Artificial intelligence (AI) seeks to simulate the human ability to reason, make decisions, and solve problems. Several AI methodologies have been introduced in forestry to reduce costs and increase accuracy in estimates. We evaluate the performance of Artificial Neural Networks (ANN) in estimating the heights of Araucaria angustifolia (Bertol.) Kuntze (Brazilian pine) trees. The trees are growing in Uneven-aged Mixed Forests (UMF) in southern Brazil and are under different levels of competition. The dataset was divided into training and validation sets. Multi-layer Perceptron (MLP) networks were trained under different Data Normalization (DN) procedures, Neurons in the Hidden Layer (NHL), and Activation Functions (AF). The continuous input variables were diameter at breast height (DBH) and height at the base of the crown (HCB). As a categorical input variable, we consider the sociological position of the trees (dominant–SP1 = 1; codominant–SP2 = 2; and dominated–SP3 = 3), and the continuous output variable was the height (h). In the hidden layer, the number of neurons varied from 3 to 9. Results show that there is no influence of DN in the ANN accuracy. However, the increase in NHL above a certain level caused the model’s over-fitting. In this regard, around 6 neurons stood out, combined with logistic sigmoid AF in the intermediate layer and identity AF in the output layer. Considering the best selected network, the following values of statistical criteria were obtained for the training dataset (R2 = 0.84; RMSE = 1.36 m, and MAPE = 6.29) and for the validation dataset (R2 = 0.80; RMSE = 1.49 m, and MAPE = 6.53). The possibility of using categorical and numerical variables in the same modeling has been motivating the use of AI techniques in different forestry applications. The ANN presented generalization and consistency regarding biological realism. Therefore, we recommend caution when determining DN, amount of NHL, and using AF during modeling. We argue that such techniques show great potential for forest management procedures and are suggested in other similar environments.

Human thymidine phosphorylase (hTP) is overexpressed in several solid tumors and is commonly associated with aggressiveness and unfavorable prognosis. 6-(((1,3-Dihydroxypropan-2-yl)amino)methyl)-5-iodopyrimidine-2,4(1H,3H)-dione (CPBMF-223) is a noncompetitive hTP inhibitor, which has been described as a tumor angiogenesis inhibitor. The present study investigated the effects of CPBMF-223 in a xenograft tumor induced by human colorectal carcinoma cells (HCT-116). Additionally, CPBMF-223 capacity to reduce cell migration, its toxicological profile, and pharmacokinetic characteristics, were also evaluated. The intraperitoneal treatment with CPBMF-223 markedly prevented the relative tumor growth with an efficacy similar to that observed for 5-fluorouracil. Interestingly, number of vessels were significantly decreased in the treated groups. Moreover, CPBMF-223 significantly reduced the migration of cell line HCT-116. In the Ames assay and in an acute oral toxicity test, the molecule did not alter any evaluated parameter. Using the zebrafish toxicity model, cardiac and locomotor parameters were slightly changed. Regarding the pharmacokinetics profile, CPBMF-223 showed clearance of 9.42 L/h/kg after intravenous administration, oral bioavailability of 13.5%, and a half-life of 0.75 h. Our findings shed new light on the role of hTP in colorectal cancer induced by HCT-116 cell in mice, pointing out CPBMF-223 as, hopefully, a promising drug candidate.

Marfan Syndrome (MFS) is a pleiotropic and autosomal dominant condition caused by pathogenic variants in FBN1. Although fully penetrant, clinical variability is frequently observed among patients and there are only few genotype-phenotype correlations described so far. Here, we describe the generation and characterization of hiPSC lines derived from two unrelated MFS patients harboring heterozygous variants in FBN1. Human iPSCs were obtained from erythroblasts reprogrammed with episomal vectors carrying the reprogramming factors OCT4, SOX2, KLF4, c-MYC and LIN-28, and characterized according to established criteria. Differentiated cells demonstrated different patterns of fibrillin-1 expression suggesting different molecular mechanisms between the two patients.

Marfan Syndrome (MFS) is an autosomal dominant connective tissue disorder caused by mutations in the FBN1 gene. To investigate the molecular mechanisms of pathogenesis for the syndrome, we genetically modified the FBN1 gene in a line of induced pluripotent stem cells (hiPSCs) derived from a healthy donor using the CRISPR/Cas9 gene editing technology. The sublines described here were characterized according to established criteria and were shown to maintain pluripotency, three germ layer differentiation potential and genomic integrity. These clones can now be used to better understand the pathogenesis of MFS in different cell types.

São Luiz do Paraitinga ficou conhecida nacionalmente depois do desastre que a assolou em 2010. Parte de seu casario tombado e da igreja Matriz ruíram ou ficaram gravemente avariados por uma enchente, obrigando várias instituições públicas a trabalharem em conjunto para minimizar os danos. Nesse contexto, ocorreu o tombamento emergencial da cidade em âmbito federal, cuja justificativa principal foi a filiação de seu traçado e de sua arquitetura aos preceitos iluministas em voga na época, que nortearam uma série de outros núcleos na segunda metade do século XVIII. Embora o Iphan venha atuando na cidade desde a década de 1950, e o Condephaat desde o momento de sua criação, na década de 1960, poucos estudos procuraram entender a história de seu patrimônio edificado. Assim, passada uma década do desastre e de seu tombamento federal, este artigo tem o objetivo de reler parte da documentação primária remanescente e lançar novos olhares sobre a criação e desenvolvimento dessa cidade, hoje patrimônio nacional.

Hypertension is a complex multifactorial disease characterized by a chronic increase of arterial pressure. Ninety percent of the cases are idiopathic and thus classified as essential hypertension. Uncontrolled arterial pressure has devasting consequences including cardiac insufficiency, stroke, dementia, chronic renal disease, ischemic heart disease and death. The hiPSC lines described here from six hypertensive patients and three controls were characterized according to established criteria and were shown to maintain pluripotency, differentiation into the three germ layers and genomic integrity. These cell lines can contribute to the understanding of the molecular mechanisms involved in hypertension in different cell types.

The dinuclear polypyridyl complex [{Ru(bipy)2}2(µ-L)](PF6)4, where bipy = 2,2'-bipyridine and L = 2,6-bis[(1,10-phenantroline-5-amine)methyl]pyridine, was prepared and characterized by UV-Vis and infrared spectroscopy. Geometry optimization revealed the possibility of three conformers, and time-dependent density functional theory calculations provided access to the correct excited state composition and unambiguous assignment of the characteristic metal-ligand charge transfer transitions. Spotted fever is the main rickettsial disease in Brazil. We report 12 cases of human parasitism by Amblyomma parkeri in the Atlantic rainforest, an area of Brazil to which spotted fever is endemic. Nine of the ticks were infected with Candidatus Rickettsia paranaensis.

HIGHLIGHTS This scoping review summarizes the findings of clinical trials using methylene blue (MB) for the treatment of various health conditions. This research method allowed mapping main findings, clarifying research topics, and identifying gaps in the literature.
