Incorporation mechanism of sulfate groups in major calcium carbonates (calcite, aragonite and vaterite) is investigated using first-principles quantum-mechanical calculations. For each mineral, the stable structure of the substituted site is determined. In calcite and aragonite, a tilting of the sulfate-group with respect to the orientation of planar carbonate groups is observed. The theoretical vibrational properties of the sulfate-bearing carbonates are determined together with those of anhydrite (CaSO4) and isolated SO2 molecule. Comparison with experimental data supports the substitution of sulfate for carbonate groups in carbonate minerals. The energetic of sulfate incorporation is increasingly unfavorable in the order vaterite, calcite, and aragonite. Regarding isotopic fractionation properties, our calculations suggest that the equilibrium S-34/S-32 isotopic fractionation factor between vaterite and calcite is small (similar to 0.5% at 0 degrees C). A slightly larger fractionation factor (similar to 1% at 0 degrees C) is expected between aragonite and calcite; whereas a negative fractionation (similar to-1.5% at 0 degrees C) is expected between anhydrite and calcite. Finally, it is suggested that the isotopic fractionation between calcite and aqueous sulfate is smaller than 4%.

Transcytosis of polymeric Ig (pIg) by mucosal epithelial cells is mediated by the polymeric Ig receptor (pIgR). Here we describe the characterization of a 3095-bp mouse pIgR cDNA, which encodes a protein of 771 amino acids. Northern blot analysis detected a single mouse pIgR transcript of 3.9 kb, expressed at high levels in small intestine and liver, and at low levels in lung. Alignment of the amino acid sequences of mouse, rat, human, bovine, and rabbit pIgR revealed that functional regions of the molecule are conserved across species. In the extracellular region, conserved motifs include: a 23-amino acid pIg-binding site, 11 intradomain disulfide bonds, consensus sites for N-glycosylation, and a putative cleavage site at which the extracellular region of pIgR (secretory component) is released from the plasma membrane. A 10-amino acid sequence within the transmembrane region is highly conserved, possibly reflecting a mechanism for transmitting signals from the extracellular region to the cytoplasmic tail. Conservation within the cytoplasmic tail of pIgR is clustered in motifs that mediate polarized sorting, endocytosis, and transcytosis.

National audience; Le modèle Safran-Idba-Modcou (SIM) permet d’analyser les sécheresses selon trois indicateurs : les précipitations, l’humidité du sol et le débit des rivières. La sécheresse de 2011 apparaît comme la plus sévère depuis le début du XXe siècle en termes d’expansion spatiale et d’intensité moyenne. Une aggravation de la fréquence et de l’intensité des événements extrêmes liés au déficit d’humidité des sols est à attendre dans la première moitié du XXIe siècle. À la fin du siècle (années 2080), des sécheresses météorologiques plus fortes apparaissent également. Des événements tels qu’observés en 1976 sont alors fréquemment dépassés.

International audience; French droughts: lessons learned from the past and expected consequences of climate change

Recent experimental studies revealed an unprecedented bimodal distribution of arsenate at the hematite (1 (1) over bar2) surface with a simultaneous adsorption of inner-sphere and outer-sphere complexes. In the present study, first-principles calculations based on density-functional theory were performed to provide detailed insights into the structural and electronic properties of such inner-sphere and outer-sphere adsorption complexes on two hydroxylated terminations of the hematite (1 (1) over bar2) surface. For bidentate corner-sharing complexes, the predicted most stable adsorption configurations display interatomic distances in good agreement with EXAFS-derived data (i.e. As-Fe distances of similar to 3.3 angstrom). Our calculations also suggest that edge-sharing bidentate complexes can form on ideal (1 (1) over bar2) hematite surfaces and do not necessarily involve step edges. These edge-sharing complexes would display two As-Fe distances at about 2.85 and 3.45 angstrom, instead of the unique short As-Fe contribution that is usually considered for interpreting EXAFS data. For outer-sphere complexes, the predicted most favorable adsorption configurations indicate that strong hydrogen bonds as well as electrostatic forces stabilize arsenate at the hematite surface. Although not investigated here, the interfacial water structure may also contribute to stabilize further OS arsenate complexes.

[Departement_IRSTEA]Eaux [TR1_IRSTEA]ARCEAU; Le projet ClimSec a étudié l'impact du changement climatique sur la sécheresse et sur l'eau du sol en France en s'appuyant sur la réanalyse de la chaîne hydrométéorologique SAFRAN-ISBA-MODCOU (SIM) depuis 1958. Des indices standardisés, " Standardized Precipitation Index " (SPI) et " Standardized Soil Wetness Index " (SSWI), ont été utilisés pour caractériser les différents types de sécheresse (météorologique et agricole) à partir d'une climatologie de 50 ans. En 2011, ces indices ont été adaptés pour le suivi hydrologique opérationnel et pour répondre aux besoins de qualification de la sécheresse printanière exceptionnelle de cette année. Ces indices ont également été calculés en climat futur à partir des différentes projections climatiques régionalisées disponibles en métropole. Trois expériences particulières en matière de scénarios socio-économiques, de modèles climatiques et de méthodes de descente d'échelle ont permis d'apprécier l'importance relative des différentes sources d'incertitude dans l'évolution attendue des sécheresses. Ces données ont aussi permis de préciser les conditions de l'évolution temporelle des sécheresses, avec une aggravation plus rapide et plus intense des événements liés au déficit d'humidité du sol plutôt qu'au déficit de précipitation. Les projections climatiques indiquent surtout que notre pays risque de connaître, lors de la seconde moitié du XXIe siècle, des sécheresses quasi continues et de grande intensité, totalement inconnues dans le climat actuel. / The ClimSec project has studied the impact of climate change on drought and soil water over France by using a climatological reanalysis of the SAFRAN/ISBA/MODCOU suite (SIM) since 1958. Standardized drought indices for precipitation (SPI) and soil moisture (SSWI) have been defined for research purposes to characterize the various kinds of events. They were then adapted for operational hydrological monitoring and used to assess the exceptional drought of spring 2011. These indices were also calculated for future climate from the various regionalized climate projections available over France. Three particular experiments in socio-economic scenarios, climate models and downscaling methods have been run to estimate the relative importance of the different uncertainties in drought evolution. The assessment of 21st century drought evolution shows a much earlier and more intense occurrence of changes for agricultural droughts linked to soil moisture deficits than for meteorological drought linked with precipitation deficits. Climate projections suggest that France could be affected on the second half of the 21st century by a quasi-continuous drought with a strong intensity, totally unknown in present climate.

Vibrational properties of diaspore, alpha-AlOOH, have been re-investigated using room-temperature single-crystal Raman spectroscopy and low-temperature powder infrared (IR) transmission spectroscopy. First-principles harmonic calculations based on density functional theory provide a convincing assignment of the major Raman peaks and infrared absorption bands. The large width of the Raman band related to OH stretching modes is ascribed to mode-mode anharmonic coupling due to medium-strength H-bonding. Additional broadening in the powder IR spectrum arises from depolarization effects in powder particles. The temperature dependence of the IR spectrum provides a further insight into the anharmonic properties of diaspore. Based on their frequency and temperature behavior, narrow absorption features at similar to 2,000 cm(-1) and anti-resonance at similar to 2,966 cm(-1) in the IR spectrum are interpreted as overtones of fundamental bending bands.

Partnerships on water resource management in France

Les études menées jusqu’à présent se sont surtout focalisées sur l’évolution des précipitations ou le débit des rivières. L’objectif du projet ClimSec était de compléter ces analyses par une étude de l’évolution passée et future des réserves en eau des couches superficielles des sols en mettant l’accent sur les évolutions attendues à la fin du siècle mais aussi aux échéances proches. L’emploi d’un modèle physique atmosphère-sol-végétation pour l’étude de la ressource en eau plutôt qu’une approche paramétrique classique basée sur l’évapotranspiration (ETP), est une des originalités de cette étude. Le modèle utilisé Safran-Isba-Modcou (SIM), fruit de travaux de recherche communautaire (Météo-France, Mines Paris Tech, Cemagref) entrepris depuis plus de 20 années, présente l’avantage d’être intégré dans la production opérationnelle quotidienne de Météo-France. La première partie de cet article présente les indicateurs retenus pour la caractérisation des sècheresses et leur application pour le suivi hydrologique opérationnel en prenant l’exemple de la sècheresse du printemps 2011. A partir de plusieurs projections climatiques sur le XXIème siècle, préalablement régionalisées sur la France, la seconde partie vise à préciser l’évolution attendue des sècheresses et les principales incertitudes associées.

[Departement_IRSTEA]Eaux [TR1_IRSTEA]ARCEAU; Le projet CLIMSEC mené par la Direction de la Climatologie de Météo-France avec le soutien de la Fondation MAIF a permis de caractériser la typologie des sécheresses en métropole sur la période 1958-2008 et d'établir un diagnostic sur leurs évolutions attendues au cours du XXIe siècle. L'étude en climat actuel s'est appuyée sur une modélisation rétrospective de l'humidité du sol sur 50 ans avec la chaîne hydrométéorologique Safran-Isba-Modcou (SIM). La réanalyse SIM établie dans le cadre du projet a fait l'objet d'une évaluation particulière pour vérifier son homogénéité temporelle et a permis d'établir une climatologie à haute résolution de l'humidité des sols en métropole. Les différents types de sécheresse (météorologique, agricole, hydrologique) ont été caractérisés à partir d'indicateurs standardisés. Ces indicateurs permettent à la fois de garantir la cohérence spatiale des analyses à différents pas de temps et la détermination des caractéristiques des évènements: début, fin, sévérité, magnitude. Cet outil a été d'abord utilisé à des fins de description comparative des épisodes marquants de sécheresse rencontrés au cours des 50 dernières années (tels que l'été 1976, les années 1989-1990, l'été 2003) en termes de caractéristiques moyennes (durée et surface moyennes, magnitude totale) mais aussi de déficits records sur la période considérée. Une climatologie spatialisée des sécheresses en métropole a ensuite été établie dont la valeur et l'originalité ont été notamment reconnues par l'attribution du prix Norbert Gerbier Mumm de l'OMM en 2010. Elle a mis en évidence les disparités régionales notamment en termes de durée, de sévérité moyenne puis a permis une qualification des événements extrêmes observés sur la période considérée. La deuxième partie de l'étude a été consacrée à l'évaluation des caractéristiques futures des sécheresses sous changement climatique à l'aide des indices standardisés développés dans le projet et en s'appuyant sur les différentes projections climatiques régionalisées disponibles en métropole et issus des travaux de l'AR4 du GIEC. La caractérisation des sécheresses a pris particulièrement en compte les différentes sources d'incertitude, liées aux modèles climatiques globaux, aux scénarios socio-économiques et aux méthodes de régionalisation. La pertinence des jeux de données et des différentes méthodes utilisées, notamment en contexte climatique non stationnaire, a fait l'objet d'une attention particulière. Les incertitudes sur l'évolution des caractéristiques des sécheresses ont été décrites à la fois dans leurs dimensions moyennes mais aussi en terme de variabilité régionale et saisonnière. Au final, les convergences des résultats issus des différentes projections climatiques permettent d'apporter un diagnostic complet sur la compréhension des éléments majeurs de l'aggravation des sécheresses au cours du XXIe siècle et une mise en perspective dans le cadre des démarches pour l'adaptation au changement climatique.

There is actually a surge for reintroducing biodiversity in agricultural systems in order to reduce chemical inputs, suppress pests, and close biogeochemical cycles (Malézieux et al., 2009). The use of cover-crops is a promising way to reintroduce biodiversity into the fields. Cover-crops have the potential to decrease chemical use against weeds (by competition) and pests (by increasing in natural enemies). To decrease herbicide use, suitable plants must be able to grow in appropriate conditions, to do not compete the cultivated crop for nutrients or water, but should compete weeds for light and space. There is a trade-off between these objectives. Banana is a semi-perennial crop, each plant develops at its own rhythm leading to an unsynchronized plant population in three years; canopy and nitrogen demand of the crop follow this unsynchronized pattern (Tixier et al., 2004). Banana cropping systems remain based on bare soil management and a large amount of herbicides is used. In tropical environment, the growth of weeds and cover-crop is complex because it is not constrained by seasons; a constant growth is possible due to relatively constant climate. In these conditions, variation in radiation due to canopy closure is one of the major drivers of their growth. First, we present the evaluation of the impact of the cover-crop on the yield in the case of two graminaceous cover-crops. Secondly, we present results of the evaluation of 11 species of potential cover-crop using measures under different levels of shade. We used a simulation model to contextualize their growth and conclude on the most appropriated species to intercrop with banana.

This paper presents a research based on data resulting from a regional project in France. The aim of the project is to evaluate the impact of the use of mathematics E-Exercises-bases (EEB) in secondary schools. Our hypothesis is that the integration of EEB disturbs the usual habits of teaching. We thus examine this integration by taking as privileged dimension the teachers’ practices. Using the model of the activity developed by Y. Engeström we study the genesis of uses of EEB among the teachers involved in the regional project. The model permits to split the situation in several poles and to inquire on these poles and their interconnections. Results show that a student autonomous work is difficult to establish and teachers have to cope with new attitudes that are not so plain., Este artigo apresenta uma investigação baseada em dados obtidos num projecto regional em França. O objectivo do projecto é avaliar o impacto do uso de bases-de-e-exercícios (EEB) de Matemática em escolas secundárias. A nossa conjectura é que a integração de EEB perturba a prática habitual de ensino. Examinamos esta integração focando-nos em particular sobre as práticas de ensino do professor. Adoptamos o modelo da actividade desenvolvido por Y. Engeström para estudar a génese dos usos dos EEB pelos professores envolvidos no projecto regional. Este modelo permite dividir a situação em vários pólos e obter informações sobre estes pólos e suas interconexões. Os resultados mostram que o trabalho autónomo dos alunos é difícil de desenvolver e que os professores têm que lidar com novas atitudes que não são tão simples.

The gene clusters encoding the lipooligosaccharide biosynthesis glycosyltransferases from Campylobacter jejuni have previously been divided in eight classes based on their genetic organization. Here, three variants of the beta1,3-galactosyltransferase CgtB from two classes were purified as fusions with the maltose-binding protein (MalE) from Escherichia coli and their acceptor preference was determined. The acceptor preference of each CgtB variant was directly related to the presence or absence of sialic acid in the acceptor, which correlated with the core oligosaccharide structure in vivo. The three variants were evaluated for their ability to use a derivitized monosaccharide, a GM2 ganglioside mimic, a GA2 ganglioside mimic as well as a peptide containing alpha-linked GalNAc. This characterization shows the flexibility of these galactosyltransferases for diverse acceptors. The CgtB variants were engineered via carboxy-terminal deletions and inversion of the gene fusion order. The combination of a 20 to 30 aa deletion in CgtB followed by MalE at its carboxy terminus significantly improved the glycosyltransferase activity (up to a 51.8-fold increase of activity compared to the full length enzyme) in all cases regardless of the acceptor tested. The improved enzyme CgtB(OH4384)DeltaC-MalE was used to galactosylate a glyco-peptide acceptor based on the interferon alpha2b protein O-linked glycosylation site as confirmed by the CE-MS analysis of the reaction products. This improved enzyme was also used successfully to galactosylate the human therapeutic protein IFNalpha2b[GalNAcalpha]. This constitutes the first report of the in vitro synthesis of the O-linked T-antigen glycan on a human protein by a bacterial glycosyltransferase and illustrates the potential of bacterial glycosyltransferases as tools for in vitro glycosylation of human proteins of therapeutic value
