Structural malformation of the oviduct has been reported in experimental animal models and women following prenatal exposure to diethylstilbestrol (DES). To study histological changes in the oviduct in the absence of gross structural malformation, neonatal CD-1 mice were treated with DES (2 micrograms/pup/day) on days 1-5 of age. Focal epithelial hyperplasia was present at 1 month of age in 16 out of 18 (89%) of the DES-treated mice. At 4 months of age, general epithelial hyperplasia with multiple gland-like structures into and through the muscle wall of the oviduct was observed in 90% of the treated mice; by 12 months of age, epithelial hyperplasia and pseudogland formation were seen in 100% of the DES-exposed animals. Epithelial hyperplasia and gland formation were not observed in control mice. The alteration induced by DES in the differentiation and proliferation of mouse oviductal epithelium suggests that the oviduct is a target for DES toxic effects. In addition, there was a progression of the epithelial changes with age. The histological changes described in this study may be partially responsible for the decreased fertility previously reported in this mouse model. Similar changes in the oviduct of DES-exposed women remain to be determined.

A severe and highly fatal disease occurred in African green monkeys (Cercopithecus aethiops). The disease was marked by an extensive maculo-vesicular rash. In fatal cases, multifocal necrosis and hemorrhage were found characteristically in many organs and tissues. New cases of the disease occurred over a 39-day period. Nine of 49 exposed African green monkeys were affected and five of the nine died. Four cynomolgus (Macaca fascicularis) and 31 stumptailed monkeys (Macaca arctoides) which were exposed were not affected. Delta Herposvirus was isolated from affected African green monkeys. Stumptailed monkeys housed in association with the African green monkeys, as well as those housed elsewhere, had significant serum concentrations of neutralizing antibodies to this agent.

Prenatal exposure of males to diethylstilbestrol (DES) results in reproductive tract teratogenesis, ie, retention of Müllerian duct remnants. The potential of these remnants to develop pathological changes has not been studied. Therefore, pregnant outbred CD-1 mice were subcutaneously injected with daily doses of DES (100 micrograms/kg) on days 9 through 16 of gestation. DES-exposed male offspring and age-matched control male mice were sacrificed at 10 to 18 mo of age and examined for reproductive tract abnormalities. Prominent Müllerian remnants were observed in 268 out of 277 (97%) of the DES-exposed male mice. These remnants differentiated into "femalelike structures" homologous to oviduct and uterus. The Müllerian remnants were often enlarged and cystic and shared supporting connective tissue with adjacent male structures. Previously reported lesions, termed "epididymal cysts," were determined histologically to be cystic "oviductlike" structures and were, therefore, considered a Müllerian duct abnormality. Pathological changes in these male oviductal and uterine homologs included benign and malignant lesions. In addition, epididymal structures were altered. Inflammation and sperm granulomas were prevalent in DES-treated mice as young as 10 mo old but were only observed in control mice at 18 mos. Cysts of epididymal duct origin, hyperplasia, and adenoma of the epididymal duct were also observed. No comparable abnormalities were noted in 122 control males of corresponding ages. The data presented in this report demonstrated that transplacental exposure to DES affected the differentiation and normal development of the male genital tract involving both the Müllerian (paramesonephric) and Wolffian (mesonephric) ducts. The long-term changes in these tissues include lesions, some of which resembled neoplasia although the natural history of the lesions is not known. Moreover, some previously described abnormalities referred to as "epididymal cysts" were associated with tissues derived from embryonic female origin.

Treatment of pregnant women with diethylstilbestrol (DES) is associated with the subsequent development of reproductive tract abnormalities such as epididymal cysts, retained hypotrophic testes and sperm abnormalities in their male offspring. It recently has been suggested that prenatal DES exposure is associated with development of testicular seminoma in humans. Studies of in utero exposure of laboratory animals to DES are few, but previous reports from our laboratory have described several abnormalities in the reproductive tract of the mouse following prenatal DES exposure. To study the possible association of testicular tumors and prenatal DES exposure in mice, pregnant outbred CD-1 mice were injected subcutaneously with daily doses of DES (100 micrograms./kg.) on days nine through 16 of gestation. DES-exposed and age-matched control male mice were sacrificed at 10 to 18 months of age and examined for testicular lesions. In addition to the nonmalignant abnormalities reported in previous studies such as 91% cryptorchidism and degenerative changes, interstitial cell tumors were observed in nine mice among 277 mice treated prenatally with DES. Two of these lesions were benign tumors and five were interstitial cell carcinomas. Rete testis adenocarcinoma was seen also in 5% of these DES-treated animals and is described in another report. The overall incidence of testicular tumors is 8% in DES-exposed male mice. No comparable lesions were seen in 122 control male mice. These results suggest that the testicular lesions that can occur following prenatal DES exposure include neoplasia. The combined prevalence of DES-induced tumors of the corpus testis and rete testis in mice suggests the male offspring may be more at risk for developing carcinoma of the reproductive tract than the female offspring.

A 7 to 8-year-old male African green monkey (Cercopithecus aethiops) was found moribund in his cage. Fluid and antibiotic therapy were administered, but the monkey dies 2 hours later. At necropsy, large septic mural thrombi obstructed the right atrioventricular orifice and the pulmonary outflow tract, and smaller septic thrombi were attached to the leaflets of the pulmonary and mitral valves. Staphylococci were isolated from the large thrombus occluding the atrioventricular orifice. Large abscesses were present in the upper and lower lobes of the right lung, and small, wedge shaped infarcts were present in the lungs and kidneys. The clinical and pathologic findings were consistent with a rapidly progressive form of bacterial endocarditis. This was the only case of vegetative bacterial endocarditis seen at this instituion in 700 necropsies of nonhuman primates.

Three rhesus (Macaca mulatta) and one cynomolgus (M. fascicularis) monkey were euthanized because of positive reactions to intradermal tests with mammalian Old Tuberculin (mOT). All the animals had gross or microscopic lesions consisting of unitized macro- or microgranulomas with central necrosis, involving the lung or mesenteric lymph nodes. Small numbers of acid fast organisms were demonstrated in the lesions. Mycobacterium avium complex, serotype 2 was isolated from three of the cases. The cases were unusual because the lesion morphology and mOT hypersensitivity were more typical of mammalian tuberculosis than avian tuberculosis in monkeys.

Since 1971, 45 of 259 male rhesus monkeys housed in a primate building have died of a chronic and progressive disease characterized by diarrhea, dehydration, weakness, gingivitis, emaciation, and alopecia. The principal necropsy finding in these monkeys, and in eight others killed for experimental purposes, was hypertrophic and hyperplastic mucinous gastropathy involving both the mucosa and submucosa. The toxic agent involved was identified as the polychlorinated biphenyl (PCB), Aroclor 1254. The suspected source of the toxic agent was a concrete sealer used during building construction.

Two groups of adult male African green grivet monkeys (12 animals in each group) were fed atherogenic diets containing 40% of total calories as fat with half of the fat calories derived from lard or menhaden oil and 0.8 mg cholesterol/kcal for a period of 10 months to ascertain the effect of such diets on lipoprotein concentrations, composition, and distribution. After 8 months on this diet, the animals' lipoproteins were isolated from their plasma by ultracentrifugation and agarose column chromatography, and these were characterized chemically. Compared with the lard-fed group, the menhaden oil-fed animals had significantly lower total plasma cholesterol (decreases 33%), HDL cholesterol (decreases 33%) and apo A-I (decreases 30%) concentrations and LDL molecular weights (decreases 15%). Although the fish-oil diets resulted in an average similar reduction of LDL cholesterol concentrations (decreases 34%), the decrease was not statistically significant. The lipid and apoprotein compositions of plasma LDL and HDL were similar between the two groups of animals. The distribution of HDL subfractions was determined by gradient gel electrophoresis and density gradient ultracentrifugation. The fish-oil group had a significant reduction in protein (decreases 46%) of HDL subfractions of intermediate size (80 to 88 A) compared to the lard-fed group. By density gradient ultracentrifugation, there was a 35% to 40% lower concentration of protein in the d = 1.10-1.13 g/ml subfraction of fish-oil-fed monkeys. Thus, feeding fish oil to nonhuman primates results in a selective decrease in the concentration of HDL subfractions of intermediate size and density compared to lard-fed animals.(ABSTRACT TRUNCATED AT 250 WORDS)

Infection with Hemobartonella sp was diagnosed in a colony-born squirrel monkey with normocytic, normochromic anemia and pronounced punctate erythrocytic basophilic stippling on Wright's-Giemsa stained blood films. The diagnosis was confirmed using transmission electron microscopy. Two randomly selected colony-born squirrel monkeys were splenectomized in an effort to activate and detect possible latent hemobartonellosis . One monkey became parasitemic 12 days following splenectomy. The second monkey was inoculated on day 14 with 1 ml of whole blood from an infected, but nonparasitemic monkey and developed overt parasitemia 3 days later (day 17 following splenectomy). Infections in the latter two monkeys were confirmed using scanning electron microscopy.

Adenocarcinoma of the rete testis is an exceptionally rare and malignant testicular neoplasm. Although treatment of pregnant women with diethylstilbestrol (DES) results in reproductive tract abnormalities in their male offspring, increased incidence of testicular tumors has not been verified. However, recently three cases of seminoma have been described in men prenatally exposed to DES, suggesting an association of prenatal DES treatment and the subsequent development of testicular tumors. This report describes the treatment of outbred pregnant CD-1 mice with DES (100 micrograms/kg) on Days 9 through 16 of gestation and its effects on their male offspring. In addition to nonmalignant abnormalities such as retained testes which have been reported in men exposed prenatally to DES, lesions resembling adenocarcinoma of the rete testis were seen in prenatally DES-treated mice at 10 to 18 mo of age (11 of 233; 5%). No comparable lesions were seen in 96 age-matched control male mice. These results suggest an association of prenatal DES exposure and the subsequent development of testicular lesions in the rete testis of mice.

Relatively few cases of myocardial infarction associated with coronary artery atherosclerosis have been described previously in macaques. In this study the authors report the prevalence and characteristics of coronary artery atherosclerosis and myocardial infarction in 10 rhesus (Macaca mulatta) and two cynomolgus (Macaca fascicularis) macaques that were fed atherogenic diets for 16 months or longer. Our findings show clearly that myocardial infarction occurs in macaques with diet-induced atherosclerosis. The frequency seems to be related to the species, composition of the atherogenic diet, and length of time fed the atherogenic diet. The myocardial lesions are remarkably similar to those described in human beings in terms of location and gross and microscopic characteristics. The characteristics of coronary artery atherosclerosis, including the occurrence of thrombosis, severe stenosis, mineralization, atheronecrosis, and sterol clefts, especially in animals fed the atherogenic diets for longer periods of time, also closely resemble those of the arterial lesions found in human beings. The greatest prevalence of myocardial infarcts was found in rhesus monkeys fed a cholesterol-containing diet with 40% of calories supplied by peanut oil and in cynomolgus macaques from Malaya that were fed the same amount of cholesterol with 40% of calories from lard. Electrocardiographic abnormalities as well as the occurrence of unexpected and relatively sudden death in several of these nonhuman primates are also consistent with signs frequently observed in human beings.

Texas health and human services programs could receive nearly half a billion dollars in additional funding over the state's 1990-1991 budget period under proposals being advocated by Comptroller of Public Accounts Bob Bullock. The proposals center on a 7-cent rise in the state's cigarette tax, with equivalent increases in other tobacco taxes. Revenue from the additional tax--estimated at $166 million in 1990-1991--would be directed into state health and human services programs eligible for federal matching funds. By directing the money in this way, the state could receive $274 million in additional federal aid. The $166 million in additional cigarette tax revenue, combined with the $274 million in increased federal aid, would yield $440 million that could be used in a variety of ways in state health and social programs.
