205. THE COPROLOGICAL DIAGNOSIS OF GASTROINTESTINAL NEMATODE INFECTIONS IN SMALL RUMINANTS

Abstract

Although representing a significant economic and welfare burden to the global ruminant livestock industry, GIN infections in small ruminants are often neglected and implementation in research, diagnosis and surveillance of these parasites is still poor, mainly in the matter of diagnostic methods and their use/interpretation. The accurate diagnosis (and interpretation) of GIN directly supports parasite control strategies and is of relevance for investigations into parasite biology, ecology and epidemiology (Roeber et al., 2013). This aspect is now particularly important given the problems associated with anthelmintic resistance (AR) in GIN populations of small ruminants worldwide (Roeber et al., 2013 a,b). Various methods are employed for the ante mortem diagnosis of GIN infections in small ruminants. These include the observation of clinical signs indicative of disease (although non-pathognomonic), coprological diagnosis (faecal egg count – FEC), biochemical and/or serological, and molecular diagnostic approaches (reviewed in Roeber et al., 2013). However, still now, faecal egg count (FEC) techniques remain the most common laboratory methods for the diagnosis of GIN in small ruminants. Also for FEC, widespread standardization of many laboratory techniques does not exist, and most diagnostic, research and teaching facilities apply their own modifications to published protocols (Kassai, 1999). Although these techniques are regarded to be standard diagnostic procedures, there is a lack of detailed studies of diagnostic performance, including the diagnostic sensitivity, specificity and/or repeatability (Roeber et al., 2013). Furthermore, many aspects concerning factors affecting FEC (e.g. season of sampling, sampling period, consistency of faeces, fecundity of worms, etc., as well as interpretation of FEC) have poorly been investigated so far. These are the reasons that motivated me in choosing “The coprological diagnosis of gastrointestinal nematode infections in small ruminants” as topic of my PhD thesis to help optimize the use and interpretation of FEC in small ruminants.

Abstract

Haemonchus contortus is a species of gastrointestinal strongyles of primary concern for sheep. This highly pathogenic, blood-feeding helminth negatively influences animal health, welfare and productivity. In order to elucidate the current scenario in terms of prevalence and intensity of H. contortus infection in sheep farms across Europe, a standardized crosssectional survey was conducted in three pilot areas in Ireland, Switzerland and Italy, all part of the EU funded GLOWORM project. Two consecutive field surveys (in 2012 and 2013) were conducted in the three countries in the same period (August-October) in 259 sheep farms in total. Harmonized, diagnostic procedures (from farm to laboratory) based on pooled samples, the FLOTAC technique and coproculture were used. The georeferenced parasitological results were modelled (at the pilot area level) following a Bayesian geostatistical approach with correction for preferential sampling and accounting for climatic and environmental covariates. The observed H. contortus prevalence rates did vary between the countries showing high values in Switzerland (77%) and Italy (73%) compared to Ireland (4%). Spatial patterns of H. contortus distribution were detected in Switzerland and Italy with a north-south gradient. The latent factor analysis highlighted the importance of seasonality and annual cyclicity within country (particularly in southern Italy), while mean temperature and rainfall dominated between country variations in the prevalence of H. contortus infection.

Abstract

The anthelminthic efficacy of the crude extracts of Hypoestes forskaolii (Vahl) R.Br (Acanthaceae) against gastrointestinal nematodes (GIN) in sheep was investigated using the in vitro egg hatch inhibition assay. Faecal samples were collected from sheep with naturally occurring infection of GIN (Trichostrongylus spp., Chabertia ovina, Cooperia spp., Haemonchus contortus and Teladorsagia spp.). Crude leaf extracts of H. forskaolii was obtained using increasing polarity solvents: n-hexane, chloroform, chloroform:methanol 9:1, methanol. Thiabendazole (0.2 μg/mL and 0.5 μg/mL) was used as a positive control and untreated GIN eggs in deionised water served as the negative control. All the extracts exhibited a weak ovicidal activity against GIN (less than 50% of egg hatch). Noteworthy, the n-hexane extract showed a percentage of inhibition of egg hatching greater than other extracts inhibiting the 30.8% at the concentration of 1 mg/mL showing a dose-dependent effect on nematode eggs hatching. Further studies are needed to investigate the effects of extracts used and to evaluate the ovicidal effects of other extracts of H. forskaolii. [ABSTRACT FROM AUTHOR]
