Disruption of DNA replication, recombination, and repair (DNA-RRR) systems has been hypothesized to cause highly elevated nucleotide substitution rates and genome rearrangements in the plastids of angiosperms, but this theory remains untested. To investigate nuclear-plastid genome (plastome) coevolution in Geraniaceae, four different measures of plastome complexity (rearrangements, repeats, nucleotide insertions/deletions, and substitution rates) were evaluated along with substitution rates of 12 nuclear-encoded, plastid-targeted DNA-RRR genes from 27 Geraniales species. Significant correlations were detected for nonsynonymous (dN) but not synonymous (dS) substitution rates for three DNA-RRR genes (uvrB/C,why1, andgyrA) supporting a role for these genes in accelerated plastid genome evolution in Geraniaceae. Furthermore, correlation between dN of uvrB/C and plastome complexity suggests the presence of nucleotide excision repair system in plastids. Significant correlations were also detected between plastome complexity and 13 of the 90 nuclear-encoded organelle-targeted genes investigated. Comparisons revealed significant acceleration of dN in plastid-targeted genes of Geraniales relative to Brassicales suggesting this correlation may be an artifact of elevated rates in this gene set in Geraniaceae. Correlation between dN of plastid-targeted DNA-RRR genes and plastome complexity supports the hypothesis that the aberrant patterns in angiosperm plastome evolution could be caused by dysfunction in DNA-RRR systems. Purpose: This article reviews the institution of mock code simulation training to improve registered nurses responder performance at a trauma 1 teaching hospital. Method: An in-situ mock code is held on random medical-surgical/telemetry units every month. Participants are asked to respond to 2 different scenarios using a 3-G Sim Man and the unit's emergency equipment. Each session is followed by debriefing, postsurveys, and evaluation. The time to cardiopulmonary resuscitation (CPR) and defibrillation is recorded for analysis. The participant's level of confidence is measured before and after simulation. Results: More than 250 nurses have participated in the Five Alive course since its initiation in March 2011. The participants have expressed more confidence in recognizing a declining patient health status after the simulation. They have also demonstrated a marked improvement in time to CPR and defibrillation. Conclusions: Initial findings reveal that in-situ medical emergency team/code simulation followed by debriefing improved the performance of responders. We are currently offering the Five Alive program monthly during four 1-hour sessions and have found this program to be an effective training tool. There is one challenge to offering the program on the off shift and weekends. That is finding a time that is not too disruptive to patient care. [ABSTRACT FROM AUTHOR]

Plastid genomes in the flowering plant family Geraniaceae are known to be highly rearranged based on complete sequences representing the four major genera Erodium, Geranium, Monsonia, and Pelargonium. In this paper we report on the genome sequence of a second species of Erodium, E. carvifolium, representing the second major clade (clade II) in the phylogeny of this genus. Comparison of this genome sequence to the previously published sequence of E. texanum from clade I demonstrates that the plastid genomes of these two species encode the same number of proteins but differ greatly in their relative degree of rearrangement; 14 kb of additional sequence in E. texanum contains complex repeats associated with rearrangement endpoints, whereas the plastid genome of E. carvifolium is streamlined at 116 kb and displays no unique alterations in gene order. Furthermore, these species from both major clades of Erodium contain intact NADH dehydrogenase ( ndh) genes, but the 11 ndh genes are represented as pseudogenes in a small clade of 13 species. It is unclear whether plastid-encoded ndh genes have been lost entirely or functionally transferred to the nucleus. This is the third report of the absence of functional ndh genes, and the current study describes the most recent loss of these genes among photosynthetic seed plants and the second such loss among angiosperms. The other ndh losses from Pinaceae/Gnetales and Orchidaceae are much more ancient. Comparative biochemistry between Erodium species with and without plastid-encoded ndh genes may elucidate changes in photosynthetic function and the role of the Ndh complex. [ABSTRACT FROM AUTHOR]

Although post-combustion emissions from power plants are a major source of air pollution, they contain excess CO2 that could be used to fertilize commercial greenhouses and stimulate plant growth. We addressed the combined effects of ultrahigh [CO2] and acidic pollutants in flue gas on the growth of Alternanthera philoxeroides. When acidic pollutants were excluded, the biomass yield of A. philoxeroides saturated near 2000μmol mol−1 [CO2] with doubled biomass accumulation relative to the ambient control. The growth enhancement was maintained at 5000μmol mol−1 [CO2], but declined when [CO2] rose above 1%, in association with a strong photosynthetic inhibition. Although acidic components (SO2 and NO2) significantly offset the CO2 enhancement, the aboveground yield increased considerably when the concentration of pollutants was moderate (200 times dilution). Our results indicate that using excess CO2 from the power plant emissions to optimize growth in commercial green house could be viable. [Copyright &y& Elsevier]

The microtubule-associated protein tau pathologically accumulates and aggregates in Alzheimer's disease (AD) and other tauopathies, leading to cognitive dysfunction and neuronal loss. Molecular chaperones, like small heat-shock proteins (sHsps), can help deter the accumulation of misfolded proteins, such as tau. Here, we tested the hypothesis that the overexpression of wild-type Hsp22 (wtHsp22) and its phosphomimetic (S24,57D) Hsp22 mutant (mtHsp22) could slow tau accumulation and preserve memory in a murine model of tauopathy, rTg4510. Our results show that Hsp22 protected against deficits in synaptic plasticity and cognition in the tauopathic brain. However, we did not detect a significant change in tau phosphorylation or levels in these mice. This led us to hypothesize that the functional benefit was realized through the restoration of dysfunctional pathways in hippocampi of tau transgenic mice since no significant benefit was measured in non-transgenic mice expressing wtHsp22 or mtHsp22. To identify these pathways, we performed mass spectrometry of tissue lysates from the injection site. Overall, our data reveal that Hsp22 overexpression in neurons promotes synaptic plasticity by regulating canonical pathways and upstream regulators that have been characterized as potential AD markers and synaptogenesis regulators, like EIF4E and NFKBIA. The plastid genome of Trifolium subterraneum is 144,763 bp, about 20 kb longer than those of closely related legumes, which also lost one copy of the large inverted repeat (IR). The genome has undergone extensive genomic reconfiguration, including the loss of six genes ( accD, infA, rpl22, rps16, rps18, and ycf1) and two introns ( clpP and rps12) and numerous gene order changes, attributable to 14–18 inversions. All endpoints of rearranged gene clusters are flanked by repeated sequences, tRNAs, or pseudogenes. One unusual feature of the Trifolium subterraneum genome is the large number of dispersed repeats, which comprise 19.5% (ca. 28 kb) of the genome (versus about 4% for other angiosperms) and account for part of the increase in genome size. Nine genes ( psbT, rbcL, clpP, rps3, rpl23, atpB, psbN, trnI-cau, and ycf3) have also been duplicated either partially or completely . rpl23 is the most highly duplicated gene, with portions of this gene duplicated six times. Comparisons of the Trifolium plastid genome with the Plant Repeat Database and searches for flanking inverted repeats suggest that the high incidence of dispersed repeats and rearrangements is not likely the result of transposition. Trifolium has 19.5 kb of unique DNA distributed among 160 fragments ranging in size from 30 to 494 bp, greatly surpassing the other five sequenced legume plastid genomes in novel DNA content. At least some of this unique DNA may represent horizontal transfer from bacterial genomes. Abstract  Harvesting fallen needles (straw) in loblolly pine (Pinus taeda L.) plantations provides forest owners with a substantial source of income, but this practice and the type of fertilizer used to replenish nutrients removed with straw harvests may alter soil microbiological and physical properties. This study was conducted to explore the influence of annual straw harvesting, fertilization, and fertilizer source (inorganic vs. broiler poultry litter) in a loblolly pine plantation in the mid-south USA on: (1) soil microbial biomass C, (2) soil dehydrogenase activity, and (3) key soil physical properties (soil strength, bulk density, porosity, aeration, soil moisture content, organic matter, and available water holding capacity). All treatments that included straw harvesting increased bulk density and reduced soil porosity. Annual straw harvesting conducted with annual fertilization of inorganic nitrogen and phosphorus fertilization was associated with the most pronounced increases in soil strength and reductions in organic matter, available water holding capacity, microbial biomass C, and dehydrogenase activity.
