm male infant, born from an improperly dispensed pregnancy. At birth, the newborn presented generalized edema and hypotonia, and abolished heart sounds, without response to stimulation. Despite the mechanical ventilation, the infant died 2 h after birth. The death was clinically presumed to be related to the maternal infection with cytomegalovirus (CMV) and Toxoplasma gondii. The infant's mother affirmed the history of 6 previous miscarriages and a non-consanguineous marriage. At autopsy, microscopic examination showed generalized vasculitis secondary to minimal calcification of the large and medium-sized vessels of the lungs, liver, and tongue. These findings supported the diagnosis of GACI. Hydrothorax, non-infective ascites, and necrosis of the brain parenchyma were also associated. The premature infant died due to tonsillar herniation associated with decreased vessel compliance and refractory pulmonary hypertension thus leading to congestive cardiac failure. CMV was not detected on histopathological assessment nor were signs of any other infections. To the best of our knowledge, this is the first case of GACI occurring in a baby from a mother co-infected with CMV and T. gondii.

Cerebral ischemia reperfusion injury is a debilitating medical condition, currently with only a limited amount of therapies aimed at protecting the cerebral parenchyma. Micro RNAs (miRNAs) are small, non-coding RNA molecules that via the RNA-induced silencing complex either degrade or prevent target messenger RNAs from being translated and thus, can modulate the synthesis of target proteins. In the neurological field, miRNAs have been evaluated as potential regulators in brain development processes and pathological events. Following ischemic hypoxic stress, the cellular and molecular events initiated dysregulate different miRNAs, responsible for long-terming progression and extension of neuronal damage. Because of their ability to regulate the synthesis of target proteins, miRNAs emerge as a possible therapeutic strategy in limiting the neuronal damage following a cerebral ischemic event. This review aims to summarize the recent literature evidence of the miRNAs involved in signaling and modulating cerebral ischemia-reperfusion injuries, thus pointing their potential in limiting neuronal damage and repair mechanisms. An in-depth overview of the molecular pathways involved in ischemia reperfusion injury and the involvement of specific miRNAs, could provide future perspectives in the development of neuroprotective agents targeting these specific miRNAs.

Introduction. Leptospirosis is a zoonosis caused by microorganisms of the genus Leptospira with a global spread. The main reservoir of leptospires are the small mammals, which survive after infection, spreading the causative agent in the environment with urine. Material and methods. The studied were conducted in 2017-2019 in different ecosystems, where 1617 small mammals were collected, investigated for the presence of specific antibodies to Leptospira spp. Ecological analysis of small mammal communities was performed and leptospire-carrier species were identified. Results. 17 species of the order Rodentia and Soricomorpha were identified. The most widespread are A. sylvaticus, A. flavicollis and A. agrarius with a frequency of 100%, abundant and dominant in most of the studied ecosystems. Antibodies specific to Leptospira spp. were detected in 9 species, the highest share of the species with leptospire belongs to A. agrarius (53.85%) and C. glareolus (11.54%). The serogroups Leptospira grippotyphosa, L. icterohaemorrhagiae and L. pomona were highlighted. Conclusions. The epizootiological process in leptospirosis is maintained active in nature by 9 species of small mammals, including one of the Europe species A. agrarius, the leptospire rate was determined at 53.85%. The intense circulation of leptospires in the small mammal population determines the need to monitor the multiannual dynamics of their in order to establish changes in natural foci of leptospirosis, forecast the epizootological situation and the risk of spreading of disease among the human population.

Unemployment is considered as a negative element of economic development that affects in varying proportions all countries, especially those underdeveloped and those in transitions to the market economy. This paper presents the "Economic and Social Effects of Unemployment in Romania", where we presented the unemployment in the contemporary world, its typology and its measurement, namely: the evolution of the number of unemployed, a rate of unemployment and its structure in Romania. Unemployment has become a problem, along with industrial development, since the second half of the 18th century, in times of recession, when industrial enterprises shrank their production and therefore released a large number of workers who became unemployed. In Romania, unemployment is partly due to the transformation of the structure of a national economy, according to the criterion of efficiency, in order to adapt to the competitive environment.
